Emergence of Endocytosis-Dependent mGlu1 LTD at Nucleus Accumbens Synapses After Withdrawal From Cocaine Self-Administration.

Extended-access cocaine self-administration induces a progressive intensification of cue-induced drug craving during withdrawal termed “incubation of cocaine craving”. Rats evaluated after >1 month of withdrawal (when incubation of craving is robust) display alterations in excitatory synapses onto medium spiny neurons (MSNs) of the nucleus accumbens (NAc), including elevated levels of Ca2+-permeable AMPA receptors (CP-AMPAR) and a transition from group I metabotropic glutamate receptor (mGluR) mGlu5- to mGlu1-mediated synaptic depression. It is important to further characterize the emergent form of mGlu1-mediated synaptic depression because it has been demonstrated that mGlu1 stimulation, by normalizing CP-AMPAR transmission, reduces cue-induced cocaine craving. In the present study, we conducted whole-cell patch-clamp recordings in NAc core MSNs, comparing rats that underwent >35 days of withdrawal from cocaine self-administration to control rats that had self-administered saline. Bath application of the nonselective group I mGluR agonist dihydroxyphenylglycine (DHPG) produced a transient mGlu5-mediated synaptic depression in saline controls, whereas a persistent mGlu1-mediated synaptic depression emerged in cocaine rats. This form of long-term depression (LTD) was abolished by the inclusion of dynamin inhibitory peptide (DIP) in the recording electrode, indicating that it is mediated by removal of CP-AMPARs through a dynamin-dependent endocytosis mechanism. We further showed that CP-AMPAR endocytosis is normally coupled to the PICK1-mediated insertion of Ca2+-impermeable AMPARs (CI-AMPAR). Interestingly, this coupling is not obligatory because disruption of PICK1-mediated CI-AMPAR insertion with pep2-EVKI spared mGlu1-mediated CP-AMPAR endocytosis. Collectively, these results reveal similarities but also differences from mGlu1-LTD observed in other brain regions, and further our understanding of a form of plasticity that may be targeted to reduce cue-induced craving for cocaine and methamphetamine.

Early-life adversity selectively impairs α2-GABAA receptor expression in the mouse nucleus accumbens and influences the behavioral effects of cocaine

Haplotypes of the Gabra2 gene encoding the α2-subunit of the GABAA receptor (GABAAR) are associated with drug abuse, suggesting that α2-GABAARs may play an important role in the circuitry underlying drug misuse. The genetic association of Gabra2 haplotypes with cocaine addiction appears to be evident primarily in individuals who had experienced childhood trauma. Given this association of childhood trauma, cocaine abuse and the Gabra2 haplotypes, we have explored in a mouse model of early life adversity (ELA) whether such events influence the behavioral effects of cocaine and if, as suggested by the human studies, α2-GABAARs in the nucleus accumbens (NAc) are involved in these perturbed behaviors. In adult mice prior ELA caused a selective decrease of accumbal α2-subunit mRNA, resulting in a selective decrease in the number and size of the α2-subunit (but not the α1-subunit) immunoreactive clusters in NAc core medium spiny neurons (MSNs). Functionally, in adult MSNs ELA decreased the amplitude and frequency of GABAAR-mediated miniature inhibitory postsynaptic currents (mIPSCs), a profile similar to that of α2 “knock-out” (α2−/−) mice. Behaviourally, adult male ELA and α2−/− mice exhibited an enhanced locomotor response to acute cocaine and blunted sensitisation upon repeated cocaine administration, when compared to their appropriate controls. Collectively, these findings reveal a neurobiological mechanism which may relate to the clinical observation that early trauma increases the risk for substance abuse disorder (SAD) in individuals harbouring haplotypic variations in the Gabra2 gene.

Estradiol impacts the endocannabinoid system in female rats to influence behavioral and structural responses to cocaine

Compared with men, women show enhanced responses to drugs of abuse, and consequently are thought to be more vulnerable to addiction. The ovarian hormone estradiol has emerged as a key facilitator in the heightened development of addiction in females. These actions of estradiol appear mediated by estrogen receptor (ER) activation of metabotropic glutamate receptor type 5 (mGluR5). However, the downstream effectors of this ER/mGluR5 signaling pathway are unknown. Here we investigate whether cannabinoid 1 receptor (CB1R) activation is a part of the mechanism whereby estradiol influences behavioral and synaptic correlates of addiction. Following repeated cocaine administration, estradiol-treated ovariectomized rats exhibited both sensitized locomotor responses and decreases in the dendritic spine density of nucleus accumbens core medium-spiny neurons in comparison to oil-treated controls. Both effects of estradiol were blocked by AM251, a CB1R inverse agonist. These results indicate that part of the signaling mechanism through which estradiol impacts behavioral and synaptic correlates of addiction in female rats requires activation of CB1Rs.

Enhanced cocaine-induced locomotor sensitization and intrinsic excitability of NAc medium spiny neurons in adult but not in adolescent rats susceptible to diet-induced obesity.

Basal and diet-induced differences in mesolimbic function, particularly within the nucleus accumbens (NAc), may contribute to human obesity; these differences may be more pronounced in susceptible populations. We examined differences in cocaine-induced behavioral plasticity in rats that are susceptible vs. resistant to diet-induced obesity and basal differences in striatal neuron function in adult and in adolescent obesity-prone and obesity-resistant rats. Susceptible and resistant outbred rats were identified based on "junk-food" diet-induced obesity. Then, the induction and expression of cocaine-induced locomotor sensitization, which is mediated by enhanced striatal function and is associated with increased motivation for rewards and reward-paired cues, were evaluated. Basal differences in mesolimbic function were examined in selectively bred obesity-prone and obesity-resistant rats (P70-80 and P30-40) using both cocaine-induced locomotion and whole-cell patch clamping approaches in NAc core medium spiny neurons (MSNs). In rats that became obese after eating junk-food, the expression of locomotor sensitization was enhanced compared to non-obese rats, with similarly strong responses to 7.5 and 15mg/kg cocaine. Without diet manipulation, obesity-prone rats were hyper-responsive to the acute locomotor-activating effects of cocaine, and the intrinsic excitability of NAc core MSNs was enhanced by ∼60% at positive and negative potentials. These differences were present in adult, but not adolescent rats. Post-synaptic glutamatergic transmission was similar between groups. Mesolimbic systems, particularly NAc MSNs, are hyper-responsive in obesity-prone individuals, and interactions between predisposition and experience influence neurobehavioral plasticity in ways that may promote weight gain and hamper weight loss in susceptible rats.

Editorial: structural plasticity induced by drugs of abuse.

The roots of addiction are often attributed to the ability of repeated drug use to compromise proper functioning of the central nervous system. Drug-induced functional changes in the brain occur on many levels, including altered expression of specific genes via genomic and epigenetic mechanisms, induction of synaptic plasticity and other cellular adaptations, and volumetric changes in discrete brain regions (Luscher and Malenka, 2011). These alterations can be drug class-specific and occur in both neuronal and non-neuronal cell populations. Drug-induced plasticity, or “rewiring” of the brain contributes to the development, maintenance, and persistence of the addicted state (Kalivas and O'Brien, 2008). The goal of this Research Topic is to assimilate recent findings related to plasticity and structural alterations produced by drug of abuse in neurons, glia, and other cell types of the brain. The mesolimbic dopamine system, originating in the ventral tegmental area (VTA) of the midbrain and projecting rostrally to the nucleus accumbens (NAc) and prefrontal cortex (PFC), mediates the acute reinforcing and incentive salience of abused drugs, as well as their aversive properties. As reviewed by Vashchinkina et al. (2014) mesolimbic dopamine neurons are highly regulated by intrinsic and extrinsic inhibitory GABAergic neurons. Vashchinkina et al. (2014) hypothesize that abused drugs induce structural and functional mesolimbic adaptations differentially via endogenous (e.g., THIP and neurosteroids) vs. exogenous (e.g., benzodiazepines and alcohol) modulators of GABAA receptors, as well as via synaptic vs. extrasynaptic GABAA receptors in the VTA. Collo et al. (2014) review evidence that drug-induced plasticity in mesolimbic dopaminergic neurons is mediated by common dopamine and brain-derived neurotrophic factor (BDNF) signaling pathways, specifically those recruiting MEK-ERK1/2, and PI3K-Akt-mTOR. Cadet and Bisagno (2014) review evidence for substantial plasticity in non-neuronal cell types in brain reward circuitry, including changes in astrocytic glutamate transporter expression, increases in pro-inflammatory cytokine production by microglia, and dysregulated oligodendrocytic myelin production. This latter topic is further elaborated on by Somkuwar et al. (2014) in their review of the proteoglycan neuron-glial antigen 2 (NG2), its expression by oligodendrocyte progenitor cells in the brain reward circuitry, and its interaction with stress-related neuromodulators. In addition, Bajo et al. (2015) provide evidence that interleukin-1β alters both basal and ethanol-facilitated GABAergic transmission in the central nucleus of the amygdala, part of the extended amygdala circuitry which is implicated in ethanol's central effects. Drugs of abuse also alter the dynamics and microstructure of both dendrites and dendritic spines, which can be interpreted as cellular (mal)adaptations that reinforce the addiction cycle (Luscher and Malenka, 2011; Gipson et al., 2014). DePoy et al. (2014) show that repeated exposure of early adolescent mice to cocaine produces lasting reductions in orbitofrontal cortex dendritic arbor length and complexity and as well as impaired reversal learning. In addition, these investigators show that while mice carrying a heterozygous deletion of the actin cytoskeleton stabilizing protein p190RhoGAP exhibit enhanced vulnerability to cocaine-induced hyperlocomotion, orbitofrontal dendritic complexity in cocaine-naive mice is intact. These findings suggest that orbitofrontal dendrite structure is impacted by repeated cocaine during adolescence, but pre-existing structural dendritic deficiencies do not account for increased behavioral sensitivity to this drug. Glutamate is the predominant excitatory amino acid in the central nervous system and mediates both normal and maladaptive cellular plasticity. Several articles in this Research Topic provide novel findings on the role of glutamate in addictive processes. Weiland et al. (2015) demonstrate that antibiotics with the glutamate and GABA modulating properties (ceftriaxone and cefazolin, respectively) attenuate cue-primed reinstatement of alcohol-seeking. Griffin et al. (2015) demonstrate that chronic intermittent ethanol exposure results in increased extracellular levels of glutamate in the NAc in ethanol-dependent mice, but these effects are not a result of locally dysregulated sodium-dependent and independent glutamate transporter function, suggesting that extrinsic corticostriatal glutamatergic pathways may contribute to this hyperglutamatergic state. Finally, McGuier et al. (2015) show that deletion of the excitatory postsynaptic scaffolding protein Homer2 is associated with an increased density of long, thin dendritic spines in NAc core medium spiny neurons, yet unexpectedly these structural modifications are not modified by repeated alcohol exposure. Together, this body of work indicates complex interactions between drugs of abuse, endogenous neuromodulators and their signaling targets, and the mechanisms underlying the functional and structural plasticity in the brain. Research into this complexity is only in its infancy, and needs to be pursued at multiple levels in order to better understand and treat addictive disorders.

Exposure to cocaine regulates inhibitory synaptic transmission from the ventral tegmental area to the nucleus accumbens

Synaptic projections from the ventral tegmental area (VTA) to the nucleus accumbens (NAc) make up the backbone of the brain reward pathway, a neural circuit that mediates behavioural responses elicited by natural rewards as well as by cocaine and other drugs of abuse. In addition to the well-known modulatory dopaminergic projection, the VTA also provides fast excitatory and inhibitory synaptic input to the NAc, directly regulating NAc medium spiny neurons (MSNs). However, the cellular nature of VTA-to-NAc fast synaptic transmission and its roles in drug-induced adaptations are not well understood. Using viral-mediated in vivo expression of channelrhodopsin 2, the present study dissected fast excitatory and inhibitory synaptic transmission from the VTA to NAc MSNs in rats. Our results suggest that, following repeated exposure to cocaine (15 mg kg(-1) day(-1) × 5 days, i.p., 1 or 21 day withdrawal), a presynaptic enhancement of excitatory transmission and suppression of inhibitory transmission occurred at different withdrawal time points at VTA-to-NAc core synapses. In contrast, no postsynaptic alterations were detected at either type of synapse. These results suggest that changes in VTA-to-NAc fast excitatory and inhibitory synaptic transmissions may contribute to cocaine-induced alteration of the brain reward circuitry.

Different Adaptations in AMPA Receptor Transmission in the Nucleus Accumbens after Short vs Long Access Cocaine Self-Administration Regimens

Ca(2+)-permeable AMPA receptors (CP-AMPARs) accumulate in the nucleus accumbens (NAc) after ∼1 month of withdrawal from a long-access cocaine self-administration regimen (6 h/d, 10d). This is functionally significant because CP-AMPARs mediate the 'incubated' cue-induced cocaine craving produced by this regimen. Our present goal was to determine if other commonly employed cocaine self-administration regimens also elicit CP-AMPAR accumulation. We compared four regimens, named according to whether sessions were short-access (ShA, 2 h) or long-access (LgA, 6 h) and the total number of sessions: LgA/10d (already shown to elicit CP-AMPAR accumulation), ShA/11d, ShA/20-24d, and LgA/20-24d. In the latter regimens, rats began with 10 days of ShA and then entered a differential phase (10-14 days) in which ShA sessions either continued or switched to LgA. Controls self-administered saline. After >40 days of withdrawal, whole-cell patch-clamp recordings were performed in NAc core medium spiny neurons to assess the contribution of CP-AMPAR transmission, based on the magnitude of synaptic suppression elicited by bath application of the selective CP-AMPAR antagonist naspm (100 μM). Naspm produced a non-significant (∼10%) attenuation of electrically evoked local excitatory postsynaptic current in the saline and ShA groups. By contrast, a significant naspm-induced synaptic attenuation (25-30%) was observed in both the LgA groups. Further analyses indicate that this emergence of CP-AMPAR transmission in the LgA groups is associated with increased baseline responsiveness of MSN to excitatory drive. Together with data on cocaine infusions in each group, our results show that CP-AMPAR accumulation and enhanced glutamate transmission is associated with longer sessions (6 h), rather than the number of sessions or cocaine infusions.
