Riboflavin improves postharvest cold tolerance in zucchini fruit inducing non-enzymatic antioxidant response and phenolic metabolism

The storage of zucchini fruit at low temperatures during postharvest induces a physiological disorder called chilling injury that drastically reduces fruit quality and shelf life. The phytohormone abscisic acid (ABA) is involved in the acquisition of cold tolerance in zucchini fruit, being the riboflavin pathway one of the most differentially induced with ABA treatment. Thus, the aim of this work was to elucidate the involvement of riboflavin in quality maintenance of zucchini fruit during postharvest cold storage. After testing different concentrations of exogenous riboflavin, 0.5 mM showed the best results. Riboflavin treatment reduced H2O2 content, but the enzymatic antioxidant defense did not change significantly. This response is due to a rise of non-enzymatic antioxidant defense, by accumulating metabolites like ascorbate and carotenoids, as well as inducing phenolic metabolism. The application of this vitamin enhanced the phenolic and flavonoid content in fruit, concomitant with an induction of PAL and C4H activities and an inhibition of PPO activity. This enhancement of the phenylpropanoid pathway resulted in high vanillic acid and quercetin levels at first day of cold storage. The induction of numerous antioxidant compounds and abscisic acid by riboflavin treatment at short-term postharvest period could be responsible for the lack of chilling injuries in zucchini fruit. Therefore, riboflavin could be successfully implemented in the food industry as an alternative to physical or chemical treatments, due to it is an innocuous additive with good water solubility and low cost, as it prolongs the shelf-life of zucchini fruit and increases its nutraceutical properties.

Oxidative Stress Associated With Increased Reactive Nitrogen Species Generation in the Liver and Kidney Caused by a Major Metabolite Accumulating in Tyrosinemia Type 1.

Tyrosinemia type 1 (TT1) is caused by fumarylacetoacetate hydrolase activity deficiency, resulting in tissue accumulation of upstream metabolites, including succinylacetone (SA), the pathognomonic compound of this disease. Since the pathogenesis of liver and kidney damage observed in the TT1-affected patients is practically unknown, this study assessed the effects of SA on important biomarkers of redox homeostasis in the liver and kidney of adolescent rats, as well as in hepatic (HepG2) and renal (HEK-293) cultured cells. SA significantly increased nitrate and nitrite levels and decreased the concentrations of reduced glutathione (GSH) in the liver and kidney, indicating induction of reactive nitrogen species (RNS) generation and disruption of antioxidant defenses. Additionally, SA decreased the GSH levels and the activities of glutathione peroxidase, glutathione S-transferase, glutathione reductase, and superoxide dismutase in hepatic and renal cells. Noteworthy, melatonin prevented the SA-induced increase of nitrate and nitrite levels in the liver. Therefore, SA-induced increase of RNS generation and impairment of enzymatic and nonenzymatic antioxidant defenses may contribute to hepatopathy and renal disease in TT1.

Exogenous silicon alleviates aluminum stress in Eucalyptus species by enhancing the antioxidant capacity and improving plant growth and tolerance quality

BackgroundAs an efficient and high-quality additive in agriculture and forestry production, silicon (Si) plays an important role in alleviating heavy metal stress and improving plant growth. However, the alleviating effect of aluminum (Al) toxicity by Si in Eucalyptus is still incomplete.ResultsHere, a study was conducted using two Al concentrations (0 and 4.5 mM) with four Si concentrations (0, 0.5, 1, and 1.5 mM) to investigate plant growth, tolerance and antioxidant defense system in four Eucalyptus species (Eucalyptus tereticornis, Eucalyptus urophylla, Eucalyptus grandis, and Eucalyptus urophylla × Eucalyptus grandis). The results showed that the stress induced by 4.5 mM Al increased oxidative damage, disturbed the balance of enzymatic and non-enzymatic antioxidant systems, and negatively affected plant growth and tolerance quality in the four Eucalyptus species. However, the addition of 0.5 mM and 1 mM Si alleviated the effects of Al toxicity on plant growth and improved plant growth quality by strengthening stress tolerance. Besides, adding Si significantly facilitated the synergistic action of enzymatic and non-enzymatic antioxidant defenses, increased the removal of reactive oxygen species, reduced lipid peroxidation, and oxidative stress, and promoted the phytoremediation rate of the four Eucalyptus species by 18.7 ~ 34.8% compared to that in the absence of Si.ConclusionsSilicon can alleviate the effect of Al toxicity by enhancing the antioxidant capacity and improving plant growth and tolerance quality. Hence, the application of Si is an effective method for the phytoremediation of Eucalyptus plantations in southern China.

Delineating drought-induced antioxidative traits as potential mechanisms for climate resilience in Argania spinosa

Drought stress ranks among the most critical environmental challenges facing agriculture today, causing significant impairments to plant growth and development. In Morocco, these negative impacts are projected to intensify further under the combined pressures of climate change and the worsening water shortage crisis. The imperative need for sustainable arganiculture (cultivation of Argane trees) in dry lands necessitates the expeditious identification of drought-tolerant elite Argania spinosa trees. In this context, we investigated the drought tolerance of 2-year-old A. spinosa seedlings from two contrasting provenances Lakhssas (LKS) and Aoulouz (ALZ) under severe stress by evaluating their antioxidative defense mechanisms. A total of 24 parameters related to reactive oxygen species (ROS), oxidative damage, and enzymatic and non-enzymatic antioxidant defense were measured and compared between both provenances. The results showed that the drought-stressed conditions significantly increased the activity of enzymatic antioxidants, including superoxide dismutase, catalase, peroxidase, polyphenoloxidase, glutathione peroxidase, glutathione S-transferase, and ascorbate-glutathione cycle enzymes, as well as the content of non-enzymatic antioxidants, including reduced glutathione (GSH), ascorbic acid (AsA), α-Tocopherols (α-toc), polyphenols, anthocyanins, and group thiols. The ability of A. spinosa trees to greatly enhance their antioxidant system to limit cellular damage caused by ROS production might be an important attribute linked to drought tolerance. Regarding the inter-provenance variation under drought stress conditions, LKS provenance exhibited superior antioxidative capacity through enhanced AsA-GSH cycle activity and elevated levels of AsA-GSH, α-toc, and polyphenols. However, ALZ demonstrated elevated anthocyanin levels and reduced peroxidative stress markers (hydrogen peroxide and malonyldialdehyde). Significant and positive correlations were recorded between studied ROS and antioxidants. Investigating the multifaceted antioxidative defense system underpinning drought tolerance in A. spinosa can facilitate the identification of drought-tolerant argane trees for the development of a future breeding program allowing sustainable arganiculture in dry lands.

Hyperhomocysteinemia-Induced Alterations in Protein Expression and Oxidative Stress Parameters in Rat Heart

Hyperhomocysteinemia (HHcy) is considered an independent risk factor of cardiovascular diseases. Among the proposed mechanisms underlying homocysteine toxicity are altered protein expression and induction of oxidative stress. In the present study, we explored protein abundance and parameters related to oxidative stress in heart homogenates of rats exposed to chronic mild HHcy. Using two-dimensional gel electrophoresis followed by MALDI-TOF/TOF mass spectrometry 22 altered proteins (6 upregulated and 14 downregulated) were identified. For eight proteins the altered abundances were validated by Western blot analysis. Identified proteins are primarily involved in energy metabolism (mainly enzymes of glycolysis, pyruvate dehydrogenase complex, citric acid cycle, and ATP synthase), cardiac muscle contraction (α-actin and myosin light chains), stress response (heat-shock protein β1 and αB-crystallin) and antioxidant defense (glutathione peroxidase 1). Diminished antioxidant defense was confirmed by decreases in total antioxidant capacity and GSH/GSSG ratio. Consistent with the decline in enzymatic and non-enzymatic antioxidant defense the protein oxidative modification, as determined by tyrosine nitration, was significantly increased. These findings suggest that both, altered protein expression and elevated oxidative stress contribute to cardiovascular injury caused by HHcy.

Redox Homeostasis, Gut Microbiota, and Epigenetics in Neurodegenerative Diseases: A Systematic Review.

Neurodegenerative diseases encompass a spectrum of disorders marked by the progressive degeneration of the structure and function of the nervous system. These conditions, including Parkinson's disease (PD), Alzheimer's disease (AD), Huntington's disease (HD), Amyotrophic lateral sclerosis (ALS), and Multiple sclerosis (MS), often lead to severe cognitive and motor deficits. A critical component of neurodegenerative disease pathologies is the imbalance between pro-oxidant and antioxidant mechanisms, culminating in oxidative stress. The brain's high oxygen consumption and lipid-rich environment make it particularly vulnerable to oxidative damage. Pro-oxidants such as reactive nitrogen species (RNS) and reactive oxygen species (ROS) are continuously generated during normal metabolism, counteracted by enzymatic and non-enzymatic antioxidant defenses. In neurodegenerative diseases, this balance is disrupted, leading to neuronal damage. This systematic review explores the roles of oxidative stress, gut microbiota, and epigenetic modifications in neurodegenerative diseases, aiming to elucidate the interplay between these factors and identify potential therapeutic strategies. We conducted a comprehensive search of articles published in 2024 across major databases, focusing on studies examining the relationships between redox homeostasis, gut microbiota, and epigenetic changes in neurodegeneration. A total of 161 studies were included, comprising clinical trials, observational studies, and experimental research. Our findings reveal that oxidative stress plays a central role in the pathogenesis of neurodegenerative diseases, with gut microbiota composition and epigenetic modifications significantly influencing redox balance. Specific bacterial taxa and epigenetic markers were identified as potential modulators of oxidative stress, suggesting novel avenues for therapeutic intervention. Moreover, recent evidence from human and animal studies supports the emerging concept of targeting redox homeostasis through microbiota and epigenetic therapies. Future research should focus on validating these targets in clinical settings and exploring the potential for personalized medicine strategies based on individual microbiota and epigenetic profiles.

Field evaluation and characterization of a novel biostimulant for broccoli (Brassica oleracea var. italica) cultivation under drought and salt stress which increases antioxidant, glucosinolate and phytohormone content

Anthropogenic global warming is affecting crop yield and thus compromising food security. Drought and salinization of the irrigation water or soil are increasingly frequent in most arable land, specifically in arid and semiarid areas such as the Mediterranean basin. Many crops have been displaced or their yield has plummeted in recent years, causing food shortages and price increases. In a context of global population growth and reduction in the availability of natural resources used in agriculture, finding novel tools that help farmers to maintain yield in an increasingly arid and saline scenario is of pivotal importance. This is particularly important in organic agriculture, where the number of available tools is very limited. Biostimulants are substances or microorganisms of natural origin whose function is to stimulate plant processes related to nutrient absorption, nutrient use efficiency, tolerance to abiotic stress or the quality of the agricultural products obtained. In the present work, we have evaluated, in field, the agronomic effectiveness of a novel biostimulant formulation in a cruciferous crop of great interest in the Mediterranean basin (broccoli) under control conditions, water and salt stress. Our research has shown that our product had a positive effect on broccoli production and in delaying flowering time. We have also found that the application of our biostimulant increased enzymatic and non-enzymatic antioxidant defense under drought conditions. Similarly, when exposed to salinity, the biostimulant increased the concentration of different phytohormones and glucosinolates. Taken together, our biostimulant increases the tolerance of broccoli to salt stress and water limitation by increasing the antioxidant response, the level of glucosinolates and eliciting the hormonal response.

Evaluation of Enzymatic and Non-enzymatic Antioxidant Defense Responses of Durum Wheat (Triticum durum Desf.) in Coping with Boron Stress

Wheat, one of the world's most important agricultural products, plays a vital role in meeting the nutritional needs of our growing global population. However, arid and semi-arid regions face a potential threat from boron (B) toxicity. While boron is an essential nutrient for plant growth and development, its excessive presence can be toxic. It disrupts physiological processes, causing chlorosis and necrosis, ultimately leading to yield loss or plant death. Although B deficiency is a problem in the soils of many countries, Türkiye is one of the few experiencing B toxicity problems in its agricultural areas. This study investigated the physiological and biochemical responses of durum wheat to various B concentrations (0-20 mg L-1) under controlled air-conditioned cabin conditions. Durum wheat exhibited a decrease in chlorophyll content, phenolic content, and antiradical capacity at B doses exceeding 10 mg L-1. However, carotene content increased steadily with increasing B concentrations. The activities of antioxidant enzymes, including superoxide dismutase (SOD), ascorbate peroxidase (APX), and glutathione S-transferase (GST), increased up to a B dose of 15 mg L-1. Catalase (CAT) and glutathione reductase (GR) activities increased up to 10 mg L-1 B dose but decreased at higher B levels. Proline content increased tenfold up to a B dose of 10 mg L-1, indicating an attempt to mitigate stress. Conversely, malondialdehyde (MDA) accumulation increased continuously (approximately 150%) with increasing B doses, suggesting membrane damage. Despite being considered B-sensitive, this study demonstrated that durum wheat can effectively cope with B stress up to a B dose of 10 mg L-1 under controlled conditions. Beyond this threshold, physiological and biochemical changes indicate a decline in stress tolerance. Many osmoregulators, carotenes, alkaloids, flavonoids, tocopherols, phenolic compounds, non-protein amino acids, and several unidentified metabolites are activated to support antioxidant defense. The SOS pathway and the released ROS force gene regulatory systems come into play. Following these, the ROS released in the organism are neutralized, and ionic homeostasis and cellular stress resistance are achieved.

Micronutrient deficiency-induced oxidative stress in plants.

Micronutrients like iron (Fe), zinc (Zn), copper (Cu), manganese (Mn), boron (B), nickel (Ni), and molybdenum (Mo) perform significant roles in the regulation of plant metabolism, growth, and development. Micronutrients, namely Fe, Zn, Cu, Mn, and Ni, are involved in oxidative stress and antioxidant defense as they are cofactors or activators of various antioxidant enzymes, viz., superoxide dismutase (Fe, Cu/Zn, Mn, and Ni), catalase (Fe), and ascorbate peroxidase (Fe). An effort has been made to incorporate recent advances along with classical work done on the micronutrient deficiency-induced oxidative stress and associated antioxidant responses of plants. Deficiency of a micronutrient produces ROS in the cellular compartments. Enzymatic and non-enzymatic antioxidant defense systems are often modulated by micronutrient deficiency to regulate redox balance and scavenge deleterious ROS for the safety of cellular constituents. ROS can strike cellular constituents such as lipids, proteins, and nucleic acids and can destruct cellular membranes and proteins. ROS might act as a signaling molecule and activate the antioxidant proteins by interacting with signaling partners such as respiratory burst oxidase homolog (RBOH), G-proteins, Ca2+, mitogen activated protein kinases (MAPKs), and various transcription factors (TFs). Opinions on probable ROS signaling under micronutrient deficiency have been described in this review. However, further research is required to decipher micronutrient deficiency-induced ROS generation, perception, and associated downstream signaling events, leading to the development of antioxidant responses in plants.

Maternal exposure to low-dose bisphenol A and its potential neurotoxic impact on male pups: A histological, immunohistochemical, and ultrastructural study

BackgroundBisphenol A (BPA) is a widely used chemical with a harmful effect on animal and human. The neonatal and juvenile period is a highly risky neurodevelopmental period. AimThis study aimed to determine how male albino rat pups’ cerebral cortex was altered by low doses of BPA given to mothers and the role of the oxidative stress. MethodsThirty pregnant rats were randomly split into three equal groups, negative control, and positive control: received 1 cc of corn oil once a day through gastric tube and BPA treated: a dose of 200 µg/kg/day (dissolved in 1 cc corn oil). The male rat pups of each group were sacrificed at 1 week, 3 weeks and 6 weeks. The cerebra were then separated from the brain for histological and biochemical studies. ResultsRats administered BPA had raised levels of lipid peroxidation marker (MDA), lower levels of enzymatic antioxidants (SOD and CAT) with decreased body, cerebral weights, and decreased levels of non-enzymatic antioxidant defense (GSH). Histo-pathologically, shrunken pyramidal cells with congested blood vessels appeared. GFAP displayed increased number of positive immune-reactive astrocytes with high statistically significant increase in the area % in BPA treated group when compared to the control groups, on contrary to MBP. Semi-thin and ultra-thin BPA-sections revealed degenerative changes in myelinated axons with tiny nucleus and broken nuclear membranes. Lysosomes, dilated endoplasmic reticulum cisternae with noticeable increase in unmyelinated nerve fibers were also observed. ConclusionThe structure of the developing cerebral cortex is negatively impacted by BPA due to oxidative stress.

Regulation of nitro-oxidative homeostasis: an effective approach to enhance salinity tolerance in plants.

Soil salinity is a major constraint for sustainable agricultural productivity, which together with the incessant climate change may be transformed into a severe threat to the global food security. It is, therefore, a serious concern that needs to be addressed expeditiously. The overproduction and accumulation of reactive oxygen species (ROS) and reactive nitrogen species (RNS) are the key events occurring during salt stress, consequently employing nitro-oxidative stress and programmed cell death in plants. However, very sporadic studies have been performed concerning different aspects of nitro-oxidative stress in plants under salinity stress. The ability of plants to tolerate salinity is associated with their ability to maintain the cellular redox equilibrium mediated by both non-enzymatic and enzymatic antioxidant defense mechanisms. The present review emphasizes the mechanisms of ROS and RNS generation in plants, providing a detailed evaluation of how redox homeostasis is conserved through their effective removal. The uniqueness of this article stems from its incorporation of expression analyses of candidate genes for different antioxidant enzymes involved in ROS and RNS detoxification across various developmental stages and tissues of rice, utilizing publicly available microarray data. It underscores the utilization of modern biotechnological methods to improve salinity tolerance in crops, employing different antioxidants as markers. The review also explores how various transcription factors contribute to plants' ability to tolerate salinity by either activating or repressing the expression of stress-responsive genes. In summary, the review offers a thorough insight into the nitro-oxidative homeostasis strategy for extenuating salinity stress in plants.

Metabolomic and lipidomic fingerprints in inflammatory skin diseases – Systemic illumination of atopic dermatitis, hidradenitis suppurativa and plaque psoriasis

Auto-inflammatory skin diseases place considerable symptomatic and emotional burden on the affected and put pressure on healthcare expenditures. Although most apparent symptoms manifest on the skin, the systemic inflammation merits a deeper analysis beyond the surface. We set out to identify systemic commonalities, as well as differences in the metabolome and lipidome when comparing between diseases and healthy controls. Lipidomic and metabolomic LC-MS profiling was applied, using plasma samples collected from patients suffering from atopic dermatitis, plaque-type psoriasis or hidradenitis suppurativa or healthy controls. Plasma profiles revealed a notable shift in the non-enzymatic anti-oxidant defense in all three inflammatory disorders, placing cysteine metabolism at the center of potential dysregulation. Lipid network enrichment additionally indicated the disease-specific provision of lipid mediators associated with key roles in inflammation signaling. These findings will help to disentangle the systemic components of autoimmune dermatological diseases, paving the way to individualized therapy and improved prognosis.

Enhancing the Storage Longevity of Apples: The Potential of Bacillus subtilis and Streptomyces endus as Preventative Bioagents against Post-Harvest Gray Mold Disease, Caused by Botrytis cinerea.

Gray mold, caused by Botrytis cinerea Pers. Fr., is one of the most vital plant diseases, causing extensive pre- and post-harvest losses in apple fruits. In the current study, we isolated and identified two potential endophytic bioagents, Bacillus subtilis and Streptomyces endus. Both bioagents exhibited a potent fungistatic effect against B. cinerea under both in vitro and in planta conditions. Moreover, two experiments were carried out; (i) the first experiment was conducted at room temperature after artificial inoculation with B. cinerea to monitor the progression of the infection and the corresponding biochemical responses of the apples. Our in vivo findings showed that the treated B. cinerea-infected apple fruits with the cell-free bacterial filtrate of B. subtilis and S. endus (dipping or wrapping) significantly reduced the rotten area of the treated apple at room temperature. Additionally, B. subtilis and S. endus enhanced the enzymatic (POX and PPO) and non-enzymatic (phenolics and flavonoids) antioxidant defense machinery in treated apples. (ii) The second experiment focused on the preventive effects of both bioagents over a 90-day storage period at 1 °C of healthy apples (no artificial inoculation). The application of both bacterial filtrates prolonged the storage period, reduced the relative weight loss, and maintained high-quality parameters including titratable acidity, firmness, and total soluble solids of apple fruits under cold storage at 1 °C. The Kaplan-Meier analysis of rotten apples over 90 days during cold storage showed that the treated apples lasted longer than the non-treated apples. Moreover, the lifespan of apple fruits dipped in the culture filtrate of B. subtilis, or a fungicide, was increased, with no significant differences, compared with the non-treated apples. The current results showed the possibility of using both bioagents as a safe and eco-friendly alternative to chemical fungicides to control gray mold disease in apples.

Tauroursodeoxycholic Acid (TUDCA) Relieves Streptozotocin (STZ)-Induced Diabetic Rat Model via Modulation of Lipotoxicity, Oxidative Stress, Inflammation, and Apoptosis.

Tauroursodeoxycholic acid (TUDCA) is approved for the treatment of liver diseases. However, the antihyperglycemic effects/mechanisms of TUDCA are still less clear. The present study aimed to evaluate the antidiabetic action of TUDCA in streptozotocin (STZ)-induced type 2 diabetes mellitus (T2DM) in rats. Fifteen adult Wistar albino male rats were randomly divided into three groups (n = five in each): control, diabetic (STZ), and STZ+TUDCA. The results showed that TUDCA treatment significantly reduced blood glucose, HbA1c%, and HOMA-IR as well as elevated the insulin levels in diabetic rats. TUDCA therapy increased the incretin GLP-1 concentrations, decreased serum ceramide synthase (CS), improved the serum lipid profile, and restored the glycogen content in the liver and skeletal muscles. Furthermore, serum inflammatory parameters (such as TNF-α, IL-6, IL-1ß, and PGE-2) were substantially reduced with TUDCA treatment. In the pancreas, STZ+TUDCA-treated rats underwent an obvious enhancement of enzymatic (CAT and SOD) and non-enzymatic (GSH) antioxidant defense systems and a marked decrease in markers of the lipid peroxidation rate (MDA) and nitrosative stress (NO) compared to STZ-alone. At the molecular level, TUDCA decreased the pancreatic mRNA levels of iNOS and apoptotic-related factors (p53 and caspase-3). In conclusion, TUDCA may be useful for diabetes management and could be able to counteract diabetic disorders via anti-hyperlipidemic, antioxidant, anti-inflammatory, and anti-apoptotic actions.

Exploring the dynamics of ISR signaling in maize upon seed priming with plant growth promoting actinobacteria isolated from tea rhizosphere of Darjeeling.

Plant growth-promoting rhizobacteria (PGPR) offer an eco-friendly alternative to agrochemicals for better plant growth and development. Here, we evaluated the plant growth promotion abilities of actinobacteria isolated from the tea (Camellia sinensis) rhizosphere of Darjeeling, India. 16S rRNA gene ribotyping of 28 isolates demonstrated the presence of nine different culturable actinobacterial genera. Assessment of the in vitro PGP traits revealed that Micrococcus sp. AB420 exhibited the highest level of phosphate solubilization (i.e., 445 ± 2.1µg/ml), whereas Kocuria sp. AB429 and Brachybacterium sp. AB440 showed the highest level of siderophore (25.8 ± 0.1%) and IAA production (101.4 ± 0.5µg/ml), respectively. Biopriming of maize seeds with the individual actinobacterial isolate revealed statistically significant growth in the treated plants compared to controls. Among them, treatment with Paenarthrobacter sp. AB416 and Brachybacterium sp. AB439 exhibited the highest shoot and root length. Biopriming has also triggered significant enzymatic and non-enzymatic antioxidative defense reactions in maize seedlings both locally and systematically, providing a critical insight into their possible role in the reduction of reactive oxygen species (ROS) burden. To better understand the role of actinobacterial isolates in the modulation of plant defense, three selected actinobacterial isolates, AB426 (Brevibacterium sp.), AB427 (Streptomyces sp.), and AB440 (Brachybacterium sp.) were employed to evaluate the dynamics of induced systemic resistance (ISR) in maize. The expression profile of five key genes involved in SA and JA pathways revealed that bio-priming with actinobacteria (Brevibacterium sp. AB426 and Brachybacterium sp. AB440) preferably modulates the JA pathway rather than the SA pathway. The infection studies in bio-primed maize plants resulted in a delay in disease progression by the biotrophic pathogen Ustilago maydis in infected maize plants, suggesting the positive efficacy of bio-priming in aiding plants to cope with biotic stress. Conclusively, this study unravels the intrinsic mechanisms of PGPR-mediated ISR dynamics in bio-primed plants, offering a futuristic application of these microorganisms in the agricultural fields as an eco-friendly alternative.

The Metabolism, Detrimental Effects, and Signal Transduction Mechanism of Reactive Oxygen Species in Plants under Abiotic Stress

Plants are a major source of bio based materials, and their productivity is significantly impacted by environmental adversities with the continuous deterioration of the environment. The disruption of reactive oxygen species (ROS) production and scavenging balance is a common consequence of various abiotic stresses in plants, resulting in the excessive accumulation of ROS. However, it is crucial to acknowledging the dual role of ROS, acting both as harmful substances and signaling molecules. ROS can interact with membrane proteins, initiating a chain reaction of membrane lipid peroxidation. This process leads to a decrease in membrane lipid unsaturation and fluidity, as well as an increase in membrane permeability. Inhibited enzyme activity even lead to loss of protein function, caused DNA damage and mutations, blocked DNA replication and transcription, eventually leading to cell death. This can result in significant reductions in crop yield and biomass in bio based agricultural production. ROS also played vital role in signal molecule transduction in the process of plant resistance to stress, and worked in concert with other signal molecules to induce the stress resistance genes’ expression and enhance plant resistance. Hereto, scavenging mechanism for ROS in plants is of the essence to regulate the concentration of ROS, thus determining which role ROS play. The thermal dissipation of excess light energy effectively avoids ROS production, and the enzymatic and non-enzymatic antioxidant defense systems in plants work cooperatively to clear excessive ROS. This review provides a comprehensive summary of the production pathway, detrimental effects, scavenging mechanism and signal transduction of ROS in plants under abiotic stress.

Arbuscular mycorrhizal fungi enhanced resistance to low-temperature weak-light stress in snapdragon (Antirrhinum majus L.) through physiological and transcriptomic responses.

Low temperature (LT) and weak light (WL) seriously affects the yield and quality of snapdragon in winter greenhouse. Arbuscular mycorrhizal fungi (AMF) exert positive role in regulating growth and enhancing abiotic stress tolerance in plants. Nevertheless, the molecular mechanisms by AMF improve the LT combined with WL (LTWL) tolerance in snapdragon remain mostly unknown. We compared the differences in root configuration, osmoregulatory substances, enzymatic and non-enzymatic antioxidant enzyme defense systems and transcriptome between AMF-inoculated and control groups under LT, WL, low light, and LTWL conditions. Our analysis showed that inoculation with AMF effectively alleviated the inhibition caused by LTWL stress on snapdragon root development, and significantly enhanced the contents of soluble sugars, soluble proteins, proline, thereby maintaining the osmotic adjustment of snapdragon. In addition, AMF alleviated reactive oxygen species damage by elevating the contents of AsA, and GSH, and the activities of superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR), monodehydroascorbate reductase (MDHAR), and glutathione reductase (GR). RNA-seq analysis revealed that AMF regulated the expression of genes related to photosynthesis (photosystem I related proteins, photosystem II related proteins, chlorophyll a/b binding protein), active oxygen metabolism (POD, Fe-SOD, and iron/ascorbate family oxidoreductase), plant hormone synthesis (ARF5 and ARF16) and stress-related transcription factors gene (bHLH112, WRKY72, MYB86, WRKY53, WRKY6, and WRKY26) under LTWL stress. We concluded that mycorrhizal snapdragon promotes root development and LTWL tolerance by accumulation of osmoregulatory substances, activation of enzymatic and non-enzymatic antioxidant defense systems, and induction expression of transcription factor genes and auxin synthesis related genes. This study provides a theoretical basis for AMF in promoting the production of greenhouse plants in winter.

Perfluorobutanoic acid triggers metabolic and transcriptional reprogramming in wheat seedlings

The environmental risks of fluorinated alternatives are of great concern with the phasing out of perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate. Here, multi-omics (i.e., metabolomics and transcriptomics) coupled with physiological and biochemical analyses were employed to investigate the stress responses of wheat seedings (Triticum aestivum L.) to perfluorobutanoic acid (PFBA), one of the short-chain per- and polyfluoroalkyl substances (PFAS) and PFOA alternatives, at environmentally relevant concentrations (0.1–100 ng/g). After 28 days of soil exposure, PFBA boosted the generation of OH and O2− in wheat seedlings, resulting in lipid peroxidation, protein perturbation and impaired photosynthesis. Non-enzymatic antioxidant defense systems (e.g., glutathione, phenolics, and vitamin C) and enzymatic antioxidant copper/zinc superoxide dismutase were strikingly activated (p < 0.05). PFBA-triggered oxidative stress induced metabolic and transcriptional reprogramming, including carbon and nitrogen metabolisms, lipid metabolisms, immune responses, signal transduction processes, and antioxidant defense-related pathways. Down-regulation of genes related to plant-pathogen interaction suggested suppression of the immune-response, offering a novel understanding on the production of reactive oxygen species in plants under the exposure to PFAS. The identified MAPK signaling pathway illuminated a novel signal transduction mechanism in plant cells in response to PFAS. These findings provide comprehensive understandings on the phytotoxicity of PFBA to wheat seedlings and new insights into the impacts of PFAS on plants.

Unravelling the combined impacts of drought and Cu in barley plants – double trouble?

The occurrence of drought in soils, particularly in those contaminated by metals, poses a current threat to crops, as these factors can interact and induce unique stress responses. Therefore, this study mainly focused on understanding the crosstalk between drought and copper (Cu) stress in the physiology of the barley (Hordeum vulgare L.) plant. Using a bifactorial experimental design, seedlings were grown in a natural soil under the following treatments: plants continuously irrigated in uncontaminated soil for 14 days (control); plants continuously irrigated in Cu-contaminated soil (115 mg Cu kg−1) for 14 days (Cu); plants only irrigated during the initials 7 days of growth in uncontaminated soil (drought); plants co-exposed to Cu and drought (combined). After 14 days of growth, the results revealed that drought prevented Cu bioaccumulation in barley roots, which were still severely affected by the metal, both individually and in combination with the water deficit. Furthermore, individual and combined exposure to these stressors resulted in impaired photosynthetic performance in barley plants. Despite the increased activation of enzymatic and non-enzymatic antioxidant defence mechanisms, particularly in the green organs, the plants co-exposed to both stress factors still showed higher oxidative damage, severely impacting biomass production.

Ecophysiological response of Populus alba L. to multiple stress factors during the revitalisation of coal fly ash lagoons at different stages of weathering.

The enormous quantities of fly ash (FA) produced by thermal power plants is a global problem and safe, sustainable approaches to reduce the amount and its toxic effects are still being sought. Vegetation cover comprising long-living species can help reduce FA dump-related environmental health issues. However, the synergistic effect of multiple abiotic factors, like drought, low organic matter content, a deficit of essential nutrients, alkaline pH, and phytotoxicity due to high potentially toxic element (PTE) and soluble salt content, limits the number of species that can grow under such stressful conditions. Thus, we hypothesised that Populus alba L., which spontaneously colonised two FA disposal lagoons at the 'Nikola Tesla A' thermal power plant (Obrenovac, Serbia) 3 years (L3) and 11 years (L11) ago, has high restoration potential thanks to its stress tolerance. We analysed the basic physical and chemical properties of FA at different weathering stages, while the ecophysiological response of P. alba to multiple stresses was determined through biological indicators [the bioconcentration factor (BCF) and translocation factor (TF) for PTEs (As, B, Cr, Cu, Mn, Ni, Se, and Zn)] and by measuring the following parameters: photosynthetic efficiency and chlorophyll concentration, non-enzymatic antioxidant defence (carotenoids, anthocyanins, and phenols), oxidative stress (malondialdehyde (MDA) concentrations), and total antioxidant capacity (IC50) to neutralise DPPH free radical activity. Unlike at L3, toxic As, B, and Zn concentrations in leaves induced oxidative stress in P. alba at L11, shown by the higher MDA levels, lower vitality, and reduced synthesis of chlorophyll, carotenoids, and total antioxidant activity, suggesting its stress tolerance decreases with long-term exposure to adverse abiotic factors. Although P. alba is a fast-growing species with good metal accumulation ability and high stress tolerance, it has poor stabilisation potential for substrates with high As and B concentrations, making it highly unsuitable for revitalising such habitats.
