Delaying panicle nitrogen application to emergence of 3rd leaf from flag leaf increases the grain-filling ability and yield of large-panicle rice by increasing stem nonstructural carbohydrates at heading

The reserve of stem nonstructural carbohydrates (NSC) at heading can enhance the sink strength and improve the grain-filling ability of large-panicle rice, which is beneficial to realizing its high-yield potential. Delaying panicle nitrogen (N) application is expected to increase stem NSC accumulation, but the relevant technical parameters and their impact on the yield of large-panicle rice remain unclear. Two-year field experiments were conducted in 2021 and 2022 with two large-panicle rice cultivars (HLY280 and YLY228) and five panicle N treatments: panicle N application at emergence of 4th leaf from flag leaf (TL4), 3rd leaf from flag leaf (TL3), 2nd leaf from flag leaf (TL2), flag leaf (TL1), and no panicle N application (CK) as the control, to study the effect of different panicle N treatments on the accumulation and translocation of stem NSC, as well as their impact on rice yield and its components. The results showed that the panicle N treatments significantly influenced rice yield. Among the panicle N application treatments, TL3 showed the highest yield for both cultivars over two years. Compared with TL4, TL3 significantly increased the percentage of filled grains and grain weight while maintaining a similar number of spikelets per panicle, which increased the yield of HLY280 and YLY228 by 10.1–13.6% and 12.4–12.7%, respectively. Among the panicle N application treatments, TL3 significantly increased the translocation of stem mass (net output of stem mass) and stem NSC (∆NSC) from heading to 15 days after heading (DAH), which led to the highest dry matter accumulation during the initiation of grain filling from heading to 15 DAH. The higher ∆NSC of TL3 was mainly due to the higher stem NSC at heading, which was contributed by the higher stem NSC accumulation from 5 days before heading (DBH) to heading. Additionally, the higher stem NSC accumulation under TL3 from 5 DBH to heading was significantly positively correlated with the higher leaf area duration (LAD) in this period. These results suggest that delaying panicle N application to TL3 can significantly increase the yield of large-panicle rice compared with TL4 by increasing the percentage of filled grains and grain weight, which is the result of increased stem NSC accumulation.

Quantifying the Individual and Combined Effects of Short-Term Heat Stress at Booting and Flowering Stages on Nonstructural Carbohydrates Remobilization in Rice

Rice production is threatened by climate change, particularly heat stress (HS). Nonstructural carbohydrates (NSCs) remobilization is a key physiological mechanism that allows rice plants to cope with HS. To investigate the impact of short-term HS on the remobilization of nonstructural carbohydrates (NSCs) in rice, two cultivars (Huaidao-5 and Wuyunjing-24) were subjected to varying temperature regimes: 32/22/27 °C as the control treatment, alongside 40/30/35 °C and 44/34/39 °C, for durations of 2 and 4 days during the booting, flowering, and combined stages (booting + flowering) within phytotrons across the years 2016 and 2017. The findings revealed that the stem’s NSC concentration increased, while the panicle’s NSCs concentration, the efficiency of NSCs translocation from the stem, and the stem NSC contribution to grain yield exhibited a consistent decline. Additionally, sugar and starch concentrations increased in leaves and stems during late grain filling and maturity stages, while in panicles, the starch concentration decreased and sugar concentration increased. The heat-tolerant cultivar, Wuyunjing-24, exhibited higher panicle NSC accumulation under HS than the heat-sensitive cultivar, Huaidao-5, which had more stem NSC accumulation. The flowering stage was the most vulnerable to HS, followed by the combined and booting stages. Heat degree days (HDDs) were utilized to quantify the effects of HS on NSC accumulation and translocation, revealing that the flowering stage was the most affected. These findings suggest that severe HS makes the stem the primary carbohydrate storage sink, and alleviation under combined HS aids in evaluating NSC accumulation, benefiting breeders in developing heat-tolerant rice varieties.

Modified TAL expression in rice plant regulates yield components and grain quality in a N-rate dependent manner

Context or problemHigher-yield and superior-quality rice are the major goals in rice breeding and nitrogen management. Genetic modification combined with optimal nitrogen (N) fertilization is expected to realize it. Objective or research questionModification in transaldolase (TAL) expression alters both the number and pattern of vascular bundles in rice plants, but its interactions with N rate remains unknown. MethodsWe employed three representative lines with different expression levels of TAL in the same genetic background (Wuyunjing 7, WT), and analyzed their responses in dry matter accumulation (DMA), carbohydrate transport, grain filling, yield, yield components, and grain quality under a wide range of N rate. ResultsIncreased expression of TAL (OE) promoted rice yield by producing more spikelets per panicle and higher 1000-grain weight, while decreased expression of TAL lowered the seed setting rate and grain weight significantly. Under a fixed N rate, when TAL expression was altered, the direct path coefficients of the seed setting rate and 1000-grain weight to the yield were higher than other yield components, especially at a high N rate. Decreased expression of TAL (RNAi) significantly reduced the starch and non-structural carbohydrates (NSC) contents in the stems, but enhanced the soluble sugar retention, whereas increased expression of TAL enhanced the translocation of NSC. The grain weight at maturity and mean grain-filling rate (mg d−1) increased significantly along with the TAL expression, but their differences among RNAi, WT and OE narrowed at higher N rate. Decreased expression of TAL enhanced the apparent amylose content (ACC) and protein (PC), reduced the starch granule size, and caused a loose starch arrangement and elevated chalkiness, as a consequence, significantly reduced the peak viscosity, hot viscosity, final viscosity, and taste value in a N rate dependent manner. The taste values of OE and WT were similar and the highest value was attained under a moderate N rate. ConclusionsTAL expression plays an important role in the regulation of assimilate supply, grain filling, yield formation, and grain quality. Increased expression of TAL alleviates competition effect among the yield components and enhances the yield potential, especially under moderate to high N rate. Higher TAL expression combined with a moderate N rate improves ECQ. Implications or significanceTAL expression and N rate interacts to regulate yield components and quality. This finding provides meaningful knowledge to administering N rate and genetically modifying TAL expression in a rice variety.

Foliar non-structural carbohydrates and resprouting ability of woody species in a tropical dry forest

Diverse disturbances contribute to biomass loss in tropical dry forests (TDF), of which slash-and-burn agriculture (SBA) may be the most severe; it is practiced by large populations of residents. Plant species capable of resprouting in response to such disturbances often possess above-ground structures that survive and mobilize resources for resprouting, in addition to below-ground structures. In Brazilian TDF, woody species regenerating from stem pieces that persist after SBA events are common. In this study, we evaluated the foliar concentrations of non-structural carbohydrates (NSC) and mineral nutrients during the first six months after an SBA event in all species that resprouted within the experimental area. Almost all the species resprouted exclusively from the stems. The measured nutrient concentrations did not change drastically after an SBA event. Two patterns were observed with respect to NSC concentrations among the species. Half of the studied species exhibited high concentrations of NSC before the SBA event, which gradually decreased three and six months after the disturbance. In contrast, the remaining species maintained consistent NSC concentrations throughout the study period. The cost of building new leaf tissue was similar for all species before the SBA event. However, after disturbance, the species invested different amounts of carbon into the new leaves. Those species that decreased NSC leaf concentration after SBA showed the highest regrowth intensity through the number of new tillers. Our results suggest that the intense translocation of NSC from the leaves before SBA may support the increased activity of the buds present in the remaining stem piece.

Close relationship between potassium accumulation in internodes and translocation of non-structural carbohydrate to grains in rice cultivars with different yields

Potassium (K) is an important macronutrient together with nitrogen and phosphorous. Potassium mostly accumulates in the internodes of rice plants during ripening stage. Since internodes are responsible for translocation of carbohydrates to the grains during ripening, the involvement of internode K in grain yield has been suggested, however, the precise mechanism remains unclear. This study aims to examine the relationship between rice grain yield in different cultivars and the dynamics of K and non-structural carbohydrates (NSC) in the internodes during the ripening period. Field experiments were conducted to investigate the differences in the dynamics of K and NSCs in internodes from heading to maturation in 10 cultivars with different yields and three mutants with different grain numbers. To reveal the localization of K in internode tissues, we conducted laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS) analysis. During the early ripening period from heading to 14 days after heading (14 DAH), the decrease in NSC contents in lower internodes was higher in the high-yielding indica cultivars than in japonica cultivars. In the late ripening period from 14 DAH to maturing, re-accumulation of NSC in lower internodes was higher in the japonica than in the indica. In contrast, the increase in K contents in the internodes was larger in the indica than in the japonica throughout the ripening stage. In the internodes of low-yielding whole crop silage (WCS) cultivar, the NSC contents were maintained at high levels during ripening, while K contents were consistently low. Similar results were observed in the mutant with low grain numbers. The mutant had lower K contents and higher NSC contents than the wild type (WT). Correlation analysis of different cultivars indicated that the change in K content in whole internodes from heading to maturing was negatively correlated with the change in NSC content. LA-ICP-MS analysis of internode tissues of the indica cultivar revealed that K accumulated more in the parenchyma cells than in large vascular bundles. These results suggest that K accumulation in the internodes is positively correlated with NSC translocation from internodes to grains, which could be through indirect effect of K accumulation in parenchyma on enhancement of long distance transport of NSC from internodes to the grains. This study provides new insights into the mechanism underlying the role of K in grain filling, which would help improving usage of K fertilizer for optimum plant growth and yields.

Carbohydrate dynamics in Populus trees under drought: An expression atlas of genes related to sensing, translocation, and metabolism across organs.

In trees, nonstructural carbohydrates (NSCs) serve as long-term carbon storage and long-distance carbon transport from source to sink. NSC management in response to drought stress is key to our understanding of drought acclimation. However, the molecular mechanisms underlying these processes remain unclear. By combining a transcriptomic approach with NSC quantification in the leaves, stems, and roots of Populus alba under drought stress, we analyzed genes from 29 gene families related to NSC signaling, translocation, and metabolism. We found starch depletion across organs and accumulation of soluble sugars (SS) in the leaves. Activation of the trehalose-6-phosphate/SNF1-related protein kinase (SnRK1) signaling pathway across organs via the suppression of class I TREHALOSE-PHOSPHATE SYNTHASE (TPS) and the expression of class II TPS genes suggested an active response to drought. The expression of SnRK1α and β subunits, and SUCROSE SYNTHASE6 supported SS accumulation in leaves. The upregulation of active transporters and the downregulation of most passive transporters implied a shift toward active sugar transport and enhanced regulation over partitioning. SS accumulation in vacuoles supports osmoregulation in leaves. The increased expression of sucrose synthesis genes and reduced expression of sucrose degradation genes in the roots did not coincide with sucrose levels, implying local sucrose production for energy. Moreover, the downregulation of invertases in the roots suggests limited sucrose allocation from the aboveground organs. This study provides an expression atlas of NSC-related genes that respond to drought in poplar trees, and can be tested in tree improvement programs for adaptation to drought conditions.

Differences and mechanisms of post-anthesis dry matter accumulation in rice varieties with different yield levels

Post-anthesis dry matter accumulation (PDMA) plays an important role in yield, but the differences and mechanisms of PDMA among rice varieties with different yield levels are still unclear. Field experiments were conducted using six rice varieties with medium, high, and super high yield levels selected to not only analyze the relationship between PDMA and yield but also compare the physiological indexes among different varieties. The effects of grain fertilizer on PDMA and yield were also observed. PDMA and apparent transferred mass of non-structural carbohydrate (NSC) in stems from heading to maturity were significantly positively correlated with yield. The physiological indexes (root oxidation activity, zeatin ​+ ​zeatin riboside contents in roots and leaves, and leaf photosynthetic rate) at 0–20 days after anthesis (DAA) of super high-yielding varieties were significantly higher than those of medium-yielding and high-yielding varieties. In addition, the α-amylase activity in stems at 30–50 DAA of super high-yielding varieties was significantly higher than other varieties. Compared with no grain fertilizer application rate (GFAR), the GFAR for maximum yield and yield increase of super high-yielding varieties were higher than other varieties. The above physiological indexes at 10–20 DAA were increased under GFAR at 54 ​kg ​ha−1. The α-amylase activity at 30–50 DAA was inhibited under GFAR at 81 ​kg ​ha−1 that hindered NSC translocation, resulting in decreased yield. Our results indicated that higher PDMA and NSC translocation were the crucial characteristics that benefited the yield formation of super high-yielding rice varieties.

Phloem Unloading in Developing Rice Caryopses and its Contribution to Non-Structural Carbohydrate Translocation from Stems and Grain Yield Formation.

Phloem unloading plays an important role in photoassimilate partitioning and grain yield improvements in cereal crops. The phloem unloading strategy and its effects on photoassimilate translocation and yield formation remain unclear in rice. In this study, plasmodesmata were observed at the interface between the sieve elements (SEs) and companion cells (CCs), and between the SE-CC complex and surrounding parenchyma cells (PCs) in phloem of the dorsal vascular bundle in developing caryopses. Carboxyfluorescein (CF) signal was detected in the phloem of caryopses, which showed that CF was unloaded into caryopses. These results indicated that the SE-CC complex was symplasmically connected with adjacent PCs by plasmodesmata. Gene expression for sucrose transporter (SUT) and cell wall invertase (CWI), and OsSUT1 and OsCIN1 proteins were detected in developing caryopses, indicating that rice plants might actively unload sucrose into caryopses by the apoplasmic pathway. Among three rice recombinant inbred lines, R201 exhibited lower plasmodesmal densities at the boundaries between cell types (SE-CC, SE-PC and CC-PC) in developing caryopses than R91 and R156. R201 also had lower expression of SUT and CWI genes and lower protein levels of OsSUT1 and OsCIN1, as well as CWI activity, than R91 and R156. These data agreed with stem non-structural carbohydrate (NSC) translocation and grain yields for the three lines. The nitrogen application rate had no significant effect on plasmodesmal densities at the interfaces between different cells types, and did not affect CF unloading in the phloem of developing caryopses. Low nitrogen treatment enhanced expression levels of OsSUT and OsCIN genes in the three lines. These results suggested that nitrogen application had no substantial effect on symplasmic unloading but affected apoplasmic unloading. Therefore, we concluded that poor symplasmic and apoplasmic unloading in developing caryopses might result in low stem NSC translocation and poor grain yield formation of R201.

Vascular Bundle Characteristics of Different Rice Variety Treated with Nitrogen Fertilizers and Its Relation to Stem Assimilates Allocation and Grain Yield

The vascular bundle plays a vital role in photoassimilates transportation in rice. However, the vascular bundle characteristics of basal internode and its relationship with stem assimilates allocation and yield remain unclear. In this study, experiments with four different types of rice varieties subjected to three nitrogen application levels were conducted. The results showed that large vascular bundles (LVB) and small vascular bundles (SVB)-related traits of basal internode exhibited genotypic differences. Indica-japonica hybrid varieties had the highest number of cross sectional areas and phloem areas of LVB and SVB, thus the highest stem nonstructural carbohydrates (NSC) translocation, grain filling percentage and grain yield, followed by indica hybrid varieties and indica conventional varieties, and those were the lowest in japonica conventional varieties. The LVB and SVB related traits were significantly and positively correlated with stem NSC translocation, grain filling percentage and grain yield, respectively These results suggested that improving the characteristics of basal internodes was beneficial to enhance stem NSC translocation and consequently increase grain yield. Nitrogen application increased LVB and SVB related traits. Therefore, varieties with developed vascular bundles of basal internodes and cultivation techniques for improving vascular bundle related traits should be considered as the effective route for increasing grain yield.

Stem small vascular bundles have greater accumulation and translocation of non-structural carbohydrates than large vascular bundles in rice.

Phloem unloading and loading are associated with stem non-structural carbohydrates (NSCs) accumulation and remobilization in rice (Oryza sativa L.). Four rice recombinant inbred lines (R032, R191, R046, and R146) derived from a cross between Zhenshan 97 and Minghui 63 were used to investigate the contributions of stem large and small vascular bundles (SVBs) to NSCs accumulation and translocation. Before heading, the parenchyma cells in stem cortex tissues (PCs) surrounding SVBs had higher starch density than those surrounding large vascular bundles (LVBs). Moreover, the protein levels of sucrose transporters (SUTs), cell wall invertase, sucrose synthase, and adenosine diphosphate glucose pyrophosphorylase, as well as the phloem plasmodesma densities were higher in SVBs than those in LVBs. After heading, starch density decreased more in PCs surrounding SVBs than in LVBs. Also, the protein levels of SUTs, α-amylase, sucrose phosphate synthase and sucrose synthase, the phloem plasmodesma densities in SVBs were higher than those in LVBs. The correlations of the number and total cross-sectional area of SVBs with mass and contribution to yield of transferred NSCs were higher than those of LVBs. Our results suggest that SVBs may have higher contributions to pre-anthesis stem NSCs accumulation and post-anthesis translocation than LVBs, which is potentially attributed to the high level of protein and enzyme involved in stem unloading and loading via apoplastic and symplastic pathways.

Genetic progress in grain yield and the associated physiological traits of popular wheat in southwestern China from 1969 to 2012

AbstractKnowledge about the key traits related to genetic gains in grain yield (GY) is necessary for understanding the main yield‐limiting factors and guiding future wheat (Triticum aestivum L.) breeding. The objective of this research was to investigate the genetic progress in GY and the associated physiological traits in popular wheat cultivars released from 1969 to 2012 in southwestern China. Field experiments included eight representative cultivars that were evaluated at three experimental sites for three growing seasons. Grain yield increased at a linear rate of 0.69% or 52.52 kg ha−1 yr−1, which was mainly attributed to the increases in dry matter (DM) accumulation at maturity, harvest index (HI), and thousand‐kernel weight (TKW). The soil and plant analyzer development (SPAD) values of flag and penultimate leaves at anthesis increased with the year of release and were significantly related to GY. Significant genetic gains were made in both nonstructural carbohydrate (NSC) translocation and NSC translocation efficiency from anthesis to maturity, and there was a trend over time towards a high net photosynthetic rate (Pn) of flag leaf and canopy apparent photosynthesis (CAP) at anthesis and at 30 d post‐anthesis (DPA). In addition, the linear increases in N uptake efficiency and N utilization efficiency were both significant and positive related to GY. Therefore, these key physiological traits can be used as selection criteria for further improving yield potential. Our results also reveal the potential for using synthetic hexaploid wheat in high‐yielding wheat breeding.

Effects of AG1 and AG2 QTLs on Nonstructural Carbohydrate and Seed Management Options for Rice Seedling Growth and Establishment under Flooding Stress

Rice (Oryza sativa) plants acquired excess photosynthates in the form of nonstructural carbohydrates (NSCs) in their stems and grain. Despite keen interest in rice NSC, the dynamics of NSC accumulation, translocation and re-accumulation have not yet been well investigated. AG1 and AG2 QTLs associated with flooding tolerance through catalyzing starch into soluble sugar in germinating seeds. Here we conducted three experiments, greenhouse and field to lay the groundwork for large-scale diversity studies on grain NSC and some agronomic traits under direct-seeded rice (DSR) system, using elite lines incorporating AG1, AG2 and AG1-AG2 QTLs into the popular varieties PSB Rc82 and Ciherang-Sub1 along with the donors Kho Hlan On (AG1) and Ma-Zhan Red (AG2). In germinating seedlings, soluble sugars increased, while starch concentration decreased gradually especially in the tolerant checks and AG1-AG2 introgression lines under flooded soil. Soluble sugar accumulation in stem started to increase from the vegetative stage and peaked at the panicle initiation stage then gradually decreased towards the maturity stage. But Sub1-AG lines had higher sugar and starch concentrations at different growth stages than other genotypes in wet season 2016 and dry season 2017. Plant survival rate was positively correlated with the stem NSC at the early vegetative stage (21 days after sowing), and stem NSC was positively associated with plant height at different growth stages. Among the tested seeding rate, the most suitable seeding rate, 4g/m2 with shallow burial depth (0.5cm), resulted in better seedling establishment, relatively higher seedling vigor index and higher leaf area index under flooding in DSR system. Introgression of AG1-AG2 QTLs had no any negative impact on nonstructural carbohydrate, germination rate, and growth and biomass production.

Effects of Elevated CO2 Concentration and Nitrogen Application Levels on the Accumulation and Translocation of Non-Structural Carbohydrates in Japonica Rice

Non-structural carbohydrates (NSC) play an important role in yield formation. In this paper, the relationships of NSC accumulation and translocation with yield formation were investigated under elevated CO2 concentrations ([CO2]) and nitrogen (N) application rates. A japonica rice (Oryza sativa L.) cultivar, “Nanjing 9108,” was grown at three [CO2]—Ambient (T0), ambient + 160 μmol·mol−1 (T1), and ambient + 200 μmol·mol−1 (T2)—in open-top chambers (OTC), with three levels of N fertilizer application rates—10 gN·m−2 (N1), 20 gN·m−2 (N2), and 30 gN·m−2 (N3)—Which were set in OTCs using pot experiments. The results showed that the concentration of NSC (CNSC) and the total mass of NSC stored in stems (TMNSC) under T1 and T2 were higher than those in the T0 treatment, and the CNSC and TMNSC of N3 were lower than those of N1 and N2 at the heading stage. The CNSC and the TMNSC were significantly positively correlated with the stem biomass during the growth period and were notably negatively correlated with the N content in leaves (Nleaf) at the heading and filling stages. The seed setting rate was significantly positively related to the apparent transferred mass of NSC from stems to grains (ATMNSC) at the filling stage, while the relationship between yield and the ATMNSC was not statistically significant. Although there was no difference in the apparent contribution of transferred NSC to grain yield (ACNSC) between treatments, NSC stored in stems further accumulated obviously during the late filling stage, implying that the grain yield of “Nanjing 9108” was predominantly sink-limited. We concluded that elevated [CO2] improved the concentration of NSC at the rice heading stage. The interaction between elevated [CO2] and N fertilizer rates significantly influenced the concentration of NSC at the filling stage. Rice stems NSC reaccumulated at the late grain filling stage, which implies a restriction on NSC transference to grain.

Senescence‐related translocation of nonstructural carbohydrate in rice leaf sheaths under different nitrogen supply

AbstractThe translocation of nonstructural carbohydrates (NSC) from leaf sheaths to filling grains after anthesis contributed greatly to the grain yield of cereal crops. In this study, the effect of nitrogen (N) supply levels on the accumulation and translocation of NSC in leaf sheath tissues and its relationship with the initiation and progression of leaf senescence during grain filling was investigated using two rice (Oryza sativa L.) genotypes, namely, premature flag leaf senescence mutant (psf) and its wild‐type. Three N treatment levels were used to examine N‐supply induced alteration in the activities of several key enzymes involved in NSC translocation and N assimilation in different leaf sheaths. The results show that the NSC translocation rate in leaf sheaths under low nitrogen (LN) treatment was significantly higher than those under normal nitrogen (NN) and high nitrogen (HN) treatments. However, the positive effect of LN on the NSC translocation in leaf sheath was closely associated with its negative effect on grain yield, due to accelerated leaf senescence and shortened leaf longevity. Comparatively, the upper‐positional sheath had a lower NSC amount and higher NSC translocation rate than the lower‐leaf sheaths after heading. High N suppressed sucrose‐phosphate synthase (SPS) activity in leaf sheaths, but enhanced the activity of key enzymes involving in N assimilation in leaf sheaths. The upper sheath had higher activity of sucrose‐metabolizing enzymes and lower activity of N‐assimilating enzymes. Hence, the upper‐leaf sheath had a relatively weak N assimilation and stronger NSC translocation than the lower‐leaf sheaths.

Non-structural carbohydrates in maize with different nitrogen tolerance are affected by nitrogen addition.

Non-structural carbohydrates (NSCs) are an important energy source for plant growth and metabolism. Analysis of NSC changes can provide important clues to reveal the adaptation mechanisms of plants to a specific environment. Although considerable differences have been reported in NSCs in response to nitrogen (N) application among crop species and cultivars, previous studies have mostly focused on the differences in leaves and stems. However, the effects of N on the characteristics of accumulation and translocation of NSC in maize with different levels of N tolerance remain unclear. To determine differences in the N levels, two cultivars (N-efficient ZH311 and N-inefficient XY508) were grown in field pots (Experiment I) and as hydroponic cultures (Experiment II) and were supplemented with different concentrations of N fertilizer. In both experiments, low-N stress significantly increased the accumulation of NSCs in maize vegetative organs and increased the translocation rate of NSCs in the stems and their apparent contribution to yield, thereby reducing the yield loss caused by low-N stress. N application had a greater effect on starch content in the vegetative organs of ZH311, but had less effect on soluble sugar (SS) and NSC content in the whole plant and starch content in the ears. ZH311 could convert more starch into SS under low N conditions to adapt to low N environments than XY508, while ensuring that grain yield and starch quantity were not affected. This is evidently an important physiological mechanism involved in this cultivar’s tolerance to low N conditions.

Quantifying the effects of short-term heat stress at booting stage on nonstructural carbohydrates remobilization in rice

Extreme heat stress events are becoming more frequent under anticipated climate change, which can have devastating impacts on rice growth and yield. To quantify the effects of short-term heat stress at booting stage on nonstructural carbohydrates (NSC) remobilization in rice, two varieties (Nanjing 41 and Wuyunjing 24) were subjected to 32/22/27 °C (maximum/minimum/mean), 36/26/31 °C, 40/30/35 °C, and 44/34/39 °C for 2, 4 and 6 days in phytotrons at booting stage during 2014 and 2015. Yield and yield components, dry matter partitioning index (DMPI), NSC accumulation and translocation were measured and calculated. The results showed that the increase of high-temperature level and duration significantly reduced grain yield by suppressing spikelet number per panicle, seed-setting rate, and grain weight. Heat stress at booting decreased DMPI in panicles, increased DMPI in stems, but had no significant effect on photosynthetic rate. Stem NSC concentration increased whereas panicles NSC concentration, stem NSC translocation efficiency, and contribution of stem NSC to grain yield decreased. Severe heat stress even transformed the stem into a carbohydrate sink during grain filling. The heat-tolerant Wuyunjing 24 showed a higher NSC transport capacity under heat stress than the heat-sensitive Nanjing 41. Heat degree-days (HDD), which combines the effects of the intensity and duration of heat stress, used for quantifying the impacts of heat stress indicates the threshold HDD for the termination of NSC translocation is 9.82 °C day. Grain yield was negatively correlated with stem NSC concentration and accumulation at maturity, and yield reduction was tightly related to NSC translocation reduction. The results suggest that heat stress at booting inhibits NSC translocation due to sink size reduction. Therefore, genotypes with higher NSC transport capacity under heat stress could be beneficial for rice yield formation.

Potassium deficiency aggravates yield loss in rice by restricting the translocation of non-structural carbohydrates under Sarocladium oryzae infection condition.

Sheath rot disease (ShR) caused by Sarocladium oryzae (S. oryzae) infection is an emerging disease that causes severe yield loss by restricting the translocation of non-structural carbohydrates (NSC). Potassium (K) nutrition plays a critical role in disease resistance and the exportation of NSC. However, the physiological mechanisms of K with respect to ShR have not been thoroughly elucidated to date. The objectives of this study were to reveal the mechanisms by which K increases ShR resistance by regulating NSC translocation of rice, therefore, a field experiment combined with an inoculation experiment was conducted. We demonstrate that ShR disease incidence and disease index decreased dramatically with an increasing K application. K deficiency sharply induced the accumulation of NSC in the flag leaf (FL) and flag leaf sheath (FLS) under S. oryzae infection condition, which reduced the contribution of transferred NSC to final yield. A permutational multivariate analysis showed that K deficiency had a greater (49.0%, P <0.001) effect on the NSC content variation in FL than that of S. oryzae infection (15.0%, P <0.001). S. oryzae infection dramatically increased the difference in apparent transferred mass of NSC and cell membrane injury of diseased organs between K-deficient and K-sufficient rice. Finally, we demonstrate that cell membrane injury was a limiting factor imposed by K deficiency, which restricts the export of NSC from source organs. This work highlights the importance of K in improving ShR resistance by regulating NSC translocation (particularly the stem NSC).

Low Nitrogen Application Enhances Starch-Metabolizing Enzyme Activity and Improves Accumulation and Translocation of Non-structural Carbohydrates in Rice Stems.

More than 4 billion inhabitants in Asia depend on rice for 35–60% of the calories consumed in their diets, but new rice cultivars frequently do not reach expected yields because of poor rice grain filling. Here, we quantified the activities of enzymes involved in starch metabolization in rice to investigate the mechanisms regulating the accumulation and translocation of stem non-structural carbohydrates (NSC) under different levels of nitrogen fertilizer application. A pot experiment was conducted using two rice cultivars, Liangyoupeijiu (LYPJ) and Shanyou63 (SY63), under high and low nitrogen applications. Compared with high nitrogen application (HN), low nitrogen application (LN) increased stem NSC concentration before the heading stage and NSC translocation during the grain filling stage; concomitantly, LN significantly shortened the active grain filling period and increased the grain filling rate in superior spikelets. Compared with the LYPJ cultivar, SY63 exhibited a higher grain weight, higher grain filling percentage, and higher stem NSC concentration before heading and greater NSC translocation after heading. During the period between panicle initiation and heading, the activities of adenosine diphosphate-glucose pyrophosphorylase (AGP), starch synthase (StS), and starch branching enzyme (SBE), all enzymes involved in starch synthesis, increased under the LN treatment and positively correlated with increases in stem NSC. During grain filling, the activities of enzymes involved in starch-to-sucrose conversion [α-amylase, β-amylase, and sucrose phosphate synthase (SPS)] increased under the LN treatment and positively correlated with stem NSC remobilization. Overall, the investigated enzymes exhibited higher activities in SY63 than in LYPJ. Our results suggest that low nitrogen increases the activities of AGP, StS, SBE, α-amylase, β-amylase, and SPS, leading to increased accumulation and remobilization of stem starch and NSC in SY63. We conclude that calculated reductions in nitrogen application and the choice of an appropriate cultivar may improve rice grain yields via enhanced stem NSC accumulation and translocation, thereby reducing the costs and increasing the sustainability of rice production.

Effects of nitrogen and phosphorus on the regulation of nonstructural carbohydrate accumulation, translocation and the yield formation of oilseed flax

Nutrients can significantly affect nonstructural carbohydrates (NSC) of crops. This field experiment was conducted to reveal how nitrogen (N) and phosphorus (P) regulated NSC accumulation, translocation as well as the seed yield formation of oilseed flax. The treatments were as follows: (1) application of 0 kg N ha−1 and 0 kg P ha−1 (N(0)P(0)), (2) application of 75 kg N ha−1 and 0 kg P ha−1 (N(75)P(0)), (3) application of 150 kg N ha−1 and 0 kg P ha−1 (N(150)P(0)), (4) application of 0 kg N ha−1 and 75 kg P ha−1 (N(0)P(75)), (5) application of 75 kg N ha−1 and 75 kg P ha−1 (N(75)P(75)), and (6) application of 150 kg N ha−1 and 75 kg P ha−1 (N(150)P(75)). Results showed that application of N decreased the concentration and content of leaves soluble sugar (SS), and increased stems SS and starch (ST) concentration and content at pre-anthesis of oilseed flax, compared to no application of N. Application of P increased the SS concentration in leaves, stems, and seeds at all growth stages, and decreased ST concentration in leaves, seeds and stems at post-anthesis, compared to no P application. Mixed application of N and P significantly improved leaves and stems SS concentration at post-anthesis, promoted more leaves and stems NSC translocation to seeds, thus seed numbers (SN) of per capsule and effective capsules (EC) significantly increased and no-effective capsules (NEC) obviously decreased, compared to only application of N or P. Meanwhile, the average seed yields were improved 14.3%–42.6% than that of only application of N or P. We also found that the SN of per capsule had significantly positive correlation with the leaves SS concentration at post-anthesis, and it had highly positive correlation with the buds and flowers SS concentration at budding and anthesis stages, respectively. The NEC had largely positive correlation with the stems ST concentration at post-anthesis. Our results suggested that N with P mixed application is an effective way for oilseed flax production in semiarid areas by increasing the SN, EC and decreasing the NEC by regulating the leaves and stems NSC at budding, anthesis and post-anthesis, and improving oilseed flax production.

Characterization of high-yielding rice cultivars with different grain-filling properties to clarify limiting factors for improving grain yield

Recently developed high-yielding rice varieties with extra-large sink capacity often have unstable grain filling. Therefore, understanding the factors that limit grain filling is essential for further improvement of rice grain yield. Because grain-filling is determined by the complex sink–source balance, grain-filling ability is very difficult to evaluate. In this study, we compared grain-filling-related traits of three high-yielding cultivars with high sink capacity. We found that the translocation of non-structural carbohydrates (NSC) from stem to panicle during early ripening and grain filling was significantly lower in Momiroman than in Hokuriku 193 and Teqing, whereas dry matter accumulation of the whole plant did not differ among the cultivars throughout ripening. The NSC-components, sucrose and starch were both remained higher in stems of Momiroman than other cultivars. ADP-glucose pyrophosphorylase (AGPase; EC 2.7.7.27) activity was not enhanced and α-Amylase (EC 3.2.1.1) and β-Amylase (EC 3.2.1.2) activities were not impeded in the stems. These data suggested lower sucrose translocation is responsible for lower NSC translocation in Momiroman, rather than too high starch synthesis and too low starch degradation activities. At early ripening, grain growth of the superior spikelets was slow in Momiroman even if carbon supply was increased by spikelet-thinning treatments. These results raise the possibility that low sink strength determines low grain filling in Momiroman, which delays grain growth and decreases sucking force of NSC from the stem.
