The Application Timing of a Cytokinin B-Mo-based Product Affects the Characteristics of Rooted Cuttings and Nonstructural Carbohydrates of Coleus (Plectranthus scutellarioides cv. Wild Lime) during Adventitious Root Development

A large group of horticulture species are propagated vegetatively through shoot-tip cuttings harvested from stock plants and planted to form adventitious roots. Adventitious rooting leads to establishing a carbohydrate sink in the region of root regeneration that is highly dependent on energy and carbon skeletons. We hypothesized that the timing of exogenous applications of cytokinin (CK) and boron (B)–molybdenum (Mo)-based products during adventitious root development can affect the flow of sugars from leaves to sinks, carbon allocation to the adventitious roots, and the quality of rooted cuttings. During this project, we aimed to determine if the application time of a CK/B-Mo-based product during the adventitious root development of unrooted cuttings would impact the source-to-sink relationship and, hence, affect plug growth and quality. A sink-strengthening commercial product based on cytokinin, B, and Mo was applied at four plug development stages plus a negative control as follows: T1, plants without product (control); T2, sticking stage (starting 24 hours after the sticking); T3, callus formation stage; T4, root development stage; and T5, toning stage. The root and shoot lengths and dry matter, number of leaves, leaf chlorophyll content, root-to-shoot ratio (based on dry matter), and nonstructural carbohydrate contents were measured. The timing of the application of the product impacted the root development, quality of the cuttings, and nonstructural carbohydrate content. Product application during the adventitious root dedifferentiation and induction phases (T2) resulted in the shortest root and shoot lengths, lowest dry matter accumulation, lowest nonstructural carbohydrate contents, and some phytotoxicity. Application during the initiation phase (T3) resulted in greater root length, total dry matter, and total soluble sugar contents compared with the control. Application during the expression phase (T4) resulted in the largest root length and mass and the highest sucrose contents. Applying the product when the roots had grown and reached all the edges of the growing media (T5) did not have any benefits compared with the control. This study provides new insights into the application timing of exogenous CKs, B, and Mo to generate a well-toned rooted coleus cutting and potential explanations in relation to nonstructural carbohydrate metabolism.

Characteristics of Isolated Lactic Acid Bacteria and Their Application in High-Moisture Broccoli Waste Silage

Four lactic acid bacteria (LAB) strains isolated from naturally ensiled broccoli waste were characterized, and their effects on the fermentation quality of high-moisture broccoli waste silage were studied. The four isolated strains were assessed using the morphological, physiological and biochemical tests. The four strains were added to broccoli waste at three moisture contents (90%, W0; 87%, W1; 80%, W2) and ensiled for 60 days. All strains (CB89, CB94, CB112, and CB120) grew at 15–45 °C, pH 3.0–7.0, and in 3.0–6.5% NaCl and were identified as Lactiplantibacillus plantarum by 16S rDNA sequencing. Inoculation of CB120 significantly (p < 0.05) increased lactic acid, starch, and non-structural carbohydrate content, and significantly (p < 0.05) decreased pH values and aerobic bacteria count compared with control (CK) at all three moisture contents. In conclusion, CB120 improved the fermentation quality and nutritional value of broccoli waste silage at three moisture contents and could be applied as a promising additive for high-moisture material.

Windthrow causes declines in carbohydrate and phenolic concentrations and increased monoterpene emission in Norway spruce.

With the increasing frequencies of extreme weather events caused by climate change, the risk of forest damage from insect attacks grows. Storms and droughts can damage and weaken trees, reduce tree vigour and defence capacity and thus provide host trees that can be successfully attacked by damaging insects, as often observed in Norway spruce stands attacked by the Eurasian spruce bark beetle Ips typographus. Following storms, partially uprooted trees with grounded crowns suffer reduced water uptake and carbon assimilation, which may lower their vigour and decrease their ability to defend against insect attack. We conducted in situ measurements on windthrown and standing control trees to determine the concentrations of non-structural carbohydrates (NSCs), of phenolic defences and volatile monoterpene emissions. These are the main storage and defence compounds responsible for beetle´s pioneer success and host tree selection. Our results show that while sugar and phenolic concentrations of standing trees remained rather constant over a 4-month period, windthrown trees experienced a decrease of 78% and 37% of sugar and phenolic concentrations, respectively. This strong decline was especially pronounced for fructose (-83%) and glucose (-85%) and for taxifolin (-50.1%). Windthrown trees emitted 25 times greater monoterpene concentrations than standing trees, in particular alpha-pinene (23 times greater), beta-pinene (27 times greater) and 3-carene (90 times greater). We conclude that windthrown trees exhibited reduced resources of anti-herbivore and anti-pathogen defence compounds needed for the response to herbivore attack. The enhanced emission rates of volatile terpenes from windthrown trees may provide olfactory cues during bark beetle early swarming related to altered tree defences. Our results contribute to the knowledge of fallen trees vigour and their defence capacity during the first months after the wind-throw disturbance. Yet, the influence of different emission rates and profiles on bark beetle behaviour and host selection requires further investigation.

Symplasmic phloem loading and subcellular transport in storage roots are key factors for carbon allocation in cassava.

Cassava (Manihot esculenta) is a deciduous woody perennial shrub that stores large amounts of carbon and water in its storage roots. Previous studies have shown that assimilate unloading into storage roots happens symplasmically once secondary anatomy is established. However, mechanisms controlling phloem loading and overall carbon partitioning to different cassava tissues remain unclear. Here, we used a combination of histological, transcriptional, and biochemical analyses on different cassava tissues and at different timepoints to better understand source-sink carbon allocation. We found that cassava likely utilizes a predominantly passive symplasmic phloem loading strategy, indicated by the lack of expression of genes coding for key players of sucrose transport, the existence of branched plasmodesmata in the companion cell/bundle sheath interface of minor leaf veins, and very high leaf sucrose concentrations. Furthermore, we showed that tissue-specific changes in anatomy and non-structural carbohydrate (NSC) contents are associated with tissue-specific modification in gene expression for sucrose cleavage/synthesis, as well as subcellular compartmentalization of sugars. Overall, our data suggest that carbon allocation during storage root filling is mostly facilitated symplasmically and is likely mostly regulated by local tissue demand and subcellular compartmentalization.

In vivo X-ray microtomography locally affects stem radial growth with no immediate physiological impact.

Micro-computed tomography (µCT) is a non-destructive X-ray imaging method used in plant physiology to visualize in-situ plant tissues that enables assessments of embolized xylem vessels. Whereas evidence for X-ray-induced cellular damage has been reported, the impact on plant physiological processes such as carbon (C) uptake, transport, and use are unknown. Yet, these damages could be particularly relevant for studies that track embolism and C fluxes over time. We examined the physiological consequences of µCT-scanning for xylem embolism over three months by monitoring net photosynthesis (Anet), diameter growth, chlorophyll concentration (Chl), and foliar non-structural carbohydrate (NSC) content in four deciduous tree species: hedge maple (Acer campestre), ash (Fraxinus excelsior), European hornbeam (Carpinus betulus), and sessile oak (Quercus petraea). C transport from the canopy to the roots was also assessed through 13C labelling. Our results show that monthly X-ray application did not impact foliar Anet, Chl, NSC content, and C transport. Although X-ray effects did not vary between species, the most pronounced impact was observed in sessile oak, marked by stopped growth and stem deformations around the irradiated area. The absence of adverse impacts on plant physiology for all the tested treatments indicates that laboratory-based µCT systems can be used with different beam energy levels and doses without threatening the integrity of plant physiology within the range of tested parameters. However, the impacts of repetitive µCT on the stem radial growth at the irradiated zone leading to deformations in sessile oak might have lasting implications for studies tracking plant embolism in the longer-term.

Metabolomics analysis of different diameter classes of Taxus chinensis reveals that the resource allocation is related to carbon and nitrogen metabolism

Taxus chinensis (Taxus cuspidata Sieb. et Zucc.) is a traditional medicinal plant known for its anticancer substance paclitaxel, and its growth age is also an important factor affecting its medicinal value. However, how age affects the physiological and metabolic characteristics and active substances of T. chinensis is still unclear. In this study, carbon and nitrogen accumulation, contents of active substances and changes in primary metabolites in barks and annual leaves of T. chinensis of different diameter classes were investigated by using diameter classes instead of age. The results showed that leaves and barks of small diameter class (D1) had higher content of non-structural carbohydrates and C, which were effective in enhancing defense capacity, while N content was higher in medium (D2) and large diameter classes (D3). Active substances such as paclitaxel, baccatin III and cephalomannine also accumulated significantly in barks of large diameter classes. Moreover, 21 and 25 differential metabolites were identified in leaves and barks of different diameter classes, respectively. The differential metabolites were enhanced the TCA cycle and amino acid biosynthesis, accumulate metabolites such as organic acids, and promote the synthesis and accumulation of active substances such as paclitaxel in the medium and large diameter classes. These results revealed the carbon and nitrogen allocation mechanism of different diameter classes of T. chinensis, and its relationship with medicinal components, providing a guidance for the harvesting and utilization of wild T. chinensis.

Non-structural carbohydrate concentrations in tree organs vary across biomes and leaf habits, but are independent of the fast-slow plant economic spectrum.

Carbohydrate reserves play a vital role in plant survival during periods of negative carbon balance. Under a carbon-limited scenario, we expect a trade-offs between carbon allocation to growth, reserves, and defense. A resulting hypothesis is that carbon allocation to reserves exhibits a coordinated variation with functional traits associated with the 'fast-slow' plant economics spectrum. We tested the relationship between non-structural carbohydrates (NSC) of tree organs and functional traits using 61 angiosperm tree species from temperate and tropical forests with phylogenetic hierarchical Bayesian models. Our results provide evidence that NSC concentrations in stems and branches are decoupled from plant functional traits. while those in roots are weakly coupled with plant functional traits. In contrast, we found that variation between NSC concentrations in leaves and the fast-slow trait spectrum was coordinated, as species with higher leaf NSC had trait values associated with resource conservative species, such as lower SLA, leaf N, and leaf P. We also detected a small effect of leaf habit on the variation of NSC concentrations in branches and roots. Efforts to predict the response of ecosystems to global change will need to integrate a suite of plant traits, such as NSC concentrations in woody organs, that are independent of the 'fast-slow' plant economics spectrum and that capture how species respond to a broad range of global change drivers.

The response of physiological and xylem anatomical traits under cadmium stress in Pinus thunbergii seedlings.

Studying the response of physiological and xylem anatomical traits under cadmium (Cd) stress is helpful to understand plants' response to heavy metal stress. Here, seedlings of Pinus thunbergii were treated with 50, 100 and 150mg/kg Cd2+ for 28days. Cd and nonstructural carbohydrate (NSC) content of leaves, stems and roots, root Cd2+ flux, Cd distribution pattern in stem xylem and phloem, stem xylem hydraulic traits, cell wall component fractions of stems and roots, phytohormonal content such as abscisic acid (ABA), gibberellic acid 3 (GA3), molecule -indole-3-acetic acid (IAA) and jasmonic acid (JA) from both leaves and roots, as well as xylem anatomical traits from both stems and roots were measured. Root Cd2+ flux increased from 50 to 100mmol/L Cd2+ stress, however, decreased at 150mmol/LCd2+. Cellulose and hemicellulose in leaves, stems and roots did not change significantly under Cd stress, while pectin decreased significantly. The NSC content of both leaves and stems showed significant changes under Cd stress while the root NSC content was not affected. In both leaves and roots, the ABA content significantly increased under Cd stress, while the GA3, IAA and JA-ME content significantly decreased. Both xylem hydraulic conductivity and xylem water potential decreased with Cd stress, however, tracheid diameter and double wall thickness of the stems and roots were not affected. High Cd intensity was found in both the stem xylem and phloem in all Cd stressed treatments. Our study highlighted the in-situ observation of Cd distribution in both the xylem and phloem, and demonstrated the instant response of physiological traits such as xylem water potential, xylem hydraulic conductivity, root Cd2+ flux, NSC content, as well as phytohormonal content under Cd stress, and the less affected traits such as xylem anatomical traits, cellulose and hemicellulose.

Effects of nitrogen addition and leaf age on needle traits and the relationships among traits in Pinus koraiensis

Leaf functional traits are essential for the growth and adaptation of trees to environmental changes. However, how needle traits and trait–trait relationships respond to different nitrogen (N) addition rates and leaf ages remain unclear for evergreen coniferous species. Our study aimed to investigate the effects of N addition and leaf age on needle traits and their bivariate correlations. Understanding resource utilization and distribution strategies during tree growth under the background of increased N deposition is important. We conducted a six-year manipulative field experiment in a Pinus koraiensis plantation in northeastern China with variable N addition levels: control (no N addition, CK); 20 kg N ha−1yr−1 (low-N, LN); 40 kg N ha−1yr−1 (medium-N, MN); and 80 kg N ha−1yr−1 (high-N, HN). The needle trait data were collected in September. The morphological traits, i.e., specific leaf area (SLA) and leaf dry matter content (LDMC); and chemical traits, i.e., leaf nitrogen concentration (LNC), leaf phosphorus concentration (LPC), and leaf nonstructural carbohydrates (NSC) concentrations, were measured to investigate the changes in needle traits and their bivariate correlations. We found that N addition and needle age significantly affected needle traits, and the effect of needle age was much stronger than that of N addition. N addition and needle age had no significant interaction effects on any of the traits except for the LNC. Due to the dispersal trait values of current-year needles, there were almost no significant bivariate correlations between traits under different N addition rates. The SLA–LDMC and LDMC–LNC relationships were found to differ among the various N addition rates, and other traittrait relationships may be relatively stable because of the weak plasticity of older needles. Additionally, needle age significantly affected the slope of the SLA–LDMC relationship and the elevation of the LDMC–LPC and NSC–LNC relationships, indicating differences in the ecological strategies of needles at different ages. In summary, our findings emphasize that trait–trait relationships in current-year and older needles were inconsistent in response to N addition and that the optimal allocation of resources in needles was based on their functional needs during leaf aging.

Different responses of oxygen and hydrogen isotopes in leaf and tree-ring organic matter to lethal soil drought.

The oxygen and hydrogen isotopic composition (δ18O, δ2H) of plant tissues are key tools for the reconstruction of hydrological and plant physiological processes and may therefore be used to disentangle the reasons for tree mortality. However, how both elements respond to soil drought conditions before death has rarely been investigated. To test this, we performed a greenhouse study and determined predisposing fertilization and lethal soil drought effects on δ18O and δ2H values of organic matter in leaves and tree rings of living and dead saplings of five European tree species. For mechanistic insights, we additionally measured isotopic (i.e. δ18O and δ2H values of leaf and twig water), physiological (i.e. leaf water potential and gas-exchange) and metabolic traits (i.e. leaf and stem non-structural carbohydrate concentration, carbon-to-nitrogen ratios). Across all species, lethal soil drought generally caused a homogenous 2H-enrichment in leaf and tree-ring organic matter, but a low and heterogenous δ18O response in the same tissues. Unlike δ18O values, δ2H values of tree-ring organic matter were correlated with those of leaf and twig water and with plant physiological traits across treatments and species. The 2H-enrichment in plant organic matter also went along with a decrease in stem starch concentrations under soil drought compared with well-watered conditions. In contrast, the predisposing fertilization had generally no significant effect on any tested isotopic, physiological and metabolic traits. We propose that the 2H-enrichment in the dead trees is related to (i) the plant water isotopic composition, (ii) metabolic processes shaping leaf non-structural carbohydrates, (iii) the use of carbon reserves for growth and (iv) species-specific physiological adjustments. The homogenous stress imprint on δ2H but not on δ18O suggests that the former could be used as a proxy to reconstruct soil droughts and underlying processes of tree mortality.

Seasonal patterns of nonstructural carbohydrate storage and mobilization in two tree species with distinct life-history traits.

Nonstructural carbohydrates (NSC) are essential for tree growth and adaptation, yet our understanding of the seasonal storage and mobilization dynamics of whole-tree NSC is still limited, especially when tree functional types are involved. Here, Quercus acutissima Carruth. and Pinus massoniana Lamb. with distinct life-history traits (i.e., a deciduous broadleaf species vs. an evergreen coniferous species) were studied to assess the size and seasonal fluctuations of organ and whole-tree NSC pools with a focus on comparing differences in carbon resource mobilization patterns between the two species. We sampled the organs (leaf, branch, stem, and root) of the target trees repeatedly over four seasons of the year. Then, NSC concentrations in each organ were paired with biomass estimates from the allometric model to generate whole-tree NSC pools. The seasonal dynamics of the whole-tree NSC of Q. acutissima and P. massoniana reached the peak in autumn and summer, respectively. The starch pools of the two species were supplemented in the growing season while the soluble sugar pools were the largest in the dormant season. Seasonal dynamics of organ-level NSC concentrations and pools were affected by organ type and tree species, with above-ground organs generally increasing during the growing season and P. massoniana roots decreasing during the growing season. In addition, the whole-tree NSC pools of P. massoniana were larger but Q. acutissima showed larger seasonal fluctuations, indicating that larger storage was not associated with more pronounced seasonal fluctuations. We also found that the branch and root were the most dynamic organs of Q. acutissima and P. massoniana, respectively, and were the major suppliers of NSC to support tree growth activities. These results provide fundamental insights into the dynamics and mobilization patterns of NSC at the whole-tree level, and have important implications for investigating environmental adaptions of different tree functional types.

Species-specific responses of C and N allocation to N addition: evidence from dual 13C and 15N labeling in three tree species

Soil nitrogen (N) availability affects plant carbon (C) utilization. However, it is unclear how various tree functional types respond to N addition in terms of C assimilation, allocation, and storage. Here, a microcosm experiment with dual 13C and 15N labeling was conducted to study the effects of N addition (i.e., control, 0 g N kg−1; moderate N addition, 1.68 g N kg−1; and high N addition, 3.36 g N kg−1 soil) on morphological traits, on changes in nonstructural carbohydrates (NSC) in different organs, as well as on C and N uptake and allocation in three European temperate forest tree species (i.e., Acer pseudoplatanus, Picea abies and Abies alba). Our results demonstrated that root N uptake rates of the three tree species increased by N addition. In A. pseudoplatanus, N uptake by roots, N allocation to aboveground organs, and aboveground biomass allocation significantly improved by moderate and high N addition. In A. alba, only the high N addition treatment considerably raised aboveground N and C allocation. In contrast, biomass as well as C and N allocation between above and belowground tissues were not altered by N addition in P. abies. Meanwhile, NSC content as well as C and N coupling (represented by the ratio of relative 13C and 15N allocation rates in organs) were affected by N addition in A. pseudoplantanus and P. abies but not in A. alba. Overall, A. pseudoplatanus displayed the highest sensitivity to N addition and the highest N requirement among the three species, while P. abies had a lower N demand than A. alba. Our findings highlight that the responses of C and N allocation to soil N availability are species-specific and vary with the amount of N addition.

External aluminium supply regulates photosynthesis and carbon partitioning in the Al-accumulating tropical shrub Melastoma malabathricum.

Aluminium (Al) is toxic to most plants, but recent research has suggested that Al addition may stimulate growth and nutrient uptake in some species capable of accumulating high tissue Al concentrations. The physiological basis of this growth response is unknown, but it may be associated with processes linked to the regulation of carbon assimilation and partitioning by Al supply. To test alternative hypotheses for the physiological mechanism explaining this response, we examined the effects of increasing Al concentrations in the growth medium on tissue nutrient concentrations and carbon assimilation in two populations of the Al-accumulator Melastoma malabathricum. Compared to seedlings grown in a control nutrient solution containing no Al, mean rates of photosynthesis and respiration increased by 46% and 27%, respectively, total non-structural carbohydrate concentrations increased by 45%, and lignin concentration in roots decreased by 26% when seedlings were grown in a nutrient solution containing 2.0 mM Al. The concentrations of P, Ca and Mg in leaves and stems increased by 31%, 22%, and 26%, respectively, in response to an increase in nutrient solution Al concentration from 0 to 2.0 mM. Elemental concentrations in roots increased for P (114%), Mg (61%) and K (5%) in response to this increase in Al concentration in the nutrient solution. Plants derived from an inherently faster-growing population had a greater relative increase in final dry mass, net photosynthetic and respiration rates and total non-structural carbohydrate concentrations in response to higher external Al supply. We conclude that growth stimulation by Al supply is associated with increases in photosynthetic and respiration rates and enhanced production of non-structural carbohydrates that are differentially allocated to roots, as well as stimulation of nutrient uptake. These responses suggest that internal carbon assimilation is up-regulated to provide the necessary resources of non-structural carbohydrates for uptake, transport and storage of Al in Melastoma malabathricum. This physiological mechanism has only been recorded previously in one other plant species, Camellia sinensis, which last shared a common ancestor with M. malabathricum more than 120 million years ago.

Non-structural carbohydrate dynamics of Pinus yunnanensis seedlings under drought stress and re-watering

Non-structural carbohydrates (NSC) are an important “buffer” for maintaining plant physiological functions under drought conditions; however, our understanding of the dynamics of NSC at the organ level during sustained drought of varying intensities and re-watering remains poor. In this study, two-year-old Pinus yunnanensis seedlings were subjected to drought and re-watering trials. Plants were subjected to three drought intensities (light drought, moderate drought, and severe drought) as well as control conditions (suitable moisture) for 51 days, including 30 days of drought followed by 21 days of re-watering for drought-treated seedlings, to study the dynamics of NSC in the leaves, stems, coarse roots, and fine roots. Changes in the distribution of NSC concentrations in the organ of P. yunnanensis seedlings under drought stress varied; in the early drought stages, the drought resistance of P. yunnanensis seedlings was enhanced by increasing soluble sugar concentrations; in later stages of drought, the stored starch in organs, stems, and coarse roots was consumed. Drought inhibited the growth of P. yunnanensis seedlings, but the maximum limit of drought tolerance was not reached under the different drought treatments after 30 days. P. yunnanensis seedlings in all treatment groups resumed growth after re-watering, and the growth of seedlings was actually promoted during re-watering in the moderate drought treatment group, indicating that drought induced the compensatory growth of seedlings. The growth of P. yunnanensis seedlings during re-watering was inhibited in the severe drought treatment group, and NSC continued to be regulated in seedlings in this group. Given that P. yunnanensis seedlings maintain growth through the consumption of coarse root starch in the late stages of drought, seedlings with a larger root-to-shoot ratio should be selected for cultivation in actual production. Based on our findings, exposure to moderate drought stress can enhance the drought tolerance of P. yunnanensis seedlings and promote the compensatory growth of seedlings.

Light availability during spring and autumn determines growth and survival of regenerated Korean pine seedlings

ABSTRACT The regeneration of Korean pine (Pinus koraiensis Sieb. et Zucc.) is a core issue for restoration from secondary to primary forests in northeastern China. However, Korean pine exhibits limited recruitment within its own canopy in primary forests, but demonstrates excellent regeneration in secondary forests. Until now, the mechanisms underlying this phenomenon have not been satisfactorily explained. Hence, we conducted studies in both primary and secondary forests to investigate whether light availability during the non-growing season determines the fate of regeneration. In spring and autumn, understory light availability in the secondary forest was significantly better than that in the primary forest. The biomass and non-structural carbohydrate (NSC) concentrations of natural pine seedlings were considerably greater in the secondary forests compared to primary forests. The carbohydrate concentrations increased during spring and autumn and decreased in summer. In the manipulation experiment, the biomass of seedlings was positively correlated with the organ NSCs in spring, while the survival rates were correlated with NSCs in autumn. The light availability during the spring and autumn seasons may have a crucial role in the regeneration of Korean pine seedlings. This finding may support our practical management approach for the transformation of secondary forests into Korean pine forests.

Effects of rainfall patterns in dry and rainy seasons on the biomass, ecostoichiometric characteristics, and NSC content of Fraxinus malacophylla seedlings.

With global climate change and rising temperatures, rainfall will change. The impact of global rainfall changes on ecosystems has prompted people to delve deeper into how changes in rainfall affect plant growth; Plant biomass, nutrient element content, and non-structural carbohydrate content are very sensitive to changes in precipitation. Therefore, understanding the impact of rainfall changes on seedlings is crucial. However, it is currently unclear how the seedlings of Fraxinus malacophylla Hemsl in rocky desertification areas respond to changes in rainfall. In this study, the response of biomass, nutrient accumulation, and NSC content of Fraxinus malacophylla Hemsl seedlings to different rainfall intervals and rainfall during the dry and rainy seasons was studied. Use natural rainfall duration of 5 days (T) and extended rainfall duration of 10 days(T+) as rainfall intervals; average monthly rainfall was used as the control (W), with a corresponding 40% increase in rainfall (W+) and a 40% decrease in rainfall (W-) as rainfall treatments. The research results indicate that the biomass of roots, stems, and leaves, as well as the accumulation of C, N, and P in Fraxinus malacophylla Hemsl seedlings increase with the increase of rainfall, while the soluble sugar and starch content show a pattern of first increasing and then decreasing. The biomass and nutrient accumulation of each organ showed root>leaf>stem. Except for the beginning of the dry season, prolonging the duration of rainfall in other periods inhibits the biomass accumulation of Fraxinus malacophylla Hemsl seedlings, and promotes the accumulation of C, N, and P nutrients and an increase in soluble sugar and starch content. There was a significant positive correlation (P<0.05) between the nutrient contents of C, N, and P in various organs, as well as between soluble sugar and starch content; And N: P>16, plant growth is limited by P element. These results indicate that changes in rainfall can affect the growth and development of Fraxinus malacophylla Hemsl seedlings, increasing rainfall can promote biomass and nutrient accumulation of Fraxinus malacophylla Hemsl seedlings, and prolonging rainfall intervals and reducing rainfall have inhibitory effects on them. The exploration of the adaptation of Fraxinus malacophylla Hemsl seedlings to rainfall patterns has promoted a basic understanding of the impact of rainfall changes on the growth of Fraxinus malacophylla Hemsl. This provides a theoretical basis for understanding how Fraxinus malacophylla Hemsl can grow better under rainfall changes and for future management of Fraxinus malacophylla Hemsl artificial forests in rocky desertification areas.

Quantifying the Individual and Combined Effects of Short-Term Heat Stress at Booting and Flowering Stages on Nonstructural Carbohydrates Remobilization in Rice

Rice production is threatened by climate change, particularly heat stress (HS). Nonstructural carbohydrates (NSCs) remobilization is a key physiological mechanism that allows rice plants to cope with HS. To investigate the impact of short-term HS on the remobilization of nonstructural carbohydrates (NSCs) in rice, two cultivars (Huaidao-5 and Wuyunjing-24) were subjected to varying temperature regimes: 32/22/27 °C as the control treatment, alongside 40/30/35 °C and 44/34/39 °C, for durations of 2 and 4 days during the booting, flowering, and combined stages (booting + flowering) within phytotrons across the years 2016 and 2017. The findings revealed that the stem’s NSC concentration increased, while the panicle’s NSCs concentration, the efficiency of NSCs translocation from the stem, and the stem NSC contribution to grain yield exhibited a consistent decline. Additionally, sugar and starch concentrations increased in leaves and stems during late grain filling and maturity stages, while in panicles, the starch concentration decreased and sugar concentration increased. The heat-tolerant cultivar, Wuyunjing-24, exhibited higher panicle NSC accumulation under HS than the heat-sensitive cultivar, Huaidao-5, which had more stem NSC accumulation. The flowering stage was the most vulnerable to HS, followed by the combined and booting stages. Heat degree days (HDDs) were utilized to quantify the effects of HS on NSC accumulation and translocation, revealing that the flowering stage was the most affected. These findings suggest that severe HS makes the stem the primary carbohydrate storage sink, and alleviation under combined HS aids in evaluating NSC accumulation, benefiting breeders in developing heat-tolerant rice varieties.

Changes in Growth Parameters, C:N:P Stoichiometry and Non-Structural Carbohydrate Contents of Zanthoxylum armatum Seedling in Response to Five Soil Types

Zanthoxylum armatum (Z. armatum) is an economic crop widely planted for both spice and medicinal purposes in Southwest China. Soil is a key environmental condition that affects seedling growth and development, and screening suitable soil types is of great significance for the large-scale cultivation of crops. This study designed growth experiments of Z. armatum seedlings in red soil (RS), yellow soil (YS), acidic purple soil (ACPS), alkaline purple soil (ALPS), and alluvial soil (AS) to screen for more suitable soil types. The growth traits of Z. armatum seedlings and the carbon (C), nitrogen (N), phosphorus (P), C:N:P stoichiometry, and non-structural carbohydrate (NSC) content of different organs were comparatively analyzed. The results showed that the morphological indexes of Z. armatum seedlings cultured in AS were better than those in the other four soils.AS and RS may be beneficial for the culture of Z. armatum seedlings due to higher nutrient levels in three organs. Two-factor ANOVA and PCA analysis showed that C, N, and P and their proportions would affect the uptake and distribution of NSC in various organs of Z. armatum seedlings. These results showed that soil types and plant organs significantly affected the accumulation and distribution of N, P, and NSC in Z. armatum seedlings. These results are conducive to screening soil types suitable for the growth and development of Z. armatum and provide data support for further large-scale cultivation of Z. armatum in suitable areas.

Responses of Fine Root Morphological and Chemical Traits among Branch Orders to Forest Thinning in Pinus massoniana Plantations

Fine roots play an essential role in biogeochemical cycling in forest ecosystems; however, little is known about the response of fine root morphology and chemistry in different root orders to forest management activities such as forest thinning. We investigated the fine root morphological and chemical traits in different root orders of Pinus massoniana under different thinning intensities, namely no thinning, low-intensity thinning (LIT), middle-intensity thinning (MIT), and high-intensity thinning (HIT) (0%, 25%, 45%, and 65% of individual trees eliminated, respectively). We found that forest thinning increased the root diameter (RD) of absorptive roots and decreased that of transport roots, while the trend for the specific root length (SRL) was the opposite. LIT and MIT could increase specific surface area (SSA), especially the SSA of absorptive roots in the MIT treatment. The root tissue density (RTD) of all root sequences in the LIT treatment decreased but increased in the HIT treatment. For the fine root chemical traits, thinning increased the root carbon concentration (RCC) of absorptive roots. The root nitrogen concentration (RNC) and root phosphorus concentration (RPC) of first- to fourth-order roots increased in the LIT and MIT treatments after thinning. Meanwhile, thinning increased root lignin, cellulose, and non-structural carbohydrate (NSC) concentrations. Soil temperature, nitrate, and microbial biomass carbon were factors affecting variations in fine root morphology and chemistry. Forest thinning was likely to shift the absorptive roots’ foraging strategy into a resource-conserving one. Thinning increased fine root chemical traits in most root orders. These findings contributed to our ability to predict how belowground ecological processes are mediated by fine roots under forest management activities.

Physiological Nitrogen Uptake and Utilisation Responses in Two Native Plants from the Qinghai-Tibet Plateau under Different Water and Fertiliser Conditions

Drought and poor soil quality are the main characteristics of extreme environments in arctic–alpine areas. Understanding how herbaceous plants in alpine grasslands maintain the normal supply and utilisation of nutrients under different rainfall conditions is key to maintaining population stability. In the present study, the native plants Poa crymophila and Stipa purpurea of the Qinghai–Tibet Plateau were used to conduct a controlled experiment involving water and fertiliser to analyse their physiological responses in terms of nutrient uptake and utilisation. The results showed that decreased soil moisture increased proline and non-structural carbohydrates in P. crymophila, mainly accumulating in the leaves and stems. Nitrogen (N) addition promoted proline accumulation, whereas nonstructural carbohydrate content decreased. However, the proline and non-structural carbohydrate contents of S. purpurea were less affected by water and fertiliser. Additionally, drought restricted rhizospheric and non-rhizospheric alkaline-hydrolysed N release, increased rapidly available phosphorus (RAP) content in rhizospheric soil, limited root growth, and reduced surface area, root length, and root volume. Both aboveground and underground N fertiliser utilisation rates decreased. Under well-hydrated conditions (WH), high N levels increased rhizospheric alkaline-hydrolysed N and urease activity while inhibiting RAP and activity of alkaline phosphatase contents, thereby limiting root growth and reducing N fertiliser utilisation. The results indicate that both plant species have relatively low overall nutrient requirements that are limited mainly by water availability. The addition of low amounts of fertiliser is beneficial for nutrient release and utilisation, improving their adaptability to arctic–alpine environments and their suitability and superiority in the community. This study has significant implications for nutrient management and ecological restoration measures in arctic–alpine grasslands.
