The goal: to study the quantitative parameters of the effector NKand NKT-cells of ulcerative colitis patients in case of different activity rate of autoimmune process. Materials and methods: From 2011 till 2012 42 ulcerative colitis (UC) patients being under treatment in the Intestines Pathology Department in CSRIG (Central Scientific Research Institute of Gastroenterology) were studied. 32 of them men, 10 women. The age of the patients ranged from 19 to 70 years, average age 41,3±2,3 (M±σ). The diagnosis was verified using data of clinical, laboratory, immunological, histological and instrumental methods of research. Immunophenotyping of NKand NKT-cells by the method of flow cytometry using twoparameter reagents (CD3-FITC/CD16/56-PE and CD3+FITC/CD16/56-PE, «Bekman Coulter») was performed. The sample was analyzed by flow cytometers FACSCalibur («Bekman Dickinson»). The statistic data analysis was performed using the computer program «STATISTICA 6.0», the statistical significance of the values was verified by the Student t-test. Results: The analysis of lymphocyte parameters determining the killer effect without the help of antigen recognizing receptor of natural killers (NK-cells) showed a significant (p<0,001) reduction of relative and absolute quantity of lymphocytes expressing CD3-CD16+56+ to 6,0±1,2% in patients with heavy UC severity comparing to the patients with mild and medium severity (12,3±1,9 and 17,5±2,5% respectively). A statistically significant reduction (up to 4,6±1,4%, p<0,001) of relative quantity of the CD3+CD16+56+NKT-cells was observed during the study of the patients with heavy UC severity comparing to the patients with mild severity (9,4±1,4%); there was no significant difference between medium and heavy UC severity (p=0,834). Insufficient functioning of natural killers (NK-cells) should be taken into account in connection to the problem of colorectal cancer development in patients with long-lasting progressive state of UC. Conclusion: The obtained data show the decrease of relative and absolute abundance of NKand NKT-cells in patients with heavy UC severity, but it should be noted that apart from the quantitative parameters an important role in the
