Genotype-specific nonphotochemical quenching responses to nitrogen deficit are linked to chlorophyll a to b ratios

Non-photochemical quenching (NPQ) protects plants from photodamage caused by excess light energy. Substantial variation in NPQ has been reported among different genotypes of the same species. However, comparatively little is known about how environmental perturbations, including nutrient deficits, impact natural variation in NPQ kinetics. Here, we analyzed a natural variation in NPQ kinetics of a diversity panel of 225 maize (Zea mays L.) genotypes under nitrogen replete and nitrogen deficient field conditions. Individual maize genotypes from a diversity panel exhibited a range of changes in NPQ in response to low nitrogen. Replicated genotypes exhibited consistent responses across two field experiments conducted in different years. At the seedling and pre-flowering stages, a similar portion of the genotypes (∼33%) showed decrease, no-change or increase in NPQ under low nitrogen relative to control. Genotypes with increased NPQ under low nitrogen also showed greater reductions in dry biomass and photosynthesis than genotypes with stable NPQ when exposed to low nitrogen conditions. Maize genotypes where an increase in NPQ was observed under low nitrogen also exhibited a reduction in the ratio of chlorophyll a to chlorophyll b. Our results underline that since thermal dissipation of excess excitation energy measured via NPQ helps to balance the energy absorbed with energy utilized, the NPQ changes are the reflection of broader molecular and biochemical changes which occur under the stresses such as low soil fertility. Here, we have demonstrated that variation in NPQ kinetics resulted from genetic and environmental factors, are not independent of each other. Natural genetic variation controlling plastic responses of NPQ kinetics to environmental perturbation increases the likelihood it will be possible to optimize NPQ kinetics in crop plants for different environments.

Nitrogen starvation causes lipid remodeling in Rhodotorula toruloides

BackgroundThe oleaginous yeast Rhodotorula toruloides is a promising chassis organism for the biomanufacturing of value-added bioproducts. It can accumulate lipids at a high fraction of biomass. However, metabolic engineering efforts in this organism have progressed at a slower pace than those in more extensively studied yeasts. Few studies have investigated the lipid accumulation phenotype exhibited by R. toruloides under nitrogen limitation conditions. Consequently, there have been only a few studies exploiting the lipid metabolism for higher product titers.ResultsWe performed a multi-omic investigation of the lipid accumulation phenotype under nitrogen limitation. Specifically, we performed comparative transcriptomic and lipidomic analysis of the oleaginous yeast under nitrogen-sufficient and nitrogen deficient conditions. Clustering analysis of transcriptomic data was used to identify the growth phase where nitrogen-deficient cultures diverged from the baseline conditions. Independently, lipidomic data was used to identify that lipid fractions shifted from mostly phospholipids to mostly storage lipids under the nitrogen-deficient phenotype. Through an integrative lens of transcriptomic and lipidomic analysis, we discovered that R. toruloides undergoes lipid remodeling during nitrogen limitation, wherein the pool of phospholipids gets remodeled to mostly storage lipids. We identify specific mRNAs and pathways that are strongly correlated with an increase in lipid levels, thus identifying putative targets for engineering greater lipid accumulation in R. toruloides. One surprising pathway identified was related to inositol phosphate metabolism, suggesting further inquiry into its role in lipid accumulation.ConclusionsIntegrative analysis identified the specific biosynthetic pathways that are differentially regulated during lipid remodeling. This insight into the mechanisms of lipid accumulation can lead to the success of future metabolic engineering strategies for overproduction of oleochemicals.

Overexpression of rice OsNRT1.1A/OsNPF6.3 enhanced the nitrogen use efficiency of wheat under low nitrogen conditions.

Overexpression of OsNRT1.1A promotes early heading and increases the tolerance in wheat under nitrogen deficiency conditions. The application of inorganic nitrogen (N) fertilizers is a major driving force for crop yield improvement. However, the overuse of fertilizers significantly raises production costs and leads to environmental problems, making it critical to enhance crop nitrogen use efficiency (NUE) for the sake of sustainable agriculture. In this study, we created a series of transgenic wheat lines carrying the rice OsNRT1.1A gene, which encodes a nitrate transporter, to investigate its possible application in improving NUE in wheat. The transgenic wheat exhibited traits such as early maturation that were highly consistent with the overexpression of OsNRT1.1A in Arabidopsis and rice. However, we also observed that overexpression of the OsNRT1.1A gene in wheat can facilitate the growth of roots under low N conditions but has no effect on other aspects of growth and development under normal N conditions. Thus, it may lead to the improvement of wheat low N tolerance,which is different from the effects reported in other plants. A field trial analysis showed that transgenic wheat exhibited increased grain yield per plant under low N conditions. Moreover, transcriptome analysis indicated that OsNRT1.1A increased the expression levels of N uptake and utilization genes in wheat, thereby promoting plant growth under low N conditions. Taken together, our results indicated that OsNRT1.1A plays an important role in improving NUE in wheat with low N availability.

Physiological and Transcriptional Characteristics of Banana Seedlings in Response to Nitrogen Deficiency Stress

Nitrogen is a crucial element for the growth and development of plants, directly affecting crop growth and yield. To investigate the physiological and molecular mechanism of nitrogen-deficiency stress, we conducted an investigation into the effects of different nitrogen levels on the growth, photosynthetic characteristics, and gene transcription levels of banana seedlings. Compared with the control group with normal nitrogen levels (NN), the height of plants receiving Reduced-N (NR), Low-N (LN), and N-Free (NF) treatments was decreased by 0.45 cm, 2.5 cm, and 3.25 cm, respectively. Their dry weight was reduced by 1.63 g, 2.99 g, and 2.88 g, respectively. Conversely, the dry weight of the underground plant part in the LN and NF treatment groups exhibited an increase of 0.13 g and 0.16 g, respectively. Regarding photosynthetic characteristics, the Specialty Products Agricultural Division (SPAD) values of the NR, LN, and NF treatments showed reductions of 15.5%, 30.4%, and 35.9%, respectively, compared with those of the control treatments. The values of maximum photosynthetic efficiency (Fv/Fm), actual photosynthetic efficiency (Y(Ⅱ)), and relative electron transfer (ETR) of the banana seedlings decreased to different degrees after NR, LN, and NF treatment, and their values were positively correlated with N levels. Gene transcription analysis showed that N transport-related proteins, including NRT1.7, NRT2.3a, NRT2.3b, and NRT2.5, were significantly up-regulated to increase the nitrogen absorption capacity of plant roots. On the other hand, various transcription factors including GRAS, MYB, and WRKY were notably up-regulated, facilitating root growth and the expanding root absorption area, thereby enhancing nitrogen uptake. Furthermore, genes associated with endogenous hormone metabolic pathways such as gibberellin (GA), strigolactone (SL), and brassinosteroids (BR) were activated in banana plants subjected to low nitrogen stress, enhancing the plant’s ability to adapt to nitrogen-deficient conditions. These findings offer valuable insights into understanding the transcriptional regulatory mechanisms governing banana responses to low nitrogen stress and breeding new varieties with improved nutrient utilization.

A new isolate cold-adapted Ankistrodesmus sp. OR119838: influence of light, temperature, and nitrogen concentration on growth characteristics and biochemical composition using the two-stage cultivation strategy.

Natural-based chemicals from microalgae such as lipids and pigments are the interests in industries and the bioeconomy. Cold-adapted Ankistrodesmus sp. OR119838, an isolated strain from Cheshmeh-Sabz Lake in northeastern Iran, was cultivated using a two-stage culture strategy under different environmental conditions. With doubling the nitrate concentration at the vegetative stage (170mg/L) and increasing the light intensity (180µmol photons/m2/s) the highest specific growth rate (0.61 ± 0.02 per day) and biomass productivity (121.1 ± 7.2 mg/L/day) were observed at 25°C. In the optimal growth condition Chl a and Chl b contents of Ankistrodesmus sp. OR119838 reached the highest amount (11.07 ± 0.14 and 11.23 ± 0.29 µg/mL, respectively) at 25°C. While carotenoid content correlated negatively with optimum biomass productivity (-0.708) and had the best value (12.23 ± 0.29µg/mL) in nitrogen deficiency (42 mg/L) and intense light conditions (180µmol photons/m2/s) at 15°C. Lipid content was increased with declined nitrate concentration (42mg/L), high light intensity, and 180µmol photons/m2/s at 25°C. The highest percentage of polyunsaturated fatty acids (71.94%) and α-linolenic acid (57.73 ± 6.63%) was observed in conditions with 170mg/L nitrate concentration and low light intensity (40µmol photons/m2/ s) at the low temperature (15°C). While saturated fatty acids content (43.27%) and palmitic acid reached the highest amount under 40µmol photons/m2/s, 42mg/L nitrate at 25°C (35.02 ± 5.33%). Biomass productivity of Ankistrodesmus sp. OR119838, as a cold-adapted strain, decreased by only 8.2% with a 10-degree decline in temperature. Therefore, this strain has good potential to grow in open ponds by tolerating the daily temperature fluctuations.

Photofermentative Biohydrogen Production Enhancement with Nanoparticle

This study will explore the use of a biological pathway known as photo fermentation for the production of biohydrogen under anoxic and nitrogen deficient conditions due to its high hydrogen production, and to emphasize the role of a dominating enzyme, the nitrogenase enzyme, in catalyzing the process of hydrogen evolution from a medium composed of Purple Non-Sulfur Bacteria (Rhodopseudomonas Palustris) with glycerol as the substrate. For Purple Non-Sulphur Bacteria, the optimal H2 production temperature ranges between 30 °C to 36 °C. Simultaneously, illumination is applied by LED lights and is varied from no string LED illumination, to single string illumination (240 W/m2), and double string illumination (480 W/m2) to simulate and examine performance in outdoor conditions and achieved a maximum of 428% increase in hydrogen productivity compared to previous studies. Consequently, this suggests that it is economically appealing to use the non-growing R. palustris bacterium as the chosen biocatalyst for continuous hydrogen production. These preliminary results constitute the development of continuous H2 production which evolves into the green fuel of the future production.

Selecting for a high lipid accumulating microalgae culture by dual growth limitation in a continuous bioreactor

A dual-growth-limited continuous operated bioreactor (chemostat) was used to enhance lipid accumulation in an enrichment culture of microalgae. The light intensity and nitrogen concentration where both limiting factors resulting in high lipid accumulation in the mixed culture. Both conditions of light and nitrogen excess and deficiency were tested. Strategies to selectively enrich for a phototrophic lipid-storing community, based on the use of different nitrogen sources (ammonium vs. nitrate) and vitamin B supplementation in the growth medium, were evaluated. The dual limitation of both nitrogen and light enhanced the accumulation of storage compounds. Ammoniacal nitrogen was the preferred nitrogen source. Vitamin B supplementation led to a doubling of the lipid productivity. The availability of vitamins played a key role in selecting an efficient lipid-storing community, primarily consisting of Trebouxiophyceae (with an 82 % relative abundance among eukaryotic microorganisms). The obtained lipid volumetric productivity (387 mg L−1 d−1) was among the highest reported in literature for microalgae bioreactors. Lipid production by the microalgae enrichment surpassed the efficiencies reported for continuous microalgae pure cultures, highlighting the benefits of mixed-culture photo-biotechnologies for fuels and food ingredients in the circular economy.

Spatio-temporal coherence of circadian clocks and temporal control of differentiation in Anabaena filaments.

Circadian clock arrays in multicellular filaments of the heterocyst-forming cyanobacterium Anabaena sp. strain PCC 7120 display remarkable spatio-temporal coherence under nitrogen-replete conditions. To shed light on the interplay between circadian clocks and the formation of developmental patterns, we followed the expression of a clock-controlled gene under nitrogen deprivation, at the level of individual cells. Our experiments showed that differentiation into heterocysts took place preferentially within a limited interval of the circadian clock cycle, that gene expression in different vegetative intervals along a developed filament was discoordinated, and that the circadian clock was active in individual heterocysts. Furthermore, Anabaena mutants lacking the kaiABC genes encoding the circadian clock core components produced heterocysts but failed in diazotrophy. Therefore, genes related to some aspect of nitrogen fixation, rather than early or mid-heterocyst differentiation genes, are likely affected by the absence of the clock. A bioinformatics analysis supports the notion that RpaA may play a role as master regulator of clock outputs in Anabaena, the temporal control of differentiation by the circadian clock and the involvement of the clock in proper diazotrophic growth. Together, these results suggest that under nitrogen-deficient conditions, the clock coherent unit in Anabaena is reduced from a full filament under nitrogen-rich conditions to the vegetative cell interval between heterocysts.IMPORTANCECircadian clocks, from unicellular organisms to animals, temporally align biological processes to day and night cycles. We study the dynamics of a circadian clock-controlled gene at the individual cell level in the multicellular filamentous cyanobacterium Anabaena, under nitrogen-stress conditions. Under these conditions, some cells along filaments differentiate to carry out atmospheric nitrogen fixation and lose their capability for oxygenic photosynthesis. We found that clock synchronization is limited to organismic units of contiguous photosynthetic cells, contrary to nitrogen-replete conditions in which clocks are synchronized over a whole filament. We provided evidence that the circadian clock regulates the process of differentiation, allowing it to occur preferentially within a limited time window during the circadian clock period. Lastly, we present evidence that the signal from the core clock to clock-regulated genes is conveyed in Anabaena as in unicellular cyanobacteria.

Phosphorus-driven regime shift from heterotrophic to autotrophic diazotrophs in a deep alpine lake

Biological nitrogen fixation plays a critical role in maintaining primary production, particularly in systematic nitrogen deficiency. However, little is known about the dynamics within diazotrophic community facing ongoing nutrient enrichment in freshwater lakes. Here, a consecutive five-year investigation on diazotrophic community was conducted in Lake Fuxian, an oligotrophic deep alpine lake on the trajectory to eutrophic state. Results showed a regime shift from heterotrophic to autotrophic diazotrophs induced by total phosphorus (TP) enrichment. Specifically, heterotrophic diazotrophs dominated the diazotrophic community when TP was lower than 21.8 μg/L, whereas heterotrophic diazotrophs or diazotrophic Cyanobacteria randomly dominated when TP ranged between 21.8 μg/L and 28.8 μg/L. When TP was higher than 28.8 μg/L, diazotrophic Cyanobacteria accounted for 60.4%-97.7% of the total N2-fixers, indicating diazotrophic biodiversity significantly declined under TP enrichment scenario. Moreover, the dominance of diazotrophic Cyanobacteria further facilitated phytoplankton growth, which strengthened positive feedback between phytoplankton and phosphorus under nitrogen deficiency conditions. This is the first report on the threshold-like state responses of freshwater diazotrophs to environmental drivers. Our study expands the knowledge of the diazotrophic dynamics in freshwater ecosystems and contributes quantitative evidence of ecological thresholds for future environmental policymaking.

EFFECTS OF NITROGEN NUTRIENT ON THE PRODUCTION OF MYCOSPORINE-LIKE AMINO ACIDS IN NOSTOC SPECIES

Mycosporine-like amino acids MAAs belong to a family of secondary metabolites produced by a wide range of different organisms. These compounds had several potential applications in biomedical, in cosmetics and toiletries. In the present study, three Nostoc strains distributed in soil and freshwater were tested for MAA production under different nitrogen conditions. UV-B radiation significantly enhanced the production of MAAs in Nostoc linckia, Nostoc muscorum and Nostoc paludosum under nitrogen sufficient and deficient conditions. The contents of MAAs significantly increased in these three Nostoc strains with prolonged UV-B treatment under both nitrogen conditions. Both UV-B exposed N. muscorum and N. paludosum produced much less MAAs under nitrogen deficiency in comparison to culture under nitrogen sufficiency, respectively. There was just little difference of MAA contents in UV-B treated N. linckia when it was cultured under both nitrogen conditions. After separation by HPLC and characterization by in-line absorption spectra and mass spectra, one type of MAA porphyra was induced by UV-B in N. linckia under nitrogen sufficient and deficient conditions, and N. muscorum synthesized another kind of MAA shinorine under nitrogen sufficient and deficient conditions. However, UV-B induced N. paludosum to produce porphyra under nitrogen sufficient condition but shinorine under nitrogen deficient condition. These results indicated the nutrient nitrogen levels could regulate the types and amounts of MAAs in Nostoc species. These strains would be a better source for the industrial production of MAAs that can be used as organisms for studying the biosynthetic route of MAAs in nitrogen conditions.

Analysis of Functional Single-Nucleotide Polymorphisms (SNPs) and Leaf Quality in Tea Collection under Nitrogen-Deficient Conditions.

This study discusses the genetic mutations that have a significant association with economically important traits that would benefit tea breeders. The purpose of this study was to analyze the leaf quality and SNPs in quality-related genes in the tea plant collection of 20 mutant genotypes growing without nitrogen fertilizers. Leaf N-content, catechins, L-theanine, and caffeine contents were analyzed in dry leaves via HPLC. Additionally, the photochemical yield, electron transport efficiency, and non-photochemical quenching were analyzed using PAM-fluorimetry. The next generation pooled amplicon-sequencing approach was used for SNPs-calling in 30 key genes related to N metabolism and leaf quality. The leaf N content varied significantly among genotypes (p ≤ 0.05) from 2.3 to 3.7% of dry mass. The caffeine content varied from 0.7 to 11.7 mg g-1, and the L-theanine content varied from 0.2 to 5.8 mg g-1 dry leaf mass. Significant positive correlations were detected between the nitrogen content and biochemical parameters such as theanine, caffeine, and most of the catechins. However, significant negative correlations were observed between the photosynthetic parameters (Y, ETR, Fv/Fm) and several biochemical compounds, including rutin, Quercetin-3-O-glucoside, Kaempferol-3-O-rutinoside, Kaempferol-3-O-glucoside, Theaflavin-3'-gallate, gallic acid. From our SNP-analysis, three SNPs in WRKY57 were detected in all genotypes with a low N content. Moreover, 29 SNPs with a high or moderate effect were specific for #316 (high N-content, high quality) or #507 (low N-content, low quality). The use of a linear regression model revealed 16 significant associations; theaflavin, L-theanine, and ECG were associated with several SNPs of the following genes: ANSa, DFRa, GDH2, 4CL, AlaAT1, MYB4, LHT1, F3'5'Hb, UFGTa. Among them, seven SNPs of moderate effect led to changes in the amino acid contents in the final proteins of the following genes: ANSa, GDH2, 4Cl, F3'5'Hb, UFGTa. These results will be useful for further evaluations of the important SNPs and will help to provide a better understanding of the mechanisms of nitrogen uptake efficiency in tree crops.

Metagenome-assembled genomes reveal carbohydrate degradation and element metabolism of microorganisms inhabiting Tengchong hot springs, China

A hot spring is a distinctive aquatic environment that provides an excellent system to investigate microorganisms and their function in elemental cycling processes. Previous studies of terrestrial hot springs have been mostly focused on the microbial community, one special phylum or category, or genes involved in a particular metabolic step, while little is known about the overall functional metabolic profiles of microorganisms inhabiting the terrestrial hot springs. Here, we analyzed the microbial community structure and their functional genes based on metagenomic sequencing of six selected hot springs with different temperature and pH conditions. We sequenced a total of 11 samples from six hot springs and constructed 162 metagenome-assembled genomes (MAGs) with completeness above 70% and contamination lower than 10%. Crenarchaeota, Euryarchaeota and Aquificae were found to be the dominant phyla. Functional annotation revealed that bacteria encode versatile carbohydrate-active enzymes (CAZYmes) for the degradation of complex polysaccharides, while archaea tend to assimilate C1 compounds through carbon fixation. Under nitrogen-deficient conditions, there were correspondingly fewer genes involved in nitrogen metabolism, while abundant and diverse set of genes participating in sulfur metabolism, particularly those associated with sulfide oxidation and thiosulfate disproportionation. In summary, archaea and bacteria residing in the hot springs display distinct carbon metabolism fate, while sharing the common energy preference through sulfur metabolism. Overall, this research contributes to a better comprehension of biogeochemistry of terrestrial hot springs.

Improved forage quality and biomass yield of alfalfa (Medicago sativa L.) by Arabidopsis QQS orphan gene

Improving the forage quality of alfalfa in terms of digestibility and crude content is essential for any alfalfa quality breeding programs. Arabidopsis thaliana orphan gene QQS (Qua-Quine Starch) has been shown to improve protein content and alter carbohydrate composition in different food crops. However, there are significant differences in agronomic traits and nutritional conditions between alfalfa and other food crops. To explore the biological function and molecular mechanisms of QQS in alfalfa, we generated QQS transgenic plants and their segregated population (T1 generation), and evaluated their performance under normal- and nitrogen-deficient conditions. Our findings indicate that QQS can significantly enhance the total nitrogen and crude protein content of alfalfa and increase nodule weight under low-nitrogen conditions. Furthermore, QQS transgenic lines also showed reduced levels of neutral detergent fiber (NDF) and lignin, improving forage digestibility. By RNA sequencing and RT-qPCR analysis, we found that QQS affected the expression of genes involved in carbon and nitrogen metabolism, lignin biosynthesis and amino acid biosynthesis and degradation pathways in alfalfa. In addition, QQS also improved alfalfa biomass yield by increasing branch number and plant height in both greenhouse and field conditions. Our results demonstrate that QQS as a useful molecular tool can improve alfalfa biomass yield and overall forage quality and could have significant implications for the alfalfa breeding industry in satisfying the constant demands for high-quality and high-yielding forage.

Identification and expression characteristics of NLP (NIN-like protein) gene family in pepper (Capsicum annuum L.).

Pepper (Capsicum annum L.) is the main crop in the vegetable industry. The growth and development of peppers are regulated by nitrate, but there is limited research on the molecular mechanisms of nitrate absorption and assimilation in peppers. A plant specific transcription factor NLP plays an important role in nitrate signal transduction. In this study, a total of 7 NLP members were identified based on pepper genome data. Two nitrogen transport elements (GCN4) were found in the CaNLP5 promoter. In the phylogenetic tree, CaNLP members are divided into three branches, with pepper NLP and tomato NLP having the closest genetic relationship. The expression levels of CaNLP1, CaNLP3, and CaNLP4 are relatively high in the roots, stems, and leaves. The expression level of CaNLP7 gene is relatively high during the 5-7days of pepper fruit color transformation. After various non-Biotic stress and hormone treatments, the expression of CaNLP1 was at a high level. The expression of CaNLP3 and CaNLP4 was down regulated in leaves, but up regulated in roots. Under conditions of nitrogen deficiency and sufficient nitrate, the expression patterns of NLP genes in pepper leaves and roots were determined. These results provide important insights into the multiple functions of CaNLPs in regulating nitrate absorption and transport.

Insights into Candida Albicans: A New Perspective on Pathogenic Factors and Regulatory Mechanisms

Candida albicans (C. albicans) is a polymorphic fungus that exists as a natural flora in the skin and mucosal surfaces of the body. However, under certain conditions, such as immunodeficiency, mucosal damage, antibiotic use, and cancer, this fungus can cause superficial and systemic infections. C. albicans is the most common opportunistic pathogenic fungus in humans and causes 60% of mucosal infections and 40% of candidemia cases. Several pathogenic factors have been identified that contribute to the pathogenic potential of this fungus. Among these factors, we can mention: hypha production, attachment, and biofilm formation, secretion of hydrolase enzymes, acquisition of micronutrients, adaptation to oxygen and nitrogen deficiency conditions, and growth at temperatures above 37 °C. This review article will investigate the pathogenic factors of C. albicans and their regulatory factors. For this purpose, articles published in national and international scientific databases, including PubMed/MEDLINE, Google Scholar, Elsevier databases, IranMedex, Scopus, SID, and Science Direct, were used. Keywords such as: “Candida,” “Fungi,” “Pathogenesis,” and “Virulence” were used to find the articles.

Comparative physiological, metabolomic, and transcriptomic analyses reveal mechanisms of apple dwarfing rootstock root morphogenesis under nitrogen and/or phosphorus deficient conditions.

Nitrogen (N) and phosphorus (P) are essential phytomacronutrients, and deficiencies in these two elements limit growth and yield in apple (Malus domestica Borkh.). The rootstock plays a key role in the nutrient uptake and environmental adaptation of apple. The objective of this study was to investigate the effects of N and/or P deficiency on hydroponically-grown dwarfing rootstock 'M9-T337' seedlings, particularly the roots, by performing an integrated physiological, transcriptomics-, and metabolomics-based analyses. Compared to N and P sufficiency, N and/or P deficiency inhibited aboveground growth, increased the partitioning of total N and total P in roots, enhanced the total number of tips, length, volume, and surface area of roots, and improved the root-to-shoot ratio. P and/or N deficiency inhibited influx into roots, and H+ pumps played a important role in the response to P and/or N deficiency. Conjoint analysis of differentially expressed genes and differentially accumulated metabolites in roots revealed that N and/or P deficiency altered the biosynthesis of cell wall components such as cellulose, hemicellulose, lignin, and pectin. The expression of MdEXPA4 and MdEXLB1, two cell wall expansin genes, were shown to be induced by N and/or P deficiency. Overexpression of MdEXPA4 enhanced root development and improved tolerance to N and/or P deficiency in transgenic Arabidopsis thaliana plants. In addition, overexpression of MdEXLB1 in transgenic Solanum lycopersicum seedlings increased the root surface area and promoted acquisition of N and P, thereby facilitating plant growth and adaptation to N and/or P deficiency. Collectively, these results provided a reference for improving root architecture in dwarfing rootstock and furthering our understanding of integration between N and P signaling pathways.

Identification and Expression Analysis of the Isopentenyl Transferase (IPT) Gene Family under Lack of Nitrogen Stress in Oilseed (Brassica napus L.).

BnIPT gene family members in Brassica napus and analyzing their expression under different exogenous hormones and abiotic stress treatments to provide a theoretical basis for clarifying their functions and molecular genetic mechanisms in nitrogen deficiency stress tolerance of B. napus. Using the Arabidopsis IPT protein as the seed sequence, combined with the IPT protein domain PF01715, 26 members of the BnIPT gene family were identified from the whole genome of the rape variety ZS11. Additionally, the physicochemical properties and structures, phylogenetic relationships, synteny relationships, protein-protein interaction network, and gene ontology enrichment were analyzed. Based on transcriptome data, the expression patterns of the BnIPT gene under different exogenous hormone and abiotic stress treatments were analyzed. We used the qPCR method to identify the relative expression level of BnIPT genes that may be related to the stress resistance of rapeseed in transcriptome analysis under normal nitrogen (N: 6 mmol·L-1) and nitrogen deficiency (N: 0) conditions and analyzed its effect on rapeseed under nitrogen deficiency stress role in tolerance. In response to nitrogen deficiency signals, the BnIPT gene showed a trend of up-regulation in shoots and down-regulation in roots, indicating that it may affect the process of nitrogen transport and redistribution to enhance the stress resistance of rapeseed to respond to the nitrogen deficiency stress. This study provides a theoretical basis for clarifying the function and molecular genetic mechanism of the BnIPT gene family in nitrogen deficiency stress tolerance in rape.

Ethanol as a carotenoid production stimulator in Dunaliella salina CCAP 19/18.

Dunaliella salina is a rich source of carotenoids. Carotenoid production is induced under specific conditions, i.e., high light intensity, high salt concentration, nutrient limitation, and suboptimal temperatures in this microalga. The control of environmental factors is vital for high productivity of carotenoids. In this paper, the effect of different ethanol concentrations in combination with nitrogen deficiency was investigated to induce carotenoid production in D. salina CCAP 19/18. Also, some biochemical and molecular parameters were investigated in response to ethanol in the cells. It was shown that ethanol at 0.5% concentration increased cell number but, at 5% concentration, reduced cell viability compared to the control. The highest carotenoid production was achieved at 3% ethanol concentration, which was 1.46 fold higher than the nitrogen deficiency condition. Investigation of the 3 carotenoid biosynthesis genes revealed that their expression levels increased at 3% ethanol concentration, and the phytoene synthase gene was the most upregulated one. Lipid peroxidation increased at both 3% and 5% ethanol concentrations. At 3% concentration, the activity of catalase and superoxide dismutase increased, but no significant changes were seen at 5% ethanol concentration. Peroxidase activity reduced at both 3% and 5% concentrations. Moreover, proline and reducing sugar content increased at 3% concentration while decreased at 5% ethanol concertation. The results showed that at 3% ethanol concentration, higher carotenoid productivity was associated with an increase in other intracellular responses (molecular and biochemical). Ethanol as a controllable element may be beneficial to increase carotenoid production even under inappropriate environmental conditions in D. salina.

Biotechnological product potential of Auxenochlorella protothecoides including biologically active compounds (BACs) under nitrogen stress conditions.

Nitrogen stress can influence microalgae's growth characteristics, and microalgae grown in nitrogen-deficient conditions may produce higher or lower levels of biotechnological products as a result of metabolic changes. In photoautotrophic and heterotrophic cultures, nitrogen limitation has been proven effective in promoting lipid accumulation. In spite of this, no study has demonstrated a significant correlation between lipid content and other biotechnological products such as bioactive compounds (BACs).This research examines a strategy for lipid accumulation as well as the potential production of BACs with antibacterial properties in parallel with that strategy. This concept involved the treatment of the microalga Auxenochlorella protothecoides with low and high concentrations of ammonium (NH4+). This particular experiment reached a maximum lipid content of 59.5% using a 0.8 mM NH4+ concentration, resulting in the yellowing of the chlorophyll levels. Agar diffusion assays were conducted to determine the antibacterial activity of different extracts derived from the biomass when stressed with different levels of nitrogen.Algal extracts prepared by a variety of solvents showed different levels of antibacterial activity against representative strains of both gram-negative (Escherichia coli) and gram-positive (Staphylococcus aureus) bacteria. Among the extracts tested, 500mg/L ethyl acetate extract had the greatest antibacterial activity against Escherichia coli. In order to identify the components responsible for the extract's antibacterial activity, fatty acid methyl ester (FAME) analysis was performed. It has been suggested that the lipid fraction may be a valuable indicator of these activities since some lipid components are known to possess antimicrobial properties. In this regard, it was found that the amount of polyunsaturated fatty acid (PUFA) significantly decreased by 53.4% under the conditions with the highest antibacterial activity observed.

Interacting Effects of CO2, Temperature, and Nitrogen Supply on Photosynthetic, Root Growth, and Nitrogen Allocation of Strawberry at the Fruiting Stage

To efficiently improve the productivity of strawberries under growing environmental change, the photosynthesis, root growth, and nitrogen allocation of strawberries (Fragaria × ananassa Duch. cv. Toyonoka) were investigated in a factorial design of CO2, temperature, and nitrogen supply. Elevated CO2 decreased the maximum CO2 assimilation rate (Amax), maximum CO2 carboxylation capacity per unit leaf area (Vcmax), and maximum CO2 carboxylation capacity per unit leaf mass (Vcm-m) by 20%, 24%, and 44%, respectively. Meanwhile, it reduced the SPAD value, maximal fluorescence level in the dark-adapted state (Fm), and maximal photochemical efficiency of PSII (Fv/Fm). Moreover, root branches, root number, root dry weight, and nitrogen-use efficiency were further increased in response to elevated CO2 under low nitrogen. When elevated CO2 was applied together with nitrogen nutrients, the Vcm-m and root nitrogen concentration (RNC) declined by 32% and 12%, respectively, but the total root dry weight (TRDW) increased by 88%. If the nitrogen nutrient was individually applied, the TRDW decreased by 16%, while the RNC increased by 21%. When the high temperature was individually applied, the TRDW increased by 104%, but the RNC decreased by 5%. Overall, elevated CO2 exacerbated photosynthetic down-regulation and significantly affected nitrogen redistribution among strawberry organs, reducing leaf nitrogen concentration and accelerating leaf senescence. However, it could increase seed quantity and improve its quality as well. In other words, under nitrogen-deficient conditions, elevated CO2 could improve the survival of offspring via the cost of the mother plant’s growth capacity.
