Protective Effects of Annona Atemoya Extracts on Inflammation, Oxidative Stress, and Renal Function in Cadmium-Induced Nephrotoxicity in Wistar Rats

Background: Cadmium (Cd), a highly toxic heavy metal from agricultural activities, and its exposure can lead to impaired renal function by increasing reactive oxygen species. The atemoya fruit is known for its high phenolic and antioxidant compounds. This study aimed to evaluate the effects of atemoya extracts on renal function, oxidative stress parameters, and inflammatory biomarkers in a cadmium-induced nephrotoxicity model. Methods: Three aqueous extracts were prepared from different parts of the atemoya fruit: seeds, peel, and pulp. Twenty-five male Wistar rats were allocated into four groups: control, seed, peel, and pulp extracts at 2 g/kg for 25 days. All treatment groups administered intraperitoneal injections of cadmium chloride (CdCl2) (2 mg/kg) to induce renal damage. Results: The cadmium-treated groups showed decreased creatinine clearance, SOD, CAT, and GPx activities (p < 0.05) and increased serum levels of TNF-α and IL-6 compared to the control group (p < 0.05). The treatment with seed, peel, and pulp extracts increased creatinine clearance (p < 0.05), increased SOD, CAT, and GPx activities (p < 0.05), and reduced serum levels of TNF-α and IL-6 compared to the Cd group (p < 0.05). Conclusions: This study supports the use of atemoya as a promising candidate for mitigating nephrotoxicity and highlights the importance of its antioxidant and anti-inflammatory properties in renal health.

Oral pre-treatment with Citronellol ameliorates Methotrexate-induced nephrotoxicity in Wistar rats via targeting oxidative stress and inflammation

Background Methotrexate (MTX) is a classical folic acid antagonist widely used in the treatment of malignant and non-malignant disorders. However, its clinical application is often restricted by concomitant adverse effects, including renal damage. Numerous studies have highlighted the role of oxidative stress and inflammation in mediating MTX-related nephrotoxicity. Therefore, the current study aimed to explore the possible renoprotective action of Citronellol (CT), a natural compound with prominent antioxidant and anti-inflammatory activities, against nephrotoxicity induced by MTX. Methods To fulfill our objective, 24 adult male Wistar rats were randomly allocated into four groups: control, MTX, 100 mg/kg CT plus MTX and 200 mg/kg CT plus MTX. At the end of the study, the experimental rats were anesthetized, blood samples were collected for biochemical assays, and the kidneys were surgically removed for biochemical and gene expression analyses, after which all rats were sacrificed by exsanguination. Results Compared to the MTX-treated group, our results revealed that pre-supplementation with 100 or 200 mg/kg CT remarkably ameliorated renal damage biomarkers, including serum urea, serum creatinine, and kidney injury molecule-1 (KIM-1). In addition, pre-treatment with 100 or 200 mg/kg CT enhanced the activity of the antioxidant enzymes superoxide dismutase (SOD) and glutathione peroxidase (GPx), diminished renal malondialdehyde (MDA) contents, and attenuated inflammation by suppressing renal nuclear factor kappa B (NF-κB) signaling and diminishing tumor necrosis factor-alpha (TNF-α) gene expression. Moreover, pre-treatment with 200 mg/kg CT markedly reduced interleukin-1 beta (IL-1β) gene expression. Conclusion Our findings demonstrate, for the first time, that CT can serve as a new promising agent for mitigating nephrotoxicity induced by MTX through its antioxidant and anti-inflammatory properties.

Assessment of Sodium Butyrate’s Pharmacological Effects on Gentamicin-induced Nephrotic Disorders in a Rat Model

Background Sodium butyrate (SB) used to increase oxidative stress was reduced and the glutathione redox pathway was boosted. It inhibits proliferation, induction of differentiation, and induction or repression of gene expression and it has also anti-inflammatory action. This is endorsed for suppression of nephrotic symptoms. Objectives The present study was carried out to evaluate the nephroprotective activity of SB on gentamicin-induced nephrotoxicity in rats. Materials and Methods Present research work was designed to estimate pharmacological potential of SB in gentamicin-induced nephrotic disorder. The pharmacological prospective was estimated in gentamicin-induced rats through the oral administration of SB formulation 200 mg/kg and 300 mg/kg BW. Results The study observed that the SB significantly protects the kidneys from gentamicin-induced nephrotic disorders. Gentamicin 100 mg/kg for 7 days was used for induction of nephrotoxicity in Wistar rats by intraperitoneal administration. The administration of SB was observed to decrease the occurrence of glomerular congestion, vascular congestion, epithelial desquamation, inflammatory cell build-up, and kidney cell necrosis induced by gentamicin. The dose of SB maintained the gentamicin-induced increased creatinine (0.62±0.05), serum urea (18.10±0.79), uric acid (2.10±0.28), and blood urea nitrogen (BUN) (17.56±0.55) levels. This fact is further sustained by the histopathological examinations. Conclusion Sodium butyrate formulation with antinephrotic activity in maintaining uremic solutes such as urea, uric acid, creatinine, BUN, ketone, urobilinogen, glucose, specific gravity, and urine pH in Wistar rats with gentamicin-induced glomerulonephritis (GN). The SB was administered to the animals at doses of 200 and 300 mg/kg body weight. Pretreatment with the drug showed promising results in maintaining elevated parameters in nephritic rats within a short duration.

Effects of orange juice intake on Doxorubicin-induced hepatotoxicity and nephrotoxicity in Wistar rats

Considering that doxorubicin (DOXO) is an anticancer agent with severe adverse effects, this preclinical study investigated the protective effect of ‘pera’ orange juice (POJ) intake on DOXO-induced hepatic and renal toxicities. To this end, male Wistar rats were treated with DOXO in chronic (4.0 mg/kg weekly for four weeks) and acute (16.0 mg/kg in the fourth week) regimens. The experimental groups received filtered water or pasteurized POJ. After euthanasia, livers and kidneys were evaluated histologically and immunohistochemically for Ki-67 and histone γ-H2A.X markers, and their data were analyzed by One-Way ANOVA followed by Tukey’s a posteriori test. In this way, it was observed that DOXO caused weight loss, histological changes and increased damage markers. POJ reduced these changes only in chronic treatment, which allows us to conclude that the orange juice may attenuate the hepatotoxicity and nephrotoxicity of DOXO at low and fractionated doses.

Therapeutic Efficacy of Helianthemum lippii Extract and Silver Nanoparticles Synthesized from the Extract against Cadmium-Induced Renal Nephrotoxicity in Wistar Rats.

This study explored the therapeutic efficacy of Helianthemum lippii and silver nanoparticles (Ag NPs) synthesized using a H. lippii extract to alleviate cadmium-induced nephrotoxicity in Wistar rats. Sub-acute toxicity assessments of H. lippii (100 mg/kg, 1000 mg/kg, and 4000 mg/kg) and Ag NPs (2 mg/kg and 10 mg/kg) did not find any significant difference, compared with untreated control rats (n = 3 animals/group). Then, the adult Wistar rats were divided into one control (untreated/unexposed) and six experimental groups (n = 5/group): Ag NPs alone, H. lippii alone, exposure to 50 mg/kg CdCl2 in drinking water for 35 days, exposure to CdCl2 for 35 days followed by treatment with 0.1 mg/kg/day Ag NPs (intraperitoneal injection) and/or 100 mg/kg/day H. lippii by gavage for 15 days. In the CdCl2-exposed group, body weight decreased; urea, creatinine, and uric acid concentrations increased (p < 0.05 vs. control), indicative of nephrotoxicity, antioxidant defenses (SOD, GSH, and CAT) were reduced, and malondialdehyde concentration increased. Moreover, the kidney's architecture in CdCl2-exposed rats was altered: fibrosis, inflammatory cell infiltration, glomerular destruction, and tubular dilatation. Treatment with H. lippii and/or Ag NPs after CdCl2 exposure improved some of the renal function and architecture alterations induced by CdCl2, and also increased body weight. This study underscores the potential therapeutic applications of H. lippii and Ag NPs to decrease oxidative stress and promote xenobiotic detoxification, in line with the growing emphasis on environmentally conscious practices in scientific research and healthcare.

Nephroprotective activity of Bilwadi Agada in Cypermethrin Induced Nephrotoxicity in Wistar Rats

Introduction: One of the most potent and widely used formulations of Agadatantra is Bilwadi Agada mentioned in the context of management of envenomation from snake in Ayurveda. The same formulation is also indicated in multiple ailments like poisoning from other animals or insects and even is general conditions of fever. Nephroprotective activity has been demonstrated by Bilwadi Agada and the same was evaluated in this experiment in Wistar rats induced by cypermethrin Aim: To evaluate the nephroprotective activity of Bilwadi Agada in toxicity induced by cypermethrin in wistar albino rats. Materials and Methods: According to OECD guidelines 407, the induction of sub‑acute toxicity was done and effect of Bilwadi Agada was evaluated in wistar albino rats. Results: Degenerative changes in kidneys were found to be increased in lower dose level but moderate reversal was observed in higher dose. Conclusion: The thorough analysis of the results shows that the test drug at double the therapeutic dose causes moderate reversal of toxicant induced changes, thereby providing evidence for presence of nephroprotective activity against cypermethrin toxicity. Thus, therapeutic effects of Bilwadi Agada along with suitable cytoprotectant will be beneficial in the management of Cypermethrin toxicity.

Evaluation of Herbal Composite for Activity Against Anti-cancer Drug-induced Nephrotoxicity in Wistar Rats

Background: Nephrotoxicity is among the significant side effects and a dose-restriction mechanism for cisplatin treatment. According to recent studies, Oxidative stress and inflammation are the causative agents for acute renal failure on Cisplatin. Curcumin and resveratrol are considered to have potent anti-inflammatory and antioxidant effects. The current investigation aimed to determine whether a composite of curcumin and resveratrol attenuates nephrotoxicity caused by Cisplatin in Wistar rats. Method: The herbal compound was administered together with 5 mg/kg of cisplatin daily for 10 days with doses of 100 and 400 mg/kg. To evaluate renal damage, serum creatinine, nitrogen in blood urea, Creatinine, and levels of nitrite were measured. The levels of renal malondialdehyde, decreased glutathione levels, catalase, and also the superoxide dismutase enzyme were measured to quantify renal oxidative stress. Result: Significant oxidative stress, severe inflammation, and nephrotoxicity were all seen after a single dosage of cisplatin. In Wistar rats, a herbal composite treatment significantly reduced inflammation, improved renal function in a dose-dependent manner, reduced oxidative stress, and provided protection from the nephrotoxicity caused by Cisplatin. Conclusion: The herbal composite of curcumin - and - resveratrol was found to act as a therapeutic adjutant against Cisplatin nephrotoxicity.

Phytochemical profiling and nephroprotective potential of ethanolic leaf extract of Polyalthia longifolia against cisplatin-induced oxidative stress in rat model

Ethnopharmacological relevanceKidney problems are becoming more common globally and are considered a major health issue in the modern world with high mortality rate. Polyalthia longifolia (Sonn.) Thwaites is a tropical ethnomedicinal plant used to treat various diseases like diabetes, hypertension and urinary disorders and possess antioxidant and anti-inflammatory properties. Aim of the studyThis study aimed to investigate the phytochemical composition of 70% ethanolic leaf extract of Polyalthia longifolia (Sonn.) Thwaites (PL) and evaluates its nephroprotective effects against cisplatin-induced nephrotoxicity in Wistar rats. Materials and methodsThe leaves of PL were extracted with 70% ethanol and performed the phytochemical profiling using Liquid Chromatography–Mass Spectrometry (LC-MS). The nephroprotective effect of PL leaf extract was evaluated at three doses (150, 300 and 600 mg/kg, p.o.) for 14 days against cisplatin toxicity (16 mg/kg, i.p., once) in male Wistar rats. Body and kidney weight indices, kidney function markers and lipid profile markers in serum, and oxidative stress markers in kidney tissue were performed along with the histopathological analysis of kidney. ResultsThe LC-MS chromatograph confirmed the presence of various phytocompounds include N-Methylhernagine (aporphine alkaloid), 4-Acetamidobutanoic acid (gamma amino acid) and choline, etc. in the PL leaf extract. Exposure of cisplatin (16 mg/kg, i.p., once only) to the animals significantly elevated the levels of kidney functional markers (i.e. serum urea, uric acid, creatinine) and the lipid markers (triglyceride and total cholesterol) in blood circulation with depletion of serum albumin which were reversed by the therapy of PL leaf extract (150, 300 and 600 mg/kg) in dose-dependent manner. The altered level of body and kidney weight in cisplatin treated group was also restored by the therapy. PL leaf extract effectively improved the antioxidant defense system of kidney at all doses by restoring the levels of tissue glutathione, superoxide dismutase, catalase, glutathione peroxidase and glutathione reductase with the dose-dependent reduction of lipid peroxidation against cisplatin-induced renal oxidative stress. The histopathological observations also showed the significant recovery in cellular morphology after PL treatment when compared to the cisplatin toxicity group. The highest dose 600 mg/kg of PL leaf extract showed more pronounced renal recovery (p < 0.001) followed by other two doses, which was similar to the silymarin treatment group (a reference drug) against nephrotoxicity. ConclusionThe results of this study revealed the nephroprotective effects of PL leaves against cisplatin-induced nephrotoxicity by reversing the level of biochemical markers and mitigating oxidative stress as well as improving the architecture of renal tissues. This renal protection by PL might be due to the synergistic effect of its phytoconstituents and antioxidant efficacy.

Renoprotective effects of Cucumeropsis mannii seed oil on cisplatin-induced nephrotoxicity in Wistar rats

BackgroundThe growing prevalence of cancer and the concomitant rise in chemotherapy use have led to an increased incidence of kidney-related diseases, including nephrotoxicity. Cisplatin (CP) is a widely used and potent anticancer drug, but nephrotoxicity limits its clinical application. PurposeOur study aimed to determine the phytochemicals and median lethal dose of Cucumeropsis mannii seed oil (CMSO) using standard methods and to further investigate the effects of CMSO on CP-induced nephrotoxicity in male Wistar rats. MethodsTwenty-one rats (100 to 150 g) were randomly divided into seven groups (n = 3) and treated with CMSO or normal saline for ten days. Group A received 1.0 mL of normal saline, irrespective of the body weight (b.w.). Groups B-D received 2500, 5000, and 7500 mg/kg b.w., respectively, of the CMSO and a single intraperitoneal dose of CP (8 mg/kg) on the seventh day. Groups E and F were administered 2500 mg/kg and 7500 mg/kg b.w., respectively, of the CMSO without CP administration. Group G received a single intraperitoneal dose (8 mg/kg b.w.) of CP on the seventh day without CMSO treatment. ResultsThe analysis of CMSO revealed the presence of various phytochemicals such as hydrogen cyanide, glycosides, saponins, steroids, tannins, alkaloids, terpenoids, phenols, and flavonoids. Acute toxicity testing demonstrated the safety of CMSO up to 5000 mg/kg b.w. We discovered that the CP administration increased serum creatinine (sCr), urea, blood urea nitrogen (BUN), and malondialdehyde (MDA) levels in rats and markedly decreased renal superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) activities and glutathione (GSH) levels. CMSO attenuated kidney dysfunction, oxidative stress, and lipid peroxidation. Interestingly, CMSO prominently decreased sCr, urea, and BUN levels, boosted the activity of SOD, CAT, and GPx, increased GSH levels, and significantly (p < 0.05) decreased MDA levels. Histological assessment corroborated these biochemical findings. ConclusionOur findings highlight the potential of CMSO as a protective agent against CP-induced nephrotoxicity. The observed effects are attributable to the rich phenolic and flavonoid content of CMSO. These findings have significant implications for developing complementary therapies to mitigate chemotherapy-associated kidney damage, potentially enhancing the safety and efficacy of cisplatin-based cancer treatments. Further investigation is needed to explore the clinical applications of CMSO for cancer patients.

The potential role of upregulated PARP-1/RIPK1 expressions in amikacin-induced oxidative damage and nephrotoxicity in Wistar rats.

This study aimed to investigate the gene expression levels associated with nephrotoxic action of amikacin, as well as the post-treatment effect of diuretics on its nephrotoxic effects. Sixty male rats were divided equally into six groups, including the control group receiving saline intra-peritoneally (ip), and the five treated groups including therapeutic and double therapeutic dose groups, injected ip (15 and 30mg/kg b.wt./day) respectively for seven days, and another two rat groups treated as therapeutic and double therapeutic dose groups then administered the diuretic orally for seven days and the last group received amikacin ip at a rate of 15mg/kg/day for seven days, then given free access to water without diuretics for another seven days and was kept as a self-recovery group. Amikacin caused kidney injury, which was exacerbated by the double therapeutic dose, as evidenced by abnormal serum renal injury biomarkers, elevated renal MDA levels, inhibition of renal catalase and SOD enzyme activities, with renal degenerative and necrotic changes. Moreover, comet assays also revealed renal DNA damage. Interestingly, amikacin administration markedly elevated expression levels of the PARP-1, RIP1, TNF-α, IL-1β, and iNOS genes as compared to the control group. However, compared to the self-recovery group, post-amikacin diuretic treatment modulates amikacin-induced altered findings and alleviates amikacin nephrotoxic effects more efficiently. Our findings suggested the potential role of PARP-1 and RIPK1 expressions that influence the expression of proinflammatory cytokines such as IL-1β and TNF-α by exaggerating oxidative stress which may contribute to the pathogenesis of amikacin-induced nephrotoxicity.

Nephroprotective effect of Pterocarpus mildbraedii leaf extract in paracetamol-induced toxicity in Wistar rats: A histopathological investigation

Pterocarpus mildbraedii leaf is among the most commonly consumed leafy vegetables in west African sub-region. In South-East and South-South geopolitical zones of Nigeria, it is used for treatment of various health disorder such as headaches, pains, fever, convulsions, and respiratory disorders. This study was aimed at evaluating the possible effect of ethanol leaf extract of Pterocarpus mildbraedii on paracetamol-induced nephrotoxicity in Wistar rats. Twenty (20) Wistar rats weighing between 190 – 240 g were used for this study and divided into five (5) groups, each group having four (4) animals, described as follows; Group A was given distilled water only (positive control). Group B was given 700 mg/kg paracetamol with no treatment (negative control); Group C was given 700 mg/kg paracetamol and treated with 60 mg/kg of plant extract. Group D was given 700 mg/kg paracetamol and treated with 30 mg/kg of plant extract. Group E given 700 mg/kg of paracetamol and treated with 15 mg/kg of plant extract. At the end of the four (4) weeks of treatment, the kidneys were removed, weighed and sacrificed for histological examinations. Coronal sections of the kidney tissues were immersion fixed in 10 % PBS-buffered formalin and embedded in paraffin. The results obtained revealed a normal cellular architecture in the groups treated with the leaf extract. Conclusively, the result suggests protective effects of ethanol extract of P. mildbraedii leaf on the paracetamol-induced toxicity on the kidney cells.

This study assessed the protective effect of glutamine (GTN) against rifampicin/isoniazid/pyrazinamide/ethambutol (RIPE)-induced nephrotoxicity in rats. Thirty adult Wistar rats (200±20 g) of both sexes were grouped into 6 of 5 rats/group. The rats were treated daily for 30 days as follows: Group 1 (Vehicle control [normal saline 0.2mL]), group 2 (GTN 200 mg/kg), group 3 (RIPE 150, 75, 400 and 275 mg/kg in vehicle), group 4 (GTN 50 mg/kg +RIPE), group 5 (GTN 100 mg/kg +RIPE) and group 6 (GTN 200 mg/kg +RIPE). After treatment, blood samples were obtained and assessed for serum renal biomarkers. Kidneys were harvested, weighed and assessed for oxidative stress markers and histology. RIPE significantly (p&lt;0.01) decreased body weight and significantly (p&lt;0.01) increased kidney weight when compared to the control. Serum urea, creatinine, uric acid levels and kidney malondialdehyde levels were significantly (p&lt;0.001) increased in RIPE-treated rats when compared to the control. Serum total protein, albumin, kidney glutathione, catalase, superoxide dismutase and glutathione peroxidase levels were significantly decreased (p&lt;0.001) in RIPE-treated rats when compared to the control. RIPE caused tubular necrosis and collapsed glomeruli in the kidneys of rats. However, body and liver weights were significantly restored in GTN 100 mg/kg +RIPE and GTN 200 mg/kg +RIPE-treated rats at p&lt;0.05 and p&lt;0.01, respectively when compared to RIPE. Serum and kidney oxidative stress markers were restored in GTN 50 mg/kg +RIPE, GTN 100 mg/kg +RIPE and GTN 200 mg/kg +RIPE-treated rats at p&lt;0.05, p&lt;0.01 and p&lt;0.001 respectively, when compared to RIPE. GTN restored kidney histology. GTN protects against RIPE-induced nephrotoxicity in a dose-related fashion.

Multi-Mechanistic and Therapeutic Exploration of Nephroprotective Effect of Traditional Ayurvedic Polyherbal Formulation Using In Silico, In Vitro and In Vivo Approaches.

Based on traditional therapeutic claims, NEERI KFT (a traditional Ayurvedic polyherbal preparation) has been innovatively developed in recent time on the decades of experience for treating kidney dysfunction. Due to the lack of scientific evidence, the present investigations are needed to support the rationale use of NEERI KFT. Considering the facts, the study investigated the nephroprotective effect of NEERI KFT against kidney dysfunction using in silico, in vitro and in vivo approaches. In this study, phytochemical and network pharmacology studies were performed for the developed formulation to evaluate the molecular mechanism of NEERI KFT in the amelioration of kidney disease. In vitro nephroprotective and antioxidant effect of NEERI KFT was determined on HEK 293 cells against cisplatin-induced cytotoxicity and oxidative stress. In vivo nephroprotective effect of NEERI KFT was determined against cisplatin-induced nephrotoxicity in Wistar rats, via assessing biochemical markers, antioxidant enzymes and inflammatory cytokines such as TNF-α, IL-1β, CASP-3, etc. The results showed that the compounds such as gallic acid, caffeic acid and ferulic acid are the major constituents of NEERI KFT, while network pharmacology analysis indicated a strong interaction between polyphenols and several genes (CASPs, ILs, AGTR1, AKT, ACE2, SOD1, etc.) involved in the pathophysiology of kidney disease. In vivo studies showed a significant (p &lt; 0.05) ameliorative effect on biochemical markers and antioxidant enzymes (SOD, CAT, GSH, etc.), and regulates inflammatory cytokine (TNF-α, IL-1β, CASP-3) expression in kidney tissue. Hence, it can be concluded that NEERI KFT subsequently alleviates renal dysfunction mediated by cisplatin via attenuating oxidative and inflammatory stress, thus preserving the normalcy of kidney function.

The characterization techniques that achieved to confirm the synthesis and the structure details of the SeNPs were: UV–Vis spectroscopy, FT-IR analysis, XRD, EDX and SEM analysis. The biological activity of the synthesized SeNPs were tested and compared to the crude extract of Adiantum capillus on gentamicin model of nephrotoxicity in Wistar rats. Sera were used to test the pro-inflammatory cytokines Tumor necrosis factor alpha (TNF-α) and Interleukin beta (IL-β) levels. Histopathology and immunohistochemistry analysis for the apoptosis regulator protein (Bcl-2) and the interstitial filament protein (Vimentin) were performed. Results revealed that the synthesized SeNPs peak appeared at 400–430 nm wave length with crystallite particle size is around 37 nm. The predominant shape is spherical and cubic at different magnification levels with a narrow size distribution of 22.04–128.43 nm. The synthesized SeNPs showed a strong protective effect against gentamicin induced toxic effects to the rat’s kidneys obtained from the (kidney function parameters, histopathology evaluation, recovery of the pro-inflammatory cytokines IL-β and TNF-α level with retrieval of Bcl-2 and vimentin protein levels proximate to the vehicle control groups). Due to the significant protective effect of SeNPs, it considered much better than the crude extract of Adiantum capillus in the treatment of kidney injury; however, additional studies are necessary to find the precise mechanism of their action.
