Association between serum concentrations of perfluoroalkyl substances and global DNA methylation levels in peripheral blood leukocytes of Japanese women: A cross-sectional study

Global DNA methylation levels in peripheral blood leukocytes can be a biomarker for cancer risk; however, levels can be changed by various factors such as environmental pollutants. We investigated the association between serum concentrations of perfluoroalkyl substances (PFASs) and global DNA methylation levels of leukocytes in a cross-sectional study using the control group of a Japanese breast cancer case-control study [397 women with a mean age of 54.1 (SD 10.1) years]. Importantly, our analysis distinguished branched PFAS isomers as different from linear isomers. The serum concentrations of 20 PFASs were measured by in-port arylation gas-chromatography negative chemical ionization mass spectrometry. Global DNA methylation levels in peripheral blood leukocytes were measured using a luminometric methylation assay. Associations between log10-transformed serum PFAS concentrations and global DNA methylation levels were evaluated by regression coefficients in multivariable robust linear regression analyses. Serum concentrations of 13 PFASs were significantly associated with increased global DNA methylation levels in leukocytes. Global DNA methylation was significantly increased by 1.45 %–3.96 % per log10-unit increase of serum PFAS concentration. Our results indicate that exposure to PFASs may increase global DNA methylation levels in peripheral blood leukocytes of Japanese women.

Development and validation of a method for the analysis of halophenols and haloanisoles in cork bark macerates by stir bar sorptive extraction heart-cutting two-dimensional gas chromatography negative chemical ionization mass spectrometry

A method has been developed for the quantitative determination in cork bark macerates of 7 halophenols and 5 haloanisoles with demonstrated or suspected contribution to the cork taint off-flavour. Macerates were extracted with stirbar (20 mm polydimethylsiloxane-coated) sorptive extraction under optimized conditions (pH 3.5, 20% NaCl and 60 min). The bars were analysed by automated thermal desorption, heart-cutting two-dimensional gas chromatography and negative chemical ionization-mass spectrometry. Matrix effects were compensated for by a “matrix matched” calibration curve. Limits of detection were in the range 0.03-0.24 ng L−1, below the corresponding odor thresholds. Linearity (0.983 ≤ R2 ≤ 0.998), intra- and inter-day precision (5.4-14.3%) and accuracy (89–126%) were satisfactory. The analysis of 48 natural cork bark samples affected/not-affected bya cryptogamic disease (yellow spot) revealed compositional differences in 2,4,6-trichloroanisole (2,4,6-TCA), 2,4,6-trichlorophenol (2,4,6-TCP) and also in 2,6-dibromophenol, 2,3,4- and 2,4,5-TCP, 2,3,4-TCA and 2,3,4,6-tetrachlorophenol (2,3,4,6-TeCP). In addition, the contents of 2,4,6-TCP and 2,4,6-TCA, and 2,3,4,6-TeCP and some TCPs presented strong linear correlations.

Compound-Specific Chlorine Isotope Analysis of Organochlorine Pesticides by Gas Chromatography-Negative Chemical Ionization Mass Spectrometry.

Compound-specific stable chlorine isotope analysis (CSIA-Cl) is an important method for identifying sources of organochlorine contaminants and helping assess their quantification of transformation processes. However, the present CSIA-Cl is challenged by either redundant conversion pretreatment or complicated mathematical correction. To overcome the mentioned problems, a novel method has been developed for the CSIA-Cl of eight organochlorine pesticides using gas chromatography-negative chemical ionization mass spectrometry (GC-NCI-qMS) in this study. The instrument parameters, acquisition mode, and required injection amounts were optimized in terms of the precision of GC-NCI-qMS. An ionization energy of 90 eV and emission current of 90 μA were selected, and the precisions for eight organochlorine pesticides were in the range of 0.37‰–2.15‰ in single ion monitoring (SIM) mode when the injected amount was 0.50 mg L−1 (viz. 0.5 ng on column). Furthermore, when standards from Supelco and O2si were calibrated using standards from AccuStandard regarded as external isotope standard, chlorine isotope composition of α-hexachlorocyclohexane (α-HCH) and 2, 2-dichloro−1, 1-bis (4-chlorophenyl) ethylene (p, p′-DDE) in Supelco and O2si was confidently differentiated. The provenance identification method was validated by three organochlorine contaminated groundwater samples and showed a prospect in identifying the source of organochlorine pesticides.

Determination of Fipronil and Four Metabolites in Foodstuffs of Animal Origin Using a Modified QuEChERS Method and GC–NCI–MS/MS

A sensitive, effective, and reliable method for the accurate determination of fipronil and four metabolites (fipronil carboxamide, fipronil sulfone, fipronil sulfide, and fipronil desulfinyl) in foodstuffs of animal origin (egg, milk, beef kidney, beef liver, chicken, and chicken liver) was developed by isotope dilution–gas chromatography–negative chemical ionization–tandem mass spectrometry (GC–NCI–MS/MS). Samples were purified by a modified QuEChERS method. Four isotopically labeled internal standards were added in the sample extraction process to compensate for the matrix effect. The average recoveries were 78.2–107.1% with RSD ≤ 8.5%. The limits of quantification (LOQs) were 0.2 μg kg−1 for fipronil carboxamide and fipronil sulfone and 0.1 μg kg−1 for fipronil, fipronil sulfide, and fipronil desulfinyl. At the same time, the fragmentation mechanism of the five target compounds was analyzed via mass spectral data to help identify the compounds.

Development of a quantitative method for determination of steroids in human plasma by gas chromatography–negative chemical ionization–tandem mass spectrometry

Sex steroids are involved in biological functions that encompass from the complete sexual development of individuals up to the deregulation of metabolic pathways leading to some pathologies. Steroids are present in blood at low concentration levels from pg mL−1 to ng mL−1. For this reason, a high sensitive and selective method based on gas chromatography–negative chemical ionization–tandem mass spectrometry (GC–NCI–MS/MS) is here proposed to quantify either androgens (androstenedione, dehydroepiandrosterone, dihydrotestosterone and testosterone), estrogens (estrone and estradiol) and a progestogen (progesterone) in human plasma. The sample preparation steps, protein precipitation and solid phase extraction, were optimized to ensure the sample matrix removal and to extract steroids with high efficiency. The NCI–MS/MS detection approach was compared with that based on electron impact to evaluate the incidence of the ionization source in the determination of steroids. The quantification limits for determination of these analytes were in a range from 10 pg mL−1 to 5 ng mL−1, with a high sensitivity for estrogens, typically found at low concentrations. The proposed method was tested for the determination of steroids in male blood samples, in which 6 out of 7 steroids were detected and quantified to report concentration values in agreement with those described in the literature.

Umbilical cord serum PBDE concentrations and child adiposity measures at 7 years

BackgroundPolybrominated diphenyl ethers (PBDEs) exist extensively in the environment. Toxicological studies suggested PBDEs may interfere with adipogenic pathways. However, few human evidence addressed PBDE exposures in utero related to childhood adiposity. ObjectiveWe assessed associations between PBDEs concentrations in cord serum and childhood adiposity measures at 7 years. MethodsAmong 318 mother-child pairs from Sheyang Mini Birth Cohort Study (SMBCS) in China, nine PBDE congener concentrations were quantified in umbilical cord serum using gas chromatography-negative chemical ionization mass spectrometry (GC-NCI-MS). Anthropometric indicators of children aged 7 years were measured, including weight, height and waist circumference. Age and sex-specific body mass index (BMI) z scores were calculated based on World Health Organization (WHO)'s child growth standards. Multivariate linear and logistic regression models adjusted for putative confounders were performed to examine associations between PBDE congeners and adiposity parameters. ResultsBDE-209 was the most abundant congener of PBDEs with a median value of 19.5 ng/g lipid. The geometric mean values of nine PBDE congeners ranged from below limit of detection (LOD) to 18.1 ng/g lipid, and the detection rates were 46.5%~96.5%. Cord serum BDE-153 and BDE-154 concentrations were associated with lower childhood BMI z score (regression coefficient, β=-0.15, 95% confidence interval: -0.29, −0.02; p=0.02; β=-0.23, 95%CI: -0.43, −0.03; p=0.03, respectively) and lower waist circumference (β=-0.75 cm, 95%CI: -1.43, −0.06; p=0.03; β=-1.22 cm, 95%CI: -2.23, −0.21; p=0.02, respectively), after controlling for potential confounders. Moreover, prenatal BDE-154 exposure was related to a decreased obesity risk of children aged 7 years (odds ratio, OR=0.46, 95%CI: 0.22, 0.94; p=0.03). These effects were only observed among boys in sex-straitified analyses. ConclusionsCord serum BDE-153 and BDE-154 concentrations were related to reduced adiposity measures at 7 years of age. Further evidence regarding the impacts of prenatal PBDE exposures on childhood development is warranted.

Derivatization gas chromatography negative chemical ionization mass spectrometry for the analysis of trace organic pollutants and their metabolites in human biological samples.

Gas chromatography negative chemical ionization mass spectrometry (GC-NCI-MS) is a preferred instrumental approach for the trace and ultra-trace analysis of various toxic organics and their metabolites in human biological fluids. Specifically, the method has played an important role in the highly sensitive and specific quantitative detection of persistent highly halogenated compounds in environmental matrices and biota during the past few decades. However, for the analysis of toxic metabolites with active hydrogen atoms, such as acids, alcohols, and phenolic compounds, from biological matrixes or organics without electronegative atoms or groups, a derivatization step is often needed prior to GC analysis. Such derivatization aims to change the properties of targets to improve their separation, increase their volatility, and enhance the sensitivity of instrumental detection. This review summarizes three derivatization strategies commonly used for GC methods, i.e., alkylation, silylation, and acylation, together with their application combined with GC-NCI-MS for the high sensitivity analysis of toxic organic metabolites in the human body. The advantages and disadvantages of each derivatization method and potential directions for future applications are discussed. Given the broad variety of applications as well as the compound-specific sensitivity for the ultra-trace analysis of target xenobiotics in human biological fluids, subsequent studies are required to develop convenient, faster derivatization procedures and reagents better suited for routine analysis. Analysis of Perfluorooctanesulfonate Isomers and Other Perfluorinated Alkyl Acids in Serum by In-Port Arylation Gas Chromatography Negative Chemical Ionization–Mass Spectrometry

Perfluorooctanesulfonate (PFOS) is a common environmental pollutant and has been detected in various environmental media. Several major isomers have been identified, and their profiles provide important signatures from the source of contamination. Gas chromatography has a significant resolving power for the isomers. However, methods for derivatizing sulfonates for a sensitive analysis are limited. In this study, in-port derivatization with diaryl iodonium salts into aryl sulfonates was used. A technical mixture of PFOS was separated on gas chromatography to afford 11 isomers. In electron-capture negative ionization, the PFOS derivatives form specific fragmented anions (sulfinate anions). The derivatization was also applied to other per- or polyfluoroalkyl acids (PFASs; 13 perfluoroalkyl carboxylic acids and 10 sulfonates). Analysis of human serum samples was achieved with as little as 50 μL of sample and indicated the presence of PFOS isomers and PFASs. The detection limits of PFOS isomers were 0.02–0.09 ng mL–¹ with sufficient recoveries (85%–98%), and those for other PFASs ranged from 0.01–1 ng mL–¹ (recoveries, 80%–100%), which are suitable for the analysis of human serum samples.

New fully automated gas chromatographic analysis of urinary S-phenylmercapturic acid in isotopic dilution using negative chemical ionization with isobutane as reagent gas.

The determination of urinary S-phenylmercapturic acid (S-PMA) represents the most reliable biomarker to monitor the intake risk of airborne benzene. Recently, the European Chemical Agency deliberated new occupational exposure limits for benzene and recommended an S-PMA biological limit value of 2-μg/g creatinine. This limit is an order of magnitude lower than the previous one, and its determination constitutes a challenge in the analytical field. We developed and validated a method that allows the fully automated and sensitive determination of S-PMA by the use of gas-chromatography negative chemical ionization tandem mass spectrometry in isotopic dilution. For negative chemical ionization, we selected a mixture of 1% isobutane in argon as reactive gas, by studying its chemical ionization mechanism and optimal parameters compared with pure isobutane or pure methane. This gas mixture produces a more abundant signal of the target analyte than isobutane or methane, and it extended the operative lifetime of the ion source, enabling us to start a high-throughput approach of the S-PMA analysis. Moreover, energy-resolved mass spectrometry experiments were carried out to refine the MS/MS analysis conditions, testing nitrogen and argon as collision gases. The method optimization was pursued by a chemometric model by using the experimental design. The quantification limit for S-PMA was 0.10 μg/L. Accuracy (between 98.3% and 99.6%) and precision (ranging from 1.6% to 6.4%) were also evaluated. In conclusion, the newly developed assay represents a powerful tool for the robust, reliable, and sensitive quantification of urinary S-PMA, and because of its automation, it is well suited for application in large environmental and biological monitoring.

Development of simple analytical methods of polychlorinated biphenyls in human serum by gas chromatography negative ion chemical ionization quadrupole mass spectrometry

In this study, we developed a highly sensitive, robust method for determining 12 congeners of two to ten chlorinated polychlorinated biphenyls (PCBs) in serum samples using gas chromatography (GC)–mass spectrometry (MS) operating in selected ion monitoring mode (SIM: m/z 35) with negative ion chemical ionization (NICI), and the results were compared with those from GC coupled with high-resolution MS (HRMS) with electron impact (EI). The recovery rates of the surrogate PCB congeners were 97.2%–112% (coefficient of variation: 5.3%–8.5%), and the method detection limits for PCBs in each matrix obtained by GC–NICI–quadrupole mass spectrometry (qMS) were 1.9–20 pg g−1 wet wt. The analytical values of the target compounds in the samples analyzed by GC–NICI–qMS and GC–EI–HRMS were comparable (Passing–Bablok regression: R = 0.888–0.967), and the analytical values obtained via GC–NICI–qMS were almost comparable with those of the certified serum samples from National Institute of Standards and Technology (NIST: SRM1957), indicating that GC–NICI–qMS is suitable for the analysis of tetra- to hepta-chlorinated PCBs in serum samples.

Simultaneous determination of cis-permethrin and trans-permethrin in rat plasma and brain tissue using gas chromatography-negative chemical ionization mass spectrometry.

A sensitive method for the simultaneous determination of cis-permethrin (cis-PERM) and trans-permethrin (trans-PERM) in small volumes (100μL) of rat plasma and brain homogenate was developed, using a liquid-liquid extraction for sample preparation and gas chromatography-negative chemical ionization mass spectrometry (GCNCI-MS) for detection. Quantitation of trace levels of the insecticide in small volumes of biological samples is essential to support toxicokinetic studies in small animals. There are currently no validated methods in the literature for determining cis-PERM and trans- PERM in volumes as low as 100μL of rat plasma or brain homogenate. The method provided a linear range of 0.2-150.0ng/mL for analytes in both matrices. The intra- and inter-batch precision (as% relative standard deviation, RSD) and accuracy (as relative error, RE) of the method were better than 20% at the limit of quantitation and better than 15% across the remaining linear range. The validated method was applied in a toxicokinetic study in adult rats with oral dosing of 10mg/kg (cis-PERM) and 100mg/kg (trans-PERM) in corn oil. cis-PERM and trans- PERM were monitored in rat plasma and brain tissue samples for 6h following dosing, and both analytes were detected in all plasma and brain samples.

Novel brominated flame retardants in food composites and human milk from the Chinese Total Diet Study in 2011: Concentrations and a dietary exposure assessment

On the basis of the fifth Chinese total diet study (TDS) performed in 2011, the dietary exposure of the Chinese population to novel brominated flame retardants (NBFRs) was assessed. Six NBFRs were determined in 80 composite samples from four animal origin food groups and 29 pooled human milk samples. Based on gas chromatography-negative chemical ionization mass spectrometry (GC-NCI/MS) analysis, the levels of the total NBFRs ranged from <LOD to 70.2ng/g lipid weight (lw) in food composites and from 2.48 to 23.9ng/g lw in human milk samples. Decabromodiphenyl ethane (DBDPE), with mean levels of 9.03ng/g lw in food composites and 8.06ng/g lw in human milk, was the most abundant compound in the total NBFRs. No obvious spatial distribution patterns in China were observed in food samples or human milk. The average estimated daily intake (EDI) of total NBFRs via food consumption for a “standard Chinese man” was 4.77ng/kg bodyweight (bw)/day, with a range of 0.681 to 18.9ng/kgbw/day. Meat and meat products were the main dietary source of NBFRs, although levels of NBFRs in aquatic food were found to be the highest among the four food groups. The average EDI of total NBFRs for nursing infants was 38.4ng/kgbw/day, with a range of 17.4 to 113ng/kgbw/day, which was approximately eight-fold higher than the EDI for adults, suggesting the heavy body burden of NBFRs on nursing infants. The levels and EDI of DBDPE in the present study were similar to or higher than those of legacy BFRs (i.e., PBDEs and HBCD) in the TDS 2007, indicating that DBDPE, as a main alternative to PBDEs, might have become the primary BFR used in China.

Determination of novel brominated flame retardants and polybrominated diphenyl ethers in serum using gas chromatography-mass spectrometry with two simplified sample preparation procedures.

Two simple and efficient pretreatment procedures have been developed for the simultaneous extraction and cleanup of six novel brominated flame retardants (NBFRs) and eight common polybrominated diphenyl ethers (PBDEs) in human serum. The first sample pretreatment procedure was a quick, easy, cheap, effective, rugged, and safe (QuEChERS)-based approach. An acetone/hexane mixture was employed to isolate the lipid and analytes from the serum with a combination of MgSO4 and NaCl, followed by a dispersive solid-phase extraction (d-SPE) step using C18 particles as a sorbent. The second sample pretreatment procedure was based on solid-phase extraction. The sample extraction and cleanup were conducted directly on an Oasis HLB SPE column using 5% aqueous isopropanol, concentrated sulfuric acid, and 10% aqueous methanol, followed by elution with dichloromethane. The NBFRs and PBDEs were then detected using gas chromatography-negative chemical ionization mass spectrometry (GC-NCIMS). The methods were assessed for repeatability, accuracy, selectivity, limits of detection (LODs), and linearity. The results of spike recovery experiments in fetal bovine serum showed that average recoveries ranged from 77.9% to 128.8% with relative standard deviations (RSDs) from 0.73% to 12.37% for most of the analytes. The LODs for the analytes in fetal bovine serum ranged from 0.3 to 50.8pg/mL except for decabromodiphenyl ethane. The proposed method was successfully applied to the determination of the 14 brominated flame retardants in human serum. The two pretreatment procedures described here are simple, accurate, and precise, and are suitable for the routine analysis of human serum. Multi-residue analysis of pesticides in traditional Chinese medicines using gas chromatography-negative chemical ionisation tandem mass spectrometry

In this study, a residue analysis method for the simultaneous determination of 107 pesticides in traditional Chinese medicines (TCMs), Angelica sinensis, A. dahurica, Leonurus heterophyllus Sweet, Pogostemon cablin and Lonicera japonica Thunb., was developed using gas chromatography coupled with tandem mass spectrometry in negative chemical ionisation mode (GC-NCI-MS/MS). NCI has advantages of high sensitivity and selectivity to chemicals with electron-withdrawing groups, and yields low background interference. For sample preparation, QuEChERS (quick, easy, cheap, effective, rugged and safe) was applied. Due to the unique characteristics of TCMs, the clean-up step was optimised by adjusting amounts of primary secondary amine, C18, graphitised carbon black and silica sorbents. Validation was mainly performed by determining analyte recoveries at four different spiking concentrations of 10, 50, 100 and 200 ng g−1, with seven replicates at each concentration. Method trueness, precision, linearity of calibration curves, lowest calibrated levels (LCLs) and matrix effects were determined to demonstrate method and instrument performance. Among the 107 pesticides tested, approximately 80% gave recoveries from 80% to 110% and < 10% relative standard deviation (RSD). The LCLs for nearly all pesticides were 5 ng g−1, and as low as 0.1 ng g−1 for dichlofenthion, endosulfan sulphate, flumetralin, isofenphos-methyl, methyl-pentachlorophenyl sulphide and trifluralin. The results indicate that GC-NCI-MS/MS is an excellent technique for quantitative and qualitative analysis of targeted GC-amenable pesticides at ultra-trace levels, especially in complex matrices such as TCMs.

Quantifying the degradation of TNT and RDX in a saline environment with and without UV-exposure

Terrorist attacks in a maritime setting, such as the bombing of the USS Cole in 2000, or the detection of underwater mines, require the development of proper protocols to collect and analyse explosive material from a marine environment. In addition to proper analysis of the explosive material, protocols must also consider the exposure of the material to potentially deleterious elements, such as UV light and salinity, time spent in the environment, and time between storage and analysis. To understand how traditional explosives would be affected by such conditions, saline solutions of explosives were exposed to natural and artificial sunlight. Degradation of the explosives over time was then quantified using negative chemical ionization gas chromatography mass spectrometry (GC/NCI-MS). Two explosives, trinitrotoluene (TNT) and cyclotrimethylenetrinitramine (RDX), were exposed to different aqueous environments and light exposures with salinities ranging from freshwater to twice the salinity of ocean water. Solutions were then aged for up to 6 months to simulate different conditions the explosives may be recovered from. Salinity was found to have a negligible impact on the degradation of both RDX and TNT. RDX was stable in solutions of all salinities while TNT solutions degraded regardless of salinity. Solutions of varying salinities were also exposed to UV light, where accelerated degradation was seen for both explosives. Potential degradation products of TNT were identified using electrospray ionization mass spectrometry (ESI-MS), and correspond to proposed degradation products discussed in previously published works [1].

Negative chemical ionization gas chromatography coupled to hybrid quadrupole time-of-flight mass spectrometry and automated accurate mass data processing for determination of pesticides in fruit and vegetables.

Gas chromatography coupled to high resolution hybrid quadrupole time-of-flight mass spectrometry (GC-QTOF MS), operating in negative chemical ionization (NCI) mode and combining full scan with MSMS experiments using accurate mass analysis, has been explored for the automated determination of pesticide residues in fruit and vegetables. Seventy compounds were included in this approach where 50% of them are not approved by the EU legislation. A global 76% of the analytes could be identified at 1μgkg(-1). Recovery studies were developed at three concentration levels (1, 5, and 10μgkg(-1)). Seventy-seven percent of the detected pesticides at the lowest level yielded recoveries within the 70%-120% range, whereas 94% could be quantified at 5μgkg(-1), and the 100% were determined at 10μgkg(-1). Good repeatability, expressed as relative standard deviation (RSD <20%), was obtained for all compounds. The main drawback of the method was the limited dynamic range that was observed for some analytes that can be overcome either diluting the sample or lowering the injection volume. A home-made database was developed and applied to an automatic accurate mass data processing. Measured mass accuracies of the generated ions were mainly less than 5ppm for at least one diagnostic ion. When only one ion was obtained in the single-stage NCI-MS, a representative product ion from MSMS experiments was used as identification criterion. A total of 30 real samples were analyzed and 67% of the samples were positive for 12 different pesticides in the range 1.0-1321.3μgkg(-1).

Simultaneous determination of trace polybrominated diphenyl ethers in serum using gas chromatography-negative chemical ionization mass spectrometry with simplified sample preparation

We have developed a simple and sensitive method which has good application value in the determination of trace PBDEs in serum.

Determination of 2-, 3-, 4-methylpentanoic and cyclohexanecarboxylic acids in wine: Development of a selective method based on solid phase extraction and gas chromatography-negative chemical ionization mass spectrometry and its application to different wines and alcoholic beverages

A method to analyse 2-methylpentanoic, 3-methylpentanoic and 4-methylpentanoic acids as well as cyclohexanecarboxylic acid has been developed and applied to wine and other alcoholic beverages. Selective isolation with solid phase extraction, derivatization with 2,3,4,5,6-pentafluorobenzyl bromide at room temperature for 30minutes, and further analysis by gas chromatography-mass spectrometry in negative chemical ionization mode provides detection limits between 0.4 and 2.4ng/L. Good linearity up to 3.6μg/L, satisfactory reproducibility (RSD<10%) and signal recovery of around 100% represent a robust method of analysis. Concentration data of these analytes in wine and other alcoholic beverages are reported for the first time. The levels found ranged from the method detection limits to 2630ng/L, 2040ng/L and 3810ng/L for 2-, 3- and 4-methylpentanoic acids, respectively, and to 1780ng/L for cyclohexanecarboxylic acid. There are significant differences depending on the type of wine or beverage. Distilled beverages, beer and aged wines have higher contents in methylpentanoic and cyclohexanecarboxylic acids.

Novel Approach to Stable Chlorine Isotope Analysis using Gas Chromatography-Negative Chemical Ionization Mass Spectrometry

A novel approach to compound-specific stable chlorine isotope analysis is presented to overcome many of the major drawbacks of existing methods. Contemporary methods often require either several laborious off-line procedures prior to isotope analysis or complicated mathematical data calculations and evaluation schemes to derive the chlorine isotope ratios from the mass spectra. In contrast to previous methods, our on-line setup was based on the use of gas chromatography-negative chemical ionization mass spectrometry. The ion traces of 35Cl−(m/z 35) and 37Cl−(m/z 37) were recorded to directly determine the isotopic ratios of chlorine. The chlorine fragments were obtained from each specific chlorinated compound using the appropriate energy and an optimized acquisition mode, and the method completely avoided isobaric interferences. The principle and the mechanism of this approach were presented and explained using trichloroethene. Optimization of the gas chromatography-negative chemical ionization mass spectrometry parameters and method validation were achieved using chlorinated alkenes, chlorinated benzenes, and organochlorine pesticides. The sensitivity of the method over previous methods was significantly improved, and the ultra-trace injection amounts of 0.4–473.5 pmol resulted in standard deviations (1σ) ranging from 0.55‰–2.15‰. The present study suggested that the gas chromatography-negative chemical ionization mass spectrometry method offered an innovative approach to compound-specific chlorine isotope analysis, and it could facilitate the application of compound-specific stable chlorine isotope analysis in environmental fields.

Levels of polybrominated diphenyl ethers in settled house dust from urban dwellings with resident preschool-aged children in Nanjing, China.

We investigated the levels and possible determinants of polybrominated diphenyl ethers (PBDEs) in the settled house-dust (SHD) of urban dwellings with resident preschool-aged children in Nanjing, China. The possible neurodevelopmental effects of house-dust PBDEs were also explored. SHD was collected from 216 urban houses. Levels of 8 PBDEs were measured by gas chromatography-negative chemical ionization mass spectrometry. The Child Behavior Checklist and the Gesell Development Inventory were used to evaluate the child's development. BDE47, BDE99, BDE153, BDE18, and BDE209 were detected in the SHD of >90 % of houses, of which BDE209 predominated. Most PBDEs were found at significantly greater levels in indoor than in outdoor dust (P < 0.05). Levels of BDE28 and BDE154 in houses with solid-wood floors were significantly greater than those in houses with plywood floors (P < 0.05). BDE154 levels in houses with wallpaper were significantly greater than those without wallpaper (P < 0.05). Greater BDE47 concentrations were found in houses with less natural ventilation time (linear trend P < 0.05). After dichotomization at the geometric mean concentration, BDE209 and total BDEs showed significant risks for depressed behavior problems and lower personal social developmental quotients (DQs); BDE99 and BDE153 indicated a risk for lower personal social DQs. In conclusion, PBDEs (especially BDE209) are ubiquitous in urban SHD in Nanjing residences. Natural ventilation and floor materials potentially influence PBDE levels in SHD. The potential adverse effect of postnatal exposure to PBDEs on the behavior and neurodevelopment of preschool-age children requires follow-up in larger studies.
