Substrate diversity affects carbon utilization rate and threshold concentration for uptake by natural bacterioplankton communities

Persistence of dissolved organic matter (DOM) in aquatic environments may in part be explained by high diversity and low concentrations of carbon substrates. However, changes in dissolved substrate quality can modify aquatic bacterial community composition and rate of carbon uptake. The aim of this study was to test if the presence of multiple simple substrates affects the turnover of organic carbon. Natural bacterial communities were grown in continuous cultures supplied with either individual carbon substrates—salicylic acid (SA), tryptophan (Trp) or tyrosine (Tyr)—or a combination of the 3 substrates. Concentrations were tracked using fluorescence spectroscopy, and steady-state concentrations of a few nanomolar were reached. Bacterial growth efficiency was dependent on which carbon sources were present and reached an intermediate level in the combined treatment. The bacterial community maintained steady-state concentrations of Trp that were lower in the combined treatment than in the individual substrate treatment. In addition, steady-state concentrations were reached faster during growth on combined carbon substrates, although the maximum utilization rate of each individual compound was lower. However, the steady-state concentration of total carbon (sum of carbon content of SA, Trp and Tyr) was higher in the combined culture than in the individual substrate treatments, and seemed to be determined by the carbon substate for which the bacteria had the lowest affinity. The results from this study indicate that persistence of dissolved organic carbon can in part be explained by vast substrate diversity, which raises the threshold concentration for utilization by natural bacterial communities.

Legacy and Emerging Pollutants in an Urban River Stretch and Effects on the Bacterioplankton Community

River contamination is due to a chemical mixture of point and diffuse pollution, which can compromise water quality. Polycyclic Aromatic Hydrocarbons (PAHs) and emerging compounds such as pharmaceuticals and antibiotics are frequently found in rivers flowing through big cities. This work evaluated the presence of fifteen priority PAHs, eight pharmaceuticals including the antibiotics ciprofloxacin (CIP) and sulfamethoxazole (SMX), together with their main antibiotic resistant genes (ARGs) and the structure of the natural bacterioplankton community, in an urbanized stretch of the river Danube. SMX and diclofenac were the most abundant chemicals found (up to 20 ng/L). ARGs were also found to be detected as ubiquitous contaminants. A principal component analysis of the overall microbiological and chemical data revealed which contaminants were correlated with the presence of certain bacterial groups. The highest concentrations of naphthalene were associated with Deltaproteobacteria and intI1 gene. Overall, the most contaminated site was inside the city and located immediately downstream of a wastewater treatment plant. However, both the sampling points before the river reached the city and in its southern suburban area were still affected by emerging and legacy contamination. The diffuse presence of antibiotics and ARGs causes particular concern because the river water is used for drinking purposes.

Insignificant Response of Bacterioplankton Community to Elevated pCO2 During a Short-Term Microcosm Experiment in a Subtropical Eutrophic Coastal Ecosystem.

Ocean acidification, as one of the major consequences of global climate change, markedly affects multiple ecosystem functions in disparate marine environments from coastal habitats to the deep ocean. Evaluation of the responses of marine microbial community to the increasing partial pressure of CO2 (pCO2) is crucial to explore the microbe-driven biogeochemical processes in the future ocean. In this study, a microcosm incubation of eutrophic coastal water from Xiamen Bay under elevated pCO2 (about 1,000 μatm) and control (ambient air, about 380–410 μatm) conditions was conducted to investigate the effect of ocean acidification on the natural bacterioplankton community. During the 5-day incubation period, the chlorophyll a concentration and bacterioplankton abundance were not significantly affected by increased pCO2. Hierarchical clustering and non-metric multidimensional scaling analysis based on Bray-Curtis similarity among the bacterioplankton community derived from the 16S rRNA genes revealed an inconspicuous impact of elevated pCO2 on the bacterial community. During the incubation period, Proteobacteria, Bacteroidetes, Actinobacteria, Cyanobacteria, and Epsilonbacteraeota were predominant in all microcosms. Despite the distinct temporal variation in the composition of the bacterioplankton community during the experimental period, statistical analyses showed that no significant difference was found on bacterioplankton taxa between elevated pCO2 and control, indicating that the bacterioplankton at the population-level were also insensitive to elevated pCO2. Our results therefore suggest that the bacterioplankton communities in the fluctuating and eutrophic coastal ecosystems appear to be adaptable to the short-term elevated pCO2.

Public good exploitation in natural bacterioplankton communities

Bacteria often interact with their environment through extracellular molecules that increase access to limiting resources. These secretions can act as public goods, creating incentives for exploiters to invade and "steal" public goods away from producers. This phenomenon has been studied extensively in vitro, but little is known about the occurrence and impact of public good exploiters in the environment. Here, we develop a genomic approach to systematically identify bacteria that can exploit public goods produced during the degradation of polysaccharides. Focusing on chitin, a highly abundant marine biopolymer, we show that public good exploiters are active in natural chitin degrading microbial communities, invading early during colonization, and potentially hindering degradation. In contrast to in vitro studies, we find that exploiters and degraders belong to distant lineages, facilitating their coexistence. Our approach opens novel avenues to use the wealth of genomic data available to infer ecological roles and interactions among microbes.

Ammonium release via dissolution and biological mineralization of food pellets used in salmon farming

We studied the input of ammonium coming from the dissolution of food pellets commonly used in salmon farming as well as its potential for mineralization via ammonification and ammonium oxidation by natural bacterioplankton communities. Through experiments in central and southern Chile, we evaluated the potential for photo‐ and bio‐ammonification of food pellets in surface and subsurface waters (30 m depth) using irradiated (PAR + UV; IPS) and non‐irradiated dissolved‐pellet solutions (NIPS). Ammonium release was observed in both IPS and NIPS treatments and occurred at short time scales (<2 hr). Experiments in southern Chile during late winter showed high rates of pellet ammonium release in surface waters (up to 3.8 µmol L⁻¹ min⁻¹) compared with deeper samples (30 m depth). In contrast, in spring higher rates were observed at 30 m depth and reached up to 45 µmol L⁻¹ min⁻¹ while in surface waters they accounted for 9 µmol L⁻¹ min⁻¹. Ammonium oxidizing bacteria and archaea showed seasonal variability, but archaea were detected in all experiments reaching high potential activity, based in RT‐qPCR of functional genes expression copy numbers, in IPS treatments during the winter season in Chiloe. Overall, our results indicate that food pellets can be a source of ammonium, which can exceed by two orders of magnitude the average natural concentrations commonly found in the euphotic layer in coastal environments. The ammonium enrichment from dissolved pellet solutions was rapidly consumed by microbial communities including nitrifying assemblages. This has implications for the evaluation of aquaculture nutrient input and its recycling in the water column in areas highly impacted by climate change and aquaculture pressure.

Coupling biogeochemical process rates and metagenomic blueprints of coastal bacterial assemblages in the context of environmental change

Bacteria are major drivers of biogeochemical nutrient cycles and energy fluxes in marine environments, yet how bacterial communities respond to environmental change is not well known. Metagenomes allow examination of genetic responses of the entire microbial community to environmental change. However, it is challenging to link metagenomes directly to biogeochemical process rates. Here, we investigate metagenomic responses in natural bacterioplankton communities to simulated environmental stressors in the Baltic Sea, including increased river water input, increased nutrient concentration, and reduced oxygen level. This allowed us to identify informative prokaryotic gene markers, responding to environmental perturbation. Our results demonstrate that metagenomic and metabolic changes in bacterial communities in response to environmental stressors are influenced both by the initial community composition and by the biogeochemical factors shaping the functional response. Furthermore, the different sources of dissolved organic matter (DOM) had the largest impact on metagenomic blueprint. Most prominently, changes in DOM loads influenced specific transporter types reflecting the substrate availability and DOC assimilation and consumption pathways. The results provide new knowledge for developing models of ecosystem structure and biogeochemical cycling in future climate change scenarios and advance our exploration of the potential use of marine microorganisms as markers for environmental conditions.

Flow cytometric monitoring of bacterioplankton phenotypic diversity predicts high population-specific feeding rates by invasive dreissenid mussels.

Species invasion is an important disturbance to ecosystems worldwide, yet knowledge about the impacts of invasive species on bacterial communities remains sparse. Using a novel approach, we simultaneously detected phenotypic and derived taxonomic change in a natural bacterioplankton community when subjected to feeding pressure by quagga mussels, a widespread aquatic invasive species. We detected a significant decrease in diversity within 1h of feeding and a total diversity loss of 11.6±4.1% after 3h. This loss of microbial diversity was caused by the selective removal of high nucleic acid populations (29±5% after 3h). We were able to track the community diversity at high temporal resolution by calculating phenotypic diversity estimates from flow cytometry (FCM) data of minute amounts of sample. Through parallel FCM and 16S rRNA gene amplicon sequencing analysis of environments spanning a broad diversity range, we showed that the two approaches resulted in highly correlated diversity measures and captured the same seasonal and lake-specific patterns in community composition. Based on our results, we predict that selective feeding by invasive dreissenid mussels directly impacts the microbial component of the carbon cycle, as it may drive bacterioplankton communities toward less diverse and potentially less productive states.

Thermal discharge-created increasing temperatures alter the bacterioplankton composition and functional redundancy.

Elevated seawater temperature has altered the coupling between coastal primary production and heterotrophic bacterioplankton respiration. This shift, in turn, could influence the feedback of ocean ecosystem to climate warming. However, little is known about how natural bacterioplankton community responds to increasing seawater temperature. To investigate warming effects on the bacterioplankton community, we collected water samples from temperature gradients (ranged from 15.0 to 18.6 °C) created by a thermal flume of a coal power plant. The results showed that increasing temperatures significantly stimulated bacterial abundance, grazing rate, and altered bacterioplankton community compositions (BCCs). The spatial distribution of bacterioplankton community followed a distance similarity decay relationship, with a turnover of 0.005. A variance partitioning analysis showed that temperature directly constrained 2.01 % variation in BCCs, while temperature-induced changes in water geochemical and grazing rate indirectly accounted for 4.03 and 12.8 % of the community variance, respectively. Furthermore, the relative abundances of 24 bacterial families were linearly increased or decreased (P < 0.05 in all cases) with increasing temperatures. Notably, the change pattern for a given bacterial family was in concert with its known functions. In addition, community functional redundancy consistently decreased along the temperature gradient. This study demonstrates that elevated temperature, combined with substrate supply and trophic interactions, dramatically alters BCCs, concomitant with decreases in functional redundancy. Roles of diatom nutrient stress and species identity in determining the short- and long-term bioavailability of diatom exudates to bacterioplankton

Phytoplankton exude carbon-rich dissolved organic matter (DOM) upon nutrient stress, yet the ecological role of this exudate in stimulating bacterial growth is not well understood. We harvested DOM produced by four coastal diatoms (Skeletonema marinoi, Chaetoceros socialis, Thalassiosira weissflogii, and Odontella aurita) subjected to depletion of nitrogen (N), silicon (Si), or N+Si simultaneously in batch culture and assessed its bioavailability to natural bacterioplankton communities using dilution batch-culture remineralization bioassays. Short-term, ecologically-relevant responses were affected both by diatom source species and by the nutrient stress under which the DOM was produced. Si-stress DOM was generally more bioavailable over the first week than that produced under N or N+Si stress, and led to higher bacterial growth efficiencies. In contrast, the amount of diatom-derived total organic carbon (TOC) that persisted over months in the same experiments differed among source diatom species, with no evidence of a nutrient stress effect. These results nuance the carbon overflow hypothesis of DOM release, which has been suggested to be maladaptive due to the possibility of DOC release allowing bacterioplankton to better compete for the same nutrient that is limiting the phytoplankton. Our results suggest that bacterial activity is most promoted when diatoms are limited by Si, a nutrient that is not subject to competition with bacteria. They imply that the identity of the nutrient that terminates a diatom bloom impacts heterotrophic activity in the short term; however, source diatom species has the greater influence on DOC persistence in surface waters and its potential export from the system.

Phytoplankton species‐specific release of dissolved free amino acids and their selective consumption by bacteria

Despite representing only a small fraction of the ocean's dissolved organic matter pool, dissolved free amino acids (DFAA) have high turnover rates and are major nitrogen and carbon sources for bacterioplankton. Both phytoplankton and bacterioplankton assimilate and release DFAA, but their consumption and production are difficult to quantify in nature due to their short residence times (min) as dissolved monomers. We segregated DFAA production by phytoplankton and bacterial consumption by measuring individual DFAA concentrations in four axenic phytoplankton cultures during the exponential growth phase, and also after 4 d incubations in the presence of a natural bacterioplankton community. The amounts and composition of the DFAA pool varied widely among phytoplankton species. The proportion of dissolved organic carbon attributed to DFAA varied among cultures. The picoeukaryotic prasinophyte, Micromonas pusilla, released higher amounts of DFAA than the other species tested (diatoms and dinoflagellate), especially alanine, which has been reported as the dominant individual DFAA in some oligotrophic environments. Community structure of heterotrophic prokaryotes responded to differences in the quality of organic matter released among microalgal species, with Roseobacter‐related bacteria responding strongly to exudate composition. Our results demonstrate the specificity of DFAA extracellular release among several algal species and their preferential uptake by members of bacterial communities.

Response of bacterioplankton activity in an Arctic fjord system to elevated &amp;lt;i&amp;gt;p&amp;lt;/i&amp;gt;CO&amp;lt;sub&amp;gt;2&amp;lt;/sub&amp;gt;: results from a mesocosm perturbation study

Abstract. The effect of elevated seawater carbon dioxide (CO2) on the activity of a natural bacterioplankton community in an Arctic fjord system was investigated by a mesocosm perturbation study in the frame of the European Project on Ocean Acidification (EPOCA). A pCO2 range of 175–1085 μatm was set up in nine mesocosms deployed in the Kongsfjorden (Svalbard). The activity of natural extracellular enzyme assemblages increased in response to acidification. Rates of β-glucosidase and leucine-aminopeptidase increased along the gradient of mesocosm pCO2. A decrease in seawater pH of 0.5 units almost doubled rates of both enzymes. Heterotrophic bacterial activity was closely coupled to phytoplankton productivity in this experiment. The bacterioplankton community responded to rising chlorophyll a concentrations after a lag phase of only a few days with increasing protein production and extracellular enzyme activity. Time-integrated primary production and bacterial protein production were positively correlated, strongly suggesting that higher amounts of phytoplankton-derived organic matter were assimilated by heterotrophic bacteria at increased primary production. Primary production increased under high pCO2 in this study, and it can be suggested that the efficient heterotrophic carbon utilisation had the potential to counteract the enhanced autotrophic CO2 fixation. However, our results also show that beneficial pCO2-related effects on bacterial activity can be mitigated by the top-down control of bacterial abundances in natural microbial communities.

Effects of UV radiation on the taxonomic composition of natural bacterioplankton communities from Bahía Engaño (Patagonia, Argentina)

In order to gain insights into the effects of solar ultraviolet radiation (UVR, 280–400nm) on the composition of marine bacterioplankton communities from South Atlantic waters – Bahía Engaño (Patagonia, Argentina), we performed microcosms experiments during the Austral summer of 2010. Water samples were exposed to three solar radiation treatments in 25 L microcosms during 8days: PAR+UV-A+UV-B (280–700nm; PAB treatment), PAR+UV-A (320–700nm; PA treatment), and PAR only (400–700nm; P treatment). The taxonomic composition of the bacterial communities, at the beginning and at the end of the experiment, were studied by the analyses of 16S rDNA gene libraries. Multivariate and phylogenetic analyses demonstrated substantial differences in the community composition so that the samples exposed to PAR and PAR+UV-A presented more similar taxa assemblages among them than compared to the PAR+UV-A+UV-B exposed one. Our results indicate that overall, exposure to different radiation treatments can shape the taxonomic composition of marine bacterial populations, grown in microcosms, from this Patagonian area.

Changes in Bacterial Community Structure after Exposure to Silver Nanoparticles in Natural Waters

Silver nanoparticles (AgNPs) are widely used in commercial products as antibacterial agents, but AgNPs might be hazardous to the environment and natural aquatic bacterial communities. Our recent research demonstrated that AgNPs rapidly but temporarily inhibited natural bacterioplankton production. The current study investigates the mechanism for the observed bacterial reaction to AgNPs by examining how AgNPs impact bacterial abundance, metabolic activity (5-cyano-2,3-ditolyl tetrazolium chloride (CTC+) cells), and 16S rRNA community composition. Natural bacterioplankton communities were dosed with carboxy-functionalized AgNPs at four concentrations (0.01-1 mg-Ag/L), incubated in triplicate, and monitored over 5 days. Ionic silver (AgNO(3)) and Milli-Q water treatments were used as a positive and negative control, respectively. Four general AgNP exposure responses, relative to the negative control, were observed: (1) intolerant, (2) impacted but recovering, (3) tolerant, and (4) stimulated phylotypes. Relationships between cell activity indicators and bacterial phylotypes, suggested that tolerant and recovering bacteria contributed the most to the community's productivity and rare bacteria phylotypes stimulated by AgNPs did not appear to contribute much to cell activity. Overall, natural bacterial communities tolerated single, low level AgNP doses and had similar activity levels to the negative control within five days of exposure, but bacterial community composition was different from that of the control.

Genomic Insights into Bacterial DMSP Transformations

Genomic and functional genomic methods applied to both model organisms and natural communities have rapidly advanced understanding of bacterial dimethylsulfoniopropionate (DMSP) degradation in the ocean. The genes for the two main pathways in bacterial degradation, routing DMSP to distinctly different biogeochemical fates, have recently been identified. The genes dmdA, -B, -C, and -D mediate the demethylation of DMSP and facilitate retention of carbon and sulfur in the marine microbial food web. The genes dddD, -L, -P, -Q, -W, and -Y mediate the cleavage of DMSP to dimethylsulfide (DMS), with important consequences for ocean-atmosphere sulfur flux. In ocean metagenomes, sufficient copies of these genes are present for approximately 60% of surface ocean bacterial cells to directly participate in DMSP degradation. The factors that regulate these two competing pathways remain elusive, but gene transcription analyses of natural bacterioplankton communities are making headway in unraveling the intricacies of bacterial DMSP processing in the ocean.

Seasonal succession and UV sensitivity of marine bacterioplankton at an Antarctic coastal site

Despite extensive microbial biodiversity studies around the globe, studies focusing on diversity and community composition of Bacteria in Antarctic coastal regions are still scarce. Here, we studied the diversity and development of bacterioplankton communities from Prydz Bay (Eastern Antarctic) during spring and early summer 2002-2003. Additionally, we investigated the possible shaping effects of solar UV radiation (UV-R: 280-400 nm) on bacterioplankton communities incubated for 13-14 days in 650-L minicosm tanks. Ribosomal DNA sequence analysis of the natural bacterioplankton communities revealed an initial springtime community composed of three evenly abundant bacterial classes: Cytophaga-Flavobacteria-Bacteroidetes (CFB), Gammaproteobacteria and Alphaproteobacteria. At the end of spring, a shift occurred toward a CFB-dominated community, most likely a response to the onset of a springtime phytoplankton bloom. The tail end of Prydz Bay clone library diversity revealed sequences related to Deltaproteobacteria, Verrucomicrobiales, Planctomycetes, Gemmatimonadetes and an unclassified bacterium (ANT4E12). Minicosm experiments showed that incubation time was the principal determinant of bacterial community composition and that UV-R treatment significantly changed the composition in only two of the four experiments. Thus, the successional maturity of the microbial community in our minicosm studies appears to be a greater determinant of bacterial community composition rather than the nonprofound and subtle effects of UV-R.

Interactive effect of temperature and resources on carbon cycling by freshwater bacterioplankton communities

Planktonic heterotrophic prokaryotes play an essential role in all aquatic ecosystems due to their short generation times and access to dissolved organic carbon and nutrients. In order to understand how rising temperatures and nutrient loading affect the biogeochemistry of whole lake ecosystems, it is essential to understand the interactive effects that temperature and resources have on the metabolism of natural bacterioplankton communities. To address this question we sampled bacterial communities from 1 mesotrophic and 1 oligotrophic lake in Clearwater County, Minnesota, USA, in winter and summer. Each community was exposed to a combination of carbon, nitrogen and phosphorus additions at 4 different levels (ambient, 2 ×, 5 ×, and 10 × ambient) and 4 temperatures (4, 14, 24 and 34°C). Community metabolic response to temperature depended on the resource treat- ment, and the season when the community was sampled. Bacterial respiration increased more with temperature than bacterial growth, resulting in decreased bacterial growth efficiencies at higher temperatures. This result was most pronounced in the high resource treatments, while at lower resource levels the results were more ambiguous. In addition, differences between seasons and lakes suggested that the bacterial communities had adapted or acclimated to in situ temperature such that communities sampled from cold temperature environments had higher respiration at high tempera- tures than those sampled from warmer environments. These results suggest that the effect of temper- ature on carbon cycling mediated by the bacterial community depends on both the in situ resource pool and the extent to which the bacterial community is acclimated to a specific temperature regime.

Bacterial Abundance, Activity, and Viability in the Eutrophic River Warnow, Northeast Germany

The River Warnow is the drinking water source for the city of Rostock. Its eutrophic status is accompanied by high amounts of bacteria, which may reach up to 24 x 10(6) cells mL(-1) as recorded during a seasonal study in 2002. Because the river is eutrophic and also heavily loaded with organic matter, this burden is a problem for drinking water purification, as it must be removed completely to not trigger new bacterial growth in the pipeline network. Therefore, restoration measures in the river have to be planned, and bacteria have to be favored as decomposers. That includes the investigation of the physiological state of bacteria in situ. Viable and active cells in the lower reaches of River Warnow were estimated using a broad set of methods. Intact bacteria were investigated by the LIVE/DEAD BacLight bacterial viability kit, containing a mixture of permeant and impermeant nucleic acid stains. Cells with ribosomes were visualized by fluorescence in situ hybridization with the EUB338 oligonucleotide probe. Intact cells and ribosome-containing bacteria represented 24% of total numbers stained by 4'6,-diamidino-2-phenylindole (DAPI) or 66 and 62%, respectively, in relation to all bacteria visualized by the LIVE/DEAD kit. Both fractions were considered as viable, although the fraction of RIB + bacteria is most likely underestimated by the protocol applied. 5-Cyano-2,3-ditolyltetrazolium chloride (CTC) was applied to mark respiring bacteria. The esterase substrate CellTracker Green 5-chloromethylfluorescein diacetate showed cells with intracellular hydrolytic activity. Whereas 1.5% of DAPI-stained bacteria were observed as respiring, 3.8% exhibited intracellular hydrolytic activity on average. If these active fractions were calculated as the percentages of intact cells, much higher fractions of 5.4% were respiring and 16% hydrolytic. Temperature was a main factor influencing total and viable cell numbers simultaneously. The results confirm that there are different states of viable and active cells in natural bacterioplankton communities. However, it remains unclear why fractions of viable and active cells were rather low in this eutrophic river in comparison to similar waters. We recommend to carefully address cells as viable in contrast to nonviable, i.e., dead. As viable cells may be active or inactive with respect to many different activities, e.g., substrate uptake, respiration, hydrolysis, and cell deviation, it is necessary to choose the method to visualize active cells according to the question to be answered.

Specific Detection, Isolation, and Characterization of Selected, Previously Uncultured Members of the Freshwater Bacterioplankton Community

High-throughput cultivation was combined with rapid and group-specific phylogenetic fingerprinting in order to recover representatives of three freshwater bacterioplankton communities. A total of 570 bacterial cultures were obtained by employing the most probable number and MicroDrop techniques. The majority of the cultured bacteria were closely related to previously uncultured bacteria and grouped with the alpha-Proteobacteria, beta-Proteobacteria, Actinobacteria, Firmicutes, or Flavobacteria-Cytophaga lineage. Correspondingly, the natural bacterioplankton community was analyzed by high-resolution phylogenetic fingerprinting of these five bacterial lineages. 16S rRNA gene fragments were generated for each lineage and subsequently separated by denaturing gradient gel electrophoresis. By the combination of five group-specific PCR protocols, the total number of 16S rRNA gene fingerprints generated from the natural communities was increased sixfold compared to conventional (eubacterial) fingerprinting. Four of the environmental alpha-Proteobacteria 16S rRNA gene sequences obtained from the natural community were found to be identical to those of bacterial isolates. One of these phylotypes was detected in 14 different cultures and hence represented the most frequently cultured bacterium. Three of these 14 strains were characterized in detail. Their complete 16S rRNA gene sequences showed only 93% similarity to that of Sandaracinobacter sibiricus, the closest relative described so far. The novel phylotype of bacterium is a strict aerobe capable of using numerous organic carbon substrates and contains bacteriochlorophyll a bound to two different photosynthetic light-harvesting complexes. Dot blot hybridization revealed that the strains occur in lakes of different trophic status and constitute up to 2% of the microbial community.

Bioluminescent Bacteria as Indicators of Chemical Contamination of Coastal Waters

The ratio of bioluminescent to total bacteria (bioluminescent ratio, BLR) as an indicator of a variety of types of anthropogenic contamination of estuarine ecosystems was evaluated through a series of laboratory and field studies. Laboratory studies indicated that the BLR of natural bacterioplankton communities was proportionally reduced in the presence of a number of contaminants including diesel fuel and saltmarsh sediments co-contaminated with mercury and polychlorinated biphenyls (PCBs). Bioluminescent ratio inhibition was observed after short-term exposure to a contaminant suggesting a physiological rather than a population response of native microbial communities. Simulated eutrophication did not suppress the BLR. Field observations of the BLR were conducted weekly for a 2-yr period in the Skidaway River estuary, Georgia, USA. These observations revealed considerable seasonal variability associated with the BLR. Bioluminescent ratios were highest during the summer (25 +/- 15%), lower in the fall (6 +/- 5%) and spring (3 +/- 2%), and near zero during the winter. Although the BLR was not significantly correlated to salinity at a single site (Skidaway River estuary), the BLR was significantly correlated with salinity when sites within the same estuary system were compared (r2 = 0.93). Variation in BLR was not correlated to standard bacteriological indicators of water quality including total and fecal coliform bacteria. Comparison of the BLR from impacted and pristine estuarine sites during the fall suggested that anthropogenically impacted sites exhibited lower BLR than predicted from salinity versus BLR relationships developed in pristine systems. These observations suggest that the BLR could be used as a simple and reliable initial indicator of chemical contamination of estuarine systems resulting from human activity.

Ecological implications of biomass and morphotype variations of bacterioplankton: an example in a coastal zone of the Northern Adriatic Sea (Mediterranean)

AbstractThis study had the objective of quantifying the variability in abundance, cell volume, morphology and C content of a natural bacterioplankton community in a coastal zone of the North Adriatic Sea during two periods (February and June) of two consequent years (1996 and 1997). We used epifluorescence microscopy with Acridine Orange staining procedures and a microphotographic technique. Low variability in bacterial abundance (range 0.3–3.1 × 105 cells ml−1) occurred between summer and winter periods. Conversely, the cell volume and the calculated carbon content changed greatly with warm and cold periods (ranges: 0.015–0.303 μm3 and 5.83–42.17 fg C cell−1, respectively). Elongated bacteria were dominant while coccoid cells prevailed only in February 1997. Biomass showed high variability (range 0.12–10.21 μg C l−1) whilst the abundance did not show noticeable differences among the sampling periods. As a consequence, quantification of bacterial biomass based solely on cell abundance must be considered with caution because the true biomass could depend on variability in cell volumes and morphotypes.
