Material extrusion additive manufacturing of poly(lactic acid)/Ti6Al4V@calcium phosphate core-shell nanocomposite scaffolds for bone tissue applications

The use of porous scaffolds with appropriate mechanical and biological features for the host tissue is one of the challenges in repairing critical-size bone defects. With today's three-dimensional (3D) printing technology, scaffolds can be customized and personalized, thereby eliminating the problems associated with conventional methods. In this work, after preparing Ti6Al4V/Calcium phosphate (Ti64@CaP) core-shell nanocomposite via a solution-based process, by taking advantage of fused deposition modeling (FDM), porous poly(lactic acid) (PLA)-Ti64@CaP nanocomposite scaffolds were fabricated. Scanning electron microscope (SEM) showed that nanostructured calcium phosphate was distributed uniformly on the surface of Ti64 particles. Also, X-ray diffraction (XRD) indicated that calcium phosphate forms an octacalcium phosphate (OCP) phase. As a result of incorporating 6 wt% Ti64@CaP into the PLA, the compressive modulus and ultimate compressive strength values increased from 1.4 GPa and 29.5 MPa to 2.0 GPa and 53.5 MPa, respectively. Furthermore, the differential scanning calorimetry results revealed an increase in the glass transition temperature of PLA, rising from 57.0 to 62.4 °C, due to the addition of 6 wt% Ti64@CaP. However, it is worth noting that there was a moderate decrease in the crystallization and melting temperatures of the nanocomposite filament, which dropped from 97.0 to 89.5 °C and 167 to 162.9 °C, respectively. The scaffolds were seeded with human adipose tissue-derived mesenchymal stem cells (hADSCs) to investigate their biocompatibility and cell proliferation. Calcium deposition, ALP activity, and bone-related proteins and genes were also used to evaluate the bone differentiation potential of hADSCs. The obtained results showed that introducing Ti64@CaP considerably improved in vitro biocompatibility, facilitating the attachment, differentiation, and proliferation of hADSCs. Considering the findings of this study, the 3D-printed nanocomposite scaffold could be considered a promising candidate for bone tissue engineering applications.

Effect of L-Glutamic Acid on the Composition and Morphology of Nanostructured Calcium Phosphate as Biomaterial.

Calcium phosphate (CaP) with several chemical compositions and morphologies was prepared by precipitation using aqueous solutions of L-Glutamic acid (H2G) and calcium hydroxide, both mixed together with an aqueous solution (0.15 M) of phosphoric acid. Plate-shaped dicalcium phosphate dihydrate (brushite) particles were obtained and identified at a lower concentration of the solution of the reactants. The Ca/P ratio deduced by EDS was ~1, as expected. The nanoscale dimension of carbonate apatite and amorphous calcium phosphate, with variable Ca/P ratios, were revealed by X-ray diffraction (XRD) and scanning electron microscopy and energy dispersive X-ray spectroscopy analysis (SEM-EDS). They were characterized in medium and high concentrations of calcium hydroxide (0.15 M and 0.20 M). The equilibria involved in all the reactions in aqueous solution were determined. The thermodynamic calculations showed a decrease in the amount of chelate complexes with an increase in pH, being the opposite of [CaPO4-] and [CaHG+]. This fluctuation showed an evident influence on the morphology and polymorphism of CaP particles obtained under the present experimental conditions, with potential use as a biomaterial.

Does Melatonin Associated with Nanostructured Calcium Phosphate Improve Alveolar Bone Repair?

Background and objectives: Calcium phosphates have been widely used as bone substitutes, but their properties are limited to osteoconduction. The association of calcium phosphates with osteoinductive bioactive molecules has been used as a strategy in regenerative medicine. Melatonin has been studied due to its cell protection and antioxidant functions, reducing osteoclastic activity and stimulating newly formed bone. This study aimed to evaluate the effect of topical application of melatonin associated with nanostructured carbonated hydroxyapatite microspheres in the alveolar bone repair of Wistar rats through histological and histomorphometric analysis. Materials and Methods: Thirty female Wistar rats (300 g) were used, divided randomly into three experimental groups (n = 10), G1: nanostructured carbonated hydroxyapatite microspheres associated with melatonin gel (CHA-M); G2: nanostructured carbonated hydroxyapatite (CHA); G3: blood clot (without alveolar filling). The animals were euthanized after 7 and 42 days of the postoperative period and processed for histological and histomorphometric evaluation. Kruskal-Wallis and Dunn's post-test were applied to investigate statistical differences between the groups at the same time point for new bone and connective tissue variables. Mann-Whitney was used to assess statistical differences between different time points and in the biomaterial variable. Results: Results showed a greater volume of residual biomaterial in the CHA-M than the CHA group (p = 0.007), and there were no significant differences in terms of newly formed bone and connective tissue between CHA and CHA-M after 42 days. Conclusions: This study concluded that both biomaterials improved alveolar bone repair from 7 to 42 days after surgery, and the association of CHA with melatonin gel reduced the biomaterial's biodegradation at the implanted site but did not improve the alveolar bone repair.

Novel rechargeable nanostructured calcium phosphate crown cement with long-term ion release and antibacterial activity to suppress saliva microcosm biofilms

ObjectiveResin cements with remineralizing and antibacterial properties are favorable for inhibition of caries. The objectives of this study were: (1) to investigate the capability of the novel dimethylaminohexadecyl-methacrylate (DMAHDM) and nano-sized amorphous calcium phosphate (NACP) containing cement to reduce saliva microcosm biofilm, and (2) to investigate the long-term ion release, recharge, and re-release of DMAHDM-NACP cement. MethodsPyromellitic glycerol dimethacrylate (PMGDM) and ethoxylated bisphenol-A-dimethacrylate (EBPADMA) were used to make PEHB monomer. Five cements were fabricated: (1) PEHB+0%NACP+0%DMAHDM (experimental control); (2) PEHB+25%NACP+0%DMAHDM, (3) PEHB+25%NACP+0%DMAHDM; (4) PEHB+25%NACP+3%DMAHDM; (5) PEHB+25%NACP+5%DMAHDM. RelyX luting cement was used as commercial control. Colony-forming units (CFU), lactic acid production, metabolic activities, and minimum inhibitory concentration (MIC) were performed. Long-term Calcium (Ca) and phosphate (P) ion release, recharge, and re-release were assessed. ResultsCompared to experimental and commercial controls, the NACP-DMAHDM cement significantly reduced CFU biofilm by 2–3 orders of magnitude, metabolic activities from 0.24±0.06 A540/cm2 to 0.03±0.01 A540/cm2, and lactic acid production from 27.7 ± 2.5 mmol/L to 5.4 ± 2.1 mmol/L (n = 6) (p<0.05). The DMAHDM showed an MIC value of 0.03 mg/L. However, when the DMAHDM was combined with PMGDM monomer, the MIC was greater than DMAHDM alone. The ion concentrations for the experimental groups significantly increased over time (1–84 days), indicating continuous ion release (n = 3) (p<0.05). Increasing the DMAHDM mass fraction from 0% to 5% and 3% to 5% significantly enhanced ion recharge and re-release at the third cycle (p<0.05). ConclusionsIncorporating DMAHDM and NACP into resin-based crown cement provides strong antibacterial action against saliva microcosm biofilm and presents a high level of Ca and P ion recharge abilities, exhibiting long-term Ca and P ion release and remineralization potential. Clinical SignificanceResin based cement containing NACP and DMAHDM were developed with remineralizing and potent antibacterial effects. This cement formulation showed ion release and remineralization potential and are promising formulations to inhibit the incidence of recurrent caries and could promote remineralization and be sustainable for the long term.

An Ammonia-Induced Calcium Phosphate Nanostructure: A Potential Assay for Studying Osteoporosis and Bone Metastasis.

Osteoclastic resorption of bones plays a central role in both osteoporosis and bone metastasis. A reliable in vitro assay that simulates osteoclastic resorption in vivo would significantly speed up the process of developing effective therapeutic solutions for those diseases. Here, we reported the development of a novel and robust nanostructured calcium phosphate coating with unique functions on the track-etched porous membrane by using an ammonia-induced mineralization (AiM) technique. The calcium phosphate coating uniformly covers one side of the PET membrane, enabling testing for osteoclastic resorption. The track-etched pores in the PET membrane allow calcium phosphate mineral pins to grow inside, which, on the one hand, enhances coating integration with a membrane substrate and, on the other hand, provides diffusion channels for delivering drugs from the lower chamber of a double-chamber cell culture system. The applications of the processed calcium phosphate coating were first demonstrated as a drug screening device by using alendronate, a widely used drug for osteoporosis. It was confirmed that the delivery of alendronate significantly decreased both the number of monocyte-differentiated osteoclasts and coating resorption. To demonstrate the application in studying bone metastasis, we delivered a PC3 prostate cancer-conditioned medium and confirmed that both the differentiation of monocytes into osteoclasts and the osteoclastic resorption of the calcium phosphate coating were significantly enhanced. This novel assay thus provides a new platform for studying osteoclastic activities and assessing drug efficacy in vitro.

Production and characterization of Ti6Al4V/CaP nanocomposite powder for powder-based additive manufacturing systems

Ti64/CaP nanocomposite powder was fabricated and characterized for use in powder-bed 3D printing. The microstructure and phase composition, morphology, particle size distribution, sphericity, flow behavior and dispersion of the as-fabricated particles on the building plate of the 3D printer were investigated. The results confirmed a uniform distribution of nanostructured calcium phosphate particles on the surface of primary Ti64 ones. Calcium phosphate appears as an octa-calcium phosphate phase. The morphology of the particles was shown as spherical, and their sphericity was better than the as-received Ti64 particles. The particle size distribution of nanocomposite powder indicated a smaller amount than the as-received Ti64 particles. Additionally, the flow behavior was improved compared to the as-received Ti64 and delivered an optimal dispersion on the building platform. Ultimately, Ti64/CaP nanocomposite powder has a very homogeneous and uniform microstructure and bears a proper flow behavior for use in powder-bed additive manufacturing machines.

Principal Physics of Oblique Sputtering of Calcium Phosphate Coatings Using RF-Magnetron Discharge Plasma

A possibility in principle is determined for the titanium surface functionalization by oblique sputtering of nanostructured bioactive calcium phosphate coatings. The coatings are deposited by the RF magnetron sputtering of the targets based on hydroxyapatite and antibacterial zinc-substituted hydroxyapatite. The coating morphology and their growth characteristics are studied as a function of such process parameters as the specimen tilt angle with respect to the particle flux from the source, the supplied power, and the distance from the target plane to the specimen surface. As the tilt angle is increased, the coating surface morphology significantly changes and represents nanosized structural elements oriented towards the particle source and ordered in accordance with the substrate surface morphology prior to sputtering. The inner coating structure consists of columnar elements tilted to the substrate plane.

In Vitro and In Vivo Evaluation of Nanostructured Biphasic Calcium Phosphate in Granules and Putty Configurations.

Synthetic biphasic calcium phosphate (BCP) granules and powder are biocompatible biomaterials with a well-known capacity for osteoconduction, presenting very satisfactory clinical and histological results. It remains unanswered if the putty configuration impacts the biological response to the material. In this study, we aimed to compare the cytocompatibility and biocompatibility of nanostructured BCP in the putty configuration (moldable nanostructured calcium phosphate, MnCaP) on the healing of critical-sized bone defects (8 mm) in rat calvaria. Cytocompatibility was determined through the viability of fibroblast cells (V-79) to the extracts of different concentrations of MnCaP. Forty-five Wistar rats were randomly divided into three groups (n = 15)—clot, MnCaP, and commercial biphasic calcium phosphate in granules configurations (Nanosynt®)—and subdivided into three experimental periods (1, 3, and 6 months). Histological, histomorphometric, and microtomographic analyses allowed the evaluation of newly formed bone, residual biomaterial, and connective tissue. The in vitro evaluation showed that MnCaP was cytocompatible. The histomorphometric results showed that the Nanosynt® group granted the highest new-formed bone values at six months (p < 0.05), although the biomaterial volume did not differ between groups. The putty configuration was easier to handle, and both configurations were biocompatible and osteoconductive, presented similar biosorption rates, and preserved the calvaria architecture.

Trabecular architecture during the healing process of a tibial diaphysis defect

The adaptation of trabecular bone microstructure to mechanical loads has been intensively investigated. However, loading-unrelated aspects of trabecular architecture remain unclear. We used synchrotron radiation-based X-ray microtomography to study the 3D microarchitecture of newly formed trabecular tissue in a defect produced in the cortical region of the rat tibia diaphysis, in the absence (7, 14, and 21 days) or the presence (21 days) of carbonated hydroxyapatite/alginate (cHA) microspheres. This work provides the first evidence that the woven bone trabecular network, formed during the healing process, displays a well-organized 3D microarchitecture consisting of nodes with 3 (3-N), 4 (4-N) and 5 (5-N) connecting trabeculae, with a mean relative abundance of (3-N)/(4-N)/(5-N) = 66/24/7, for the analyzed periods. The measured inter-trabecular angles (ITA) distribution presented a Gaussian profile, with mean value at 115° for 3-N nodes, and 105° for 4-N nodes, close to the angles of idealized 3D regular structures (120° and 109.5°, respectively). Changes in the dispersion of ITA distribution suggested that a highly symmetric trabecular fabric organized under tensegrity principles is formed early during the bone healing process. Post-implantation, cHA disaggregated into multiple fragments (~20–400 μm), stimulating osteoconduction and bone growth toward the interior of the medullary cavity. The presence of biomaterials in bone defects affected the trabecular dimensions; however, it did not interfere with the formation of geometrical motifs with topological parameters similar to those found in the sham-defects. Statement of SignificanceThe trabecular bone microstructure enables the tissue to meet the necessary mechanical and functional demands. However, the process of trabecular microarchitecture formation during healing, in the absence or presence of a bone graft, is not yet well understood. This work demonstrated that, from the beginning of its formation in cortical bone defects, the woven-bone trabecular network is spatially organized according to the principle of tensegrity. This microarchitecture is comprised of highly symmetric geometric motifs and is an intrinsic characteristic of trabecular growth, regardless of hierarchical scale or mechanical stimulation. The addition of a biodegradable nanostructured calcium phosphate graft did not disrupt trabecular microarchitecture; however, graft biodegradation should be controlled to optimize the reproduction of intrinsic trabecular motifs throughout the defect.

Healing of osteoporotic bone defects by micro-/nano-structured calcium phosphate bioceramics

It is a particularly critical challenge to achieve enough bone regeneration in osteoporotic fractures with bone defects. In the present study, we designed a micro-/nano-structured calcium phosphate bioceramic composed of a nanoparticle-reinforced micro-whisker backbone (nwCaP). This sandwich-structured bioceramic exhibited a higher compressive strength, a suitable degradation rate and better cell attachment than traditional or intermediate bioceramics. In a rat model of osteoporotic bone defects, the nwCaP group showed a reduced fracture occurrence and an effective new bone substitution rate, as characterized by micro-CT analysis. The increased bone formation rate and greater amount of new bone formed within the defected area of the nwCaP group was revealed by the serum PINP level and histological staining. Moreover, a gene microarray study indicated that the promotion of osteogenesis might be attributed to selectively upregulated fibroblast growth factor 23 (FGF23) expression in cells co-cultured with the nwCaP bioceramic. Furthermore, the JAK2 signal pathway was confirmed to be involved in the nwCaP-induced elevation of FGF23 expression using primary osteoblasts derived from osteoporotic rats. Collectively, the findings suggested that the micro-/nano-structured bioceramic could enhance osteoporotic bone regeneration and presents a promising strategy for healing bone defects in osteoporosis.

Cytocompatibility and Matrix Properties of Surfaces of Nanostructured Calcium Phosphate Cements

There is a growing interest in bioactive materials of medicinal use, because their employment gives large opportunities in the recovery of the integrity of bone tissue, which is dictated by their biocompatibility and bioactivity. The authors have compared cytocompatibility and matrix properties of a series of nanostructured calcium phosphate cements and have elaborated recommendations on improvement of primary in vitro screening of the materials dedicated for the replacement of osteochondral defects.

Osteogenesis by foamed and 3D-printed nanostructured calcium phosphate scaffolds: Effect of pore architecture

There is an urgent need of synthetic bone grafts with enhanced osteogenic capacity. This can be achieved by combining biomaterials with exogenous growth factors, which however can have numerous undesired side effects, but also by tuning the intrinsic biomaterial properties. In a previous study, we showed the synergistic effect of nanostructure and pore architecture of biomimetic calcium deficient hydroxyapatite (CDHA) scaffolds in enhancing osteoinduction, i.e. fostering the differentiation of mesenchymal stem cells to bone forming cells. This was demonstrated by assessing bone formation after implanting the scaffolds intramuscularly. The present study goes one step forward, since it analyzes the effect of the geometrical features of the same CDHA scaffolds, obtained either by 3D-printing or by foaming, on the osteogenic potential and resorption behaviour in a bony environment. After 6 and 12 weeks of intraosseous implantation, both bone formation and material degradation had been drastically affected by the macropore architecture of the scaffolds. Whereas nanostructured CDHA was shown to be highly osteoconductive both in the robocast and foamed scaffolds, a superior osteogenic capacity was observed in the foamed scaffolds, which was associated with their higher intrinsic osteoinductive potential. Moreover, they showed a significantly higher cell-mediated degradation than the robocast constructs, with a simultaneous and progressive replacement of the scaffold by new bone. In conclusion, these results demonstrate that the control of macropore architecture is a crucial parameter in the design of synthetic bone grafts, which allows fostering both material degradation and new bone formation.Statement of Significance3D-printing technologies open new perspectives for the design of patient-specific bone grafts, since they allow customizing the external shape together with the internal architecture of implants. In this respect, it is important to design the appropriate pore geometry to maximize the bone healing capacity of these implants. The present study analyses the effect of pore architecture of nanostructured hydroxyapatite scaffolds, obtained either by 3D-printing or foaming, on the osteogenic potential and scaffold resorption in an in vivo model. While nanostructured hydroxyapatite showed excellent osteoconductive properties irrespective of pore geometry, we demonstrated that the spherical, concave macropores of foamed scaffolds significantly promoted both material resorption and bone regeneration compared to the 3D-printed scaffolds with orthogonal-patterned struts and therefore prismatic, convex macropores.

Strontium-Doped Amorphous Calcium Phosphate Porous Microspheres Synthesized through a Microwave-Hydrothermal Method Using Fructose 1,6-Bisphosphate as an Organic Phosphorus Source: Application in Drug Delivery and Enhanced Bone Regeneration.

Nanostructured calcium phosphate porous microspheres are of great potential in drug delivery and bone regeneration due to their large specific surface area, biocompatibility, and similarity to inorganic component of osseous tissue. In this work, strontium (Sr)-doped amorphous calcium phosphate porous microspheres (SrAPMs) were synthesized through a microwave-hydrothermal method using fructose 1,6-bisphosphate trisodium salt as the source of phosphate ions. The SrAPMs showed a mesoporous structure and a relatively high specific area. Compared with the hydroxyapatite nanorods prepared by using Na2HPO4·12H2O as the phosphorus source, the SrAPMs with a higher specific surface area were more effective in drug loading using vancomycin as the antiobiotics of choice and consequently having a higher antibacterial efficiency both on agar plates and in broths. Furthermore, to assess the potential application of SrAPMs in bone defect repair, a novel biomimetic bone tissue-engineering scaffold consisting of collagen (Coll) and SrAPMs was constructed using a freeze-drying fabrication process. Incorporation of the SrAPMs not only improved the mechanical properties, but also enhanced the osteogenesis of rat bone marrow mesenchymal stem cells. The in vivo experiments demonstrated that the SrAPMs/Coll scaffolds remarkably enhanced new bone formation compared with the Coll and APMs/Coll scaffolds in a rat critical-sized calvarial defect model at 8 weeks postimplantation. In summary, SrAPMs developed in this work are promising as antibiotic carriers and may encourage bone formation when combined with collagen.

Molecular designing of nanoparticles and functional materials

The interdisciplinary research team implemented the program titled ?Molecular designing of nanoparticles with controlled morphological and physicochemical characteristics and functional materials based on them? (MODENAFUNA), between 2011 and 2016, gaining new knowledge significant to the further improvement of nanomaterials and nanotechnologies. It gathered under its umbrella six main interrelated topics pertaining to the design and control of morphological and physicochemical properties of nanoparticles and functional material based on them using new methods of synthesis and processing: 1) inorganic nanoparticles, 2) cathode materials for lithium-ion batteries, 3) functional ceramics with improved electrical and optical properties, 4) full density nanostructured calcium phosphate and functionally-graded materials, 5) nano-calcium phosphate in bone tissue engineering and 6) biodegradable micro- and nano-particles for the controlled delivery of medicaments.

Dense nanostructured calcium phosphate coating on titanium by cold spray

This article deals with the understanding of building-up mechanisms of bioactive nanocrystalline hydroxyapatite coatings by Cold Spray, revealing very promising results in contrast to more conventional techniques such as Plasma Spray. A full characterization of feedstock and coatings is provided. The agglomerated structure of the powder proved to be suitable to obtain successfully thick hydroxyapatite coatings. A crystallite size below ∼20nm in the powder and the as-sprayed coatings is calculated by the Rietveld X-ray refinement method and agreed by Transmission Electron Microscopy. Some wipe tests were carried out on Ti6Al4V substrates in order to study the deposition of single particles and the nanoscale features were evaluated. The resulting structure indicates that there is no delimitation of particle boundaries and the overall coating has been formed by effective compaction of the original nanocrystallites, leading to consistent and consolidated layers.

Synthesis and Characterization of Nanostructured Calcium Phosphates: Colloidal Suspension Rheological Study

Calcium phosphates bioceramics are a broad class of biomaterials, known by properties like biocompatibility and bioactivity in living tissues. The aim of this paper was the synthesis by wet method and characterization of nanostructured calcium phosphate powders in the Ca/P ratio of 1.67molar and rheological evaluation of the colloidal suspension during the synthesis: suspension viscosity with variable shear rate. Morphological characterization was performed for the hydrated nanostructured calcium phosphate powder by scanning electron microscopy with field effect, crystallography by X-Ray diffractometry, chemical characterization by Fourier Transformed Infrared Spectroscopy. The results demonstrated that varying the shear rate has a direct influence on the viscosity values according to time. This result may contribute to the development of a synthesis methodology for nanostructured calcium phosphate powders.

In situ formation of nanostructured calcium phosphate coatings on porous hydroxyapatite scaffolds using a hydrothermal method and the effect on mesenchymal stem cell behavior

The micro/nano-structure of calcium phosphate has been demonstrated to play an essential role in determining cell behavior. However, it is difficult to fabricate micro/nano-structures on the surface of hydroxyapatite (HA) scaffolds. In this study, we have developed a new hydrothermal method for fabricating nanostructures on HA scaffolds with the assistance of 1,2,3,4,5,6-cyclohexanehexacarboxylic acid (H6L). We controlled the nanostructures of the calcium phosphate coatings deposited on the scaffolds by modulating the H6L concentration. The results demonstrated that with an increase in the H6L concentration, the calcium phosphate coatings gradually changed from plate-like, to wire-like, and further to spherical morphology. In vitro mesenchymal stem cell (MSC) culture indicated that cell differentiation was significantly enhanced when cultured on the spherical nanostructured coating compared with culture on a plate-like or wire-like nanostructure. Cell proliferation on coatings with different nanostructures was not statistically different with prolonged culture time. These findings indicate that different scaffold surface characteristics, including surface morphology and chemical properties, result in a significant difference in cell differentiation. The small molecule-assisted hydrothermal deposition method provides a simple and controllable route to engineer the nanostructure of HA scaffolds and to construct micro-environments for bone tissue engineering.

Sirolimus-loaded CaP coating on Co-Cr alloy for drug-eluting stent.

To achieve polymer-free and controllable drug-eluting system, there have been many efforts to modify the surface composition and topography of metal stent. Recently, calcium phosphate is commonly applied to metallic implants as a coating material for fast fixation and firm-implant bone attachment on the account of its demonstrated bioactive and osteoconductive properties. In the present study, the release of sirolimus could be controllable because of immobilization of sirolimus during the process of biomimetic CaP coating forming. A completely new concept is the drug carrier of biomimetic CaP coating with sirolimus for an absorbable drug eluting system, which in turn can serve as a drug reservoir. We here describe the characteristic, mechanisms and drug release in vitro of new drug-eluting system in comparison to conventional system equivalent. Nano-structured calcium phosphate (CaP) coating was formed on the cobalt–chromium (Co-Cr) alloy substrate. By immersing coated sample in solution with sirolimus (rapamycin), the sirolimus could be immobilized in the newly formed CaP layer. The morphology, composition and formation process of the coating were studied with scanning electron microscopy, energy dispersive spectrometer, X-ray diffraction and X-ray photoelectron spectroscopy. The results showed that a uniform CaP coating incorporated with sirolimus was observed on Co-Cr alloy.
