Comparison between mycobiota diversity and fungi and mycotoxin contamination of maize and wheat

Abstract The aim of this study was to establish the differences in fungi and mycotoxin contamination and mycobiota biodiversity between wheat and maize after harvesting. Determination of fungal contamination of wheat and maize is crucial, given that these grains are extensively used in food and feed production. Some moulds can produce toxic secondary metabolites, such as mycotoxins, which can cause serious health problems in humans and animals. Due to the metabolic activities of moulds, undesirable changes in food and feed colour, taste, and smell can occur. In this study, samples of wheat and maize, harvested during 2021 in the Northern Croatia, were analysed for mycological and mycotoxin contamination before storage. Also, potentially mycotoxigenic mould genera were identified at the species level in order to uncover the presence of toxigenic species. The presence of the following mycotoxins was examined: zearalenone, deoxynivalenol, ochratoxin, aflatoxin B1, T-2, and HT-2 toxin. The results indicate that both wheat and maize samples were contaminated with yeasts, with maize exhibiting a higher level of contamination as compared to wheat (1.5 × 104 to 1.8 × 106 vs 1.7 × 103 to 9.0 × 105). In wheat, the predominant mycotoxigenic mould species was Alternaria, as opposed to maize in which the Penicillium genus was the most represented. Mycotoxin analysis revealed that both maize and wheat were contaminated with Fusarium mycotoxins. Aflatoxin B1 and ochratoxin were not determined in any of the tested samples. The results point out that grains carry a different risk of contamination with Fusarium mycotoxins and that maize is more susceptible to fungus spoilage.

Occurrence and Fate Analysis of Mycotoxins in Maize During the Post-Harvest Period

The consumption of agricultural products contaminated with mycotoxins poses a significant threat to the health of both humans and animals. Maize frequently becomes contaminated with toxic fungi while it is still growing in the field. Therefore, more proactive measures should be implemented to reduce mycotoxin levels during the storage and processing of maize after harvest. This article analyzes the prevalent mycotoxins found in maize, specifically aflatoxins, ochratoxins, trichothecenes, fumonisins, and zearalenone. The study provides a comprehensive analysis of the occurrence of mycotoxins in maize during storage, as well as fate analysis of them during processing. It summarizes the impacts of storage time, environmental conditions, storage methods, and agricultural practices on mycotoxin occurrence during storage in the post-harvest period. Furthermore, the different distribution of mycotoxins across various fractions during both dry- and wet-milling processes in the post-harvest processing period is analyzed. Additionally, the strategies to control mycotoxins in maize are also proposed during the post-harvest period. This review offers valuable insights for future research on mycotoxin contamination in maize during the post-harvest period.

Effect of Nixtamalization on Aflatoxins, Functional and Acceptability of Maize from Mulindi, Rwanda

Rwanda relies highly on maize for human food as well as for animal feed and industrial uses. However, high temperature and wet climate conditions in Rwanda favor the growth of fungi that may lead to mycotoxin contamination in maize and subsequent degradation of its quality. Therefore, this study aimed to assess the effect of nixtamalization on the aflatoxins, the functional and acceptability of maize grains from Mulindi, Rwanda. The maize samples were collected from the Mulindi market in Rwanda; representative samples were collected by the guidelines for sampling food for mycotoxin determination. Subsequently, the treated maize samples were dry-milled to produce fine flour, and the flour obtained was analyzed for swelling capacity, water absorption capacity, water solubility, acceptability, and aflatoxins, which were quantified by the HPLC method. The HPLC results showed that the highest total aflatoxin level in maize grains was 44.99 ppb. Cooking maize in water and calcium hydroxide of 1% resulted in a reduction of aflatoxin with degradation percentages of 25% and 88%, respectively. However, in all treatments, aflatoxins B1 and B2 were less reduced compared to aflatoxins G1 and G2. Cooking maize in pure water showed the lowest reduction of aflatoxin compared to the other treatments. However, cooking maize in water with 1% calcium hydroxide decreased aflatoxin to a higher percentage. Cooking maize in lime enhanced the swelling capacity, water absorption capacity, and water solubility of maize, as well as its acceptability. However, the nixtamalization process was observed to reduce aflatoxins levels in maize and improve the functional properties of maize, and therefore, its application could be encouraged for safe products in the market.

Aspergillus and Fusarium Mycotoxin Contamination in Maize (Zea mays L.): The Interplay of Nitrogen Fertilization and Hybrids Selection.

Maize plays a significant global role as a food source, feed, and as a raw material in industry. However, it is affected by toxin-producing fungi, mainly Fusarium graminearum, Fusarium verticillioides, and Aspergillus flavus, which compromise its quality. This study, conducted in 2022 and 2023 at the Látókép long-term research site of the University of Debrecen, Hungary, investigated the effects of different nitrogen fertilization rates (0, 90 and 150 Kgha-1 N) on mycotoxin contamination (DON vs. FB vs. AFB1) in the kernels of three (3) maize hybrids: DKC4590 (tolerant), GKT376 (sensitive), and P9610 (undefined). The results showed a significant (p = 0.05) influence of nitrogen fertilization and maize genotype on mycotoxin levels. Sole nitrogen impacts were complex and did not define a clear trend, contrary to the hybrids selected, which followed superiority to resistance. Increased nitrogen fertilization was associated with higher DON production, while hybrid selection demonstrated a clearer trend in resistance to mycotoxins. Therefore, to maximize yield and minimize mycotoxin contamination, future research should focus on optimizing nitrogen application rates and breeding for resistance to balance yield and mycotoxin management. These results suggest that while nitrogen fertilization is crucial for maximizing yield, selecting less susceptible maize hybrids remains vital for minimizing mycotoxin contamination.

Characterization and diversity of potential mycotoxin producing endophytic fungi isolated from maize husk leaves

Maize (Zea mays L.) is an important staple food due to its good nutritional profile. Unfortunately, maize can be vulnerable to contamination by fungi, which are capable of producing mycotoxins that have an impact on human and animal health, crop productivity and lead to economic losses. In addition, the ubiquitous presence of mycotoxin-producing fungi in maize poses significant challenges to food safety and economic stability. Mycotoxins are toxic secondary metabolites produced by certain fungi, which, when ingested by humans or animals through contaminated maize products can lead to a variety of health issues. There is a shift of non-pathogenic microbes to pathogenic state due to unfavourable conditions arising. The aim of this study was to characterize and identify fungi that have the potential to produce mycotoxins isolated from maize husks. The study identified Fusaria sp., Aspergillus and Penicillium sp. as target species since they are often regarded as primary mycotoxin producing fungi, which often contaminates the maize plants. Ten (n=10) healthy (disease-free) and mature maize samples were obtained, and a total of 200 maize husk segments were utilized to isolate eighty-four (n=84) endophytic fungi. Preliminary identification was conducted using macroscopic and microscopic characteristics. Molecular processes using the Internal transcribed spacer region (ITS1 and ITS4) were used for PCR amplification as a confirmatory test. A total of 13 isolates were identified by macroscopic and microscopic characterization based on morphological features and their identification was confirmed using molecular techniques. Fusaria sp. were found to be predominant (46%), followed by Aspergillus sp. (39%) and Penicillium sp. (15%). The findings suggest a community of mycotoxin producing fungi inhibited within the maize husks. Under unfavourable conditions, these fungi might be the source of contamination and produce mycotoxins. In the face of changing environmental conditions and evolving fungal threats, our research serves as a valuable resource for understanding and addressing the challenges posed by mycotoxin contamination in maize. Effective strategies and interventions are essential to mitigate the impact of these fungi on food security, human health, and the global maize industry. Preharvest and postharvest management practices related to mycotoxin contamination in maize in Ethiopia – a review

Abstract Mycotoxins are fungal metabolites that commonly contaminate food crops such as maize. Conducive climatic conditions together with improper crop value chain practices are favorable for mycotoxin contamination. Previous studies in Ethiopia have indicated that mycotoxin contamination in maize is prevalent. For the implementation of proper mycotoxin prevention and control strategies, identifying the current local value chain practices that are related to mycotoxin contamination is needed. This review investigates current preharvest and postharvest management practices of maize cultivation in Ethiopia in relation to mycotoxin contamination and identifies gaps in knowledge and priority areas for future research. Findings indicate that the majority of applied preharvest and postharvest practices of maize in Ethiopia seem to favor mycotoxin contamination. Recent developments in grain drying and storage technologies, which are also potential mycotoxin management strategies, are facing constraints for proper implementation in subsistence farmers’ level.

Mycotoxins and Crop Yield in Maize as Affected by Irrigation Management and Tillage Practices

In addition to the weather conditions, agronomic practices can have a major influence on maize crop yield and contamination with mycotoxins. In this work, the effect of different irrigation systems (flood vs. sprinkler irrigation), sprinkler irrigation management (low vs. high frequency, daytime vs. nighttime irrigation) and tillage practices (conventional tillage, no tillage with or without crop stover) on crop yield and the contamination with aflatoxins (AFs), fumonisins (FUM) and deoxynivalenol (DON) were evaluated in the maize grain from two experimental maize fields. No aflatoxins were detected in any of the samples analyzed. DON and FUM levels were significantly higher when the sprinkler irrigation was performed at nighttime (0.54 and 1.21 mg kg−1, respectively) as compared to daytime (0.38 and 0.45 mg kg−1). Likewise, DON and FUM were greater when irrigation frequency was low (0.61 and 1.09 mg kg−1, respectively) in comparison with high frequency (0.30 and 0.57 mg kg−1). DON concentrations were significantly higher in fields with sprinkler irrigation (0.53 mg kg−1) as compared to flood irrigation (0.19 mg kg−1), while the levels of FUM were very similar regardless of the maize irrigation system. Mycotoxin concentrations were not affected by the different soil tillage practices. This highlights the importance of implementing crop management practices to minimize the risk of mycotoxin contamination in maize.

Segmentation of mycotoxin's contamination in maize: A deep learning approach

Maize is the main staple food and feed inSub-Saharan African countries and is highly susceptible to mycotoxin contamination under opportune environmental conditions. The presence of mycotoxins in maize affects the health of consumers and impacts global trade. According to the literature, the lack of mycotoxin awareness and the existence of strategies that are labor- and cost-prohibitive have led to the ongoing mycotoxin contamination in maize. Therefore, this study developed a cost-effective deep learning-based mobile application for segmentation of mycotoxin contamination in maize; using the RESNET152 model with performance rates of accuracy, test accuracy, epochs, time used, loss and image size results at 99.5%, 99.9%, 40, 07:30 min, and 0.051; and 460 respectively and performance evaluation metrics of F1-Score and sensitivity 0.62 and 0.997 respectively. During, the development processes, a total of 4800 images were collected and augmented. Then, the resulting 9600 data points were randomly shuffled and then split into the ratio of 70%:20:10% for training, validation, and testing datasets in order to avoid overfitting and biases in the resulting model. Lastly, the average result of model validation was 89% which was conducted among the farmers in the Maize area, Maize entrepreneurs, ICT experts, decision-makers from the Government, and policymakers. Therefore, the study recommends the collection of quality data which can be in the form of images, satellite, and biochemical properties of mycotoxin in order to enable researchers to analyze the contamination of mycotoxin and its linkages with environmental factors such as weather, soil characteristics, geographical position, and other unexpected events.

Biofumigation for the Management of Fusarium graminearum in a Wheat-Maize Rotation.

Fusarium graminearum is the most important causal agent of head blight in wheat, and stalk and ear rot in maize. A field experiment was conducted to investigate the effect of incorporation of Brassicaceae cover crops on Fusarium graminearum in a wheat-maize rotation. Five species belonging to Brassicaceae (Brassica juncea, Eruca sativa, Raphanus sativus, B. carinata, B. oleracea var. caulorapa L.) were used in the field experiment to investigate their potential to suppress F. graminearum inoculum in soil, disease incidence in maize and to reduce subsequent mycotoxin contamination in maize. Brassica juncea was found to contain the highest glucosinolate concentration in shoots (31 µmol g-1). Severity of ear rot and stalk rot in maize was not significantly reduced in the amended plots. Incorporation of R. sativus 'Terranova' significantly decreased the amount of F. graminearum DNA by 58% compared with the cultivated fallow treatment, however the DNA concentration was not significantly different to fallow uncultivated. Fusarium graminearum DNA and deoxynivalenol in maize was 50% lower after incorporation of B. oleracea var. caulorapa L. compared to after fallow treatment but the difference was not significant. The brassica crops used in the present field experiment were not effective in suppressing F. graminearum, therefore further studies to optimise the current approach are recommended.

Harvest and post-harvest handling practices associated with fumonisin B1 contamination in maize (Zea mays L.): dietary exposure and risk characterization in eastern Ethiopia.

Maize is the main staple food crop in the eastern part of Ethiopia. However, maize loss is a major issue due to fungal contamination especially at the post-harvest stage owing to inadequate handling practices. This study aimed to assess post-harvest handling and awareness against fungal development and fumonisin B1 (FB1) in maize and to calculate risk exposures of FB1. A total of 197 maize samples (grain and flour) were collected from five districts (Haramaya, Kersa, Meta, Oda Bultum, and Tullo). FB1 was detected using LC-MS/MS qTRAP. Exposure assessment was done based on the maize consumption rate per day in Ethiopia for different age groups (infants, children, and adults). Risk characterization depends on the margin of exposure (MoE) combined with the lower confidence limit of the benchmark dose level (BMDL). About 81% of farmers were not physically separating undamaged maize ears with damaged from either birds or fungi. Moreover, 100% were not using improved storage material. In storage samples, FB1 was detected as high as 1058μg/kg ± 234 in the Kersa district while a minimum of 22.60μg/kg ± 5.27 in Meta. In flour samples, the maximum FB1 (327μg/kg) was detected from the Oda Bultum district. The maximum exposure of infants was estimated at Kersa (1131µg/kg bw/day), followed by Oda Bultum (1073µg/kg bw/day) and Haramaya (854µg/kg bw/day). Overall, FB1 exposures ranged from 6.09 to 1131µg/kg bw/day, which is 3 to 500µg/kg bw/day higher than the maximum tolerable daily intake of 2µg/kg bw/day recommended by the World Health Organization. The MoE ranged from 0.15 to 176, with infants being at higher risk than adults. The study highlights the urgent need to enhance growers' awareness and knowledge of good post-harvest practices to reduce mycotoxin contamination in maize. Further biomarker analysis must be pursued to determine the risk exposure assessment for different age groups in these areas with a priority for the Kersa district.

Influence of Agronomic Factors on Mycotoxin Contamination in Maize and Changes during a 10-Day Harvest-Till-Drying Simulation Period: A Different Perspective.

Agronomic factors can affect mycotoxin contamination of maize, one of the most produced cereals. Maize is usually harvested at 18% moisture, but it is not microbiologically stable until it reaches 14% moisture at the drying plants. We studied how three agronomic factors (crop diversification, tillage system and nitrogen fertilization rate) can affect fungal and mycotoxin contamination (deoxynivalenol and fumonisins B1 and B2) in maize at harvest. In addition, changes in maize during a simulated harvest-till-drying period were studied. DON content at harvest was higher for maize under intensive tillage than using direct drilling (2695 and 474 μg kg−1, respectively). We found two reasons for this: (i) soil crusting in intensive tillage plots caused the formation of pools of water that created high air humidity conditions, favouring the development of DON-producing moulds; (ii) the population of Lumbricus terrestris, an earthworm that would indirectly minimize fungal infection and mycotoxin production on maize kernels, is reduced in intensive tillage plots. Therefore, direct drilling is a better approach than intensive tillage for both preventing DON contamination and preserving soil quality. Concerning the simulated harvest-till-drying period, DON significantly increased between storage days 0 and 5. Water activity dropped on the 4th day, below the threshold for DON production (around 0.91). From our perspective, this study constitutes a step forward towards understanding the relationships between agronomic factors and mycotoxin contamination in maize, and towards improving food safety.

Pre-harvest and post-harvest farmer experiences and practices in five maize growing regions in Ghana.

Maize is a major staple crop mainly produced by smallholder farmers in developing nations. Grain losses happen in Sub-Saharan Africa, and therefore the objective of this study was to assess the different kinds of pre-harvest and post-harvest losses that maize farmers in Ghana encounter. The storage practices, and farmers' awareness and knowledge of mycotoxin contamination in maize were also assessed. The study area had five regions, and three districts per region. The study sites were selected purposefully because of the prior knowledge of farmers on maize production. A semi-structured questionnaire was used to collect the data, and a purposive sampling technique was used to select 75 maize farmers for the interview. The male maize farmers were many compared to females. Over 70% of farmers were at least 40 years. Over 50% of farmers had basic education except those in the northern region. Grain yields were generally low, and at least 60% of farmers experienced post-harvest loss. The period of grain storage and the storage techniques were the prerogatives of the farmers but largely dependent on farmers' financial status. Farmers basically used synthetic chemicals, and a few of the farmers decided to use plant materials during grain treatment.

Harvesting and phytosanitary parameters with particular regard to mycotoxin content of maize as a function of different seasonal, fertilisation and hybrid effect

The aim of our three consecutive years (2017–2019) field trial was to obtain information as to the effect of weather conditions of the actual year as well as to assess the impact of some technological parameters such as fertilisation, the choice on the hybrid type on the yield parameters, phytosanitary conditions and mycotoxin contamination of maize. According to our results, the climatic characteristics of the years, the examined hybrid characters (FAO 310 and 490) and the fact of N-fertilisation had significant effects on yield parameters and grain moisture content. The additional N-supply did not affect the development or severity of stem rot in any of the hybrid effects. In this respect, the year effect appeared to be the decisive factor since much higher stem rot values were recorded in the plots of the longer growing season hybrids. Among the mycotoxins examined, only zearalenone and fumonisin found in the harvest were significantly influenced by the effect of the year, the length of the growing season as well as nutrient replenishment. It can be stated that the applied technological parameters have a major effect on the expression of this toxin load in maize. Dry maize stocks that have lost their water in the vegetation are predisposing factors for toxin accumulation. N-content of soil and that of plants can play a different role in mycotoxin accumulation in maize plants.

Mycotoxin contamination of maize (Zea mays L.) samples in Hungary, 2012–2017

Mycotoxin contamination of maize often raises risks for human and animal health. The most frequently detected mycotoxins in maize are trichothecenes, fumonisins, and aflatoxin. A total number of 17,011 maize samples were tested by SGS for their mycotoxin content between 2012 and 2017. The toxin results clearly show that the southern areas of the country had higher levels of toxin contamination than the average. According to the dataset, aflatoxin contamination has become regular but the appearance of fumonisins was also more frequent. Deoxynivalenol toxin accumulation in crops can also reach dangerous levels under favorable ecological conditions. The fluctuation between years and regisons is decisively shaped by the weather conditions. However, the two pathogens with less virulence (Fusarium verticillioides and Aspergillus flavus) must be taken into account and the contribution of insect pests. 72.63% of the total fumonisin concentration was defined as fumonisin B1, 20.34% as fumonisin B2, and 7.03% as fumonisin B3. The correlations between the three fumonisins analogs were higly significant (P = 0.001), and correlation coefficient varied between 0.961 and 0.998 across the six years of evaluation. This is the first complex evaluation of deoxynivalenol, fumonisin, and aflatoxin contamination of maize samples in Hungary.

The East African Community (EAC) is a regional Inter-Governmental body comprising of Uganda, Kenya, Tanzania, Rwanda, Burundi, and South Sudan. This region produces and extensively consumes maize and peanuts as staple foods.

Impact of push–pull cropping system on pest management and occurrence of ear rots and mycotoxin contamination of maize in western Kenya

AbstractPush–pull involves intercropping of cereals with Desmodium as a “push” crop and planting Napier grass/Brachiaria as the “pull” crop at the border. The technology has been reported to effectively control stemborers, striga weed, and fall armyworm (FAW), and to improve soil nutrition, resulting in increased grain yield. This study evaluated the impact of stemborer and FAW management using this technology on incidence of maize ear rots and preharvest contamination of grains with aflatoxin and fumonisin in western Kenya. The study was conducted during three cropping seasons on maize grown under the push–pull system and as a monocrop. Incidence of stemborer and FAW damage was determined as percentage of damaged plants, while incidence of ear rots was determined as percentage of ears with symptoms. At harvest, fungi were isolated from kernels and aflatoxin and fumonisin were quantified using enzyme‐linked immunosorbent assay. Stemborer and FAW damage was significantly (p = .001) reduced by over 50% under the push–pull system. There was also a significant (p < .001) reduction in the incidence of Fusarium verticillioides (60%) and Aspergillus flavus (86%), which was reflected in a reduced incidence of ear rots (50%) with the push–pull system (p = .001). Fumonisin in maize from push‐pull farms was significantly (p = .048) reduced (39%) but the technology had no significant (p > .05) effect on aflatoxin. The study showed that push–pull is an effective strategy for managing maize ear rots and fumonisins, and therefore could play a role in improving food safety among smallholder maize farmers in the region.

Low-cost grain sorting technologies to reduce mycotoxin contamination in maize and groundnut

The widespread contamination of foods by mycotoxins continues to be a public health hazard in sub-Saharan Africa, with maize and groundnut being major sources of contamination. This study was undertaken to assess the hypothesis that grain sorting can be used to reduce mycotoxin contamination in grain lots by removing toxic kernels. We tested a set of sorting principles and methods for reducing mycotoxin levels in maize and groundnut from a variety of genotypes and environments. We found that kernel bulk density (KBD) and 100-kernel weight (HKW) were associated with the levels of aflatoxins (AF) and fumonisins (FUM) in maize grain. A low-cost sorter prototype (the ‘DropSort’ device) that separated maize grain based on KBD and HKW was more effective in reducing FUM than AF. We then evaluated the effectiveness of DropSorting when combined with either size or visual sorting. Size sorting followed by DropSorting was the fastest method for reducing FUM to under 2 ppm, but was not effective in reducing AF levels in maize grain to under 20 ppb, especially for heavily AF-contaminated grain. Analysis of individual kernels showed that high -AF maize kernels had lower weight, volume, density, length, and width and higher sphericity than those with low AF. Single kernel weight was the most significant predictor of AF concentration. The DropSort excluded kernels with lower single kernel weight, volume, width, depth, and sphericity. We also found that visual sorting and bright greenish-yellow fluorescence sorting of maize single kernels were successful in separating kernels based on AF levels. For groundnut, the DropSort grouped grain based on HKW and did not significantly reduce AF concentrations, whereas size sorting and visual sorting were much more effective.

Fusarium species and mycotoxin contamination in maize in Buenos Aires province, Argentina

Maize is one of the most important crops in Argentina. This crop can be affected by the presence of different Fusarium species, causing two distinct diseases known as Fusarium ear rot and Gibberella ear rot. In this study, a total of 90 samples from commercial fields of Buenos Aires province in Argentina were collected during the 2015, 2016 and 2017 harvest seasons with the aim to evaluate Fusarium species presence, mycotoxin content and to establish a relationship between the main Fusarium mycotoxins infecting maize grains and the environmental conditions. The results showed that F. verticillioides was the most isolated species followed by F. subglutinans, species of Fusarium graminearum species complex, F. proliferatum, and F. cerealis varying their presence according to the harvest season evaluated. Regarding mycotoxins, deoxynivalenol and 3-acetyl-deoxynivalenol showed content from 281.26 ± 22.51 to 359.02 ± 42.57 μg/kg in the 3 yrs evaluated, while 15-acetyl-deoxynivalenol, nivalenol, and zearalenone were not found. With regard to B1 and B2 fumonisins, these mycotoxins were detected below the detection limit of 0.3 μg/kg in some maize samples in the 3 yrs. The selection of maize genotypes, cultural practices, and climatic conditions could be responsible for the Fusarium species found and their respective mycotoxin production. Due to worldwide concern about food security and the potential risk the global climate change could impart, it will be necessary to develop predictive models based on meteorological conditions to determine the risk of Fusarium presence and mycotoxin production in maize similar to those developed in other crops.

Occurrence of Maize Ear Rot Caused by Fusarium fujikuroi in China

Maize (Zea mays L.) is one of the most important crops in China. Ear rot is a serious disease that often leads to a decline in maize yield and quality and is a hazard to human and livestock health owing to potential mycotoxin contamination of grain. The incidence of maize ear rot usually ranges from 20 to 40% at different fields, even reaching as high as 70% at certain plots in Chongqing and surrounding areas (Zhou et al. 2018). In August 2016, a survey to determine population composition and distribution of Fusarium spp. causing maize ear rot was conducted in 22 counties in Chongqing. Maize ears with white, pink, or salmon-colored mold were collected from fields for further analysis. Symptomatic kernels were soaked in 2% sodium hypochlorite solution for 3 min, rinsed with sterile water three times, placed on acidified potato dextrose agar (PDA), and incubated at 26 ± 1°C for 3 to 5 days. The newly grown-out fungal colonies displaying morphological characteristics of Fusarium spp. were transferred onto fresh PDA and Spezieller Nahrstoffarmer agar and purified by the single-spore isolation method (Yang et al. 2008). Fusarium spp. were identified based on morphological characteristics (Leslie and Summerell 2006) and sequence analysis of the translation elongation factor-1α (TEF-1α), β-tubulin, and mitochondrial small subunit (mtSSU) genes (Jiang et al. 2018). The predominant Fusarium spp. were Fusarium verticillioides, F. proliferatum, and F. meridionale, which were the main causal agents of maize ear rot in these areas. In addition, morphological characteristics of several isolates from Beibei, Jiangjin, Xiushan, Rongchang, Kaixian, and Wanzhou Districts were found to be identical to those of Fusarium fujikuroi Nirenberg, which grouped into the Gibberella fujikuroi species complex. Colonies on PDA produced abundant white aerial mycelia initially, but the mycelia became pale pinkish and dark violet with age. Microconidia were abundant and small in size (4.9 to 11.3 × 2.2 to 3.2 µm), colorless, oval to club shaped, and 0- to 1-septate. Macroconidia were slender, almost straight, sickle shaped, medium length, with three to five septa (17.1 to 39.2 × 2.6 to 3.3 µm). No chlamydospores were observed. Identity of the fungus was also confirmed by sequence comparison of the partial TEF-1α, β-tubulin, and mtSSU genes of two representative isolates. The resulting sequences of the TEF-1α, β-tubulin, and mtSSU genes showed 100, 100, and 99% sequence identity to those of F. fujikuroi (GenBank accession nos. MH263736.1, MH263737.1, and MF984420.1, respectively). DNA sequences of partial TEF-1α, β-tubulin, and mtSSU genes from two representative isolates (FCQD90 and FCQD97) were deposited in GenBank under accession numbers MK328875 and MK328876, MK385626 and MK385627, and MK385628 and MK385629, respectively. A pathogenicity test was performed on maize inbred lines B73 and Ye 107. Four days after silk emergence, 2 ml of conidial suspension (1 × 10⁶ conidia /ml) of each isolate was injected into each of 10 maize ears through the silk channel. Control plants were inoculated with sterile distilled water. The typical Fusarium ear rot symptom (white or white-pink mold) was observed on inoculated ears and the infected kernels. No symptom developed on the water-inoculated controls. F. fujikuroi was reisolated from the symptomatic ears but not from the control. F. fujikuroi was previously reported associated with seedling damping-off on soybean, stem wilt of Canna edulis, and root rot of Reineckia carnea (Jiang et al. 2018; Pedrozo et al. 2015; Sun et al. 2018). To our knowledge, this is the first report of F. fujikuroi causing maize ear rot in China, and this disease should be paid sufficient attention owing to a serious risk of mycotoxin contamination in maize.
