Expression of IGF-II/Man-6-P receptors on rat, rabbit, and human colon epithelial cells

Previous experiments from this laboratory have established the presence of receptors for insulin and insulin-like growth factor I (IGF-I) on apical membranes prepared from rabbit colon epithelial cells; however, no receptors for multiplication-stimulating activity (MSA), the rat peptide hormone equivalent of human IGF-II, were found in this tissue. In the current studies, radioligand binding assays, covalent cross-linking experiments, and immunoblot analyses using a polyclonal rabbit antiserum that recognizes the IGF-II/mannose 6-phosphate (Man-6-P) receptor, all confirmed the presence of IGF-II/Man-6-P receptors on membranes prepared from rat and human colon epithelial cells. Exposure of rat colon epithelial cell membrane fractions to 5 mM Man-6-P before incubation with 125I-labeled IGF-II increased radioligand binding. Immunoblot analysis indicated that IGF-II/Man-6-P receptors were present in both unfractionated rat colon membranes and fractions enriched with apical membranes. Rabbit and human colon epithelial cells displayed a different pattern of receptor distribution than rat colon epithelial cells, with more insulin receptors but relatively few IGF-II/Man-6-P receptors. Immunohistochemical studies using a rabbit polyclonal antiserum confirmed that IGF-II/Man-6-P receptors were present on both the apical and the basolateral surfaces of colon epithelial cells.

Stimulation of DNA synthesis in rat uterine cells by growth factors and uterine extracts

Replicative DNA synthesis, as measured by thymidine incorporation, has been measured in rat uterine cells in primary culture in response to growth factors. Insulin, insulin-like growth factor-I (IGF-I), multiplication-stimulating activity (MSA) and platelet-derived growth factor (PDGF) stimulated DNA synthesis, while estradiol, epidermal growth factor (EGF), transforming growth factor-β (TGF-β), basic fibroblast growth factor (bFGF) and relaxin did not stimulate or did so weakly and only at very high concentrations. Uterine acid extracts also stimulated DNA synthesis. IGF-I stimulated at concentrations consistent with its acting through the IGF-I receptor; however, insulin stimulated at concentrations higher than expected for its acting through its receptor and thus its action may be mediated through the IGF-I receptor. IGF-I was found in uterine tissue by radioimmunoassay (RIA). There was a 5- to 10-fold increase in IGF-I in the uteri from ovariectomized rats that had been treated with estradiol 24 h earlier. This is analogous to the increase in growth factor activity found previously in rat uterus after 24-h estradiol treatment (Beck, C.A. and Garner, C.W. (1989) Mol. Cell. Endocrinol. 63, 93–101). These data are consistent with the hypothesis that estradiol effects in the uterus are in part mediated through IGF-I.

Intrinsic Bioactivity of Insulin-Like Growth Factor-Binding Proteins from Vascular Endothelial Cells*

Conditioned medium from cultured vascular endothelial cells contains material capable of stimulating acute metabolic processes in endothelial cells. The bioactivity of the conditioned medium is not caused by the copurification of known growth factors produced by the cells, in particular platelet-derived growth factor, basic fibroblast growth factor, or insulin-like growth factor (IGF)-I/II. We now demonstrate that the bioactivity is directly due to an IGF-binding protein(s) (ECBP) and, further, that the bioactive domain of the binding protein differs from the IGF-binding domain. Binding proteins (BPs) from cultured pulmonary artery endothelial cells were purified by sequential passage over sizing, multiplication-stimulating activity affinity, and hydrophobic columns. BP fractions were separated into those with and those without biological activity. The bioactive binding protein(s) was cross-linked with disuccinimidyl suberate to IGF-I or the recombinant IGF analog [1-27,Gly4,38-70]IGF-I (Analog). The IGF-I Analog, by itself, had minimal interaction with the type I IGF receptor in cultured microvessel endothelial cells and no intrinsic bioactivity, but did bind with high affinity to ECBP. All free BP and free IGF-I/Analog were removed from the cross-linked mixture by passage over gel filtration and IGF affinity columns. The cross-linked BP-IGF-I complex did not bind to the type I receptor of cultured endothelial cells, but did stimulate glucose and alpha-aminoisobutyric acid uptake in endothelial cells (approximately 2-fold increase); the magnitude of the response was nearly equal to the effect of ECBP or IGF-I alone. The BP-Analog complex also stimulated glucose and alpha-aminoisobutyric acid uptake, with the magnitude of the response approaching the effect of ECBP alone. The BP-Analog complex also did not react with type I IGF receptors on the cultured endothelial cells. We conclude 1) IGF-BP produced by endothelial cells possess intrinsic biological activity; 2) bioactivity of the BP(s) is retained when the IGF-binding domain of the BP is occupied by IGF-I or an inactive IGF-I analog; and 3) IGF-I bound to the bioactive BP does not react with its receptor and possesses minimal, if any, bioactivity in vitro.

Expression of Atypical and Classical Insulin Receptors in Chinese Hamster Ovary Cells Transfected with Cloned cDNA for the Human Insulin Receptor*

Human placenta and IM-9 lymphocytes contain subpopulations of atypical insulin receptors which differ from classical insulin receptors in their higher binding affinity for insulin-like growth factors I and II (IGF-I and IGF-II). Both types of insulin receptors may be derived from different but related genes, or may represent alternative post-translational modifications of the same gene product. To test these possibilities, we have examined the IGF binding characteristics of the human insulin receptors expressed in Chinese hamster ovary (CHO) cells which had been stably transfected with cloned human insulin receptor cDNA (CHO-T cells). The parent CHO cells contained 3 x 10(3) rodent insulin receptors/cell, and the CHO-T cells, 2.0 x 10(6) human insulin receptors/cell. Competition binding studies showed that the binding of [125I]IGF-I and [125I]multiplication stimulating activity (MSA/rat IGF-II) to parent CHO cells was primarily to type I and II IGF receptors, which cross-react poorly or not at all with insulin. However, competition binding studies with CHO-T cells showed that [125I]IGF-I binding was displaced 60-70%, and [125I]MSA binding, 50-55%, by low concentrations of insulin (20 ng/ml) and no further by higher concentrations of insulin (500 ng/ml). The insulin-insensitive IGF binding sites corresponded to the rodent type I and II IGF receptors; the insulin-sensitive IGF binding sensitive sites resembled the human atypical insulin receptors in that they bound IGF-I and MSA with moderately high affinity and reacted with insulin, MSA, and IGF-I in that order of potency. Atypical insulin receptors were also demonstrated by insulin-sensitive [125I]IGF-I and [125I]MSA binding to solubilized CHO-T proteins adsorbed to microtiter wells coated with monoclonal antibodies specific for the human insulin receptor. These results suggest that atypical human insulin receptors are generated by differential post-translational processing of the same gene product as classical human insulin receptors.

Polypeptide growth factors in metanephric growth and segmental nephron differentiation

Although the developing nephron expresses receptors for various polypeptide growth factors, the specific roles of such factors in renal organogenesis are unknown. Therefore, the effects of epidermal growth factor (EGF) (8.2 x 10(-11) M-1.6 x 10(-8) M), multiplication stimulating activity (MSA) (6.6 x 10(-10) M-1.3 x 10(-8) M) and transforming growth factor beta (TGF-beta) (1 x 10(-12) M-1 x 10(-9) M) on organotypic renal growth and segmental nephron differentiation were studied in a serum-free hormone-supplemented, murine metanephric organ culture system. Following culture in control or growth-factor-supplemented medium, explant growth was assessed, and explant growth and differentiation were determined morphometrically in four defined neprhon segments which were identified morphologically or immunohistologically with segment-specific antibodies and/or lectins: glomeruli, proximal tubules, thick ascending limb-early distal tubules, and collecting tubules. Results showed that EGF increased overall renal growth and specific differentiation of distal elements, but retarded differentiation of glomeruli and proximal tubules. EGF also induced hyperplastic cystic malformation in proximal tubules. MSA stimulated explant growth and promoted segmental differentiation of all tubular segments. TGF-beta globally retarded in vitro nephrogenesis. Such data demonstrate that polypeptide growth factors have multiple and often disparate effects on overall renal growth in relation to differentiation of discrete nephron segments and provide insight into the factors which may regulate normal and abnormal renal embryogenesis.

Autocrine Regulation of Human Tumor Cell Proliferation by Insulin-Like Growth Factor II: anin-VitroModel*

We have shown that a pleomorphic cell line of abnormal human karyotype derived from a stomach carcinoma (LIM-1839) proliferates in serum-free medium, expresses insulin-like growth factor II (IGF-II) mRNA, and secretes IGF-II (up to 56 ng/ml in serum-free conditioned medium, as measured in a rat liver RRA. No detectable levels of IGF-I can be measured in serum-free conditioned medium by RIA. These cells also secrete IGF-binding proteins, detected by a charcoal adsorption assay. The release of IGF-II and IGF binding proteins into serum-free conditioned medium (1.7 pmol/10(6) cells.24 h and 0.8 pmol binding sites/10(6) cells.24 h for 3 days, respectively) is inhibited 80% by cycloheximide (10 micrograms/ml). The LIM-1839 cells have type I and type II IGF receptors, determined by affinity cross-linking and competition binding studies. These cells proliferated 1.6-fold over 4 days in serum-free medium, with fresh medium changes on days 0 and 2: their growth was inhibited 56% by 40 micrograms/ml Sm 1.2, a monoclonal antibody which recognizes IGF-I and IGF-II. The addition of 20 and 50 ng/ml multiplication stimulating activity (rat IGF-II) caused 1.8- and 1.7-fold increases in cell growth between days 0 and 4 compared to controls, while [Thr59]IGF-I, at 20 and 50 ng/ml, caused 1.6- and 2.0-fold increases. Insulin, at 2 and 10 micrograms/ml, had no significant effect. The stimulatory effects of endogenous and exogenous IGFs on LIM-1839 cell proliferation were inhibited by a monoclonal antibody to the type I IGF receptor, alpha IR-3. These results suggest that the LIM-1839 cells are biologically responsive to endogenously produced IGF-II, and may thereby provide an in vitro model for autocrine regulation of human tumor growth by IGF-II.

1,25-Dihydroxyvitamin D3[1,25-(OH)2D3] Increases Insulin-Like Growth Factor I (IGF-I) Receptors in Clonal Osteoblastic Cells. Study on Interaction of IGF-I and 1,25-(OH)2D3*

We previously reported a cooperative effect between insulin-like growth factor I (IGF-I) and 1,25-dihydroxy-vitamin D3 [1,25-(OH)2D3] in murine clonal osteoblastic cells, MCT3T3-E1. In the present study, the possible mechanism of interaction between these hormones was investigated. The effect of IGF-I on 1,25-(OH)2D3 receptors in MC3T3-E1 cells was examined. The affinity and hormone binding capacity of 1,25-(OH)2D3 receptors were not altered by IGF-I. Immunoblot analysis showed about 54 kilodaltons (kDa) 1,25-(OH)2D3 receptors, similar to that observed for mouse fibroblasts. The synthesis of IGF-I by the cells under a serum-free condition was determined by RIA. The assay revealed immunoreactive IGF-I secreted by MC3T3-E1 cells (1.79 +/- 0.04 x 10(-9) M, mean +/- SE, n = 5). Rat GH significantly increased the concentration of IGF-I, but 1,25-(OH)2D3 did not. IGF-I radioligand-receptor assay revealed specific binding of IGF-I to MC3T3-E1 cells. The relative potency of IGF-I-related peptides to bind with the cells was in the order of IGF-I much greater than multiplication-stimulating activity (the rat homologue of IGF-II) greater than insulin, and the receptor protein migrated as a 130-kDa band in autoradiography. Scatchard analysis showed a significant increase in IGF-I binding sites by 50% after 3-day treatment with 5 x 10(-11) M 1,25-(OH)2D3, without any change in affinity. These results indicate that the interaction of IGF-I and 1,25-(OH)2D3 in the culture of MC3T3-E1 cells may be mediated by the effect of 1,25-(OH)2D3 on IGF-I receptors.

Insulin-like growth factor binding to the atypical insulin receptors of a human lymphoid-derived cell line (IM-9)

The cells of the IM-9 human lymphocyte-derived line contain a sub-population of insulin-binding sites whose immunological and hormone-binding characteristics closely resemble those of the atypical insulin-binding sites of human placenta. These binding sites, which have moderately high affinity for multiplication-stimulating activity [MSA, the rat homologue of insulin-like growth factor (IGF) II] and IGF-I, are identified on IM-9 cells by 125I-MSA binding. They account for approximately 30% of the total insulin-receptor population, and do not react with a monoclonal antibody to the type I IGF receptor (alpha IR-3). The relative concentrations of unlabelled insulin, MSA and IGF-I required to displace 50% of 125I-MSA from these binding sites (1:4.7:29 respectively) are maintained for cells, particulate membranes, Triton-solubilized membranes precipitated either by poly(ethylene glycol) or a polyclonal antibody (B-10) to the insulin receptor, and receptors purified by insulin affinity chromatography. Because the atypical insulin/MSA-binding sites outnumber the type I IGF receptors in IM-9 cells by approximately 10-fold, they also compete with the latter receptors for 125I-IGF-I binding. Thus 125I-IGF-I binding to IM-9 cells is inhibited by moderately low concentrations of insulin (relative potency ratios for insulin compared with IGF-I are approx. 1/14 to 1/4) and is partially displaced (65-80%) by alpha IR-3. When type I IGF receptors are blocked by alpha IR-3 or removed by B-10 immunoprecipitation or insulin affinity chromatography, the hormone-displacement patterns for 125I-IGF-I binding resemble those of the atypical insulin/MSA-binding sites.

Characterization of the Change in Type I and II Insulin-Like Growth Factor Receptors of Bovine Mammary Tissue during the Pre- and Postpartum Periods*

Type I and II receptors for the insulin-like growth factors (IGF) were characterized in microsomes from bovine mammary tissue. Specific binding of [125I]IGF-I increased linearly with increasing microsomal protein concentrations. In contrast, IGF-II binding showed a curvilinear relationship over the concentration range tested, with a maximum at 120 micrograms/ml. Kinetic studies conducted at 4 C showed the binding reactions to be reversible. Competition studies showed recombinant human IGF-I (rh-IGF-I) to be 8-, 18-, and 1000-fold more potent at inhibiting [125I]rh-IGF-I binding than recombinant bovine IGF-II (rb-IGF-II), multiplication-stimulating activity, and insulin, respectively. rbIGF-II was 1.8- and 6-fold more potent at inhibiting [125I]rbIGF-II binding than rhIGF-II and multiplication-stimulating activity. Specific binding of [125I]IGF-I and -II was measured in microsomes prepared from cows (n = 47) ranging from 138 days prepartum to 411 days postpartum. IGF-I binding declined during the prepartum period, increased 75% with the onset of lactation, and then declined during the postpartum period. Scatchard analysis showed the presence of a single class of high affinity binding sites for both ligands, with type II receptors being about 10-fold more prevalent than type I receptors. The survey and Scatchard data support the conclusion that the onset of lactation coincides with an increase in the number of type I receptors present in mammary tissue. This increase supports the contention that IGF-I may play an important role in modulating the metabolic activity of the bovine mammary gland.

Insulin-Like Growth Factors and the Developing and Mature Rat Small Intestine: Receptors and Biological Actions

To determine if insulin-like growth factor-I (IGF-I) or multiplication stimulating activity (MSA, rat IGF-II) might directly influence small intestinal epithelium, we studied the distribution of IGF binding sites during development of the rat intestine. Cell membranes from suckling rat mucosa bound 2-6 times as much 125I-IGF-I and 3-5 times as much 125I-MSA as did adult membranes. Isolated villus cells from suckling and adult rats specifically bound both IGFs. IGF-I binding tended to remain high during suckling, whereas MSA binding fell progressively from the early suckling period. Competitive displacement studies with insulin, IGF-I and MSA demonstrated the presence of type-I and type-II IGF receptors. In vitro autoradiography of 125I-IGF-I binding sites in adult and suckling rat jejunum showed highest binding in the submucosa with extensions up into the lamina propria. Immunocytochemical localization of type-II receptors showed highest density in villus epithelium and vessel walls. Administration of MSA by oral and IGF-I by oral and parenteral routes (1 microgram/day for 6 days) to suckling rats stimulated jejunal brush border enzymes, but not intestinal growth. Developmental changes in receptor density and effects on brush border enzymes suggest a specific role for IGFs in post-natal development of the rat intestine.

Insulin-Like Growth Factor-Binding Proteins from Vascular Endothelial Cells: Purification, Characterization, and Intrinsic Biological Activities*

Insulin-like growth factor (IGF)-binding proteins are produced by several cell types, including vascular endothelial cells. The production of IGF-binding proteins by endothelial cells is of particular interest, since these cells are directly bathed by the circulating IGFs and form the initial barrier to the passage of circulating IGFs from the bloodstream to subendothelial tissues. We have purified IGF-binding proteins from medium conditioned by cultured bovine endothelial cells by sequential passage over Bio-Gel P-60, multiplication-stimulating activity affinity, anion exchange (DEAE cellulose) and/or hydrophobic (phenyl-Sepharose) chromatography. Two peaks of IGF-binding activity were eluted from the phenyl-Sepharose column. After cross-linking, each peak contained two to five protein bands on gels that specifically bound IGF-I and -II with mol wt ranging from about 28-44K. Material in peak 1 bound IGF-I congruent to IGF-II and had no affinity for insulin and proinsulin. Peak 2 IGF-binding proteins bound IGF-II with substantially higher affinity than IGF-I and did not recognize insulin or proinsulin. Peak 1 material from phenyl-Sepharose chromatography was a potent stimulator of both glucose transport and aminoisobutyric acid (AIB) uptake in microvessel endothelial cells, with maximal stimulation of both processes being 300-400% of control values. In contrast, peak 2 material either had no intrinsic bioactivity or was slightly inhibitory or stimulatory, depending on the concentration of peak 2 material that was added. The bioactivity in peak 1 was not due to copurification of other endothelial proteins capable of stimulating glucose and AIB uptake, such as IGF-I/-II, platelet-derived growth factor, and basic fibroblast growth factor, since bioactivity was retained after acid treatment, antibody neutralization, and selective affinity chromatography to deplete these other factors. When peak 1 material was added to IGF-I the bioeffects (glucose and AIB uptake) of IGF and binding proteins were additive, and in some experiments the binding proteins potentiated the effect of IGF on endothelial cells, suggesting that the binding protein-IGF complex may retain the bioactivity of both the binding protein(s) and the IGF.

Effects of insulin-like growth factors I and II on growth and differentiation of transplanted rat embryos and fetal tissues.

The objective of our study was to obtain comparative information on the roles of insulin-like growth factors (IGFs) I and II in the growth and differentiation of rat embryos and fetuses in an in vivo experimental system. Ten-day-old rat embryos or 16-day-old fetal rat paws were transplanted under the capsule of both kidneys of syngeneic hosts, where they grew rapidly, and tissue differentiation occurred in an essentially normal manner. Infusion of a rabbit antiserum to human (h) IGF-I for 6 days into the renal artery of one kidney caused only a slight inhibition (approximately 20%) of growth of transplanted embryos on that kidney. However, 9 days of treatment with the antiserum caused highly significant growth retardation of transplanted embryos by 40-45%. Exposure of fetal paws to the same antiserum for 9 days inhibited their growth by 27%. Infusion of synthetic hIGF-II or partially purified rat IGF-II [(MSA)multiplication-stimulating activity], for 6 days stimulated the growth of embryo transplants directly, and MSA was more effective than hIGF-II. By contrast, infusion of purified natural hIGF-I did not promote their growth. Growth of fetal paw transplants in hypophysectomized (HX) hosts was reduced by 70% compared to that in intact hosts. In the HX hosts infusion of the MSA was more effective than the hIGF-I at promoting the growth of paw transplants. Both MSA and IGF-I promoted ossification in the fetal paws in the HX hosts. These results suggest that both IGFs could be involved in regulating the growth and differentiation in prenatal rats, and IGF-II may be the more effective of the two factors as a growth-promoting agent in utero.

Schistosoma mansoni: Effect of Transferrin and Growth Factors on Development of Schistosomula In vitro

We have studied the role of human transferrin and of exogenous iron on early growth and development of schistosomula of Schistosoma mansoni in vitro, as determined by developmental achievement and thymidine incorporation. To help define an adequate serumless medium we also utilized fibroblast growth factor, platelet-derived growth factor, epidermal growth factor, and multiplication-stimulating activity, all present in the hepatic environment of developing schistosomula, as well as certain commercial serum substitutes. Supplementation of basal medium with transferrin in the range of 250-1,000 micrograms/ml provided development equal or superior to serum-supplemented medium, at least within the first 2 wk of culture. Serum-free medium supplemented with Nutridoma-HU or -SP stimulated development to a lesser extent. The stimulatory effect of iron (as ferric sulfate) on thymidine incorporation was removed by deferroxamine, a chelating agent. Competitive inhibition studies with 125I-labeled and unlabeled transferrin indicated the presence of nonsaturable binding sites on schistosomula.

Stimulation of human synovial fibroblast DNA synthesis by platelet-derived growth factor and fibroblast growth factor. Differences to the activation by IL-1.

The pronounced synovial hyperplasia often found in the joints of patients with rheumatoid arthritis could be explained partially by the action of monocyte-macrophage polypeptides (monokines). This report demonstrates that two cytokines which may be derived from monocyte-macrophage populations, namely platelet-derived growth factor (PDGF) and basic fibroblast growth factor (bFGF), stimulate the DNA synthesis and proliferation of human synovial fibroblast-like cells cultured in low (i.e., 1%) fetal bovine serum. Epidermal growth factor, insulin-like growth factor-I, insulin-like growth factor-II (multiplication stimulating activity) and substance P were inactive. Unlike IL-1, PDGF and FGF do not also stimulate PGE2, plasminogen activator, and hyaluronic acid levels. Thus PDGF and FGF, arising from stimulated monocyte-macrophages, may play a role in the stimulation of mesenchymal cell proliferation that often accompanies chronic inflammatory arthritic disease. The synovial cells respond to a variety of cytokines in different ways suggesting multiple-signaling pathways.

The binding characteristics and biological effects in FRTL5 cells of placental protein-12, an insulin-like growth factor-binding protein purified from human amniotic fluid.

We have studied the binding of recombinant human insulin-like growth factor I (hIGF-I), and hIGF-II, and rat IGF-II [rIGF-II (multiplication-stimulating activity)] to the human amniotic fluid IGF-binding protein placental protein-12 (PP12). PP12 displayed a 5- to 10-fold higher affinity for IGF-I compared to hIGF-II or rIGF-II. These differences in binding affinity were confirmed by both saturation binding analysis and competitive binding analysis using 125I-labeled IGF-I, hIGF-II, and rIGF-II and each of the unlabeled ligands. PP12 produced dose-dependent inhibition of IGF-I-stimulated [3H]thymidine incorporation in the rat thyroid follicular cell line FRTL5. Inhibition of IGF-I-stimulated thymidine incorporation paralleled the ability of PP12 to inhibit IGF-I binding to the surface of FRTL5. At a high concentration, PP12 also inhibited TSH-stimulated DNA synthesis but did not inhibit the binding of 125I-labeled TSH to FRTL5. Insulin did not inhibit the binding of 125I-labeled IGFs to PP12, and PP12 did not inhibit the ability of insulin to stimulate DNA synthesis. These data suggest that the ability of PP12 to inhibit TSH-stimulated DNA synthesis is through the inactivation of IGF produced endogenously by FRTL5. Low concentrations of PP12 produced a statistically significant enhancement of TSH-stimulated DNA synthesis; the mechanism by which this occurs remains unclear.

A monoclonal antibody to human insulin-like growth factor-I: characterization, use in radioimmunoassay and effect on the biological activities of the growth factor

A monoclonal antibody (BPL-M23) to insulin-like growth factor-I (IGF-I) was obtained following immunization of BALB/c mice with human IGF-I conjugated to ovalbumin. The affinity constant of BPL-M23 for IGF-I was 10.5 litres/nmol and the cross-reactivities of IGF-II, multiplication-stimulating activity III-2 and insulin were 0.8, 0.03 and less than 0.0001% respectively. Porcine, bovine, ovine and rabbit sera, but not rat or mouse sera, showed substantial reactivity with the antibody. Comparison of radioimmunoassay analyses of 54 human serum samples from normal subjects and acromegalic and GH-deficient patients using BPL-M23 and a polyclonal rabbit antiserum (R557A) to human IGF-I showed a high correlation, indicating the usefulness of the monoclonal antibody in radioimmunoassay. Monoclonal antibody BPL-M23 was capable of abolishing the sulphation, mitogenic and insulin-like activities of IGF-I in in-vitro bioassays, suggesting that these activities may rely upon the same receptor-binding site which is near to the antibody-binding site.

Identification of a Family of Insulin-Like Growth Factor II Secreted by Cultured Rat Epithelial-Like Cell Line 18,54-SF: Application of a Monoclonal Antibody

Somatomedin/insulin-like growth factor (IGF)-like polypeptides (designated SMP) were purified from the serum-free conditioned medium of cultured rat epithelial-like cells, 18,54-SF. A monoclonal antibody (MAb) was produced against partially purified SMP. The antibody was immunoglobulin G1 relatively specific for multiplication-stimulating activity III-2 (rat IGF-II), with a Kd value of 5.6 X 10(-9) M. The antibody showed 100% cross-reactivity with human IGF-II and 10% cross-reactivity with human IGF-I, but did not cross-react with insulin. For purification of SMP, therefore, immunoaffinity chromatography on Sepharose coupled with the MAb was used besides a procedure including ion exchange chromatography, gel filtration, and reverse phase HPLC. The purified SMP (at least five polypeptides) each gave a single peak on reverse phase HPLC and appeared as a single band with an apparent mol wt of 5000-8000 on sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The major components of SMP (designated HP1-SMP and HP3-SMP), which were purified about 100-fold from conditioned medium, stimulated DNA synthesis in human fibroblasts in culture and sulfation in chick embryonic cartilage in culture. These polypeptides showed almost the same cross-reactivity as multiplication-stimulating activity III-2 on RIA with the MAb. The partial amino acid sequences of HP1- and HP3-SMP were determined, and these polypeptides were identified with rat IGF-II.

Growth promoting effects of IGF-I on fetal hypothalamic cell lines under serum-free culture conditions

Recent evidence indicates that the insulin-like family of peptides may act as endogenous trophic factors in the central nervous system. To further examine this possibility we have investigated the effects of three insulin-like peptides on thein vitro growth of fetal hypothalamic cell lines. Two virally transformed rat hypothalamic cell lines which have been developed in our laboratory (A-6 and F-12) were used. Cells were plated at varying densities and cultured in the presence or absence of either insulin-like growth factor I (IGF-I), insulin, or multiplication stimulating activity (MSA or IGF-II), in serum-free medium for 1 wk. Cell growth was assessed by counting or by measuring cellular incorporation of3H-thymidine. Of the three peptides tested IGF-I was the most potent in eliciting cell growth. Insulin also stimulated growth of both cell lines, but was 100 times less potent for A-6 cells while it was equipotent with IGF-I in F-12 cells. MSA had no effect on either cell line. Both IGF-I and insulin showed dose-response effects in increasing cell growth. We also found that the two cell lines had the greatest response to IGF-I at low cell densities. Finally, time-course experiments suggested that a continued presence of the peptide is essential for the growth-promoting effects. We conclude that IGF-I is a potent growth factor for virally transformed cell lines derived from the rat fetal hypothalamus. Since both IGF-I immunoreactivity and IGF-I receptors have been located in this diencephalic area these results suggest that IGF-I may constitute a mitogenic signal for hypothalamic cells during neurogenesis.

Growth requirements, growth factor responsiveness, and growth factor secretion of three human embryonal carcinoma cell lines.

The in vitro growth requirements of three human embryonal carcinoma cell lines (H 12.7, 2102 EP, 1428 A) were investigated. The basal medium DME/F12 supplemented with insulin, transferrin, and low-density and high-density lipoproteins was sufficient to support substantial multiplication of all three lines. The most efficient attachment factor was either fibronectin (for 2102 EP and 1428 A) or collagen type I (H 12.7). In a serum-free system the influence of epidermal growth factor (EGF), insulin-like growth factor I, multiplication stimulating activity (MSA), a platelet extract, and the glucocorticoids dexamethasone and hydrocortisone, as determined by the DNA synthesis rate of the cells, was generally minimal. However, the DNA synthesis rate of cell lines H 12.7 and 2102 EP was increased by MSA, and the line with the highest potential to differentiate (H 12.7) was stimulated by EGF. All three cell lines secreted growth factors in a heterologous stimulation assay. Insulin-like growth factors I and II were not part of the growth promoting activity. The inhibitory effect of a monoclonal anti-EGF antibody on the 3H-thymidine incorporation of cell line 2102 EP might indicate autocrine secretion of EGF or an EGF-like factor by this cell line.

Demonstration of type I and type II somatomedin receptors on bovine growth plate chondrocytes.

The chondrocytes of the epiphyseal growth plate are the presumed target cells for hormones regulating skeletal growth. The somatomedins, a family of low molecular weight peptides, are thought to play a stimulatory role in this regulation. The cellular actions of the somatomedins are themselves determined by binding to specific receptors on target cells. Previous studies have characterized a specific receptor for somatomedin-C (Sm-C) or insulin-like growth factor I (IGF-I) on bovine growth plate chondrocytes (GPCs). We now report the characterization of a second type of somatomedin receptor on these cells that is more specific for another class of somatomedin represented by multiplication-stimulating activity (MSA) or rat insulin-like growth factor II (rIGF-II). Binding of [125I]MSA/rIGF-II to isolated GPCs was time dependent and saturable. Unlabeled Mr 7,100 MSA/rIGF-II and Sm-C/IGF-I were approximately equipotent in competing with [125I]MSA/rIGF-II for binding, while Mr 8,600 MSA/rIGF-II was an order of magnitude less potent. Low levels of competition by insulin appeared in some studies at concentrations of 10(-7) M and higher, suggesting displacement of [125I]MSA/rIGF-II binding to the Sm-C/IGF-I receptor. In affinity-labeling studies, [125I]MSA/rIGF-I labeled a complex of Mr greater than 300,000 (unreduced) and of Mr 140,000 (reduced), consistent with a type I somatomedin receptor composed of disulfide-linked subunits. [125I]MSA/rIGF-II labeled a Mr 240,000 moiety (unreduced) and Mr 260,000 (reduced), consistent with a type II somatomedin receptor. Both affinity-labeling and kinetic data revealed cross-binding of MSA/rIGF-II and insulin with the type I receptor and of Sm-C/IGF-I with the type II receptor. In contrast, the type II receptor did not recognize insulin. These data suggest a complex pattern of graded specificity of these receptors for their ligands. These data are consistent with the hypothesis that IGF-II as well as Sm-C/IGF-I participate in the stimulation of skeletal growth.
