Mangrove sediments-associated bacterium (Bacillus sp. SW7) with multiple plant growth-promoting traits promotes the growth of tomato (Solanum Lycopersicum)

Global food production intensification presents a major hurdle to ensuring food security amidst a growing world population. Widespread use of chemical fertilizers in recent decades has risked soil fertility, compounded by the challenges posed by climate change, particularly in arid regions. To address these issues, adopting plant growth-promoting (PGP) bacteria stands out as a promising solution, offering multifaceted benefits to arid agroecosystems. We isolated a bacterial strain, SW7, from mangrove sediment, characterised the entire genome followed by phylogenetic analyses, and evaluated its in-vitro PGP activity. Subsequently, we examined its impact on tomato seed germination and plant growth. The strain SW7 exhibited growth on 11% NaCl, survival at 50°C, and possessed multiple PGP traits such as significant increase in seed germination rate (60.60 ± 38.85%), phosphate (83.3 g L−1) and potassium (39.6 g L−1) solubilization and produced indole acetic acid (3.60 ppm). Additionally, strain SW7 tested positive for ammonia, catalase, and oxidase enzyme production. The strain SW7 genome consists of 5.1 MB with 35.18% G+C content. Through genome-based phylogenetic and orthoANI analyses, the strain was identified as a novel Bacillus species, designated herein as Bacillus sp. SW7. In an eight-week shade-house experiment, inoculation of strain SW7 improved, leaf number, leaf density, leaf area index and mass water of tomatoes. Additional parameters, like chlorophyll a, chlorophyll b and carotenoids were not affected in SW7-inoculated tomatoes. In conclusion, Bacillus sp. SW7 exhibits multiple PGP traits and an adaptive capacity to high temperature and salinity, positioning it as a potential candidate for elevating the productivity of arid agroecosystems.

Understanding the Molecular Mechanism of PGPR Strain Priestia megaterium from Tea Rhizosphere for Stress Alleviation and Crop Growth Enhancement

The study aimed to assess the plant growth-promoting (PGP) capabilities and abiotic stress tolerance of strain IHB B 7164 with a focus on identifying associated genes through genomic analysis. Isolated from the tea rhizosphere, strain underwent screening for multiple PGP traits and stress tolerance against various abiotic factors. Taxonogenomic analysis of the complete genome identified the bacterium as Priestia megaterium exhibiting high similarity (97.04 % ANI, 74.3 % in silico DDH) to P. megaterium ATCC 14581. Functional annotations of the genome highlighted gene clusters associated with PGP traits and stress tolerance substantiating the strain's potential in promoting plant growth under stress conditions. The strain demonstarted robust growth and PGP activities under stressed culture conditions such as acidity, temperature fluctuations, desiccation, salinity, and exposure to calcium, aluminum, and iron salts, affirming its status as a stress-tolerant rhizobacterium. Screening of PGP traits under acidic conditions revealed phosphate solubilization, ACC-deaminase activity and production of IAA-like auxins. Notably gluconic and oxalic acids were consistently detected during solubilization of tri-calcium phosphate (TCP), aluminum phosphate (Al-P), and iron phosphate (Fe-P). Inoculation with the strain led to significant enhancements in tea nursery plants including a 9 % increase in plant height, 10 % increase in leaf number, and substantial improvements in leaf fresh and dry weights. Similarly, pea and wheat yields recorded notable increases of 16.9 % and 18.3 %, respectively under field conditions. These findings underscore the efficacy of P. megaterium strain IHB B 7164 as an abiotic stress-tolerant and broad-spectrum plant growth promoting rhizobacteria (PGPR) capable of enhancing plant growth and productivity across diverse agricultural settings.

Revealing the diversity of Jojoba-associated fungi using amplicon metagenome approach and assessing the in vitro biocontrol activity of its cultivable community.

Jojoba shrubs are wild plants cultivated in arid and semiarid lands and characterized by tolerance to drought, salinity, and high temperatures. Fungi associated with such plants may be attributed to the tolerance of host plants against biotic stress in addition to the promotion of plant growth. Previous studies showed the importance of jojoba as jojoba oil in the agricultural field; however, no prior study discussed the role of jojoba-associated fungi (JAF) in reflecting plant health and the possibility of using JAF in biocontrol. Here, the culture-independent and culture-dependent approaches were performed to study the diversity of the jojoba-associated fungi. Then, the cultivable fungi were evaluated for in-vitro antagonistic activity and in vitro plant growth promotion assays. The metagenome analysis revealed the existence of four fungal phyla: Ascomycota, Aphelidiomycota, Basidiomycota, and Mortierellomycota. The phylum Ascomycota was the most common and had the highest relative abundance in soil, root, branch, and fruit samples (59.7%, 50.7%, 49.8%, and 52.4%, respectively). Alternaria was the most abundant genus in aboveground tissues: branch (43.7%) and fruit (32.1%), while the genus Discosia had the highest abundance in the underground samples: soil (24%) and root (30.7%). For the culture-dependent method, a total of 14 fungi were isolated, identified, and screened for their chitinolytic and antagonist activity against three phytopathogenic fungi (Fusarium oxysporum, Alternaria alternata and Rhizoctonia solani) as well as their in vitro plant growth promotion (PGP) activity. Based on ITS sequence analysis, the selected potent isolates were identified as AspergillusstellatusEJ-JFF3, Aspergillusflavus EJ-JFF4, Stilbocreasp. EJ-JLF1, Fusariumsolani EJ-JRF3, and AmesiaatrobrunneaEJ-JSF4. The endophyte strain A. flavus EJ-JFF4 exhibited the highest chitinolytic activity (9 Enzyme Index) and antagonistic potential against Fusarium oxysporum, Alternaria alternata, and Rhizoctonia solani phytopathogens with inhibitory percentages of 72, 70, and 80 respectively. Also, A. flavus EJ-JFF4 had significant multiple PGP properties, including siderophore production (69.3%), phosphate solubilization (95.4 µg ml-1). The greatest production of Indol-3-Acetic Acid was belonged to A.atrobrunnea EJ-JSF4 (114.5 µg ml-1). The analysis of FUNGuild revealed the abundance of symbiotrophs over other trophic modes, and the guild of endophytes was commonly assigned in all samples. For the first time, this study uncovered fungal diversity associated with jojoba plants using a culture-independent approach and in-vitro assessed the roles of cultivable fungal strains in promoting plant growth and biocontrol. The present study indicated the significance of jojoba shrubs as a potential source of diverse fungi with high biocontrol and PGP activities.

Diversity and functional traits of seed endophytes of Dysphania ambrosioides from heavy metal contaminated and non-contaminated areas.

Seed endophytes played a crucial role on host plants stress tolerance and heavy metal (HM) accumulation. Dysphania ambrosioides is a hyperaccumulator and showed strong tolerance and extraordinary accumulation capacities of multiple HMs. However, little is known about its seed endophytes response to field HM-contamination, and its role on host plants HM tolerance and accumulation. In this study, the seed endophytic community of D. ambrosioides from HM-contaminated area (H) and non-contaminated area (N) were investigated by both culture-dependent and independent methods. Moreover, Cd tolerance and the plant growth promoting (PGP) traits of dominant endophytes from site H and N were evaluated. The results showed that in both studies, HM-contamination reduced the diversity and richness of endophytic community and changed the most dominant endophyte, but increased resistant species abundance. By functional trait assessments, a great number of dominant endophytes displayed multiple PGP traits and Cd tolerance. Interestingly, soil HM-contamination significantly increased the percentage of Cd tolerance isolates of Agrobacterium and Epicoccum, but significantly decreased the ration of Agrobacterium with the siderophore production ability. However, the other PGP traits of isolates from site H and N showed no significant difference. Therefore, it was suggested that D. ambrosioides might improve its HM tolerance and accumulation through harboring more HM-resistant endophytes rather than PGP endophytes, but to prove this, more work need to be conducted in the future.

Pseudomonas sp. AMGC1 takes on rice blast: Broad-spectrum antimicrobial activity underpins plant growth and disease tolerance.

A diverse group of gram-negative bacteria called Pseudomonas have been extensively researched for their ability to promote plant growth and suppress pathogens. The aim of the study was to identify the strain AMGC1 as a biocontrol agent against rice blast disease and a plant growth-promoting agent. The activity of AMGC1 against phytopathogens and its plant growth promotion activity were tested in vitro. Molecular identification by 16S rRNA gene sequencing, detection of antifungal antibiotic genes, biochemical characterization and antibiotic resistance testing were performed. Moreover, an in-planta biocontrol experiment with a blast susceptible rice cultivar was conducted to assess the effectiveness in managing the disease. Thin-layer chromatography was used to verify the presence of antifungal compounds. The results showed that AMGC1 had strong antimicrobial activity and multiple plant growth promotion traits. The strain was identified as Pseudomonas sp. and it contained genes for phenazine-1-carboxamide, hydrogen cyanide, and 2-hexyl, 5-propyl resorcinol antifungals. In the in-planta suppression experiment, applying AMGC1 strain two days before pathogen inoculation reduced the disease severity by 28% and disease incidence by 29% compared to control. Applying AMGC1 two days after pathogen inoculation resulted in lower reductions in disease severity (17.10%) and disease incidence (18.5%). Two putative bioactive compounds, phenazine-1-carboxamide and 2-hexyl, 5-propyl resorcinol, were isolated and purified from the AMGC1 growth medium. AMGC1 showed varied carbohydrate utilization patterns and both resistance and susceptibility to different antibiotics. The strain had multiple features that enhance plant growth and resistance against various phytopathogens, making it a potential biocontrol agent for sustainable agriculture.

Cyanobacteria Based Nanoformulation of Biogenic CuO Nanoparticles for Plant Growth Promotion of Rice Under Hydroponics Conditions.

Synthesizing nanoparticles through a green synthesis approach is common nowadays. Cyanobacteria have attained great importance in the field of biosynthesis of nanoparticles as there is no use of toxic chemicals as reducing or capping agents for the synthesis of metal oxide nanoparticles. Micronutrient-based nano-formulations have become a topic of great interest in recent times due to their various advantageous properties and applications in agriculture. The current study aims to exploit the potential cyanobacterial strains isolated from different locations such as freshwater and soil ecosystems. The potential cyanobacterial isolates were screened based on their multiple plant growth promoting (PGP) attributes such as Indol acetic acid (IAA) production, siderophores, and phosphate solubilization. After the screening of cyanobacteria based on multiple PGP activities, the cyanobacterial strain was identified at the species level as Pseudanabaena foetida RJ1, based on microscopy and molecular characterization using 16S rRNA gene sequencing. The cyanobacterial biomass extract and cell-free extracts are utilized for the synthesis of CuO micronutrient Nanoparticles (NPs). The cyanobacterial strain Pseudanabaena foetida RJ1 possesses plant growth-promoting (PGP) attributes that provide reduction and capping for CuO NPs. The synthesized NPs were characterized and subjected to make a nano-formulation, utilizing the cyanobacteria-mediated CuO NPs along with low-cost zeolite as an adsorbent. The application of cyanobacterial biomass extract and cell-free extract provided an excellent comparative aspect in terms of micronutrient NP synthesis. The NPs in the form of formulations were applied to germinated paddy seeds (Pusa Basmati -1509) with varying concentrations (5, 10, 15mg/l). Effects of cyanobacteria based CuO NPs on hydroponically grown paddy crops were analyzed. The application of nano-formulations has shown a significant increase in plant growth promotion in rice plants under hydroponics conditions. There is no such type of comparative investigation reported earlier, and NPs of micronutrients can be utilized as a new economic nanofertilizer and can be applied to plants for their growth promotion.

Effect of osmotic stress on in vitro plant growth hormone production by osmotolerant bacteria isolated from chilli phyto microbiome

The present study was conducted to determine the effect of osmotic stress on the plant growth hormone production by six osmotolerant plant growth promoting bacterial strains. These strains originated from the phytomicrobiome of chilli cultivated in the drought prone areas of Andhra Pradesh. They possessed multiple plant growth promotion traits including the ability to produce a variety of plant growth hormones. The effect of osmotic stress on the plant growth hormone production was determined by High Performance Liquid Chromatography (HPLC) under normal and in vitro osmotic stress conditions using 25% Poly Ethylene Glycol (PEG) 8000. In general, it was observed that osmotic stress impacted the plant growth hormone production of the isolates, but nevertheless plant hormones were detected in all the bacterial strains. An exception to this was the cytokinin molecule zeatin riboside, which was produced at higher levels by five of the six bacterial isolates under osmotic stressed conditions.

Functional Characterization of Rhizobia for Multiple Plant Growth Promoting Activities

This research focuses on the functional characterization of 19 rhizobia strains isolated from Southern Rajasthan's Udaipur and Dungarpur districts, with an emphasis on their multiple Plant Growth Promoting (PGP) activities. Through a series of tests encompassing Indole-3-Acetic Acid (IAA) production, siderophore production, phosphate solubilization, ammonia production, and hydrogen cyanide (HCN) production, this study elucidates the diverse PGP potential of these rhizobia strains. The results showcase variations in IAA production, highlighting their ability to enhance root development and overall plant growth, while also revealing strains capable of siderophore production, indicating potential iron uptake facilitation. Furthermore, some strains exhibit phosphate solubilization, ammonia production, and HCN production capabilities, signifying their potential to improve nutrient availability and protect plants from pathogens. These findings underscore the promise of harnessing the PGP activities of these rhizobia strains for sustainable agricultural practices in Southern Rajasthan and similar agroecological regions, with implications for enhanced crop yields and soil health.

Characterization and utilization of multi-trait plant growth promoting rhizobacteria from arid soils of western Rajasthan for enhancing drought resilience in an arid legume

In the study, plant growth promoting rhizobacteria (PGPR) indigenous to arid western Rajasthan were isolated and screened for plant growth promoting (PGP) capabilities. Ten isolates showing multiple PGP activities were identified as Pseudomonas sp. G6-3, Staphylococcus sp. G6-4, Bacillus badius G8-6, Agrobacterium sp. G8-7, Bacillus pseudomycoides G10-1, Staphylococcus capitis subsp. capitis G11-4, Bacillus paralicheniformis G11-5, Bacillus subtilis G12-3, Stenotrophomonas pavanii G12-4, and Solibacillus isronensis G21-9. Physiological profiling of these isolates demonstrated them to be tolerant to drought (25% PEG-6000), high temperature (40 °C), alkalinity (pH 10.0), and salinity (4.5% NaCl). These PGPR significantly improved clusterbean seedling characteristics in moisture stress-induced hydroponic flask cultures and pots. PGPR inoculation of clusterbean in drought-induced pots significantly enhanced the plant dry weight by 15–33%, plant water potential by 5–8%, and the relative water content of leaves by 4–16%. There was also a significant reduction in the levels of anti-oxidant enzymes and proline in plant tissues under drought conditions, which indicated lower levels of stress experienced by the treated plants. These were translated into yield enhancements by 13–15% under rainfed conditions. The study demonstrates the ability of stress tolerant PGPR from arid western Rajasthan in enhancing drought resilience of arid legumes besides promoting their growth, and their potential utility in sustainable arid agriculture systems.

Biofortification of Broccoli Microgreens (Brassica oleracea var. italica) with Glucosinolates, Zinc, and Iron through the Combined Application of Bio- and Nanofertilizers.

There is a severe need to develop a sustainable, affordable, and nutritious food supply system. Broccoli microgreens have attracted attention due to their rich nutritional content and abundant bioactive compounds, constituting an important opportunity to feed the ever-increasing population and fight global health problems. This study aimed to measure the impact of the combined application of biofertilizers and zinc and iron nanofertilizers on plant growth and the biofortification of glucosinolates (GLSs) and micronutrients in broccoli microgreens. Biofertilizers were based on plant growth-promoting (PGP) bacterial consortia previously isolated and characterized for multiple PGP traits. Nanofertilizers consisted of ZnO (77 nm) and γ-Fe2O3 (68 nm) nanoparticles synthesized with the coprecipitation method and functionalized with a Pseudomonas species preparation. Treatments were evaluated under seedbed conditions. Plant growth parameters of plant height (37.0-59.8%), leaf diameter (57.6-81.1%) and fresh weight (112.1-178.0%), as well as zinc (122.19-363.41%) and iron contents (55.19-161.57%), were mainly increased by nanoparticles subjected to the functionalization process with Pseudomonas species and uncapped NPs applied together with the biofertilizer treatment. Regarding GLSs, eight compounds were detected as being most positively influenced by these treatments. This work demonstrated the synergistic interactions of applying ZnO and γ-Fe2O3 nanofertilizers combined with biofertilizers to enhance plant growth and biofortify micronutrients and glucosinolates in broccoli microgreens.

Quorum sensing activities and genomic insights of plant growth-promoting rhizobacteria isolated from Assam tea.

Secretion of quorum sensing (QS) molecules is important for the effective colonization of host plants by plant growth-promoting rhizobacteria. The current study aims at the isolation and characterization of tea rhizo bacteria, which produce the QS molecules, acyl homoserine lactone (AHLs), along with multiple plant growth-promoting (PGP) activities. Thirty-one isolates were isolated from the tea rhizosphere, and screening for PGP activities resulted in the selection of isolates RTE1 and RTE4 with multiple PGP traits, inhibiting the growth of tea fungal pathogens. Both isolates also showed production of AHL molecules when screened using two biosensor strains, Chromobacterium violaceum CV026 and Escherichia coli MT 102(jb132). The isolates identified as Burkholderia cepacia RTE1 and Pseudomonas aeruginosa RTE4 based on genome-based analysis like phylogeny, dDDH, and fastANI calculation. Detailed characterization of AHLs produced by the isolates using reverse-phase TLC, fluorometry, and LC-MS indicated that the isolate RTE1 produced a short chain, C8, and a long chain C12 AHL, while RTE4 produced short-chain AHLs C4 and C6. Confocal microscopy revealed the formation of thick biofilm by RTE1 and RTE4 (18 and 23μm, respectively). Additionally, we found several genes involved in QS, and PGP, inducing systemic resistance (ISR) activities such as lasI/R, qscR, pqq, pvd, aldH, acdS, phz, Sod, rml, and Pch, and biosynthetic gene clusters like N-acyl homoserine lactone synthase, terpenes, pyochelin, and pyocyanin. Based on the functional traits like PGP, biofilm formation and production of AHL molecules, and genetic potential of the isolates B. cepacia RTE1 and P. aeruginosa RTE4 appear promising candidates to improve the health and growth of tea plantations.

The combination of multiple plant growth promotion and hydrolytic enzyme producing rhizobacteria and their effect on Jerusalem artichoke growth improvement

Rhizobacteria are well recognized for their beneficial multifunctions as key promoters of plant development, suppressing pathogens, and improving soil health. In this study, experiments focused on characterizing the plant growth promotion (PGP) and extracellular hydrolase production traits of rhizobacteria, and their impact on Jerusalem artichoke growth. A total of 50 isolates proved capable of either direct PGP or hydrolase-producing traits. Two promising strains (Enterobactercloacae S81 and Pseudomonasazotoformans C2-114) showed potential on phosphate and potassium solubilization, IAA production, and 1-aminocyclopropane-1-carboxylic acid deaminase activity and hydrolase production. A hydrolase-producing strain (Bacillussubtilis S42) was able to generate cellulase, protease, amylase, β-glucosidase, and phosphatase. These three selected strains also gave positive results for indirect PGP traits such as siderophore, ammonia, oxalate oxidase, polyamine, exopolysaccharide, biofilm, motility, and tolerance to salinity and drought stress. Colonization was observed using a scanning electron microscope and rhizobacteria appeared at the root surface. Interestingly, inoculation with consortia strains (S42, S81, and C2-114) significantly increased all plant parameters, including height, biomass, root (length, surface, diameter, and volume), and tuber fresh weight. Therefore, we recommend that potential consortia of PGP and hydrolase-producing rhizobacteria be employed as a biofertilizer to improve soil and boost crop productivity.

Endorhizosphere of indigenous succulent halophytes: a valuable resource of plant growth promoting bacteria

The adaptability of halophytes to increased soil salinity is related to complex rhizosphere interactions. In this study, an integrative approach, combining culture-independent and culture-dependent techniques was used to analyze the bacterial communities in the endorizosphere of indigenous succulent halophytes Salicornia europaea, Suaeda maritima, and Camphorosma annua from the natural salt marshes of Slano Kopovo (Serbia). The 16 S rDNA analyses gave, for the first time, an insight into the composition of the endophytic bacterial communities of S. maritima and C. annua. We have found that the composition of endophyte microbiomes in the same habitat is to some extent influenced by plant species. A cultivable portion of the halophyte microbiota was tested at different NaCl concentrations for the set of plant growth promoting (PGP) traits. Through the mining of indigenous halotolerant endophytes, we obtained a collection representing a core endophyte microbiome conferring desirable PGP traits. The majority (65%) of the selected strains belonged to the common halotolerant/halophilic genera Halomonas, Kushneria, and Halobacillus, with representatives exhibiting multiple PGP traits, and retaining beneficial traits in conditions of the increased salinity. The results suggest that the root endosphere of halophytes is a valuable source of PGP bacteria supporting plant growth and fitness in salt-affected soils.

Alleviation of cold stress in wheat with psychrotrophic phosphorus solubilizing Acinetobacter rhizosphaerae EU-KL44.

Low-temperature stress can seriously impair plant physiology. Chilling injury leads to a complex array of cellular dysfunctions, and symptoms include chlorosis, sterility, loss of vigor, wilting, and even death of the plants. Furthermore, phosphorus limitations additionally halt the growth of plants. Low-temperature adaptive plant growth-promoting microbes through various direct and indirect mechanisms help in the survival of plants under stress conditions. The present investigation deals with isolation of P-solubilizing psychrotrophic bacteria from diverse cultivars of wheat grown in the Keylong region of Himachal Pradesh. A total of 33 P-solubilizing bacterial isolates were obtained. P-solubilizers were screened for different plant growth-promoting (PGP) attributes of K and Zn solubilization, production of IAA, siderophores, and different hydrolytic enzymes. Among 33 P-solubilizers, 8 efficient strains exhibiting multiple PGP attributes were used as bioinoculants for wheat under low-temperature stress in different in vitro and in vivo experiments. The psychrotrophic bacterial isolates positively influenced the growth and physiological parameters as well as nutrient uptake and yield of wheat and efficiently alleviated low-temperature stress. The potential of low-temperature stress adaptive and PGP microbes can be utilized in agricultural sector for amelioration of low-temperature stress and plant growth promotion. The present study deals with the isolation of psychrotrophic P-solubilizers with multiple PGP attributes and their role in alleviation of cold stress in wheat.

Effect of seed biopriming with selected endophytes on the growth and chilling tolerance of rice plants.

The aim of this study was to develop an efficient bioinoculant for amelioration of adverse effects from chilling stress (10°C), which are frequently occurred during rice seedling stage. Seed germination bioassay under chilling condition with rice (Oryza sativa L.) cv. Tainan 11 was performed to screen for plant growth-promoting (PGP) bacteria among 41 chilling-tolerant rice endophytes. And several agronomic traits were used to evaluate the effects of bacterial inoculation on rice seedling, which were experienced for 7-d chilling stress in walk-in growth chamber. The field trials were further used to verify the performance of potential PGP endophytes on rice growth. A total of three endophytes with multiple PGP traits were obtained. It was demonstrated that Pseudomonas sp. CC-LS37 inoculation led to 18% increase of maximal efficiency of Photosystem II (PSII) after 7-d chilling stress and 7% increase of chlorophyll a content, and 64% decline of malondialdehyde content in shoot after 10-d recovery at normal temperature in walk-in growth chamber. In field trial, biopriming of seeds with strain CC-LS37 caused rice plants to increase shoot chlorophyll soil plant analysis development values (by 2.9% and 2.5%, respectively) and tiller number (both by 61%) under natural climate and chilling stress during the end of tillering stage, afterward 30% more grain yield was achieved. In conclusion, strain CC-LS37 exerted its function in increase of tiller number of chilling stress-treated rice seedlings via improvement of photosynthetic characteristics, which in turn increases the rice grain yield. This study also proposed multiple indices used in the screening of potential endophytes for conferring chilling tolerance of rice plants.

Biofertilizer Based on Biochar and Metal-Tolerant Plant Growth Promoting Rhizobacteria Alleviates Copper Impact on Morphophysiological Traits in Brassica napus L.

Metal tolerant plant growth-promoting (PGP) rhizobacteria are promising for enhancing plant productivity under copper (Cu) stress. Present pot scale experiment was conducted on Brassica napus L. to check the efficiency of rhizobacteria isolated from the rhizosphere of Tussilago farfara L. growing on Cu-contaminated soils. Out of fifty Cu tolerant strains, three isolates which showed multiple PGP traits such as indole-3-acetic acid (IAA) synthesis, phosphate (PS) solubilization, siderophore and ammonia production were identified preliminarily by morphological and physiological characteristics followed by 16S rRNA gene sequencing. The best Bacillus altitudinis strain TF16a which showed IAA: 15.5 mg L-1, PS: 215 mg L-1, siderophore halo zone ratio of 3.0 with high ammonia production was selected to prepare a biochar-based biofertilizer (BF). Seedling test showed maximum growth of B. napus shoot and root in presence of 5% of BF and this concentration was selected for further experiment. The pot experiment included four treatments: control (soil), 100Cu (100 mg Cu kg-1 soil), 5%BF (v/v), and 5%BF+100Cu, which were carried out for 30 days, after which the morphological, physiological, and biochemical parameters of B. napus were studied. The Cu treatment caused its accumulation in shoot and root up to 16.9 and 30.4 mg kg-1 DW, respectively, and increased malondialdehyde (MDA) content by 20%. Application of BF with copper led to the decrease in the Cu accumulation by 20% for shoot and 28% for root while MDA content was the same as in the control. Both treatments of BF with and without Cu increased chlorophyll a and b content by 1.3 times on average as well as non-enzymatic antioxidants such as soluble phenolic compounds (1.3 times) and free proline (1.6 times). Moreover, BF + Cu led to the increase in the biomass of shoot and root by 30 and 60%, respectively, while there was no significant effect on the growth characteristics of plants after the addition of BF without Cu. The study elucidates that BF based on B.altitudinis strain TF16a and biochar can be a promising bioformulation which could increase rapeseed growth under the moderate Cu concentration in soil.

Abundant and diverse endophytic bacteria associated with medicinal plant Arctium lappa L. and their potential for host plant growth promoting.

Arctium lappa L. is one of the medicinal and food homologous plants in China, which is rich in nutrients and medicinal ingredients. The use of plant growth-promoting (PGP) endophytic bacteria is an alternative to reducing chemical fertilizers in agricultural production. The aim of this study was to analyze the diversity of endophytic bacteria in different cultivars of A. lappa L. collected from two geographical locations in China and evaluate PGP traits of the isolates and their potential PGP ability in greenhouse condition. Endophytic bacterial community was investigated by culture-dependent and culture-independent methods. Isolates were screened and investigated for multiple PGP traits, and representative strains were inoculated host seedlings to evaluate the growth promoting effect. A total of 348 endophytic bacteria were obtained and they were distributed into 4 phyla and 30 genera. In addition, highthroughputsequencing revealed more abundant bacterial community, including 17 bacterial phyla and 207 genera. A high proportion of PGP traits were detected, including production of indole acetic acid, siderophore, ammonia and phosphate solubilization. Four representative strains with multiple PGP traits of the most dominant genera (Bacillus, Pantoea, Microbacterium and Pseudomonas) were further selected for host inoculation and growth promoting evaluation, and they significantly increase seedlings length, root length and fresh weight. This study demonstrated that A. lappa L. harbors abundant endophytic bacteria, and some endophytic bacteria showed good potential for the development of microbial fertilizer in the future.

Potential use of endophytic and rhizosheath bacteria from the desert plant Stipagrostis pennata as biostimulant against drought in wheat cultivars

The use of bacteria associated with desert plants seems to have a higher potential to alleviate the negative effects of drought stress on water deficit-stressed non-desert plants. The aim of this study was to investigate the potential effect of superior endophytic (E1) and rhizosheath (R11) bacterial strains (B0-control, E1, R11 and E1 + R11), selected based on their tolerance to drought stress and plant growth-promoting (PGP) traits, from the root and rhizosheath of Stipagrostis pennata, a pioneer plant species in the desert, on growth indices (morphological, biophysiological and nutritional responses) of two wheat cultivars (Sirvan, resistant to drought, and Sivand, sensitive to drought) under water-deficit stress conditions (50–55% FC, water deficit stress and 80–88% FC, non-stress). This evaluation was carried out in the form of a completely randomized design in factorial arrangement with three replications under greenhouse conditions. The results showed that with the application of water-deficit stress, wheat growth indices decreased in both cultivars, but this decrease was more in Sivand cultivar. However, with the use of superior endophytic (E1) and rhizosheath (R11) bacterial strains and a mixture of these two strains with a greater effect, the negative impact of water-deficit stress on the growth indices of two cultivars (an increase in plant height by 27%, in root length by 27%, in shoot dry weight by 56%, in root dry weight by 50%, in chlorophyll index by 30%, in superoxide dismutase activity by 8% and in the concentration of shoot N, P and K by 30%, 56% and 31%, respectively, and a decrease in proline concentration by 59%) was alleviated. Bacterial performance also was variable depending on stress level and cultivar, as the effect of these isolates on Sirvan cultivar was more than Sivand cultivar. This can be a challenge for producing biofertilizers for cultivars whose tolerance to drought is unknown. This limitation can probably be managed by developing biofertilizers composed of different bacterial strains containing a wide range of PGP traits and compatible with different plant cultivars. Based on 16S rRNA sequence analysis, E1 and R11 isolates were identified as Pseudomonas sp. and Pantoea agglomerans, respectively. In general, this study showed that the rhizosheath and endosphere of the desert plant S. pennata can be a suitable source for isolating drought-resistant bacteria with multiple PGP traits. These bacteria can also play an important role in promoting the tolerance of non-desert plants (e.g., wheat) to drought and can be used as biofertilizers to increase the growth of the plants in arid and semi-arid regions after their field assays. The results of this study also emphasize on considering the best bacterial strain-wheat cultivar-stress level combination to increase wheat yield.

Synergistic Effect of Azotobacter nigricans and Nitrogen Phosphorus Potassium Fertilizer on Agronomic and Yieldtraits of Maize (Zea mays L.).

Plant growth-promoting bacteria (PGPB) Azotobacter spp. is the most promising bacteria among all microorganisms. It is an aerobic, free-living, and N2-fixing bacterium that commonly lives in soil, water, and sediments. It can be used as a biofertilizer for plant growth and nutrient utilization efficiency. Maize is the highly consumed cereal food crop of the cosmopolitan population, and the sustainable maize productivity achieved by applying bacteria in combination with nitrogen phosphorus potassium (NPK) is promising. In the present study, a bacterial isolate (PR19). Azotobacter nigricans, obtained from the soil of an organic farm was evaluated for its plant growth promoting potential alone and in combination with an inorganic fertilizer (NPK) included. The bacterial cultue (PR19) was screened for its morphological, biochemical, and plant growth-promoting characteristics, sequenced by the 16S rDNA method, and submitted to NCBI for the confirmation of strain identification. Further, the inoculation effect of the bacterial culture (PR19) in combination with NPK on growth and yield parameters of maize under pot were analyzed. Based on phenotypic and molecular characteristics, PR19 was identified as Azotobacter nigricans it was submitted to NCBI genbank under the accession No. KP966496. The bacterial isolate possessed multiple plant growth-promoting (MPGP) traits such as the production of ammonia, siderophore, indole-3-acetic acid (IAA), and ACC Deaminase (ACCD). It showed phosphate solubilization activity and tolerance to 20% salt, wide range of pH 5–9, higher levels of trace elements and heavy metals, and resistance to multiple antibiotics. PR19 expressed significantly increased (p < 0.001) antioxidant enzyme activities (SOD, CAT, and GSH) under the abiotic stress of salinity and pH. In vitro condition, inoculation of maize with the PR19 showed a significant increase in seed germination and enhancement in elongation of root and shoot compared to untreated control. The combined application of the PR19 and NPK treatments showed similar significant results in all growth and yield parameters of maize variety SHIATS-M S2. This study is the first report on the beneficial effects of organic farm isolated PR19-NPK treatment combinations on sustainable maize productivity.

Bacterial Mitigation of Drought Stress in Plants: Current Perspectives and Future Challenges.

Climate change is emerging as a crucial issue with global attention and leading to abiotic stress conditions. There are different abiotic stress which affects the crop production among which drought is known to be most destructive stress affecting crop productivity and world's food security. Different approaches are under consideration to increase adaptability of the plants under drought stress with plant-microbe interactions being a greater area of focus. Stress-adaptive microbes either from the rhizosphere, internal tissue, or aerial parts of plants have been reported which through different mechanisms help the plants to cope up with drought and also promote their growth. These mechanisms include the accumulation of osmolytes, decrease in the inhibitory levels of ethylene by aminocyclopropane-1-carboxylate (ACC) deaminase enzyme, and furnishing the unavailable nutrients to plants. Microbial genera including Azotobacter, Bacillus, Ochrobactrum, Pseudomonas, and Serratia are known to be self-adaptive and growth promoters under drought stressed conditions. Stress-adaptive plant growth promoting (PGP) microbes thus are excellent candidates for stress alleviation in drought environment to provide maximum benefits to the plants. The present review deals with the effect of the drought stress on plants, biodiversity of the drought-adaptive microbes, mechanisms of the drought stress alleviation through enhancement of stress alleviators, reduction of the stress aggravators, and modification of the molecular pathways as well as the multiple PGP attributes of the drought-adaptive microbes.
