Mechanical stability of polarization signatures in biological tissue characterization.

Mueller matrix imaging polarimetry (MMIP) is a promising technique for investigating structural abnormalities in pathological diagnosis. The characterization stability of polarization signatures, described by Mueller matrix parameters (MMPs), correlates with the mechanical state of the biological medium. In this study, we developed an MMIP system capable of applying quantitative forces to samples and measuring the resulting polarization signatures. Mechanical stretching experiments were conducted on a mimicking phantom and a tissue sample at different force scales. We analyzed the textural features and data distribution of MMP images and evaluated the force effect on the characterization of MMPs using the structural similarity index. The results demonstrate that changes in the mechanical microenvironment (CMM) can cause textural fluctuations in MMP images, interfering with the stability of polarization signatures. Specifically, parameters of anisotropic orientation, retardance, and optical rotation are the most sensitive to CMM, inducing a dramatic change in the overall image texture, while other parameters (e.g., polarization, diattenuation, and depolarization) exhibit locality in their response to CMM. For some MMPs, CMM can enhance regional textural contrasts. This study elucidates the mechanical stability of polarization signatures in biological tissue characterization and provides a valuable reference for further research toward minimizing CMM influence.

Mueller-matrix imaging polarimetry elevated by wavelet decomposition and polarization-singular processing for analysis of specific cancerous tissue pathology.

Mueller-matrix polarimetry is a powerful method allowing for the visualization of malformations in biological tissues and quantitative evaluation of alterations associated with the progression of various diseases. This approach, in fact, is limited in observation of spatial localization and scale-selective changes in the poly-crystalline compound of tissue samples. We aimed to improve the Mueller-matrix polarimetry approach by implementing the wavelet decomposition accompanied with the polarization-singular processing for express differential diagnosis of local changes in the poly-crystalline structure of tissue samples with various pathology. Mueller-matrix maps obtained experimentally in transmitted mode are processed utilizing a combination of a topological singular polarization approach and scale-selective wavelet analysis for quantitative assessment of the adenoma and carcinoma histological sections of the prostate tissues. A relationship between the characteristic values of the Mueller-matrix elements and singular states of linear and circular polarization is established within the framework of the phase anisotropy phenomenological model in terms of linear birefringence. A robust method for expedited (up to ) polarimetric-based differential diagnosis of local variations in the poly-crystalline structure of tissue samples containing various pathology abnormalities is introduced. The benign and malignant states of the prostate tissue are identified and assessed quantitatively with a superior accuracy provided by the developed Mueller-matrix polarimetry approach.

Polarization clustering of biological structures with Mueller matrix parameters.

Mueller matrix imaging polarimetry (MMIP) is a promising technique for the characterization of biological tissues, including the classification of microstructures in pathological diagnosis. To expand the parameter space of Mueller matrix parameters, we propose new vector parameters (VPs) according to the Mueller matrix polar decomposition method. We measure invasive bladder cancer (IBC) with extensive necrosis and high-grade ductal carcinoma in situ (DCIS) with MMIP, and the regions of cancer cells and fibrotic stroma are classified with the VPs. Then the proposed and existing VPs are mapped on the Poincaré sphere with 3D visualization, and an indicator of spatial feature is defined based on the minimum enclosing sphere to evaluate the classification capability of the VPs. For both IBC and DCIS, the results show that the proposed VPs exhibit evident contrast between the regions of cancer cells and fibrotic stroma. This study broadens the fundamental Mueller matrix parameters and helps to improve the characterization ability of the MMIP technique.

Combined Mueller matrix imaging and artificial intelligence classification framework for Hepatitis B detection.

.SignificanceThe combination of polarized imaging with artificial intelligence (AI) technology has provided a powerful tool for performing an objective and precise diagnosis in medicine.AimAn approach is proposed for the detection of hepatitis B (HB) virus using a combined Mueller matrix imaging technique and deep learning method.ApproachIn the proposed approach, Mueller matrix imaging polarimetry is applied to obtain Mueller matrix images of 138 HBsAg-containing (positive) serum samples and 136 HBsAg-free (negative) serum samples. The kernel estimation density results show that, of the 16 Mueller matrix elements, elements and provide the best discriminatory power between the positive and negative samples.ResultsAs a result, and are taken as the inputs to five different deep learning models: Xception, VGG16, VGG19, ResNet 50, and ResNet150. It is shown that the optimal classification accuracy (94.5%) is obtained using the VGG19 model with element as the input.ConclusionsOverall, the results confirm that the proposed hybrid Mueller matrix imaging and AI framework provides a simple and effective approach for HB virus detection.

Feasibility of Hepatitis B virus infection detection using Mueller matrix decomposition method and Mueller matrix imaging polarimetry technique

The ability to distinguish and characterize differences in the optical properties of biological samples is crucial in various disease diagnosis applications. The present study investigates the feasibility for utilizing the Mueller matrix decomposition method based on a UV light Stokes polarimeter and a Mueller matrix imaging polarimetry technique based on a He–Nelaser diode to detect Hepatitis B virus (HBV) infection in human plasma samples. In the Mueller matrix decomposition method, the orientation angle of linear birefringence (α), phase retardance (β), optical rotation angle of circular birefringence (γ), orientation angle of linear dichroism (θd), linear dichroism (D), and circular dichroism (R) are extracted for negative and positive human plasma HBV samples and the differences between them are analyzed. It is shown that statistically significant differences exist between the β, γ and R values of the negative and positive samples, respectively. Moreover, for both methods, the positive HBV samples show a greater degree of anisotropy than the negative samples. In general, the results confirm that both methods provide a feasible approach for distinguishing between negative and positive HBV samples and thus serve as potential tools for HBV detection and diagnosis.

Study of the spatial scale stability of Mueller matrix parameters for textural characterization of biological tissues.

Mueller matrix imaging polarimetry (MMIP) is a promising technique for the textural characterization of biological tissue structures. To reveal the influence of imaging magnification on the robustness of Mueller matrix parameters (MMPs), the spatial scale stability of MMPs was studied. We established a new MMIP detector and derived the mathematical model of the spatial scale stability of MMPs. The biological tissues with well-defined structural components were imaged under different magnifications. Then, we compared and analyzed the textural features of the MMPs in the resulting images. The experimental results match the predictions of the mathematical model in these aspects: (a) magnification exhibits a strong nonlinear effect on the textural contrasts of MMPs images; (b) higher magnification does not necessarily lead to superior contrast for textural characterization; and (c) for different biological tissues, MMPs contrasts can be optimized differently, with some showing superior results. This study provides a reference for the experimental design and operation of the MMIP technique and is helpful for improving the characterization ability of MMPs.

Evolution of raw meat polarization-based properties by means of Mueller matrix imaging.

The possibilities of using Mueller matrix (MM) imaging polarimetry to assess meat quality have not yet been sufficiently explored. In the current study, the fresh porcine muscles are imaged at room temperature with a wide-field MM imaging polarimeter over 26 hours to visualize dynamics of tissue optical properties through applying Lu-Chipman decomposition. The frequency distribution histograms (FDHs) and statistical analysis of the MM elements show prominent changes over time. The wavelength spectra of both total depolarization and scalar retardance have dips at 550 nm whereas their values continuously increase with time; the former is referred to the increase of number of scattering events and decrease of myoglobin absorption in the red part of visible spectra related to meat color and freshness, while the latter is associated with the increase in birefringence and meat tenderness. The obtained results are promising to develop a novel fast noncontact optical technique for monitoring of meat quality.

Different orders of scattering through time-resolved Mueller matrix imaging estimates of pre-malignancy in human cervical tissues.

Time-resolved Mueller matrix (MM) imaging polarimetry in transmission mode has been implemented in both epithelium and stromal regions of cervical tissues to explore the various polarization dynamics in connection with the diagnosis of cervical precancer. The picosecond-resolved intensity patterns of various MM elements, resulting from the various orders of scattering, at different time delays provide clear demarcation between the epithelium and stroma of cervical tissue. The time dependent depolarization and retardance maps are seen to differentiate the epithelium from stroma. The average values of time dependent linear, linear-45, and circular depolarization and linear, circular, and scalar retardance parameters in different regimes of scattering from the optically anisotropic stromal region identify the pre-malignancy in cervical tissue. As the disease evolves, time dependent linear depolarization varies to larger values as compared to time dependent circular depolarization. Interestingly, the chirality of the collagen network that rotates the plane of polarized light in either direction in normal samples is limited to only the clockwise direction during the progression of the disease. These results show potential in the early detection and understanding of the mechanisms of morphological changes in cervical cancer development.

Evaluation of structural and molecular variation of starch granules during the gelatinization process by using the rapid Mueller matrix imaging polarimetry system.

Starch is an essential and widely distributed natural material, but its detailed conformation and thermal transition properties are not yet well understood. We present a rapid Mueller matrix imaging system to explore the structural characteristics of starch granules by using 16 measurements with different incoming and outgoing polarizations. Due to the minimum rotation of the optical elements and the self-calibration ability of this system, the full Mueller matrix images can be accurately obtained within ten-odd seconds. Both structural and molecular features of the starch granule were investigated in the static state and gelatinization process by means of multiple optical characteristics deduced from the Mueller matrix. The experimental results for the structural changes during the gelatinization were close to other nonlinear optical approaches; moreover, the crystallinity and optical rotation of the starch granule are also determined through the use of this approach.

Optical techniques for cervical neoplasia detection

This paper provides an overview of the current research in the field of optical techniques for cervical neoplasia detection and covers a wide range of the existing and emerging technologies. Using colposcopy, a visual inspection of the uterine cervix with a colposcope (a binocular microscope with 3- to 15-fold magnification), has proven to be an efficient approach for the detection of invasive cancer. Nevertheless, the development of a reliable and cost-effective technique for the identification of precancerous lesions, confined to the epithelium (cervical intraepithelial neoplasia) still remains a challenging problem. It is known that even at early stages the neoplastic transformations of cervical tissue induce complex changes and modify both structural and biochemical properties of tissues. The different methods, including spectroscopic (diffuse reflectance spectroscopy, induced fluorescence and autofluorescence spectroscopy, Raman spectroscopy) and imaging techniques (confocal microscopy, optical coherence tomography, Mueller matrix imaging polarimetry, photoacoustic imaging), probe different tissue properties that may serve as optical biomarkers for diagnosis. Both the advantages and drawbacks of these techniques for the diagnosis of cervical precancerous lesions are discussed and compared.

Measurement of microfibril angles in bamboo using Mueller matrix imaging.

The microfibril angle (MFA) giving the orientation of cellulose chains in hard sclerenchymatous bamboo fibers is one of the most important parameters determining the overall strength of the bamboo culm. In this work, Mueller matrix imaging polarimetry is implemented for determining MFA measured over a transverse section of group of fibers and parenchyma cells in bamboo of Dendrocalamus strictus species. The method, based on the Stokes-Mueller formalism, decouples the birefringence exhibited by crystalline cellulose from the clumped polarization parameters using 16 images taken with different polarization states at subcellular resolution. Retardance values, obtained from polar decomposition of the Mueller matrix, are extracted from different locations in the specimen, and distribution of MFA over the entire section is presented. The method permits simultaneous measurement of MFA in a transverse section of several fibers and parenchyma cells. The range of MFA obtained for bamboo fibers from Mueller matrix imaging is verified with the results obtained through x-ray diffraction using the pole figure method.

Dichroism in Helicoidal Crystals.

Accounting for the interactions of light with heterogeneous, anisotropic, absorbing, optically active media is part of the characterization of complex, transparent materials. Stained biological structures in thin tissue sections share many of these features, but systematic optical analyses beyond the employ of the simple petrographic microscopes have not be established. Here, this accounting is made for polycrystalline, spherulitic bundles of twisted d-mannitol lamellae grown from melts containing light-absorbing molecules. It has long been known that a significant percentage of molecular crystals readily grow as helicoidal ribbons with mesoscale pitches, but a general appreciation of the commonality of these non-classical crystal forms has been lost. Helicoidal crystal twisting was typically assayed by analyzing refractivity modulation in the petrographic microscope. However, by growing twisted crystals from melts in the presence of dissolved, light-absorbing molecules, crystal twisting can be assayed by analyzing the dichroism, both linear and circular. The term "helicoidal dichroism" is used here to describe the optical consequences of anisotropic absorbers precessing around radii of twisted crystalline fibrils or lamellae. d-Mannitol twists in two polymorphic forms, α and δ. The two polymorphs, when grown from supercooled melts in the presence of a variety of histochemical stains and textile dyes, are strongly dichroic in linearly polarized white light. The bis-azo dye Chicago sky blue is modeled because it is most absorbing when parallel and perpendicular to the radial axes in the respective spherulitic polymorphs. Optical properties were measured using Mueller matrix imaging polarimetry and simulated by taking into account the microstructure of the lamellae. The optical analysis of the dyed, patterned polycrystals clarifies aspects of the mesostructure that can be difficult to extract from bundles of tightly packed fibrils.

Rapid full Mueller matrix imaging polarimetry based on the hybrid phase modulation technique

In this work, we present a novel method of Mueller matrix imaging polarimetry, which comprises dual liquid crystal variable retarders at the polarization generation portion and a photoelastic modulator at the polarization analysis portion. The light source can be operated either in the continuous mode, which provides an in-situ calibration process for the liquid crystal variable retarders, or in the pulse mode to deduce the full two-dimensional Mueller matrix with 16 images from the camera. We measured the Mueller matrix images of air as a standard test, as well as a quarter wave plate to determine its azimuthal angle and phase retardation by the polar decomposition technique. Finally, the decomposed Mueller matrix images of a biopolymer specimen with the conformational change produced by heat treatment are presented.

Development of a Mueller matrix imaging system for detecting objects embedded in turbid media

In this research, the Mueller matrix (MM) imaging polarimetry technique is used to experimentally measure the MMs corresponding to the surface reflectance of turbid media in which an opaque object is immersed. Depending on the penetration depth of light in the medium and the location of the object, the MM may be sensitive enough as to detect not only its presence and location but also its characteristics, according to the nature of its interaction with the incident light. A Dual Rotating Compensator Polarimeter (DRCP) has been used to obtain accurate surface MMs for either reflecting or absorbent objects. It is shown how the comparison of the experimental MMs and the corresponding object-free MMs can be made by means of a relative matrix. The results show how this new tool may assess the enhanced contrast that this technique provides.

Bernauer′s Bands

Ferdinand Bernauer proposed in his monograph, "Gedrillte" Kristalle (1929), that a great number of simple, crystalline substances grow from solution or from the melt as polycrystalline spherulites with helically twisting radii that give rise to distinct bull's-eye patterns of concentric optical bands between crossed polarizers. The idea that many common molecular crystals can be induced to grow as mesoscale helices is a remarkable proposition poorly grounded in theories of polycrystalline pattern formation. Recent reinvestigation of one of the systems Bernauer described revealed that rhythmic precipitation in the absence of helical twisting accounted for modulated optical properties [Gunn, E. et al. J. Am. Chem. Soc. 2006, 128, 14234-14235]. Herein, the Bernauer hypothesis is re-examined in detail for three substances described in "Gedrillte" Kristalle, potassium dichromate, hippuric acid, and tetraphenyl lead, using contemporary methods of analysis not available to Bernauer, including micro-focus X-ray diffraction, electron microscopy, and Mueller matrix imaging polarimetry. Potassium dichromate is shown to fall in the class of rhythmic precipitates of undistorted crystallites, while hippuric acid spherulites are well described as helical fibrils. Tetraphenyl lead spherulites grow by twisting and rhythmic precipitation. The behavior of tetraphenyl lead is likely typical of many substances in "Gedrillte" Kristalle. Rhythmic precipitation and helical twisting often coexist, complicating optical analyses and presenting Bernauer with difficulties in the characterization and classification of the objects of his interest.

Polarimetric characterization of liquid-crystal-on-silicon panels

Mueller matrix imaging polarimetry of liquid-crystal-on-silicon (LCoS) panels provides detailed information useful for the diagnosis of LCoS problems and to understand the interaction of LCoS panels with other projector components. Data reduction methods are presented for the analysis of LCoS Mueller matrix images yielding contrast ratio, efficiency, spatial uniformity, and the calculation of optimum trim retarders. The effects of nonideal retardance, retardance orientation, and depolarization on LCoS system performance are described. The white-state and dark-state Mueller matrix images of an example LCoS panel are analyzed in terms of LCoS performance metrics typical for red-green-blue wavelengths of 470, 550, and 640 nm. Variations of retardance, retardance orientation, and depolarization are shown to have different effects on contrast ratio, efficiency, and brightness. Thus Mueller matrix images can diagnose LCoS problems in a way different from radiometric testing. The calculation of optimum trim retarders in the presence of spatial variations is discussed. The relationship of the LCoS retardance in single-pass (from front to back) to the double-pass retardance (from entrance to exit) is established and used to clarify coordinate system issues related to Mueller matrices for reflection devices.

Double-pass axially resolved confocal Mueller matrix imaging polarimetry

We present experimental depth-resolved complete polarization-sensitive measurements of a stack of linear retarders and glass plates by using what is to the best of our knowledge the first combination of a confocal imaging system with a complete Mueller matrix polarimeter. The axially resolved Mueller matrices were compared with a forward simulation, with good agreement.

Time-resolved Mueller matrix imaging polarimetry

We present a new method of time-resolved Mueller matrix imaging polarimetry for spatial and temporal characterization of the polarization effects in backscattering from turbid media. The technique allows measuring the time evolution of spatially varying polarization patterns of diffusely backscattered light with picosecond resolution. A series of time-resolved polarization patterns are obtained at various time delays, are analyzed in sequence, and used to separate the polarimetric contributions of different scattering paths. Specific features of the 2D Mueller matrix components corresponding to light backscattered from colloidal suspensions were determined and characterized. The temporally-and spatially-resolved measurements permit detailed analysis of the changes in the magnitude, sign, and the general symmetry properties of Mueller matrix components.

Polarization properties of the in vitro old human crystalline lens.

We have studied the spatially resolved polarization properties of the in vitro intact old human crystalline lens (from 56 to 88 years old) by using Mueller-matrix imaging polarimetry. Analysis was performed within an average of 54 h of death. Results show that the overall retardation is small (7 degrees on average) and decreases from the centre of the lens to the periphery. Lenticular birefringence is linear but has a spatial dependence, reducing outwards along the radius. The distribution of azimuthal angle of the birefringent structure of the crystalline lens changes depending on each individual lens. Diattenuation and polarizance were found to be small, however, depolarization was about 35% for the set of lenses studied here.
