From Hagfish to Humans

With more clinical information for trainees to master in the face of increasing time pressures, discussions about underlying physiology and the mechanisms of disease seem to have been de-emphasized during residency training. This reduced focus on pathophysiology and basic science may weaken trainees' clinical effectiveness and reduce their interest in pursuing research careers.In response, the authors helped to develop in 2006 a one-week immersive comparative physiology course for second- and third-year internal medicine residents at Beth Israel Deaconess Medical Center. The course, held at the Mount Desert Island Biological Laboratory, includes four modules (hematology, vascular physiology, secretory physiology, and salt and water homeostasis), each composed of basic science experiments using aquatic species and accompanied by clinical correlation discussions and group presentations.From 2007 to 2010, 72 residents rotated through the course. Most reported that it enhanced their understanding of the mechanisms of disease in their patients. After the course, residents reported that physiology played a more prominent role in their teaching and clinical decision making during both ward and intensive care unit rotations. They also reported being more likely than before the course to read about the pathophysiology of disease when faced with a clinical problem.Although cost-intensive and geographically unique, this model for teaching the mechanisms of disease could be applied elsewhere with the help of physician-scientists and clinician-educators. In this article, the authors describe the development of the course, share preliminary data evaluating progress toward its goals, and discuss future directions and lessons learned.

Characterization of aquaporin 8 isoform expression in eel (Anguilla sp.) intestine

Euryhaline eels are placed under severe osmotic stress when they migrate from freshwater to seawater during their lifecycle. In seawater, the animals drink to maintain equilibrium; to obtain water to replace continual losses occurring due to osmosis across permeable body surfaces. A significant increase in the intestine's capacity to absorb water from imbibed fluid consequently occurs and this may involve upregulation of aquaporin water channels. Studies using quantitative polymerase chain reaction or Northern blotting have shown increases in aquaporin 8 and 8b isoform mRNA levels in intestinal segments following seawater acclimation of eels. Custom-made isoform-specific polyclonal antibodies have been raised against aquaporin 8 and 8b, and these show intracellular staining in intestine surface epithelial cells. Additionally, there are indications of a lateral plasma membrane localization of aquaporin 8 and 8b isoforms in rectal surface epithelial cells, suggesting a role for aquaporin 8 and 8b isoforms in water absorption mechanisms. Cell physiology at the Mount Desert Island Biological Laboratory: a brief look back and forward.

The Mount Desert Island Biological Laboratory (MDIBL) has played important roles in the development of modern physiological concepts and tools, particularly in the fields of kidney and epithelial cell physiology. Over the last decade, MDIBL has undergone remarkable growth and evolution. This article will briefly review MDIBL's past and outline its future directions. It is hoped that this overview will renew and stimulate interest in MDIBL and, in particular, will encourage an even wider community of physiologists to participate in its ongoing growth and development.

The Community Outreach and Education Program (COEP) of the Center for Membrane Toxicity Studies, housed at Mount Desert Island Biological Laboratory in Bar Harbor, Maine, is using its open-air Myers Marine Aquarium Visitors Center to maximize this combination and get kids excited about learning during the summer months. The Center for Membrane Toxicity Studies, one of the NIEHS Marine and Freshwater Biomedical Sciences Centers, educates children about the environment and their own role in keeping it healthy.For about five years, the COEP has provided environmental health enrichment to the surrounding community through the Myers Marine Aquarium Visitors Center. Aquarium staff provide weekly programs in the summer for local and tourist visitors to learn about the center’s research. In a typical tour, visitors learn about water pollutants and how they affect our health and environment, and are also given information on water quality management.Because school-aged children are the target audience, the message delivery is kept simple and fun. The aquarium has several tanks that allow for viewing and touching of the lab’s research species and other sea life from the Gulf of Maine. Cartoon-emblazoned posters illustrate center findings on how pollutants are processed in the body and describe in nonspecialist language our own responsibility in safeguarding the environment we live in. As part of the tour, center staff describe their research projects and allow visitors to view specimens through microscopes.“When we talk about environmental stewardship, I am always amazed by the reaction of the children,” says Jeri Bowers, the COEP associate director. “They love to get involved and to feel like they are playing an important role in maintaining a healthy environment; they take it very seriously. Our hope is that we can capture that enthusiasm and interest while they’re young—and influence their behavior well into adulthood.”The aquarium attracts large numbers of student groups. One particularly memorable visit was a May 2004 tour by a group of 40 children with Almström syndrome, a rare hereditary disorder that can affect multiple organ systems and cause blindness, hearing impairment, type 2 diabetes mellitus, heart failure, and liver disease. The visit was arranged by researchers at the nearby Jackson Laboratory, who discovered the gene for Almström syndrome and provide their own outreach to children affected by the illness. Bowers says the tactile component of the aquarium experience was especially important and beneficial for these children, most of whom can’t see or hear.Along with the strong community outreach it offers through the aquarium and labs, Mount Desert Island also has exciting environmental health research in progress that staff hope will someday become part of the children’s experience. Earlier this year, the lab set up a website ( http://ctd.mdibl.org/) for the Comparative Toxicogenomics Database, a data system being created to provide centralized data on diverse organisms to scientists worldwide. The lab has also received NIH approval to sequence the genome of the skate, data that will eventually be entered into the Comparative Toxico-genomics Database. One of the future goals of the researchers is to incorporate the electronic data into the visitors program so that children can access the database on computers set up as part of the aquarium tour.Diving in. DATABASE: Bad for the Genes

Need to know which genes the neurotoxic pesticide DDT meddles with? Wondering how the toxic metal cadmium affects erythropoietin, a hormone that spurs blood cell production? Drop by the prototype Comparative Toxicogenomics Database, sponsored by the National Institute for Environmental Health Sciences and the Mount Desert Island Biological Laboratory in Maine. The collection allows you to determine which genes respond to a particular compound, to find out which species the interaction has been studied in, and to answer other questions about hundreds of chemicals. The information comes from papers cited in PubMed and in databases such as GenBank and SwissProt, but the site's creators hope that researchers will contribute their own findings.

The Comparative Toxicogenomics Database (CTD).

The Mount Desert Island Biological Laboratory in Salsbury Cove, Maine, USA, is developing the Comparative Toxicogenomics Database (CTD), a community-supported genomic resource devoted to genes and proteins of human toxicologic significance. CTD will be the first publicly available database to a) provide annotated associations among genes, proteins, references, and toxic agents, with a focus on annotating data from aquatic and mammalian organisms; b) include nucleotide and protein sequences from diverse species; c) offer a range of analysis tools for customized comparative studies; and d) provide information to investigators on available molecular reagents. This combination of features will facilitate cross-species comparisons of toxicologically significant genes and proteins. These comparisons will promote understanding of molecular evolution, the significance of conserved sequences, the genetic basis of variable sensitivity to environmental agents, and the complex interactions between the environment and human health. CTD is currently under development, and the planned scope and functions of the database are described herein. The intent of this report is to invite community participation in the development of CTD to ensure that it will be a valuable resource for environmental health, molecular biology, and toxicology research.

Exploiting Genome Data to Understand the Function Regulation and Evolutionary Origins of Toxicologically Relevant Genes

The wealth of new information coming from the many genome sequencing projects is providing unprecedented opportunities for major advances in all areas of biology, including the environmental health sciences. To facilitate this discovery process, experts in the fields of functional genomics and informatics and the emerging field of toxicogenomics recently gathered at the Mount Desert Island Biological Laboratory in Salisbury Cove, Maine, site of a National Institute of Environmental Health Sciences Marine and Freshwater Biomedical Science Center, to share their ideas and latest research findings. The goal of the symposium was to highlight approaches that may be used to identify and characterize toxicologically relevant genes being discovered in the genome sequencing projects. Many of the approaches rely heavily on comparative models as a way of identifying gene homology, ontology, and physiologic function, and on the availability of databases that facilitate storage, analysis, interpretation, and widespread dissemination of relevant data.
