Honokiol trimers and dimers via biotransformation catalyzed by Momordica charantia peroxidase: Novel and potent α-glucosidase inhibitors

Ten honokiol oligomers (1–10), including four novel trimers (1–4) and four novel dimers (5–8), were obtained by means of biotransformation of honokiol catalyzed by Momordica charantia peroxidase (MCP) for the first time. Their structures were established on the basis of spectroscopic methods. The biological results demonstrated that most of the oligomers were capable of inhibiting α-glucosidase with significant abilities, which were one to two orders of magnitude more potent than the substrate, honokiol. In particular, compound 2, the honokiol trimer, displayed the greatest inhibitory activity against α-glucosidase with an IC50 value of 1.38μM. Kinetic and CD studies indicated that 2 inhibited α-glucosidase in a reversible, mixed-type manner and caused conformational changes in the secondary structure of the enzyme protein. These findings suggested that 2 might be exploited as a promising drug candidate for the treatment of diabetes.

Dimerization of piceatannol by Momordicacharantia peroxidase and α-glucosidase inhibitory activity of the biotransformation products

Stilbenes, especially those oligomers, have great potential to be antihyperglycemic agents. In this study, eight stilbene dimers, including five new ones, were obtained by biotransformation of piceatannol using Momordicacharantia peroxidase (MCP) for the first time. Their structures were established on the basis of spectroscopic evidences. These piceatannol dimers displayed potential α-glucosidase inhibitory activities, and trans double bond, tetrahydrofuran ring, and free adjacent phenolic dihydroxyls were found to be important for their activities. Enzymatic biotransformation of stilbenes by M.charantia peroxidase (MCP) was showed to be a prominent way to produce oligomeric stilbenes for antihyperglycemic development.

Biocatalytic Production of Acyclic Bis[bibenzyls] from Dihydroresveratrol by Crude Momordica charantia Peroxidase

Biotransformation of dihydroresveratrol by crude Momordica charantia peroxidase provided six new acyclic bis[bibenzyls] 1-6. Their structures were established on the basis of NMR and MS analyses as C-C, C-O-C, and C-CH(2)-C dimers of dihydroresveratrol. Compounds 1-6 were tested for antiproliferative activity against human prostate cancer PC3 cell line in vitro, and 2 and 6 were found to be more potent than the parent compound.

Biotransformation of caffeic acid by Momordica charantia peroxidase

Biotransformation of caffeic acid with H2O2/Momordica charantia peroxidase at pH 5.0, 25 °C in the presence of acetone has resulted in the isolation of three caffeic acid trimers 1–3, five caffeic acid dimers 4–8, and one monomer 9. Among them, seven (1–7) are new compounds, and two (8 and 9) are known compounds, including one compound (9), which was obtained by biotransformation for the first time. The structures were established by 2D NMR techniques, such as HSQC, HMBC, and NOESY measurements. The possible mechanism for the formation of the products is also discussed. This is the first time that caffeic acid trimers have been obtained through the biotransformation of catalyzed by peroxidase in vitro. Compound 5 has shown much stronger anti-oxidative activity than the other caffeic acid polymers obtained.Key words: Momordica charantia peroxidase, caffeic acid, polymers of caffeic acid, biotransformation, antioxidant activity.

Biotransformation of Sinapic Acid Catalyzed by Momordica charantia Peroxidase

Biotransformation of sinapic acid (1) with H2O2/Momordica charantia peroxidase, which exists in the widely used food M. charantia, at pH 5.0, 43 degrees C, in the presence of acetone resulted in six compounds, including four new compounds. Compound 2 was the first isolation of a new unique sinapic acid tetrameric derivate, which is formed by peroxidase catalysis in vitro. Besides 2, three other new sinapic acid dimers, 3-5 have also been isolated. Their structures were established on the basis of spectroscopic data. Compound 5 showed a stronger antioxidative activity than the parent sinapic acid (1). Compounds 4 and 5 significantly inhibited the growth of HL-60 cell at the concentration of 10-5 microl/L.

Biotransformation of Ferulic Acid into Two New Dihydrotrimers by Momordica charantia Peroxidase

As a result of a continuing investigation of the biotransformation of ferulic acid with H2O2/Momordica charantia peroxidase at pH 5.0 in the presence of acetone, triFA1 (1) and triFA2 (2), two new FA dehydrotrimers, have been isolated. The structures were elucidated from the spectroscopic data including 2D-NMR such as HSQC, HMBC and NOESY data. TriFA1 (1) is a unique FA dehydrotrimer possessing a 3a,9b-dihydro-1H-furo[3,4-c][1]benzopyran-3(4H)-one ring system. The possible mechanism for the formation of triFA1 was also discussed.

Oxidation of Ferulic Acid by Momordica charantia Peroxidase and Related Anti-inflammation Activity Changes

Plant peroxidases were found to play an important role in plant physiology such as the metabolism and transformation of small complexes. In the present research, a novel Momordica charantia peroxidase (MCP) from fruits was purified to electrophoretic homogeneity by combining consecutive treatment of ammonium sulfate fractionation, ion exchange chromatography on DEAE-Sepharose FF, affinity chromatography on concanavalin A (Con A) Sepharose and gel filtration on Sephadex G-150. The physical and chemical characters of MCP were also investigated. MCP catalyzed the oxidation of ferulic acid (FA) to dehydrodimer (FA-2) in aqueous acetone system at pH 5.0. Its structure was identified by spectral analyses including IR, 1H-, 13C-NMR and electrospray ionization mass spectroscopy (ESI-MS). The anti-inflammatory activities of FA, FA-2 and other derivatives were examined. FA-2 significantly inhibited the release of proinflammatory factors such as TNF-alpha, NO and proliferation of spleen cells induced by phytohemagglutinin (PHA) and Con A and promoted a greater DNA fragmentation of spleen cells than that of other complexes. These results suggested that MCP as a tool enzyme transformed some complexes such as FA to more active derivatives, and that FA-2 was a potential inhibitor on inflammation through interference with immune response in the process of inflammation, which maybe was associated with apoptosis of immune related cells induced by FA-2.
