Engineering Escherichia coli-Derived Nanoparticles for Vaccine Development

The development of effective vaccines necessitates a delicate balance between maximizing immunogenicity and minimizing safety concerns. Subunit vaccines, while generally considered safe, often fail to elicit robust and durable immune responses. Nanotechnology presents a promising approach to address this dilemma, enabling subunit antigens to mimic critical aspects of native pathogens, such as nanoscale dimensions, geometry, and highly repetitive antigen display. Various expression systems, including Escherichia coli (E. coli), yeast, baculovirus/insect cells, and Chinese hamster ovary (CHO) cells, have been explored for the production of nanoparticle vaccines. Among these, E. coli stands out due to its cost-effectiveness, scalability, rapid production cycle, and high yields. However, the E. coli manufacturing platform faces challenges related to its unfavorable redox environment for disulfide bond formation, lack of post-translational modifications, and difficulties in achieving proper protein folding. This review focuses on molecular and protein engineering strategies to enhance protein solubility in E. coli and facilitate the in vitro reassembly of virus-like particles (VLPs). We also discuss approaches for antigen display on nanocarrier surfaces and methods to stabilize these carriers. These bioengineering approaches, in combination with advanced nanocarrier design, hold significant potential for developing highly effective and affordable E. coli-derived nanovaccines, paving the way for improved protection against a wide range of infectious diseases.

From Knallgas Bacterium to Promising Biomanufacturing Host: The Evolution of Cupriavidus necator.

The expanding field of synthetic biology requires diversification of microbial chassis to expedite the transition from a fossil fuel-dependent economy to a sustainable bioeconomy. Relying exclusively on established model organisms such as Escherichia coli and Saccharomyces cerevisiae may not suffice to drive the profound advancements needed in biotechnology. In this context, Cupriavidus necator, an extraordinarily versatile microorganism, has emerged as a potential catalyst for transformative breakthroughs in industrial biomanufacturing. This comprehensive book chapter offers an in-depth review of the remarkable technological progress achieved by C. necator in the past decade, with a specific focus on the fields of molecular biology tools, metabolic engineering, and innovative fermentation strategies. Through this exploration, we aim to shed light on the pivotal role of C. necator in shaping the future of sustainable bioprocessing and bioproduct development.

Innovative Bacterial Therapies and Genetic Engineering Approaches in Colorectal Cancer: A Review of Emerging Strategies and Clinical Implications.

Colorectal cancer (CRC) is considered a widespread cancer, ranking second in mortality and incidence among cancer patients worldwide. CRC develops from adenoma to carcinoma through the dynamic interplay of genetic and environmental factors. The conventional modes of treatment, including operation, chemotherapy, and irradiation, are associated with significant challenges, such as drug resistance and toxicity, necessitating the exploration of new treatment modalities. These difficulties reveal the necessity of the emergence of new therapeutic approaches. This review mainly emphasizes the bacterial-based therapies that have recently developed like the engineered bacteriophage therapy and bacterial immunotherapy that pale the existing chemotherapy in terms of toxicity but are effective in killing tumor cells. Also, it also investigates various molecular genetic engineering strategies such as CRISPR-Cas9, CRISPR prime editing and gene silencing to achieve better targeting of CRC. Implementing these new approaches into the forefront of CRC treatment may bring better, more effective therapy with fewer side effects on patients' quality of life.

Achieving Controllable Thermochromic Fluorescence via Synergistic Intramolecular Charge Transfer and Molecular Packing

Thermochromic fluorescent materials (TFMs) have attracted significant attention due to their unique fluorescent colorimetric response to temperature. However, existing TFMs still suffer from weak stimulus responsiveness, broad temperature response ranges, uncontrollable emission color changes, and low quantum yields. In this study, we address these issues by designing and synthesizing three diketone‐boron complexes with distinct emission wavelengths (NWPU‐(2‐4)). Utilizing a molecular engineering strategy to manipulate intramolecular charge transfer transitions and molecular packing modes, our synthesized complexes exhibit efficient fluorescence emission in both solution and solid states. Moreover, their emission wavelengths are highly sensitive to environmental polarity. By incorporating these compounds into thermosensitive matrices of long‐chain alkanes, we produced TFMs with varied fluorescence emission peak variation ranges. Notably, the TFM based on NWPU‐4, owing to its strong charge transfer transitions and dense J‐aggregate packing configuration, not only exhibits intense fluorescence emission spanning the deep red to near‐infrared spectrum but also displays a remarkable 90 nm broad range of thermochromic properties. Ultimately, it was successfully applied to programmable, thermally controlled, multi‐level information encryption.

Enhancing Magnetic Ordering in Two-Dimensional Metal-Organic Frameworks via Frontier Molecular Orbital Engineering.

Two-dimensional (2D) metal-organic frameworks (MOFs) have promise for use in lightweight permanent magnets in contrast to inorganic solid- or molecule-based magnets, but the realization of 2D MOF magnets with a high ordering temperature is limited by the typically weak spin exchange interactions. Here, we have proposed a frontier molecular orbital engineering strategy for modulating magnetism in 2D MOFs. It shows that the magnetic ground state can be mediated by two intra-atomic spin exchange pathways in organic ligands, akin to the Bloch and Heisenberg models, depending on the shape of the frontier orbitals of the organic ligands. By engineering the shape of the lowest unoccupied molecular orbital (LUMO) via chemical hydrogenation, we achieved a nearly 11-fold increase in the ordering temperature. In particular, a quantitative analysis shows that the ordering temperature increases linearly with the orbital delocalization index of the ligands' LUMO. This work suggests a general frontier orbital engineering approach for modulating the spin exchange interaction in 2D MOF magnets.

Distortion Engineering: A Strategy to Modulate Molecular Upconversion with Molecular Cluster-Aggregates.

The rational engineering of molecules is a powerful chemistry tool of pivotal importance in the fields of molecular magnetism and luminescence. Hence, systems that can be modulated via molecular engineering and composition control are expected to present extra versatility regarding the tunability of their properties. This is the case with molecular cluster aggregates (MCAs), high nuclearity molecular compounds. Herein, we demonstrate how the union of both strategies, namely, composition control and molecular engineering, can be employed to enhance molecular upconversion in MCAs. This was achieved by doping a {Gd8Er2Yb10} MCA with CeIII ions. By replacement of the optically silent GdIII ions with CeIII, the upconversion mechanism is modified due to CeIII-mediated cross-relaxation. In addition to this effect, we could also engineer the degree of metal site distortion due to the larger size of CeIII ions, relaxing the selection rules and impacting the upconversion quantum yield and luminescent thermometry. Opto-structural correlations demonstrate that the presented molecular engineering strategy can be used to enhance the performance of molecular upconverters.

Ultrahigh Responsivity and Robust Semiconducting Fiber Enabled by Molecular Soldering-Governed Defect Engineering for Smart Textile Optoelectronics.

Semiconducting fibers (SCFs) are of significant interest to design next-generation wearable and comfortable optoelectronics that seamlessly integrate with textiles. However, the practical applications of current SCFs are always limited by poor optoelectronic performance and low mechanical robustness caused by uncontrollable multiscale structural defects. Herein, a versatile in situ molecular soldering-governed defect engineering strategy is proposed to construct ultrahigh responsivity and robust wet-spun MoS2 SCFs, by using a π-conjugated dithiolated molecule to simultaneously patch microscale sulfur vacancies within MoS2 nanosheets, diminish mesoscale interlayer voids/wrinkles, promote macroscale orientation, build long-range photoelectron percolation bridges, and provide n-doping effect. The derived MoS2 SCFs exhibit over two orders of magnitude higher responsivity (144.3 A W-1) than previously reported fiber photodetectors, 37.3-fold faster photoresponse speed (52ms) than pristine counterpart, and remarkable bending robustness (retain 94.2% of the initial photocurrent after 50000 bending-flattening cycles). Such superior robustness and photodetection capacity of MoS2 SCFs further enable large-scale weaving of reliable smart textile optoelectronic systems, such as direction-identifiable wireless light alarming system, modularized mechano-optical communication system, and indoor light-controlled IoT system. This work offers a universal strategy for the scalable production of mechanically robust and high-performance SCFs, opening up exciting possibilities for large-scale integration of wearable optoelectronics.

Boosting Zn-Ion Storage Behavior of Pre-Intercalated MXene with Black Phosphorus toward Self-Powered Systems.

MXene-based Zn-ion capacitors (ZICs) with adsorption-type and battery-type electrodes demonstrate high energy storage and anti-self-discharge capabilities, potentially being paired with triboelectric nanogenerators (TENGs) to construct self-powered systems. Nevertheless, inadequate interlayer spacing, deficient active sites, and compact self-restacking of MXene flakes pose hurdles for MXene-based ZICs, limiting their applications. Herein, black phosphorus (BP)-Zn-MXene hybrid is formulated for MXene-based ZIC via a two-step molecular engineering strategy of Zn-ion pre-intercalation and BP nanosheet assembly. Zn ions as intercalators induce cross-linking of MXene flakes with expandable interlayer spacing to serve as scaffolds for BP nanosheets, thereby providing sufficient accessible active sites and efficient migration routes for enhanced Zn-ion storage. The density functional theory calculations affirm that zinc adsorption and diffusion kinetics are significantly improved in the hybrid. A wearable ZIC with the hybrid delivers a competitive areal energy of 426.3µWhcm-2 and ultra-low self-discharge rate of 7.0mVh-1, achieving remarkable electrochemical matching with TENGs in terms of low energy loss, matched capacity, and fast Zn-ion storage. The resultant self-powered system efficiently collects and stores energy from human motion to power microelectronics. This work advances the Zn-ion storage of MXene-based ZICs and their synergy with TENG in self-powered systems.

Molecular Engineering of Multivariate Porous Aromatic Frameworks for Recovery of Dispersed Uranium Resources

AbstractUranium plays an indispensable role in the sustainable development of nuclear industry, and the recovery of uranium from nuclear wastewater is one way to solve the shortage of uranium resources. However, the recovery of uranium resources from nuclear wastewater remains a challenge due to the interfere of coexisting ions. Herein, ratios of the adsorption group and photocatalytic enrichment group are well‐designed and controllably assembled into the porous aromatic frameworks (PAFs) using molecular engineering strategy to realize high uranium recovery efficiency and high resistance to interfering ions. The optimally coordinated multivariate PAF material PAF‐AN2T8‐AO exhibits bifunctions of adsorption and photocatalytic enrichment and thus, achieves high enrichment selectivity and high uranium recovery efficiency of 99% under simulated sunlight irradiation. Compared to PAFs with only adsorption group or photocatalytic group, the uranium recovery capacity of the optimized PAF is enhanced by 1.2 times and 2.3 times, respectively. Mechanistic studies reveal that the product of photocatalytic uranium on PAF‐AN2T8‐AO is air‐stabilized solid crystal metastudtite ((UO2)O2∙2H2O). This work provides a guiding method for designing adsorption‐photocatalytic bifunctional materials and proposes a more environmentally adaptable strategy for dispersed uranium resource recovery.

A Molecular Engineered Strategy to Remolding Architecture of RuP2 Nanoclusters for Sustainable Hydrogen Evolution

AbstractTransition metal phosphides (TMPs) are promising hydrogen evolution reaction (HER) electrocatalysts, but the unsatisfying activity and durability at industrial‐scale current densities hinder their application. Downsizing TMPs to nanoclusters can substantially enhance their activity; yet, the high surface free energy tends to initiate agglomeration thereby limiting their service life. Herein, a molecular engineering strategy to synthesize robust sulfur‐doped RuP2 (S‐RuP2) nanoclusters, which can continuously provide ampere‐level current densities of 1.0, 2.0, and 3.0 A cm−2 for at least 480 h with low overpotentials of 55.6 ± 1.0, 90.6 ± 1.2, and 122.8 ± 1.0 mV, respectively, is proposed. In particular, it exhibits a remarkable charge transfer amount (representing a long service life), outperforming all reported alkaline HER electrocatalysts. Remoulding the RuP2 architecture by sulfur atom can significantly lift the d‐band center of Ru and the p‐band center of P, therefore strengthening the adsorption of H2O, H, and OH to reduce the barriers of water dissociation and H migration. The authors‘ research unveils the enormous potential of molecule‐based porous materials for designing high‐performance nano‐structured catalysts.

Remotely Modulating the Optical Properties of Organic Charge-Transfer Crystallites via Molecular Packing.

Organic charge-transfer complex (CTC) formation has emerged as an effective molecular engineering strategy for achieving the desired optical properties via intermolecular interactions. By synthesizing organic CTCs with carbazole-based electron donors and a 7,7,8,8-tetracyanoquinodimethane acceptor and adopting a molecular linker located remotely from the charge-transfer interface within the donors, we were able to modulate near-infrared absorptive and short-wavelength infrared emissive properties. Structural characterizations performed by using single-crystal X-ray diffraction confirmed that the unique molecular arrangements induced by the steric hindrance from the remotely located linker significantly influence the electronic interactions between the donor and acceptor molecules, resulting in different photophysical properties. Our findings offer an improved understanding of the interplay between molecular packing and optoelectronic properties, providing a foundation for designing advanced materials for optoelectronic applications.

Dual–Acceptor Engineering in Pyrene‐Based Covalent Organic Frameworks for Boosting Photocatalytic Hydrogen Evolution

AbstractThe integration of electron donor (D) and acceptor (A) units into covalent organic frameworks (COFs) has received increasing interest due to its potential for efficient photocatalytic hydrogen (H2) evolution from water. Nevertheless, the advancement of D–A COFs is still constrained by the limited investigations on acceptor engineering, which enables the highly effective charge transfer pathways in COFs to deliver photoexcited electrons in a preferential orientation to enhance photocatalytic performance. Herein, two systems with D–A and D–A–A configurations based on the acceptor molecular engineering strategy are proposed to construct three distinct COFs. Specifically, TAPPy‐DBTDP‐COF merging one pyrene‐based donor and two benzothiadiazole acceptors realized an average H2 evolution rate of 12.7 mmol h−1 g−1 under visible light, among the highest ever reported for typical D–A‐type COF systems. The combination of experimental and theoretical analysis signifies the crucial role of the dual‐acceptor arrangement in promoting exciton dissociation and carrier migration. These findings underscore the significant potential of D–A–A structural design, which is conducive to the efficient separation of photoexcited electrons and holes resulting in superior photocatalytic activities.

Side-chain-engineered fluorescent dyes for 3D and long-term dynamic tracking of the plasma membrane in living cells

The plasma membrane involves in many important biological events such as cell fusion and programmed cell death, but most of current plasma membrane probes cannot meet the requirement of long-term specific anchoring to the plasma membrane. Herein, we propose a molecular side-chain engineering strategy to modulate the long-term imaging performance of fluorescent dyes to the plasma membrane by regulating the cell permeability and anchoring ability. A series of FMR dyes with different lengths of side chains were designed and synthesized, and their transmembrane behaviours and staining performance were evaluated in living HeLa cells. We found that short-chain and medium-chain FMR dyes have excellent cell permeability without the labeling ability to the plasma membrane while the long-chain FMR dyes specifically stain the plasma membrane and can be firmly anchored to the plasma membrane for a long period of time. These long-chain FMR dyes have high stain specificality to the plasma membrane, and C10-FMR can be anchored to the plasma membrane of living cells for 2 h, which enables it to continuously monitor dynamic changes of the plasma membrane. The three-dimensional precision imaging of various cells was achieved using C10-FMR, which provides an opportunity to obtain complete information on the three-dimensional spatial morphology of the plasma membrane. The PEG-induced cell fusion of chicken red blood cells and H2O2-induced apoptosis of HeLa cells were monitored by real-time tracking of dynamic changes of the plasma membrane during these processes, which provide solid examples to prove the usefulness of these fluorescent dyes as long-term imaging tools. This work validates the hypothesis that cell permeability of membrane dyes can be readily regulated by tuning the side chains, and provides the effective design strategy of fluorescent dyes for 3D and long-term dynamic tracking of the plasma membrane of diverse animal cells.

Molecular Engineering Strategies of Spectral Matching and Structure Optimization for Efficient Metal-Free Organic Dyes in Dye-Sensitized Solar Cells: A Theoretical Study.

Metal-free organic dyes are promising dyes that can be applied widely in dye-sensitized solar cells (DSSCs). The rational design and selection of dyes with complementary absorption can promote the development of methods that can enhance the utilization of incident light by DSSCs, such as cosensitization and tandem devices. Based on these opinions, the structure of the reported high-performance metal-free organic dye ZL003 is used as a template to design two new metal-free organic dyes, HX-1 and HX-2, by replacing its donor unit with a 2-phenothiazine-phenylamine unit and fusing its three independent π-bridge units into a whole with the aim of driving the red shift and the blue shift of the absorption spectra of ZL003, respectively. Through theoretical investigation, it is demonstrated that the perfect complementary optical absorption of HX-1 and HX-2 can be realized as the shift of the absorption spectra of ZL003 to different directions, which means their feasibility to the application in cosensitization or tandem dye-sensitized solar cells (T-DSSCs). Furthermore, it is hypothesized that HX-1 may be the dye with better photovoltaic performance than ZL003 by modeling their intramolecular charge-transfer (ICT) processes, TiO2 surface adsorption, and photovoltaic parameters. The short-circuit current density (Jsc) and photoelectric conversion efficiency (PCE) of HX-1 are 23.10 mA·cm-2 and 21.26% in theory, compared to those of 20.68 mA·cm-2 and 19.64% in ZL003 at the same computational level, respectively. In view of the complementary optical properties, the combination of HX-1 with HX-2 may be a reasonable option for dyes for the development of a highly efficient cosensitization system or T-DSSCs in the future. In terms of such findings, these two novel metal-free organic dyes may have bright prospects in the research of highly efficient DSSCs, and this work can provide a reference for the design of dyes with complementary absorption through simple structural adjustments of the realistic dyes with high photovoltaic performance.

A Molecular Engineering Approach to Achieve Wavelength-Selective Photorelease Using a Dynamic Photocage.

We introduce a molecular engineering strategy for wavelength-selective photorelease of alcohols and carboxylic acids, employing a dynamic photocage featuring an (E)-3-(6-acetyl-2-hydroxynaphthalen-1-yl)acrylate moiety. Initially activated by visible light (λ ≥ 410 nm), it releases alcohol, generating a second photocage. Subsequent exposure to shorter wavelengths (λ ≤ 365 nm) selectively releases carboxylic acid, albeit at a reduced rate.

A Review on Small Molecule Based Fluorescence Chemosensors for Bioimaging Applications.

This review provides a thorough examination of small molecule-based fluorescence chemosensors tailored for bioimaging applications, showcasing their unique ability to visualize biological processes with exceptional sensitivity and selectivity. It explores recent advancements, methodologies, and applications in this domain, focusing on various designs rooted in anthracene, benzothiazole, naphthalene, quinoline, and Schiff base. Molecular Engineering of Hydrogen-Bonded Organic Framework for Enhanced Nitrate Electroreduction to Ammonia.

Electrocatalytic nitrate reduction is an efficient way to produce ammonia sustainably. Herein, we rationally designed a copper metalloporphyrin-based hydrogen-bonded organic framework (HOF-Cu) through molecular engineering strategies for electrochemical nitrate reduction. As a result, the state-of-the-art HOF-Cu catalyst exhibits high NH3 Faradaic efficiency of 93.8%, and the NH3 production rate achieves a superior activity of 0.65 mmol h-1 cm-2. The in situ electrochemical spectroscopic combined with density functional theory calculations reveals that the dispersed Cu promotes the adsorption of NO3- and the mechanism is followed by deoxidation of NO3- to *NO and accompanied by deep hydrogenation. The generated *H participates in the deep hydrogenation of intermediate with fast kinetics as revealed by operando electrochemical impedance spectroscopy, and the competing hydrogen evolution reaction is suppressed. This research provides a promising approach to the conversion of nitrate to ammonia, maintaining the nitrogen balance in the atmosphere.

Defect Engineering of MoTe2 via Thiol Treatment for Type III van der Waals Heterojunction Phototransistor.

Molybdenum ditelluride (MoTe2) nanosheets have displayed intriguing physicochemical properties and opto-electric characteristics as a result of their tunable and small band gap (Eg ∼ 1 eV), facilitating concurrent electron and hole transport. Despite the numerous efforts devoted to the development of p-type MoTe2 field-effect transistors (FETs), the presence of tellurium (Te) point vacancies has caused serious reliability issues. Here, we overcome this major limitation by treating the MoTe2 surface with thiolated molecules to heal Te vacancies. Comprehensive materials and electrical characterizations provided unambiguous evidence for the efficient chemisorption of butanethiol. Our thiol-treated MoTe2 FET exhibited a 10-fold increase in hole current and a positive threshold voltage shift of 25 V, indicative of efficient hole carrier doping. We demonstrated that our powerful molecular engineering strategy can be extended to the controlled formation of van der Waals heterostructures by developing an n-SnS2/thiol-MoTe2 junction FET (thiol-JFET). Notably, the thiol-JFET exhibited a significant negative photoresponse with a responsivity of 50 A W-1 and a fast response time of 80 ms based on band-to-band tunneling. More interestingly, the thiol-JFET displayed a gate tunable trimodal photodetection comprising two photoactive modes (positive and negative photoresponse) and one photoinactive mode. These findings underscore the potential of molecular engineering approaches in enhancing the performance and functionality of MoTe2-based nanodevices as key components in advanced 2D-based optoelectronics.
