Antiglycation and antioxidant activity of herbal medicines used in the treatment of climacteric symptoms

ObjectivesThis work had the objective to evaluate the antiglycation and antioxidant potential of Cimicifuga racemosa L., Trifolium pratense L., Humulus lupulus L. and Morus nigra L., commonly used in traditional medicine for the treatment of climacteric symptoms. MethodsAntioxidant activity was evaluated by 2,2-Diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, ferric reducing antioxidant power, inhibition of lipid peroxidation thiobarbituric acid reactive substances (TBARS), nitric oxide (NO) radical scavenging and inhibition of oxidative haemolysis test. The evaluation of antiglycation activity was performed by the determination of relative electrophoretic mobility of bovine serum albumin (BSA) with ribose and BSA with methylglyoxa in the presence and absence of Cu2+ ion. Total polyphenol and flavonoid contents were determined by spectrophotometric methods. ResultsThe highest antioxidant activity for DPPH and ferric reducing antioxidant power test was detected in Humulus lupulus (83.56% and 46.95 µM TE/g dry extract, respectively). For the inhibition of lipid peroxidation, the lowest percentages of TBARS were found in M. nigra (4.82%) and H. lupulus (18.26%). C. racemosa presented the lowest nitrite formation (34.57 µmol/mL nitrite formed) in nitric oxide test. T. pratense presented the highest polyphenol (24.61 mg GAE/g dry extract) and flavonoid (22.79 mg RE/g dry extract) contents. For the oxidative haemolysis assay, the extracts showed concentration-dependent inhibitory capacity, while for the antiglycation activity all extracts showed antiglycation potential against glycation of BSA promoted by Ribose evaluated by the electrophoretic profile. ConclusionsIn view of the results obtained in this study, it was possible to verify that the dry extracts of herbal medicines used for climacteric symptoms present antioxidant and antiglycation activities, mainly due to the presence of phenols compounds in their composition.

Binding of serum albumin to the anthelmintic drugs albendazole, triclabendazole and their sulphoxides

The binding of drugs to plasma proteins – especially serum albumin – is an important factor in controlling the availability and distribution of these drugs. In this study we have investigated the binding of two drugs commonly used to treat liver fluke infections, albendazole (ABZ) and triclabendazole (TCBZ), and their sulphoxide metabolites to bovine serum albumin (BSA). Both ABZ and TCBZ caused shifts in the mobility of BSA in native gel electrophoresis. No such changes were observed with the sulphoxides under identical conditions. The drugs, and their sulphoxides, caused quenching of the intrinsic tryptophan fluorescence of BSA, indicating association between the drugs and this protein. Quantification of this quenching suggested a 5–10-fold reduction in affinity of the sulphoxides compared to the parent compounds. These results are discussed in respect to previous work on the pharmacodynamics of these fasciolicides and will inform the design of novel anthelmintics.

Antifungal activity of cerebrospinal fluid against Cryptococcus neoformans and Candida species

The effect of human cerebrospinal fluid (CSF) on the growth of Cryptococcus neoformans and Candida species was tested in RPMI-1640. CSF alone was highly fungistatic for both yeasts and inhibited growth in a concentration-dependent manner. Unlike human serum, CSF did not collaborate with fluconazole for killing C. neoformans. Molecular sieve fractionation of CSF on a G-200 Sephadex column yielded a highly antifungal fraction with a molecular weight around 66 kDa. On SDS-PAGE this fraction migrated as a major and a minor band corresponding to the mobility of bovine serum albumin. These novel findings suggest that CSF contains a factor(s) that provides resistance to the growth of C. neoformans or Candida species.

Multichannel Flow Electrophoresis in an Alternating Electric Field

Abstract Investigation on the mass transfer behavior of multichannel flow electrophoresis (MFE) has confirmed the existence of concentration polarization in the separation process, which is characterized by a reduction of the relative protein migration flux corresponding to an increase in the protein concentration in the feed. The mobility of bovine serum albumin (BSA) in a gel membrane, as interpreted from its elution curve, was about one-sixth of its mobility in solution as determined by high capillary electrophoresis. This indicates that accumulation of charged protein onto the membrane surface occurs in MFE. An alternating electric field was applied instead of the steady one in MFE. The negative part of the alternating electric field reduced the polarization layer periodically. Continuous Separation of Proteins by Multichannel Flow Electrophoresis

Abstract A new preparative electrophoresis method, multichannel flow electrophoresis (MFE), is proposed in this paper. Mechanisms for different operation modes of MFE are described and a simulation of continuous MFE is proposed. A membrane spaced 5-compartment electrolyzer is developed in which the central compartment and its neighboring compartments are respectively used for sample loading and product discharging. The mass transfer behavior of MFE is investigated by measuring the equivalent migration speed and mobility of bovine serum albumin (BSA) and hemoglobin bovine blood (HBB) at different buffering pH, electric field strength, and sample loading flow rate. The continuous separation of a mixture composed of BSA and HBB, buffered with 0.01 M Tns-HAc at pH 5.9, yields 13.2 mg BSA and 20.0 mg HBB per hour.
