Automated measurement of blood flow velocity and direction and hemoglobin oxygen saturation in the rat lung using intravital microscopy.

Intravital microscopy of the pulmonary microcirculation in research animals is of great scientific interest for its utility in identifying regional changes in pulmonary microcirculatory blood flow. Although feasibility studies have been reported, the pulmonary window can be further refined into a practical tool for pharmaceutical research and drug development. We have established a method to visualize and quantify dynamic changes in three key features of lung function: microvascular red blood cell velocity, flow direction, and hemoglobin saturation. These physiological parameters were measured in an acute closed-chest pulmonary window, which allows real-time images to be captured by fluorescence and multispectral absorption microscopy; images were subsequently quantified using computerized analysis. We validated the model by quantifying changes in microcirculatory blood flow and hemoglobin saturation in two ways: 1) after changes in inspired oxygen content and 2) after pharmacological reduction of pulmonary blood flow via treatment with the β1 adrenergic receptor blocker metoprolol. This robust and relatively simple system facilitates pulmonary intravital microscopy in laboratory rats for pharmacological and physiological research.

Inhibition of Leukocyte Adherence Enables Venular Control of Capillary Perfusion in Streptozotocin‐Induced Diabetic Rats

Vasoactive molecules can diffuse from venules to dilate closely paired arterioles and enhance capillary perfusion. Venular control of capillary flow has been found to be dependent on nitric oxide (NO), which might be scavenged rapidly in diabetic microvasculature due to the presence of activated leukocytes. This study attempts to improve venular control of capillary flow using fucoidan, which inhibits venular leukocvte adhesion. Microvascular red blood cell velocity was measured in the mesentery of streptozotocin-induced diabetic rats, with and without fucoidan treatment, and in normal rats. Arteriolar pathways leading to branching capillaries were videotaped to measure the percent of the surrounding area occupied by a venule (% pairing). Microvascular wall NO was measured using fluorescent diaminofluorescein-2-diacetate in diabetic rats, with and without fucoidan treatment. In normal rats, close pairing of venules to arterioles resulted in faster capillary flow. However, after 4-5 weeks of diabetes, the correlation between capillary velocity and % pairing was no longer significant. Capillary velocity and % pairing decreased approximately 50% in comparison to normal rats. Treatment of diabetic rats with fucoidan restored venular control of capillary flow and increased NO levels. Leukocyte-derived mediators that scavenge NO may lead to inadequate venular control of capillary flow in diabetes.

New Model for in Vivo Quantification of Microvascular Embolization, Thrombus Formation, and Recanalization in Composite Flaps

Background. Microthrombi are suggested to be involved in the pathogenesis of composite flap failure. Due to the lack of appropriate experimental models, however, the significance of microvascular thrombus formation and microthromboembolization in free flap failure remains poorly understood. The purpose of this study was therefore to develop a rat hindlimb model that allows tissue-confined in vivo analysis of thrombus formation, thromboembolization, and recanalization within the microcirculation of osteomyocutaneous flaps using intravital fluorescence microscopy. Materials and methods. Thrombus formation was induced photochemically in individual arterioles and venules of muscle, subcutis, and periosteum. To study thromboembolization, autologous arterial thrombi (40 μm) were preformed in vitro and were injected into the femoral artery supplying the osteomyocutaneous flap. Results. First platelet deposition was found independent from microvascular red blood cell velocity, while the subsequent growth of thrombus correlated inversely with red blood cell flow measured in the respective microvessel. Time required for complete thrombotic arteriolar occlusion exceeded 700 s, whereas thrombus growth in venules was found to be significantly accelerated (∼300 s) without differences between the individual tissues analyzed. The embolization resulted in a complete shutdown of capillary perfusion in muscle, subcutis, skin, and periosteum. During subsequent spontaneous recanalization, capillary perfusion increased in all tissues to approximately half of baseline, however, without further recovery during the 4-h postembolization period. Conclusion. The model presented is suitable to quantitatively study the pathophysiology of microvascular thrombus formation, thromboembolization, and recanalization in composite flaps, and may thus be used to evaluate the effectiveness of novel therapeutic strategies to prevent flap failure.

Systemic and subcutaneous microvascular Po2 dissociation during 4-h hemorrhagic shock in conscious hamsters.

The relationship between systemic and microvascular Po2 in subcutaneous connective tissue and hemodynamics was investigated during 4-h hemorrhagic shock (40 mmHg) in conscious Syrian hamsters (n = 66) fitted with a dorsal skinfold window. Systemic blood gases, metabolic parameters, arteriolar, venular, and tissue Po2, microvascular red blood cell velocity, and blood flow were evaluated in survivors (S) and nonsurvivors (NS). Surviving animals were resuscitated with shed blood. Microvascular and tissue Po2 were measured by phosphorescence decay of Pd-meso-tetra (4-carboxyphenyl) porphyrin (30 mg/kg body wt iv). Shock caused a significant dissociation between systemic arterial and microvascular arteriolar Po2 levels. Arterial Po2 increased from 59.7 +/- 12.0 to 110.8 +/- 19.7 mmHg (S) and from 64.0 +/- 13.7 to 128.5 +/- 10.1 mmHg (NS), whereas Po2 in large arterioles decreased from 56.9 +/- 5.5 (control) to 29.5 +/- 20.1 (S) and 6.0 +/- 5.7 mmHg (NS). Correspondingly, tissue Po2 fell from 24.1 +/- 6.8 (control) to 0.9 +/- 0.6 (S) and 0.4 +/- 0.3 mmHg (NS). Venous Po2 decreased from 28.8 +/- 3.7 to 20.4 +/- 4.1 (S) and from 28.0 +/- 2.9 to 16.3 +/- 0.5 mmHg (NS). Shock outcome and tissue oxygenation were predicted by arterial blood gases and metabolic and microcirculatory conditions but not by central venous Po2.
