Event Abstract Back to Event Hippocampal injection of in vitro aggregated Tau fragments initiates tauopathy and neuronal loss. Astrid Bottelbergs1, Eve Peeraer1, Diederik Moechars1* and Xavier Langlois1 1 Janssen Pharmaceutica, Belgium Along with amyloid plaques, neurofibrillary tangles are a hallmark of Alzheimer’s disease. In the TauP301L mouse model these tangles, mainly composed of tau, are mimicked. A major disadvantage of this model is the late onset of tauopathy. To accelerate pathology, three month old TauP301L mice were injected with in vitro aggregated K18, a fragment of the microtubule binding domain of tau. These fragments were injected into the hippocampus or cortex respectively and the brains were investigated at different time points after injection, mainly by immunohistochemistry. Phosphorylated tau, recognized by the AT8 antibody, was observed within neurons from two weeks after injection. Initially, AT8 was only observed at the site of injection, followed by a spread toward other brain regions. After cortical injection, the maximum number of AT8 positive cells was reached at one month after injection, followed by a steady-state at two and three months. Hippocampal injection also resulted in pronounced AT8 immunoreactivity at one month after injection. However, this was followed by a decrease at two and three months, mainly in the CA1 region of the hippocampus. The decreased number of AT8 positive neurons in the hippocampus correlates with neuronal loss, as indicated by reduced neuronal cell soma (NeuN) and dendrite (MAP2) labeling. Acute degenerating neurons were observed by Fluorojade B staining at one month after injection. In summary, injection of aggregated K18 fragments into the hippocampus or cortex triggers tauopathy. In the hippocampus tauopathy results in neuronal loss.
