Assessing biomass and primary production of microphytobenthos in depositional coastal systems using spectral information.

In depositional intertidal coastal systems, primary production is dominated by benthic microalgae (microphytobenthos) inhabiting the mudflats. This benthic productivity is supporting secondary production and supplying important services to humans including food provisioning. Increased frequencies of extreme events in weather (such as heatwaves, storm surges and cloudbursts) are expected to strongly impact the spatiotemporal dynamics of the microphytobenthos and subsequently their contribution to coastal food webs. Within north-western Europe, the years 2018 and 2019 were characterized by record-breaking summer temperatures and accompanying droughts. Field-calibrated satellite data (Sentinel 2) were used to quantify the seasonal dynamics of microphytobenthos biomass and production at an unprecedented spatial and temporal resolution during these years. We demonstrate that the Normalized Difference Vegetation Index (NDVI) should be used with caution in depositional coastal intertidal systems, because it may reflect import of remains of allochthonous pelagic productivity rather than local benthic biomass. We show that the reduction in summer biomass of the benthic microalgae cannot be explained by grazing but was most probably due to the high temperatures. The fivefold increase in salinity from January to September 2018, resulting from reduced river run-off during this exceptionally dry year, cannot have been without consequences for the vitality of the microphytobenthos community and its resistance to wind stress and cloud bursts. Comparison to historical information revealed that primary productivity of microphytobenthos may vary at least fivefold due to variations in environmental conditions. Therefore, ongoing changes in environmental conditions and especially extreme events because of climate change will not only lead to changes in spatiotemporal patterns of benthic primary production but also to changes in biodiversity of life under water and ecosystem services including food supply. Satellite MPB data allows for adequate choices in selecting coastal biodiversity conservation and coastal food supply.

Spatiotemporal variability in microphytobenthic primary production across bare intertidal flat, saltmarsh, and mangrove forest of Asia and Australia

The ecological role of intertidal microphytobenthos (MPB) is increasingly recognized in coastal production systems. MPB primary production (PP) measured in coastal wetlands of Korea, Cambodia, and Australia confirmed large variability at the global scale. Surprisingly, MPB biomass in mangrove forests almost doubled those measured in nearby bare tidal flats. However, MPB productivity (Pb) in vegetated habitats was significantly reduced (by ~50%) compared to that on bare tidal flats. Extensive measurements of MPB biomass, PP, and Pb across 12 Korean tidal flats revealed large spatiotemporal variations, suggesting complex sediment-MPB coupled dynamics. The key factors included sediment type, tide, bed elevation, irradiation, temperature, and vegetation. Winter MPB blooms and the elevated Pb seem to be unique characteristics of the Korean intertidal flats. The present study provides the baseline data of MPB PPs in mudflat, saltmarsh, and mangrove habitats in the highly productive zones of the Western Indo-Pacific Rim.

Water column dissolved silica concentration limits microphytobenthic primary production in intertidal sediments.

Primary production of microphytobenthos (MPB) contributes significantly to the total production in shallow coastal environments. MPB is a diverse community in which diatoms are usually the main microalgal group. Diatoms require N, P, and other nutrients as with other autotrophs, but in addition require silicate to create their outer cell wall. Therefore, dissolved silica (DSi) might be a potential limiting factor for benthic primary production in areas with reduced freshwater input. To test this hypothesis, a microcosm experiment was conducted using intact sediment cores collected from an intertidal mudflat in the Bay of Cádiz and supplied with increasing concentrations of DSi (0, 5, 10, 25, and 45μmol·L-1 ). After 7d of enrichment, we determined chlorophyll a and c (Chl a, c) contents, metabolic rates (Net [Pn ] and Areal Gross [PgA ] Production and Light [RL ] and Dark [RD ] Respiration), as well as fluxes of inorganic nutrients across the sediment-water interface. Chl a and c contents increased significantly with respect to the initial conditions but no differences between treatments were found. Both Pn and PgA showed a saturating-like pattern with silicate concentration, reaching maximum rates at a DSi concentration of 45μmol·L-1 . The addition of DSi also resulted in an increase of DSi and ammonium uptake by the sediment, which was significantly higher in light than in darkness. Our results clearly show that water column DSi concentrations have a direct impact on benthic primary production, also controlling other related processes such as inorganic nutrient fluxes.

Development of temperature-based algorithms for the estimation of microphytobenthic primary production in a tidal flat: A case study in Daebu mudflat, Korea

This study presents the results of field experiments that were designed to investigate the photophysiological characteristics of microphytobenthos (MPB) and to estimate primary production (PP) in Daebu mudflat, which is located at the west coast of Korea. A typical seasonal (or monthly) fluctuation of intertidal MPB PP was found in association with biotic (benthic Chl-a) and/or abiotic parameters (irradiance and temperature) over a period of three years. From a series of field-laboratory experiments using the oxygen micro-profiling method (totaling 28 surveys), three consistent phenomena were observed: 1) winter to early spring algal blooms, 2) seasonal changes in Q10, and 3) temperature dependent MPB photosynthesis-irradiance (P-I). In particular, both the chlorophyll-specific maximum photosynthetic capacity (Pbmax) and the saturated light intensity (Ik), derived from 126 P-I curves (1870 data sets of oxygen micro-profiling in the sediment), were significantly correlated with sediment temperature (p < 0.01). To develop an empirical MPB PP model, the relationships between P-I parameters and environmental variables were parameterized following established exponential forms (e.g., Q10). It was possible to estimate the MPB PP in Daebu mudflat area by using easily accessible explanatory factor, suitable to be used for future explorations of parameters such as sediment temperature, irradiance, chlorophyll concentration, and tidal height. The estimated annual MPB PP based on the empirical PP model were found to be greater than that in the Wadden Sea and average annual PP in the temperate zones of the world. Authors believe that the present approach of the MPB PP estimation could be combined with remote-sensing techniques (e.g., satellites) to support coastal ecosystem management.

Microphytobenthos primary production estimated by hyperspectral reflectance.

The use of remote sensing techniques allows monitoring of photosynthesis at the ecosystem level and improves our knowledge of plant primary productivity. The main objective of the current study was to develop a remote sensing based method to measure microphytobenthos (MPB) primary production from intertidal mudflats. This was achieved by coupling hyperspectral radiometry (reflectance, ρ and second derivative, δδ) and PAM-fluorometry (non-sequential light curves, NSLC) measurements. The latter allowed the estimation of primary production using a light use efficiency parameter (LUE) and electron transport rates (ETR) whereas ρ allowed to estimate pigment composition and optical absorption cross-section (a*). Five MPB species representative of the main growth forms: epipelic (benthic motile), epipsammic (benthic motile and non motile) and tychoplanktonic (temporarily resuspended in the water column) were submitted to increasing light intensities from dark to 1950 μmol photons.m-2.s-1. Different fluorescence patterns were observed for the three growth-forms and were linked to their xanthophyll cycle (de-epoxydation state). After spectral reflectance measurements, a* was retrieved using a radiative transfer model and several radiometric indices were tested for their capacity to predict LUE and ETR measured by PAM-fluorometry. Only one radiometric index was not species or growth-form specific, i.e. δδ496/508. This index was named MPBLUE and could be used to predict LUE and ETR. The applicability of this index was tested with simulated bands of a wide variety of hyperspectral sensors at spectral resolutions between 3 and 15 nm of Full Width at Half Maximum (FWHM).

The Relationship between Primary Production of Microphytobenthos and Tidal Cycle on the Hwaseong Mudflat, West Coast of Korea

The relationship between primary production of microphytobenthos and tidal cycle on the Hwaseong mudflat, west coast of Korea.The present study examined variation in sediment temperature and irradiance with respect to microphytobenthos primary production at the Hwaseong mudflat, west coast of Korea, under different tidal cycles. The study was carried out during March and April 2009, which encompassed two spring tides and one neap tide. It was not possible to continuously measure primary production in situ across the tidal cycles; hence, photosynthesis–irradiance (P-I) curves at different temperatures were obtained experimentally in the laboratory. These readings were then used to calculate primary production, by projecting in situ mooring records of irradiance and temperature onto the P-I curves. The sediment temperature and irradiance were measured continuously, and the averages of 10-minute intervals from the mooring records were used. The results show that changes in primary production simulated from the P-I curves were strongly associated with the tidal cycle. During the two spring tides that were recorded, mean daily primary production was 1005 and 1125 mg C m−2 d−1, respectively. However, primary production was lower during the neap tide (898 mg C m−2 d−1). Temperature appeared to influence these differences in mean daily production. In situ mean daily temperatures during exposure were 14°C and 20°C during the first and second spring tides, respectively. In contrast, mean daily total irradiance was noticeably higher in the first spring tide compared with the second spring tide. During the neap tide, the site was exposed in the morning and afternoon, when light intensity and temperature were relatively low. In contrast, during the spring tide, the site was exposed at noon, when light intensity and temperature were high. The current study provided new insights about how changes in environmental parameters during tidal cycles affect microphytobenthic primary production, indicating the complexity of this process and the requirement for further fine-scale research.

Impact of microphytobenthos on the sediment biogeochemical cycles: A modeling approach

In marine biogeochemical modeling, the sediment is usually represented by diagenetic models, but in shallow ecosystems these models are incomplete, because they do not take into account the benthic primary production. While microphytobenthos (MPB) is known to strongly impact mineralization pathways and nutrient fluxes, MPB is rarely integrated as an explicit variable. To investigate the impact of microphytobenthos on early diagenesis in sediment, we built a fine-scale dynamic model, based on the diagenetic model OMEXDIA and including MPB and associated processes. The model outputs were similar to a data set of MPB-colonized sediment sampled in Florida Bay, suggesting that the model can recreate a realistic situation. The model showed that MPB activities induced a strong diurnal rhythm on concentration profiles, fluxes, and mineralization processes. When MPB was present at the sediment surface, the total mineralization was strongly enhanced thanks to the supply of labile organic matter. In contrast, coupled nitrification–denitrification was inhibited by a factor of 3.8. This inhibition can be explained by the competition for nitrate and ammonium between MPB and bacteria. Nitrogen uptake of MPB represented 96% of the daily supply of dissolved inorganic nitrogen. This was more than 50 times greater than N consumption by denitrification. With MPB, sediment nitrogen flux to the water column was reduced by a factor of 70, suggesting that sediment colonized by MPB represents a minor source of nutrients for phytoplankton and bacterioplankton. Results showed that current diagenetic models are not well-suited for shallow ecosystems with significant MPB primary production.

Toxic effects of isoproturon on periphyton communities – a microcosm study

Coastal and estuarine ecosystems are increasingly exposed to herbicide contamination. To study the potential risks of these pollutants, the establishment of periphyton communities under exposure of a concentration series of isoproturon in a range of 0.0024–0.312 mg L −1 was investigated in a microcosm study. After two weeks of growth, chronic effects on biomass development, measured as Chl a fluorescence, taxonomic composition and photosynthetic capacity of PS II of attached microalgae were analysed using a pulse-amplitude modulated (PAM) fluorescence-based method. Algal classes were differentiated according to their pigment systems using four excitation wavelengths. Biomass remained constant up to pre-exposure concentrations of 0.02 mg L −1 isoproturon but decreased within one order of magnitude at the highest test concentration. Algal classes shifted from diatoms to chlorophytes at pre-exposure concentrations of 0.02–0.39 mg L −1. Community structure of diatoms representing the dominant group of attached algae at lower test concentrations was studied by microscopic analysis revealing that species numbers were highest at lowest test concentrations as compared to controls. At higher test concentrations Navicula halophila dominated with 89% of the diatom biomass but was abnormally shaped. Pollution-induced community tolerance (PICT) of periphyton to isoproturon was determined by short-term inhibition tests of photosynthesis. Pre-exposed communities increased their effect concentrations (EC 50) up to threefold within the 0.039 mg L −1 pre-treatment. Our results suggest that attached microalgal communities have a high potential of tolerance development to isoproturon at lower levels of contamination due to the replacement of sensitive species by more tolerant algae, although these species seemed to be affected too. We conclude that herbicide concentrations in river basins, estuaries or coastal zones may change community structure and primary production of microphytobenthos even when the contamination does not exceed levels of acute toxicity.

Seasonal and interannual patterns of intertidal microphytobenthos in combination with laboratory and areal production estimates

From April 1994 to December 1997, we studied the microphytobenthic assemblages in sur- face (0 to 0.5 cm) and subsurface (0.5 to 2 cm) sediments at spring low tide along a transect of 5 stations in an estuarine sandflat of the Seto Inland Sea, Japan. At the innermost sampling station, microphyto- benthos biomass (chl a) was also investigated in a vertical profile to 10 cm depth from December 1994 to April 1996. The chl a contents at the 2 uppermost layers were well correlated with each other, with a mean decrease of 34% from the surface to subsurface layer. Chl a tended to decrease rapidly through the vertical profile and was reduced to 3.2 ± 1.4% SD in the 9 to 10 cm layer. There was a progressive de- crease in the chl a content every year in fall and the occurrence of major peaks in early spring and/or summer. This was accompanied by a significant increase in microphytobenthos biomass from 1994 to 1995 and from 1995 to both 1996 and 1997. The microphytobenthos biomass in surface sediments (mean of 5 stations) ranged between 27.7 (October 1994) and 120 mg chl a m -2 (July 1997), or between 3.9 (November 1994) and 20.3 µg chl a g -1 dry wt (July 1996). Annual mean (1995 to 1997) biomass was 72.3 ± 27.1 mg chl a m -2 and 11.0 ± 4.3 µg chl a g -1 dry wt. These values rank in the mid-upper range of microphytobenthic biomass for intertidal sediments. In addition to the field investigations, we conducted laboratory experiments on a dominant diatom species, Navicula sp. The photosynthetic rate of Navi- cula sp. was saturated at a light intensity of 165 µE m -2 s -1 at 21°C. No photoinibition was found at higher light intensities up to 400 µE m -2 s -1 . The relationship between temperature and photosynthetic rate was positive and linear within a temperature range between 10 and 35°C at 55 µE m -2 s -1 . Areal 'potential' primary production of microphytobenthos was between 0.32 (December 1994) and 3.0 g C m -2 d -1 (July 1997), with an annual mean of 1.2 g C m -2 d -1 . Uni- (summer) or bi-modal (spring and summer) peaks of microphytobenthos biomass and primary production highlighted a marked interannual variability. Marked seasonal patterns were also recognizable, with primary production of microphytobenthos significantly higher both in spring and summer than in winter and fall.

Seasonal variation in primary production of microphytobenthos at the Isshiki intertidal flat in Mikawa Bay

Seasonal variations in photosynthetic rates by microphytobenthos and phytoplankton at the Isshiki tidal flat in Mikawa Bay were measured with a 14C combustion method. In addition, diurnal variations in the photosynthetic rate and photosynthesis versus irradiance (P-I) curves were obtained through in situ incubation. The photosynthetic rate of microphytobenthos (annual average, 13.9 ± 6.4 mg C m−2 h−1) did not show a remarkable change, and they maintained a higher production rate than phytoplankton (annual average 9.0 ± 5.1 mg C m−2 h−1) throughout the year. The P-I curves from in situ experiments showed that the photosynthetic activity of microphytobenthos at the laboratory irradiance (250 μE m−2 s−1) was 56% of that at the maximum irradiance (1200 μE m−2 s−1) in situ. In the in situ experiments, the chlorophyll a concentration, photosynthetic rate, and activity of microphytobenthos varied greatly throughout the day, influenced by tidal submersion/emersion and daylight. From an analysis of these results, it is considered that microphytobenthos contributed greatly to primary production in this ecosystem throughout the year by adapting suitably to intertidal environments.

Assessment of the Colonization and Primary Production of Microphytobenthos in the Littoral of Acidic Mining Lakes in Lusatia (Germany)

In 3 extremely acidic mining lakes (pH 2.6-3.1) in Lusatia (Brandenburg, Germany) microphytobenthos colonization was characterized by a very low species diversity, and partly high abundances of the dominant species. Eunotia exigua (Bacillariophyceae) was frequent in all lakes. The abundance of Nitzschia paleaeformis (Bacillariophyceae) clearly increased with increasing pH. Euglena mutabilis (Euglenophyceae) occured mainly in the littoral of Lake 108 with a strongly varying spatial and temporal distribution. Mean concentrations of microphytobenthic biomass, measured as chlorophyll a (Chl), showed marked variations between the lakes and ranged between 1.3 - 17.9 µg Chl g dw-1 or 5.1 - 120 mg Chl m-2. Primary production of microphytobenthos was measured using polyethylene foils as artificial substrates or by direct measurements of sediment surface samples. Primary production rates and specific production rates ranged between 0.99 - 5.1 mg C m-2h-1 and 0.04 - 0.33 mg C (mg Chl)-1h-1, respectively. The impact of the sediment chemistry (especially the available carbon) is discussed as determining factor for the species composition and activity of the microphytobenthic species. First insights can be given concerning the relevance of benthic algae for the carbon metabolism of the investigated acidic mining lakes.

Physicochemical Features and Primary Production of Microphytobenthos and Phytoplankton at Wakaura Tidal Flat in Japan.

The photosynthetic rates of phytoplankton and microphytobenthos were measured with a combustion method of 14C in a tidal flat of the Wakaura estuary, Central Japan. In addition, chlorophyll a concentra-tion and a set of environmental factors (water temperature, salinity, nutrient concentrations) in the water and the sediment were measured in order to investigate factors affecting their photosynthetic rates. The chlorophyll a concentration of microphytobenthos (average 20 mg chl a m-2) was several times as high as that of phytoplankton (average 3.0 mg chl a m-2). The photosynthetic rate of microphytobenthos, which showed a significant positive correlation only with chlorophyll a concentration in the sediment, ranged from 1.1 to 19 mg C m-2 hr-1 (average 8.7 mg C m-2 hr-1), which was equal to or greater than the photosynthetic rate of phytoplankton at that same time. The photosynthetic activity of microphytobenthos (average 1.5 mg C mg chl a-1 hr-1) was lower than that of phytoplankton (average 2.3 mg C mg chl a-1 hr-1). During the observation period, the chlorophyll a concentration, photosynthetic rate and photosynthetic activity of phytoplankton dropped with decreasing water temperature, whereas those of microphytobenthos were almost unchanged. From these results, it was considered that microphytobenthos, particularly in winter, greatly contributed to primary production at the Wakaura tidal flat due to the large amount of biomass, though the growth rate of microphytobenthos was small.

Interrelationships Between Microbiological and Chemical Parameters of Sandy Beach Sediments, a Summer Aspect

At 12 stations located in sandy beach sediments of the brackish water Kiel Fjord and Kiel Bight (Baltic Sea, FRG), variations and interrelationships of microbiological, chemical, and physicochemical parameters were monitored. Depending upon location, wide variations of a number of parameters reflecting dissolved organic and inorganic nutrients, chlorophyll a, microbial number, and uptake activity of glucose were measured. Whereas most of the parameters generally showed the tendency to decrease from the inner to the outer Kiel Fjord, individual parameters (oxygen, particulate nitrogen, ribose, chlorophyll a, glucose/fructose ratio) increased with increasing distance from the inner Kiel Fjord. Similarities in the local variation pattern demonstrated various relationships between individual parameters. Among those, dissolved organic nutrients on the one hand and inorganic nutrients on the other hand were closely linked together. Variations of organic and inorganic nutrients corresponded to variations of microbial activity and physicochemical parameters. By comparing standing stock carbon with carbon production, a microbial biomass turnover time of about 100 h was calculated. Approximately 50% of the microphytobenthos primary production was fixed by microorganisms. Daily microbial carbon production (43 mg of C per m) was in the range of meiofauna carbon (35 mg of C per m).
