Astaxanthin-mediated Nrf2 activation ameliorates glucocorticoid-induced oxidative stress and mitochondrial dysfunction and impaired bone formation of glucocorticoid-induced osteonecrosis of the femoral head in rats

BackgroundOsteonecrosis of the femoral head caused by glucocorticoids (GIONFH) is a significant issue resulting from prolonged or excessive clinical glucocorticoid use. Astaxanthin, an orange-red carotenoid present in marine organisms, has been the focus of this study to explore its impact and mechanism on osteoblast apoptosis induced by dexamethasone (Dex) and GIONFH.MethodsIn this experiment, bioinformatic prediction, molecular docking and dynamics simulation, cytotoxicity assay, osteogenic differentiation, qRT-PCR analysis, terminal uridine nickend labeling (TUNEL) assay, determination of intracellular ROS, mitochondrial function assay, immunofluorescence, GIONFH rat model construction, micro-computed tomography (micro-CT) scans were performed.ResultsOur research demonstrated that a low dose of astaxanthin was non-toxic to healthy osteoblasts and restored the osteogenic function of Dex-treated osteoblasts by reducing oxidative stress, mitochondrial dysfunction, and apoptosis. Furthermore, astaxanthin rescued the dysfunction in poor bone quality, bone metabolism and angiogenesis of GIONFH rats. The mechanism behind this involves astaxanthin counteracting Dex-induced osteogenic damage by activating the Nrf2 pathway.ConclusionAstaxanthin shields osteoblasts from glucocorticoid-induced oxidative stress and mitochondrial dysfunction via Nrf2 pathway activation, making it a potential therapeutic agent for GIONFH treatment.

Modelling of microscopic crack initiation behaviour of fretting fatigue based on X-ray micro-computed tomography scan

Fretting fatigue is a particularly destructive form of fatigue failure that has drawn significant attention from researchers. This paper proposes a numerical modelling method based on tomography scan to investigate the impact of inclusions on fretting fatigue crack initiation behaviour. The results reveal that inclusions can affect the crack initiation direction and reduce the initiation lifetime. The difference between the properties of inclusions and matrix leads to local mechanical response mismatch, resulting in local stress, strain, and damage concentration. The mismatch in strain and damage follows a gradient distribution along the thickness as the distribution in these mechanical responses. These could explain why particles closer to the surface are more likely to cause damage. Additionally, the aggregation distribution of inclusions results in merging and connecting local regions with high stress, strain, and damage concentration, ultimately promoting fracture behaviour. Finally, the numerical models are validated with experimental data from fretting fatigue tests. Compared to the homogeneous model, the numerical model that considers inclusions from topography data can more accurately analyse the material damage and provide a more comprehensive understanding of fretting fatigue behaviour.

Nondestructive internal disorders detection of ‘Braeburn’ apple fruit by X-ray dark-field imaging and machine learning

'Braeburn' apples are susceptible to internal browning disorders when stored under controlled atmosphere (CA) conditions with unfavorable gas compositions. The progression of CA-related disorders in apple tissues is dynamic, noting a decrease in porosity during early storage due to cellular breakdown and pore flooding, and an increase in porosity in later stages due to structural collapse and cavity formation. Utilizing grating-based X-ray dark-field radiography, which leverages X-ray small-angle scattering to detect microstructural changes below the pixel scale, this study assesses the technique's efficacy in identifying internal disorders in 'Braeburn' apples at both early and later stages. A machine learning approach was applied to compare the diagnostic capabilities of dark-field imaging with those of X-ray absorption radiography at identical image resolutions. Results indicate that for early-stage disordered fruit detection, X-ray dark field radiography is 10 % more accurate than absorption radiography, regardless of the machine learning classifiers that were applied. In the later stage of browning, dark-field imaging performs similarly to absorption imaging. High-resolution micro-computed tomography scans suggested that the distinct detection performance of dark-field imaging may be attributed to the more pronounced microstructural differences between healthy and early-stage defective tissues than those between healthy and later-stage defective tissues. The insights from this work will guide the application of X-ray dark-field systems in fruit quality assurance, particularly in detecting internal disorders.

Multiscale porosity characterization in additively manufactured polymer nanocomposites using micro-computed tomography

Extrusion-based additive manufacturing (AM) of polymer composites exhibits complex thermally driven phenomena that introduce severe discontinuities in the internal structure across length scales, especially voids or porosity. This study utilizes a high-throughput porosity characterization technique to analyze large datasets from numerous micro-computed tomography (μCT) scans to capture the influence of AM print parameters on the size, shape, and location of porosity across multiple print layers (up to a few cm) on fused granular fabrication (FGF) printers. The materials investigated include nanocomposite formulations based on commercially relevant nylon-12 and polyether ketone ketone materials (PEKK) comprising nano- or micro-sized fillers. The estimated global porosity follows an inverse linear correlation against the bulk density of the printed samples. Increasing the extrusion multiplier (EM) and the nozzle temperature while decreasing the print speeds reduces the global porosity. Outlier analyses (local porosity morphology) show that faster print speeds and higher extrusion rates result in long, slender inter-layer voids, while lower nozzle temperatures lead to large, symmetrical, inter-bead voids (at the bead junction). Lack of active chamber temperature increases inter-layer and intra-bead voids with a two-fold increase in global porosity. Overall, the micro filler-reinforced composites exhibit higher global porosity than nanofiller-reinforced composites, which is attributed to the increased mismatch in the thermal expansion coefficient between the filler and the polymers used in the study.

A Hybrid Scaffold Induces Chondrogenic Differentiation and Enhances In Vivo Cartilage Regeneration.

Extensively researched tissue engineering strategies involve incorporating cells into suitable biomaterials, offering promising alternatives to boost tissue repair. In this study, a hybrid scaffold, Gel-DCM, which integrates a photoreactive gelatin-hyaluronic acid hydrogel (Gel) with an oriented porous decellularized cartilage matrix (DCM), was designed to facilitate chondrogenic differentiation and cartilage repair. The Gel-DCM exhibited excellent biocompatibility in vitro, promoting favorable survival and growth of human adipose-derived stem cells (hADSCs) and articular chondrocytes (hACs). Gene expression analysis indicated that the hACs expanded within the Gel-DCM exhibited enhanced chondrogenic phenotype. In addition, Gel-DCM promoted chondrogenesis of hADSCs without the supplementation of exogenous growth factors. Following this, invivo experiments were conducted where empty Gel-DCM or Gel-DCM loaded with hACs/hADSCs were used and implanted to repair osteochondral defects in a rat model. In the control group, no implants were delivered to the injury site. Interestingly, macroscopic, histological, and microcomputed tomography scanning results revealed superior cartilage restoration and subchondral bone reconstruction in the empty Gel-DCM group compared with the control group. Moreover, both hACs-loaded and hADSCs-loaded Gel-DCM implants exhibited superior repair of hyaline cartilage and successful reconstruction of subchondral bone, whereas defects in the control groups were predominantly filled with fibrous tissue. These observations suggest that the Gel-DCM can provide an appropriate three-dimensional chondrogenic microenvironment, and its combination with reparative cell sources, ACs or ADSCs, holds great potential for facilitating cartilage regeneration.

Optical method to preserve residual hearing in patients receiving a cochlear implant

IntroductionWorldwide, thousands of patients with severe to profound hearing loss restore their hearing with cochlear implant (CI) devices. Newer developments in electrode design and manufacturing and a better understanding of cochlear mechanics allow for conserving critical structures, often translating into serviceable residual hearing and improving device performance. Monitoring insertion speed and intraluminal pressure helps mitigate some of these challenges. However, the information becomes available after irreparable damage has occurred.MethodsWe developed and tested a high-resolution optical system to navigate the intricate anatomy of the cochlea during electrode insertion. The miniaturized optical system was integrated in conventional cochlear implants electrode arrays and custom-made cochlear probes. Electrode insertion were conducted in eight cadaveric human temporal bones and video recordings were acquired. Micro-computed tomography (μCT) scans were performed to evaluate the position of the modified electrode arrays.ResultsFull insertions of the modified CI electrode were successfully conducted and verified by μCT scans. Video recordings of the cochlear structures visible in scala tympani were acquired, and no scala migration was detected.DiscussionSurgeons can now follow the CI electrode's path during its insertion and reduce cochlear damage through early interventions and steering of the CI electrode. Our device will be compatible with robotic platforms that are already available to insert these electrodes.

Effect of radiation cross-linked collagen scaffold in alveolar ridge preservation of extraction socket.

This study aimed to evaluate the properties of radiation cross-linked collagen scaffold (RCS) and its efficacy for alveolar ridge preservation (ARP). RCS was prepared from collagen dispersion by electron beam irradiation and freeze-drying. The microstructure, swelling ratio, area alteration and mechanical properties of RCS were characterized. Fifty-four New Zealand rabbits performing incisor extraction on maxilla and mandible were randomly assigned into positive, sham operation or treatment groups. Micro-computed tomography (micro-CT) scans, performed after 1, 4, and 12 weeks of surgery, were to assess changes in ridge height at buccal and palatal side, in ridge width and in micromorphological parameters. Histological analysis accessed socket microarchitecture. The results showed that RCS had stable mechanical properties and morphologic features that provided a reliable physical support for ARP. Dimensional changes in treatment group revealed significantly greater vertical height at buccal (5.32 [3.37, 7.26] mm, p < .0001) and palatal (4.37 [2.66, 6.09] mm, p < .0001) side, and horizontal width at the maxilla (0.16 [0.04, 0.28] mm, p < .01) and mandible (0.33 [0.11, 0.54] mm, p < .01) than those in sham operation group after 12 weeks. The treatment group had advantage than positive group in vertical height preservation, quantitatively. The order and density of bone trabeculae were improved in treatment group. These findings indicated that RCS had the potential to serve as an effective scaffold for ARP.

Scaling of buccal mass growth and muscle activation determine the duration of feeding behaviours in the marine mollusc Aplysia californica.

The mechanical forces experienced during movement and the time constants of muscle activation are important determinants of the durations of behaviours, which may both be affected by size-dependent scaling. The mechanics of slow movements in small animals are dominated by elastic forces and are thus quasistatic (i.e. always near mechanical equilibrium). Muscular forces producing movement and elastic forces resisting movement should scale identically (proportional to mass2/3), leaving the scaling of the time constant of muscle activation to play a critical role in determining behavioural duration. We tested this hypothesis by measuring the duration of feeding behaviours in the marine mollusc Aplysia californica whose body sizes spanned three orders of magnitude. The duration of muscle activation was determined by measuring the time it took for muscles to produce maximum force as A. californica attempted to feed on tethered inedible seaweed, which provided an in vivo approximation of an isometric contraction. The timing of muscle activation scaled with mass0.3. The total duration of biting behaviours scaled identically, with mass0.3, indicating a lack of additional mechanical effects. The duration of swallowing behaviour, however, exhibited a shallower scaling of mass0.17. We suggest that this was due to the allometric growth of the anterior retractor muscle during development, as measured by micro-computed tomography (micro-CT) scans of buccal masses. Consequently, larger A. californica did not need to activate their muscles as fully to produce equivalent forces. These results indicate that muscle activation may be an important determinant of the scaling of behavioural durations in quasistatic systems.

Radiological, histological and clinical side effects of incisor trimming in rabbits.

Incisor malocclusion in rabbits (Oryctolagus cuniculus) is a common clinical problem seen in general practice. Given that the growth rate is about 2 mm per week, a lack of wear quickly leads to feeding difficulties and soft tissue injuries. Therefore, pathologically elongated incisors must be shortened every three to six weeks. The goal of this study was to assess the potential adverse effects on dental and periodontal tissues associated with the three most commonly used trimming methods: nail cutter, diamond-coated cutting disc and diamond burr. The left mandibular incisor of 28 healthy New Zealand rabbits was subjected to four trimmings with one of the three cutting methods. After the fattening period, the mandibles were collected and both mandibular incisors were investigated on dental radiographs, micro-computed tomography scans and histological sections. Dental and periodontal tissue changes were evaluated. This study allowed a more accurate statement of the potential short-term adverse effects of the three trimming methods. At the clinical level, the nail cutter caused the formation of an irregular occlusal surface with sharp edges. Both engine-drive methods allowed the attainment of a smooth surface but the disc was less accurate. Histological evaluation revealed that the primary modifications, including coronal fractures, periodontal ligament widening and inflammation, reparative osteodentine, paracementosis and biofilm accumulation, were found in the nail cutter group.

Trueness, physical properties, and surface characteristics of additive-manufactured zirconia crown

ObjectiveThis study aimed to conduct a comparison of trueness and physical and surface properties among five distinct types of additive manufactured (AM) zirconia crowns and zirconia crowns produced using the subtractive manufacturing (SM). Material and methodsZirconia crowns were fabricated using five distinct techniques, each varying in the method of slurry transfer and photocuring source. Each experimental group utilized either one of the four digital light processing (DLP)-based techniques (DLP spreading, DLP spreading gradation, DLP vat and DLP circular spreading) or the stereolithography (SLA)-based technique (SLA spreading). The control (CON) group employed SM. To assess accuracy, trueness was measured between the scan and reference data. To analyze the physical properties, voids were examined using high-energy spiral micro-computed tomography scans, and the crystal structure analysis was performed using X-ray diffraction (XRD). Surface roughness was assessed through laser scanning microscopy. ResultsDifferences in the trueness of internal surfaces of crowns were found among the groups (P < 0.05). Trueness varied across the measurement surfaces (occlusal, lateral, and marginal) in all the groups except for the DLP spreading gradation group (P < 0.05). Voids were observed in all AM groups. All groups showed similar XRD patterns. All AM groups showed significantly greater surface roughness compared to the CON group (P < 0.001). ConclusionThe AM zirconia crowns showed bubbles and a rougher surface compared to the SM crowns. All groups exhibited typical zirconia traits and trueness levels within clinically acceptable limits, suggesting that current zirconia AM techniques could be suitable for dental applications.

Investigation on the contribution of swim bladder to hearing in crucian carp (Carassius carassius).

The swim bladder in some teleost fish functions to transfer the sound energy of acoustic stimuli to the inner ears. This study uses the auditory evoked potential tests, micro-computed tomography scanning, reconstruction, and numerical modeling to assess the contribution of the swim bladder to hearing in crucian carp (Carassius carassius). The auditory evoked potential results show that, at the tested frequency range, the audiogram of fish with an intact swim bladder linearly increases, ranging from 100 to 600 Hz. Over this frequency, the sound pressure thresholds have a local lowest value at 800 Hz. The mean auditory threshold of fish with an intact swim bladder is lower than that of fish with a deflated swim bladder by 0.8-20.7 dB. Furthermore, numerical simulations show that the received pressure of the intact swim bladders occurs at a mean peak frequency of 826 ± 13.6 Hz, and no peak response is found in the deflated swim bladders. The increased sensitivity of reception in sound pressure and acceleration are 34.4 dB re 1 μPa and 40.3 dB re 1 m·s-2 at the natural frequency of swim bladder, respectively. Both electrophysiological measurement and numerical simulation results show that the swim bladder can potentially extend hearing bandwidth and further enhance auditory sensitivity in C. carassius.

Evolution of pore structure and reactive transport parameters during acid leaching of sandstone uranium ore

The changes in the micro-scale pore structure during the acid leaching of sandstone uranium ore significantly affect its reactive transport parameters, which, in turn, directly influence the leaching efficiency of uranium. Consequently, understanding and managing the dynamic evolution of the pore structure of sandstone at micro-scale during acid leaching is beneficial for enhancing the leaching efficiency of uranium. This paper employs a dynamic continuous water-rock reaction experiment on sandstone uranium ore to simulate the in-situ acid leaching process. Additionally, X-ray micro-computed tomography (μCT) scanning was utilized to produce three-dimensional (3D) images at various leaching stages. Subsequently, image processing techniques were employed to characterize and parameterize the pore structure within these images. The results revealed that, during the leaching process, mineral dissolution led to an increase in the interconnected pores, while there was a decrease in the isolated pores. Nonetheless, at the leading edge of the sandstone uranium ore, detachment of mineral grains occurred during the leaching process along the direction of fluid flow. The migration of mineral grains resulted in a reduction of interconnected pores and an increase in the isolated pores. The analysis of 3D images indicated that porosity, permeability, the percentage of connected pore area and the reactive surface area exhibited similar variation trends throughout the leaching process. Furthermore, the permeability, the percentage of connected pore area and the reactive surface area displayed a positive correlation with porosity. The study holds valuable insights to develop a deeper understanding of the evolutionary patterns regarding the pore structure during the in-situ acid leaching of sandstone uranium ore.

Fabrication and application of salicin-polycaprolactone 3D-printed scaffold in the healing of femur bone defects

Polycaprolactone (PCL) is a suitable material for bone repair due to good biocompatibility and mechanical properties. However, low bioactivity and hydrophobicity pose major challenges for its biomedical applications. To overcome these limitations, PCL-based scaffolds loaded with bioactive agents have been developed. Salicin (Sal) is an anti-inflammatory and analgesic herbal glycoside with osteogenic potential. In the present study, we aimed to produce a Sal-laden PCL (PCL–Sal) scaffold for bone healing applications. Three-dimensional scaffolds were produced and their biocompatibility, and physical-chemical characteristics were determined. The osteogenic potential of the PCL (PCL) and PCL–Sal scaffolds was evaluated using bone marrow mesenchymal stem cells (BMSCs). Scaffolds were implanted into a 5 mm bone defect created in the femur of adult rats, and the new bone fraction was determined using micro-computed tomography scanning at one-month follow-up. PCL–Sal scaffold had a structure, porosity, and fiber diameter suitable for bone construction. It also possessed a higher rate of hydrophilicity and bioactivity compared to the PCL, providing a suitable surface for the proliferation and bone differentiation of BMSCs. Furthermore, PCL–Sal scaffolds showed a higher capacity to scavenge free radicals compared to PCL. The improved bone healing potential of the PCL–Sal scaffold was also confirmed according to in vivo implantation results. Our findings revealed that the Sal-laden implant could be considered for bone repair due to desirable characteristics of Sal such as hydrophilicity, surface modification for cell attachment, and antioxidant properties.

Engineering Precise Interconnected Porosity in β-Tricalcium Phosphate (β-TCP) Matrices by Means of Top-Down Digital Light Processing.

This study evaluated the biocompatibility and accuracy of 3D-printed β-tricalcium phosphate (β-TCP) pure ceramic scaffolds. A specific shaping process associating a digital light processing (DLP) 3D printer and a heat treatment was developed to produce pure β-TCP scaffolds leaving no polymer binder residue. The β-TCP was characterised using X-ray diffraction, infrared spectroscopy and the detection of pollutants. The open porosity of produced matrices and their resorption were studied by hydrostatic weighing and calcium release measures. The biocompatibility of the printed matrices was evaluated by mean of osteoblast cultures. Finally, macroporous cubic matrices were produced. They were scanned using a micro-Computed Tomography scanner (micro-CT scan) and compared to their numeric models. The results demonstrated that DLP 3D printing with heat treatment produces pure β-TCP matrices with enhanced biocompatibility. They also demonstrated the printing accuracy of our technique, associating top-down DLP with the sintering of green parts. Thus, this production process is promising and will enable us to explore complex phosphocalcic matrices with a special focus on the development of a functional vascular network.

Abstract 2336: A prototype for a machine-learning assisted image analysis tool for preclinical in vivo studies

Abstract The purpose of this study was to build a software tool to test the feasibility of machine-learning (ML) assisted analysis of in vivo 3D images in oncology research studies. Conventionally, in vivo image segmentations are performed by hand, and are a bottleneck for data analysis, taking time and introducing both inter and intra-user variability. Recently, advances in ML have shown promise in accelerating similar clinical image analysis tasks. Our objective was to work toward making this technology accessible to preclinical cancer researchers. 3D image data across multiple mouse models, organs, modalities, and imaging systems were used. ML segmentation models were trained with user-defined masks on images of mouse bladders (N=134), kidneys (N=131), livers (N=65), and lungs (N=26) from either ultrasound or micro-computed tomography (uCT) scanners. Models were developed using PyTorch/MONAI, hosted in an AWS cloud environment, and served to users via a custom extension for 3D Slicer (www.slicer.org). Performance was assessed as the difference between the human vs. ML organ volumes on a test set unseen by the models during training. Lastly, the functionality of on-the-fly segmentation improvement tools through iterative user feedback was assessed. Results demonstrated that generating segmentations with the prototype ML software took 2-6 seconds per image running on a modern GPU (NVIDIA RTX 3090), compared to 3-7 minutes if performed manually by an expert user (&amp;gt;30x improvement). The Dice scores between the ML generated and human segmentations ranged from 0.81 (liver ultrasound) to 0.84 (bladder ultrasound) but could be improved using ML-based on-the-fly mask improvement tools. Using data from this pilot study, we estimate that the threshold to achieve 90% accuracy on first-pass segmentations is between 200-300 training images, depending on the variance observed in the image data. In summary, the prototype software was able to decrease the manual analysis time across a variety of preclinical in vivo image modalities, which is broadly relevant to preclinical oncology research. The next steps for this project will be to extend the platform to additional orthotopic tumor models (e.g. pancreatic, liver, prostate, etc.) and deploy to the field to enable access to this toolset by preclinical imaging scientists across the world. Experimental Investigation of Injection and Production Cycles for Limestone Reservoirs via Micro-CT: Implications for Underground Gas Storage

Global demand for underground gas storage (UGS) is steadily increasing, with the limestone-based UGS system situated in the Sichuan Basin of China gathering considerable interest in recent years. However, studies focusing on the fundamental mechanisms of the injection-production process in these systems are limited. Moreover, existing studies utilizing physical experimental methods frequently fall short in effectively visualizing micro-flow or incorporating real core samples from the reservoir. To address these gaps, we performed a coreflood experiment, integrating micro-Computed Tomography (CT) scanning to investigate mechanisms of fluid flow and storage capacity during the injection and production cycles in limestone reservoirs. Our approach involved utilizing core plugs with artificially engraved fracture-vuggy structures, which mimic the characteristics of the reservoir. Micro-CT scans were performed to visualize the microscopic changes in fractured-vuggy structures and the distribution of irreducible water during each cycle. This study reveals that increased cycles correspondingly affect gas storage capacity, particularly by expanding it in relative larger vuggy structures while reducing it in finer fissure network structures. The amount of irreducible water decreases after injection-production cycles, likely being expelled alongside the extracted dry gas. This plays a critical role in expanding the storage capacity in larger vuggy systems. Conversely, there is a decrease in storage capacity within fissure network systems, as the irreducible water is replaced by gas. This leads to a reduction in the opening force of the fine conduit. The dense matrix has a very limited effect on the flow mechanism and its influence on storage capacity. Overall, these findings offer practical insights for optimizing injection and production strategies in limestone UGS systems within the Sichuan Basin, contributing to a deeper understanding and efficient utilization of this vital infrastructure.

Spatiotemporal analysis of the effects of exercise on the hemodynamics of the aorta in hypertensive rats using fluid-structure interaction simulation

Abstract Background and Objective Hemodynamic changes that lead to increased blood pressure represent the main drivers of organ damage in hypertension. Prolonged increases to blood pressure can lead to vascular remodeling, which also affects vascular hemodynamics during the pathogenesis of hypertension. Exercise is beneficial for relieving hypertension, however the mechanistic link between exercise training and how it influences hemodynamics in the context of hypertension is not well understood. Methods n exercise model was developed using spontaneously hypertensive rats (SHR) subject to a 12-week treadmill training regime. The heart rates and blood pressures of rats were measured using the tail cuff method, while micro-computed tomography (CT) scanning was used to develop three-dimensional structures of rat aorta, and ultrasound was used to detect rat aortic blood flow and changes to vessel wall structures. Computational fluid dynamics (CFD) and fluid-structure interaction (FSI) models were used to simulate and measure hemodynamic parameters of the rat aortic vessels. In parallel, Masson staining was performed on fixed samples of blood vessels to investigate collagen volume fraction. Hypertensive rats in the sedentary and long-term exercise training groups were subjected to a single bout exercise training, and their aortic hemodynamic parameters were analyzed before, 5 min, 24 h, and 72 h after the single bout exercise. Results Of the two models, in comparison to actual ultrasonic measurement values recorded, we found that numerical simulation results from the FSI model could more accurately model blood flow in the ascending aorta of hypertensive rats, compared to the CFD model. Moreover, longterm exercise training improved local hemodynamic parameters of blood vessels, and led to improvements in adverse hemodynamic features documented, including time-averaged wall shear stress (TAWSS), oscillatory shear index (OSI), and relative residence time (RRT). Longterm exercise training of SHR also improved local vascular collagen deposition in the aorta, while improvements in vascular remodeling were also correlated with favorable hemodynamic parameters. Compared with sedentary SHR, signals for low TAWSS regions of the aortic arch in SHR on the long-term exercise regime shifted to the position of the ascending aorta after a single bout of exercise. Conclusions This study demonstrates that FSI is informative to study the spatiotemporal effects of long-term exercise training on hemodynamic changes within the aortas of hypertensive rats, and that long-term exercise is beneficial through its effects to modulate vascular hemodynamics in hypertension.

Effect of Injectable Acellular Adipose Matrix on Soft Tissue Reconstruction in a Murine Model.

The extracellular matrix isolated from adipose tissue, known as acellular adipose matrix (AAM), represents a novel biomaterial. AAM functions as a scaffold that not only supports stem cell proliferation and differentiation but also induces adipogenesis and angiogenesis. This study aims to investigate the volumetric effects and microenvironmental changes associated with injectable AAM in comparison to conventional fat grafting. AAM was manufactured from fresh human abdominoplasty fat using a mechanically modified method and then transformed into an injectable form. Lipoaspirate was harvested employing the Coleman technique. A weight and volume study was conducted on athymic nude mice by injecting either injectable AAM or lipoaspirate into the scalp (n=6 per group). After eight weeks, graft retention was assessed through weight measurement and volumetric analysis using micro-computed tomography (micro-CT) scanning. Histological analysis was performed using immunofluorescence staining for perilipin and CD31. Injectable AAM exhibited similar weight and volume effects in murine models. Histological analysis revealed comparable inflammatory cell presence with minimal capsule formation when compared to conventional fat grafts. Adipogenesis occurred in both AAM-injected and conventional fat graft models, with no significant difference in the blood vessel area (%) between the two. In summary, injectable AAM demonstrates effectiveness comparable to conventional fat grafting concerning volume effects and tissue regeneration in soft tissue reconstruction. This promising allogeneic injectable holds the potential to serve as a safe and effective "Off-the-Shelf" alternative in both aesthetic and reconstructive clinical practices. Contribution of small airway inflammation to the development of COPD

BackgroundLittle attention has been paid to the pathophysiological changes in the natural history of chronic obstructive pulmonary disease (COPD). The destructions of the small airways were visualized on thoracic micro-computed tomography scan. We investigated whether small airway inflammation (SAI) was the risk for the development of COPD.MethodsA total of 1062 patients were enrolled and analyzed in the study. The partitioned airway inflammation was determined by exhaled nitric oxide (NO) of FnNO, FeNO50, FeNO200, and calculated CaNOdual. Both FeNO200 and CaNOdual were compared to detect the promising predictor for peripheral airway/alveolar inflammation in COPD. The correlation between exhaled NO and white cell classification was evaluated to determine the inflammation type during the development of COPD.ResultsExhaled NO levels (FnNO, FeNO50, FeNO200, and CaNOdual) were the highest in the COPD group compared with all other groups. Furthermore, compared with controls, exhaled NO levels (FeNO50, FeNO200, and CaNOdual) were also significantly higher in the emphysema, chronic bronchitis, and smoking groups. FeNO200 was found to be a promising predictor for peripheral airway/alveolar inflammation (area under the curve [AUC] of the receiver operating characteristic [ROC] curve, area under the curve [AUC] = 0.841) compared with CaNOdual (AUC ROC = 0.707) in COPD. FeNO200 was the main risk factor (adjusted odds ratio, 2.191; 95% CI, 1.797–2.671; p = 0.002) for the development of COPD. The blood eosinophil and basophil levels were correlated with FeNO50 and FeNO200.ConclusionThe complete airway inflammations were shown in COPD, whereas SAI was the main risk factor for the development of COPD, which might relate to eosinophil and basophil levels.

Comparative three-dimensional jaw muscle anatomy of marsupial carnivores (Dasyurus spp.) and the termite-eating numbat (Myrmecobius fasciatus).

Among marsupials, the endangered numbat (Myrmecobius fasciatus) is the only obligate myrmecophage with a diet comprised strictly of termites. Like many other specialised myrmecophagous mammals, numbats have a gracile and highly specialised skull morphology with an elongated rostrum and small braincase. Myrmecobiidae is one of four taxonomic families within the Australasian marsupial order Dasyuromorphia, and to date, the muscular anatomy of any member of this group is relatively poorly known. We utilised microdissection and contrast-enhanced microcomputed tomography scanning to provide the first comprehensive qualitative and quantitative descriptions of jaw muscle anatomy in numbats and quolls (Dasuyrus species). The arrangement of the jaw muscles across these species was conservative, both in gross anatomy and muscle proportions, corresponding to a 'generalised' mammalian pattern. In contrast to Dasyurus, the jaw muscles of the numbat were greatly reduced. Many aspects of the muscle anatomy of the numbat were similar to patterns reported in other myrmecophagous species, particularly a greatly reduced temporalis muscle. Unusually, the digastric muscle in the numbat was comprised of a single, large anterior belly while the posterior belly was absent. We propose that the enlarged anterior belly of the digastric may be linked to jaw stabilisation and coordination of tongue movements during feeding. The lateral insertion and fascial connection of the digastric to the tongue in numbats may also aid in distributing stress evenly across the jaw and minimise muscle fatigue. The muscle descriptions and three-dimensional models provided in this study will facilitate further analysis of musculoskeletal adaptation and evolution within the Dasyuromorphia.
