Colorimetric detection of cysteine based on dispersion–aggregation mechanism of chitosan stabilized gold nanoparticles

A simple and selective colorimetric method for detection of cysteine using chitosan stabilized gold nanoparticles has been developed. L-cysteine is a sulfur containing amino acid. Cysteine binds with the gold nanoparticles through a sulfur group and induces aggregation of the nanoparticles, which leads to a visible colour change from pale red to blue. A good linearity was observed from 0.1 to 30 μmol/L (R = 0.9958) cysteine, and the limit of detection was found to be 0.1 μmol/L. The gold nanoparticles showed a high selectivity towards the detection of cysteine even in the presence of 10 000 fold higher concentration of common interferences such as glutathione, methionine, and homocysteine. A plausible mechanism for the selective detection of cysteine in presence of interferences based on a dispersion–aggregation mechanism has been proposed.

A simple green route to prepare stable silver nanoparticles with pear juice and a new selective colorimetric method for detection of cysteine

In this work, a new cost-effective, rapid and simple method for the preparation of stable silver nanoparticles (AgNPs) was developed, which can be completed within 15 minutes at room temperature by oxidizing the reductants in pear juice with AgNO3. Compared with the most used citrate-capped AgNPs, the as-prepared AgNPs showed high stability, good biocompatibility and enhanced antibacterial activity. Based on the formation of Ag-S covalent bonds between cysteine and AgNPs as well as the electrostatic interaction of COO(-) and NH4(+) between cysteine molecules, which selectively lead to the aggregation of the as-prepared AgNPs and give a specific yellow-to-red colour change, a new selective colorimetric method for detection of cysteine was proposed with the as-prepared AgNPs by coupling the decrease of the characteristic localized surface plasmon resonance (LSPR) absorption at 406 nm of the as-prepared AgNPs and the increase of the new aggregation-induced band at 530 nm. The ratio of the absorbance at 530 nm to 406 nm (A530/A406) was found to be linearly dependent on the cysteine concentrations in the range of 5.0 × 10(-7) to 1.0 × 10(-5) M with a limit of detection of 6.8 × 10(-8) M.

Electrochemical oxidation of dopamine in the presence of sulfhydryl compounds: Application to the square-wave voltammetric detection of penicillamine and cysteine

Electro-oxidation of dopamine at a glassy carbon electrode was investigated in the presence of some biologically important thiols (R-SH), e.g. cysteine and penicillamine. Results of cyclic voltammetric studies together with the spectrophotometric foundations via Ellman's test during the controlled-potential coulometry show a nucleophilic addition/reduction of thiol to the electrochemically generated dopaminoquinone by a 1 + 4 Michael addition. The resulting ring substituted substrate (as RS-form) is more easily oxidized leading to an increase in the anodic current of dopamine, which is proportional to the concentration of thiol. The square-wave voltammetry (SWV) were applied as a very sensitive voltammetric detection method for the detection of cysteine and penicillamine. A linear response from 0.10 to 2.5 μM with a detection limit of 0.08 μM is resulted by the voltammetric determinations for penicillamine. The effect of other biological thiols, such as N-acetyl cysteine, glutathione, methionine, captopril and mercaptoglycine were also assessed and found to present no appreciable change in the voltammetric response of dopamine. The proposed method can be considered as a relatively selective method for the voltammetric detection of penicillamine and cysteine. The practical utility of the method was investigated in the determination of thiol species in pharmaceutical preparations.
