Risk Assessment and Heavy Metals Accumulation in Organs of Clarias Gariepinus and Heterobranchus Longifilis from Omambala River, Anambra State Nigeria

Heavy metals are toxic to organisms even at low concentrations. In recent times, there has been the high occurrence of cancer cases locally, traceable to heavy metal contaminations of foods consumed by man and animals. The experimental site for this study was Omambala River which serves as a source point of many fresh fishes as well as its distribution throughout the state both in dry and rainy seasons. One hundred (100) samples each of Clarias gariepinus and Heterobranchus longifilis, with the size of 17–21 cm were selected from Omambala River. Heavy metal determination was done using the digestion method. All the digested samples were analyzed three times for heavy metals using SP-V21A Vis Spectrophotometer and the instrument was calibrated with standard solutions. For Risk assessment analyses, the target hazard quotients (THQ) index was applied to assess the potential non-cancer risk associated to consumption of the different species of marine organisms sampled. While the total target hazard quotient (TTHQ) was calculated as the arithmetic sum of each THQ value. The lifetime cancer risk (TR) was calculated by multiplying the daily dose by the cancer slope factor (CSF) derived by response-dose curve for toxicant ingestion. The bioaccumulation factor (BAF) was determined by dividing the metal concentration in the tissue of the organism by the metal concentration in the environment or medium. From the correlation analysis, there was a positive correlation between the four different organs of the fish species and the selected heavy metals. The toxicity of magnesium and zinc are higher in C. gariepinus while the toxicity of iron is higher in H. longifilis. The degree of toxicity is zinc > iron > magnesium > lead > cadmium > copper > mercury > chromium. The water quality index (WQI) for all the heavy metals in four different organs of the two fish species was found to be less than 1 (WQI < 1) meaning low pollution; which further means that the consumption of C. gariepinusand H. longifilis may not pose any health risk to the people of the area. However, there are reports of lifetime cancer risk on daily consumption of Chromium, Cadmium, Lead and Mercury in fish. Chemicals when found in large quantities.

Evaluation of proximate compositions and heavy metal concentrations of Amaranthus spinosus L. and Talinum fruticosum (L.) Juss. harvested on some poultry dumpsites in Badagry, Lagos State, Nigeria

Abstract Poultry waste as enhancement for crop development has become a norm for nutrients’ source but their pollution potentials is a source for concern. Thus, this study aimed at assessing proximate compositions and heavy metal accumulation in organs of Amaranthus spinosus and Talinum fruticosum handpicked from three poultry faecal sites in Badagry, Lagos State. Leaves and roots of A. spinosus and T. fruticosum as well as soil samples were collected from Yafin, Agric and Badagry dumpsites in Badagry Local Government and these were subjected to proximate and heavy metal analyses using standard analytical procedures. Data obtained were analyzed using mean-standard deviation. Proximate analyses of A. spinosus and T. fruticosum from three faecal sites contained appreciable Moisture, protein, carbohydrate, fiber, ash, and fat contents. Also, heavy metal analyses of the vegetables from the three faecal sites ranged as follows: Cd: 0.01-0.05mg/100g in leaves; 0.04-0.12mg/100g in roots, Co: 0.04-0.05 in leaves; 0.04-0.12mg/100g in roots, Cr: 0.58-0.84mg/100g in leaves; 0.91-1.01mg/100g in roots, Cu: 0.27-70.51mg/100g in leaves; 0.50-58.55mg/100g in roots for A. spinosus; Cd: 0.04-0.07mg/100g in leaves; 0.06-0.12mg/100g in roots, Co: 0.02-0.06mg/100g in leaves; 0.08-0.34mg/100g in roots, Cr: 0.05-0.12mg/100g in leaves; 0.04-0.23mg/100g in roots, Cu: 61.55-121.81mg/100g in leaves; 48.45-147.45mg/100g in roots for T. fruticosum and Cd: 3.53-9.05mg/100g, Co: 0.14-0.69mg/100g, Cr:0.14-4.22mg/100g: and Cu: 2.59-8.11mg/100g in soils for all three dumpsites sampled. However, all metals analysed were below WHO limits for vegetable and soils except cobalt, copper and chromium. Thus, locals should be advised against consumption of vegetables from dumpsites since toxic substances often accumulates in the environment and maybe hazardous to man’s health.

Fish response of metal bioaccumulation to reduced toxic load on long-term contaminated lake Imandra.

The present study analysed the response of whitefish (Coregonus lavaretus L.) to reduced toxicity after the long-term contamination of subarctic Lake Imandra. High concentrations of Ni, Al, and Sr in fish organs and tissues were accompanied by nephrocalcinosis, scoliosis, and myopathy during the period of intense contamination. After reduction of the toxic impact on the lake, the accumulation of Cu, Al, Sr, Cr, Pb, and Hg in the kidney, which is the target organ for toxicity, was two-fold less and that of Cd was 10-fold less in whitefish from the contaminated part of the lake compared with those in whitefish from the non-contaminated parts of the lake. The ecological success of whitefish from the contaminated part of the lake was associated with the limited accumulation of metals in organs and tissues and a more favourable physiological state compared with whitefish from non-contaminated parts of the lake. Redistribution models were constructed for essential and non-essential metals in the fish liver and kidney depending on the physiological state of fish. The results revealed changes in Fe and Zn metabolism: an increase in Fe and a decrease in Zn accumulation in the liver with increasing stage of liver disease and decreasing blood haemoglobin concentration. Furthermore, under the sub-toxic (except for Cu) conditions of Lake Imandra, the strategy of adaptive fish response is to preserve and maintain acid-base regulation system.

The article presents the review of scientific works on ecology and biology of sterlet aspects of its distribution, spawning, migration, feeding and accumulation of heavy metals in organs and tissues. It has been stated that in natural populations there are two morphs - with a sharp (long) and blunt (short) rostrum. It has been assumed that they are characterized by the separation of spawning period into time intervals. In artificial aquaculture sterlet individuals show a decrease in morphometric traits, which may result from the identical farming conditions or the reduction of genetic diversity. The background color of the tanks used for cultivation can affect the linear-weight characteristics of fish: in light colored tanks there are fish of smaller sizes, in dark colored ones - fish of larger sizes. It has been found that the color of the species body similar to the bottom substrate acts as s a highly effective protection against predators in turbid waters. The study of sterlet population of in the Ob’-Irtysh and Yenisei basins showed that the former population has a higher genetic diversity index. Migration length of sterlet is insignificant: up to 250 km. It spawns in the spring-summer period, in winter period it concentrates in the wintering holes. The leading role in sterlet feeding belongs to the representatives of benthos and zooperiphyton, which are characteristic of lithophilic, lythoreophilic and psammoreophilic biocenoses. Maximum concentrations of heavy metals have been registered in the liver and gills, minimal in the muscles. The largest accumulation of elements is noted for Fe, Al and Zn, the minimum for Cd and Co.

Ecosystem and human health assessment in relation to aquatic environment pollution by heavy metals: case study of the Murmansk region, northwest of the Kola Peninsula, Russia

Throughout the Euro-Arctic region of Russia (Murmansk region), there is a substantial increase of metal concentrations in water, which are related to local discharges from the metallurgical and mining industry, transboundary pollution, as well as indirect leaching of elements by acid precipitation. This study collates data to investigate the relationship between surface water contamination by metals, and fish and human health. Fish are used as a biological indicator to show the impact of water pollution by metals on the ecosystem’s health. The etiology of fish and human diseases are related to the water pollution and accumulation of metals in organisms. High concentrations of Ni and Cd in water drives an accumulation of these elements in organs and tissues of fish, especially in kidneys. The relation between the accumulation of Ni in kidneys and the development of fish nephrocalcinosis and fibroelastosis was established. Statistical analysis demonstrated that human populations in cities close in proximity to smelters show the highest incidence of disease. The results of histological, clinical, and post-mortem examination of patients shows the highest content of toxic metals, especially Cd, in livers and kidneys. Our complex investigation of a set of disorders observed in fish and human populations indicates that there is a high probability that the negative impact on human health is caused by prolonged water contamination by heavy metals. As a novel finding, this paper shows that based on the similarity of pathological processes and bioaccumulation of metals in fish and humans, examining the content of heavy metals in fish can be used to confirm etiology and evaluate the potential risk to human health by pollution of surface waters.

Subcellular partitioning of cadmium and zinc in mealworm beetle (Tenebrio molitor) larvae exposed to metal-contaminated flour

By studying the internal compartmentalization of metals in different subcellular fractions we are able to better understand the mechanisms of metal accumulation in organisms and the transfer of metals through trophic chains. We investigated the internal compartmentalization of cadmium (Cd) and zinc (Zn) in mealworm beetle (Tenebrio molitor) larvae by breeding them in flour contaminated with either Cd at 100, 300 and 600mgkg−1, or Zn at 1000 and 2000mgkg−1. We separated the cellular components of the larvae into 3 fractions: the S1 or cytosolic fraction containing organelles, heat-sensitive and heat-stable proteins, the S2 or cellular debris fraction and the G or metal-rich granule fraction. The concentration of Cd and Zn in each fraction was measured at 0, 7, 14 and 21 days of being fed the flour.The concentration of Cd in the flour affected the concentration of Cd measured in each larval subcellular fraction (p≤0.0001), while the concentration of Zn in the flour only affected the Zn concentration in the S2 and G fractions (p≤0.02). Both Cd and Zn concentrations in mealworms remained relatively constant during the exposure (days 7, 14 and 21) in all three fractions, but the Cd concentrations were much higher than those found in larvae before the exposure (day 0). The concentration of Cd in the flour, however, did not affect the percentage of Cd in the S1 fraction. The contribution of Cd in the G fraction to the total Cd amount was similar (30–40%) in all Cd treatments. The percentage of Zn in all three fractions was not affected by the concentration of Zn in the flour and the relative contributions of each subcellular fraction to the total burden of Zn remained generally constant for both control and treated larvae. In general, larvae sequestered approximately 30% of Cd and Zn in the S1 fraction, which is important for the transport of metals to higher trophic levels in a food web.

Perspectives on the nanotechnology applications of for the analytical detection of heavy metals in marine organisms

Heavy metals accumulate in organisms throughout the food chain and eventually end up in humans. Heavy metals can cause severe diseases and may even result in death. Therefore, concerns about heavy metal accumulation in marine organisms have increased in recent years. To determine solutions to this concern, the sensitive detection of heavy metals in marine organisms is required. Current detection techniques for heavy metals present in marine organisms have several limitations, such as complicated pre-treatment steps and a lengthy analysis time. Thus, there are increasing needs for the newly developed methods of detecting heavy metals in marine organisms. In this review, we focus here on (1) the current detection techniques available and (2) the application of newly emergent nanotechnology for the sensitive detection of heavy metals in marine organisms.

Organ- and species-specific accumulation of metals in two land snail species (Gastropoda, Pulmonata)

In order to evaluate the usefulness of terrestrial gastropods as bioindicators there is a need for studies that simultaneously compare (1) concentrations of metals in reference and polluted plots, (2) species within the same polluted habitat, (3) metal accumulation patterns in different organs and (4) metal accumulation patterns in relation to soil physicochemical properties. This study aims to assess metal accumulation patterns in two land snail species. Instead of analyzing an organism as a whole, investigating the partitioning of metals in different organs can provide information on the actual toxicological relevant fractions. Therefore, concentrations of Ag, Cd, Cr, Cu, Ni and Zn were examined in five different organs of Cepaea nemoralis, as well as in the foot and the body of Succinea putris. Snails were sampled at four polluted dredged sediment disposal localities and three adjacent less polluted reference plots situated along waterways in Flanders, Belgium. Due to the small size and problematic dissection of S. putris only the concentrations in the foot of both species could be compared. For this reason only, C. nemoralis can be described as a better bioindicator species that allows a far more detailed analysis of organ metal accumulation. This study showed that organs other than the digestive gland may be involved in the immobilization and detoxification of metals. Furthermore, pH, soil fractionation (clay %, silt %, sand %) and organic matter, correlate with metal accumulation in organs. However, most often the soil metal concentrations did not correlate with the concentrations found in snail organs. Metal concentrations in organs of both species (1) differed among polluted plots but rarely between polluted and reference plots within a locality, (2) were organ-specific (digestive gland>foot>albumen gland=spermoviduct=ovotestis), (3) were species-specific and (4) depended on the metal type (high Cd and Cu concentrations were observed in the digestive gland and foot respectively). Our study emphasizes that background metal levels should be taken into account when using invertebrates as bioindicators of metal contamination and that bioindicators may show substantial differences in accumulation patterns even if they have a highly comparable ecology.

Seasonal variation of Sarpa salpa fish toxicity, as related to phytoplankton consumption, accumulation of heavy metals, lipids peroxidation level in fish tissues and toxicity upon mice

The aim of this work was to investigate for Sarpa salpa the seasonal trend in the food sources, heavy metals bioaccumulation and the oxidative stress in the organs. In addition, the toxicity was assessed by mouse bioassay of extract of the fish's organs collected in autumn, the peak of occurrence of hallucinatory syndrome. The toxicity was further studied for compounds present in epiphyte collected from the sea at the end of spring and in summer that are digested by the S. salpa in these seasons. We observed a higher lipid peroxydation in different tissues of S. salpa compared to the control fish Diplodus annularis. Furthermore, heavy metals accumulation in organs of these fish showed a significant variation between the two species (P < 0.05). The lethal dose (LD50%) determined for crude ciguatoxin (neurotoxins) extracts of viscera, liver, brain and muscle of S. salpa were as follows: 1.217, 2.195, 14.395, 18.645 g/kg mouse, respectively. We noticed a significant correlation (P < 0.05) between the total amount of toxic dinoflagellates and the level of TBARS in the liver, the brain and the muscle, this for all seasons and all sizes. Moreover, the cytotoxic effect observed for epiphytes extract confirms the transfer of toxins originating from toxic dinoflagellates, which live as epiphytes on P. oceanica leaves, to the fish by grazing. Our work indicates that, toxic phytoplanktons and heavy metals accumulation are responsible for the increase of oxidative stress in the organs of S. salpa. Hence, the edible part of S. salpa, especially the viscera and liver, can cause a threat to human health, and consumption should, for this reason, be dissuaded.

Decreased metal accumulation in passerines as a result of reduced emissions

Technological advances in industrial processes have resulted in reduced atmospheric emissions from metal industries all over the globe, but the response of the environment is not well known. The authors studied metal (As, Cd, Cu, Ni, Pb, and Se) accumulation in passerine birds (pied flycatcher, Ficedula hypoleuca, and great tit, Parus major) following almost 20 years of reduced metal emissions from the largest nonferrous smelter in Finland. Close to the industry, emission reductions resulted in reduced exposure to several of the elements and, more importantly, reduced accumulation by 58 to 95% in liver tissue. Albeit showing significant tissue reductions, nestlings had elevated concentrations of arsenic, cadmium, lead, and selenium close to the industry. Single-element concentrations were below critical levels associated with subclinical effects, but the mixture of toxic elements indirectly affected health and reproduction. Our study indicates that in environments with moderate duration of industrial activity, impact, and soil pollution, metal accumulation in organisms can decrease relatively rapidly when atmospheric emissions are reduced.

Assessment of Trace Metal Levels in Catfish (Cathorops spixii) from Sal River Estuary, Aracaju, State of Sergipe, Northeastern Brazil

The concentrations of cadmium, copper, nickel, zinc, and lead were determined in the catfish (Cathorops spixii) from the Sal River estuary, Brazil, to evaluate the potential role of domestic and industrial effluents released without treatment on the quality of the estuarine environment with consequences to fish resources. Muscle, liver, and kidney samples were analyzed for trace metal composition by flame atomic absorption spectrophotometry. The mean concentration (microg/g wet weight) of trace metals in the tissues of C. spixii were as follows: cadmium = 1.65, 2.81, and 0.71 microg/g; copper = 29.07, 4.38, and 3.40 microg/g; zinc = 229, 312, and 15.57 microg/g; nickel = 1.22, 1.04, and 117 microg/g; and lead = 7.09, 10.77, and 11.18 microg/g. The results show that metal accumulation in organs indicates the difference between them. The results showed high concentrations of copper, zinc, nickel, lead, and cadmium in the Cathorops spixii compared with the maximum values registered from fish in studies reported in other coastal regions of Brazil.

The barnacle Balanus amphitrite as a biomonitor for Cd: Radiolabelled experiments

Radiolabelled experiments were carried out to measure necessary parameters in the development of a biodynamic ecotoxicological simulation model of Cd accumulation in the barnacle biomonitor Balanus amphitrite. The Cd uptake rate constant from the dissolved phase, the Cd assimilation efficiency (AE) from suspended particulate matter (SPM) and the efflux rate constant were obtained using 109Cd. A Cd uptake rate constant from the dissolved phase ( k u) of 0.0072 L g −1 h −1 was determined for the barnacle under environmentally realistic dissolved Cd concentrations (maximum of 400 ng L −1). Cd AE from SPM was determined from the barnacle feeding on SPM with low and high chl a concentrations, resulting in AEs of 39.0% and 48.7%, respectively, and an efflux rate of 0.0072 d −1. The difference between the AEs resulted from differences in chl a:SPM ratios suggesting a general tendency of higher AE when SPM is enriched with chl a. These results reinforce that the accuracy of ecotoxicological models for metal accumulation in organisms depends on how representative the selected food items are of the organism’s natural diet.

Influence of a Mine Tailing Accident Near Do�ana National Park (Spain) on Heavy Metals and Arsenic Accumulation in 14 Species of Waterfowl (1998 to 2000)

This article presents the impact on waterbirds in Doñana National Park (Spain) of an accidental release of 5 million m3 acid waste produced by the processing of pyrite ore. Heavy metals (zinc, copper, cadmium, and lead) and arsenic were measured in several soft tissues (liver, kidney, and muscle) taken from 14 waterfowl species collected between April 1998 and May 2000. The main source of copper and zinc found in the waterfowl species examined was the spill waste, whereas cadmium, lead, and arsenic could also came from other sources. Kidney was the primary organ for cadmium and lead accumulation, whereas liver accumulated the most zinc and copper. Arsenic was concentrated in both muscle and liver tissue. The degree of contamination of the area where the birds lived, their age, their sex their size, and the time since the spill were found to have less influence than species and trophic level on the accumulation of metal in organs and tissues. Four species (Anser anser, Ciconia ciconia, Larus ridibundus, and Porphyrio porphyrio) were found to have the highest levels of the 5 elements.

Metal accumulation in three species of passerine birds (Emberiza cia, Parus major, and Turdus merula) subjected to air pollution from a coal-fired power plant

Metal accumulation in three passerine bird species, Great Tit (Parus major), Rock Bunting (Emberiza cia), and Blackbird (Turdus merula), was analyzed in a polluted zone where there is a coal-fired power plant (Cercs, Northeast Spain), and a non-polluted zone located 40 km from the source of pollution. Rainwater from the polluted area was acidic (pH 5.2) but the calcareous composition of the soil in this zone buffered the soil and water pools; there were no significant differences between soil pH in the two areas. Metal accumulation in organs and tissues was dependent upon the species and the diet. Blackbirds did not show any significant difference between sites, but Rock Buntings from the polluted zone had higher levels of Cr in feathers and Al in bone, and lower levels of Mn in muscle. Great Tits from the polluted zone showed significantly higher levels of Cr in feathers than those from the unpolluted area. Concerning interspecific differences, Blackbird accumulated significantly more metals than Great Tit or Rock Bunting: higher levels of Cd and Cu in the liver, higher levels of Cr, Pb, Zn and S in bone, and higher levels of S in muscle.

Relationship between heavy metal pollution and water productivity in Xiamen estuarine harbor

This paper is based on the results of simulation experiments and annual observations on the effect of Cr, Cu, Hg, and Zn on diatom (Chaetoceros sp.Skeletonema costatum andMelosira sulcata) growth. Laboratory experiments reveal many physico-chemical parameters play an important role in the toxicity and accumulation of metals in organisms. The mesotrophic level in the water environment is related to phytoplankton growth. A higher nutrient level can have a negative impact on diatom production. Several of the above metals coexist in the investigated region and exert mainly a negative effect on growth.

Heavy metals in animals of Lake Balaton

Hg, Cd, Cu, Pb, Zn, Fe and Mn concentrations were measured in Chironomidae larvae, Crustacea plankton, as well as in various organs of mussels and fishes collected from the open water area of Lake Balaton. The values were comparatively low. However, there were significant differences in the metal concentrations of species and tissue. Most Cd was found in the gill of Unio pictorum L., most Pb, Zn and Mn in the gill of Anodonta cygnea L., most Cu in Crustacea plankton and in the liver of Abramis brama L., and most Hg and Fe in Chironomidae larvae. Low concentrations occurred in fishes, the lowest in the flesh of Stizostedion lucioperca L. Accumulation of metals in organs as compared with their concentration in the lake water was in the range of 3.4 × 10 2−2.6 × 10 3 for Hg; 7.0 × 10 2−6.8 × 10 3 for Cd; 3.3 × 10 2−9.7 × 10 3 for Cu; 6.9 × 10 2−8.1 × 10 3 for Zn; 1.5 × 10 3−2.4 × 10 4 for Pb; 3.5 × 10 2−3.1 × 10 4 for Fe; and 5.0 × 10 1−3.5 × 10 5 for Mn.
