Environmental assessment of the central Atlantic coast of Morocco using a multibiomarker approach in Mytilus galloprovincialis.

A multibiomarker approach helps assess environmental health as it provides a complete tool to understand the effects of environmental stressors on ecosystems and human health. We applied this approach in the central Atlantic Ocean of Morocco, an area subjected to the impact of many types of pollutants, threatening the durability of its resources. In this study, four biomarkers acetylcholinesterase (AChE), glutathione-s-transferase (GST), metallothioneins (MTs), and catalase (CAT) were measured in the digestive gland of the mussel Mytilus galloprovincialis collected from four sites: Imsouane (S1), Cap Ghir (S2), Imi Ouaddar (S3), and Douira (S4). These sites were chosen due to the diversity of impacts ranging from industrial to agricultural and touristic. We also assembled all the enzymatic responses (AChE, GST, CAT, and MTs), using the integrated biomarker response (IBR), to estimate the degree of impact of pollutants at the prospected sites to reveal all the complex interactions between biomarkers and to classify sites via the integrated approach. Results show a seasonal change in biomarker responses with variability between sites. We also recorded the highest levels of AChE inhibition and GST induction in S1, higher levels of catalase activity in S4, and a significant impact on metallothionein concentration in S1 and S3. This project highlights the interest in using a multibiomarker approach to ensure accurate interpretation of biomarker variation to protect the Moroccan coast and its resources.

Differential effects of oxytetracycline on detoxification and antioxidant defense in the hepatopancreas and intestine of Chinese mitten crab under cadmium stress

This study aims to evaluate the effects of oxytetracycline (OTC) on detoxification and oxidative defense in the hepatopancreas and intestine of Chinese mitten crab (Eriocheir sinensis) under cadmium (Cd) stress. The crab was exposed to 0.6 μM Cd, 0.6 μM OTC, and 0.6 μM Cd plus 0.6 μM OTC for 42 days. Our results showed that in the intestine, OTC alone enhanced protein carboxylation (PC) and malondialdehyde (MDA) contents, which was associated with the increased OTC accumulation. Compared to Cd alone, Cd plus OTC increased Cd and OTC contents, and reduced detoxification (i.e., glutathione (GSH) content, gene expressions of cytochrome P450 (CYP) isoforms, 7-ethoxyresorufin O-deethylase (EROD) activity, mRNA levels and activities of glutathione peroxidase (GPx), glutathione reductase (GR) and glutathione-S-transferase (GST)), and antioxidant defense (i.e., gene expressions and activities of catalase (CAT) and superoxide dismutase (SOD)) in the intestine, leading to the increased in PC and MDA contents, suggesting that OTC had a synergistic effect on Cd-induced oxidative damage. In the hepatopancreas, although OTC alone increased OTC accumulation, it did not affect PC and MDA contents. Compared to Cd alone, Cd plus OTC reduced MDA content, which was closely related to the improvement of detoxification (i.e., GSH content, mRNA levels of CYP isoforms, EROD activity, gene expressions and activities of GPx, GR and GST), and antioxidant defense (gene expressions and activities of CAT and SOD, metallothionein content). Aryl hydrocarbon receptor (AhR) and nuclear factor E2-related factor 2 (Nrf2) transcriptional expressions were positively correlated with most detoxification- and antioxidant-related gene expressions, respectively, indicating that AhR and Nrf2 were involved in the regulation of these gene expressions. Our results unambiguously demonstrated that OTC had tissue-specific effects on Cd-induced toxicological effect in E. sinensis, which contributed to accurately evaluating Cd toxicity modulated by TCs in crab.

Influence of biochar on the remediation of cadmium-contaminated soil in an Eisenia fetida–Solanum nigrum system

Biochar, plants, and earthworms have good remediation effects on cadmium (Cd)-contaminated soils. However, few studies have combined all three technologies to explore the treatment of Cd-contaminated soils. This study investigated the effect of corn straw biochar addition (1% and 5% mass ratios) on soil Cd treatment in an Eisenia fetida–Solanum nigrum system. The addition of corn straw biochar increased soil pH, total nitrogen (TN), total phosphorus (TP), and soil organic carbon (SOC); adding 5% (w/w) biochar under Cd stress resulted in significant increases (P < 0.05) of soil pH, TN, TP, and SOC. Adding 5% (w/w) biochar under Cd stress increased Cd enrichment by E. fetida and S. nigrum and significantly reduced the soil total and available Cd contents (P < 0.05). The addition of biochar increased the metallothionein content of E. fetida, which functions to resist Cd stress in high-Cd environments (P < 0.05); with the addition of 5% (w/w) biochar, the metallothionein content was 1.55 times higher than in the 1% (w/w) biochar treatment, at 23.78 ng L−1. Adding 5% (w/w) biochar significantly increased the Cd enrichment coefficient and transfer coefficient values of S. nigrum under high-Cd stress (P < 0.05), reaching 7.37 and 1.89, respectively. Adding 5% (w/w) biochar significantly reduced the exchangeable and acid-soluble fraction of Cd, increased the oxidizable fraction, reduced Cd bioavailability, and mitigated physiological damage (P < 0.05). The present study demonstrated that adding biochar to the E. fetida–S. nigrum system could effectively reduce the soil Cd pollution level, providing a new method and scientific guidance for the remediation of heavy metal-polluted soil.

Inoculation of heavy metal resistant bacteria alleviated heavy metal-induced oxidative stress biomarkers in spinach (Spinacia oleracea L.)

Most vegetable crops are severely affected by the uptake of heavy metals from the soil. Heavy metals in vegetable bodies generate reactive oxygen species (ROS) that unbalance the antioxidant defense system. This study was initiated to determine the physiological and biochemical characteristics of spinach plants grown on soil contaminated with heavy metals and responding to Bacillus cereus and Bacillus aerius were isolated from soil contaminated with heavy metals. Heavy metal contamination led to a significant reduction in seed germination, seedling biomass, protein, and total nitrogen content of spinach plants grown in contaminated soils compared to control soils. In contrast, a significant increase in the content of metallothioneins and antioxidant enzymes was observed. Plants inoculated with B. cereus and B. aerius significantly reduced the oxidative stress induced by heavy metals by improving seed germination (%), seedling growth, nitrogen, and protein content. The content of metallothioneins and the activities of antioxidant enzymes were reduced in spinach plants grown from seeds inoculated with bacterial strains. In addition, plants inoculated with, B. cereus and B. aerius showed greater stomata opening than plants grown on soil contaminated with heavy metals, whose stomata were almost closed. These results suggested that both bacterial strains enhanced plant growth by reducing oxidative stress caused by metals.

Vitamin E: An assistant for black soldier fly to reduce cadmium accumulation and toxicity

Cadmium (Cd) is a toxic heavy metal associated with osteoporosis, liver, and kidney disease. The black soldier fly (BSF) Hermetia illucens may be exposed to Cd during the transformation of livestock manure. The BSF has a high tolerance to Cd. In the previous work of the laboratory, we found that vitamin E (VE) may play a role in the tolerance of BSF to Cd exposure. The main findings are as follows: The BSF larvae pretreated with exogenous VE had heavier body weight, lower content and toxicity of Cd under similar Cd exposure. Even in high Cd exposure at the concentrations of 300 and 700 mg/kg, the BSF larvae pretreated with exogenous VE at a concentration of 100 mg/kg still reduced the Cd toxicity to 85.33 % and 84.43 %, respectively. The best-fitting models showed that metallothionein (MT) content, oxidative damage (8-hydroxydeoxyguanosine content, malondialdehyde content), antioxidant power (total antioxidant power, peroxidase activity) had a great influence on content and toxicity of Cd bioaccumulated in the larvae. The degree of oxidative damage was reduced in the larvae with exogenous VE pretreatments. This variation can be explained by their changed MT content and increased antioxidant power because of exogenous VE. These results reveal the roles of VE in insects defense against Cd exposure and provide a new option for the prevention and therapy of damage caused by Cd exposure.

Antioxidant system alterations, oxidative, and genotoxic effects in Danio rerio (zebrafish) exposed to leachate from a dumpsite.

Water body contamination by leachate originated from dumpsites is a concern for municipal solid waste (MSW) management. In this context, this study aimed to evaluate antioxidant system alterations and oxidative and genotoxic effects in Danio rerio (zebrafish) exposed to leachate from a closed dumpsite. Groups comprising 50 fish were exposed (96 h) to different leachate concentrations (5, 15, 30, and 50%) to evaluate effects on liver and brain superoxide dismutase (SOD), catalase (CAT), and glutathione-S-transferase (GST) activities and reduced glutathione (GSH) and metallothionein (MT) concentrations, as well as malondialdehyde (MDA) and protein carbonylation (PTC) levels. Blood genotoxicity was evaluated by the comet assay. The investigated dumpsite leachate pond presented high chloride concentrations (Cl-; 2288.4 ± 69.5 mg L-1) and high electrical conductivity (EC; 8434.0 mS cm-1), indicating the presence of leachate. Concerning Danio rerio exposure, higher SOD (37%), CAT (67%), and GST (39%) activities and higher GSH (57%) concentrations were observed in liver following exposure to 50% leachate, while decreased brain GST (42%) activities and GSH (90%) levels were observed at the same leachate concentration. A significant increase in the olive tail moment (OTM; 280%) indicative of genotoxicity in blood was observed. A principal component analysis indicated that increased enzymatic activities and high levels of both GSH and MT were not sufficient to prevent the accumulation of reactive oxygen species, resulting in PTC and genotoxicity. Therefore, leachate exposure causes sublethal Danio rerio effects, altering the antioxidant system, increasing ROS production, and leading to PTC and genotoxicity. The findings demonstrate the need to further develop sublethal level assessments in zebrafish using leachate from different sources to subsidize risk assessments regarding MSW management.

Characteristics and Mechanisms of Soil Co-Contamination Affecting the Transfer of Cadmium and Arsenic in Peanut (Arachis hypogaea L.)

Soil co-contamination with cadmium (Cd) and arsenic (As) occurs frequently and has caused increasing concern. This study aimed to explore the transfer characteristics and the chemical forms, subcellular distribution of Cd and As, as well as the synthesis of phytochelatins (PCs) and other chelates in peanut (Arachis hypogaea L.) plants grown in a Cd and As co-contaminated soil, shedding light on the mechanisms involved. Compared with the single Cd contamination, Cd–As co-contamination led to a higher accumulation of Cd in peanut plants. Conversely, compared to the single As contamination, the As content increased in peanut shoots but decreased in roots and grains under Cd–As co-contamination. Furthermore, the Cd–As interaction resulted in notable changes in peanut plants’ physiological and biochemical responses. In the roots and shoots, there was an 81.8% and 60.0% increase in water-soluble Cd. In the roots, metallothioneins (MTs) content increased by 50%, while PCs increased by 6.4% in the shoots. These changes promoted the translocation of Cd from roots to grains. The Cd–As interaction also influenced the synthesis of MTs in the roots, showing a 41.2% increase, and facilitated the transfer of As to the shoots. In peanut shoots, Cd increased the cell wall fraction of As by 34.5%, decreased the proportion of water-soluble As by 31.8%, and increased PCs content by 6.9%. These changes inhibited the migration of As from shoots to grains. Overall, Cd–As co-contamination increased Cd in peanut grains by increasing water-soluble forms and MTs in roots, while Cd–As co-contamination decreased As in peanut grains by increasing cell wall fractions and PCs in shoots. These findings provide a theoretical basis for understanding Cd–As interactions in soil–peanut systems.

Changes in the level of metallothioneins, zinc and copper in blood plasma of patients with chronic lymphocytic leukemia

The relationship between the content of metallothioneins, copper and zinc ions in peripheral blood plasma in chronic lymphocytic leukemia (CLL) depending on the disease progression was studied. It was shown that in patients with CLL in blood plasma with a progressive disease, partial and complete remission, the values of the zinc-to-copper ratios significantly decreased in comparison to similar indicators of practically healthy donors. It was established that in patients with CLL, the level of metallothioneins in blood plasma decreases with the disease progression compared to other studied groups, which indicates a reduced function of cysteine-containing metallothionein proteins. Based on the obtained data, it can be concluded that the zinc-to-copper concentration ratio and the level of metallothionenins in peripheral blood plasma are potential diagnostic and prognostic markers in chronic leukemia.

Do Nanoparticles of Calcium Disodium EDTA Minimize the Toxic Effects of Cadmium in Female Rats?

The present study aims to investigate the ability of CaNa2EDTA (ethylenediaminetetraacetic acid) macroparticles and nanoparticles to treat cadmium-induced toxicity in female rats and to compare their efficacies. Forty rats were divided into 4 equal groups: control, cadmium, cadmium + CaNa2EDTA macroparticles and Cd + CaNa2EDTA nanoparticles. Cadmium was added to the drinking water in a concentration of 30 ppm for 10 weeks. CaNa2EDTA macroparticles and nanoparticles (50 mg/kg) were intraperitoneally injected during the last 4 weeks of the exposure period. Every two weeks, blood and urine samples were collected for determination of urea, creatinine, metallothionein and cadmium concentrations. At the end of the experiment, the skeleton of rats was examined by X-ray and tissue samples from the kidney and femur bone were collected and subjected to histopathological examination. Exposure to cadmium increased the concentrations of urea and creatinine in the serum and the concentrations of metallothionein and cadmium in serum and urine of rats. A decrease in bone mineralization by X-ray examination in addition to various histopathological alterations in the kidney and femur bone of Cd-intoxicated rats were also observed. Treatment with both CaNa2EDTA macroparticles and nanoparticles ameliorated the toxic effects induced by cadmium on the kidney and bone. However, CaNa2EDTA nanoparticles showed a superior efficacy compared to the macroparticles and therefore can be used as an effective chelating antidote for treatment of cadmium toxicity.

Chromium(VI) Toxicity and Active Tolerance Mechanisms of Wheat Plant Treated with Plant Growth-Promoting Actinobacteria and Olive Solid Waste.

The present study aimed to assess the potential of plant growth-promoting Actinobacteria and olive solid waste (OSW) in ameliorating some biochemical and molecular parameters of wheat (Triticum aestivum) plants under the toxicity of high chromium levels in the soil. With this aim, a pot experiment was conducted, where the wheat plants were treated with a consortium of four Actinobacterium sp. (Bf treatment) and/or OSW (4% w/w) under two levels of nonstress and chromium stress [400 mg Cr(VI) per kg of soil] to estimate the photosynthetic traits, antioxidant protection machine, and detoxification activity. Both Bf and OSW treatments improved the levels of chlorophyll a (+47-98%), carotenoid (+324-566%), stomatal conductance (+17-18%), chlorophyll fluorescence (+12-28%), and photorespiratory metabolism (including +44-72% in glycolate oxidase activity, +6-72% in hydroxypyruvate reductase activity, and +5-44% in a glycine to serine ratio) in leaves of stressed plants as compared to those in the stressed control, which resulted in higher photosynthesis capacity (+18-40%) in chromium-stressed plants. These results were associated with an enhancement in the content of antioxidant metabolites (+10-117%), of direct reactive oxygen species-detoxifying enzymes (+49-94%), and of enzymatic (+40-261%) and nonenzymatic (+17-175%) components of the ascorbate-glutathione cycle in Bf- and OSW-treated plants under stress. Moreover, increments in the content of phytochelatins (+38-74%) and metallothioneins (+29-41%), as markers of detoxification activity, were recorded in the plants treated with Bf and OSW under chromium toxicity. In conclusion, this study revealed that the application of beneficial Actinobacteria and OSW as biofertilization/supplementation could represent a worthwhile consequence in improving dry matter production and enhancing plant tolerance and adaptability to chromium toxicity.

Metal and metallothionein concentrations in mesozooplankton from an oligotrophic offshore area in the eastern Mediterranean Sea (Cretan Passage/Levantine Sea)

Spatial variability of Cd, Cu, Cr, Ni, Zn, Fe, Mn, Pb and metallothionein (MT) concentrations were determined in mesozooplankton samples along the west-east axis of the Cretan Passage in the western Levantine Sea (Eastern Mediterranean). Metal and MT values from the present study are proposed as background levels, due to the lack of substantial anthropogenic activities in the area, where only maritime traffic and atmospheric deposition could be potential sources. Higher concentrations, of both metals and MTs, were recorded mainly at the western stations indicating higher metal bioavailability than in the eastern part. An inverse relationship of the metal zooplankton levels with zooplankton biomass, abundance and vital rates (production, respiration and ingestion rates), as well as salinity, was evident. We discuss the hypothesis that physical and biological characteristics of the marine environment, affecting growth dynamics of phytoplankton and zooplankton communities, may also act on metal uptake in oligotrophic marine systems.

The Effects of Zinc Supplementation on Growth, Diarrhea, Antioxidant Capacity, and Immune Function in Holstein Dairy Calves.

The current study examined the effects of supplementary zinc proteinate (ZnPro) and zinc oxide (ZnO) on growth performance, diarrhea, antioxidant capacity, immune function, and mineral element concentrations of calves aged 1 to 28 days. A total of twenty-four newborn calves were divided randomly into 3 groups (n = 8; 2 males and 6 females per group), and each received: 0 mg/d Zn (CON), 627 mg/d ZnPro (80 mg/d Zn; ZnPro group), and 101 mg/d ZnO (80 mg/d Zn; ZnO group). The calves received the additive in their milk during the first 28 days of life. Compared with the CON group: ZnPro and ZnO improved average daily gain (ADG) and decreased the feed:gain ratio (FGR) between days 1 and 14 (p < 0.05), while the ADG increased and FGR decreased only in the ZnPro group between days 1 and 28 (p < 0.05). The incidence of diarrhea decreased (p < 0.05) in the ZnPro and ZnO groups between days 15 and 28 as well as days 1 and 28, but decreased (p < 0.05) only in the ZnPro group between days 1 and 14. The serum immunoglobulin G (IgG) concentration of the ZnPro and ZnO groups increased on days 14 and 28 (p < 0.05). ZnPro supplementation increased serum IgM concentration during the whole study, while ZnO enhanced serum IgM concentration only on day 14 (p < 0.05). In the ZnO group, the serum concentration of cytokines interleukin (IL)-10 increased on day 14, while that of IL-1β increased on day 28 (p < 0.05). In addition, ZnPro reduced the serum malondialdehyde (MDA) concentration on days 14 and 28 (p < 0.05). Both ZnPro and ZnO increased the serum concentrations of alkaline phosphatase (ALP) and metallothionein (MT) on day 14 (p < 0.05). With zinc supplementation, plasma Zn concentration increased (p < 0.05) on days 14 and 28 of age. We concluded that supplementary ZnPro and ZnO reduced incidences of diarrhea and promoted the immune function, but ZnPro improved the growth performance and antioxidant capacity of Holstein dairy calves to a greater extent.

Combined toxic effects of cadmium and environmental microplastics in Aphanius fasciatus (Pisces, Cyprinodontidae)

Microplastics (MPs), plastic particles smaller than 5 mm in diameter, have received extensive attention as new environmental pollutants with still unexplored potential ecological risks. The main objective of the present study is to see if the concomitant exposure to MPs and Cd is more toxic than that to MPs or Cd separately in Aphanius fasciatus. Immature female were exposed to Cd and/or MPs for 21 days, and the subsequent effects were monitored by a combination of biochemical, histological and molecular toxicity markers. Exposure to Cd, but not to MPs, increased metallothioneins content and mRNA levels of the metallothioneins gene MTA both in liver and gills. In addition, we observed a significant oxidative stress response at histological, enzymatic (Catalase and Superoxide dismutase), non-enzymatic (proteins sulfhydryl and malondialdehyde) and gene expression levels to both toxicants in both tissues, particularly in gills, but no clear evidence for interaction between the two factors. Our results indicate a major effect of MPs on gills at different organizational levels. Finally, exposure to both MPs and Cd induced spinal deformities, although bone composition was only altered by the latter, whereas MTA mRNA bone levels were only increased realtive to controls in doubly-exposed samples. Interestingly, the simultaneous use of both pollutants produced the same effects as Cd and MPs alone, probably due to reduced bioavailability of this heavy metal.

Biochemical responses of the Dreissena polymorpha from municipal pond to caffeine, microplastics, and heating in single and combined exposures

Background. Pharmaceuticals have become the aquatic pollutants of growing concern. Caffeine is one of the most widely distributed in the surface waters among them. However, the environmentally relevant models of its effect, which include combined exposures with probable confounding factors, are unknown. Microplastics are a suspected vector that influences caffeine bioavailability. The temperature dependence of response, considering the increase of temperature in surface waters, can also be anticipated. The aim of this study was to analyze the input of caffeine, microplastics and elevated temperature into their combined effect on the zebra mussel Dreissena polymorpha (Pallas, 1771). Materials and Methods. Molluscs were exposed to caffeine (Caf, 20.0 μg·L−1), microplastics (MP, 1 mg·L−1, 2 μm in size), or elevated temperature (T, 25 °C) in the single and combined (Mix- and MixT-) exposures for 14 days. The concentrations of metallothioneins, metallothioneins-bound zinc, total Zn and Cu concentration in the tissue, total glutathione level, antioxidant (superoxide dismutase) and metabolic (citrate synthase) enzymes activities, acid phosphatase activity as the lysosomal functionality marker were determined. Results and Discussion. The decrease in Zn/Cu concentrations ratio in the soft tissues shared the common response in all exposures, reflecting the metal imbalance as the most sensitive marker. The MP-group was distinguished by the decrease in the levels of total Zn and extra lysosomal acid phosphatase activity, proving injury of Zn transportation and Zn-related enzyme activities. All other exposures (T-, Mix-, MixT-) caused citrate synthase and superoxide dismutase activation. Caf-related groups demonstrated the elevation of the levels of phosphatase lysosomal membrane-linked latency, metallothionein total protein and its apo-form. However, glutathione level was stable in all exposures. Conclusion. These data revealed the adverse effect of MP and shared beneficial effects in the exposures that involved caffeine, which can be explained by the antioxidant activity of this substance. Exposure to elevated temperature partially alleviated the effect of caffeine in the mixture. Thus, the results indicate the importance of multi-stress exposures modeling, which allows the evaluation of environmentally realistic responces of an organism to xenobiotics.

Heavy metal accumulation, biochemical and transcriptomic biomarkers in earthworms Eisenia andrei exposed to industrially contaminated soils from south-eastern Tunisia (Gabes Governorate)

Heavy metal pollution is causing harmful consequences on soil fertility, and earthworms are frequently employed as test organisms to evaluate the ecotoxicity of polluted soils. In this study, Eisenia andrei was exposed for 7 and 14 days to polymetallic contaminated soils collected from an industrial zone in the south-eastern Tunisia. Earthworm growth, heavy metal accumulation, genotoxicity, cytotoxicity, biochemical and transcriptional responses were determined. Results revealed a higher accumulation of heavy metals in earthworms after 14 than 7 days of soil exposure, a reduction in lysosomal membrane stability (LMS), besides an increase in micronuclei frequency (MN). Moreover, earthworm oxidative status was affected in terms of increases in malondialdehyde (MDA) and metallothionein (MTs) content, and enhancement of catalase (CAT) and glutathione-S-transferase (GST) activities. An inhibition of acetylcholinesterase (AChE) activity was also observed in treated earthworms, whereas transcriptional data demonstrated an up-regulation of cat, gst, mt, p21 and topoisomerase genes. Overall, these findings support the use of earthworms as suitable bioindicator species for pollution monitoring and assessment, advance our understanding of the interaction between heavy metals and earthworms, and provide valuable information about the harmful impact of biota exposure to naturally contaminated soils.

Exposure to Nickel Oxide Nanoparticles Induces Alterations in Antioxidant System, Metabolic Enzymes and Nutritional Composition in Muscles of Heteropneustes fossilis.

The current study was performed to explore potential toxic effect of nickel oxide nanoparticles (NiO NPs) on muscle tissue of catfish, Heteropneustes fossilis. Fishes were exposed to different concentrations of NiO NPs (12mg/L, 24mg/L, 36mg/L and 48mg/L) for a period of 14 days. Results revealed that NiO NPs caused significant increase in Ni accumulation, metallothionein content, lipid peroxidation and activity of different antioxidant enzymes (catalase, glutathione s transferase and glutathione reductase) while decrease in activity of superoxide dismutase (p < 0.05). Data also reported induction of Na+/K+ ATPase activity initially and then its decrease in concentration dependent manner. Fourier transform infrared spectroscopy revealed shift and changes in spectra of muscle of NiO NPs treated fishes. Fluctuations in activity of aspartate amino transferase, alanine amino transferase and alkaline phosphatase were also noticed. Nutritional contents like protein, lipid, and moisture significantly reduced while glucose and ash percent increased.

Tumor Necrosis Factor-α Alters Maternal and Embryonic Zinc Metabolism and Is Developmentally Toxic in Mice

We tested the hypothesis that tumor necrosis factor-α (TNF-α) would be teratogenic in mice due in part to its effects on zinc metabolism. In Experiment 1, nonpregnant mice were injected with a single dose of TNF-α (40,000 U) or PBS and then received a 65Zn-labeled meal. Mice killed 10 h after TNF-α treatment had high liver 65Zn and low plasma 65Zn, compared with controls. In Experiment 2, gestation day 8 (GD 8) mice were injected with PBS or TNF-α and then received a 65Zn-labeled meal. Dams killed 10 h after TNF-α treatment had higher liver and kidney 65Zn and lower plasma and embryonic 65Zn accumulation than controls. In Experiment 3, TNF-α dosing from GD 7–12 was associated with high maternal liver Zn and metallothionein concentrations on GD 13 and a high frequency of exencephaly on GD 18. In Experiment 4, dams fed diets containing 4.5, 12.5 or 50.0 µg Zn/g were given PBS or TNF-α on GD 7–12. Gross fetal defects were not observed in the PBS-treated litters evaluated on GD 18. In contrast, TNF-α-treated litters were characterized by multiple defects, with the incidence and severity being highest in the low Zn diet group. In Experiment 5, embryos cultured in serum from TNF-α-treated animals exhibited a high frequency of defects; the developmental toxicity of this serum was ameliorated when it was supplemented with Zn. Thus, the developmental toxicity of TNF-α is due in part to its influence on Zn metabolism.

Supplemental Dietary Cystine Elevates Kidney Metallothionein in Rats by a Mechanism Involving Altered Zinc Metabolism

Dietary sulfur-containing amino acids influence zinc and copper status. This study was conducted to investigate the relationship between the status of these elements and tissue metailothionein in rats fed diets supplemented with sulfur-containing amino acids. In a series of experiments, a diet containing 100 g casein/kg diet was unsupplemented or supplemented with L-cystine (3–50 g/kg diet) or L-methionine (3 or 30 g/kg diet). Kidney concentrations of zinc and copper in rats fed the diet supplemented with high levels of cystine (25 or 50 g/kg) were significantly higher than those in rats fed the unsupplemented diet. Kidney concentrations of metallothionein and metallothionein mRNA were also significantly higher in rats fed cystine-supplemented diets. There was a correlation (r = 0.838, P < 0.01) between the levels of zinc and metallothionein in kidney of rats fed the diets with different levels of cystine and zinc. However, in the rats fed the diets with different levels of cystine and copper, the correlation was not as strong (r = 0.587, P < 0.01). The changes in kidney metallothionein concentration due to the addition of 3 or 30 g/kg cystine were associated with parallel changes in serum zinc concentration and in apparent absorption of zinc, but not in serum copper concentration or apparent absorption of copper. Addition of 3 or 30 g methionine/kg diet to the diet did not affect kidney concentrations of metallothionein or zinc. These results indicate that supplementation of cystine to the diet can induce kidney metallothionein through a mechanism involving altered zinc metabolism.

Chronic Corticosterone Exposure Suppresses Copper Transport through GR-Mediated Intestinal CTR1 Pathway in Mice.

Numerous studies have discovered that chronic stress induces metabolic disorders by affecting iron and zinc metabolism, but the relationship between chronic stress and copper metabolism remains unclear. Here, we explore the influence of chronic corticosterone (CORT) exposure on copper metabolism and its regulatory mechanism in mice. Mice were treated with 100 μg/mL CORT in drinking water for a 4-week trial. We found that CORT treatment resulted in a significant decrease in plasma copper level, plasma ceruloplasmin activity, plasma and liver Cu/Zn-SOD activity, hepatic copper content, and liver metallothionein content in mice. CORT treatment led to the reduction in duodenal expression of copper transporter 1 (CTR1), duodenal cytochrome b (DCYTB), and ATPase copper-transporting alpha (ATP7A) at the mRNA and protein level in mice. CORT treatment activated nuclear glucocorticoid receptor (GR) and down-regulated CRT1 expression in Caco-2 cells, whereas these phenotypes were reversible by an antagonist of GR, RU486. Chromatin immunoprecipitation analysis revealed that GR bound to the Ctr1 promoter in Caco-2 cells. Transient transfection assays in Caco-2 cells demonstrated that the Ctr1 promoter was responsive to the CORT-activated glucocorticoid receptor, whereas mutation/deletion of the glucocorticoid receptor element (GRE) markedly impaired activation of the Ctr1 promoter. In addition, CORT-induced downregulation of Ctr1 promoter activity was markedly attenuated in Caco-2 cells when RU486 was added. These findings present a novel molecular target for CORT that down-regulates intestinal CTR1 expression via GR-mediated trans-repression in mice.

Production of Intestinal Mucins, sIgA, and Metallothionein after Administration of Zinc and Infection of Ascaridia galli in Chickens: Preliminary Data.

The effect of inorganic zinc and Ascaridia galli infection was studied on MUC1, MUC2 (mucin), sIgA (secretory immunoglobulin A), and metallothionein in the intestines of broilers. Thirty-five-day-old chickens (n = 24), COBB 500 breed, were included in a 14-day experiment. Chickens were divided into 4 groups of 6 chickens each: control ©, Ascaridia galli (AG), Zinc group (Zn), and combined group (AG + Zn). Samples from the intestine for determination of MUC1, MUC2, sIgA, and metallothionein were taken at 7 and 14 days during necropsy. Samples from the jejunum for determination of MUC1, MUC2, sIgA, and metallothionein were taken at 7 and 14 days during necropsy. The results demonstrated that 12 days' administration of inorganic zinc increased production of MUC1 (p &lt; 0.0001) and MUC2 (p &lt; 0.001) in the Ascaridia galli-infected group (Ag + Zn) in comparison to control (C). The beneficial effect of zinc was also revealed in the production of sIgA (p &lt; 0.0001) in the combined group (AG + Zn) at 7 days. The concentration of metallothionein increased mainly in the zinc group (p &lt; 0.01) of first sampling and was upregulated in Zn and AG + Zn groups. The obtained data indicate the use of inorganic zinc as a suitable immunomodulator of intestinal immunity in Ascaridia galli-infected chickens.
