Bioaccumulation and metabolism of polybrominated diphenyl ethers (PBDEs) in coenobitid hermit crabs from marine litter-polluted beaches in remote islands

Plastic litter containing additives is potentially a major source of chemical contamination in remote areas. We investigated polybrominated diphenyl ethers (PBDEs) and microplastics in crustaceans and sand from beaches with high and low litter volumes on remote islands that were relatively free of other anthropogenic contaminants. Significant numbers of microplastics in the digestive tracts, and sporadically higher concentrations of rare congeners of PBDEs in the hepatopancreases were observed in coenobitid hermit crabs from the polluted beaches than in those from the control beaches. PBDEs and microplastics were detected in high amounts in one contaminated beach sand sample, but not in other beaches. Using BDE209 exposure experiments, similar debrominated products of BDE209 in field samples were detected in the hermit crabs. The results showed that when hermit crabs ingest microplastics containing BDE209, BDE209 leaches out and migrates to other tissues where it is metabolized.

The toxicity, bioaccumulation and debromination of BDE-47 and BDE-209 in Chlorella sp. under multiple exposure modes

Polybrominated diphenyl ethers (PBDEs) are a series of important persistent organic pollutants (POPs) in marine environments. Microalgae are the start of PBDEs bioaccumulated and bioconcentrated along the marine food web. In order to investigate the variations of PBDEs bioaccumulation by microalgae and its influencing factors, we set up a series of experiments with Chlorella sp. under different BDE-47 or BDE-209 exposure modes to measure their toxicity, bioaccumulation and degradation patterns. The inhibition effect on cell growth was much more obvious in BDE-47 than BDE-209, with the EC50 values at 96 h calculated as 64.7 μg L−1 and 4070 μg L−1, respectively. Microalgal uptake rates showed BDE-209 diffused less into cells than BDE-47, with highest measured uptake rates of 0.145 × 10−7 μg h−1 cell−1 and 0.45 × 10−7 μg h−1 cell−1, respectively. The bioaccumulation amount by unit microalgal cell varied with PBDE concentrations and culture time, which appeared to be related to the changes of extracellular polymeric substances (EPS) and cellular neutral lipids under the toxicity of PBDEs. Finally, we found Chlorella sp. delayed the debromination patterns of BDE-209 compared to seawater. This study linked the toxicity, microalgal bioaccumulation and metabolism of PBDEs, provided new insights in the research of POPs by microalgae and marine food webs.

Retention of polybrominated diphenyl ethers and hydroxylated metabolites in paired human serum and milk in relation to CYP2B6 genotype

Polybrominated diphenyl ethers (PBDEs) and their hydroxylated metabolites (OH-BDEs) are endocrine disrupting compounds prevalent in human serum and breast milk. Retention of PBDEs and OH-BDEs in humans may be affected by differences in PBDE metabolism due to variants in cytochrome P450 2B6 (CYP2B6). The objectives of this study are to assess the partitioning profiles of PBDEs and OH-BDEs in forty-eight paired human serum and milk samples, and to evaluate the relationship between variants in CYP2B6 genotype and PBDE and OH-BDE accumulation in humans. Results show that the geometric mean (GM) concentrations of PBDEs are similar in serum (GM = 43.4 ng/g lipid) and milk samples (GM = 52.9 ng/g lipid), while OH-BDEs are retained primarily in serum (GM = 2.31 ng/g lipid), compared to milk (GM = 0.045 ng/g lipid). Participants with CYP2B6*6 genotype had a greater relative retention of PBDEs in serum and milk, and significant relationships (p < 0.05) were also observed for PBDE-47, 5-OH-BDE-47 and 6-OH-BDE-47 concentrations relative to CYP2B6*5 and CYP2B6*6 genotypes. These results are the first to show that CYP2B6 genotype is significantly related to the relative retention of PBDEs in humans, which may have direct implications for variability in the susceptibility of individuals to the potential adverse effects of these contaminants.

Novel Interactions between Gut Microbiome and Host Drug-Processing Genes Modify the Hepatic Metabolism of the Environmental Chemicals Polybrominated Diphenyl Ethers.

The gut microbiome is a novel frontier in xenobiotic metabolism. Polybrominated diphenyl ethers (PBDEs), especially BDE-47 (2, 2', 4, 4'-tetrabromodiphenyl ether) and BDE-99 (2, 2', 4, 4',5-pentabromodiphenyl ether), are among the most abundant and persistent environmental contaminants that produce a variety of toxicities. Little is known about how the gut microbiome affects the hepatic metabolism of PBDEs and the PBDE-mediated regulation of drug-processing genes (DPGs) in vivo. The goal of this study was to determine the role of gut microbiome in modulating the hepatic biotransformation of PBDEs. Nine-week-old male C57BL/6J conventional (CV) or germ-free (GF) mice were treated with vehicle, BDE-47 or BDE-99 (100 μmol/kg) for 4 days. Following BDE-47 treatment, GF mice had higher levels of 5-OH-BDE-47 but lower levels of four other metabolites in liver than CV mice; whereas following BDE-99 treatment GF mice had lower levels of four minor metabolites in liver than CV mice. RNA sequencing demonstrated that the hepatic expression of DPGs was regulated by both PBDEs and enterotypes. Under basal conditions, the lack of gut microbiome upregulated the Cyp2c subfamily but downregulated the Cyp3a subfamily. Following PBDE exposure, certain DPGs were differentially regulated by PBDEs in a gut microbiome-dependent manner. Interestingly, the lack of gut microbiome augmented PBDE-mediated upregulation of many DPGs, such as Cyp1a2 and Cyp3a11 in mouse liver, which was further confirmed by targeted metabolomics. The lack of gut microbiome also augmented the Cyp3a enzyme activity in liver. In conclusion, our study has unveiled a novel interaction between gut microbiome and the hepatic biotransformation of PBDEs.

Oxidation reactivity of 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE) by Compound I model of cytochrome P450s

Alternative brominated flame retardants (BFRs) have become prevalent as a consequence of restrictions on the use of polybrominated diphenyl ethers (PBDEs). For risk assessment of these alternatives, knowledge of their metabolism via cytochrome P450 enzymes is needed. We have previously proved that density functional theory (DFT) is able to predict the metabolism of PBDEs by revealing the molecular mechanisms. In the current study, the reactivity of 1,2-bis(2,4,6-tribromophenoxy)ethane and structurally similar chemicals with the Compound I model representing the active site of P450 enzymes was investigated. The DFT calculations delineated reaction pathways which lead to reasonable explanations for products that were detected by wet experiments, meanwhile intermediates which cannot be determined were also proposed. Results showed that alkyl hydrogen abstraction will lead to bis(2,4,6-tribromophenoxy)ethanol, which may undergo hydrolysis yielding 2,4,6-tribromophenol, a neurotoxic compound. In addition, a general pattern of oxidation reactivity regarding the 2,4,6-tribromophenyl moiety was observed among several model compounds. Our study has provided insights for convenient evaluation of the metabolism of other structurally similar BFRs.

Species-specific and structure-dependent debromination of polybrominated diphenyl ether in fish by in vitro hepatic metabolism.

To explore the cause of species-specific differences and structure-activity relationships in the debromination of polybrominated diphenyl ethers (PBDEs) in fish, a series of in vitro measurements of hepatic metabolism of PBDE were made using crucian carp (Carassius carassius) and catfish (Silurus asotus) and the activity of deiodinase in liver microsomes was measured. Debromination was observed in the crucian carp but not in the catfish. No difference was found in total deiodinase activity despite the activity of type 1 deiodinase in crucian carp being twice that of catfish. It is difficult to determine whether the differences in deiodinase activity were responsible for the species-specific differences observed. In crucian carp, penta-brominated diphenyl ether congeners exhibited the highest debromination rates, and the transformation rate decreased with an increasing number of substituted bromines. Adjacent bromine substitution in the phenyl ring was a necessary, but insufficient, condition for debromination in crucian carp. Doubly flanked bromine was always preferentially removed, while single-flanked bromine, meta-substituted bromine, was debrominated the most, followed by para- and then ortho-bromine. No debromination was observed for single-flanked bromine when there was a symmetrical structure with (2, 4, 6) bromine substitutions in 1 phenyl ring, indicating that this structure can improve resistance to debromination metabolism. Effects of Ni/Fe bimetallic nanoparticles on phytotoxicity and translocation of polybrominated diphenyl ethers in contaminated soil

In vivo studies of the interactions of polybrominated diphenyl ethers (PBDEs) in plants have generally focused on uptake, translocation, metabolism and accumulation, but there were limited reports about the phytotoxicity and translocation of PBDEs in contaminated soil with the effects of nanoparticles. In this study, the effects of Ni/Fe bimetallic nanoparticles on translocation of polybrominated diphenyl ethers (PBDEs) in contaminated soil and its phytotoxicity to Chinese cabbage were investigated by soil culture experiments. The results showed that the plant biomass, germination rate, and shoot and root lengths of treated soil (S-5) increased by 0.0044 g, 15%, and 5 and 6 mm, respectively, compared with untreated soil (S-2B). The average Ni and Fe contents of the edible parts(stem and leaf) of the S-5 sample, which contained 0.03 g/g Ni/Fe and 10 mg/kg BDE209, were measured at 1.71 and 184 mg/kg, respectively. The superoxide dismutase, peroxidase and catalase activities in the S-5 sample decreased by 12%, 6.1% and 5.9%, respectively, while compared with the S-2B sample. In all treatments, the contents of BDE209 and the total PBDEs in sample S-5 were lowest, suggesting that the fresh Ni/Fe nanoparticles had higher toxicity than that of the aged nanoparticles. And the lower brominated PBDEs (tri-to nona-) were detected in samples, indicating uptake, debromination and/or metabolism of PBDEs existed in plants. The phytotoxicity and translocation of BDE209 in the contaminated soil decreased as a result of the effects of the Ni/Fe bimetallic nanoparticles.

Development of an analytical method to quantify PBDEs, OH-BDEs, HBCDs, 2,4,6-TBP, EH-TBB, and BEH-TEBP in human serum.

Polybrominated diphenyl ethers (PBDEs) flame retardants (FRs) were phased-out in the mid-2000s (penta- and octaBDE) and 2013 (decaBDE); however, their hydroxylated metabolites (OH-BDEs) are still commonly detected in human serum. Today, novel FRs such as Firemaster® 550, a mixture that contains two brominated compounds, EH-TBB and BEH-TEBP are used as replacements for PBDEs in some applications, and there is a need to develop a comprehensive analytical method to assess exposure to both legacy PBDEs and novel FRs. This study developed a solid-phase extraction (SPE)-based method to analyze PBDEs, OH-BDEs, 2,4,6-tribromophenol (TBP), hexabromocylcododecane isomers (HBCDs), EH-TBB, and BEH-TEBP in human serum. Briefly, serum proteins were first denatured with formic acid, and then the target analytes were isolated using a SPE column. Finally, the extract was cleaned and fractioned using a silica SPE column. Method performance was assessed by spiking fetal bovine serum with 1-2ng of the target analytes, and method accuracy was quantified by comparison to a serum Standard Reference Material (SRM). The developed method showed good recovery and accuracy for all target analytes with the exception of the very low and very high molecular weight PBDE congeners. Using this method, 43 serum samples collected from the Healthy Pregnancy, Healthy Baby Study (HPHB) cohort in Durham, NC, USA were analyzed for FRs. A novel finding was the ubiquitous detection of 2,4,6-TBP, at levels greater than the individual PBDE congeners. Furthermore, 2,4,6-TBP was positively correlated with PBDEs, suggesting that they may have a similar source of exposure, or that 2,4,6-TBP may result from metabolism of PBDEs in vivo.

Levels of PBDEs in plasma of juvenile starlings (Sturnus vulgaris) from British Columbia, Canada and assessment of PBDE metabolism by avian liver microsomes

In this study, the levels of polybrominated diphenyl ethers (PBDEs), HO-PBDEs, and bromophenols were monitored in starling chick plasma samples collected in Delta (British Columbia, Canada) close to the Vancouver municipal landfill and in Glen Valley, a rural area in British Columbia. The in vitro formation of hydroxylated metabolites of 2,2′,4,4′-tetrabromodiphenyl ether (BDE-47) and 2,2′,4,4′,5-pentabromodiphenyl ether (BDE-99) was also investigated using starling chick liver microsomes. Total PBDE plasma levels were approximately 60 times higher in starling chicks from Delta than from Glen Valley, suggesting that the Delta site is a major source of PBDEs for the local population of starlings and that PBDEs previously measured in starling eggs are bioavailable to chicks. In both locations, BDE-47 and BDE-99 were the two major congeners present at similar concentrations, suggesting contamination with the Penta-BDE mixture. Among the several possible hydroxylated metabolites of PBDEs monitored in starling plasma, only 2,4,5-tribromophenol was detected and its levels did not exceed 18±7pg/mL. Also, several hydroxylated metabolites of BDE-47 and BDE-99 were formed by starling chick liver microsomes, but in low amounts. Therefore, our data consistently suggest that oxidative metabolism of PBDEs in starling chicks proceeds at low rate in vivo and in vitro. In conclusion, the landfill located in Delta is a relevant source of bioavailable PBDEs for the local starling population. Because of the limited ability of starling chicks to metabolize PBDEs, these compounds are likely to bioaccumulate in starlings over time. The possible toxicological implications of PBDEs bioaccumulation in starlings are currently unknown and require further research.

Fast liquid chromatographic–tandem mass spectrometric method using mixed-mode phase chromatography and solid phase extraction for the determination of 12 mono-hydroxylated brominated diphenyl ethers in human serum

Hydroxylated polybrominated diphenyl ethers (OH-PBDEs) are formed from the oxidative metabolism of polybrominated diphenyl ethers (PBDEs) in humans, rats and mice, but their quantitation in human blood and other matrices with liquid chromatography–mass spectrometric techniques has been a challenge. In this study, a novel analytical method was developed and validated using only 250μL of human serum for the quantitation of twelve OH-PBDEs, fully chromatographically separated in a 15min analytical run. This method includes two novel approaches: an enzymatic hydrolysis procedure and a chromatographic separation using a mixed mode chromatography column. The enzymatic hydrolysis (EH) was found critical for 4′-OH-BDE17, which was not detectable without it. For the sample clean up, a solid phase extraction protocol was developed and validated for the extraction of the 12 congeners from human serum. In addition, for the first time baseline resolution of two components was achieved that correspond to a single peak previously identified as 6′-OH-BDE99. The method was validated for linearity, accuracy, precision, matrix effects, limit of quantification, limit of detection, sample stability and overall efficiency. Recoveries (absolute and relative) ranged from 66 to 130% with relative standard deviations <21% for all analytes. Limit of detection and quantitation ranged from 4 to 90pgmL−1 and 6–120pgmL−1, respectively, with no carry over effects. This method was applied in ten commercially available human serum samples from the general US population. The mean values of the congeners detected in all samples are 4′-OH-BDE17 (34.2pgmL−1), 4-OH-BDE42 (33.9pgmL−1), 5-OH-BDE47 (17.5pgmL−1) and 4′-OH-BDE49 (12.4pgmL−1).

Alterations of endogenous metabolites in urine of rats exposed to decabromodiphenyl ether using metabonomic approaches

There is large usage of polybrominated diphenyl ethers (PBDEs) especially for decabromodiphenyl ether (BDE-209, Deca-BDE) in controlling the risks of fire. The toxicological effects of PBDEs are worth being concerned about. Female SD rats were daily gavaged with BDE-209 ether at the dose of 100 mg/kg for 20 days. Histological observation was performed for the screening of the target organs for BDE-209 exposure. The distribution and metabolism of PBDEs in the exposed main organs were evidenced by HRGC-HRMS. Alterations of the endogenous metabolite concentrations in urine were investigated using metabonomic approaches based on 1H NMR spectrum. Histopathological changes including serious edema in kidney, hepatocellular spotty necrosis and perivasculitis in liver indicated that BDE-209 caused potential influences on endogenous metabolism in the exposed liver and the kidney. BDE-209 was found to be highly accumulated in lipid, ovary, kidney and liver after 20 days' exposure. Occurrence of other lower brominated PBDEs in the rats demonstrated that reductive debromination process happened in vivo. Hydroxylated and methoxylated–BDEs, as metabolism products, were also detected in the rat tissues. A total of 12 different endogenous metabolites showed obvious alterations in urine from the exposed rats, indicating the disturbance of the corresponding internal biochemical processes induced by BDE-209 exposure. These findings in vivo suggested the potential health risk might be of concern due to the toxicological effects of BDE-209 as a ubiquitous compound in the environment.

In Vivo Metabolism of 2,2′,4,4′-Tetrabromodiphenyl Ether (BDE-47) in Young Whole Pumpkin Plant

Polybrominated diphenyl ethers (PBDEs) are widely distributed persistent organic pollutants. In vitro and in vivo research using various animal models have shown that PBDEs might be transformed to hydroxylated PBDEs, but there are few studies on in vivo metabolism of PBDEs by intact whole plants. In this research, pumpkin plants (Cucurbita maxima × C. moschata) were hydroponically exposed to 2,2',4,4'-tetrabromodiphenyl ether (BDE-47). A debromination product (BDE-28) and four hydroxylated metabolites (5-OH-BDE-47, 6-OH-BDE-47, 4'-OH-BDE-49, and 4-OH-BDE-42) were detected in different parts of the whole plant. In addition, 4-methoxylated-2,2',3,4'-tetraBDE (4-MeO-BDE-42) was observed as a methoxylation product. Root exudates in solution were found to play an important role in metabolizing BDE-47 to a specific OH-PBDE: 4'-OH-BDE-49. BDE-28 was found to translocate more easily and accumulate in shoots than BDE-47 due to the lower hydrophobicity and molecular weight. The concentration ratio between metabolites and parent compound BDE-47 was lower for OH-PBDEs than that for both BDE-28 and 4-MeO-BDE-42. The metabolism pathway of BDE-47 in young whole plants was proposed in this study.

Biological metabolic transformation of polybrominateddiphenyl ethers

As widely used flame retardants polybrominateddiphenyl ethers(PBDEs)and their hydroxyl and methoxyl derivatives(OH-PBDEs and MeO-PBDEs)have been detected from various kinds of environmental media, different organisms and human bodies. The sources of low brominated PBDEs, OH-PBDEs, and MeO-PBDEs detected in organisms are not clearly elucidated, which may either be uptaken from the surrounding environment or formed via biotransformation in vivo . Since the biotransformation mechanism and transformation products of PBDEs in organism are closely related with their toxicities, it has been the focus of many literatures. Based on the analysis of distributions and sources of PBDEs and their metabolic derivatives in organism, the research progresses of reductive debromination and oxidation metabolism of PBDEs in organisms have been reviewed. The metabolism, pathways, and key enzymes involved in the biotransformation of PBDEs both in vivo and in vitro are explored. Directions for further studies on the bioaccumulation and biotransformation of PBDEs are proposed.

Tissue accumulation and species‐specific metabolism of technical pentabrominated diphenyl ether (DE‐71) in two predator fish

The tissue-specific accumulation and species-specific metabolism of polybrominated diphenyl ethers (PBDEs) in two predator fish species (redtail catfish and oscar fish) feeding on the same prey (tiger barb) that was exposed to technical pentabrominated diphenyl ether (DE-71) in the laboratory were investigated. The trends in the wet-weight tissue concentration of PBDEs in two predatory fish species suggested that the tissue distribution of PBDEs occurs through a series of events involving passive diffusion to the lipid compartment. A comparison of the fugacities of PBDEs in various tissues and in the serum revealed that the liver, gill, and perivisceral adipose tissue readily achieved equilibrium with the serum, but the muscle, kidney, and intestine exhibited the potential to accumulate PBDEs. The lower fugacities of PBDEs in the intestine may have significance in the transportation of PBDEs from prey to predatory fish. No tissue-specific differences in PBDE congener profiles were found, while interspecies differences in PBDE profiles were evident. The difference in profiles between two species could be attributed to species-specific debromination of PBDE. No metabolic debromination of PBDE was observed in redtail catfish, but extensive debromination of PBDEs occurred in oscar fish. Several hydroxylated PBDEs (OH-PBDEs) were detected in serum samples from the two fish species, but no methoxylated PBDEs were found. The similarities in the OH-PBDE congener profile and the ratio of OH-PBDEs to total PBDEs between the two fish species indicated that the hydroxylation of PBDEs might not be species-specific.

Synthesis and Characterization of Bromophenol Glucuronide and Sulfate Conjugates for Their Direct LC-MS/MS Quantification in Human Urine as Potential Exposure Markers for Polybrominated Diphenyl Ethers

Bromophenol glucuronide and sulfate conjugates have been reported to be products of mammalian metabolism of polybrominated diphenyl ethers (PBDEs), a group of additive flame-retardants found ubiquitously in the environment. In order to explore their occurrence in human urine, four water-soluble bromophenol conjugates, namely, 2,4-dibromophenyl glucuronide, 2,4,6-tribromophenyl glucuronide, 2,4-dibromophenyl sulfate, and 2,4,6-tribromophenyl sulfate, were synthesized, purified, and characterized. An analytical protocol using solid-phase extraction and ion-paired liquid chromatography-electrospray tandem mass spectrometry (LC-ESI-MS/MS) quantification has been developed for the direct and simultaneous determination of these glucuronide and sulfate conjugates in human urine samples. The limit of detections for all analytes were below 13 pg mL(-1), with 73-101% analyte recovery and 7.2-8.6% repeatability. The method was applied to analyze 20 human urine samples collected randomly from voluntary donors in Hong Kong SAR, China. All the samples were found to contain one or more of the bromophenol conjugates, with concentration ranging from 0.13-2.45 μg g(-1) creatinine. To the best of our knowledge, this is the first analytical protocol for the direct and simultaneous monitoring of these potential phase II metabolites of PBDEs in human urine. Our results have also suggested the potential of these bromophenol conjugates in human urine to be convenient molecular markers for the quantification of population exposure to PBDEs.

Oxidative Metabolism of BDE-99 by Human Liver Microsomes: Predominant Role of CYP2B6

Hydroxylated polybrominated diphenyl ethers (PBDEs) have been found in human serum, suggesting that they are formed by in vivo oxidative metabolism of PBDEs. However, the biotransformation of 2,2',4,4',5-pentabromodiphenyl ether (BDE-99), a major PBDE detected in human tissue and environmental samples, is poorly understood. In the present study, the oxidative metabolism of BDE-99 was assessed using pooled and single-donor human liver microsomes, a panel of human recombinant cytochrome P450 (CYP) enzymes, and CYP-specific antibodies. Hydroxylated metabolites were quantified using a liquid chromatography/tandem mass spectrometry-based method. In total, 10 hydroxylated metabolites of BDE-99 were produced by human liver microsomes. Six metabolites were identified as 2,4,5-tribromophenol (2,4,5-TBP), 4-OH-BDE-90, 5'-OH-BDE-99, 6'-OH-BDE-99, 4'-OH-BDE-101, and 2-OH-BDE-123 using authentic standards. Three monohydroxy- and one dihydroxy-pentabrominated metabolites were unidentified. Rates of formation of the three major metabolites (2,4,5-TBP, 5'-OH-BDE-99, and 4'-OH-BDE-101) by human liver microsomes ranged from 24.4 to 44.8 pmol/min/mg protein. Additional experiments demonstrated that the dihydroxylated metabolite was a primary metabolite of BDE-99 and was not produced by hydroxylation of a monohydroxy metabolite. Among the panel of recombinant CYP enzymes tested, formation of all 10 hydroxylated metabolites was catalyzed solely by CYP2B6. A combined approach using antibodies to CYP2B6 and single-donor liver microsomes expressing a wide range of CYP2B6 levels confirmed that CYP2B6 was responsible for the biotransformation of BDE-99. Collectively, the results show that the oxidative metabolism of BDE-99 by human liver microsomes is catalyzed solely by CYP2B6 and is an important determinant of the toxicity and bioaccumulation of BDE-99 in humans.

Debrominated, hydroxylated and methoxylated metabolism in maize (Zea mays L.) exposed to lesser polybrominated diphenyl ethers (PBDEs)

A hydroponic experiment was conducted to investigate the debrominated, hydroxylated and methoxylated metabolism of polybrominated diphenyl ethers (PBDEs, BDE-15, -28 and -47) in maize. A total of six debrominated metabolites (de-PBDEs), seven hydroxylated PBDEs (OH-PBDEs, including two unidentified OH-di-PBDEs and one unidentified OH-tri-PBDE) and four methoxylated PBDEs (MeO-PBDEs) were determined in the exposed plants. The metabolic products were detected in maize only after 12h of exposure to the PBDEs. However, the concentration of each type of the metabolites (de-PBDEs, OH-PBDEs or MeO-PBDEs) decreased at the later exposure time, possibly due to further metabolism. The removal of a bromine atom or the introduction of a hydroxyl/methoxy group was easier at the ortho-positions on the biphenyl structure than at the para-positions. Concentration ratios of the total debrominated, hydroxylated or methoxylated metabolites to the parent congener (BDE-28 or -47) generally followed the order of leaves>stems≫roots, and MeO-PBDEs>de-PBDEs≫OH-PBDEs. These results suggest that metabolism occurred preferentially in leaves and stems than in roots. Less transformation and shorter elimination half-life of OH-PBDEs would contribute to the lower concentrations of OH-PBDEs than of de-PBDEs or MeO-PBDEs in maize.

Metabolism of polybrominated diphenyl ethers and tetrabromobisphenol A by fish liver subcellular fractions in vitro

Polybrominated diphenyl ethers (PBDEs) and tetrabromobisphenol A (TBBPA) are two major flame retardants that accumulate in fish tissues and are potentially toxic. Their debrominated and oxidated derivatives were also reported in fish tissues although the sources of theses derivatives were unidentified. Our study was to determine whether PBDEs and TBBPA could be metabolized by fish liver subcellular fractions in vitro and to identify what types of metabolites were formed. Liver microsomes and S9 fractions of crucian carp (Carassius auratus) were exposed to 4,4′-dibromodiphenyl ether (BDE 15), 2,2′,4,4′-tetrabromodiphenyl ether (BDE 47) or TBBPA solutions for 4h. Exposure of liver subcellular fractions to BDE 15 resulted in the formation of bromophenol and two monohydroxylated dibromodiphenyl ether metabolites. Neither in microsomes nor in S9 studies has revealed the presence of hydroxylated metabolites with BDE 47 exposure which indicated that the oxidation reactions in vitro were hindered by the increased number of bromine substituents on the PBDEs. TBBPA underwent an oxidative cleavage near the central carbon of the molecule, which led to the production of 2,6-dibromo-4-isopropyl-phenol and three unidentified metabolites. Another metabolite of TBBPA characterized as a hexa-brominated compound with three aromatic rings was also found in the liver subcellular fractions. These results suggest that the biotransformation of BDE 15 and TBBPA in fish liver is mediated by cytochrome P450 (CYP450) enzymes, as revealed by the formation of hydroxylated metabolites and oxidative bond cleavage products. Moreover, further studies on the identification of specific CYP450 isozymes involved in the biotransformation revealed that CYP1A was the major enzyme responsible for the biotransformation of BDE 15 and TBBPA in fish liver subcellular fractions and CYP3A4 also played a major role in metabolism of TBBPA.

Decabromobiphenyl, Polybrominated Diphenyl Ethers, and Brominated Phenolic Compounds in Serum of Cats Diagnosed With the Endocrine Disease Feline Hyperthyroidism

The incidence of cats being diagnosed with feline hyperthyroidism (FH) has increased greatly since it was first described in 1979. The cause of FH has not been established. Hypothetically, there is a link between increasing FH and exposure to brominated flame retardants. Much greater polybrominated diphenyl ethers (PBDE) concentrations have been reported in cat serum compared with human serum, likely due to cat licking behaviour. This study aimed to extend the present identification of brominated compounds in cat serum, with a focus on hydroxylated metabolites of PBDE, to improve the understanding of feline metabolism of PBDEs. A pooled serum sample from 30 Swedish pet cats with FH was analysed, and brominated species were identified. The results showed exposure to the discontinued flame retardant decabromobiphenyl (BB-209) and technical penta- and octa-BDEs. Altogether 12 PBDE congeners were identified along with 2'-MeO-BDE68. Furthermore, 2,4-dibromophenol, 2,4,6-, 2,4,5- and 2,3,4-tribromophenol plus 2'-OH-BDE68, 6-OH-BDE47, 5-OH-BDE47, 4'-OH-BDE49 were identified. 2,4,6-tribromophenol and 6-OH-BDE47 were the most prominent species in cat serum. Considering that these are natural products, it can be concluded that metabolism of PBDEs to OH-PBDEs is not a major route of PBDE elimination in cats. It is notable that BB-209, 6-OH-BDE47, and 2,4,6-tribromophenol all suggested that endocrine-disrupting chemicals were present in high concentrations in cat serum.

Uptake, translocation and metabolism of polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs) in maize (Zea mays L.)

A hydroponic experiment was conducted in the present study to investigate and compare plant uptake, translocation and metabolism of polybrominated diphenyl ethers (PBDEs) of BDE-15, BDE-28 and BDE-47 and polychlorinated biphenyls (PCBs) of PCB-15, PCB-28 and PCB-47 in maize. Root concentrations of BDE-15, BDE-28 and BDE-47 were consistently higher than PCB-15, PCB-28 and PCB-47, respectively. A significantly positive correlation was found between logRCF (root concentration factor) and logKow of these PBDEs and PCBs, suggesting a control role of their partitioning in plant uptake. The translocation factors (TFs, Cstem/Croot) of PBDEs were generally lower than those of PCBs of the same halogen-substitutions, demonstrating easier transport of PCBs than PBDEs. Metabolites mono-, di- and tri-BDEs and PCBs were detected, suggesting the existence of in vivo metabolism of PBDEs and PCBs in maize. Dehalogenation and rearrangement of halogen atoms were identified, and some similarities but also significant differences existed between the PBDEs and PCBs. PBDEs in maize were, in general, more susceptible to metabolism compared with PCBs of the same halogen-substitutions. This is the first comparative report on the uptake, translocation and metabolism of PBDEs and PCBs in plants.
