Correction of cellular immunity with biferon-C in piglets in case of specific prevention of circovirosis and mycoplasmosis

The results of studying the effect of biferon-C containing recombinant porcine interferons -alpha and -gamma on cellular nonspecific and adaptive immunity in piglets in case of specific prevention of circovirosis and mycoplasmosis are presented. The corrective effect of the drug on cellular immunity was stated, manifested by an increase in the absorption and functional and metabolic activity of phagocytes, the relative and absolute number of T- and B-lymphocytes, T-cells with helper activity and a lower formation of the level of T-suppressors, an increase in the T-helpers/T-suppressors ratio.

The ancient Thracian endemic plant Haberlea rhodopensis Friv. and related species: A review.

Haberlea rhodopensis (HR) use dates back to the Thracian and Roman periods. Bulgarians call it Orpheus flower and exploit its leaves for making tea and extracts with detoxifying, tonic, restorative and rejuvenating effects. HR was traditionally applied in wound healing and treatment of cattle diseases. The general aim of the review was to analyze the progress of phytochemical and pharmacological studies on HR, focusing on its radioprotective and immunomodulating effects. The main source material for the review was collected using several global search engines with the phrase: Haberlea rhodopensis, as well as Bulgarian books and dissertations. HR metabolite profile includes large amounts of free sugars, polyols, polysaccharides (PS), flavonoids, phenolic acids and carotenoids. The radioprotective effect of 70% ethanolic leaf extract (70HREE) is explained by preservation of lymphocytes, other blood cells and testicular tissue from aberration under γ-radiation via stimulation of antioxidant enzymes and neutralization of free radicals. The extract immunomodulating activity results from raised antibody response, stem and neutrophil cell count, complement system activation, anti-tumour and anti-inflammatory effects. The detoxifying, restorative, rejuvenating and wound healing plant properties known to ethnomedicine were supported by radioprotective and immunomodulating studies. Metabolites of phenolic origin involved in HR resurrection are supposed to contribute to its radioprotective, immunomodulatory, anti-mutagenic and anti-aging effects. However, there is no chemical characterization of 70HREE in the investigations with humans and animals. Structure-activity relationship studies on HR immunomodulating and radioprotective compounds, and on their mode of action are required. They should include not only phenols but PS and other unexplored molecules. The metabolic activity of phagocytes, platelets and lymphocytes triggered by HR extracts has to be examined to elucidate their immunostimulatory potential. HR formulations can be tested in cosmetic, food and medical products as adjuvants to treat infectious, chronic inflammatory and tumour diseases, and especially in patients undergoing radiotherapy.

Influence of beta-glucan and vaccination against Lawsonia intracellularis on selected immune indices in weaned piglets

Abstract The aim of this paper was to study the immunostimulating effects of feed additive containing beta-glucan given to pregnant sows and consecutive specific immunomodulation by vaccination of their suckling piglets. Thirty-five suckling piglets from the control and experimental group of sows were divided into four groups. Selected blood indices of immune profile (TIg, IMA, SI), specific antibodies against Lawsonia intracellularis formed in the blood serum of suckling piglets after vaccination, as well as production indices were observed. In all the groups, birth body weights were similar. After weaning, average daily weight gains were significantly higher (p < 0.05) in the group of vaccinated suckling piglets, as well as in suckling piglets from experimental sows. The same trend was recorded in average TIg values. Significant effects (p < 0.01) of beta-glucan and vaccination were observed in the changes of the index of metabolic activity of phagocytes (IMA) and less markedly in the index of stimulation activity of lymphocytes (SI). The least favorable results were observed in the group without both vaccination and beta-glucan. The effect of vaccination was manifested by ELISA estimation of specific antibodies (PI), when in both non-vaccinated groups (A and D) we observed minimum antibodies levels compared with vaccinated groups (B and C), which showed a high positivity without significant differences between the beta-glucan stimulated (C) and the non-stimulated group (B). These results suggest that beta-glucan given to sows and consecutive vaccination of suckling piglets can stimulate their non specific and specific immunity.

The effect of monotherapy and combined therapy with NSC-631570 (Ukrain) on growth of low- and high-metastasizing B16 melanoma in mice

NSC-631570 (ukrain) is a semisynthetic derivative of the Chelidonium majus alcaloids and the alkylans thiotepa. It exerts a selective cytotoxic effect on tumor cells in vitro and in vivo and shows the ability to modulate immunocyte functions. Purpose. The aim of our work was to carry out a comparative investigation of the effects of NSC-631570 alone or in combination with pathogen-associated molecules (PAM) on the growth of low- and high-metastasizing melanoma B16 in mice. NSC-631570 was administered intravenously and PAM intramuscularly to tumor-bearing mice seven times every third day, starting from the second day after the transplantation of tumor cells. The effect of monotherapy and combined therapy on tumor growth was evaluated by the indices of tumor growth inhibition in experimental animals. Cell cycle distribution of cancer cells was determined by flow cytometry. TAP1 and TAP2 expression was evaluated by RT-PCR. The metabolic activity of phagocytes was determined by NBT-test, phagocytosis was tested by flow cytometry, and arginase activity was estimated by colorimetric determination of urea. Combined therapy and monotherapy with NSC-631570 resulted in significant inhibition of tumor growth in melanoma-bearing mice. Monotherapy with Ukrain was more effective in mice with high-metastasizing tumors. The therapeutic efficacy of NSC-631570 used in combination with PAM was more expressed in mice with low-metastasizing melanoma. The effectiveness of monotherapy and combined therapy with NSC-631570 in the treatment of melanoma B16 depends on the biological properties of the tumor and the immune state of the organism.

Effects of Short-Term Hypothermal and Contrast Exposure on Immunophysiological Parameters of Laboratory Animals

Experiments on inbred animals showed that short-term exposure in cold water significantly modified structural and functional parameters of the immune system at different levels of its organization, from bone marrow hemopoiesis to effector stage of the immune response to antigen. The thermal factor caused changes in nonspecific and specific mechanisms of the immune system. Hypothermal exposure (7-9 degrees C, 5 sec) increased the thymic index and bone marrow lymphocyte count, reduced absorption capacity and stimulated metabolic activity of phagocytes, stimulated cell-mediated and suppressed humoral immunity. Contrast exposure in cold and hot water (7-9 degrees C, 5 sec/40-42 degrees C, 30 sec) increased monocyte count in bone marrow and reduced it in the their peripheral blood, reduced metabolic activity of phagocytes, stimulated cell-mediated and suppressed humoral immunity. These data demonstrate physiological mechanisms of interactions between the thermoregulatory and immune systems.

Influence of Linseed and Fish Oil on Metabolic and Immunological Indicators of Laying Hens

The metabolic and immunological indicators were determined in the blood of laying hens of ISA BROWN hybrid divided into three groups, the control group and two experimental groups. The administration of n-3 polyunsaturated fatty acids (PUFA) in the form of linseed (Group 1) and fish oil (Group 2) and α-tocopherol as antioxidant to laying hens resulted in a significant increase in concentrations of high density lipoproteins (HDL) cholesterol (P &lt; 0.05), eicosapentaenoic (EPA), docosahexaenoic (DHA) and α-linolenic acids (AA) in blood in comparison to the control group. Significantly lower levels of cholesterol (CHOL) were determined in both experimental groups at the third sampling (P &lt; 0.05) and arachidonic acid (AA) in the fish group (P &lt; 0.01). The metabolic activity of phagocytes and polyclonal activation of lymphocytes showed no significant differences and remained within the physiological range. Oral administration of n-3 PUFA showed no significant increase of the immune response of experimental animals.

Effects of supplementation of organic-bound trace elements on blood and tissues - micromineral profile and immune parameters of piglets novotný j, pistl j and kováČ g

An 8-week experiment was carried out on piglets to observe the effects of organic trace element supplementation (Piglet Booster Alttech, Inc) on the micromineral profile of blood serum, immunological parameters, body weight (before and after weaning), and tissue concentrations of Zn, Fe, Cu, Mn, and Se. Forty piglets from 4 sows were divided into 2 groups (control – 19 piglets, experimental – 21). Organic trace element supplementation affected positively the body weight of the experimental animals. The metabolic activity of phagocytes and polyclonal activation of lymphocytes were significantly increased (p< 0.05 – 0.01) in the second phase of the experiment. Significant increases in Fe, Cu, and Se in the blood serum (p<0.05 – 0.001) and Zn, Se, and Mn in the heart muscle (p<0.05) were recorded. Increased concentrations of minerals were observed in the heart (Fe), liver (Zn, Fe, and Se), kidneys (Zn, Fe, Se, and Mn), and loin muscles (Fe, Se, and Mn).

Effect of oil containing n-3 polyunsaturated fatty acids (PUFA) on the immune response and growth factors in piglets.

Oral administration of n-3 polyunsaturated fatty acids (PUFA) to piglets slightly enhances the immune response. As compared to the control, in the experimental piglets the absolute values of monocytes in the peripheral blood were significantly increased (P < 0.05), while the metabolic activity of phagocytes and the number of lymphocytes within the individual subpopulations were slightly higher. The level of growth factors, determined on the basis of somatomedin in the blood serum, was significantly higher in the experimental group (P < 0.05). n-3 PUFA interfere with the synthesis of prostaglandins and influence the metabolism of fatty acids. This finding may contribute to the therapy of inflammatory processes influencing immune and growth factors in piglets.

Measurement of whole blood phagocyte chemiluminescence in the Wistar rat.

The factors influencing the rat whole blood chemiluminescence (CL): concentrations of blood, luminol, zymosan or opsonized zymosan, volume of the reaction mixture, storage time of blood samples and the presence of anticoagulants were evaluated. The CL micromethod described provides a fast and sensitive tool for the determination of metabolic activity of phagocytes in the microlitre range of rat whole blood.

Effect of prostatilen on the metabolic activity of phagocytes in patients with chronic pyelonephritis

The investigation of the metabolic activity of leukocytes and the effect of bioregulation peptide of the prostate prostatilen on it is performed. It is found that suppression of oxygendependent microbicide systems of phagocytes, more pronounced in the initial form of the disease, occurs in patients with chronic pyelonephritis. The differences in the activity of oxygenindependent systems of phagocytes in patients with chronic pyelonephritis and healthy persons are not revealed. The increase of NST-test values after the use of prostatilen is noted.

Metabolic activity of phagocytes in experimental sporotrichosis.

Phagocytosis plays an important role as a protective mechanism against infections, since polymorphonuclear leukocytes (PMN) and macrophages are the first cellular lines opposed to aggressive microorganisms. In patients with sporotrichosis a diminished capability of killing engulfed yeast by their PMN has been described, but the origin of this deficiency remains unknown. In this work, partial aspects of the oxidative metabolism of PMN leukocytes and peritoneal macrophages of mongolian gerbils experimentally infected with sporotrichosis were studied. For this purpose the nitroblue tetrazolium (NBT) test as described by Baehner and Nathan (1) and myeloperoxidase activity measured according to Kaplow's method were utilized. The PMN and macrophages of mongolian gerbils infected with sporotrichosis showed increased reduction of NBT when compared with the phagocytic cells of normal ones, as is usually observed in most infections. Myeloperoxidase activity was diminished in both PMN and macrophages, but this diminution was statistically significant only in PMN leukocytes. These results show that part of the oxidative mechanisms of phagocytic cells can be impaired in experimental sporotrichosis, and could be correlated with the diminished fungicidal activity of PMN leukocytes obtained from patients infected with sporotrichosis.
