Pushed to the edge: Environmental factors drive ecological responses of Aphanius fasciatus when in sympatry with invasive Gambusia holbrooki

Abstract Interspecific competition may be particularly exacerbated when occurring between native and invasive alien species, with environmental conditions affecting drivers of natural selection and shaping spatio‐temporal dynamics of animal communities. The effects of invaders are particularly evident in aquatic ecosystems with delicate and complex balances, such as Mediterranean coastal wetlands, characterized by unpredictable water flows and variable reserves. Understanding how native species respond to human‐induced biological invasions is of paramount importance for planning management measures aiming at conservation purposes. This study investigated whether the occurrence of a competing invader species resulted in changes in the ecological adaptations of a native species by comparing two coastal wetland fish species: the invasive eastern mosquitofish Gambusia holbrooki and the native Mediterranean killifish Aphanius fasciatus of conservation concern. The hypothesis was tested by sampling fishes at a coastal Ramsar site in central Italy, over 2 years. Generalized linear mixed models were used to examine the occurrence of killifish in relation to water parameters and the presence of its putative competitor. The aim was to determine whether salinity, temperature, and water depth influenced the presence and abundance of killifish when in sympatry with mosquitofish. Both the probability of presence and abundance of killifish increased with water salinity, but this increase was stronger when mosquitofish was present. Increasing water temperature and water depth were associated with a higher probability of presence and abundance of killifish respectively, although the effect of water depth was small. Both salinity and the presence of the invader acted synergistically in characterizing and shaping the population structure of the native species. Living in the most extreme and stressed environments, the killifish is potentially at higher risk of extinction. These results highlight the importance of considering the interplay between multiple abiotic stressors combined with comparative data between invasive and native species as increasingly critical to aid in conservation efforts.

A comparative analysis of habitat quality between artificial and natural creeks in the Mediterranean killifish Aphanius fasciatus: Implications for conservation

Abstract In transitional water ecosystems, habitat loss is one of the major threats to fish biodiversity. This is particularly true in the case of temperate salt marshes. Good quality artificial novel habitats may compensate for the loss of natural habitats, acting as a refuge for fish populations. The aim of this study was to use coloured ornamental traits exhibited by the males of a small lagoon‐resident fish, the Mediterranean killifish Aphanius fasciatus (Valenciennes, 1821), to assess habitat quality in both the natural and artificial creeks of the Venice lagoon. The conspicuousness of two secondary sexual traits, the yellow/orange caudal fin pigmentation and the contrast between white and dark vertical bars on body sides, proved to be greater in artificial habitats compared to natural salt marshes, highlighting the refuge function of artificial creeks. The use of secondary sexual traits could be extended to other fish species and to other aquatic habitats as a non‐invasive approach. This would allow the identification of good quality habitats in the context of fish population management and conservation.

Aphanius fasciatus: a molecular model of scoliosis?

Observational study of Killifish with spinal deformities OBJECTIVE: To evaluate the morphology and molecular biology of Aphanius fasciatus with severe spine deformities. Idiopathic Scoliosis affects 3% of the population and is an abnormal three-dimensional curvature of the spine with unknown cause. The lack of a model system with naturally occurring spinal curvatures has hindered research on the etiology of IS. The Mediterranean killifish Aphanius fasciatus, collected from the coast of Sfax (Tunisia), which has an inborn skeletal deformity was chosen. We used morphologic features to evaluate the severity of scoliosis according to the different types and performed a biochemical analysis using factors previously studied in humans (estradiol, melatonin and Insulin Growth Factor 1 "IGF-1"). We have detected relevant molecular deviations that occur in Killifish deformities and the fish with severe scoliosis are smaller and less old than the ones with milder scolioses. Furthermore, a significant change in levels of ovarian estradiol, liver IGF-1 and brain melatonin was noted between deformed and normal fish. Aphanius fasciatus could be used as a molecular model system to study the etiology of IS in humans as the characterization of the Aphanius fasciatus scoliosis syndrome has revealed morphological and biochemical parallels to IS. However, it is important to note the limitations of the proposed model, including the short lifespan of the fish. III.

The occurrence, distribution and biology of invasive fish species in fresh and brackish water bodies of NE Morocco

Monitoring the presence and expansion of alien species and upgrading their biological and ecological knowledge seems crucial to mitigate their possible impact on native communities. Within inland superficial waters, alien fish represent an important threat to the biodiversity and studies on their impact on native communities have increased around the world in the last years. However, little is known about their occurrence, biology and influences in North Africa in general, and more specifically in Morocco. In the present work we aimed to: 1) investigate the presence of any native Aphanius species, especially the Mediterranean killifish Aphanius fasciatus recorded from the lower basin of the Moulouya River (NE Morocco); 2) monitor the presence and expansion of two invasive species, the eastern mosquitofish Gambusia holbrooki and the mummichog Fundulus heteroclitus; and 3) contribute to the understanding of the ecological and abiotic affinities that govern the distribution of these alien fishes in North Africa. To achieve these goals, several field sampling campaigns were carried out between 2014 and 2018 across eastern Morocco, comprising the administrative Oriental Region and the Moulouya River Basin and covering an area of 119,268 km2. No native Aphanius species were found. The eastern mosquitofish has invaded the freshwater hydrosystems of the northern part of Morocco, including the study area, while the mummichog is currently limited to the brackish and salty wetlands of Lower Moulouya. Age, growth and reproduction of the Mediterranean killifish Aphanius fasciatus Nardo, 1827 in Mellah Lagoon (Eastern Algeria)

This paper studies the biology of reproduction and growth of the Mediterranean killifish Aphanius fasciatus in Mellah Lagoon (Algeria). This work helps to complete data lacks for this species concerning West Mediterranean and especially the westmost known distributions limit. A total of 1169 individuals (14.3 mm ≥ TL ≥ 54.4 mm, 0.06 g ≥ TW ≥ 3.89 g) caught monthly from January to December 2012, were examined. The population has a 6-year life cycle. Length–weight relationship was estimated as TW = 0.0093 LT3.513 (r2 = 0.804) for males and TW = 0.0091 TL3.504 (r2 = 0.766) for females. The von Bertalanffy growth function fitted to back-calculated size at age data was: Lt = 49.45 [1 - e-0.274 (t + 1.177)] for males, Lt = 59.03 [1 - e-0.179 (t + 1.514)] for females. The growth performance index (Φ) indicate that females (Φ = 6.67) grew slightly faster than males (Φ = 6.50). Sex-ratio was 1:1.6 in favor of females. The reproductive season extended from February to July. The length at first sexual maturity was 3.84 cm for males and 4.14 cm for females.

The role of habitat choice in micro-evolutionary dynamics: An experimental study on the Mediterranean killifish Aphanius fasciatus (Cyprinodontidae).

Habitat choice is defined as a nonrandom distribution of genotypes in different microhabitats. Therefore, it could exert a great impact on the genetic variance of natural populations by promoting genetic divergence, local adaptation, and may even lead to sympatric speciation. Despite this potential role in micro‐ and macro‐evolutionary processes, there is little empirical evidence that the various genotypes within a population may differ in habitat choice‐related behaviors. Here, we tested whether habitat choice may have contributed to genetic divergence within a local population of the Mediterranean killifish Aphanius fasciatus, which emerged between groups inhabiting microhabitats with different oxygen concentrations during previous field studies. In a first experiment, we studied the distribution of individuals in conditions of hypoxia and normoxia to test whether they had a different ability to shy away from a hypoxic environment; in a second experiment, we analyzed the individual behavior of fish separately in the two conditions, to verify whether they showed peculiar behavioral responses linked to a possible differential distribution. We then analyzed the six allozyme loci, whose allelic and genotypic frequencies were significantly divergent in the previous studies. In the first test, we found that the distribution of the two homozygote genotypes of the glucose‐6‐phosphate isomerase‐1 locus (GPI‐1) was significantly different between the hypoxic and the normoxic conditions. During the second test, all individuals were more active in hypoxic conditions, but the two GPI‐1 homozygotes showed a significant difference in time spent performing surface breathing, which was consistent with their distribution observed in the first experiment. These results provide evidence that individual behavioral traits, related to genetic features, may lead to a nonrandom distribution of genotypes in heterogeneous although contiguous microhabitats and, consequently, that habitat choice can play a significant role in driving the micro‐evolutionary dynamics of this species.

Life history of the Mediterranean killifish Aphanius fasciatus in brackish water habitat of Algerian low Sahara

The Mediterranean tooth carp Aphanius fasciatus, a strongly eurythermic and euryhaline species, is an important component of the ichthyofauna in the valley of Oued Righ, situated in Algerian low Sahara. This study aims to understand the life history of this species in arid environment. A total of 1868 individuals (16.7–60.2 mm TL, 0.04–26.8 g TW) were collected between November 2010 and November 2011 from Ayata Lake (Algerian low Sahara). Individual age was determined by scalimetry. Some aspects of the sexual cycle are studied. The population was composed of 6 age classes in both sexes. Back-calculations of total length-at-age were suitably adjusted to von Bertalanffy growth model with: Lt = 80.00 [1−e−0.172 (t + 1.377)] for females and Lt = 77.58 [1−e−0.138(t + 2.236)] for males. The fitted total length-total weight relationship was W = 0.013 L2.848 for females, W = 0.013 L2.809 for males. Spawning occurs once a year between February and July with peak activity in May for Females and in April for Males. Gonado-somatic index reaches a maximum of 4.16% in males and 15.73% in females. The length at the first sexual maturity is 45 mm for both sexes.

Environmental heterogeneity promotes microgeographic genetic divergence in the Mediterranean killifish Aphanius fasciatus (Cyprinodontidae)

Environmental stress can promote evolutionary forces and genetic divergence, but at which microgeographic scale divergence may arise, even in the presence of gene flow, remains poorly known. We studied the effects of eutrophication in a saltwork over a period of 6 years on the gene pool of a local population of the Mediterranean killifish Aphanius fasciatus, a species resistant to extreme conditions. We hypothesised that during eutrophication, the environmental stress may have acted differently along a gradient of salinity and oxygen concentration in promoting evolutionary forces, generating divergence over a small spatial scale (2 km), despite gene flow. We analysed 24 allozymes in three temporal samples each composed of four spatial sub-samples, collected along the gradient, during eutrophication (2003 and 2005) and after a recovery project (2008). The results suggest that eutrophication promoted natural selection, originating a genetic cline on one locus (adenosine deaminase) significantly linked to salinity and oxygen concentration. Together with selection, both genetic drift and gene flow contributed to shaping the genetic structure under stress by further promoting the genetic heterogeneity and giving rise to deficits of heterozygotes as a secondary effect of the divergence. Environmental stress thus increased diversification, with the effects of selection and drift prevailing on gene flow. When environmental stress was relaxed (2008), allele and genotype frequencies became homogeneous, likely because under less extreme conditions the gene flow prevailed. These results improve our understanding of microgeographic divergence, and highlight the role of environmental stress in moulding microevolutionary dynamics and genetic patterns of animal populations.

The influence of habitat structure on energy allocation tactics in an estuarine batch spawner

Trade-off between fecundity and survival was tested in a batch spawner, the Mediterranean killifish Aphanius fasciatus, using an integrated modelling-data approach based on previously collected empirical data. Two sites of the lagoon of Venice (Northern Adriatic sea, Italy) were selected in order to compare the energy allocation between growth and reproduction in two contrasting habitats. These were characterised by high and comparable level of richness in basal resources, but showed two different mortality schedules: an open natural salt marsh, exposed to high level of predation, and a confined artificial site protected from piscivorous predation. By means of a bioenergetic Scope for Growth model, developed and calibrated for the specific goals of this work, we compared the average individual life history between the two habitats. The average individual life history is characterised by a higher number of spawning events and lower per-spawning investment in the confined site exposed to lower predation risk, compared to the site connected with the open lagoon. Thus, model predictions suggest that habitat structure with different extrinsic mortality schedules may shape the life history strategy in modulating the pattern of energy allocation. Model application highlights the central role of energy partitioning through batch spawning, in determining the life history strategy. The particular ovary structure of a batch spawner seems therefore to allow the fish to modulate timing and investment of spawning events, shaping the optimal life history in relation to the environmental conditions.

Lunar-induced reproductive patterns in transitional habitats: Insights from a Mediterranean killifish inhabiting northern Adriatic saltmarshes

Estuaries and coastal lagoons play a key role in the functioning of coastal ecosystems and represent important natural areas for fish communities. Species living in these habitats often show specialised ecological and life history traits. The reproductive periodicity of a northern population of the Mediterranean killifish Aphanius fasciatus was assessed using the male courtship behaviour as indicator of reproductive motivation under laboratory conditions. Timing of egg development was also checked to further support the existence of a tidal-related periodicity. Using a statistical model applied to the intensity of courtship behaviour measured over a two-month period, a semilunar periodicity was revealed, with two main peaks corresponding with full moons. Both behaviours and timing of egg development were consistent with a periodicity of 14 days, suggesting that the tidal cycle drives the reproduction of A. fasciatus in Northern Adriatic saltmarshes. The findings are assessed with a comparative approach within both European and North American transitional waters.

Mitochondrial phylogeography of the killifish Aphanius fasciatus (Teleostei, Cyprinodontidae) reveals highly divergent Mediterranean populations

We studied the genetic structure of the Mediterranean killifish Aphanius fasciatus. Analysis of the sequence variation in a 372-bp portion of the mitochondrial control region in 623 fish from 27 sampling sites along the species’ distributional range (Tyrrhenian coast, Sardinia, Sicily, Adriatic coast, Malta, Tunisia, and Greece) yielded 120 distinct haplotypes. Most of the haplotypes are unique, and only 15 % are shared among different populations. The high F ST value (=0.80) suggests a strong population genetic structuring. The phylogenetic analysis based on Bayesian inference, maximum likelihood and maximum parsimony, and the median-joining network show a sharp separation of the Southeastern Sicilian populations (belonging to the Hyblean region) and of the fluvial Tunisian population of Rio Melah from the others. The Adriatic, the Eastern Sicilian, and the Greek populations are well differentiated, while the group of populations from the Central-Western Mediterranean does not show a clear pattern of differentiation. Our findings indicate that the current genetic structuring of A. fasciatus reflects historical geographical patterns occurring within the Mediterranean basin from the Late Miocene to the Pleistocene. The presence of divergent evolutionary entities in the Hyblean region and the Tunisian Rio Melah supports their inclusion as target areas for the conservation of A. fasciatus.

Linking habitat structure to life history strategy: Insights from a Mediterranean killifish

Modern theories of life history evolution deal with finding links between environmental factors, demographic structure of animal populations and the optimal life history strategy. Small-sized teleost fish, occurring in fragmented populations under contrasting environments, have been widely used as study models to investigate these issues. In the present study, the Mediterranean killifish Aphanius fasciatus was used to investigate the relationships between some habitat features and life history strategy. We selected four sites in the Venice lagoon inhabited by this species, exhibiting different combinations of two factors: overall adult mortality, related to intertidal water coverage and a consequent higher level of predator exposure, and the level of sediment organic matter, as indicator of habitat trophic richness. Results showed that these were the two most important factors influencing demography and life history traits in the four sites. Fish from salt marshes with high predator pressure were smaller and produced a higher number of eggs, whereas bigger fish and a lower reproductive investment were found in the two closed, not tidally influenced habitats. Habitat richness was positively related with population density, but negatively related with growth rate. In particular the synergy between high resources and low predation level was found to be important in shaping peculiar life history traits. Results were discussed in the light of the interactions between selective demographic forces acting differentially on age/size classes, such as predation, and habitat trophic richness that may represent an important energetic constraint on life history traits. The importance to link habitat productivity and morphology to demographic factors for a better understanding of the evolution of life history strategy under contrasting environments was finally suggested.

Heavy metal levels in gonad and liver tissues—effects on the reproductive parameters of natural populations of Aphanius facsiatus

The impact of heavy metal levels (Cd, Cu, and Zn) on the reproductive parameters in natural populations of the Mediterranean killifish Aphanius fasciatus was monitored in four sites of the Tunisian coast. Our study covered a period of the reproductive cycle, from April to November 2009. The significantly higher levels of Cd, Cu, and Zn in the liver and gonads for both sexes (p<0.05) were observed in the most polluted site (Sfax coast). During the reproductive cycle of A. fasciatus, an unbalanced sex ratio in favor to females was observed for all the studied populations. The progression of the hepato- (HSI), gonadosomatic (GSI), and K-factor revealed a spatio-temporal variation between populations during the sampling period. Estradiol-17β amounts in the gonad tissues of females captured from the studied sites showed a significant difference, and the highest concentrations were noticed in the control site (Luza). Taking together, our data provide the first evidence of a physiologically stress and a disturbance of the reproductive status in natural populations of A. fasciatus.

Metallothionein genes in both testes and ovaries showed elevated expression in males and females from the more contaminated areas. In addition, males from these areas showed elevated levels of vitellogenin synthesis in the liver. These results were correlated with heavy metals and polycyclic aromatic hydrocarbon levels in water and sediment samples from the sampling sites. They represent the first evidence of an incipient feminization of marine fish along the coast of Tunisia and confirm the utility of RNA quantification-based markers for monitoring environmental impact on small animals and in species for which very little biological information is available. Evaluation of environmental impact on natural populations of the Mediterranean killifish Aphanius fasciatus by quantitative RNA biomarkers

The Mediterranean killifish, Aphanius fasciatus (Valenciennes, 1821) represents an excellent sentinel species for analysis of environmental impact in coastal areas. Quantitative changes on liver mRNA levels of five stress-related genes – metallothionein (metal exposure), vitellogenin, (estrogenic effects), cytochrome P4501A (CYP1A, dioxin-like compounds), superoxide dismutase (oxidative stress) and HSP70 (general stress) – were tested as markers of exposure to pollutants both in natural populations and in experimental treatments with model effectors. Analysis of fish from a metal-contaminated site in the Tunisian coast (Sfax) indicated that fish presenting spinal deformities (eight times more frequent at this site than in reference sites) showed increased levels of hepatic CYP1A mRNA levels compared to non-deformed fish from the same site or from the reference Louza site, suggesting a combined impact by metals and organic pollutants. Due to the strategy used in their design, these quantitative RNA markers will likely be useful to analyze environmental impacts on other related fish species.

Relationships between relative size of sexual traits and male mating success in the Mediterranean killifish Aphanius fasciatus (Nardo, 1827)

Some relationships between the relative size of secondary sexual traits and male mating success were investigated in the killifish Aphanius fasciatus (Nardo, 1827). The relative ability of a male to compete with another male for access to a ripe female was related to the relative size of male sexual traits by means of a behavioural experiment under controlled laboratory conditions. Furthermore, the covariation in the expression of sexual traits was analysed from a sample of field-collected males. Results of the behavioural experiment showed that male dominance and success were positively related to the relative height of the dorsal fin, but not to the pattern of vertical bars along body sides. Within the analysed male population, males with larger fins tended to have thinner bars, whereas there was no statistically significant correlation with bar number or with bar asymmetry. The results were discussed in terms of advantages conferred by larger dorsal fins in killifish breeding systems, and developmental constraints defining a multiple set of traits correlated to male quality were also hypothesised.

Small-scale morphological and genetic differentiation in the Mediterranean killifish Aphanius fasciatus (Cyprinodontidae) from a coastal brackish-water pond and an adjacent pool in northern Sardinia

Two samples of Aphanius fasciatus collected in the Pilo pond (northern Sardinia, Italy) and in an adjacent pool of small surface area were analysed morphologically (235 individuals) and genetically (a subsample of 58 individuals). The aims of the present study were (i) to test the hypothesis that different predation pressures may be associated with morphological and/or genetic differences between samples from each habitat and (ii) to assess the level of divergence between the two populations. Morphological analysis was based on the relative size of fins because it has been shown to be associated with predation pressure. The relative caudal fin area (caudal fin area/total body surface) was smaller in specimens from the pool, in both males and females, whereas no differences were found for the dorsal and anal fin areas. Caudal fins with higher aspect ratio (fin depth/fin length) were found in fish from the pool but not in the pond, due to a higher fin depth. We hypothesised that specimens from the pool would show smaller caudal fin area, since they are subject to lower predation pressure. Random amplification of polymorphic DNA (RAPD) analysis revealed a relatively high degree of both within- and between-sample genetic heterogeneity. The pond and pool samples exhibited heterozygosities, which did not differ significantly by t-test. Between-sample genetic divergence was highlighted by the coancestry coefficient (θ = 0.301 ± 0.059, P < 0.001) and analysis of molecular variance (AMOVA) (variance between sites = 41%, P < 0.001). Genetic divergence between sites with a relatively high genetic diversity within both samples suggested that the population in the pool did not originate from a single colonisation event with a small number of founders. The genetic divergence between the two populations is consistent with their differences in fin size.
