Evaluation of alpha-glucosidase inhibition activity and glycosides in the Syringa pubescens Turcz from different geographical origin

Inhibition of α-glucosidase is an attractive strategy for the control of hyperglycaemia. Naturally occurring bioactive compounds are potential sources of α-glucosidase inhibitors. The Syringa pubescens Turcz (SP) is commonly used as Chinese folk medicine for treatment of hepatitis and cirrhosis. However, the anti-hyperglycemic activity of SP was lack of study and the report on compounds of α-glucosidase inhibitory activity of SP was also insufficient. In this study, the α-glucosidase inhibitory activity of SP extract was firstly assessed in vitro. The quantification analysis of bioactive glycosides in SP collected at different geographical origins and its correlation with α-glucosidase inhibitory activities were investigated. As the results, six glycosides including salidroside, echinacoisde, forsythoside B, verbascoside, isoacteoside and oleuropein were simultaneously determined using HPLC-DAD method. Moreover, SP collected from different geographical origins exhibited different concentrations of bioactive glycosides and had potential α-glucosidase inhibitory activity. The results of correlation analysis indicated that the amounts of glycosides were positively correlated with the α-glucosidase inhibitory activities of SP. Then, the evaluation of α-glucosidase inhibitory activity of six glycosides was performed. And docking study was conducted to predict the possible binding sites of forsythoside B. Forsythoside B might be the most possible α-glucosidase inhibitor in SP.

Jasmonates promote enhanced production of bioactive caffeoylquinic acid derivative in Eclipta prostrata (L.) L. hairy roots

Eclipta prostrata (L.) L. is widely used in traditional medicine for treatment of hepatitis, poisoning from snake bites and viral infections. Pharmacological studies confirmed its antioxidant, anti-inflammatory and anticancer activities. The efficacy of E. prostrata (L.) L. extracts has been correlated to phenylpropanoids such as flavonoids, coumestans and caffeoylquinic acid derivatives. In this work, the production of wedelolactone, demethylwedelolactone and 3,5-di-O-caffeoylquinic acid (3,5-diCQA) in hairy root cultures of E. prostrata (L.) L. C19 clone was increased after addition of eliciting agents jasmonic acid (JA) or methyl jasmonate (MeJA) at multiple concentrations. Cultures elicited with 100 μM of JA saw a 5.2 fold increase in wedelolactone (from 0.72 to 3.72 mg/g d.w.), a 1.6 fold increase in demethylwedelolactone (from 5.54 to 9.04 mg/g d.w.) and a 2.47 fold increase in 3,5-diCQA (from 18.08 to 44.71 mg/g d.w.). Obtained data validate the potential of E. prostrata (L.) L. hairy root cultures as a production system of wedelolactone, demethylwedelolactone and especially 3,5-diCQA, which has recently been reported to possess activity against coronavirus disease (Covid-19) by in silico computational studies.Supplementary InformationThe online version contains supplementary material available at 10.1007/s11240-021-02201-4.

Optimization of ultrasonic extraction by response surface methodology combined with ultrafast liquid chromatography–ultraviolet method for determination of four iridoids in Gentiana rigescens

Gentiana rigescens is a rich source of iridoids and is commonly used as a folk medicine for treatment of hepatitis and cholecystitis for over 1000 years. A rapid ultrafast liquid chromatography–ultraviolet method was developed for simultaneous determination of four major iridoid glycosides in G. rigescens. Response surface methodology based on the Box–Behnken design was applied to optimize the extraction conditions of iridoid glycosides. Using the Shim-Pack XR-ODS III, four iridoid glycosides were efficiently separated with an acetonitrile:0.1% formic acid aqueous solution gradient at a flow rate of 0.25 mL/min for 8 minutes. All the regression equations revealed a good linear relationship (R2 > 0.9995). The intraday and interday variations were <1.95%. The recoveries ranged from 99.7% to 103.2%. The optimal extraction conditions were as follows: methanol concentration, 82%; the ratio of liquid to solid material, 68:1 (mL/g); and extraction time, 32 minutes. The yield of the four iridoid glycosides under the optimal process was found to be 63.08 mg/g, which was consistent with the predicted yield. In addition, the total content of 50 cultivated samples from Lincang, Yunnan, China, was within the range of 33.6–113.26 mg/g, which provides a more reasonable foundation for utilization of G. rigescens.

Establishment and application of an experimental model of human fetal hepatocytes for investigation of the protective effects of silybin and polyporus umbellalus polysaccharides.

To establish a new experimental model system of human fetal hepatocytes to study the mechanisms underlying the protective effect of silybin and polyporus umbellalus polysaccharides (PSP) on the cellular ultrastructure. Human fetal hepatocytes were obtained from the liver of a human fetus that resulted from a medically necessary induced labor; the mother provided informed consent for sampling, experimental use and publication of findings. The hepatocytes were cultured and then pretreated with silybin or PSP or without either (control), after which the treated cells were exposed to CCl4 for 4 h. Changes in cellular ultrastructure were observed by scanning electron microscopy and transmission electron microscopy, and changes in alanine aminotransferase (ALT), aspartate aminotransferase (AST) and superoxide dismutase (SOD) were assayed. Levels of ALT and AST were significantly decreased, and level of SOD was elevated in the two pretreatment groups following CCl4 exposure, as compared to the control group. The cellular integrity and ultrastructure were well preserved in the two pretreatment groups but were seriously damaged in the control group. The CCl4-induced hepatotoxic cell model system of human fetal hepatocytes is an effective tool for studying the hepatoprotective effect of drugs and may be applicable for studies to screen medicines for treatment of hepatitis.

Application of countercurrent chromatography/thermospray mass spectrometry for the identification of bioactive lignans from plant natural products

The versatility and resolving power of countercurrent chromatography (CCC) has been demonstrated with a newly developed analytical high-speed planet centrifuge system. Interfacing countercurrent chromatography with mass spectrometry (MS) provides a new analytical methodology that integrates the advantages of countercurrent chromatography with the low detection limit and identification capability of mass spectrometry. In this paper the capability of thermospray CCC/MS is demonstrated in identifying and validating the bioactive and structurally known lignans from a crude extract of Schisandra rubriflora Rhed et Wils, a traditional Chinese herbal medicine for treatment of hepatitis.
