The Antibacterial Activity of Rhazya stricta Extracts against Klebsiella pneumoniae Isolated from Some Soil Invertebrates at High Altitudes.

Klebsiella is a common dangerous pathogen for humans and animals and is widely present in the digestive system. The genus Klebsiella is ubiquitous, as it is endemic to surface water, soil, and sewage. In this study, 70 samples were obtained from soil-dwelling invertebrates from September 2021 to March 2022 from Taif and Shafa in different altitudinal regions of Saudi Arabia. Fifteen of these samples were identified as Klebsiella spp. The Klebsiella isolates were genetically identified as Klebsiella pneumoniae using rDNA sequencing. The antimicrobial susceptibility of the Klebsiella isolates was determined. Amplification of virulence genes was performed using PCR. In this study, 16S rDNA sequencing showed a similarity from 98% to 100% with related K. pneumonia from the NCBI database, and the sequences were deposited in the NCBI GenBank under accession numbers ON077036 to ON077050. The growth inhibition properties of ethanolic and methanolic extracts of the medicinal plant Rhazya stricta's leaves against K. pneumoniae strains using the minimum inhibitory concentration (MIC) method and disc diffusion were evaluated. In addition, the biofilm inhibitory potential of these extracts was investigated using crystal violet. HPLC analysis identified 19 components divided into 6 flavonoids, 11 phenolic acids, stilbene (resveratrol), and quinol, and revealed variations in the number of components and their quantities between extracts. Both extracts demonstrated interesting antibacterial properties against K. pneumoniae isolates. The 2 extracts also showed strong biofilm inhibitory activities, with percentages of inhibition extending from 81.5% to 98.7% and from 35.1% to 85.8% for the ethanolic and methanolic extracts, respectively. Rhazya stricta leaf extract revealed powerful antibacterial and antibiofilm activities against K. pneumoniae isolates and could be a good candidate for the treatment or prevention of K. pneumonia-related infections.

In silico screening of some compounds derived from the desert medicinal plant Rhazya stricta for the potential treatment of COVID-19

The latest coronavirus pandemic (SARS-CoV-2) poses an exceptional threat to human health and society worldwide. The coronavirus (SARS-CoV-2) spike (S) protein, which is required for viral–host cell penetration, might be considered a promising and suitable target for treatment. In this study, we utilized the nonalkaloid fraction of the medicinal plant Rhazya stricta to computationally investigate its antiviral activity against SARS-CoV-2. Molecular docking and molecular dynamics simulations were the main tools used to examine the binding interactions of the compounds isolated by HPLC analysis. Ceftazidime was utilized as a reference control, which showed high potency against the SARS-CoV-2 receptor binding domain (RBD) in an in vitro study. The five compounds (CID:1, CID:2, CID:3, CID:4, and CID:5) exhibited remarkable binding affinities (CID:1, − 8.9; CID:2, − 8.7; and CID:3, 4, and 5, − 8.5 kcal/mol) compared to the control compound (− 6.2 kcal/mol). MD simulations over a period of 200 ns further corroborated that certain interactions occurred with the five compounds and the nonalkaloidal compounds retained their positions within the RBD active site. CID:2, CID:4, and CID:5 demonstrated high stability and less variance, while CID:1 and CID:3 were less stable than ceftazidime. The average number of hydrogen bonds formed per timeframe by CID:1, CID:2, CID:3, and CID:5 (0.914, 0.451, 1.566, and 1.755, respectively) were greater than that formed by ceftazidime (0.317). The total binding free energy calculations revealed that the five compounds interacted more strongly within RBD residues (CID:1 = − 68.8, CID:2 = − 71.6, CID:3 = − 74.9, CID:4 = − 75.4, CID:5 = − 60.9 kJ/mol) than ceftazidime (− 34.5 kJ/mol). The drug-like properties of the selected compounds were relatively similar to those of ceftazidime, and the toxicity predictions categorized these compounds into less toxic classes. Structural similarity and functional group analyses suggested that the presence of more H-acceptor atoms, electronegative atoms, acidic oxygen groups, and nitrogen atoms in amide or aromatic groups were common among the compounds with the lowest binding affinities. In conclusion, this in silico work predicts for the first time the potential of using five R. stricta nonalkaloid compounds as a treatment strategy to control SARS-CoV-2 viral entry.

A Review of Rhazya stricta Decne Phytochemistry, Bioactivities, Pharmacological Activities, Toxicity, and Folkloric Medicinal Uses.

The local medicinal plant Rhazya stricta Decne is reviewed for its folkloric medicinal, phytochemical, pharmacological, biological, and toxicological features. R. stricta has been used widely in different cultures for various medical disorders. The phytochemical studies performed on the R. stricta extract revealed many alkaloidal and fatty acid compounds. Moreover, several flavonoid and terpenoid compounds were also detected. Pharmacological activates of R. stricta extracts are approved to possess antimicrobial, antioxidant, anticancer, antidiabetic, and antihypertensive activities. Additionally, R. stricta extract was found to hold biological activates such as larvicidal and phytoremediation activates R. stricta extract was found to be toxic, genotoxic, and mutagenic. R. stricta contains novel phytochemical compounds that have not been investigated pharmacologically. Further research is needed through in vitro and in vivo experiments to pave the road for these compounds for medical, veterinary, and ecological uses.

Biodiversity in Bacterial Phyla Composite in Arid Soils of the Community of Desert Medicinal Plant Rhazya stricta

Rhazya stricta is an important medicinal plant used as native herbal drug to cure various diseases across Asia. Plant-microbe interactions represented by different environment abiotic stress factors that shows effects also on the host plants or on the associated microbial communities. Soil microorganism can be adapted to specific stress conditions and evolve tolerance to this stress. The adjustment of the plant microbiome can productively improve the yield production of essential crops. In the present study, metagenomics analysis of bacterial communities associated with five desert plants (Rhazya stricta, Enneapogon desvauxii, Citrullus colocynthis, Senna italica, and Zygophyllum simplex) was used to highlights the bacterial diversity in the communities of these desert plants, as they also have an importance as folkloric medicinal plants, in addition to their important roles in the ecological and agricultural applications. The results of the bacterial community’s taxonomy showed eleven phyla in each sample, and the most abundant were highlighted for further analysis as it was unassigned phyla. Results indicated that the most commonlyknownphylawere; Actinobacteria, Proteobacteria, Chloroflexi, Firmicutes, Acidobacteria, Gemmatimonadetes, and Bacteroidetes. Further bioinformatics and functional analysis of bacteria are needed to investigate their effectiveness in promoting plant growth and understanding symbiotic relationships under drought-stress conditions, meanwhile, introducing the pinpointed ones in this study will help in achieving this task.

Biotechnology of the medicinal plant Rhazya stricta: a little investigated member of the Apocynaceae family.

Rhazya stricta Decne. (Apocynaceae) is an important medicinal plant that is widely distributed in the Middle East and Indian sub-continent. It produces a large number of terpenoid indole alkaloids (TIAs) some of which possess important pharmacological properties. However, the yields of these compounds are very low. Establishment of a reliable, reproducible and efficient transformation method and induction of hairy roots system is a vital prerequisite for application of biotechnology in order to improve secondary metabolite yields. In the present review, recent biotechnological attempts and advances in TIAs production through transformed hairy root cultures in R. stricta are reviewed to draw the attention to its metabolic engineering potential.

An epidemiological survey of extended-spectrum β-lactamases producing bacteria genotypes and the evaluation of the antimicrobial effect of Rhazya stricta leaf extract

An epidemiological survey and evaluation of the anti-bacterial growth effect of the aqueous leaf extract of the traditional medicinal plant Rhazya stricta were undertaken for extended-spectrum β-lactamase’s producing bacteria isolates collected from the King Abdulaziz Hospital and Oncology Center. Results showed high rates of extended-spectrum β-lactamase (ESBL) isolates (49.2%) compared to non-ESBL isolates (50.8%). The surgical intensive care unit was the hospital site showing the highest rate (15.1%) of infection, followed by the women's medical ward (11.2%). Urine samples were the highest source of infection (40%). Prevalence among Saudis was 66.67% as compared to non-Saudis (33.33%). Prevalence according to gender was 51.94% females and 48.06% males. Age groups 51-60 years, 21-30 years, and 61 years and over showed the highest rates (20.93%, 20.54% and 20.16%, respectively) compared with other age groups. E. coli resistance rates were 94.5% for ceftazidime, 98.18% for cefotaxime, 92.73% for cefepime and 93.75% for aztreonam. On the other hand, in cases of K. pneumoniae resistance, rates were 100% for ceftazidime, 100% for cefotaxime, 98.68% for cefepime and 97.29% for aztreonam. Genetic analysis showed 8 genotypes, 4 of them were for E. coli and 4 for K. pneumoniae. Analysis also showed genetic diversities between the ESBL and the non-ESBL types in their genomic DNA. There was no antimicrobial effect of the plant extract against all study isolates, although the antimicrobial growth had an effect upon some of the control strains.

The effect on plasma glucose, insulin and glucagon levels of treatment of diabetic rats with the medicinal plant Rhazya stricta and with glibenclamide, alone and in combination.

Because many diabetic patients in the United Arab Emirates use medicinal plants as a supplement to treatment with insulin or oral hypoglycaemic agents, the effect on plasma glucose, insulin and glucagon concentrations of simultaneous treatment of streptozotocin-diabetic rats with Rhazya stricta extract and glibenclamide has been examined. Treatment of control rats with the extract at oral doses of 0.5, 2.0 and 4.0 g kg-1 did not significantly affect the concentration of glucose, insulin or glucagon for up to 4 h after administration of the extract. The same doses in diabetic rats reduced the glucose level 1 h (2 and 4 g kg-1) and 2 h (4 g kg-1) after administration of the extract. This was accompanied by significant increases in insulin concentration 1, 2 and 4h after administration of the extract at doses of 2 and 4 g kg-1. Glibenclamide (2.5, 5.0 and 10.0 mg kg-1) dose-dependently reduced glucose and glucagon levels, and increased that of insulin in normal and diabetic rats. Simultaneous treatment of normal and diabetic rats with the plant extract (0.5, 2.0 and 5.0 g kg-1) and glibenclamide (5.0 mg kg-1) significantly exacerbated the effects on glucose, insulin and glucagon induced by the extract or by glibenclamide when given separately. When the plant extract was given at doses of 0.5, 2 and 4 g kg-1 per day for 6 consecutive days the glucose level was reduced by approximately 6, 8 and 30%, respectively. No significant effect was seen on the levels of cholesterol or protein. These results imply that co-administration of the extract with glibenclamide might adversely interfere with glycaemic control in diabetic patients.

Phytochemical and enzyme inhibitory studies on indigenous medicinal plant Rhazya stricta

A new triterpenoid 3α-hydroxy-ursane-5-ene (1) and a known steroid stigma sterol (2) have been isolated for the first time from the fruit of Rhazya stricta. The structures of these compounds were elucidated and identified by spectral studies. The crude ethanolic extract of R. stricta fruit has led to the determination of good results of antibacterial, lipoxygenase and acetylcholinesterase activities. In addition, we also reported the acetylcholinesterase activity of stigma sterol (2) isolated from this plant. Stigma sterol (2) was found to be an acetyl-cholinesterase inhibitor with IC50 at a concentration of 644.0 ± 11.75 µM. Eserine [(−)-physostigmine, IC50 = 61 nM)] was used as a positive control.

The effect of a strongly basic alkaloidal fraction of Rhazya stricta, a traditional medicinal plant, on cytochrome P450-mediated metabolism of theophylline in mice.

The strongly basic alkaloidal fraction of the traditional medicinal plant Rhazya stricta (RS) was given orally to mice, in a single dose of 10 mg/kg (group 1) or, twice daily for 3 days at the same dose (group 2). A third group (control) received normal saline. Liver homogenates from all animals were used to assess the microsomal activity of cytochrome P450 and its isoforms as well as its catalytic activity (using theophylline as a substrate). RS alkaloidal fraction had no significant effect on the total amount of microsomal cytochrome P450, but it caused a significant increase in the cytochrome P450 isoforms CYPs 1A1 and 1A2. It also significantly increased the concentrations of some metabolites of theophylline. These results suggest that RS has the potential to interact with other drugs that are biotransformed by cytochrome P450, when given concomitantly with it.

The effect of treatment with the medicinal plant Rhazya stricta decne on gentamicin nephrotoxicity in rats.

Generation of oxygen free radicals in kidney cortex plays an important role in the pathogenesis of gentamicin (GM) nephrotoxicity, and the leaf extract of the medicinal plant Rhazya stricta has been shown to have an anti-oxidant action in rats. Therefore, in the present work we aimed at testing, in this species, the possible protective effect of R. stricta extract on GM nephrotoxicity. Crude water extract of R. Stricta leaves (0.25, 0.5 and 1 g/Kg) was given orally to rats four days before GM treatment, and thereafter, concomitantly with GM (80 mg/Kg/day) for another six days. Nephrotoxicity was evaluated histopathologically by light microscopy, and biochemically by measuring the concentrations of urea and creatinine in serum, reduced glutathione (GSH), lipid peroxidation and superoxide dismutase (SOD) activity in kidney cortex. The results suggested that the plant extract (0.25 g/Kg) was ineffective in significantly altering the indices of GM-induced nephrotoxicity. However, a dose-related amelioration in the indices of toxicity was noted when the two higher doses of the plant extract were given. The plant extract, at the three doses used, had no significant adverse effect on the body weight of treated rats or on the measured hepatic and renal functions in serum. However, the two higher doses, significantly and dose-dependently increased SOD activity and GSH concentration, and decreased that of lipid peroxides in the kidney cortex. These results suggest that R. stricta water extract may contain compounds that could potentially ameliorate GM nephrotoxicity in rats.

Phytochemical, pharmacological and toxicological properties of the medicinal plant Rhazya stricta Decne. are reviewed. Several types of alkaloids and a few flavonoids have been isolated and their structures and stereochemistry characterized. However, in most cases the biological activity of these compounds has not been studied. Most of the pharmacological activity of the plant resides in its alkaloidal fractions which cause depression of the central nervous system and hypotension. Extracts of R. stricta appear to have low toxicity, although its use in pregnant women may be inadvisable.

Antioxidant action of extract of the traditional medicinal plant Rhazya stricta Decne. in rats.

The effects of a leaf extract of the traditional medicinal plant Rhazya stricta (0.25, 1.0 and 4.0 g/kg/day for 3 days) on reduced glutathione (GSH), lipid peroxidation (LP) and ascorbic acid (AA) concentrations in the liver and kidneys were studied in rats 24 h after the last dose. The plant extract, at a dose of 0.25 g/kg, did not significantly affect the concentrations of GSH, LP or AA in the liver or kidneys. At a dose of 1.0 g/kg, the plant extract significantly increased the GSH concentration in the liver, but did not affect the GSH concentration in the kidneys, or LP or AA in the liver or kidneys. The plant extract (4.0 g/kg) significantly increased the GSH and decreased LP peroxidation, but did not affect the AA concentrations in the liver and kidneys. It may be concluded that the R. stricta extract, at some of the doses used, has antioxidant actions in the rat.

The Effect of Rhazya stricta Decne, a Traditional Medicinal Plant, on Spontaneous and Drug-Induced Alterations in Activity of Rats

The effect of acute and chronic treatment of rats with a lyophilized extract of the leaves of the medicinal plant Rhazya stricta on total and ambulatory activity was studied. Given acutely at single oral doses of 1, 2, 4, and 8 g/kg, the extract produced dose-dependent decreases in total activity and ambulatory activity. Diazepam (20 mg/kg, orally) produced a decrease in rat activity comparable to that produced by a dose of 1 g/kg of the extract. When given daily at an oral dose of 2 g/kg for 21 consecutive days, the extract produced, on the last day of treatment, significant decrease in activity amounting to about 30% of control activity levels. Subcutaneous (SC) treatment of rats with caffeine (7.5, 15, and 30 mg/kg), dose-dependently and significantly increased total activity and ambulatory activity. These effects were dose-dependently attenuated when the extract was given concommittantly with caffeine at oral doses of 1, 2, and 4 mg/kg. Treatment of rats with zoxazolamine alone (10, 20, or 40 mg/kg, SC) or R. Stricta (1 and 4 g/kg orally) alone significantly decreased total and ambulatory activities. Concomittant treatment with zoxazolamine and R. Stricta decreased the rats activity to a greater degree than with either treatment given alone.

The Effect of Rhazya stricta Decne, a Traditional Medicinal Plant, on the Forced Swimming Test in Rats

Immobility induced by forced swimming is well known as an animal model of depression. Using this paradigm, we have, in the present work, tested the possibility that the medicinal plant Rhazya stricta, which has previously been found to affect the monoamine oxidase inhibitory activity in rat brain, may have an antidepressant-like action. Rats were pretreated with various doses (0.025–6.4 g/kg) of the lyophilized extract of the plant leaves, or with desipramine (10, 20, and 40 mg/kg) and were subjected to the forced swimming test. The results indicated that the plant extract produced a biphasic (bell-shaped) effect on the immobility time. The lower doses (0.1, 0.2, and 0.4 g/kg) elicited a highly significant and inversely dose-dependent decrease in immobility time, and the higher doses (0.8, 1.6, and 6.4 g/kg) showed a dose-dependent decrease in immobility time. Under the same experimental conditions desipramine (20 and 40 mg/kg) produced dose-dependent significant decreases in immobility time. Following administration of R. stricta (6.4 g/kg) the immobility time recovered progressively with time, and 4 h after its administration the immobility time was about 70% of the control level (statistically insignificant). It is concluded that R. stricta extract [or component(s) thereof] may possess an antidepressant-like effect.

Stricticine - a new alkaloid from the leaves of rhazya stricta

On reinvestigation of the chemical constituents of a medicinal plant Rhazya stricta , a new alkaloid stricticine (1) has been isolated. The stereochemistry was elucidated with the help of 1H-NMR (homodecoupling, COSY-45°, NOE difference) and 13C-NMR (DEPT) spectra.
