Total oxidant capacity, total antioxidant capacity, ischemic modified albumin, microRNA levels, and their relationship with psoriasis area and severity index.

Aims To examine the differences in the levels of microRNA, ischemic modified albumin (IMA), total oxidant capacity (TOC), and total antioxidant capacity (TAC) of persons with and without psoriasis and, in the case group, the relationship between these parameters and psoriasis area and severity index (PASI). Methods Blood samples were collected from patients and healthy participants to examine levels of these parameters. Results The mean serum TOC level was higher in the case group. The mean serum TAC and IMA levels were significantly lower in the case group (P <0.001). It was observed that the mean serum miR-203 and miR-146a levels were increased in psoriasis patients. It was determined that there was only a significant positive weak correlation between miR-203 and PASI (r = 0.232, P = 0.027). Limitations The small sample size, not controlling serum albumin and not evaluating the effects of the treatment agents used by the patients on oxidative and inflammatory processes. Conclusion In the case group changes in the mean serum TOC and TAC levels provide evidence that oxidative stress may play a critical role in disease pathogenesis. The increase in the mean serum miR-203 and miR-146a levels suggest the possibility of therapies targeting these microRNAs as a new option.

Measurement of oxidative stress and total antioxidant capacity in hyperthyroid patients following treatment with carbimazole and antioxidant

Hyperthyroidism is a common endocrine disorder in which the thyroid produces too many hormones, resulting in the metabolism speed up. The present study was designed to measure oxidative stress and total antioxidant capacity in hyperthyroid patients following treatment with carbimazole and antioxidants supplements. This randomized clinical trial study was conducted to compare Malondialdehyde (MDA) and total antioxidant capacity (TAC) among 25 newly diagnosed hyperthyroid patients (Group A), 25 hyperthyroid patients treated with carbimazole (Group B) and 25 hyperthyroid patients treated with carbimazole and antioxidants supplement (Group C) of both sexes. In this study, the mean serum malondialdehyde (MDA) of the three groups were 4.60 ± 1.08 μmol/L (Group A), 2.79 ± 0.58 μmol/L (Group B), and 1.57 ± 0.29 μmol/L (Group C). We found the mean MDA level was significantly higher in Group A than Group B and Group C. This study found the MDA level was significantly higher in hyperthyroid patients treated with carbimazole alone (Group B, 2.79 ± 0.58 umol/L) than hyperthyroid patients treated with carbimazole and antioxidant combined (Group C, 1.57 ± 0.29 umol/L) among the study groups (p < 0.001). The results showed that the mean serum TAC was significantly lower in newly diagnosed hyperthyroid (Group A, 527.8 ± 78.44 umol/L] patients compared to carbimazole treated alone (Group B, 951.80 ± 99.67 umol/L) and combination with the antithyroid drug (carbimazole) and antioxidant treated (Group C, 1113.56 ± 121.69 umol/L). There was more improvement found in the treatment combined with the antithyroid drug (carbimazole) and antioxidant (Group C).Conventional treatment of hyperthyroid patients significantly reduced oxidative stress and elevated serum TAC but not up to normal level. Therefore, the supplementation of antioxidants could be utilized to improve thyroid function in hyperthyroid patients by boosting antioxidants and restoring oxidant-antioxidant balance. However, further studies are required to determine the optimal dosage, route of administration, and timing of antioxidant therapy needed before this supplementation could be officially recommended as adjuvant therapy.

Evaluation of Salivary and Serum Total Antioxidant Capacity and Lipid Peroxidation in Postmenopausal Women.

Objective In menopause, reduction of estrogen hormone affects oxidative stress process in serum. Oxidative stress in saliva plays a significant role in the pathogenesis of oral diseases. The aim of this study was to investigate the total antioxidant capacity and lipid peroxidation in the serum and saliva of premenopausal and postmenopausal women. Methods In this case control study, 50 postmenopausal women (case group) and 48 premenopausal women (control group) were selected. The unstimulated whole saliva and serum of the postmenopausal and premenopausal women were collected. The total antioxidant capacity (TAC) of the saliva and serum was measured by ferric reducing antioxidant power (FRAP). Also, malondialdehyde (MDA) was measured by thiobarbituric acid reactive substance (TBARS) method for serum and saliva. Then, the obtained data were analyzed by SPSS 17, whereby Mann–Whitney test and Spearman's correlation test were used. P < 0.05 was considered statistically significant. Results The postmenopausal group had significantly lower mean serum TAC and higher mean serum MDA than the control group (P < 0 < 001 and P < 0.01, respectively). The mean salivary TAC and MDA, however, did not differ significantly between the case and control group (P = 0.64 and P = 0.08, respectively). Conclusion In postmenopausal women, with elevation of serum MDA and reduction of serum TAC, the extent of serum oxidative stress grows, but MDA and TAC levels of saliva do not change.

Relationship Between Oxidative Stress Concentration and Postpartum Depression: A Cohort Study

Background: Postpartum depression is a mood disorder that has harmful effects on infants and family relationships. However, its low diagnostic rate (50%) limits the proper treatment of women suffering from this disorder. The identification of women who are at risk can be useful. Oxidative stress may be a risk factor for depression. Oxidative stress occurs when the production of reactive species exceeds the capacity of cellular antioxidant defenses to remove these toxic species. However, little is known about the peripheral oxidative stress in depressive disorder. Objectives: This study aimed to determine the relationship between oxidative stress during pregnancy and postpartum depression. Methods: This prospective cohort study was conducted in the health centers affiliated to Shahid Beheshti University of Medical Sciences from January to May 2017. Eighty pregnant women were assessed for serum total antioxidant capacity and Malondialdehyde levels at 38 - 40 weeks of pregnancy. They were also assessed for postpartum blues (using Maternity Blues Questionnaire of Stein) and postpartum depressive symptoms (using Edinburgh Postnatal Depression Rating Scale, EPDRS) on day 3 and at 4 - 6 weeks after delivery, respectively. The chi-square, Mann-Whitney, and linear regression were used to analyze the relationship between oxidative stress and depressive symptoms. Results: The prevalence of postpartum blues and postpartum depression was 45% and 27%, respectively. The mean serum total antioxidant capacity was significantly lower in the depression group (P < 0.05). There was a significant reverse correlation between serum total antioxidant capacity and EPDRS scores (P < 0.05). Conclusions: The present study demonstrated a relationship between the serum total antioxidant capacity and postpartum depression.

Oxidative stress markers and disease severity among children with Sickle Cell Anaemia

Background: Sickle cell anaemia has been associated with oxidative stress. Total Antioxidant Capacity (TAC), Total Oxidant Status (TOS) and Oxidative Stress Index (OSI) are cumulative markers of oxidative stress. Objective: To evaluate the serum levels of oxidative stress markers in children with sickle cell anaemia (SCA) and determine the relationship between these markers and disease severity. Method: One hundred and fifty-six children, comprising 78 with SCA, aged 1 - 15 years and 78 age- and sex-matched Haemoglobin AA controls were studied. Serum TOS, OSI, and TAC were determined using ELISA kits. The severity of the SCA was determined using clinical and laboratory parameters. Result: Children with SCA had lower mean serum TAC (0.83±0.31UAE) than controls (1.19±0.24UAE) with p&lt;0.001. However, the mean serum TOS and OSI of children with SCA was higher than among the controls (13.33±4.64U/ml vs. 9.70±2.72U/ml and 20.95±16.75 vs. 8.68±3.76 respectively) with p&lt;0.001. SCA subjects with mild disease had higher mean serum TAC (0.91 ± 0.27UAE) than those with moderate disease (0.54±0.27UAE) (p&lt;0.001). On the other hand, the mean TOS and OSI were lower in children with mild disease compared to those with moderate disease (12.64±4.32U/ml vs. 15.63±5.07U/ml, p = 0.016 and 16.26±10.25 vs. 36.61±23.89 p&lt;0.001 respectively). Sickle cell disease severity score had negative correlation with TAC (r = -0.60, p &lt; 0.001) but positive correlation with TOS (r = 0.3, p = 0.008) and OSI (r = 0.6, p &lt; 0.001). Conclusion: Children with SCA had lower TAC but higher TOS and OSI than matched controls. Oxidative stress markers had a significant relationship with SCD severity.

Effects of Chicory Leaf Extract on Serum Oxidative Stress Markers, Lipid Profile and Periodontal Status in Patients With Chronic Periodontitis

ABSTRACTAim: The aim of present study was to evaluate the effects of chicory leaf extract on serum oxidative stress markers, lipid profile, and periodontal status in patients with chronic periodontitis.Methods: In this double-blind, randomized controlled clinical trial 40 patients with chronic periodontitis were allocated to intervention and control groups. The intervention group received a 1-gram chicory leaf methanolic extract capsule twice daily for 8 weeks. In the control group, participants received a placebo capsule (containing 1 gram wheat flour) twice daily for 8 weeks. All participants had nonsurgical periodontal therapy during the study. Anthropometric measurements, dietary intake, total antioxidant capacity (TAC), malondialdehyde (MDA), uric acid, lipid profile (total cholesterol [TC], triglycerides [TG], low-density lipoprotein cholesterol [LDL-C], and high-density lipoprotein cholesterol [HDL-C]), and pocket depth (PD) were assessed before and after intervention.Results: The results showed that mean serum TAC, uric acid, and HDL-C increased and mean serum MDA, TG, LDL-C, and TC decreased significantly in the intervention group compared to their baseline and the control group post-intervention. A significant difference was observed in mean PD between the two groups.Conclusion: Chicory leaf extract as an adjunct nutritional approach with nonsurgical periodontal therapy may be helpful in controlling periodontal status.

Effects of Elaeagnus angustifolia L. powder supplementation on serum total antioxidant capacity and malondialdehyde levels in females with knee osteoarthritis

AimThe present study aimed to determine the effects of Elaeagnus angustifolia L. supplement on serum antioxidant status in females with knee osteoarthritis. MethodsIn this 8-week randomized clinical trial, 90 females with knee osteoarthritis were randomly assigned to one of three groups—medulla powder (15g medulla powder), whole fruit powder (15g whole fruit powder) or placebo. Information on dietary intake was collected using 24-h recall method for 3 days (one weekend day included). The serum levels of total antioxidant capacity (TAC) and malondialdehyde (MDA) were measured using the Randox® assay and the fluorometry method respectively. Paired sample t-test, ANOVA and ANCOVA were used for statistical analysis. ResultsThe mean serum TAC level in the medulla, whole fruit and placebo groups increased from 1.12±0.34 to 1.28±0.27, 1.30±0.28 to 1.50±0.28 and 1.18±0.19 to 1.30±0.22mmol/L respectively. In terms of the TAC level, the significant increase was only observed in the whole fruit group (P=0.001). The mean serum MDA level in the medulla, whole fruit and placebo groups decreased from 3.80±0.33 to 3.30±0.41, 3.25±0.51 to 2.45±0.43 and 3.38±0.63 to 2.56±0.41, respectively. A statistically significant reduction was observed only in the whole fruit group (P=0.009). DiscussionBased on the results, E. angustifolia L. in the form of whole fruit had beneficial effects on serum TAC and MDA levels in females with knee OA. This may show a possible disease modifying effect of E. angustifolia L. in knee osteoarthritis.

Effects of &lt;i&gt;Ganoderma lucidum&lt;/i&gt; Spent Mushroom Substrate Extract on Milk and Serum Immunoglobulin Levels and Serum Antioxidant Capacity of Dairy Cows

Purpose: To investigate the effects of Ganoderma lucidum hot water extract (HWE) from spent mushroom substrate on milk and serum immunoglobulin levels and serum antioxidant capacity.Methods: Forty cows within the same parity and stage of lactation and with similar body weight were randomly divided into four groups of 10 cows each. The daily dietary dosage of HWE was 0, 33, 67, and 100 g for control and the three experimental groups EG1, EG2, and EG3, respectively. Serum antioxidant activity as well as milk and serum immunoglobulin levels were measured by enzyme-linked immunosorbent assay (ELISA).Results: After administration for 60 days, no significant differences in milk IgG, IgA, or IgM concentrations were detected among the experimental groups, but serum IgA concentration was significantly higher in EG1, EG2, and EG3 groups compared with control group (p &lt; 0.01). Additionally, significant differences were detected in serum total antioxidant capacity (TAC) and mean serum TAC concentration among control, EG1, EG2, and EG3 groups (p &lt; 0.05).Conclusion: The results suggest that when utilized as a feed additive, HWE may enhance immunity and antioxidant capacity in dairy cows, and subsequently improve milk quality.Keywords: Ganoderma lucidum, Spent mushroom substrate, Immunoglobulin, Antioxidant capacity, Dairy cows

Serum total antioxidant capacity in oral carcinoma patients.

Context:Oxidative stress is a major determinant in carcinogenesis and serum total antioxidant capacity (TAC) indirectly reflects the level of oxidative stress. Although oral cancer is the third most frequent cancer in Indian population, there are no standard noninvasive marker for early detection and monitoring therapeutic response in oral cancer patients.Aims:This study was carried out to investigate implications of serum TAC in oral cancer by evaluating pre- and post-operative levels in oral cancer patients.Settings and Designs:It was a prospective single blinded study.Subjects and Methods:Serum TAC was measured using ferric-reducing antioxidant power assay. Pre- and post-operative values were estimated and compared in 30 oral cancer patients who underwent surgery.Statistical Analysis Used:Paired t-test was used to compare pre- and post-operative values.Results:Compared to the normal value, both pre- and post-operative serum TAC were significantly low in oral cancer patients. However, there was no statistically significant difference between pre- and postoperative levels.Conclusions:In this study, low mean serum TAC was detected in oral cancer patients. However, its diagnostic and prognostic significance in oral cancer needs further investigation.

Serum Thyroid Stimulating Hormone and Antioxidant Status in Patients with Suspected Thyroid Dysfunction

The incidence of thyroid disorders is reported in significant numbers globally. Patients referred for thyroid function tests to the Nuclear Medicine unit, University of Peradeniya, Sri Lanka were assessed for antioxidant status in addition to routine investigations. Among the patients investigated 41.9% had enlarged thyroid glands while others (58.1%) did not show any thyroid enlargement. Majority (79.5%) of the patients with enlarged thyroid, had multi nodular goiter, while only 12.8% had diffused enlargement and 7.7% had solitary thyroid nodules. Ninety three subjects formed the study group and their mean Thyroid stimulating hormone (TSH) concentration was 5.4 ± 12.3 mU/L with a range from 0.1 to 58.0 mU/L. Their mean serum Total antioxidant capacity (TAC) was 719.1 ± 164.5 µmol/L and mean serum Protein thiol concentration was 517.7 ± 89.8 µmol/L. Age had a significant positive correlation with TAC (r= 0.268, P= 0.009), and a significant negative correlation with protein thiol (r= -0.481, P < 0.001). BMI of the patients increased significantly with age. Among the patients, 9 (9.7%) were hyperthyroid, 68 (73.1%) were euthyroid while 16 (17.2%) were hypothyroid. There was a significant positive correlation between TSH and TAC (r=0.526, P=0.036) in hypothyroid patients which had not been reported previously. The mean serum TAC in males (928.7 ± 60.4 µmol/L) was significantly higher than that of females (699.4 ± 150.5 µmol/L) (P =0.006). Similarly, mean serum protein thiol concentration of males (570.7 ± 23.6 µmol/L) was significantly higher than that of females (512.7 ± 90.0 µmol/L) (P=0.049). No significant differences in TAC and protein thiol concentration were evident among euthyroid, hyperthyroid and hypothyroid patients which suggest that the antioxidant status does not differ among the different categories of patients investigated.

Oxidative Stress in Alopecia Areata: A Case–Control Study

Increased reactive oxygen species (ROS) and lipid peroxidation are seen in many dermatologic disorders, including atopic dermatitis, psoriasis, vitiligo, acne vulgaris, pemphigus vulgaris, and lichen planus. In alopecia areata (AA), there is increased production of ROS from perifollicular inflammatory cells. The aim of this study was to determine the oxidative stress index (OSI) and lipid peroxidation by studying serum total oxidant capacity (TOC), total antioxidant capacity (TAC), and malondialdehyde (MDA) values in AA patients. The study included 35 AA patients and a control group consisting of 30 age- and sex-matched healthy volunteers. The serum TOC, TAC, and MDA values were measured, and the OSIs were calculated and compared in both groups. The mean serum TOC (p<0.001), MDA (p<0.001), and OSI (p<0.001) values were found to be significantly higher in AA patients than in the control group. The mean serum TAC value was significantly lower (p<0.05) in cases than in controls. Significantly higher MDA (p<0.001), TOC (p<0.001), and OSI values (p<0.001) and lower TAC values (p<0.01) were found in severe AA than in mild or moderate AA. The demonstrated results confirmed the presence of oxidative stress and lipid peroxidation in AA. Whether these changes play a role in disease pathogenesis or result from the inflammatory process requires further investigation.

Serum total antioxidant capacity in patients with multiple sclerosis

Multiple sclerosis (MS) is a chronic inflammatory disease of the central nervous system (CNS). It is characterized by loss of myelin, the fatty tissue that surrounds and protects nerve fibres allowing them to conduct electrical impulses. Recent data indicate that oxidative stress (OS) plays a major role in the pathogenesis of multiple sclerosis (MS). The aim of this study was to estimate level of serum total antioxidative capacity in patients with multiple sclerosis. Our cross-sectional study included 33 patients with MS and 24 age and sex matched control subjects. All our patients had a Poser criteria for definite diagnostic categories of multiple sclerosis. Serum total antioxidant capacity (TAC) was measured by quantitative colorimetric determination, using Total antioxidant Capacity-QuantiCromAntioxidant Assay Kit (BioAssay systems, USA; DTAC-100). Mean serum TAC in multiple sclerosis group of patients was 119.2 mM Trolox equivalents and was significantly lower (p<0.001) compared to the control group of subjects (167.1 mM Trolox equivalents). Our results showed that oxidative stress plays an important role in pathogenesis of multiple sclerosis. This finding, also, suggests the importance of antioxidants in diet and therapy of MS patients.
