The effects of melatonin on prolactin and growth hormone secretion in ewes under different photoperiods, during the early post partum period

The effects of melatonin on the secretion of prolactin (PRL) and growth hormone (GH) were studied in ewes (Polish Longwool, n = 20) nursing lambs, under different day length (photoperiod) regimes. The animals were divided into four groups: (i) long day control group (LDC, n = 5); (ii) long day melatonin-treated (LDM, n = 5) group, which had lambed in March (spring); (iii) short day control group (SDC, n = 5) and (iv) short day melatonin-treated (SDM, n = 5) group, which had lambed in November (autumn). Blood samples were collected from all ewes 5 days after lambing. Four blood collections were performed at 10-day intervals, over a 40-day time period. Sampling started at sunset and continued for 6 h at 20 min intervals. Subcutaneous melatonin implants were inserted in ewes of the LDM and SDM groups after first blood collection. The plasma concentrations of PRL and GH were assayed in samples using RIA. In ewes from the LDC group, the mean plasma PRL concentration increased gradually, reaching a significantly ( P < 0.001) higher level, after 3 weeks. In contrast, in the LDM group, the PRL concentration decreased significantly ( P < 0.001) after 10 days. The mean plasma GH concentration was significantly ( P < 0.001) higher in the LDC than in the LDM group, for the entire experimental period. However, a gradual decrease in the concentration was recorded in both groups. In the SDC and SDM groups, the plasma PRL concentrations did not decrease significantly 2 weeks after the onset of the trial, and did not differ significantly in these groups. The mean plasma GH concentration increased significantly ( P < 0.001) in the SDM group, compared to the SDC group, after the third week. The mean plasma GH concentration in the SDM and the SDC group reached a similar level by the end of the trial. It would appear that melatonin may effectively inhibit PRL secretion in ewes during the early post partum period under long photoperiod, and stimulate GH release in such ewes under short photoperiod. The inhibition of PRL secretion in nursing ewes during increasing photoperiod (long days) occurred, despite the strong stimulation of suckling. It can therefore be assumed that melatonin via decreasing PRL concentrations, may be used to shorten the nursing period in early-lactating ewes.

Opioid-salsolinol relationship in the control of prolactin release during lactation

Endogenous opioid peptides (EOP) and dopamine (DA)-derived salsolinol are implicated in the suckling-induced prolactin surge. The aim of this study was to investigate the relationship between the opioidergic and salsolinergic activity in the mediobasal hypothalamus of nursing sheep. The sheep were infused intracerebroventricularly with opioid receptors antagonists: naloxone (all types of receptors, n=6); naloxonazine (μ receptor, n=6) or the vehicle (control, n=6) in a series of five 30-min infusions (60 μg/60 μl) from 10:00 to 15:00, at 30-min intervals. The period of the experiment included the non-suckling (10:00–12:30) and suckling (12:30–15:00) periods. Simultaneously, a push–pull perfusion of the infundibular nucleus/median eminence was performed in every sheep to study the dopaminergic system activity. Blood samples were also collected at 10-minute intervals to determine plasma prolactin concentration. Both the mean perfusate salsolinol and plasma prolactin concentrations were higher during the suckling vs. non-suckling ( P<0.001) period in the control. The perfusate DA concentration was below the detection limit in this group. Treatment with either naloxone or naloxonazine significantly ( P<0.01) diminished plasma prolactin concentration, as compared with the controls and blocked the prolactin surge during suckling. In drug-infused sheep, the perfusate salsolinol concentration was below the detection limit but the increased DA and its metabolite 3,4-dihydroxyphenylacetic acid concentrations were observed. In conclusion, the stimulatory action of EOP on prolactin secretion in nursing females is mediated, at least in part, by salsolinol, and the ligands for μ opioid receptor may be the primary factors of this relationship, especially with respect to the suckling-induced prolactin surge.

Cystic ovaries in women affected with hereditary angioedema.

Polycystic ovary (PCO) syndrome is biochemically characterized by abnormal gonadotropin secretion and polycystic ovaries associated with increase in size and functional activity of stromal tissue; multifollicular ovaries (MFO) are defined by the presence of multiple cysts with no increase in stromal tissue. A central (hypothalamic-pituitary) abnormality, including high plasma beta-endorphin (BE) concentrations without simultaneous elevation of ACTH, was reported for subjects with PCO syndrome. Since we have found the presence of high plasma BE concentrations in hereditary angioedema (HANE) during attacks as well as during symptom-free periods, we studied, by means of pelvic ultrasound scanning employed to determine the prevalence of PCO and of MFO, 13 women of reproductive age affected with HANE who were not on oral contraceptives. We have found PCO in 5/13 (38.4%) and MFO in 7/13 (53.8%) HANE patients. Nine patients had oligomenorrhoea (five with PCO, three with MFO, one with normal ovaries), five (three with PCO, two with MFO) were hirsute and only one (with MFO) had weight loss. No patient was obese. Mean plasma LH, testosterone, prolactin, cortisol and ACTH concentrations were normal, while FSH was significantly reduced and LH/FSH ratio increased. BE concentrations were significantly high in all the patients studied. Our results clearly demonstrate that women with HANE frequently have cystic ovaries (polycystic or multifollicular) in the presence of high BE concentrations.

Identification of salsolinol in the mediobasal hypothalamus of lactating ewes and its relation to suckling-induced prolactin and GH release

The push-pull perfusions of the infundibular nucleus-median eminence (IN/ME) were made in lactating ewes (n=7) twice, to identify dopamine (DA)-derived salsolinol and the changes in its extracellular concentration in response to suckling. The perfusate collecting period in every ewe consisted of control non-suckling period, 1000-1230 h (five perfusates), and suckling period, 1230-1500 h (next five perfusates). Simultaneously, blood samples were collected from 1000 to 1500 h at 10-min intervals. The perfusate concentrations of salsolinol and DA were measured by HPLC, and plasma prolactin and GH concentrations were assayed by the RIA. Mean concentrations of salsolinol in perfusates collected from the anterior and posterior parts of the IN/ME (according to post-mortem localization of a perfusion site) increased significantly (P<0.05 and P<0.001 respectively) during the suckling period, when compared with those noted during the non-suckling period. While no DA was found in the anterior part, only vestigial amounts of DA were found in a few perfusates collected from the posterior part. Salsolinol was not detected in the IN/ME of ewes 10 weeks after weaning (seasonal anoestrus). Mean plasma prolactin and GH concentrations during suckling were significantly (P<0.001) higher than those noted during the non-suckling period. In conclusion, our current study reveals that salsolinol is present in the IN/ME of lactating ewes and that its extracellular concentration increases during suckling. Moreover, it supports the role of salsolinol as a neurotransmitter involved in the regulatory process of prolactin secretion at least during lactation.

Seasonal variation and circadian rhythmicity of the prolactin profile during the summer months in repeat-breeding Murrah buffalo heifers

The present study was undertaken to determine a detailed endocrine profile for prolactin and progesterone during the oestrous cycle in repeat-breeding Murrah buffalo heifers during summer and winter. Hormone concentrations were quantified in blood plasma samples collected over the oestrous cycle in both winter and summer, as well as in samples collected during the summer months to observe circadian rhythmicity. The mean plasma prolactin concentration during the winter months ranged from 3.10 +/- 0.48 to 9.17 +/- 1.39 ng mL(-1); during the summer months, plasma prolactin concentrations ranged from 248.50 +/- 16.03 to 369.63 +/- 25.13 ng mL(-1). During the winter months, the mean plasma progesterone concentration ranged from 0.20 +/- 0.00 to 3.04 +/- 0.34 ng mL(-1), which was significantly higher (P < 0.01) than the prolactin concentrations recorded in the summer months (range 0.20 +/- 0.00 to 1.48 +/- 0.13 ng mL(-1)). Plasma prolactin and progesterone concentrations were negatively correlated (r = -0.24) during the summer oestrous cycle, which indicates prolactin-induced suppression of progesterone secretion through poor luteal development. During the summer, a circadian rhythmicity was observed in buffaloes and the results indicate that high prolactin secretion contributes to poor fertility by lowering gonadal hormone (progesterone) secretion. It was concluded from the present study that prolactin and progesterone profiles during the summer and winter months are directly correlated with the reproductive performance of buffaloes. The finding also validates the hypothesis that hyperprolactinaemia may cause acyclicity/infertility in buffaloes during the summer months due to severe heat stress.

Development and validation of a highly sensitive economic enzymeimmunoassay for prolactin determination in blood plasma of mithun ( Bos frontalis) and its application during milk let down and cyclicity

The objective of the present study was to develop and validate highly sensitive and economic enzymeimmunoassay (EIA) for prolactin determination in mithun blood plasma on microtitreplates using the biotin–streptavidin amplification system and second antibody coating technique and to apply this procedure during milk let down and cyclicity in mithuns ( Bos frontalis), a semi-wild ruminant. Biotin was coupled to prolactin and used to bridge between streptavidin peroxidase and immobilized antiserum in competitive assay. The EIA was carried out directly in 50 μl mithun plasma. The sensitivity of the EIA procedure was 0.1 ng/ml plasma. Plasma volumes viz., 12.5, 25 and 50 μl did not influence much the shape of standard curve though a slight drop in the OD 450 was seen with higher plasma volumes. A parallelism test was carried out to compare the endogenous mithun plasma prolactin with bovine prolactin standards used in the assay. It showed good parallelism with the bovine standard curve. Plasma prolactin was estimated in six cyclic mithun cows during an estrous cycle. Mean plasma prolactin concentrations around the day of estrus were recorded to be higher than any other day of the cycle. Prolactin profiles were also obtained in three mithuns before, during and after milking. A sharp release of prolactin shortly after udder stimulation was observed. High levels of prolactin were maintained during milking, falling sharply thereafter. In conclusion, the EIA developed for prolactin determination in mithun blood plasma is sufficiently reliable, economic and sensitive enough to estimate prolactin in all physiological variation in mithun.

Differential regulation of suppressor of cytokine signaling-3 in the liver and adipose tissue of the sheep fetus in late gestation

It is unknown whether the JAK/STAT/suppressor of cytokine signaling-3 (SOCS-3) intracellular signaling pathway plays a role in tissue growth and metabolism during fetal life. We investigated whether there is a differential profile of SOCS-3 expression in the liver and perirenal adipose tissue during the period of increased fetal growth in late gestation and the impact of fetal growth restriction on SOCS-3 expression in the fetal liver. We also determined whether basal SOCS-3 expression in the fetal liver and perirenal adipose tissue is regulated by endogenous fetal prolactin (PRL). SOCS-3 mRNA abundance was higher in the liver than in the pancreas, spleen, and kidney of the sheep fetus during late gestation. In the liver, SOCS-3 mRNA expression was increased (P < 0.05) between 125 (n = 4) and 145 days (n = 7) gestation and lower (P < 0.05) in growth-restricted compared with normally grown fetal sheep in late gestation. The relative expression of SOCS-3 mRNA in the fetal liver was directly related to the mean plasma PRL concentrations during a 48-h infusion of either a dopaminergic agonist, bromocriptine (n = 7), or saline (n = 5), such that SOCS-3 mRNA expression was lower when plasma PRL concentrations decreased below approximately 20 ng/ml [y = 0.99 - (2.47/x) + (4.96/x(2)); r(2) = 0.91, P < 0.0001, n = 12]. No relationship was shown between the abundance of phospho-STAT5 in the fetal liver and circulating PRL. SOCS-3 expression in perirenal adipose tissue decreased (P < 0001) between 90-91 (n = 6) and 140-145 days (n = 9) gestation and was not related to endogenous PRL concentrations. Thus SOCS-3 is differentially expressed and regulated in key fetal tissues and may play an important and tissue-specific role in the regulation of cellular proliferation and differentiation before birth.

Application of sensitive enzymeimmunoassays for oxytocin and prolactin determination in blood plasma of yaks ( Poephagus grunniens L.) during milk let down and cyclicity

Highly sensitive and specific enzymeimmunoassays for oxytocin and prolactin determination in yak plasma using the biotin–streptavidin amplification system and the second antibody coating technique were validated and applied for determining their profiles during milk let down and cyclicity in yaks. Oxytocin EIA was conducted taking duplicate 200 μl of unknown plasma samples and standards per well. The lowest detection limit was 0.2 pg/well, which corresponded to 1 pg/ml plasma. Prolactin EIA was carried out directly in 50 μl of yak plasma. The sensitivity of EIA procedure was 5 pg/well prolactin, which corresponded to 0.1 ng/ml plasma. Mean plasma prolactin concentrations although high at estrus were not statistically different ( P > 0.05) from the hormone concentrations on other days. Mean plasma prolactin concentrations during non-breeding season were significantly higher ( P < 0.001) than that recorded in breeding season. Oxytocin and prolactin profiles were also obtained in two yaks before, during and after milking. A sharp release of oxytocin and prolactin shortly after udder stimulation was observed. High levels of oxytocin and prolactin were maintained during milking, falling sharply thereafter.

Circadian variations in plasma concentrations of melatonin and prolactin during breeding and non-breeding seasons in yak ( Poephagus grunniens L.)

Circadian variations of plasma melatonin and prolactin concentrations were determined during breeding as well as non-breeding seasons in yak. Blood samples (5 ml) were collected during different phases of estrous cycle, viz. early (0–6 days), mid (7–12 days) and late luteal (13–19 days) at 2 h interval for 24 h from eight yaks during one breeding month (November); the same yaks were bled at 2 h interval during one non-breeding month (February) for 24 h. Plasma melatonin concentrations rose sharply ( P < 0.01) after sunset to record peak concentrations between midnight and 2 a.m. declining sharply thereafter in both breeding as well as non-breeding seasons. Basal melatonin concentrations were recorded between 0600 and 1600 h. Stage of luteal phase did not influence the diurnal hormone change ( P < 0.01). In the breeding season, mean plasma prolactin concentrations displayed circadian variations with maximum value at 0400 h (41.22 ± 1.5 ng/ml) and minimum at 1400 h (12.0 ± 4.02 ng/ml). In the non-breeding season plasma prolactin concentrations showed circadian variation with maximum value at 0000 h (59.9 ± 10.5 ng/ml) and minimum at 1200 h (32.13 ± 3.2 ng/ml). A positive correlation in breeding ( r = 0.75) and in non-breeding season ( r = 0.65) between circadian changes in mean plasma prolactin and melatonin concentrations were seen. Circadian changes of mean plasma melatonin concentrations during breeding and non-breeding seasons were not different ( P > 0.05). However, mean plasma prolactin concentrations were found to be higher ( P < 0.01) in the non-breeding season. Three conclusions were drawn from the study: (i) melatonin and prolactin concentrations followed a circadian pattern of secretion (ii) melatonin and prolactin secretion may be closely interrelated and (iii) higher prolactin concentrations during the non-breeding season could be due to nutritional and environmental stress and hence might be contributing to lack of cyclicity.

Genistein, a Phytoestrogen, Effectively Modulates Luteinizing Hormone and Prolactin Secretion in Ovariectomized Ewes during Seasonal Anestrus

Through binding with estrogen receptors, phytoestrogens, plant-derived estrogen-like compounds, affect numerous reproductive functions. It is not known whether these compounds are capable of evoking effective changes in luteinizing hormone (LH) and prolactin (PRL) secretion in ewes by acting directly within the central nervous system (CNS). The hypothesis studied was that genistein, infused for several hours into the third ventricle, could immediately affect LH and PRL secretion in ovariectomized (OVX) ewes during seasonal anestrus. Two doses of genistein, 1 µg/100 µl/h (total 4 µg, n = 7) and 10 µg/100 µl/h (total 40 µg, n = 7), were infused intracerebroventricularly from 12.00 to 16.00 h and blood samples were collected from 8.00 to 20.00 h at 10-min intervals. Randomly selected ewes were infused with a vehicle (control, n = 5). The mean plasma LH concentration in control ewes was significantly (p < 0.01) higher during infusion of the vehicle than before the infusion. It remained on an insignificantly changed level after the infusion. The frequency of LH pulses in control ewes did not differ significantly before, during, or after vehicle infusion. In ewes infused with a lower dose of genistein, plasma LH concentrations decreased significantly (p < 0.001) after the infusion, as compared with the values noted before and during genistein infusion. Only a tendency towards a decrease in LH pulse frequency occurred after infusion of a lower dose of genistein. In ewes infused with a higher dose of genistein, the plasma LH concentration decreased significantly (p < 0.01) after phytoestrogen administration as compared with the values noted before and during infusion. The frequency of LH pulses was also significantly (p < 0.01) lower after genistein administration. Because the changes in PRL secretion were more dynamic in response to genistein infusion, the statistical analysis included 2-hour periods. The mean plasma PRL concentration in control animals was significantly enhanced (p < 0.01) only during the first 2-hour period of sampling. After that it decreased and remained on an unchanged level up to the end of sampling. Similar changes in PRL secretion were observed in both experimental groups before genistein infusion. In contrast, significant (p < 0.01 to p < 0.001) increases in PRL concentration were noted regularly during and shortly after the genistein infusion in either low-dose or high-dose genistein-infused ewes, compared with the concentrations noted before genistein treatment. Plasma PRL concentrations during and after genistein infusion in both experimental groups were also significantly higher than the control (p < 0.01 to p < 0.001). The presented data demonstrate that genistein, a phytoestrogen, may effectively modulate LH and PRL secretion in OVX ewes by acting within the CNS.

Low doses of bromocriptine shorten the interestrous interval in the bitch without lowering plasma prolactin concentration

In order to investigate the effect of different doses of bromocriptine on plasma prolactin concentration and the interestrous interval, beagle bitches were treated twice daily with 5 μg (5-group), 20 μg (20-group), or 50 μg (50-group) bromocriptine per kg body weight orally, starting 28 days after ovulation. In the 5-group, the difference between the mean plasma prolactin concentration before and that during bromocriptine treatment was not significant. In contrast, mean plasma prolactin concentration decreased significantly after the start of bromocriptine treatment in the 20- and 50-groups. The mean interestrous interval was significantly shorter in all three groups than in untreated bitches in the same colony. The mean interestrous interval in the 20-group and that in the 50-group were similar, but both were significantly shorter than that in the 5-group. The results of this study indicate that bromocriptine shortens the interestrous interval in the bitch even when the dose is so low that it does not lower plasma prolactin concentration. Induction of estrus in the bitch by bromocriptine therefore involves a mechanism other than via the lowering of plasma prolactin concentration. Furthermore, this study shows that the extent of shortening of the interestrous interval by bromocriptine is dose dependent.

Short-term modulation of prolactin secretion by melatonin in anestrous ewes following dopamine- and opiate receptor blockade.

This study tested the hypothesis that the dopaminergic and opioidergic systems are not involved in the short-term stimulatory action of melatonin (MLT) on the secretion of prolactin in anestrous ewes. Thus, MLT should stimulate prolactin release after blockade of either dopamine (DA) or opiate receptors with specific antagonists at the level of the pituitary gland and central nervous system (CNS), respectively. During afternoon intracerebroventricular (icv.) infusion of MLT, the mean plasma prolactin concentration increased significantly (P < 0.001) as compared with the concentrations noted before and during the infusion of the vehicle (veh.). As a result of subcutaneous (s.c.) injection of sulpiride (SULP, DA antagonist), an increase in plasma prolactin concentration was observed, followed by a gradual decrease during the icv. infusion of the vehicle. MLT infused icv. significantly increased (P < 0.001) the secretion of prolactin in SULP + MLT-treated ewes, as compared with the concentration of prolactin noted during infusion of the vehicle in SULP + veh.-treated ewes. Naloxone (NAL, opioid antagonist) infused icv. did not significantly affect the secretion of prolactin, however, a significant (P < 0.01) increase in the concentration was observed after the infusion. In MLT + NAL-treated ewes, the plasma prolactin concentration increased significantly (P < 0.001) during the infusion, as compared with the concentration noted before and that in NAL-alone infused ewes. These results demonstrate that melatonin stimulates prolactin release after the pharmacological exclusion of the dopaminergic input with the DA antagonist sulpiride and also despite the presence of DA activity in the hypothalamus after NAL treatment. Secondly, endogenous opioid peptides are not a major component of this melatonin action.

The rôles of photoperiod and nutrition in the seasonal increases in growth and insulin-like growth factor-1 secretion in male red deer

AbstractYoung male red deer follow a seasonal growth pattern that can be shifted by altering the photoperiod they experience. An increase in photoperiod to 16 h of light per day (16L : 8D) during winter advances the onset of rapid growth and high food intake that normally commences in spring. These changes are associated with increased growth hormone (GH) and insulin-like growth factor-1 (IGF-1) secretion. The GH/IGF-1 axis is acutely sensitive to the level of nutrition and the relative rôles of photoperiod and nutrition in determining the spring IGF-1 rise is unknown. The present experiment set out to examine this by exposing two groups of deer (no. = 8 per group) to a photoperiod shift during their 1st year of life (16L : 8D from 2 June), designed to cause accelerated growth and increased food intake after approximately 7 weeks. However, after 6 weeks the food intake (pellets containing 11 MJ metabolizable energy and 160 g crude protein per kg dry matter (DM)) of one group (LDRES) was clamped, thereby preventing the intake component of the response. The intake of the other group (LDAL) remained ad libitum for a further 12 weeks until 6 October, when the experiment concluded.During the first 6 weeks of 16L : 8D, growth rate (118 (s.e. 15·4) g/day) and food intake (1·37 (s.e. 0·031) kg DM per head per day) did not differ between the groups. Food intake following the clamp in LDRES averaged 1·40 (s.e. 0·015) kg per head per day. The intake of LDAL increased 2 weeks after the clamp and thereafter was higher than LDRES (P &lt; 0·001). Food intake of LDAL averaged 2·13 (s.e. 0·051) kg during the nutritional clamp period. Growth rates increased in both groups during the first 3 weeks of the clamp, averaging 237 (s.e. 25·0) g/day, then growth slowed in LDRES and live weights diverged. Growth rates until the end of the experiment (147 (s.e.23·0) g/ day v. 299 (s.e. 12·5) g/day, P &lt; 0·001) and mean live weight over the last 5 weeks of the experiment were lower (P &lt; 0·05) in LDRES than LDAL, weights reaching 88·3 (s.e. 1·86) kg and 97·9 (s.e. 2·74) kg respectively on the final sampling date. Metatarsal bone length grew more in LDAL than in LDRES (3·1 v. 2·2 cm, s.e.d. = 0·23, P &lt; 0·01). Prior to the nutritional clamp, mean plasma prolactin and IGF-1 concentrations increased at 3 and 6 weeks after 16L : 8D respectively, in both groups. Prolactin concentrations were lower in LDRES than LDAL on two occasions, at weeks 3 and 7 after the onset of the nutritional clamp, and IGF-1 concentrations were lower in LDRES than LDAL (676 v. 872 ng/ml, s.e.d. = 73·8, P &lt; 0·05) over the last 7 weeks of sampling.In summary, a photoperiodically driven increase in IGF-1 occurred even when the usual associated increase in food intake was prevented. This indicates that the seasonal IGF-1 rise in red deer is not a consequence of the increased food intake, although the latter appears necessary to maintain elevated IGF-1 concentrations. The rise in IGF-1 may therefore be considered as a component of the photoperiodically entrained seasonal drive to grow, and the increase in food intake a response to satisfy the increased energy demand.

Secretion of Prolactin and Growth Hormone in Relation to Ovarian Activity in the Dog

In pregnant bitches an apparent increase in plasma prolactin concentrations is observed during the second half of pregnancy, mean plasma prolactin concentrations peak on the day of parturition, fall for the next 24-48 h and then rise again. During lactation, high plasma prolactin concentrations are observed. Plasma prolactin levels in non-pregnant bitches appear to be lower than in pregnant animals, particularly in the last part of the luteal phase. Pulsatile secretion of prolactin has been observed during the luteal phase and mid-anoestrus. Progression of the luteal phase is found to be associated with an increase in prolactin release. The association of a strong increase of prolactin release and a decrease of plasma progesterone concentrations has also been demonstrated in overtly pseudopregnant bitches. Elevated prolactin secretion during progression of the luteal phase in the bitch may play a role in mammogenesis and is important because of the luteotrophic action of prolactin. Acromegaly is a syndrome of tissue overgrowth and insulin resistance due to excessive growth hormone (GH) production. In the bitch, acromegaly can be induced either by endogenous progesterone or by exogenous progestagens. Progestagen-induced GH production in this species originates from foci of hyperplastic ductular epithelium of the mammary gland. Pulsatile secretion of GH has been observed in normal cyclic bitches. In contrast with the pulsatile GH secretion seen in healthy dogs, the progestagen-induced plasma GH levels in bitches with acromegaly do not have a pulsatile secretion pattern. Just as with prolactin, the plasma progesterone levels influence the secretion pattern of GH in the bitch. The pulsatile secretion pattern of GH changes during the progression of the luteal phase in healthy cyclic bitches, with higher basal GH secretion and less GH being secreted in pulses during the first part of the luteal phase. The progesterone-induced GH production may promote the proliferation and differentiation of mammary gland tissue during the luteal phase of the bitch by local autocrine/paracrine effects and may exert endocrine effects.

Reproductive seasonality in domestic sows kept outdoors without boars

Plasma progesterone, LH and prolactin concentrations were measured twice a week in mature sows kept outdoors without boars in two experiments to examine whether perception of daylength change underlies seasonal infertility in domestic pigs. In Expt 1, melatonin implants inserted on 12 April or 22 May to block perception of the increasing daylength did not affect the oestrous cycle significantly, since only two untreated control sows became seasonally anoestrous. In Expt 2, all control sows became anoestrous for 131 +/- 42.5 days (n = 11). Melatonin implants inserted at the spring equinox (n = 9) prevented seasonal anoestrus (P < 0.001), but timed daily oral melatonin administration was less effective (P < 0.05): 5 of 11 sows became anoestrous for 132 +/- 45.6 days. In both experiments, there were significant low-amplitude seasonal rhythms in mean plasma prolactin and LH concentrations. Prolactin reached maximum concentrations 2-4 weeks before the summer solstice, whereas LH reached a nadir 4-6 weeks after the summer solstice. Neither anoestrus nor melatonin administration altered these patterns consistently. Endogenous plasma melatonin was higher (P < 0.001) during darkness (12.5 ng l(-1)) than during daytime (8.9 ng l(-1)) in untreated sows, but only clearly so during spring and summer. Melatonin implants increased mean daily plasma melatonin to 146 ng l(-1), whereas melatonin fed at 15:00 h increased values to 40-60 ng l(-1) 2-4 h after administration, but daytime concentrations were unchanged. Melatonin administration, despite decreasing seasonal anoestrus, did not prevent the seasonal increase in plasma prolactin and had no significant effect on plasma LH; therefore, its role in regulating seasonal changes in the reproduction of domestic sows remains uncertain.

The effects of perphenazine and bromocriptine on follicular dynamics and endocrine profiles in anestrous pony mares

Nineteen anestrous pony mares were used in a project designed to determine the effects of altered prolactin concentrations on follicular dynamics and endocrine profiles during spring transition. The dopamine antagonist, perphenazine, was administered daily to mares (0.375 mg/kg body weight) in Group A (n = 6), while Group B mares (n = 7) received 0.08 mg/kg metabolic weight (kg 75) dopamine agonist, 2-bromo-ergocriptine, intramuscularly twice daily. Mares in Group C (n = 6) received 0.08 mg/kg 75, im., saline twice daily. Treatment began January 20, 1994, and continued until ovulation occurred. Mares were teased 3 times weekly with an intact stallion. The ovaries of the ponies were palpated and imaged weekly using an ultrasonic B-mode unit with a 5 Mhz intrarectal transducer until they either exhibited estrual behavior and had at least a 20-mm follicle, or had at least a 25-mm follicle with no signs of estrus. At this time, ovaries were palpated and imaged 4 times weekly. Blood samples were obtained immediately prior to ultrasonic imaging for measurement of prolactin, FSH and estradiol-17β. Perphenazine treatment advanced the spring transitional period and subsequent ovulation by approximately 30d. Group A exhibited the onset of estrual behavior earlier (P < 0.01) than control mares. In addition, Group A mares developed large follicles (> 30 mm) earlier (P < 0.01) than Group B mares, with least square means for Groups A and B of 47.0 ± 8.8 vs 88.1 ± 8.2 d, respectively. Control mares developed 30-mm follicles intermediate to Groups A and B at 67.3 ± 8.8 d. Bromocriptine decreased (P < 0.05) plasma prolactin levels throughout the study, while perphenazine had no significant overall effect. However, perphenazine treatment did increase (P < 0.05) mean plasma prolactin concentrations from Day 31 to 60 of treatment. There were no differences in mean plasma FSH or estradiol-176 between treatment groups. We concluded that daily perphenazine treatment hastened the growth of follicles and subsequent ovulation while bromocriptine treatment appeared to delay the growth of preovulatory size follicles without affecting the time of ovulation.

Short-term inhibition of prolactin secretion by naloxone treatment in the pregnant gilt

Abstract The involvement of endogenous opioids in modulation of prolactin (PRL) secretion during pregnancy in the pig was studied. Twenty-four crossbred pregnant gilts (150 ± 10 kg) were cannulated via the cephalic vein 24–48 h before treatment with 1 mg kg −1 body weight of naloxone (NAL) or 3 ml of saline (CONT) i.v. at Day 40 (NAL, n = 6; CONT, n = 6) or Day 70 (NAL, n = 6; CONT, n = 6) of pregnancy. Blood plasma was collected at 15 min intervals from 1 h before to 3 h after treatment with NAL or saline. At Day 40 of pregnancy, administration of NAL caused a decrease in mean plasma PRL concentrations at 60 min, 120 min and 180 min post-treatment (NAL, 19.1 ± 1.3 ng ml −1 , P −1 , P −1 , P −1 , 21.6 ± 0.6 ng ml −1 and 22.4 ± 0.5 ng ml −1 , respectively). Mean plasma estradiol concentration was higher ( P P −1 , 16.2 ± 1.5 ng ml −1 and 14.8 ± 0.4 ng ml −1 , respectively) compared with the CONT group (33.4 ± 1.7 ng ml −1 , 34.1 ± 1.3 ng ml −1 and 29.1 ± 0.9 ng ml −1 , respectively). Estradiol concentrations were not different ( P > 0.05) between groups in this stage of gestation. Mean concentrations of progesterone were similar during the pre- and post-treatment periods in both stages of pregnancy. These data would suggest a possible role of the opioids in modulation of PRL secretion at these stages of pregnancy in the pig.

Long-term effects of time, medical treatment and pregnancy in 176 hyperprolactinemic women

The changes in plasma prolactin (PRL) concentrations were studied in 176 hyperprolactinemic women over periods of 6–180 months, to evaluate the independent effects of time, drugs and pregnancy on the evolution of prolactinemia. CT scans showed pituitary adenoma in 87 (9 macroadenoma), the clinical presentations for 110 patients there amenorrhea, for 37 abnormal cycles and 29 had anovulatory sterility as an isolated symptom. 107 women underwent 191 cycles of dopaminergic treatment and 73 had pregnancies (86), either spontaneously or as a consequence of the treatment. Changes in prolactin induced by medical treatment and pregnancy were recorded and the spontaneous changes in prolactin in 38 patients (17 with adenoma) were followed over periods of 6–72 months. Final mean PRL concentrations were lower than basal though not significantly, in both ‘functional’ (54.4 vs. 79.2 ng/ml) and prolactinoma patients (87.3 vs. 116.4 ng/ml). Separate calculation of changes in prolactin after the course of medical treatment, pregnancies or ‘just waiting’ periods showed mean PRL concentrations to be significantly lower only for ‘functional’ patients after pregnancy. On the other hand, PRL variations in individual patients revealed that: 1. (1) spontaneously, PRL rarely becomes lower over a few years; 2. (2) dopaminergic treatment was associated with normalization of PRL in 13% of women; and 3. (3) pregnancy normalized prolactin concentrations in 29% of the patients. Chi-square analysis of the PRL-lowering frequencies in functional patients showed a high cure rate for pregnancy ( P < 0.0001) and a lesser but still significant effect of drugs ( P < 0.025).

Effects of bromocriptine and perphenazine on prolactin and progesterone concentrations in pregnant pony mares during late gestation

Pregnant pony mares in Group A (n = 4) received i.m. injections at 07:00 and 17:00 h of 0.8 mg bromocriptine/kg body weight 0.75 per day beginning on Day 295 of gestation and continuing until parturition. Group B (n = 4) was treated similarly, but perphenazine was administered orally at 0.375 mg/kg body weight twice a day beginning on Day 305 of gestation and continuing until parturition. Mares in Group C (n = 3) received i.m. injections of saline. Mean plasma prolactin and progesterone concentrations were greater (P less than 0.05) for mares in Group C than in Groups A and B from 295 to 309 days of gestation. From 305 days of gestation, plasma prolactin and progesterone concentrations were greater (P less than 0.05) in Group B and C than in Group A mares. Progesterone and prolactin concentrations increased over this period for Group B and Group C mares, but remained constant in Group A mares. From 10 days pre partum through foaling, mares in Group A had lower progesterone (P less than 0.05) and prolactin (P less than 0.01) concentrations than Group B and C mares. All mares in Group A were agalactic at foaling, while all mares in Groups B and C had normal milk secretion. Gestation was longer (P less than 0.05) in Group A than in Group C mares. In Group A, 2 mares retained the placenta for greater than 3 h, 3 mares had dystocia and all 4 mares had thickened, haemorrhagic placentae.(ABSTRACT TRUNCATED AT 250 WORDS)

Daily rhythm of prolactin and corticosterone in unrestrained, incubating turkey hens

Plasma concentrations of prolactin and corticosterone were determined in hourly samples collected over a 25-hr period from unrestrained turkey hens exhibiting incubation (broody) behavior. Hens were maintained in cages on a 14 hr light: 10 hr dark photoperiod. Mean plasma prolactin concentration increased significantly late in the dark period to reach maximum daily levels of 496 ng/ml at the start of the photophase. However, a well-defined daily rhythm in prolactin secretion was not evident. Plasma corticosterone concentration showed a significant daily rhythm characterized by a major secretory peak (7.14 ng/ml) in the middle of the light period and a smaller, less well-defined peak (4.11 ng/ml) during the dark period. Both prolactin and corticosterone secretion ranged widely throughout the day in individual hens in a manner suggestive of pulsatile hormone secretion. This study demonstrates that the extremely high prolactin concentrations characteristic of the incubating turkey hen reflect large, dynamic changes in plasma prolactin concentrations occurring throughout the photoperiod, rather than a constantly high level of prolactin secretion.
