Abstract PO4-28-11: Murine Breast Cancer Cell Culture Supernatant Induces CAF-like and TAM-like Traits in Normal Cells

Abstract Several studies in the past have demonstrated the role of cell-to-cell interaction (crosstalk) between tumor cells and normal (nontumorigenic) cells in cancer progression and metastasis. Within the tumor microenvironment (TME), these interactions have a potential to transform the phenotypes and the behaviors of normal cells. Application of the 2D in-vitro cultures has been limited due to its inability to replicate the complex in-vivo TME. Conditioned Medium (CM) obtained from cultured cancer cells contains secreted growth factors that potentially regulate the phenotype and the functionality of normal cells. In this study, a culture of normal murine fibroblast NIH3T3 and macrophage RAW 264.7 cells with conditioned medium (CM) obtained from malignant mammary epithelial 4T1 cells (4T1CM) resulted in an altered phenotype with increased cell viability. 4T1CM treated NIH3T3 and RAW 264.7, in comparison with the respective control cells, resulted in an upregulation of the genes including- α-smooth muscle actin (αSMA), IL-10, CD206, and vascular endothelial growth factor (VEGF). In addition, 4T1.CM treated NIH3T3 showed an EMT phenotype as indicated by the regulation of EMT markers such as, E-cadherin, β-catenin, N-cadherin, and Vimentin. Interestingly, an upregulation of cyclooxygenase-2 (COX-2) and programmed death-ligand 1 (PDL-1) was observed in 4T1CM treated RAW 264.7, a tendency towards exhibiting an inhibitory immune response. Intriguingly, NIH3T3 cells conditioned with 4T1CM demonstrated an upregulation of stemness markers including- sex determining region Y-box 2, and Aldehyde dehydrogenase. Collectively, our study suggested a role for 4T1CM in transforming the normal NIH3T3 and RAW 264.7 into cancer-associated fibroblasts (CAFs) and tumor-associated macrophages (TAMs). RNAseq experiments are underway to map differentially expressed genes (DEGs) that potentially regulate the 4T1CM induced transformation of NIH3T3 and RAW 264.7 cells. Furthermore, invitro drug testing in 3D model of these transformed cells to target the pathway intermediates may provide novel therapeutic intervention strategies. Abstract 4219: Establishing CAF-like and TAM-like transformation induced by TNBC culture supernatant in 3D in-vitro culture as a model for targeted drug testing

Abstract Several previous studies have highlighted the significance of cell-to-cell communication, or crosstalk, between tumor cells and non-tumorigenic cells in the context of cancer progression and metastasis. Within the tumor microenvironment (TME), these interactions have the potential to alter the phenotypes and behaviors of normal cells. The use of 2D in-vitro cultures has been limited due to their inability to accurately replicate the intricate in-vivo TME. Conditioned medium (CM) derived from cultured cancer cells contains secreted factors that may influence the phenotype and functionality of normal cells. In our investigation, the exposure of normal murine fibroblast NIH3T3 and macrophage RAW 264.7 cells to conditioned medium (CM) obtained from malignant mammary epithelial 4T1 cells (4T1CM) resulted in a modified phenotype with enhanced cell viability. Treatment with 4T1CM led to the upregulation of genes, including α-smooth muscle actin (αSMA), IL-10, CD206, and vascular endothelial growth factor (VEGF), in NIH3T3 and RAW 264.7 cells compared to their respective control cells. Additionally, NIH3T3 cells treated with 4T1CM exhibited an epithelial-mesenchymal transition (EMT) phenotype, as indicated by the regulation of EMT markers such as E-cadherin, β-catenin, N-cadherin, and Vimentin. To further investigate, a series of experiments using mono- and co-cultures of these cells are underway, specifically targeting the transformed macrophages and fibroblasts with the aim of either re-educating or eliminating them. This treatment involves drug combinations that specifically target the identified signaling pathway intermediates, offering novel therapeutic intervention strategies. Differential regulation of fibronectin expression and fibrillogenesis by autocrine TGF-β1 signaling in malignant and benign mammary epithelial cells

Remodeling of the extracellular matrix (ECM) is a key hallmark of cancer progression. A critical component of ECM remodeling is the assembly of the glycoprotein fibronectin (FN) into insoluble fibrils, which provide a scaffold for invading vascular endothelial cells and escaping cancer cells, as well as a framework for collagen deposition and oncogenic cytokine tethering. FN fibril assembly is induced by Transforming Growth Factor-β1 (TGF-β1), which was originally identified for its role in malignant transformation. Addition of exogenous TGF-β1 drives FN fibril assembly while also upregulating endogenous TGF-β1 expression and autocrine signaling. In the current study, we sought to determine if autocrine TGF-β1 signaling plays a role in FN fibril formation in either MCF10A mammary epithelial cells, which behave similarly to healthy epithelia, or malignant MDA- MB-231 breast cancer cells. Our results show two interesting findings: first, malignant MDA-MB- 231 cells assemble less FN into fibrils, despite expressing and secreting more soluble FN; second, autocrine TGF-β1 signaling is required for FN fibril formation in MCF10A epithelial cells, even in the presence of exogenous, active TGF-β1. This suggests that autocrine TGF-β1 is signaling through distinct pathways from active exogenous TGF-β1. We hypothesized that this signaling was mediated by interactions between the TGF-β1 latency associated peptide (LAP) and αv integrins; indeed, incubating MCF10As with soluble LAP, even in the absence of the active TGF-β1 ligand, partially recovered FN fibril assembly. Taken together, these data suggests that autocrine TGF-β1 plays a critical role in FN fibril assembly, and this interaction is mediated by LAP-integrin signaling.

OSR1 phosphorylates the Smad2/3 linker region and induces TGF-β1 autocrine to promote EMT and metastasis in breast cancer.

Oxidative stress-responsive kinase 1 (OSR1) plays a critical role in multiple carcinogenic signal pathways, and its overexpression has been found in various types of cancer; however, the pathophysiological role of OSR1 in breast cancer has not been evaluated. This study aims to elaborate on the role of OSR1 in breast cancer metastasis and the specific regulatory mechanism. Our results showed that OSR1 mRNA and protein were upregulated in both human breast cancer samples and cell lines. Moreover, phosphorylated OSR1 (p-OSR1) was an independent poor prognostic indicator in patients with breast cancer. OSR1 upregulation induced epithelial-to-mesenchymal transition (EMT) in normal and malignant mammary epithelial cells with the increasing metastatic capacity. In contrast, deleting OSR1 in aggressive breast cancer cells inhibited these phenotypes. OSR1 is the critical activator for transcription factors of EMT. Mechanistically, we found that OSR1 can directly interact and phosphorylate the linker region of Smad2 at Thr220 and Smad3 at Thr179. Phosphorylated Smad2/3 translocated into the nucleus to enhance transforming growth factor-β1 (TGF-β1) autocrine signalling and increase the transcription of EMT regulators. Importantly, interruption of the OSR1-Smad2/3-TGF-β1 signalling axis elicited a robust anti-EMT and anti-metastatic effect in vitro and in vivo. Taken together, we conclude that OSR1-mediated Smad2/3-TGF-β1 signalling promotes EMT and metastasis representing a promising therapeutic target in breast cancer treatment.

LEFTY1 Is a Dual-SMAD Inhibitor that Promotes Mammary Progenitor Growth and Tumorigenesis.

SMAD pathways govern epithelial proliferation, and transforming growth factor β (TGF-β and BMP signaling through SMAD members has distinct effects on mammary development and homeostasis. Here, we show that LEFTY1, a secreted inhibitor of NODAL/SMAD2 signaling, is produced by mammary progenitor cells and, concomitantly, suppresses SMAD2 and SMAD5 signaling to promote long-term proliferation of normal and malignant mammary epithelial cells. In contrast, BMP7, a NODAL antagonist with context-dependent functions, is produced by basal cells and restrains progenitor cell proliferation. In normal mouse epithelium, LEFTY1 expression in a subset of luminal cells and rare basal cells opposes BMP7 to promote ductal branching. LEFTY1 binds BMPR2 to suppress BMP7-induced activation of SMAD5, and this LEFTY1-BMPR2 interaction is specific to tumor-initiating cells in triple-negative breast cancer xenografts that rely on LEFTY1 for growth. These results suggest that LEFTY1 is an endogenous dual-SMAD inhibitor and that suppressing its function may represent a therapeutic vulnerability in breast cancer.

Malignant canine mammary epithelial cells shed exosomes containing differentially expressed microRNA that regulate oncogenic networks

BackgroundBreast (mammary) cancers in human (BC) and canine (CMT) patients share clinical, pathological, and molecular similarities that suggest dogs may be a useful translational model. Many cancers, including BC, shed exosomes that contain microRNAs (miRs) into the microenvironment and circulation, and these may represent biomarkers of metastasis and tumor phenotype.MethodsThree normal canine mammary epithelial cell (CMEC) cultures and 5 CMT cell lines were grown in serum-free media. Exosomes were isolated from culture media by ultracentrifugation then profiled by transmission electron microscopy, dynamic light scattering, and Western blot. Exosomal small RNA was deep-sequenced on an Illumina HiSeq2500 sequencer and validated by qRT-PCR. In silico bioinformatic analysis was carried out to determine microRNA gene and pathway targets.ResultsCMEC and CMT cell lines shed round, “cup-shaped” exosomes approximately 150–200 nm, and were immunopositive for exosomal marker CD9. Deep-sequencing averaged ~ 15 million reads/sample. Three hundred thirty-eight unique miRs were detected, with 145 having > ±1.5-fold difference between one or more CMT and CMEC samples. Gene ontology analysis revealed that the upregulated miRs in this exosomal population regulate a number of relevant oncogenic networks. Several miRNAs including miR-18a, miR-19a and miR-181a were predicted in silico to target the canine estrogen receptor (ESR1α).ConclusionsCMEC and CMT cells shed exosomes in vitro that contain differentially expressed miRs. CMT exosomal RNA expresses a limited number of miRs that are up-regulated relative to CMEC, and these are predicted to target biologically relevant hormone receptors and oncogenic pathways. These results may inform future studies of circulating exosomes and the utility of miRs as biomarkers of breast cancer in women and dogs.

Abstract B67: The DLK1-DIO3 imprinted region regulates long-term proliferation in normal and malignant breast epithelium

Abstract DNA methylation changes are common in cancers, but their consequences are not fully understood. The DLK1-DIO3 locus, whose transcripts are controlled by the imprinted control region IG-DMR, is hypomethylated in 8% of breast cancer patients. Moreover, breast cancer patients with increased copy number of miR-127, an miR encoded in the DLK1-DIO3 locus, had worse disease-specific survival. IG-DMR reporter mice revealed that the elevated expression of miR-127 in immature normal and malignant mammary epithelial cells is controlled by allele-specific DNA methylation. Aberrant expression of miR-127 led to the de novo acquisition of long-term proliferation by normal and malignant mammary epithelial cells. Mechanistically, miR-127 suppresses translation of SETD8 and subsequent H4K20me1 deposition at LATS2, resulting in an effective activation of the Hippo target genes. Our results demonstrate that the DLK1-DIO3 region epigenetically regulates mammary stem cells, contributing to increased proliferative and tumorigenic capacity partially via regulation of the Hippo signaling pathway. Invasion in breast lesions: the role of the epithelial-stroma barrier.

Despite the significant biological, behavioural and management differences between ductal carcinoma in situ (DCIS) and invasive carcinoma of the breast, they share many morphological and molecular similarities. Differentiation of these two different lesions in breast pathological diagnosis is based typically on the presence of an intact barrier between the malignant epithelial cells and stroma; namely, the myoepithelial cell (MEC) layer and surrounding basement membrane (BM). Despite being robust diagnostic criteria, the identification of MECs and BM to differentiate in-situ from invasive carcinoma is not always straightforward. The MEC layer around DCIS may be interrupted and/or show an altered immunoprofile. MECs may be absent in some benign locally infiltrative lesions such as microglandular adenosis and infiltrating epitheliosis, and occasionally in non-infiltrative conditions such as apocrine lesions, and in these contexts this does not denote malignancy or invasive disease with metastatic potential. MECs may also be absent around some malignant lesions such as some forms of papillary carcinoma, yet these behave in an indolent fashion akin to some DCIS. In Paget's disease, malignant mammary epithelial cells extend anteriorly from the ducts to infiltrate the epidermis of the nipple but do not typically infiltrate through the BM into the dermis. Conversely, BM-like material can be seen around invasive carcinoma cells and around metastatic tumour cell deposits. Here, we review the role of MECs and BM in breast pathology and highlight potential clinical implications. We advise caution in interpretation of MEC features in breast pathology and mindfulness of the substantive evidence base in the literature associated with behaviour and clinical outcome of lesions classified as benign on conventional morphological examination before changing classification to an invasive lesion on the sole basis of MEC characteristics.

Abstract 215: Metabolic and mitochondrial consequences of p53 upregulation and their relation to cell fate outcomes

Abstract Background: p53 regulates a plethora of biological processes including cellular metabolism and mitochondrial function. Using the small molecule MDM2 agonist MI-63, we previously demonstrated differential p53 upregulation and induction of global metabolomics profiles in malignant MCF-7 and non-malignant MCF-10A mammary epithelial cells undergoing different cell fates (apoptosis vs. cytostasis). In particular, p53 upregulation suppressed glycolytic metabolite pools in MCF-10A, while increasing their levels in MCF-7 cells. We postulated that this opposing metabolic response may be linked to their different cell fate outcomes. In this study, we evaluated the association between p53 induced effects on live cell metabolic flux and cell fate using different p53wt cell lines subjected to comparable p53 upregulation. Methods: We employed four p53wt cell lines: MCF-7, ZR-75-1, SJSA-1 and MCF-10A. Cell fate outcomes upon p53 upregulation (24hr, 10μM MI63) were assessed by Western blotting for cytostasis (p21) and apoptosis markers (cleaved PARP, PUMA). Expression changes in enzymes involved in proline metabolism were similarly measured. Real-time measurements of oxygen consumption rates (OCR) and extracellular acidification rates (ECAR) under control and MI-63 treatments were assessed by the Seahorse Extracellular Flux Analyzer, and specific acidification contributions from respiration and glycolysis to total ECAR were computed. Results: p53 upregulation induced apoptosis in the malignant MCF-7, ZR-75-1 and SJSA-1 cells, but not in non-malignant MCF-10A cells. MI-63 treatment decreased basal OCR in MCF-7 cells (t-test p = 0.046), but not the other cell lines. No significant differences in maximum OCR were observed between control and MI-63 treatments in any of the cell lines tested. As well, p53 upregulation did not produce any significant change in total ECAR. However, in MCF-7 cells, glycolytic ECAR was significantly increased (MI-63/Control: 4.36, Tukey test p = 0.001), consistent with our earlier metabolomics conclusions. In contrast, no significant changes in glycolytic ECAR were produced in MCF-10A, ZR-75-1 or SJSA-1 cells (MI-63/Control: 1.27, 1.32 and 1.33 respectively). Interestingly, p53 upregulation induced expression of proline dehydrogenase (PRODH) in all three malignant cell lines, but not in MCF-10A cells; and coordinated changes in other proline catabolizing enzymes (P5C dehydrogenase, P5C reductase) were seen in MCF-7 cells. Conclusion: These findings indicate that p53 induced effects on oxygen consumption and glycolysis are cell context dependent but are not likely responsible for the different cell fate outcomes observed following p53 upregulation. Rather, our evidence suggests a potential mechanistic link between p53 induced effects on proline metabolism and cell fate outcomes, indicating that this stress-activated mitochondrial pathway deserves further study. Collective migration in tissues.

The Minisymposium on “Cell Migration in Tissues” focused on analysis of dynamic collective cell migration in several model systems and was cochaired by Alissa M. Weaver (Vanderbilt University Medical Center) and Celeste M. Nelson (Princeton University). The program highlighted both the physical forces exerted during migration and the signaling pathways involved in the process. Celeste Nelson (Princeton University) presented results suggesting that cells migrate collectively through fibrous extracellular matrix (ECM) by exerting tensile forces at the leading edge of the cohort. Three-dimensional traction force microscopy was used to quantify the physical forces exerted by normal and malignant mammary epithelial cells undergoing collective migration through collagen gels (Gjorevski et al., 2015 ). The forces and displacements were surprisingly dynamic, with the cohort periodically relaxing as it migrated forward. Jessica Konen (Marcus laboratory, Emory University) described a new technique called spatiotemporal genomic analysis, which was used to select rare populations of leader cells from three-dimensional (3D) lung cancer spheroids and compare their gene expression patterns to that of follower cells. Leader cells were found to express higher levels of genes related to cell adhesion and vascular sprouting. These gene expression changes persisted in purified cultures. Eliah Shamir (Ewald laboratory, Johns Hopkins University) showed that in 3D culture of mammary epithelial organoids, the myoepithelial layer was required to prevent invasion and dissemination of Twist1-expressing luminal epithelial cells. Surprisingly, Twist1-induced invasion required the expression of E-cadherin, and knocking down expression of this cell–cell adhesion protein prevented dissemination downstream of Twist1 (Shamir et al., 2014 ). Joshua Bloomekatz (Yelon laboratory, University of California, San Diego) showed a critical role for platelet-derived growth factor (PDGF) in the movement of cardiomyocytes during cardiac fusion. A forward genetic screen was used to identify a mutation called refuse-to-fuse, which causes inhibition of cardiac fusion and the subsequent formation of a bifurcated ventricle. This mutation was found to disrupt splicing of pdgfra, leading to expression of a truncated form of the PDGF receptor. Expression of PDGF ligands in relatively medial locations suggested a model in which PDGF signaling coordinates the movement of cardiomyocytes toward the midline during cardiac fusion. Bong Hwan Sung (Weaver laboratory, Vanderbilt University) showed that directionally persistent migration of cancer cells is promoted by autocrine secretion of exosomes, late endosome–derived extracellular vesicles (Sung et al., 2015 ). Exosomes were shown to contain ECM proteins as cargo and to be secreted at sites of nascent adhesions. Accordingly, exosome secretion promoted adhesion assembly, migrational polarity, and the stability of lamellipodia. David Matus (Stony Brook University) used an RNA interference screen to identify a transcriptional program that maintains invasive anchor cells in a cell cycle–arrested state during uterine-vulval attachment in Caenorhabditis elegans. Nuclear hormone receptor nhr-67 (TLX) expression leads to up-regulation of cyclin-dependent kinase inhibitor cki-1, causing cell cycle arrest in G1 (Matus et al., 2015 ). Cells that lack nhr-67 are proliferative and noninvasive, failing to express matrix or form invadopodia. The Minisymposium concluded with three lightning talks. Micah Webster (Fan laboratory, Carnegie Institution for Science) showed a role for “ghost fibers,” ECM remnants of damaged muscle fibers, in organizing skeletal muscle progenitor migration, division, and fusion during regeneration in mice (Webster et al., 2016 ). Rebecca Green (Oegema laboratory, University of California, San Diego) developed a four-dimensional high-content imaging approach to screen for developmentally important genes within C. elegans embryos. Yutaka Matsubayashi (Stramer laboratory, King’s College London) showed that formation of the basement membrane in the Drosophila embryo requires rapid anterior–posterior dispersal of macrophages (hemocytes), which first appear in the head of the embryo.

Abstract A63: Clonal analysis of normal and malignant human mammary epithelial cell responsiveness to radiation

Abstract Knowledge gap: Fatal breast cancers are characterized by biological, genomic and extensive treatment heterogeneity. Although many breast cancers can now be cured by established therapies, treatment failure remains a major problem and is difficult to predict. In the current era of “personalized medicine”, a possible solution is to develop a large-scale system for quantifying responses to candidate treatments of individual malignant human mammary cells with in vivo clonogenic activity. Such cells can be detected by their ability to produce uniquely barcoded clones of progeny in xenografted immunodeficient mice and the clones obtained can be assessed for their size and number using next generation sequencing of tumor extracts. However, to pursue this approach it is first critical to establish how the clone content of a tumor may vary according to the number or type of competent tumorigenic and/or other cells that are present in the inoculum used to initiate tumor formation, and hence whether and how these parameters may influence assessment of the treatment responsiveness of these cells. Approach/methods: Here we describe the development and initial testing of a method to measure the treatment responsiveness of large numbers of tumorigenic cells using radiation as a prototypic treatment. Treatment sensitivity of in vitro colony-forming cells (CFCs) will then be compared with future measurements of in vivo clone-initiating tumorigenic cells obtained by sequencing the progeny of DNA-barcoded input cells. Results: In an initial series of experiments we showed that normal human luminal progenitor (LP) CFCs are ~1.5-fold more radioresistant than basal cell (BC) CFCs, and both are more sensitive than either type of mouse mammary CFCs. In vitro CFC assays of 2 human breast cancer cell lines (MDA MB231 and SUM149, with in vitro CFC frequencies of 70% and 40%, respectively) showed these to be 1.2- and 1.5-fold more radioresistant than normal LPs. Limiting dilution analysis showed the corresponding frequency of in vivo tumor-initiating cells in these 2 cell lines to be 1/6 and 1/47. Assessment of their response to radiation is complicated by the finding that the barcoded clone content of tumors initiated with >20,000 of these cells (untreated) is inversely related to the number injected and, at these input cell doses, very heterogeneous clone dynamics are also seen in successive passages. However, evidence of a positive linear cell dose-clone yield relationship is seen at input transplants of <1,000 cells from these 2 lines and this relationship would be predicted to extend to initial transplants of <2-5,000 cells. Recently we have found that normal human mammary LPs and BCs transduced with KRASG12D generate tumors efficiently in immunodeficient mice. Thus a next step is to determine if and how the radiosensitivity of the tumorigenic cells changes during the transformation process. Conclusion: These results highlight the complex clonal dynamics already operative in the growth of tumorigenic cells present in relatively homogeneous established human mammary cell lines and set the stage for future measurements of clone yields from irradiated cells derived from mammary tumors of different origins. Abstract P4-05-08: Cellular localization dictates the role for c-Abl in breast cancer

Abstract c-Abl is a ubiquitously expressed non-receptor tyrosine kinase that regulates numerous aspects of normal mammary epithelial cell (MEC) physiology. However, the exact role of c-Abl during mammary tumorigenesis remains controversial. Clinical trials utilizing the c-Abl antagonist, imatinib mesylate, for the treatment of metastatic breast cancer (BC) patients failed due to disease progression. Additionally, our previous studies overexpressing a constitutively active c-Abl mutant in aggressive murine triple negative breast cancer (TNBC) cells abrogated tumorigenesis when injected into syngeneic mice. Through data mining we found c-Abl expression to be down-regulated across BC subtypes and this correlated with a significantly worse relapse free survival rate for patients of triple negative and luminal A BC subtypes. Furthermore, through in silico analyses we determined that BC patients with high c-Abl expression responded to docetaxel treatment, whereas their low c-Abl expressing counterparts did not. Taken together these findings support a tumor suppressive role for c-Abl in BC and that its targeted activation could inhibit BC tumorigenesis as well as sensitize BC patients to docetaxel treatment. We show here that shRNA mediated depletion of c-Abl elicited docetaxel resistance in malignant MECs through three dimensional (3-D) culture and clonogenic assays on plastic. Furthermore, we show through data mining that c-Abl expression is down-regulated in response to docetaxel treatment in a cohort of BC patients. This finding was recapitulated in malignant MECs through which docetaxel treatment selected for cells that retained lower c-Abl expression highlighting the importance of c-Abl in this cell death pathway. Targeted activation of c-Abl utilizing a novel small molecule activator, DPH, presents a novel therapeutic strategy for sensitizing BC patients to docetaxel treatment. However, we show DPH-mediated c-Abl activation to inhibit the proliferation of luminal A MCF7 BC cells, but enhance proliferation of the TNBC MDA-MB-231-L2 cell line in vitro. The cellular localization of c-Abl is crucial for its tumor suppressive functions. In Philadelphia chromosome positive chronic myelogenous leukemia (CML) the constitutively active BCR-Abl gene fusion has a strict cytoplasmic localization, however when nuclear expression of BCR-Abl is enforced CML cells undergo apoptosis. Cellular fractionation of normal murine MECs cells indicate that c-Abl expression is detected both in cytoplasmic and nuclear compartments before and with greater amounts detected in the nucleus upon activation. A similar trend is seen in the luminal A MCF7 BC cell line, however c-Abl is predominantly expressed in the cytoplasm basally in the TNBC MDA-MB-231-L2 cell line and retains a strict cytoplasmic localization upon activation with DPH. These findings suggest a similar mechanism as CML of cytoplasmic isolation as a means of circumventing the tumor suppressive role of c-Abl across different BC subtypes. Data from ongoing immunohistochemical analyses of a cohort of BC patient tissue microarrays will inform whether nuclear versus cytoplasmic c-Abl will correlate with available patient outcome data to determine if cellular localization dictates a tumor suppressive or tumor promoting role for c-Abl. Lysosomal protein turnover contributes to the acquisition of TGFβ-1 induced invasive properties of mammary cancer cells.

BackgroundNormal epithelial cells and carcinoma cells can acquire invasiveness by epithelial-to-mesenchymal transition (EMT), a process of considerable cellular remodeling. The endosomal/lysosomal compartment is a principal site of intracellular protein degradation. Lysosomal cathepsin proteases are secreted during cancer progression. The established pro-metastatic role of specific cysteine cathepsins has until now been ascribed to their contribution to extracellular matrix remodeling. We hypothesized that cysteine cathepsins affect transforming growth factor β-1 (TGFβ-1)-induced EMT of normal and malignant mammary epithelial cells.MethodsThe role of lysosomal proteolysis in TGFβ-1-induced EMT and invasion was investigated in a normal and a novel malignant murine mammary epithelial cell line. The contribution of cysteine cathepsins was determined by addition of the general cysteine cathepsin inhibitor E64d. Hallmarks of EMT were analyzed by molecular- and cell-biologic analyses including real-time cell migration/invasion assays. A quantitative proteome comparison using stable isotopic labeling with amino acids in culture (SILAC) showed the effect of E64d on TGFβ-1 induced proteome changes. Lysosomal patterning and junctional adhesion molecule A (Jam-a) localization and abundance were analyzed by immunofluorescence.ResultsWe found increased lysosome activity during EMT of malignant mammary epithelial cells. Cysteine cathepsin inhibition had no effect on the induction of the TGFβ-1-induced EMT program on transcriptional level. Protease inhibition did not affect invasion of TGFβ-1 treated normal mammary epithelial cells, but reduced the invasion of murine breast cancer cells. Remarkably, reduced invasion was also evident if E64d was removed 24 h before the invasion assay in order to allow for recovery of cathepsin activity. Proteome analyses revealed a high abundance of lysosomal enzymes and lysosome-associated proteins in cancer cells treated with TGFβ-1 and E64d. An accumulation of those proteins and of lysosomal vesicles was further confirmed by independent methods. Interestingly, E64d caused lysosomal accumulation of Jam-a, a tight junction component facilitating epithelial cell-cell adhesion.ConclusionOur results demonstrate an important role of lysosomal proteolysis in cellular remodeling during EMT and a pivotal contribution of lysosomal cysteine cathepsins to TGFβ-1 induced acquisition of breast cancer cell invasiveness. CUL4A Induces Epithelial–Mesenchymal Transition and Promotes Cancer Metastasis by Regulating ZEB1 Expression

The ubiquitin ligase CUL4A has been implicated in tumorigenesis, but its contributions to progression and metastasis have not been evaluated. Here, we show that CUL4A is elevated in breast cancer as well as in ovarian, gastric, and colorectal tumors in which its expression level correlates positively with distant metastasis. CUL4A overexpression in normal or malignant human mammary epithelial cells increased their neoplastic properties in vitro and in vivo, markedly increasing epithelial-mesenchymal transition (EMT) and the metastatic capacity of malignant cells. In contrast, silencing CUL4A in aggressive breast cancer cells inhibited these processes. Mechanistically, we found that CUL4A modulated histone H3K4me3 at the promoter of the EMT regulatory gene ZEB1 in a manner associated with its transcription. ZEB1 silencing blocked CUL4A-driven proliferation, EMT, tumorigenesis, and metastasis. Furthermore, in human breast cancers, ZEB1 expression correlated positively with CUL4A expression and distant metastasis. Taken together, our findings reveal a pivotal role of CUL4A in regulating the metastatic behavior of breast cancer cells.

TRAF4 Is a Novel Phosphoinositide-Binding Protein Modulating Tight Junctions and Favoring Cell Migration

Author SummaryTumor necrosis factor (TNF) receptor-associated factor 4, also known as TRAF4, is an unusual member of the TRAF protein family. While TRAFs are primarily known as regulators of inflammation, antiviral responses, and apoptosis, research on TRAF4 has identified its involvement in development and cancer. Importantly TRAF4 overexpression has been associated with a poor prognosis in breast cancers. TRAF4 has multiple subcellular localizations: to the plasma membrane in tight junctions (TJs), cytoplasmic and nuclear. The recruitment mechanisms and the functional impact of these distinct localizations are not completely understood. Here we investigate how TRAF4 is recruited to TJs and its involvement in cell–cell contacts in mammary epithelial cells (MECs). We show that the TRAF domain of all TRAFs contains a lipid binding module, and that TRAF4 uses this domain to form a trimeric complex that associates with phosphoinositides at the plasma membrane. Moreover this interaction is necessary for its recruitment to TJs. Additionally, we show that through its interaction with lipids TRAF4 delays TJ assembly and increases cell migration. We propose that TRAF4 has an important function in cancer progression by destabilizing TJs and favoring cell migration.

Hyperglycemia Enhances the Proliferation of Non-Tumorigenic and Malignant Mammary Epithelial Cells through Increased leptin/IGF1R Signaling and Activation of AKT/mTOR

Obesity and diabetes are associated with increased breast cancer risk and worse disease progression once cancer is diagnosed; however, the exact etiology behind these observations remains to be fully elucidated. Due to the global obesity/diabetes pandemic, it is imperative to understand how these diseases promote and enhance breast cancer and other common cancers. In this study we demonstrate that hyperglycemia promotes breast cancer by altering leptin/IGF1R and AKT/mTOR signaling. To our knowledge, we show for the first time that in breast epithelial cells, hyperglycemia alone directly impacts leptin signaling. Hyperglycemia increased proliferation of both non-tumorigenic and malignant mammary epithelial cells. These observations coincided with increased leptin receptor and IGF1R receptor, as well as, increased levels of GRB2, pJAK2, pSTAT3, pIRS1/2, pAKT, and p-mTOR. Moreover, pJAK2 was almost completely colocalized with leptin receptor under high glucose conditions. These results demonstrate how hyperglycemia can potentially increase the risk of breast cancer in premalignant lesions and enhance cancer progression in malignant cells.

The independent roles of mechanical, structural and adhesion characteristics of 3D hydrogels on the regulation of cancer invasion and dissemination

Metastasis begins with the escape, or dissemination, of cancer cells from the primary tumor. We recently demonstrated that tumors preferentially disseminate into collagen I and not into basement membrane protein gels (Matrigel). In this study, we used synthetic polymer systems to define material properties that could induce dissemination into Matrigel. We first specifically varied rigidity by varying the crosslinking density of poly(ethylene glycol) (PEG) networks within Matrigel scaffolds. Increased microenvironmental rigidity limited epithelial growth but did not promote dissemination. We next incorporated adhesive signals into the PEG network using peptide-conjugated cyclodextrin (α-CDYRGDS) rings. The α-CDYRGDS rings threaded along the PEG polymers, enabling independent control of matrix mechanics, adhesive peptide composition, and adhesive density. Adhesive PEG networks induced dissemination of normal and malignant mammary epithelial cells at intermediate values of adhesion and rigidity. Our data reveal that microenvironmental signals can induce dissemination of normal and malignant epithelial cells without requiring the fibrillar structure of collagen I or containing collagen I-specific adhesion sequences. Finally, the nanobiomaterials and assays developed in this study are generally useful both in 3D culture of primary mammalian tissues and in the systematic evaluation of the specific role of mechanical and adhesive inputs on 3D tumor growth, invasion, and dissemination.

Targeted inactivation of β1 integrin induces β3 integrin switching, which drives breast cancer metastasis by TGF-β

Mammary tumorigenesis and epithelial-mesenchymal transition (EMT) programs cooperate in converting transforming growth factor-β (TGF-β) from a suppressor to a promoter of breast cancer metastasis. Although previous reports associated β1 and β3 integrins with TGF-β stimulation of EMT and metastasis, the functional interplay and plasticity exhibited by these adhesion molecules in shaping the oncogenic activities of TGF-β remain unknown. We demonstrate that inactivation of β1 integrin impairs TGF-β from stimulating the motility of normal and malignant mammary epithelial cells (MECs) and elicits robust compensatory expression of β3 integrin solely in malignant MECs, but not in their normal counterparts. Compensatory β3 integrin expression also 1) enhances the growth of malignant MECs in rigid and compliant three-dimensional organotypic cultures and 2) restores the induction of the EMT phenotypes by TGF-β. Of importance, compensatory expression of β3 integrin rescues the growth and pulmonary metastasis of β1 integrin-deficient 4T1 tumors in mice, a process that is prevented by genetic depletion or functional inactivation of β3 integrin. Collectively our findings demonstrate that inactivation of β1 integrin elicits metastatic progression via a β3 integrin-specific mechanism, indicating that dual β1 and β3 integrin targeting is necessary to alleviate metastatic disease in breast cancer patients.

Activation-induced cytidine deaminase (AID) is necessary for the epithelial–mesenchymal transition in mammary epithelial cells

Activation-induced cytidine deaminase (AID), which functions in antibody diversification, is also expressed in a variety of germ and somatic cells. Evidence that AID promotes DNA demethylation in epigenetic reprogramming phenomena, and that it is induced by inflammatory signals, led us to investigate its role in the epithelial-mesenchymal transition (EMT), a critical process in normal morphogenesis and tumor metastasis. We find that expression of AID is induced by inflammatory signals that induce the EMT in nontransformed mammary epithelial cells and in ZR75.1 breast cancer cells. shRNA-mediated knockdown of AID blocks induction of the EMT and prevents cells from acquiring invasive properties. Knockdown of AID suppresses expression of several key EMT transcriptional regulators and is associated with increased methylation of CpG islands proximal to the promoters of these genes; furthermore, the DNA demethylating agent 5 aza-2'deoxycytidine (5-Aza-dC) antagonizes the effects of AID knockdown on the expression of EMT factors. We conclude that AID is necessary for the EMT in this breast cancer cell model and in nontransformed mammary epithelial cells. Our results suggest that AID may act near the apex of a hierarchy of regulatory steps that drive the EMT, and are consistent with this effect being mediated by cytosine demethylation. This evidence links our findings to other reports of a role for AID in epigenetic reprogramming and control of gene expression.

Molecular genetic analysis of VRK1 in mammary epithelial cells: depletion slows proliferation in vitro and tumor growth and metastasis in vivo

The vaccinia-related kinases (VRKs) comprise a branch of the casein kinase family. VRK1, a ser/thr kinase with a nuclear localization, is the most well-studied paralog and has been described as a proproliferative protein. In lower eukaryotes, a loss of VRK1 activity is associated with severe mitotic and meiotic defects. Mice that are hypomorphic for VRK1 expression are infertile, and depletion of VRK1 in tissue culture cells can impair cell proliferation and alter several signaling pathways. VRK1 has been implicated as part of a ‘gene-expression signature' whose overexpression correlates with poor clinical outcome in breast cancer patients. We present here our investigation of the role of VRK1 in the growth of normal (MCF10) and malignant (MDA-MB-231) human mammary epithelial cells, and demonstrate that shRNA-mediated depletion of VRK1 slows their proliferation significantly. Conversely, stable overexpression of a FLAG-tagged VRK1 transgene imparts a survival advantage to highly malignant MDA-MB-231 cells under conditions of nutrient and growth factor deprivation. Moreover, in a murine orthotopic xenograft model of breast cancer, we demonstrate that tumors depleted of VRK1 show a 50% reduction in size from 4–13 weeks postengraftment. The incidence and burden of distal metastases in the lungs and brain was also significantly reduced in mice engrafted with VRK1-depleted cells. These studies demonstrate that VRK1 depletion or overexpression has an impact on the proliferation and survival of cell lines derived from normal or malignant mammary tissue, and moreover show that depletion of VRK1 in MDA-MB-231 cells reduces their oncogenic and metastatic properties in vivo.
