Characterization of male killer whale ( Orcinus orca) sexual maturation and reproductive seasonality

Longitudinal serum testosterone concentrations ( n = 10 males) and semen production ( n = 2 males) in killer whales were evaluated to: (1) characterize fluctuations in serum testosterone concentrations with respect to reproductive maturity and season; (2) compare morphologic changes to estimated age of sexual maturity, based on changes in serum testosterone concentrations; and (3) evaluate seasonal changes in sperm production. Classification of reproductive status and age class was based on differences ( P < 0.05) in serum testosterone concentrations according to age; juvenile males ranged from 1 to 7 years (mean ± S.D. testosterone, 0.13 ± 0.20 ng/mL), pubertal males from 8 to 12 years (2.88 ± 3.20 ng/mL), and sexually mature animals were 13 years and older (5.57 ± 2.90 ng/mL). For captive-born males, serum testosterone concentrations, total body length and height to width ratio of the dorsal fin were 0.7 ± 0.7 ng/mL, 495.6 ± 17.5 cm and 1.14 ± 0.13 cm, respectively, at puberty; at sexual maturity, these end points were 6.0 ± 3.3 ng/mL, 548 ± 20 cm and 1.36 ± 0.1 cm. Serum testosterone concentrations were higher ( P < 0.05) from March to June than from December to February in pubertal animals (4.2 ± 3.4 ng/mL versus 1.4 ± 2.6 ng/mL) and than from September to December in sexually mature animals (7.2 ± 3.3 ng/mL versus 4.0 ± 2.0 ng/mL). Ejaculates ( n = 90) collected from two males had similar ( P > 0.05) sperm concentrations across all months. These data represent the first comprehensive study on male testosterone concentrations during and after sexual maturation, and on reproductive seasonality in the killer whale.

Wolbachia variant that induces two distinct reproductive phenotypes in different hosts

Wolbachia is an intracellular endosymbiont that induces a variety of reproductive alterations in diverse arthropods. The almond moth, Cadra cautella, is double infected with two Wolbachia variants, wCauA and wCauB, and expresses complete cytoplasmic incompatibility (CI). The individual contribution of wCauA and wCauB to the expression of CI are unclear, however, because the two variants have not been separated in this host. The effect of wCauA is of particular interest because it induces male killing when transferred into the Mediterranean flour moth, Ephestia kuehniella. In the present study, we generated C. cautella infected with only wCauA by treating double-infected insects with tetracycline. Single-infected C. cautella exhibited strong CI, demonstrating that wCauA induces two distinct reproductive phenotypes in different hosts: CI in C. cautella and male killing in E. kuehniella. CI was also observed in the cross of double-infected males and single-infected females. Comparison of the single- and double-infected insects by real-time quantitative polymerase chain reaction suggested that the wCauA density is not affected much by the presence or absence of wCauB.

Prevalence and penetrance variation of male‐killingWolbachiaacross Indo‐Pacific populations of the butterflyHypolimnas bolina

Male-killing bacteria are generally thought to attain low to intermediate prevalence in natural populations, with only mild effects on the host population sex ratio. This view was recently challenged by reports of extremely high infection frequencies in three butterfly species, raising the prospect that male killers, by making males rare, might drive many features of host ecology and evolution. To assess this hypothesis, it is necessary to evaluate how often male killers actually produce a highly female-biased population sex ratio in nature, which requires both high prevalence of infection and high penetrance of action. To this end, we surveyed South Pacific and Southeast Asian populations of Hypolimnas bolina, a butterfly in which extreme prevalence of male-killing Wolbachia bacteria has recently been recorded. Our results indicate that highly female-biased populations are common in Polynesia, with 6 out of 12 populations studied having in excess of 70% of females infected with a fully efficient male killer. However, heterogeneity is extreme in Polynesia, with the male-killing Wolbachia absent from three populations. In contrast to the Polynesian situation, Wolbachia does not kill males in any of the three Southeast Asian populations studied, despite its very high prevalence there. We conclude that male killers are likely to have significant ongoing ecological and evolutionary impact in 6 of the 15 populations surveyed. The causes and consequences of the observed spatial variation are discussed with respect to host resistance evolution, host ecology and interference with additional symbionts.

Widespread prevalence of wolbachia in laboratory stocks and the implications for Drosophila research.

Wolbachia is an intracellular microbe harbored by a wide variety of arthropods (including Drosophila) and filarial nematodes. Employing several different strategies including male killing, induced parthenogenesis, cytoplasmic incompatibility, and feminization, and acting by as-yet-unknown mechanisms, Wolbachia alters host reproduction to increase its representation within a population. Wolbachia is closely associated with gametic incompatibility but also interacts with Drosophila in other, little understood ways. We report here significant and widespread infection of Wolbachia within laboratory stocks and its real and potential impact on Drosophila research. We describe the results of a survey indicating that approximately 30% of stocks currently housed at the Bloomington Drosophila Stock Center are infected with Wolbachia. Cells of both reproductive tissues and numerous somatic organs harbor Wolbachia and display considerable variation in infection levels within and between both tissue types. These results are discussed from the perspective of Wolbachia's potential confounding effects on both host fitness and phenotypic analyses. In addition to this cautionary message, the infection status of stock centers may provide further opportunities to study the genetic basis of host/symbiosis.

Distribution, Expression, and Motif Variability of Ankyrin Domain Genes inWolbachia pipientis

The endosymbiotic bacterium Wolbachia pipientis infects a wide range of arthropods, in which it induces a variety of reproductive phenotypes, including cytoplasmic incompatibility (CI), parthenogenesis, male killing, and reversal of genetic sex determination. The recent sequencing and annotation of the first Wolbachia genome revealed an unusually high number of genes encoding ankyrin domain (ANK) repeats. These ANK genes are likely to be important in mediating the Wolbachia-host interaction. In this work we determined the distribution and expression of the different ANK genes found in the sequenced Wolbachia wMel genome in nine Wolbachia strains that induce different phenotypic effects in their hosts. A comparison of the ANK genes of wMel and the non-CI-inducing wAu Wolbachia strain revealed significant differences between the strains. This was reflected in sequence variability in shared genes that could result in alterations in the encoded proteins, such as motif deletions, amino acid insertions, and in some cases disruptions due to insertion of transposable elements and premature stops. In addition, one wMel ANK gene, which is part of an operon, was absent in the wAu genome. These variations are likely to affect the affinity, function, and cellular location of the predicted proteins encoded by these genes.

Wolbachia, sex ratio bias and apparent male killing in the harlequin beetle riding pseudoscorpion

Bacterial endosymbionts that manipulate host reproduction are now known to be widespread in insects and other arthropods. Since they inhabit the cytoplasm and are maternally inherited, these microorganisms can enhance their fitness by biasing host sex ratio in favour of females. At its most extreme, sex ratio manipulation may be achieved by killing male embryos, as occurs in a number of insect species. Here, we provide evidence for the first case of male killing by a tetracycline-sensitive microbe in pseudoscorpions. Using a combination of inheritance studies, antibiotic treatment and molecular assays, we show that a new strain of Wolbachia is associated with extreme female bias in the pseudoscorpion, Cordylochernes scorpioides. In a highly female-biased line, sex ratio distortion was maternally inherited, and occurred in conjunction with a high rate of spontaneous abortion and low reproductive success. Antibiotic treatment cured females of the Wolbachia infection, restored offspring sex ratio to 1:1, and significantly enhanced female reproductive success. The discovery of apparent male-killing in C. scorpioides is of interest because pseudoscorpions are viviparous. Theory predicts that male killing should be favoured, if male death enhances the fitness of infected female siblings. In a live-bearing host, reallocation of maternal resources from dead male embryos to their sisters provides a direct, physiological mechanism through which fitness compensation could favour male killing by cellular endosymbionts. Our results suggest, however, that fitness compensation and the spread of male-killing endosymbionts may be undermined by a high rate of spontaneous abortion in infected females of this viviparous arthropod.

Transinfection reveals the crucial importance of Wolbachia genotypes in determining the type of reproductive alteration in the host

Wolbachia , a group of endosymbiotic bacteria in arthropods, alter the reproduction of their hosts in various ways. A Wolbachia strain (wSca) naturally infecting the adzuki bean borer moth Ostrinia scapulalis induces male killing, while another strain (wKue) infecting the Mediterranean flour moth Ephestia kuehniella induces cytoplasmic incompatibility (CI) in the resident host. Transinfection of Wolbachia can be a powerful tool to elucidate the relative importance of Wolbachia and the host in determining the type of reproductive alterations. Recently, male killing was shown to occur in E. kuehniella transinfected with w Sca. In the present study, we transferred w Kue to O. scapulalis by embryonic microinjection. In the O. scapulalis transinfected with wKue, CI, but not male killing occurred. Thus, in addition to wSca, wKue was shown to induce the same type of alteration in a foreign host as in its natural host. These results demonstrate the crucial role of the Wolbachia genotype in determining the type of reproductive alteration. However, the present study also revealed the involvement of host factors. First, the degree of incompatibility induced by wKue in O. scapulalis was stronger than that in E. kuehniella , indicating that host factors can affect the level of CI. Second, the vertical transmission rate of wKue in O. scapulalis was generally low, suggesting that the host affects the dynamics of Wolbachia transmission.

Infection dynamics of different Wolbachia-types within one host population

Wolbachia are widespread intracellular symbionts of arthropods which are known to cause several reproductive manipulations in their hosts, the commonest of which being cytoplasmic incompatibility (CI), male killing (MK), and the induction of parthenogenesis (PI). Strains of endosymbionts inducing one of these effects can be referred to as `Wolbachia-types'. Here, we try to ascertain whether two of these Wolbachia-types can stably coexist within one population. We investigate this question by means of two discrete-time mathematical models which describe the dynamics of an infection of a host population with either CI- and MK- or CI- and PI-Wolbachia. We derive analytical solutions for two special cases of each model showing that stable coexistence of the respective Wolbachia-types is not possible if no doubly infected individuals occur within the population and that stable coexistence is possible when doubly infected hosts do exist and transmission of the endosymbionts is perfect. Moreover, we show that a population infected with either CI- or MK-Wolbachia at equilibrium can resist invasion of the respective other Wolbachia-type as a single infection. In contrast, a population infected with CI-Wolbachia can be invaded by PI-Wolbachia as a single infection with the CI-Wolbachia going extinct. Computer simulations confirmed these findings for the general models. We discuss our results with respect to the prevalence of the Wolbachia-types considered here and the emergence of PI- from CI-Wolbachia.

Sexy sons: a dead end for cytoplasmic genes

Critics of sexual conflict theory argue that females may gain a net reproductive benefit from mating with manipulative males because the direct costs that they suffer may be offset by the production of sexy, i.e. manipulative, sons. However, this exclusive focus on nuclear gene effects represents an incomplete view of female fitness. Females differ fundamentally from males in transmitting not only nuclear genes but also a wide range of cytoplasmic genetic elements (CGEs) that can have profound effects, from male killing to influencing development of the nervous system and cognitive ability. Maternal transmission of CGEs has two major implications for sexual selection. First, the evolution of male fitness traits, such as sperm competitive ability, may be constrained because response to selection on mitochondrial genomes can occur only through the female line. Second, CGEs bear the direct costs of male manipulation but gain no indirect benefits when females produce sexy sons. This should result in perpetual antagonistic coevolution between nuclear genes involved in male manipulation and CGEs that promote female resistance to male sexually selected traits. Explicit consideration of the consequences of selection acting on CGEs is therefore necessary for a better understanding of the relationship between sexual selection and sexual conflict.

PBDEs, PBBs, and PCNs in three communities of free-ranging killer whales (Orcinus orca) from the northeastern Pacific Ocean.

Polybrominated diphenyl ethers (PBDEs), polybrominated biphenyls (PBBs), and polychlorinated naphthalenes (PCNs) were quantified in blubber biopsy samples collected from free-ranging male and female killer whales (Orcinus orca) belonging to three distinct communities (southern residents, northern residents, and transients) from the northeastern Pacific Ocean. High concentrations of sigmaPBDE were observed in male southern residents (942+/-582 ng/g Iw), male and female transients (1015+/-605 and 885+/-706 ng/g Iw, respectively), and male and female northern residents (203+/-116 and 415+/-676 ng/g Iw, respectively). Because of large variation within sample groups, sigmaPBDE levels generally did not differ statistically with the exception of male northern residents, which had lower sigmaPBDE concentrations than male southern residents, male transients, and female transients, perhaps reflecting the consumption of less contaminated prey items. Male transient killer whales, which consume high trophic level prey including other cetaceans and occasionally spend time near populated areas, had sigmaPBDE concentrations approximately equal to southern residents. No significant age-related relationships were observed for sigmaPBDE concentrations. sigmaPBDE concentrations were approximately 1-3 orders of magnitude greater than those of sigmaPBB (3.0-31 ng/g Iw) and sigmaPCN (20-167 ng/g Iw) measured in a subset of samples, suggesting that PBDEs may represent a contaminant class of concern in these marine mammals.

“Who Kills Whom” Revisited

This article addresses the extraordinarily high ratio of females to males who kill domestic partners in the United States. Using a theoretical perspective conceptualizing violence between intimates as a form of self-help social control and as involving a pattern of sex-specific motives, the authors developed an explanatory model of variance in the rates of female and male spouse killing and thereby, in the spousal sex ratio of killing (SROK). Applied to cities in the United States with populations of 100,000 or more, the model predicted a significant proportion of variance in the rates of female and male killers (1984-1996) and in the SROK. Examination of the net differences in effects on sex-specific rates allowed for the interpretation of why the SROK varies so dramatically across communities and what factors contribute to the high spousal SROK in the United States. Directions for further sociological investigation of this question are discussed.

Low temperature cure of a male killing agent in Drosophila melanogaster

Environmental factors can affect transmission or phenotype expression of selfish cytoplasmic endosymbionts such as embryonic male killers. Temperature is one factor that usually affects the transmission rate of selfish cytoplasmic endosymbionts. Heat cures have been described for several host-parasite systems, cold cures, however, are rare. We report a temperature cure of the Drosophila melanogaster male-killing agent, which occurs when flies are raised at 16.5 °C. Flies grown at 20, 24, and 28 °C maintained an extremely female biased sexual proportion.

Distribution and Prevalence of &lt;I&gt;Wolbachia&lt;/I&gt; Infections in Native Populations of the Fire Ant &lt;I&gt;Solenopsis invicta&lt;/I&gt; (Hymenoptera: Formicidae)

Wolbachia are endosymbiotic bacteria that commonly infect arthropods. These bacteria induce a number of phenotypes in their hosts, including cytoplasmic incompatibility, thelytokous parthenogenesis, feminization, and male killing. We surveyed native South American populations of the fire ant Solenopsis invicta Buren for Wolbachia infections by using a diagnostic polymerase chain reaction assay. In addition, we determined the fidelity of vertical transmission of the bacteria from mother to offspring in this species by assaying daughters in 24 simple-family (monogyne) colonies. Infections were common in many parts of the extensive native range of S. invicta. However, the proportion of individuals infected varied greatly among samples, ranging from zero in several populations from the northerly parts of the range to >90% in more southerly populations. Possible explanations for this variation in the prevalence of Wolbachia infections are discussed. A survey of the two social forms of S. invicta from four geographic areas showed that the prevalence of Wolbachia infections consistently was higher in the monogyne form (single queen per colony) than the sympatric polygyne form (multiple queens per colony). One likely explanation for this trend is that the selective regimes acting on Wolbachia in the two forms differ because of the dissimilar reproductive strategies used by each form. Finally, overall transmission efficiency was found to be very high (>99%), making it unlikely that imperfect transmission prevents the spread of the microbe to near fixation in native populations.

Sperm chromatin remodelling and Wolbachia-induced cytoplasmic incompatibility in Drosophila.

Wolbachia pipientis is an obligate bacterial endosymbiont, which has successfully invaded approximately 20% of all insect species by manipulating their normal developmental patterns. Wolbachia-induced phenotypes include parthenogenesis, male killing, and, most notably, cytoplasmic incompatibility. In the future these phenotypes might be useful in controlling or modifying insect populations but this will depend on our understanding of the basic molecular processes underlying insect fertilization and development. Wolbachia-infected Drosophila simulans express high levels of cytoplasmic incompatibility in which the sperm nucleus is modified and does not form a normal male pronucleus when fertilizing eggs from uninfected females. The sperm modification is somehow rescued in eggs infected with the same strain of Wolbachia. Thus, D. simulans has become an excellent model organism for investigating the manner in which endosymbionts can alter reproductive programs in insect hosts. This paper reviews the current knowledge of Drosophila early development and particularly sperm function. Developmental mutations in Drosophila that are known to affect sperm function will also be discussed.incompatibility.

Diurnal and annual changes in serum cortisol concentrations in Indo-Pacific bottlenose dolphins Tursiops aduncus and killer whales Orcinus orca

Until present, fundamental studies on cortisol secretory patterns have not been conducted in cetaceans. The objectives of this study were: (1) to examine diurnal changes in serum cortisol concentrations in Indo-Pacific bottlenose dolphins Tursiops aduncus and killer whales Orcinus orca, (2) to investigate annual cortisol changes in killer whales, and (3) to investigate the relationship between cortisol and sex steroids (testosterone and progesterone) concentrations in killer whales. Diurnal changes in serum cortisol concentrations were investigated at various intervals in the two species. In Indo-Pacific bottlenose dolphins, serum cortisol levels exhibited the same episodic fluctuations for 24 h as did diurnal terrestrial mammals: cortisol levels were lower at 18:00 h and higher in the early morning. In killer whales, cortisol concentrations continued to decrease until 18:00 h, after which they fluctuated, and then increased in the next morning. Annual changes in cortisol levels were investigated by collecting blood samples every two weeks from two male killer whales and a pregnant female one twice per day (during 09:00–10:00 and 16:00–17:00 h) throughout a one-year period. Regarding sera collected during 09:00–10:00 h from the female, cortisol concentrations showed cyclic changes having about 4-month intervals. In males, cortisol showed higher concentrations in winter and lower concentrations during the summer season. There was a negative correlation between cortisol and progesterone levels in the female and a negative correlation was also observed between cortisol and testosterone in male no. 2. In the female and male no. 1, cortisol levels during 09:00–10:00 h were significantly higher than those during 16:00–17:00 h, and their data are considered to support observations regarding diurnal changes in cortisol levels in the two cetacean species.

Population dynamics of male-killing and non-male-killing spiroplasmas in Drosophila melanogaster.

The endosymbiotic bacteria Spiroplasma spp. are vertically transmitted through female hosts and are known to cause selective death of male offspring in insects. One strain of spiroplasma, NSRO, causes male killing in Drosophila species, and a non-male-killing variant of NSRO, designated NSRO-A, has been isolated. It is not known why NSRO-A does not kill males. In an attempt to understand the mechanism of male killing, we investigated the population dynamics of NSRO and NSRO-A throughout the developmental course of the laboratory host Drosophila melanogaster by using a quantitative PCR technique. In the early development of the host insect, the titers of NSRO were significantly higher than those of NSRO-A at the first- and second-instar stages, whereas at the egg, third-instar, and pupal stages, the titers of the two spiroplasmas were almost the same. Upon adult emergence, the titers of the two spiroplasmas were similar, around 2 x 10(8) dnaA copy equivalents. However, throughout host aging, the two spiroplasmas showed strikingly different population growth patterns. The titers of NSRO increased exponentially for 3 weeks, attained a peak value of around 4 x 10(9) dnaA copy equivalents per insect, and then decreased. In contrast, the titers of NSRO-A were almost constant throughout the adult portion of the life cycle. In adult females, consequently, the titer of NSRO was significantly higher than the titer of NSRO-A except for a short period just after emergence. Although infection of adult females with NSRO resulted in almost 100% male killing, production of some male offspring was observed within 4 days after emergence when the titers of NSRO were as low as those of NSRO-A. Based on these results, we proposed a threshold density hypothesis for the expression of male killing caused by the spiroplasma. The extents of the bottleneck in the vertical transmission through host generations were estimated to be 5 x 10(-5) for NSRO and 3 x 10(-4) for NSRO-A.

Genetic conflicts over sex ratio: mite-endosymbiont interactions.

Nucleocytoplasmic genetic conflicts arise as a result of asymmetric transmission of cytoplasmic and nuclear genes. Spread of a cytoplasmic element promoting female-biased sex ratios creates selection on nuclear genes for mechanisms that decrease the bias. Here we investigate the conflict over sex ratio between the cytoplasmic bacterium Wolbachia and the two-spotted spider mite Tetranychus urticae Koch. We show that, first, infected females produce significantly more female-biased sex ratios than uninfected (cured) females. Second, this effect is not due to parthenogenesis, male killing, or feminization, phenotypes commonly associated with infection by Wolbachia. Third, sex ratio is a trait with a heritable component in this species; thus, it can evolve under selection. Fourth, the sex ratio produced by uninfected (cured) females changes over time, approaching the sex ratio produced by females from the infected culture. On the basis of these results, we suggest that after sex ratio manipulation by Wolbachia, a host compensatory mechanism evolved that allows infected females to produce the sex ratio favored by nuclear genes. We discuss the evolution of "mutualism" with respect to the evolution of host mechanisms that compensate for effects induced by vertically transmitted "parasites."

Superinfection of Laodelphax striatellus with Wolbachia from Drosophila simulans.

Wolbachia are maternally inherited, intracellular alpha-proteobacteria that infect a wide range of arthropods. They manipulate the reproduction of hosts to facilitate their spread into host populations, through ways such as cytoplasmic incompatibility (CI), parthenogenesis, feminization and male killing. The influence of Wolbachia infection on host populations has attracted considerable interest in their possible role in speciation and as a potential agent of biological control. In this study, we used both microinjection and nested PCR to show that the Wolbachia naturally infecting Drosophila simulans can be transferred into a naturally Wolbachia-infected strain of the small brown planthopper Laodelphax striatellus, with up to 30% superinfection frequency in the F(12) generation. The superinfected males of L. striatellus showed unidirectional CI when mated with the original single-infected females, while superinfected females of L. striatellus were compatible with superinfected or single-infected males. These results are, to our knowledge, the first to establish a superinfected horizontal transfer route for Wolbachia between phylogenetically distant insects. The segregation of Wolbachia from superinfected L. striatellus was observed during the spreading process, which suggests that Wolbachia could adapt to a phylogenetically distant host with increased infection frequency in the new host population; however, it would take a long time to establish a high-frequency superinfection line. This study implies a novel way to generate insect lines capable of driving desired genes into Wolbachia-infected populations to start population replacement.

Space and the persistence of male-killing endosymbionts in insect populations.

Male-killing bacteria are bacteria that are transmitted vertically through the females of their insect hosts. They can distort the sex ratio of their hosts by killing infected male offspring. In nature, male-killing endosymbionts (male killers) often have a 100% efficient vertical transmission, and multiple male-killing bacteria infecting a single population are observed. We use different model formalisms to study these observations. In mean-field models a male killer with perfect transmission drives the host population to extinction, and coexistence between multiple male killers within one population is impossible; however, in spatially explicit models, both phenomena are readily observed. We show how the spatial pattern formation underlies these results. In the case of high transmission efficiencies, waves with a high density of male killers alternate with waves of mainly wild-type hosts. The male killers cause local extinction, but this creates an opportunity for uninfected hosts to re-invade these areas. Spatial pattern formation also creates an opportunity for two male killers to coexist within one population: different strains create spatial regions that are qualitatively different; these areas then serve as different niches, making coexistence possible.
