Accumulation of geranylgeranylated chlorophylls in the pigment-protein complexes of Arabidopsis thaliana acclimated to green light: effects on the organization of light-harvesting complex II and photosystem II functions

Light quality significantly influences plant metabolism, growth and development. Recently, we have demonstrated that leaves of barley and other plant species grown under monochromatic green light (500–590 nm) accumulated a large pool of chlorophyll a (Chl a) intermediates with incomplete hydrogenation of their phytyl chains. In this work, we studied accumulation of these geranylgeranylated Chls a and b in pigment-protein complexes (PPCs) of Arabidopsis plants acclimated to green light and their structural–functional consequences on the photosynthetic apparatus. We found that geranylgeranylated Chls are present in all major PPCs, although their presence was more pronounced in light-harvesting complex II (LHCII) and less prominent in supercomplexes of photosystem II (PSII). Accumulation of geranylgeranylated Chls hampered the formation of PSII and PSI super- and megacomplexes in the thylakoid membranes as well as their assembly into chiral macrodomains; it also lowered the temperature stability of the PPCs, especially that of LHCII trimers, which led to their monomerization and an anomaly in the photoprotective mechanism of non-photochemical quenching. Role of geranylgeranylated Chls in adverse effects on photosynthetic apparatus of plants acclimated to green light is discussed.

Probing the picosecond kinetics of the photosystem II core complex in vivo

Photosystems I (PSI) and II (PSII) are two major pigment-protein complexes of photosynthetic organisms that function in series to convert sunlight energy into chemical energy. We have studied the picosecond fluorescence behaviour of the core of both photosystems in vivo by using two Synechocystis PCC 6803 mutants: BE cells contain PSI but are lacking both PSII and the light-harvesting complexes called phycobilisomes (PBs) whereas PAL cells contain both PSI and PSII but lack the PBs. Measurements were performed at room temperature and at 77 K. The fluorescence kinetics of PSI and PSII can nicely be separated, en passant providing the PSI/PSII ratio. At room temperature, the PSI kinetics are identical to those of isolated PSI whereas the PSII kinetics can equally well be described by the in vitro trap-limited model of Y. Miloslavina, M. Szczepaniak, M. G. Muller, J. Sander, M. Nowaczyk, M. Rogner and A. R. Holzwarth, Biophys J., 2009, 96(2), 621-631, and by the transfer-to-the-trap-limited model of C. D. van der Weij-de Wit, J. P. Dekker, R. van Grondelle and I. H. M. van Stokkum, J. Phys. Chem. A, 2011, 115(16), 3947-3956, albeit that the in vivo kinetics turn out to be somewhat slower. At 77 K several low-energy pigments are observed in both photosystems which complicate the overall dynamics but the PSII kinetics can still be described by both a trap-limited and a transfer-to-the-trap-limited model.

Electronic Spectra of Structurally Deformed Lutein

Quantum chemical calculations have been employed for the investigation of the lowest excited electronic states of lutein, with particular reference to its function within light harvesting antenna complexes of higher plants. Through comparative analysis obtained by using different methods based on gas-phase calculations of the spectra, it was determined that variations in the lengths of the long C-C valence bonds and the dihedral angles of the polyene chain are the dominant factors in determining the spectral properties of Lut 1 and Lut 2 corresponding to the deformed lutein molecules taken from crystallographic data of the major pigment-protein complex of photosystem II. By MNDO-CAS-CI method, it was determined that the two singlet B(u) states of lutein (nominally 1B(u)(-)* and 1B(u)(+)) arise as a result of mixing of the canonical 1B(u)(-) and 1B(u)(+) states of the all-trans polyene due to the presence of the ending rings in lutein. The 1B(u)(-)* state of lutein is optically allowed, while the 1B(u)(-) of a pure all-trans polyene chain is optically forbidden. As demonstrated, the B(u) states are much more sensitive to minor distortions of the conjugated chain due to mixing of the canonical states, resulting in states of poorly defined particle-hole symmetry. Conversely, the A(g) states are relatively robust with respect to geometric distortion, and their respective inversion and particle-hole symmetries remain relatively well-defined.

Structural analysis of photosystem II in far-red-light-adapted thylakoid membranes. New crystal forms provide evidence for a dynamic reorganization of light-harvesting antennae subunits.

We studied two-dimensional crystals of the major pigment-protein complex, photosystem II, in far-red-light-adapted thylakoid membranes of the viridis-zb63 mutant of barley. Significantly larger grana membranes were produced with an increased synthesis of the entire photosystem II complex. These red-light-adapted membranes also contained two-dimensional crystals with a high frequency. Three different crystal forms of photosystem II were observed, providing the following data which further our understanding of the architecture of the native complex. (a) The oligomeric form of photosystem II in the membrane was monomeric in all crystal forms, but with a clear non-crystallographic pseudo-twofold symmetry. This was more apparent on the lumenal face of the complex. (b) The variability of unit cell contacts in different crystal forms implied that the peripheral light-harvesting antenna complex and the core of the complex were loosely connected. These peripheral subunits were predicted to rearrange so that they can either encircle the core complex or associate in parallel channels separated by lines of core complexes. (c) Grana membranes were found to retain a double-layered inside-out character, with a stromal face-to-stromal face packing. However, the presence of a crystal in one membrane did not necessarily impose crystallinity on its pair.

Differential control of xanthophylls and light-induced stress proteins, as opposed to light-harvesting chlorophyll a/b proteins, during photosynthetic acclimation of barley leaves to light irradiance

Barley (Hordeum vulgare L.) plants were grown at different photon flux densities ranging from 100 to 1800 &mgr;mol m-2 s-1 in air and/or in atmospheres with reduced levels of O2 and CO2. Low O2 and CO2 partial pressures allowed plants to grow under high photosystem II (PSII) excitation pressure, estimated in vivo by chlorophyll fluorescence measurements, at moderate photon flux densities. The xanthophyll-cycle pigments, the early light-inducible proteins, and their mRNA accumulated with increasing PSII excitation pressure irrespective of the way high excitation pressure was obtained (high-light irradiance or decreased CO2 and O2 availability). These findings indicate that the reduction state of electron transport chain components could be involved in light sensing for the regulation of nuclear-encoded chloroplast gene expression. In contrast, no correlation was found between the reduction state of PSII and various indicators of the PSII light-harvesting system, such as the chlorophyll a-to-b ratio, the abundance of the major pigment-protein complex of PSII (LHCII), the mRNA level of LHCII, the light-saturation curve of O2 evolution, and the induced chlorophyll-fluorescence rise. We conclude that the chlorophyll antenna size of PSII is not governed by the redox state of PSII in higher plants and, consequently, regulation of early light-inducible protein synthesis is different from that of LHCII.

Effects of ultraviolet-B radiation on phycobilisomes of Synechococcus PCC 7942: alterations in conformation and energy transfer characteristics.

Phycobilisomes (PBS), the major light harvesting antenna of the cyanobacterium Synechococcus contain phycocyanin (PC) and allophycocyanin (APC) as major pigment-protein complexes. PBS also absorb ultraviolet-B (280-320 nm) radiation. Exposure of Synechococcus PBS to low dose of UV-B (approximately 0.28 mw.cm-2) for 90 min induced change in absorption, emission and excitation characteristics of PBS and these changes got enhanced after 3 h of exposure. Room temperature excitation and emission spectra clearly indicated uncoupling of energy transfer from PC to APC on exposure to UV-B. Also, the 77K emission spectra suggested that F682 emission originating from APC decreased by 42% after 3 h of exposure. Circular dichroism (CD) spectra of UV-B exposed PBS indicated changes (14% decrease) in the alpha-helical content after 90 min treatment. SDS-PAGE analysis indicated degradation of a 75 kDa polypeptide (which appear to be a linker polypeptide) on UV-B treatment. The degradation of this polypeptide seems to induce changes in pigment-protein interaction and decoupling of energy transfer within the PBS. Our results for the first time clearly indicate that the PBS of Synechococcus are targets for UV-B damage.

Functional analysis of the iron-stress induced CP 43? polypeptide of PS II in the cyanobacterium Synechococcus sp. PCC 7942

Under conditions of iron-stress, the Photosystem II associated chlorophyll a protein complex designated CP 43', which is encoded by the isiA gene, becomes the major pigment-protein complex in Synechococcus sp. PCC 7942. The isiB gene, which is located immediately downstream of isiA, encodes the protein flavodoxin, which can functionally replace ferredoxin under conditions of iron stress. We have constructed two cyanobacterial insertion mutants which are lacking (i) the CP 43' apoprotein (designated isiA (-)) and (ii) flavodoxin (designated isiB (-)). The function of CP 43' was studied by comparing the cell characteristics, PS II functional absorption cross-sections and Chl a fluorescence parameters from the wild-type, isiA (-) and isiB (-) strains grown under iron-stressed conditions. In all strains grown under iron deprivation, the cell number doubling time was maintained despite marked changes in pigment composition and other cell characteristics. This indicates that iron-starved cells remained viable and that their altered phenotype suggests an adequate acclimation to low iron even in absence of CP 43' and/or flavodoxin. Under both iron conditions, no differences were detected between the three strains in the functional absorption crossection of PS II determined from single turnover flash saturation curves of Chl a fluorescence. This demonstrates that CP 43' is not part of the functional light-harvesting antenna for PS II. In the wild-type and the isiB (-) strain grown under iron-deficient conditions, CP 43' was present in the thylakoid membrane as an uncoupled Chl-protein complex. This was indicated by (1) an increase of the yield of prompt Chl a fluorescence (Fo) and (2) the persistence after PS II trap closure of a fast fluorescence decay component showing a maximum at 685 nm.

The highly abundant chlorophyll-protein complex of iron-deficient Synechococcus sp. PCC7942 (CP43') is encoded by the isiA gene.

A chlorophyll (Chl)-protein complex designated CPVI-4 becomes the major pigment-protein complex in Synechococcus sp. PCC7942 cells grown under conditions of iron limitation. Work by Laudenbach et al. (J Bacteriol [1988] 170: 5018-5026) has identified an iron-repressible operon, designated isiAB, containing the flavodoxin gene and a gene predicted to encode a Chl-binding protein resembling CP43 of photosystem II. To test the hypothesis that the CP43-like protein is a component of the CPVI-4 complex, we have inactivated the isiAB operon in Synechococcus sp. PCC7942 using directed insertional mutagenesis. Mutant cells grown under conditions of iron limitation exhibit pronounced changes in their spectroscopic and photosynthetic properties relative to similarly grown wild-type cells. Notably, the strong 77 K fluorescence emission at 685 nm, which dominates the spectrum of iron-deficient wild-type cells, is dramatically reduced in the mutant. The loss of this emission appears to unmask the otherwise obscured photosystem II emissions at 685 and 695 nm. Most importantly, mildly denaturing gel electrophoresis shows that mutant cells no longer express the CPVI-4 complex, indicating that the isiA gene encodes a component of this abundant Chl-protein complex.

Enhancement of carotenoid-to-chlorophyll singlet energy transfer by carotenoid-carotenoid interaction

The apparent quantum yield of singlet-singlet spirilloxanthin-to-bacteriochlorophyll a energy transfer increases linearly with the residual spirilloxanthin content in Rhodospirillum rubrum membrane vesicles from which this carotenoid has been partially removed. Since it has been previously shown that carotenoid-carotenoid interaction is a linear function of the residual spirilloxanthin level in the major pigment-protein complex of those vesicles (Zurdo, J., R. M. Lozano, C. Fernandez-Cabrera, and J. M. Ramirez. 1991. Biochem. J. 274:881-884), it appears that such degenerate interaction enhances singlet energy transfer. Part of the enhancement may be explained if the energy donor is the spirilloxanthin 1Bu----1Ag (S2----S0) transition, because exciton coupling probably brings its energy closer to that of the Qx (S2----S0) transition of bacteriochlorophyll. In contrast, it seems that the possible stabilization of the spirilloxanthin 2Ag (S1) state would hardly improve energy transfer, because this hidden state probably lies below the S1 bacteriochlorophyll state. In any case, the stabilizing effects of carotenoid-carotenoid interactions seem insufficient to explain the enhancement of energy transfer. Direct or indirect effects of carotenoid dimerization on the three-dimensional structure of the pigment cluster appear to be required to account for such enhancement.

Thylakoid membrane development in pigment-deficient wheat chloroplasts

The thylakoid membranes of higher plant chloroplasts contain at least two major pigment protein complexes, photosystem I (PSI), and photosystem II (PSII). The mature apoprotein of these complexes (involved in the initial reactions of photosynthesis) bind specific chlorophylls (Chl) and specific carotenoids in an unknown manner. It has been suggested, however, that the synthesis of pigments is normally coordinated with that of apoproteins. We have examined the effect of gabaculine (3-amino-2, 3-dihydrobenzoic acid) on granal thylakoid stacking as well as pigment and apoprotein accumulations for PSI and PSII in wheat.Gabaculine (0.5mM) was applied with nutrient solution to 6.5 day-old wheat seedlings maintained in a growth chamber at 23C. One seedling lot grown under continuous light (400 μmol photons s-1 m-2) possessed green primary leaves at time of treatment whereas another seedling lot, dark grown, possessed only etiolated primary leaves. Twelve hours after treatment, the etiolated seedlings were transferred into continuous light. The primary and secondary leaves were subsequently harvested from 14 day-old seedlings of both lots.

Assembly of the barley light-harvesting chlorophyll a/b proteins in barley etiochloroplasts involves processing of the precursor on thylakoids

A barley gene encoding the major light-harvesting chlorophyll a/b-binding protein (LHCP) has been sequenced and then expressed in vitro to produce a labelled LHCP precursor (pLHCP). When barley etiochloroplasts are incubated with this pLHCP, both labelled pLHCP and LHCP are found as integral thylakoid membrane proteins, incorporated into the major pigment-protein complex of the thylakoids. The presence of pLHCP in thylakoids and its proportion with respect to labelled LHCP depends on the developmental stage of the plastids used to study the import of pLHCP. The reduced amounts of chlorophyll in a chlorophyll b-less mutant of barley does not affect the proportion of pLHCP to LHCP found in the thylakoids when import of pLHCP into plastids isolated from the mutant plants is examined. Therefore, insufficient chlorophyll during early stages of plastid development does not seem to be responsible for their relative inefficiency in assembling pLHCP. A chase of labelled pLHCP that has been incorporated into the thylakoids of intact plastids, by further incubation of the plastids with unlabelled pLHCP, reveals that the pLHCP incorporated into the thylakoids can be processed to its mature size. Our observations strongly support the hypothesis that after import into plastids, pLHCP is inserted into thylakoids and then processed to its mature size under in vivo conditions.

Interaction of Chloroplasts with Inhibitors

A primary symptom of diuron (DCMU) phytotoxicity in plants is the destruction of chlorophyll. To study this process in vitro, chloroplasts from pea leaves (Pisum sativum L.) have been incubated in the light with DCMU for periods of up to 34 hours. The sequence of photodestruction of chlorophylls and carotenoids has been followed to try and establish the nature of the chloroplast protection mechanisms that are destroyed by DCMU. beta-Carotene decays most rapidly, followed by chlorophyll a and xanthophylls which are destroyed in a constant ratio, followed finally by chlorophyll b. Bypassing the DCMU block in the electron transport system with an artificial electron donor provides complete protection against chlorophyll and carotenoid photodestruction. The same protection by this electron donor system is afforded to stroma-free lamellae from which soluble reductants have been removed so that NADPH formation, which has been proposed as an essential part of a protective xanthophyll cycle, is not possible. Both this and the simultaneous loss of chlorophyll a and xanthophylls tend to preclude the breakdown of a xanthophyll cycle from the possible protective mechanisms inhibited or destroyed by DCMU.Cofactors of cyclic electron transport also protect against DCMU-induced photodestruction of pigments. Their concentration dependence for this protection appears to reflect their various abilities to catalyze cyclic photophosphorylation. The extent to which the chlorophylls are destroyed in the major pigment-protein complexes from chloroplasts illuminated with and without DCMU has been measured. In the absence of DCMU, the light-harvesting chlorophyll a/b protein complex is destroyed most rapidly. In the presence of DCMU, the losses of chlorophyll a from the photosystem I P700-chlorophyll a protein and the chlorophyll a/b complex are about the same. Chlorophyll losses are matched by simultaneous losses of the protein moieties; spectral analyses show that the remaining chlorophyll a is held in a loose association with the protein. Phenazine methosulfate protects the chlorophyll of the light-harvesting complex in DCMU-treated chloroplasts more than it protects that in photosystem I. Data published on DCMU-induced fluorescence and its quenching are used to interpret the longer term DCMU-induced chlorosis and its protection. By blocking electron transport, conformational changes in the membrane that allow spillover of excitation energy from photosystem II to photosystem I (and quenching of fluorescence by this means) are prevented. The mechanism that normally protects the chloroplast against excessive illumination is then overloaded which impairs the harmless dissipation of absorbed light energy; consequently, the pigments are destroyed. When photosystem I is allowed to function again through cyclic electron flow, a necessary conformational change is believed to be reintroduced that once again allows the harmless dissipation of excitation energy through spillover. A functional electron transport system associated with photosystem I will protect against DCMU-induced chlorosis when the thylakoid membranes are intact, but when the P700-chlorophyll a protein complex is in isolation, there is only a limited degree of protection.
