Chemically Powered Nanomotors with Magnetically Responsive Function for Targeted Delivery of Exosomes.

Janus structure plays a crucial role in achieving chemically driven nanomotors with exceptional motion performance. However, Janus-structured chemically driven nanomotors with magnetic responsiveness are commonly fabricated by sputtering metal films. In the study, a self-assembly technique is employed to asymmetrically modify the surfaces of magnetic silica (SiO2@Fe3O4) nanoparticles with platinum nanoparticles, resulting in the formation of this kind nanomotors. Compared to platinumfilm, platinum nanoparticles exhibit a larger surface area and a higher catalytic activity. Hence, the nanomotors demonstrate improved diffusion capabilities at a significantly lower concentration (0.05%) of hydrogen peroxide (H2O2). Meanwhile, exosomes have gained attention as a potential tool for the efficient delivery of biological therapeutic drugs due to their biocompatibility. However, the clinical applications of exosomes are limited by their restricted tropism. The previously obtained nanomotors are utilized to deliver exosomes, greatly enhancing its targetability. The drug doxorubicin (DOX) is subsequently encapsulated within exosomes, acting as a representative drug model. Under the conditions of H2O2 concentration at the tumor site, the exosomes exhibited a significantly enhanced rate of entry into the breast cancercells. The utilization of the nanomotors for exosomes presents a novel approach in the development of hybrid chemically and magnetically responsive nanomotors.

MRI-traceable multifunctional magnetic mesoporous silica nanoparticles (MMSN) capped with graphene quantum dots (GQD) as a theragnostic system in colorectal cancer treatment

In the study, a multifunctional nanosystem composed of magnetic core-mesoporous silica nanoparticles (MMSN) capped with graphene quantum dots (GQD) was proposed as a theragnostic platform against colorectal cancer cells. The theragnostic carrier was well characterized and had an average particle size of 140 nm, with a specific surface of 475.82 m2 g−1. The superparamagnetic nanoparticles showed a hysteresis loop, with a maximum magnetization of 14.93 emu.g−1 and a remanence of 2.64 emu.g−1. The functionalization of MMSN was confirmed using the ninhydrin test. Regorafenib (REGO), an anticancer drug, was used with a loading capacity of 77.5 μg mg−1. The drug release profiles from the carrier and the sensitivity of the interaction between MMSN and GQD to pH were examined, revealing that more drug release was observed in acidic pH. The cytotoxicity of the MMSN-REGO/GQD system was evaluated on colorectal cancer cell line SW-480 using MTT assay, and the results indicated that no cytotoxicity was observed for the MMSN/GQD carrier. Furthermore, the IC50 of REGO was significantly reduced from 85 μg ml−1 to 6 μg ml−1 in the loaded form. The response of MMSN/GQD nanoparticles to the external magnetic field exhibited the capability of the theragnostic system as an MRI contrast agent. Generally, biocompatible, theragnostic, pH-responsive, multifunctional MMSN/GQD can be considered promising candidates for treatment, MRI imaging, and hyperthermia in colorectal cancer treatment.

New Cu(II) Schiff-base complex modified on magnetic nanoparticles: Oxidation and DFT investigation

A copper magnetic nanocatalyst was facilely synthesized by surface modification of silica magnetic nanoparticles (Fe3O4@SiO2) via reaction with 3-aminopropyltrimethoxysilane (Fe3O4@SiO2@APTMS), followed by a subsequent reaction with thiophene-2-carbaldehyde to form a Schiff base (Fe3O4@SiO2@SB). Sequentially, reaction with Cu(CH3COO)2·H2O in ethanol yielded Fe3O4@SiO2@[SB@Cu] (CH3COO)2. This nanomagnetic catalyst was comprehensively characterized using Fourier Transform Infrared Spectroscopy (FT-IR), X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM), Thermogravimetric Analysis (TGA), and Vibrating Sample Magnetometry (VSM) techniques. The catalytic proficiency of these nanoparticles was evaluated through alkene and alcohol oxidation reactions. Under optimized conditions, the nanocatalyst exhibited significant efficacy in oxidizing styrene, cyclohexene, and benzyl alcohol, with notable selectivity values of 88 %, 100 %, and 100 % for the respective formation of styrene epoxide, 2-cyclohexene-1-one, and benzoic acid. The heterogeneous copper nanocatalyst offers advantages, including high conversion rates, facile separation, and exceptional selectivity in alkene and alcohol oxidation. Additionally, this study investigated density functional theory (DFT) methods, encompassing geometric optimization, natural charge analysis, molecular electrostatic potential maps, and vibrational normal modes of the chelating ligands.

An ultra-sensitive SARS-CoV-2 antigen optical biosensor based on angiotensin converting enzyme 2 (ACE-2) functionalized magnetic-fluorescent silica nanoparticles.

This work reports on the design and synthesis of an angiotensin-converting enzyme 2 (ACE-2) functionalized magnetic fluorescent silica nanoparticles (Fe-FSNP) as a biosensing platform to detect severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) antigen. Iron oxide (Fe3O4) nanoparticles were synthesized via ultrasonic-assisted coprecipitation and then coated with fluorescent silica nanoparticles (FSNP) through thesol-gelmethod forming the Fe-FSNP samples. Silica obtained from local geothermal powerplant was used in this work and Rhodamine B was chosen as the incorporated fluorescent dye, hence this reports for the first time ACE-2 was immobilized on the natural silica surface. The Fe-FSNP nanoparticle consists of a 18-25 nm magnetic core and a silica shell with a thickness of 30 nm as confirmed from the transmission electron microscopy image. Successful surface functionalization of the Fe-FSNP with ACE-2 as bioreceptor was conducted through hydrosylilation reaction and confirmed through the Fourier transform infrared spectroscopy. The detection of SARS-Cov-2 antigen by Fe-FSNP/ACE2 was measured through the change in its maximum fluorescence intensity at 588 nm where fluorescence- quenching had occurred. The biosensing platform showed a rapid response at 30 min with a linear range of 10-6to 10-2μg ml-1. The magnetic-fluorescent properties of the nanoparticle enables an ultra-sensitive detection of SARS-Cov-2 antigen with the limit of detection as low as 2 fg ml-1.

Stimuli-responsive magnetic silica-poly-lactic-co-glycolic acid hybrid nanoparticles for targeted cancer chemo-immunotherapy.

Chemotherapy and immunotherapy are two important modalities in cancer management. However, due to multiple reasons, a monotherapy is only partially effective. Hence, if used concurrently in targeted and stimuli-responsive manner, it could have been superior therapeutically. To facilitate co-delivery of chemotherapeutic and immunotherapeutic agent to the target cancer cells, engineered nanoparticles, i.e., a pH-responsive polymer PLGA-coated magnetic silica nanoparticles (Fe3O4-SiO2-PLGA-PDA-PTX-siRNA NPs) encapsulating paclitaxel (PTX) and siRNA against programmed cell death ligand-1 (PD-L1) are synthesized and characterized. Developed nanoparticles demonstrated pH-sensitive sustained drug release up to 10 days. In vitro 4T1 cell line studies showed efficient cellular uptake, PD-L1 gene downregulation, and apoptosis. Further, in vivo efficacy studies carried out in the mice model demonstrated a significant reduction of tumor growth following treatment with dual-Fe3O4-SiO2-PLGA-PDA-PTX-siRNA NPs as compared with monotherapy with Fe3O4-SiO2-PLGA-PDA-PTX NPs. The high therapeutic efficacy observed with dual-Fe3O4-SiO2-PLGA-PDA-PTX-siRNA NPs was mainly due to the cytotoxic effect of PTX combined with targeted silencing of the gene of interest, i.e., PD-L1, which in turn improve CD8+ T cell-mediated cancer cell death as evident with increased proliferation of CD8+ T cells in co-culture experiments. Thereby, dual-Fe3O4-SiO2-PLGA-PDA-PTX-siRNA NPs may have a promising anti-cancer treatment potential against breast cancer; however, the beneficial effects of dual loading of PTX + PD-L1 siRNA may be corroborated against other cancer models such as lung and colorectal cancer models as well as in clinical trials.

Network–Polymer–Modified Superparamagnetic Magnetic Silica Nanoparticles for the Adsorption and Regeneration of Heavy Metal Ions

Superparamagnetic magnetic nanoparticles (MNPs, Fe3O4) were first synthesized based on a chemical co–precipitation method, and the core–shell magnetic silica nanoparticles (MSNPs, Fe3O4@SiO2) were obtained via hydrolysis and the condensation of tetraethyl orthosilicate onto Fe3O4 seed using a sol–gel process. Following that, MSNPs were immobilized using a three–step grafting strategy, where 8-hloroacetyl–aminoquinoline (CAAQ) was employed as a metal ion affinity ligand for trapping specific heavy metal ions, and a macromolecular polymer (polyethylenimine (PEI)) was selected as a bridge between the surface hydroxyl group and CAAQ to fabricate a network of organic networks onto the MSNPs’ surface. The as–synthesized MSNPs–CAAQ nanocomposites possessed abundant active functional groups and thus contained excellent removal features for heavy metal ions. Specifically, the maximum adsorption capacities at room temperature and without adjusting pH were 324.7, 306.8, and 293.3 mg/g for Fe3+, Cu2+, and Cr3+ ions, respectively, according to Langmuir linear fitting. The adsorption–desorption experiment results indicated that Na2EDTA proved to be more suitable as a desorbing agent for Cr3+ desorption on the MSNPs–CAAQ surface than HCl and HNO3. MSNPs–CAAQ exhibited a satisfactory adsorption capacity toward Cr3+ ions even after six consecutive adsorption–desorption cycles; the adsorption efficiency for Cr3+ ions was still 88.8% with 0.1 mol/L Na2EDTA as the desorbing agent. Furthermore, the MSNPs–CAAQ nanosorbent displayed a strong magnetic response with a saturated magnetization of 24.0 emu/g, and they could be easily separated from the aqueous medium under the attraction of a magnet, which could facilitate the sustainable removal of Cr3+ ions in practical applications.

Solid-phase extraction of gallic acid and ascorbic acid in fruits using polymerized deep eutectic solvent-modified substrate.

Aim: Novel substrates were synthesized by porous and nonporous polymerization of deep eutectic solvents on magnetic silica nanoparticles and introduced for dispersive solid-phase extraction of two analytes. Materials & methods: The prepared substrates were characterized, and an extraction procedure was implemented to select the best substrates and eluent. The central composite design acted to optimize the effects of parameters that influenced the extraction efficiencies. Results: For gallic and ascorbic acids, the limits of detection were obtained at 0.136 and 0.165μM, respectively, with linear ranges of 0.6-125.2 and 0.5-106.8μM, respectively. Conclusion: The substrate produced good extractions even after being used three-times and was successfully applied for the analysis of real samples.

Effective myocardial infarction treatment by targeted accumulation of Sulforaphane using porous magnetic silica nanoparticles

Myocardial infarction (MI) is a common cardiovascular pathology that induces extensive sterile inflammation during its early stages, posing a severe threat to human health. Effectively modulating cardiac inflammation may improve post-MI outcomes. Unfortunately, owing to the side effects of therapeutic drugs and cardiac coronary artery occlusion, current MI drugs are sub-optimal for the clinical management of ischemic myocardia. Sulforaphane (SFN) has been adopted for MI treatment due to its myocardial protective effects and low toxicity. However, the targeted accumulation of SFN in infarcted areas remains challenging. Herein, porous magnetic silica nanoparticles (PMSNs) were synthesized and loaded with SFN to improve the specificity of targeted SFN delivery to infarcted areas in mouse models of MI. PMSNs loaded with SFN (PMSNs + SFN) decreased the levels of pro-inflammatory cytokines, thus leading to the improvement of cardiac function and cell survival without adverse effects. To further explore SFN’s mechanisms of action in MI, a cellular (in vitro) model was established via oxygen and glucose deprivation (OGD). HSF1 and Nrf2 knockdown resulted in a decrease of SFN-induced HSP70 expression in OGD cells. Moreover, as a result of HSP70 knockdown, the pro-survival and anti-inflammatory effects of SFN were blocked in OGD cells. The level of pro-inflammatory cytokines decreased upon HSP70 overexpression, and cell survival rate increased under OGD conditions. In summary, the results confirm that PMSNs are capable of transporting SFN to infarcted areas in the myocardium, where the drug exerts cardioprotective effects against myocardial injury by up-regulating HSP70 through Nrf2/HSF1.

Preparation and PET/CT imaging of implant directed 68Ga-labeled magnetic nanoporous silica nanoparticles

BackgroundImplant infections caused by biofilm forming bacteria are a major threat in orthopedic surgery. Delivering antibiotics directly to an implant affected by a bacterial biofilm via superparamagnetic nanoporous silica nanoparticles could present a promising approach. Nevertheless, short blood circulation half-life because of rapid interactions of nanoparticles with the host’s immune system hinder them from being clinically used. The aim of this study was to determine the temporal in vivo resolution of magnetic nanoporous silica nanoparticle (MNPSNP) distribution and the effect of PEGylation and clodronate application using PET/CT imaging and gamma counting in an implant mouse model.MethodsPEGylated and non-PEGylated MNPSNPs were radiolabeled with gallium-68 (68Ga), implementing the chelator tris(hydroxypyridinone). 36 mice were included in the study, 24 mice received a magnetic implant subcutaneously on the left and a titanium implant on the right hind leg. MNPSNP pharmacokinetics and implant accumulation was analyzed in dependence on PEGylation and additional clodronate application. Subsequently gamma counting was performed for further final analysis.ResultsThe pharmacokinetics and biodistribution of all radiolabeled nanoparticles could clearly be visualized and followed by dynamic PET/CT imaging. Both variants of 68Ga-labeled MNPSNP accumulated mainly in liver and spleen. PEGylation of the nanoparticles already resulted in lower liver uptakes. Combination with macrophage depletion led to a highly significant effect whereas macrophage depletion alone could not reveal significant differences. Although MNPSNP accumulation around implants was low in comparison to the inner organs in PET/CT imaging, gamma counting displayed a significantly higher %I.D./g for the tissue surrounding the magnetic implants compared to the titanium control. Additional PEGylation and/or macrophage depletion revealed no significant differences regarding nanoparticle accumulation at the implantation site.ConclusionTracking of 68Ga-labeled nanoparticles in a mouse model in the first critical hours post-injection by PET/CT imaging provided a better understanding of MNPSNP distribution, elimination and accumulation. Although PEGylation increases circulation time, nanoparticle accumulation at the implantation site was still insufficient for infection treatment and additional efforts are needed to increase local accumulation.

Synergistic Effect of Magneto-Mechanical Bioengineered Stem Cells and Magnetic Field to Alleviate Osteoporosis.

Therapeutic bioengineering based on stem cell therapy holds great promise in biomedical applications. However, the application of this treatment is limited in orthopedics because of their poor survival, weak localization, and low cell retention. In this work, magneto-mechanical bioengineered cells consisting of magnetic silica nanoparticles (MSNPs) and mesenchymal stem cells (MSCs) are prepared to alleviate osteoporosis. The magneto-mechanical bioengineered MSCs with spatial localization, cell retention, and directional tracking capabilities could be mediated by a guided magnetic field (MF) in vitro and in vivo. Furthermore, high uptake rates of the MSNPs ensure the efficient construction of magnetically controlled MSCs within 2 h. In conjunction with external MF, the magneto-mechanical bioengineered MSCs have the potential for the activation of the YAP/β-catenin signaling pathway, which could further promote osteogenesis, mineralization, and angiogenesis. The synergistic effects of MSNPs and guided MF could also decline bone resorption to rebalance bone metabolism in bone loss diseases. In vivo experiments confirm that the functional MSCs and guided MF could effectively alleviate postmenopausal osteoporosis, and the bone mass of the treated osteoporotic bones by using the bioengineered cells for 6 weeks is nearly identical to that of the healthy ones. Our results provide a new avenue for osteoporosis management and treatment, which contribute to the future advancement of magneto-mechanical bioengineering and treatment.

Microwave-assisted alkali pre-treatment medium for fractionation of rice straw and catalytic conversion to value-added 5-hydroxymethyl furfural and lignin production

In the current study, the use of microwave-assisted sodium hydroxide medium (MWSH) for pre-treatment and saccharification of rice straw to obtain sugar syrup for the production of 5-hydroxymethyl furfural (5-HMF) was investigated. The optimization of the MWSH pre-treatment was carried out using central composite methodology, resulting in a maximum reducing sugar yield of 350 mg/g of treated rice straw (TRS) and a glucose yield of 255 mg/g of TRS under the conditions of a microwave power of 681 W, a NaOH concentration of 0.54 M, and a pre-treatment time of 3 min. Additionally, the microwave assisted transformation of sugar syrup with titanium magnetic silica nanoparticle as catalyst, producing 41.1 % yield of 5-HMF from the sugar syrup after 30 min microwave irradiation at 120 °C with catalyst loading of 2.0:200 (w/v)). The structural characterization of the lignin was analysed using 1H NMR techniques, and the surface carbon (C1s spectra) and oxygen (O1s spectra) composition changes of the rice straw during pre-treatment were analysed using X-ray photoelectron spectroscopy. The rice straw based bio-refinery process which contains MWSH pretreatment followed by dehydration of sugars achieved high efficiency of 5-HMF production.

Polyethylenimine–Silica–Magnetic Nanoparticle Composites for Efficient Extraction of Dissolved Copper from Aqueous Solutions and Multiphase Mixtures

Design of micro/nanoadsorbents developed for effective copper extraction should consider methods of their retrieval from relevant copper-rich environments, which are commonly complex multiphase mixtures, such as slurries of polluted soils and sediments, copper minerals including low-grade ores, and industrial and mining wastes. This work reports integration of copper-chelating polyethylenimine (PEI) with a robust silica network and highly magnetic magnetite nanoparticles, resulting in magnetic PEI–silica nanocomposite microparticles that enable efficient binding of copper ions in both aqueous solutions and complex, multiphase mixtures with subsequent retrieval using magnetic force. Produced via simple emulsion templating, these nanocomposite particles can withstand highly acidic conditions and multiple copper extraction cycles, without loss of efficiency after five consecutive cycles of copper extraction. PEI–silica–magnetic nanoparticles composites effectively bind all copper ions leached into an aqueous phase and allow their rapid magnetically induced separation from aqueous solutions and mixtures of both coarse and fine solid particles, demonstrating promising results for progress toward more sustainable copper extraction techniques.

Comparative X-Ray Diffraction (XRD) Study on the Synthesis of Natural Based Magnetic Silica Nanoparticles

The aim of this study was to compare the synthesis process of magnetic silica nanoparticles (Fe-SNP) through the analysis of X-ray Diffraction (XRD) results. Fe3O4 magnetic nanopowders was synthesized by ultrasonic assisted co-precipitation and Fe-SNP was synthesized by direct mixing method of sodium silicate (Na2SiO3) and magnetite (Fe3O4) and the sol-gel method. Silica sludge was used as a silica source from Indonesia geothermal power plant waste. The synthesized of Fe-SNP is the functionalization of Fe3O4 with silica. Variations concentration of Na2SiO3 is used for the direct mixing method and variations of Fe3O4 form is used for the sol-gel method. Particles formed and particle size were characterized by XRD. The XRD results showed that there is no SiO2 phase in the sample synthesized by direct mixing method while two phases of SiO2 and Fe3O4 were found in the sample synthesized by sol-gel method. The size of the Fe3O4 nanoparticles calculated with Scherer’s formula and it obtained 19.9 nm, while the Fe3O4 nanoparticles with the addition of 20 mL and 6 mL Na2SiO3 concentrations were 6,53 nm and 10,23 nm. For the sol-gel method the size of Fe3O4 nanoparticles obtained was 11,03 using Fe3O4 powders and 9,86 using Fe3O4 solutions.

Influence of PEG Chain Length of Functionalized Magnetic Nanoparticles on the Cytocompatibility and Immune Competence of Primary Murine Macrophages and Dendritic Cells.

A major drawback of nanoparticles (NPs) for biomedical applications is their preferential phagocytosis in immune cells, which can be avoided by surface modifications like PEGylation. Nevertheless, examinations of different polyethylene glycol (PEG) chain lengths on the competence of immune cells as well as possible immunotoxic effects are still sparse. Therefore, primary murine macrophages and dendritic cells were generated and incubated with magnetic nanoporous silica nanoparticles (MNPSNPs) modified with different mPEG chains (2 kDa, 5 kDa, and 10 kDa). Cytotoxicity, cytokine release, and the formation of reactive oxygen species (ROS) were determined. Immune competence of both cell types was examined and uptake of MNPSNPs into macrophages was visualized. Concentrations up to 150 µg/mL MNPSNPs showed no effects on the metabolic activity or immune competence of both cell types. However, ROS significantly increased in macrophages incubated with larger PEG chains, while the concentration of cytokines (TNF-α and IL-6) did not indicate a proinflammatory process. Investigations on the uptake of MNPSNPs revealed no differences in the onset of internalization and the intensity of intracellular fluorescence. The study gives no indication for an immunotoxic effect of PEGylated MNPSNPs. Nevertheless, there is still a need for optimization regarding their internalization to ensure an efficient drug delivery.

Organosilane based magnetic silica nanoparticles for the detection of Al(iii) ions and construction of a molecular logic gate

The work describes organosilane magnetic silica nanoparticles, functioning as a fluorescent sensor: ‘ON’ for Al(iii) and ‘OFF’ for 4-NP. These nanoparticles also serve as an IMPLICATION logic gate by responding to added Al(iii) and 4-NP.

Sol-gel polymerization of silylated amino acids around a protein template yields selective biomimetic imprints

The development of artificial receptors able to selectively recognize a target protein is of particular interest in separation, diagnostics, and therapeutics fields. Herein, we disclose a method to prepare biomimetic and functionalized protein imprints in biocompatible conditions avoiding any protein denaturation. For that purpose, a set of different hybrid silylated amino acid derivatives were synthesized and used without tetraethyl orthosilicate to prepare our molecularly imprinted polymers, allowing to reduce to a minimum of the silicon amount, in order to obtain imprints made almost entirely of amino acids to mimic paratope surfaces of antibodies. Such functional building blocks were polymerized on the surface of magnetic silica nanoparticles at pH 8.5 in ultrapure water in the presence of two globular proteins: cytochrome C or lysozyme. The resulting imprinted hybrid materials were evaluated for their adsorption capacity, specificity, and selectivity by quartz-crystal microbalance with dissipation and magnetic enzyme-linked immunosorbent assay (ELISA) assays. High imprinting factors of 8.7 were measured for these biomimetic hybrid materials (corresponding to approximately 4000 and 450 ng of protein per cm2 immobilized on molecularly imprinted polymers and non-imprinted polymer nanoparticles, respectively), representing a significant breakthrough in sol-gel-based molecular imprinting materials. Moreover, competition experiments performed by magnetic ELISA (mELISA) show very good specificity of our imprints at the usual concentrations of ELISA measurements.

Upconversion nanoparticles as an immunocomplexing agent for selective detection of caspases via sandwich-like supracomplexes

In this study, a nanoparticle-based sandwich-like immunoassay was designed in dispersion medium to precisely detect apoptosis over caspase antibodies in order to overcome the disadvantages of traditional apoptosis determination methods such as high cost, large sampling requirement, and appropriate laboratory and equipment conditions. For this purpose, a complementary particulate system including magnetic (MNPs) and upconversion silica (UC-SiNPs) nanoparticles while immobilizing antibodies (primary antibody to MNPs, secondary antibody to UC-SiNPs) were synthesized and characterized. Optimization and selectivity studies of the complex formed by primary antibody immobilized MNPs with standard caspase proteins were examined by the HPLC system. Within the scope of optimization studies, protein concentrations, optimal duration, and temperature parameters were evaluated. Optimal conditions were determined for pH, initial concentration, time, and temperature as 7.4, 5.6 μg/mL, 45 min, and room temperature, respectively. Furthermore, the adsorption of competitive proteins was investigated in selectivity studies as well. Moreover, the primary antibody immobilized MNPs were treated with standard caspase proteins under optimal conditions; subsequently, they were interacted with secondary antibody immobilized UC-SiNPs to demonstrate the supracomplex formation meanwhile zeta potential/size measurements and fluorescence emission spectrometry analyses were performed. As a result of these analyses, it was observed that the sandwich-like supracomplexes were successfully formed that significantly varied upconversion emission intensities of UC-SiNPs in dependence on the amounts of caspase proteins. Because this approach enabled a quantitative result, the nanoparticle-based sandwich-like immunoassay should be classified as an easy-to-handled, fast, and promising alternative to benchmark apoptosis assays.

Application of functionalized magnetic silica nanoparticles for selective induction of three coumarin metastable polymorphs

The precise control of active pharmaceutical ingredient (API) crystal nucleation and polymorphism is a key consideration in pharmaceutical manufacturing. In this study, tunable nanoparticles were developed to regulate the nucleation process of coumarin. Magnetic silica nanoparticles with four different functional groups (–NH 2 , –COOH, –SH, –NCO) were prepared and coated on the substrate for inducing the crystallization of coumarin. Confined melt crystallization and microspacing sublimation crystallization methods were used to investigate the regulation mechanism. The results indicated that three metastable forms of coumarin can be obtained as pure components based on the combined influence of crystallization methods and functionalized nanoparticles. Form II could be selectively obtained by microspacing sublimation crystallization on Fe 3 O 4 @SiO 2 –SH substrates, and Form IV could be obtained by confined melt crystallization on Fe 3 O 4 @SiO 2 –NCO substrates. Form III could be obtained by further heating Form IV crystals to 52 °C on Fe 3 O 4 @SiO 2 –NCO substrates. Moreover, the polarized light microscopy results also indicated that the introduction of nanoparticles could also increase the stability of the metastable crystalline forms of coumarin. Finally, the diffusion and surface dynamics during nanoparticle induced crystallization were comparatively investigated and the corresponding polymorphic selectivity mechanism was proposed.
