A “Talking” between Gold Nanoparticle and a Luminescent Iridium(III) Complex: A Study of the Effect Due to the Interaction between Plasmon Resonance and a Fluorophore

This paper explores a novel synthesis and characterization of silica-coated gold nanorods (AuNRs) embedding a highly emissive cyclometalated iridium(III) complex, denoted as Ir1. We investigate the optical properties and the interplay between the metal compound and gold plasmon, observing how the emission of Ir1 incorporated into the nanoparticles shows two emission bands, one in the blue and the other in the green-orange range of the visible spectrum. To obtain a clearer picture of what we were observing, we synthesized analogous nanosystems, from which it was possible to highlight the effect of different features. Based on what we observed, we proposed that the fraction of the iridium(III) complex in direct contact with the surface of the gold nanoparticle undergoes a “demixing” of the excited state, which, for cyclometalated iridium complexes, is generally considered a mixed LC+MLCT state. This preliminary study sheds light on the complexity of the “talking” between a fluorophore and a plasmonic system, highlighting the importance of considering the emitter typology when modeling such systems.

Experimental and theoretical studies of pH-responsive iridium(III) complexes of azole and N-heterocyclic carbene ligands.

A series of nine luminescent iridium(III) complexes with pH-responsive imidazole and benzimidazole ligands have been prepared and characterized. The first series of complexes were of the form [Ir(ppy)2(N^N)]+ or [Ir(ppy)2(C^N)]+ (where ppy is 2-phenylpyridine and N^N is 2-(2-pyridyl)imidazole or 2-(2-pyridyl)benzimidazole and C^N represents a pyridyl-triazolylidene-based N-heterocyclic carbene ligand). For these complexes, the benzimidazole group was either unsubstituted or substituted with electron-withdrawing (Cl) or electron-donating (Me) groups. The second series of complexes were of the form [Ir(phbim)2(N^N)]+ or [Ir(phbim)2(C^N)]+ (where phbim is 2-phenylbenzimidazole and N^N is either 2,2'-bipyridine or 1,10-phenanthroline and C^N is either a pyridyl-imidazolylidene or pyridyl-triazolylidene N-heterocyclic carbene ligand). UV-visible and photoluminescence pH titration studies showed that changing the protonation state of these complexes results in significant changes in the photoluminescence emission properties. The pKa values of prepared complexes were estimated from the spectroscopic pH titration data and these values show that the nature of the pH-sensitive ligands (either main or ancillary ligands) resulted in a significant capacity to modulate the pKa values for these compounds with values ranging from 5.19-11.22. Theoretical investigations into the nature of the electronic transitions for the different protonation states of compounds were performed and the results were consistent with the experimental results.

Thermally activated delayed fluorescence in a deep red dinuclear iridium(iii) complex: a hidden mechanism for short luminescence lifetimes

Thermally activated delayed fluorescence (TADF) is found to play an important role in accelerating the rate of radiative decay in luminescent iridium(iii) complexes, shown in this instance for dinuclear complexes formed with high stereoselectivity.

A new family of luminescent iridium complexes: synthesis, optical, and cytotoxic studies.

By using N,N-dibutyl-2,2'-bipyridine-4,4'-dicarboxamide as a diimine (dbbpy) and distinctive cyclometalated groups, this work reports a new family of cationic phosphorescent Ir(III) cyclometalated [Ir(C^N)2(N^N)]X compounds [C^N = difluorophenylpyridine (dfppy) a, 2,6-difluoro-3-(pyridin-2-yl)benzaldehyde (CHO-dfppy) b, and 2,6-difluoro-3-pyridin-2-yl-benzoic acid (COOH-dfppy) c; X = Cl-2a,b,c-Cl; X = PF6-2b,c-PF6]. For comparative purposes, the related complex [Ir(dfppy)2(H2dcbpy)]+ (3a-PF6) incorporating 3,3'-dicarboxy-2,2'-bipyridine as an auxiliary ligand (N^N = H2dcbpy) is also presented. All complexes have been fully characterized and their photophysical properties were investigated in detail. The theoretically calculated results obtained by density functional theory (DFT) and time-dependent density functional theory (TD-DFT) studies indicate that luminescence is derived from mixed 3ML'CT (Ir → N^N)/3LL'CT (C^N → N^N) excited states with the predominant metal-to-diimine charge transfer character. Their antineoplastic activity against tumour cell lines A549 (lung carcinoma) and HeLa (cervix carcinoma), as well as the nontumor BEAS-2B (bronchial epithelium) cell line was assessed and fluorescence microscopy studies were performed for their cellular localization. Among them, 2a-Cl exhibited the most potent anticancer activity, being higher than cisplatin. However, 2b-Cl and 2c-Cl,-PF6 were the least toxic, while 2b-PF6 and 3a-PF6 exhibited only moderate activity. Confocal microscopy studies for 2a-Cl suggest that complexes localize preferentially in the lysosomes and to a lesser extent in the cytoplasm, but ultimately causing damage to the mitochondria. Finally, the potential photodynamic behaviour of scarcely toxic complexes 2b-Cl, 2b-PF6, 2c-Cl and 3a-PF6 was also studied.

Affinity-Based Luminescent Iridium(III) Complexes for the Detection of Disease-Related Proteins

The occurrence of diseases is usually accompanied by changes in protein levels and types. These differentially expressed proteins can be used as biomarkers for the diagnosis and treatment of diseases. In recent years, luminescent iridium(III) complexes have attracted much attention in the field of protein-based disease diagnosis due to their excellent optical properties. In particular, affinity-based luminescent iridium(III) complexes have the advantage of evaluating protein information with minimal interference on their biological activities. In this review, we summarize the current advances in affinity-based luminescent iridium(III) complexes for the detection of disease-related proteins. Moreover, the future perspective for affinity-based iridium(III) complexes is discussed.

Luminescent Iridium Complexes with a Sulfurated Bipyridine Ligand: PCET Thermochemistry of the Disulfide Unit and Photophysical Properties.

Molecular systems combining light harvesting and charge storage are receiving great attention in the context of, for example, artificial photosynthesis and solar fuel generation. As part of ongoing efforts to develop new concepts for photoinduced proton-coupled electron transfer (PCET) reactivities, we report a cyclometallated iridium(III) complex [Ir(ppy)2(S-Sbpy)](PF6) ([1]PF6) equipped with our previously developed sulfurated bipyridine ligand S-Sbpy. A new one-step synthetic protocol for S-Sbpy is developed, starting from commercially available 2,2'-bipyridine, which significantly facilitates the use of this ligand. [1]+ features a two-electron reduction with potential inversion (|E1| > |E2|) at moderate potentials (E1 = -1.12, E2 = -1.11 V versus. Fc+/0 at 253 K), leading to a dithiolate species [1]-. Protonation with weak acids allows for determination of pKa = 23.5 in MeCN for the S-H···S- unit of [1H]. The driving forces for both the H atom and the hydride transfer are calculated to be ∼60 kcal mol-1 and verified experimentally by reaction with a suitable H atom and a hydride acceptor, demonstrating the ability of [1]+ to serve as a versatile PCET reagent, albeit with limited thermal stability. In MeCN solution, an orange emission for [1]PF6 from a triplet-excited state was found. Density functional calculations and ultrafast absorption spectroscopy are used to give insight into the excited-state dynamics of the complex and suggest a significantly stretched S-S bond for the lowest triplet-state T1. The structural responsiveness of the disulfide unit is proposed to open an effective relaxation channel toward the ground state, explaining the unexpectedly short lifetime of [1]+. These insights as well as the quantitative ground-state thermochemistry data provide valuable information for the use of S-Sbpy-functionalized complexes and their disulfide-/dithiol-directed PCET reactivity.

Cathodoluminescence imaging of cellular structures labeled with luminescent iridium or rhenium complexes at cryogenic temperatures

We report for the first time the use of two live-cell imaging agents from the group of luminescent transition metal complexes (IRAZOLVE-MITO and REZOLVE-ER) as cathodoluminescent probes. This first experimental demonstration shows the application of both probes for the identification of cellular structures at the nanoscale and near the native state directly in the cryo-scanning electron microscope. This approach can potentially be applied to correlative and multimodal approaches and used to target specific regions within vitrified samples at low electron beam energies.

Acid-base-induced fac → mer isomerization of luminescent iridium(iii) complexes.

Luminescent Ir(C^N)3 complexes (C^N = cyclometalated arylpyridine ligand) exist in the form of two stable isomers with distinct photophysical and electrochemical properties: fac and mer. Herein, we show that fac-Ir(C^N)3 complexes can be converted into the thermodynamically less stable mer forms by a consecutive reaction with first acid and then base. The chemically induced isomerization is fast, quantitative, and stereoselective, and it can be inversed by light. The new isomerization process opens the possibility to use highly luminescent Ir(C^N)3 complexes as molecular switches.

Iridium complexes having phosphrous ligands of large steric hindrance: Photophysical comparison between solution state, solid state and electrospun fibers

In this work, two luminescent iridium complexes were synthesized with 2-phenyl pyridine (ppy) and two phosphine-based ligands. There was large steric hindrance in these phosphine-based ligands, so that there would be enough time for the excited state based on highly luminescent 3MLLCT (metal-to-ligand-ligand-charge-transfer) electronic transformation It was found that there was limited aggregation-induced phosphorescence emission (AIPE) effect in these two iridium complexes since their ligands with large steric hindrance minimized the intermolecular (π-π) interactions between molecules. A distorted octahedral coordination field was found in the single crystals of these two iridium complexes. A systematical comparison on the photophysical parameters of these two iridium complexes in solution state, in solid state and in electrospun fiber was carried out, so that the limited aggregation-caused quenching (ACQ) could be confirmed. The potential surface crossing and energy transfer in the excited state of these two iridium complexes were analyzed by means of density functional theory calculation and temperature-dependent emission spectra. The limited ACQ effect in these two iridium complexes was finally confirmed due to the large ligand steric hindrance, which allowed phosphorescent decay of excited state. High emission quantum yield (0.27) and long emissive lifetime (0.43 µs) in solid state were observed.

Long-lived cyclometallated iridium complexes: Synthesis, structure, DFT and photocatalytic aspects

Herein, we present synthesis, photophysical and photocatalytic aspects of four mononuclear luminescent iridium complexes ([1]+ - [4]+) incorporating cyclometallating benzimidazole-based (L1 and L2) and phenanthroline pyrazine-based (L3 – L5) ligands. Several analytical techniques were used for the characterization of the complexes. The structural elucidation of [2]+ and [3]+ using x-ray crystallography show a distorted Oh structure with nitrogen donors of cyclometallating ligands are in trans-direction. All the complexes are emitting in the orange-red region and displayed moderately good quantum yield. The complexes have also shown moderately good luminescence lifetime (τ = 80–1900 ns) in different solvents. Importantly, the complex [3]+ exhibits comparative longer emission lifetime (τ = 1800 ns) in water at ambient condition. The complexes have shown moderately good photocatalytic efficacy towards azo dyes degradation in aqueous solution. The mechanistic investigation suggests that in the presence of light in aqueous medium the complexes induce the generation of (HO•/(O2•−) species which is responsible for the photodecomposition of azo dyes.

Two Luminescent Iridium Complexes with Phosphorous Ligands and Their Photophysical Comparison in Solution, Solid and Electrospun Fibers: Decreased Aggregation-Caused Emission Quenching by Steric Hindrance

In this paper, we prepared two phosphorescent Ir complexes with ligands of 2-phenyl pyridine (ppy), and two phosphorous ligands with large steric hindrance, hoping to allow enough time for the transformation of the highly phosphorescent 3MLLCT (metal-to-ligand-ligand-charge-transfer) excited state. Their large steric hindrance minimized the π-π interaction between complex molecules, so that the aggregation-induced phosphorescence emission (AIPE) influence could be minimized. Their single crystals indicated that they took a distorted octahedral coordination mode. Photophysical comparison between these Ir complexes in solution, in the solid state and in electrospun fibers was performed to confirm the realization of limited aggregation-caused quenching (ACQ). The potential surface crossing and energy transfer from 3MLBPECT/3MLBPELppyCT to 3MLppyCT in these Ir complexes were revealed by density functional theory calculation and temperature-dependent emission. It was confirmed that these two phosphorous ligands offered large steric hindrance, which decreased the ACQ effect, allowing the efficient emissive decay of the 3MLppyCT excited state. This is one of the several luminescent Ir complexes with a high emission yield (Φ = 0.27) and long emission lifetime (0.43 μs) in the solid state.

Spectroscopic and Theoretical Investigation of Color Tuning in Deep-Red Luminescent Iridium(III) Complexes.

A series of heteroleptic, neutral iridium(III) complexes of the form [Ir(L)2(N^O)] (where L = cyclometalated 2,3-disubstituted quinoxaline and N^O = ancillary picolinate or pyrazinoate) are described in terms of their synthesis and spectroscopic properties, with supporting computational analyses providing additional insight into the electronic properties. The 10 [Ir(L)2(N^O)] complexes were characterized using a range of analytical techniques (including 1H, 13C, and 19F NMR and IR spectroscopies and mass spectrometry). One of the examples was structurally characterized using X-ray diffraction. The redox properties were determined using cyclic voltammetry, and the electronic properties were investigated using UV–vis, time-resolved luminescence, and transient absorption spectroscopies. The complexes are phosphorescent in the red region of the visible spectrum (λem = 633–680 nm), with lifetimes typically of hundreds of nanoseconds and quantum yields ca. 5% in aerated chloroform. A combination of spectroscopic and computational analyses suggests that the long-wavelength absorption and emission properties of these complexes are strongly characterized by a combination of spin-forbidden metal-to-ligand charge-transfer and quinoxaline-centered transitions. The emission wavelength in these complexes can thus be controlled in two ways: first, substitution of the cyclometalating quinoxaline ligand can perturb both the highest occupied molecular orbital (HOMO) and lowest unoccupied molecular orbital levels (LUMO, Cl atoms on the ligand induce the largest bathochromic shift), and second, the choice of the ancillary ligand can influence the HOMO energy (pyrazinoate stabilizes the HOMO, inducing hypsochromic shifts).

Luminescent complexes of iridium(iii) with aliphatic amines and detection of biogenic amines

The straightforward reaction of [Ir2(ppy)4(µ-Cl)2] with an excess of aliphatic amines yields luminescent iridium complexes of general formula [IrCl(ppy)2(amine)] [amine = n-octylamine (1), t-butylamine (2), piperidine (3)]. The higher sterical hindrance of the amine in complex 2 was the responsible of its equilibrium with the starting materials. The luminescence of 1 and 3 has been studied showing emission at 508 and 509 nm respectively. As the aliphatic amines can be considered models of biogenic amines, this luminescence has been used to explore the viability of this reaction in the detection of biogenic amines. The exposition to vapors of biogenic amines of a solution of [Ir2(ppy)4(µ-Cl)2] in CH2Cl2 or in different solid supports, showed that it was possible to detect the amines in a quick and easy way, with limits of detection value (in solution of methylene chloride) of 4.8 µM for cadaverine.

Targeted cell imaging properties of a deep red luminescent iridium(iii) complex conjugated with a c-Myc signal peptide.

A nuclear localisation sequence (NLS) peptide, PAAKRVKLD, derived from the human c-Myc regulator gene, has been functionalised with a long wavelength (λ ex = 550 nm; λ em = 677 nm) cyclometalated organometallic iridium(iii) complex to give the conjugate Ir-CMYC. Confocal fluorescence microscopy studies on human fibroblast cells imaged after 18-24 h incubation show that Ir-CMYC concentrations of 80-100 μM promote good cell uptake and nuclear localisation, which was confirmed though co-localisation studies using Hoechst 33342. In comparison, a structurally related, photophysically analogous iridium(iii) complex lacking the peptide sequence, Ir-PYR, showed very different biological behaviour, with no evidence of nuclear, lysosomal or autophagic vesicle localisation and significantly increased toxicity to the cells at concentrations >10 μM that induced mitochondrial dysfunction. Supporting UV-visible and circular dichroism spectroscopic studies show that Ir-PYR and Ir-CMYC display similarly low affinities for DNA (ca. 103 M-1), consistent with electrostatic binding. Therefore the translocation and nuclear uptake properties of Ir-CMYC are attributed to the presence of the PAAKRVKLD nuclear localisation sequence in this complex.

Highly Selective Aluminum(III) Ion Sensing with Luminescent Iridium(III) Complexes Bearing a Distorted 2,2'-Bipyridine-3,3'-diol Moiety Utilizing a Rigidified Seven-Membered Chelate Ring.

To create an ion sensor utilizing a rigidified seven-membered chelate ring, we developed two Ir(III) complexes with 2,2'-bipyridine-3,3'-diol (bpy(OH)2, bpydL) ligands as reaction centers, namely Ir1 ([Ir(ppy)2{bpy(O-)(OH)}], ppy = 2-phenylpyridine) and Ir2 ([Ir(bzq)2{bpy(O-)(OH)}], bzq = benzo[h]quinoline), and evaluated their reactivities toward metal ions by spectrophotometry. When they are reacted with Al3+, these complexes exhibit dramatic enhancements in emission intensity (775-fold for Ir1 and 51.0-fold for Ir2) and distinct orange to green changes in emission color. The reactions of Ir1 and Ir2 with Al3+ were found to barely be affected by nearly all common metal ions. We conclude that these high selectivities arise from the high affinities of the (O,O) atoms in bpydL for hard metal ions and the increased strain of the seven-membered chelate ring due to the coordination of bpydL to the Ir(III) center in each complex, which excludes large metal ions out of the chelate ring.

Catalyst-Controlled Regiodivergent C–H Arylation Site of Fluorinated 2-Arylpyridine Derivatives: Application to Luminescent Iridium(III) Complexes

Regiodivergent C–H bond arylation of fluorinated 2-arylpyridines and 2-arylquinolines has been developed. The use of Pd catalyst allows functionalization of the C–H bond of the aryl flanked by two fluorine atoms (most acidic position), while using Ru catalyst, the arylation takes place on the aryl unit at the ortho position of the heterocycle. Both reaction conditions exhibit a good functional group tolerance. The synthetically useful selectivity observed with Pd catalyst was applied to design C^N ligands for the preparation of luminescent cationic iridium(III) complexes. The influence of the incorporated aryl group onto the fluorinated phenyl unit and the fluorine position patterns on the photophysical properties is described.

Iridium-based probe for luminescent nitric oxide monitoring in live cells

Nitric oxide (NO) is an intra- and extracellular messenger with important functions during human physiology process. A long-lived luminescent iridium(III) complex probe 1 has been designed and synthesized for the monitoring of NO controllably released from sodium nitroprusside (SNP). Probe 1 displayed a 15-fold switch-on luminescence in the presence of SNP at 580 nm. The probe exhibited a linear response towards SNP between 5 to 25 μM with detection limit at 0.18 μM. Importantly, the luminescent switch-on detection of NO in HeLa cells was demonstrated. Overall, complex 1 has the potential to be applied for NO tracing in complicated cellular environment.

Synthesis, structure, photo- and electroluminescent properties of bis(2-phenylpyridinato-N,c2′)[2-(2′-tosylaminophenyl)benzoxazolato-N,N′]iridium(III)

A new highly luminescent iridium complex, bis(2-phenylpyridine-N,C2′)[2-(2′-tosylaminophenyl)benzoxazole-N,N′]iridium(III) [Ir(ppy)2(TAPBO)] (TAPBO- 2-(2′-tosylaminophenyl)benzoxazole) has been synthesized and its molecular structure determined using single crystal X-ray diffraction analysis. The Ir(III) complex displayed intense yellow photophosphorescence emission that manifested its potential for organic electroluminescence. Photo- and electroluminescent properties of the OLEDs fabricated on the basis of [Ir(ppy)2(TAPBO)] with doping concentration of Ir(ppy)2(TAPBO) varied from 3 to 20 wt% have been investigated. The OLED with 9 wt% Ir(ppy)2(TAPBO) exhibited maximum luminance of 9000 cd m−2 at 180 mA/cm−2 and had sufficiently low turn-on voltage of ca. 6 V.

Synthesis, Characterization, and Optoelectronic Properties of Iridium Complexes Bearing Nonconjugated Six-Membered Chelating Ligands.

We report the synthesis, characterization, and optoelectronic properties of a series of four new luminescent iridium(III) complexes, 1-4, of the form [Ir(C∧N)2(N∧N)]PF6 (where C∧N is the nonconjugated benzylpyridinato (bnpy) and N∧N is a neutral diimine ancillary ligand) with the goal of investigating the effect of the methylene spacer between the coordination moieties of the C∧N ligand on the optoelectronic properties of the complexes. The crystal structures of 1-3 illustrate two possible orientations of the methylene unit of the bnpy ligand. The formation of these two separate conformers is a result of the conformational flexibility of the bnpy ligand. In complexes 3 and 4, mixtures of the two conformers were observed by 1H NMR spectroscopy in CDCl3 at room temperature, whereas only a single conformer is detected for 1 and 2. Detailed DFT calculations corroborate NMR experiments, accounting for the presence and relative populations of the two conformers. The optoelectronic properties of all four complexes, rationalized by the theoretical study, demonstrate that the interruption of conjugation in the C∧N ligands results in a reduced electrochemical gap but similar triplet state energies and lower photoluminescence quantum yields in comparison to the reference complexes R1-R4. Depending on the nature of the N∧N ligand, we observe (1) marked variations of the ratio of the conformers at ambient temperature and (2) phosphorescence ranging from yellow to red.
