Association of Omega-3 Polyunsaturated Fatty Acids with Sarcopenia in Liver Cirrhosis Patients with Hepatocellular Carcinoma.

Sarcopenia is associated with the prognosis of patients with liver cirrhosis and hepatocellular carcinoma (HCC). Given their diverse physiological activities, we hypothesized that plasma fatty acids might influence the progression of sarcopenia. This study aimed to clarify the association between fatty acids and sarcopenia in cirrhotic patients with HCC. In this single-center retrospective study, we registered 516 cases and analyzed 414 cases of liver cirrhosis and HCC. The skeletal muscle mass index was measured using a transverse computed tomography scan image at the third lumbar vertebra. The cutoff value for sarcopenia followed the criteria set by the Japan Society of Hepatology. Fatty acid concentrations were measured by gas chromatography. Fatty acid levels, particularly omega-3 (n-3) polyunsaturated fatty acid (PUFA), were lower in patients with poor liver function (Child-Pugh grade B/C) and were negatively correlated with the albumin-bilirubin score (p<0.0001). The prognosis of HCC patients with low PUFA levels was significantly worse. Among the different fatty acid fractions, only n-3 PUFAs significantly correlated with skeletal muscle mass index (p=0.0026). In the multivariate analysis, the n-3 PUFA level was an independent variable associated with sarcopenia (p=0.0006). A low level of n-3 PUFAs was associated with sarcopenia in patients with liver cirrhosis and HCC.

Fatty acid composition and biophysical characteristics of the cell membrane of feline spermatozoa

Sperm membrane composition and biophysical characteristics play a pivotal role in many physiological processes (i.e. sperm motility, capacitation, acrosome reaction and fusion with the oocyte) as well as in semen processing (e.g. cryopreservation). The aim of this study was to characterize the fatty acid content and biophysical characteristics (anisotropy, generalized polarization) of the cell membrane of domestic cat spermatozoa. Semen was collected from 34 adult male cats by urethral catheterization. After a basic semen evaluation, the fatty acid content of some of the samples (n = 11) was evaluated by gas chromatography. Samples from other individuals (n = 23) were subjected to biophysical analysis: membrane anisotropy (which is inversely proportional to membrane fluidity) and generalized polarization (describing lipid order); both measured by fluorimetry at three temperature points: 38 °C, 25 °C and 5 °C. Spermatozoa from some samples (n = 10) were cryopreserved in TRIS egg yolk-glycerol extender and underwent the same biophysical analysis after thawing. Most fatty acids in feline spermatozoa were saturated (69.76 ± 24.45%), whereas the polyunsaturated fatty acid (PUFA) content was relatively low (6.12 ± 5.80%). Lowering the temperature caused a significant decrease in membrane fluidity and an increase in generalized polarization in fresh spermatozoa, and these effects were even more pronounced following cryopreservation. Anisotropy at 38 °C in fresh samples showed strong positive correlations with viability and motility parameters after thawing. In summary, feline spermatozoa are characterized by a very low PUFA content and a low ratio of unsaturated:saturated fatty acids, which may contribute to low oxidative stress. Cryopreservation alters the structure of the sperm membrane, increasing the fluidity of the hydrophobic portion of the bilayer and the lipid order in the hydrophilic portion. Because lower membrane fluidity in fresh semen was linked with better viability and motility after cryopreservation, this parameter may be considered an important factor in determination of sperm cryoresistance.

Comparative Analysis of Fatty Acids Concentration in Liver and Muscle Tissues of Rats and Mice

This study conducted a comparative analysis of fatty acids (FAs) concentration derived from polar (PL) and non-polar (NPL) lipids in the liver and muscle tissues of rats and mice. The objective was to elucidate species-specific differences in tissue FA distribution. Employing targeted GC/MS-based methodology, the study aimed to provide insights into conserved and species-specific aspects of lipid metabolism, thereby enhancing future experimental design, linked with comprehension of the interactions between diet, metabolism, and health. Results revealed markedly higher levels of total fatty acids (TFAs) derived from PL in mice skeletal muscle compared to rats and elevated saturated fatty acids (SFAs) levels in mice. Unsaturated fatty acid levels, mainly monounsaturated fatty acids (MUFAs) and polyunsaturated fatty acids (PUFAs), were substantially higher in mice muscle across all lipid classes, resulting in a higher PUFA/TFA ratio in mice muscle. Detailed analysis of specific unsaturated fatty acids (C16:1, C18:1n9c, C18:2n6c, C22:6n3) indicated elevated levels in mice relative to rats. Conversely, rats exhibited higher SFA levels derived from the NPL fraction in the liver, particularly in myristic (C14:0), stearic (C18:0), and tricosanoic (C23:0) acids. Mice liver PL fractions displayed significantly elevated unsaturated FA levels, with notably higher MUFAs and lower PUFAs in NPL fractions compared to rats. Analysis of specific unsaturated FAs revealed higher levels of palmitoleic acid (C16:1) in mice, while rats exhibited increased linoleic (C18:2n6c) and linolenic (C18:3n3) acids. In conclusion, significant differences in FAs tissue distribution between rats and mice underscore the importance of considering species-specific FAs variations when utilizing these animal models and interpreting experimental results related to FA metabolism.

Investigation of Crypthecodinium cohnii High-Cell-Density Fed-Batch Cultivations

Crypthecodinium cohnii is a marine microalga that can accumulate high amounts of polyunsaturated fatty acids (PUFAs) and thus replace conventional routes of fish oil production. They are associated with the destruction of marine resources and multiple downstream/purification complications. The major drawbacks of using C. cohnii for industrial-scale production are associated with low PUFA productivity. One of the means of increasing the PUFA synthesis rate is to maintain the medium component concentrations at optimal values throughout cultivation, thus increasing PUFA production efficiency, which can result in the successful transfer of the process to pilot and/or industrial scale. The goal of the present research was to develop techniques for increasing the efficiency of PUFA production via C. cohnii cultivation. Multiple experiments were carried out to test and fine-tune the cultivation medium composition and oxygen transfer factors. The biomass yields from individual components, yeast extract, sea salts, and glucose amounted to 5.5, 0.65, and 0.61 g·g−1, respectively. C. cohnii cell susceptibility to mechanical damage was experimentally evaluated. Power inputs of &lt;276.5 W/m3 did not seem to promote cell destruction when Pitched-blade impellers were used. The obtained cultivation conditions were shown to be efficient in terms of increasing the biomass productivity and the omega-3 fatty acid content in C. cohnii. By using the applied methods, the maximal biomass productivity reached 8.0 g·L−1·day−1, while the highest obtained biomass concentration reached 110 g·L−1. A steady increase in the concentration of PUFAs during cultivation was observed from the FTIR data.

Women with Gestational Diabetes Mellitus do not have compromised polyunsaturated fatty acid status

Pregnant women who develop pre-eclampsia (PE) and/or intra-uterine growth restriction (IUGR) have reduced polyunsaturated fatty acid (PUFA) status compared to healthy pregnancy(1). It is unknown if pregnant women diagnosed with Gestational Diabetes Mellitus (GDM), and their offspring, also have compromised PUFA status. To determine if women with GDM, and their offspring, have altered PUFA status compared to healthy pregnancy. Pregnant women were recruited from Glasgow Scotland, and Brisbane, Australia from antenatal clinics for this cross sectional study. Third trimester maternal blood samples were collected after an overnight fast and cord blood samples were collected at delivery. Plasma fatty acids were analysed using gas chromatography from women with GDM (n = 37) and healthy pregnancies (n = 27) and their respective offspring (n = 31, from women with GDM, and n = 27 from healthy women). T-tests were used to determine significant differences between maternal with GDM and healthy pregnancy, as well as for their offspring and significance was set at p&lt;0.05. Previously, erythrocyte fatty acids were analysed from women with PE (n = 21), IUGR (n = 13) and healthy pregnancies (n = 86)(1). All results were expressed as mol percent of total fatty acids. There were no differences in maternal plasma arachidonic acid (4.51 ± 1.23 vs. 4.72 ± 0.64, p = 0.39) and plasma EPA &amp; DHA (2.33 ± 0.74 vs 2.69 ± 1.04, p = 0.14) in women with GDM and healthy pregnancies, respectively. There were no differences in fetal plasma arachidonic acid (11.58 ± 2.26 vs. 12.63 ± 1.69, p = 0.08) and plasma EPA &amp; DHA (4.44 ± 1.17 vs. 4.44 ± 1.00, p = 0.89) in offspring from women with GDM and healthy pregnancies, respectively. Women with PE and IUGR had approximately 25% lower erythrocyte EPA &amp; DHA and 35% lower erythrocyte arachidonic acid compared to healthy pregnant(1). Offspring from women with PE and IUGR had approximately 25% lower erythrocyte EPA &amp; DHA and 22% lower erythrocyte arachidonic acid compared to healthy pregnancy(1).Women with PE and IUGR had lower PUFA status likely due to reduced PUFA synthesis(1) and offspring from women with PE and IUGR had reduced PUFA status likely due to ectopic fat in placenta tissue(2). Women with GDM do not have compromised PUFA status suggesting there is no reduced synthesis and transport of PUFA. Offspring from women with GDM do not have reduced PUFA status suggesting there is no problem with PUFA transport across the placenta, unlike offspring from women with PE and/or IUGR. Women with GDM, and their offspring, do not have compromised plasma PUFA status compared to healthy pregnancy.

Fatty acid profiles and stable isotope composition of Antarctic macroalgae: a baseline for a combined biomarker approach in food web studies

AbstractThe Western Antarctic Peninsula supports a diverse assemblage of &gt; 100 described macroalgal species that contribute to the base of coastal food webs, but their contribution to local nearshore food webs is still uncertain across larger spatial scales. The analysis of biomarkers, specifically fatty acids and stable isotopes, offers a tool to clarify the trophic role of Antarctic macroalgae. The aim of this study was to describe the fatty acid profiles and stable isotope values of 31 algal species from three divisions (Chlorophyta—1, Ochrophyta—8, Rhodophyta—22) collected at the same sites for both biomarkers. Of these, 13 species had no previously published fatty acid profiles. Most species were rich in polyunsaturated fatty acids (PUFAs), suggesting they are highly nutritious and could be a resource of essential fatty acids for consumers. This was specifically noticeable in the overall low PUFA ratio (∑ω6/∑ω3 ratio), with low ratios being an indicator of high nutritional quality for consumers. Fatty acid profiles of macroalgae grouped strongly by phylogeny (at the levels of division, order, and family), while stable isotope groupings were more driven by the physiological properties of the species. Specifically, some closely related red algal species exhibited very different stable isotope values based on their carbon concentrating mechanisms, with highly 13C-depleted values in several Rhodophyta species. The fact that the two biomarker approaches created different groupings of Antarctic macroalgae collected at the same locations emphasizes that their combined application can be a powerful tool in Antarctic coastal food web studies.

Land use alters cross-ecosystem transfer of high value fatty acids by aquatic insects

BackgroundMany aquatic insects emerge as adults from water bodies to complete parts of their life cycle in terrestrial ecosystems and are potential prey for riparian predators. The benefits of riparian predators from aquatic insects include higher contents of long-chain polyunsaturated fatty acids (PUFA) compared to terrestrial insects. Aquatic insects are therefore considered a high-quality food. Food containing high levels of PUFA can enhance growth and immune response of spiders. However, agricultural stressors like nutrient increase, pesticides and habitat degradation can affect the biomass of aquatic insects and in turn the diet of spiders. Studies quantifying the influence of land use on fatty acid (FA) profiles of emergent aquatic insects and riparian predators are lacking. We quantified differences in exports of FA, saturated FA, monounsaturated FA, and PUFA, FA profiles of aquatic insects and spiders between forested and agricultural sites over the primary emergence period within one year. The FA export to the riparian food web is crucial to understand energy fluxes between ecosystems. Furthermore, we monitored environmental variables to identify associations between agricultural stressors and FA profiles of aquatic insects and spiders.ResultsWe found differences in FA export and profiles of aquatic insects between land-use types. The quantity of total FA export via aquatic insects was lower in agricultural sites (95% CI 1147–1313 µg m−2) in comparison to forested sites (95% CI 1555–1845 µg m−2), while the biomass export was higher in agricultural sites. Additionally, in spring the PUFA export was significantly lower (up to 0.06 µg d−1 m−2) in agricultural than forested sites. Agricultural stressors explained only little variation in the FA profiles of aquatic insects, e.g., 4% for caddisflies and 12% for non-biting midges. Percentage of shading and pool habitats were identified as most important variables explaining the variation in FA profiles.ConclusionThe quality of aquatic insects as food source for riparian spiders was smaller in agricultural than forested sites, which can decrease the fitness of riparian predators. To improve our capacity to predict potential adverse effects in the riparian food web, future studies should identify the mechanisms underlying a lower PUFA content.

Associations of fatty acids with the risk of biliary tract calculus and inflammation: a Mendelian randomization study

BackgroundThe presence of gallstones in both the gallbladder and bile ducts is referred to as cholelithiasis. The prevalence of cholecystolithiasis and bile duct stones differs. Observational and Mendelian randomization (MR) studies have elucidated the significant contributing role of numerous fatty acids (FAs) in the development of cholelithiasis. Despite numerous studies about cholelithiasis, evidence on the relationship between serum FA levels and cholecystolithiasis, as well as bile duct stones with or without inflammation, remains insufficient.MethodsA two-sample MR study was designed to clarify the impact of serum FA levels on various bile duct inflammatory diseases. The summary statistics of single nucleotide polymorphisms (SNPs) associated with fatty acids were obtained from the UK Biobank (UKB) and included data from 114,999 participants. The researchers obtained GWAS summary statistics for cholecystolithiasis and bile duct stones in 463,010 and 361,194 European participants, including cases with and without inflammation. No sample overlap between the exposure and outcome was verified through the “mr-lap” package. The SNPs were screened to identify instrumental variables (IVs). Cochran’s Q test was applied for heterogeneity assessment. Inverse variance weighting (IVW) (fixed effects or random effects), MR-Egger regression and weighted median methods were used for MR. Multivariable MR was applied to determine the direct effect of each exposure on the outcome. A false discovery rate (FDR) was applied to adjust for multiple testing correction based on the Benjamini-Hochberg method. Finally, the FinnGen Consortium was used to validate some results.ResultsThe overall concentration of polyunsaturated fatty acids (PUFAs) in the serum was negatively associated with the risk of calculus of the gallbladder with acute cholecystitis (IVW, OR = 0.996, P = 0.038, CI 0.992–0.999; weighted median, OR = 0.995, P = 0.025, CI 0.991–0.999). The percentage of PUFAs to total monounsaturated fatty acids(MUFAs) (IVW, OR = 0.998, P = 0.045, CI 0.997–0.999) and the percentage of PUFAs to total FAs (IVW, OR = 0.997, P = 0.025, CI 0.995–0.999) had a protective role against cholecystitis. The percentage of PUFAs to total FAs had a protective role against calculus of the gallbladder without cholecystitis (IVW, OR = 0.995, P = 0.026, CI 0.990–0.999; MR Egger, OR = 0.99, P = 0.03, CI 0.982–0.998; weighted median, OR = 0.991, P = 5.41e-06, CI 0.988–0.995). Conversely, the percentage of MUFAs to total FAs increased the risk for cholecystitis (IVW, OR = 1.001, P = 0.034, CI 1.0001–1.002). However, there were no causal effects of the above exposures on the outcomes through multivariable MR and multiple testing correction. Finally, the causal effects of the above exposures on cholecystitis were validated in the FinnGen Consortium, which suggested that the percentage of PUFAs to total FAs (IVW, OR = 0.744, P = 0.021, CI 0.579–0.957) had a protective role against cholecystitis.ConclusionThese Mendelian randomization findings suggested that more attention should be focused on people who have low serum PUFA levels, which may have a potential role in the occurrence of calculus of the gallbladder or cholecystitis rather than calculus of the bile duct without cholangitis or cholecystitis.

Fatty acid profiling of almond germpalsm grown in the Western Himalayan region of India

Aim: Nutritional profiling of almond genetic resources for their utilization and further use in the breeding programme. Methodology: The nuts of 32 almond genotypes were collected from the experimental farm of ICAR-CITH, Srinagar. Using a Soxhlet fat apparatus and 100ml of petroleum ether as a solvent, almond oil was extracted from 5 g of ground kernel. The fatty acid content of almond oil was analyzed by GC 30 (Perkin Elmer Autosystem XL) equipped with a CP-Wax 52 CB column (Varian Inc.) (50 x 0.25 mm, 0.2 μm). Results: The total oil content in these genotypes ranged between 41.82% (CITH-A-21) and 51.86% (Waris). The major fatty acids found were oleic acid (53.70-80.81%), linoleic acid (11.63-36.88%), and palmitic acid (5.19-8.13%). Stearic acid (1.01-2.53%) was estimated at a lower proportion, while α-linolenic acid was found in trace amounts (0.05-0.18%). The oleic: linoleic acid ratio varied from 1.46 (CITH-A-21) to 6.95 (CITH-A-06), conversely the highest (115.42) and lowest (93.96) iodine values were recorded in CITH-A-21 and CITH-A-06, respectively. The lowest content of saturated fatty acids (6.32%) and the highest content of unsaturated fatty acids (93.69%) were found in CITH-A-17 whereas contrasting results were observed in Nonpareil. The highest monounsaturated fatty acids (80.81%) and lowest polyunsaturated fatty acids (11.71%) were found in CITH-A-6. Oleic acid content was negatively correlated with linoleic acid (r = -1.00), palmitic acid (r = -0.67) and stearic acid (r = -0.68). Principal component analysis indicated that among the fatty acids, the oleic acid, linoleic acid, and Oleic/Linoleic ratio were largely responsible for the separation on the PC 1. Among the different almond genotypes, CITH-A-17 and CITH-A-06 were found to be promising in terms of fatty acid composition. Interpretation: The present study identified the genotypes with a high Oleic/Linoleic acid ratio, which is important for crop improvement programme and commercialization of these genotypes. Further, the present study also confirms that the Kashmir region is suitable for growing almonds with fatty acids composition at par with commercial almond cultivars grown in major almond-growing countries in the world. Role of Polyunsaturated Fat in Modifying Cardiovascular Risk Associated With Family History of Cardiovascular Disease: Pooled De Novo Results From 15 Observational Studies.

It is unknown whether dietary intake of polyunsaturated fatty acids (PUFA) modifies the cardiovascular disease (CVD) risk associated with a family history of CVD. We assessed interactions between biomarkers of low PUFA intake and a family history in relation to long-term CVD risk in a large consortium. Blood and tissue PUFA data from 40 885 CVD-free adults were assessed. PUFA levels ≤25th percentile were considered to reflect low intake of linoleic, alpha-linolenic, and eicosapentaenoic/docosahexaenoic acids (EPA/DHA). Family history was defined as having ≥1 first-degree relative who experienced a CVD event. Relative risks with 95% CI of CVD were estimated using Cox regression and meta-analyzed. Interactions were assessed by analyzing product terms and calculating relative excess risk due to interaction. After multivariable adjustments, a significant interaction between low EPA/DHA and family history was observed (product term pooled RR, 1.09 [95% CI, 1.02-1.16]; P=0.01). The pooled relative risk of CVD associated with the combined exposure to low EPA/DHA, and family history was 1.41 (95% CI, 1.30-1.54), whereas it was 1.25 (95% CI, 1.16-1.33) for family history alone and 1.06 (95% CI, 0.98-1.14) for EPA/DHA alone, compared with those with neither exposure. The relative excess risk due to interaction results indicated no interactions. A significant interaction between biomarkers of low EPA/DHA intake, but not the other PUFA, and a family history was observed. This novel finding might suggest a need to emphasize the benefit of consuming oily fish for individuals with a family history of CVD.

Liver and Plasma Fatty Acid Characterization in Cultured Brown Trout at Distinct Reproductive Stages.

Fatty acids are energy sources, and their profiles are used as biomarkers of metabolic status and physiological changes in fish. Within this context, the main aim of this study was to identify the fatty acids that best discriminate the reproductive status of male and female farmed brown trout. The fatty acid composition in liver and plasma samples from the adults of both sexes was monitored along four distinct reproductive stages, namely the spawning capable (December), regressing (March), regenerating (July), and developing (November) stages. Irrespective of the sex and stage, the most representative fatty acids were palmitic acid (16:0), stearic acid (18:0), oleic acid (18:1 n-9), arachidonic acid (20:4 n-6), eicosapentaenoic acid (EPA, 20:5 n-3), and docosahexaenoic acid (DHA, 22:6 n-3). There were no significant sex differences in fatty acid classes in the liver and plasma. Despite this, there were several changes in individual fatty acid levels between the sexes. In the liver, both males and females showed high monounsaturated fatty acid and low polyunsaturated fatty acid (PUFA) levels during the regressing and regenerating stages. At spawning capable and developing stages, a reverse profile was noted. The plasma profiles were mainly influenced by changes in saturated fatty acids and PUFAs in males and by PUFA in females. Based on the most representative fatty acids, four patterns were established for female plasma samples, one for each reproductive stage. This scenario suggests that female plasma samples are promising for the discrimination of gonadal reproductive status, and this potential can be further explored in aquaculture and environmental monitoring studies.

Inclusion of Sainfoin in the Concentrate of Finishing Lambs: Fatty Acid Profiles of Rumen, Plasma, and Muscle.

The effects of sainfoin inclusion (Onobrychis viciifolia) in the finishing concentrate for light lambs on the fatty acid (FA) composition of the ruminal digesta, plasma, and meat were evaluated. Twenty-six weaned male lambs were divided into three groups and fed individually ad libitum for 40 days with one of three concentrates differing in the level of sainfoin inclusion: 0% (0SF), 20% (20SF), and 40% (40SF). The rumen digesta showed an increase in C18:3 n-3 concentration and a decrease in C18:1 t10 concentration when sainfoin was included in the concentrate regardless of the level of inclusion. However, the highest C18:1 t11 and the lowest C18:2 n-6 proportions were obtained only in the 40SF rumen, showing a stronger t11 biohydrogenation pathway. In plasma, most effects were associated with changes in the levels of polyunsaturated FA (PUFA) n-3. The meat FA profile of 40SF lambs presented higher percentages of PUFA n-3 and CLA c9,t11 and a lower PUFA n-6/PUFA n-3 ratio compared with those from 0SF and 20SF diets because of the potentiation of the ruminal t11 pathway. Inclusions of 20 and 40% sainfoin both showed beneficial effects on meat quality; furthermore, these effects were most marked in the 40% sainfoin diet.

Selection of candidate varieties of garden egg (Solanum aethiopicum) in an on-station trial using multi-disciplinary approach

The on-station trial was conducted at the National Horticultural Research Institute, Ibadan during the rainy season of the year 2021. Seven improved varieties selected from crosses generated through half-diallel mating design and two local checks were used in this study. The study aimed to select the best candidate varieties for multi-locational trials. The trial was laid out in a Randomized Complete Block Design with three replications. Data were collected on agronomic and yield characters. The nine varieties of garden egg were screened for tolerance to Anthracnose and Tuta absoluta. Fatty acid Methyl Esters were also analyzed and sensory evaluation was done using a twelve-member trained panel of judges. The results showed that there were significant variations among all the agronomic and yield-related characters except for plant height, stem girth and days to flowering. L03 was the best in yield and related characters, followed by L06 and L01. All the seven improved garden egg varieties were tolerant to Athracnose while the highest disease incidence was recorded in YALO. The mining activities of Tuta absoluta were more pronounced in L03 while the least was observed in L07. The improved variety L03 had the highest Saturated Fatty Acids (SFAs) and very low Polyunsaturated Fatty Acids (PUFAs). YALO had the best low SFAs and appreciable levels of PUFAs. This was followed by L06 and L08 with moderate SFAs and high PUFAs. L02 was the best in overall acceptability during sensory evaluation but had very high SFAs. L06 was second best in overall acceptability while L01 was third. However, L03 was the least accepted. Based on the overall assessment, L06, L08 and L01 were selected for multi-locational trials and future release.

Abstract P165: Hypertension Increases Susceptibility To Experimental Malaria In Mice

Despite available treatments, global prevalence of hypertension is on the rise and burdening healthcare, especially in under-developed countries where infectious diseases are also rampant. Previously we reported that hypertension is associated with abnormal red blood cell (RBC) physiology and anemia. Since RBCs are target host cells for malarial pathogen, Plasmodium , we hypothesized that hypertensive patients with abnormal RBC physiology are at an increased risk of susceptibility to Plasmodial infection. Methods: To test this hypothesis, eight weeks old hypertensive BPH/2J and normotensive BPN/3J female mice (n=6) were first characterized for their RBC physiology and subsequently infected with Plasmodium yoelii (P. yoelii) , a murine-specific non-lethal strain. Results: Compared to BPN mice, BPH mice displayed comparable RBC count, but lower hemoglobin (BPH 16.3±0.8, vs BPN 16.5±1.0 mg/dL; p&lt;0.01); lower % hematocrit (BPH 59.6 ±2.4 vs BPN 61.0 ± 2.7; p&lt;0.01), and lower mean corpuscular volume (BPH 52.6±3.13 vs BPN 54.2±1.6 fl; p&lt;0.005), indicating microcystic anemia. Further, RBCs from BPH mice were resistant to osmotic hemolysis in hypotonic (0.42%) NaCl solution (% hemolysis BPH 43.2±1.6% vs 97.3±1.8% BPN; p&lt;0.001). Lower membrane fluidity was observed in the RBCs of BPH mice as noted through higher levels of the saturated fatty acid, stearate (BPH 16.2±0.1% vs BPN 13.9±0.16%; p&lt;0.001), phospholipids (BPH 554±18.3 vs BPN 496±6.2 μg/mg protein p=0.02) and lower polyunsaturated fatty acids like arachidonic acid (BPH 13.7±0.1% vs BPN 15.1±0.5%, BPN; p=0.03). Further, upon infection for 7 days with P. yoelii , BPH mice experienced severe body weight loss compared to minimal change in body weight noted in the BPN mice (BPH Day 0 18.53±0.32 g vs Day 7 16.43±0.71 g vs BPN Day 0 22.5±1.28 g vs Day 7 22.43±1.30 g; p&lt;0.01). This was accompanied by a significantly decreased number of RBC compared to BPN mice (BPH 3.4±1.3х10 12 vs BPN 5.5±1.2х10 12 cells/L; p=0.03), indicating that BPH mice were more susceptible to malaria infection. Conclusion: Collectively, these data demonstrate that aberrant RBC physiology observed in hypertensive mice contributes to an increased susceptibility to experimental malaria.

Environmental drivers alter PUFA content in littoral macroinvertebrate assemblages via changes in richness and abundance

Shallow littoral areas in lakes are productive and highly diverse ecotonal zones, providing habitats for both invertebrate and vertebrate species. We developed a Bayesian modeling framework to elucidate the relationships between environmental drivers (lake typology, habitat, water chemistry, and latitude) and taxon richness, abundance, as well as the content of polyunsaturated fatty acids (PUFAs) in littoral macroinvertebrate communities in 95 boreal lakes. PUFAs, particularly arachidonic acid (ARA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), are critical micronutrients to maintain normal physiological functions in consumers. Lake typology was a significant predictor for PUFA content in the invertebrate assemblages, which was connected to taxon richness and/or abundance. Benthic communities in large humus-poor or nutrient-rich lakes displayed higher abundance, taxon richness, and more PUFA-rich taxa, whereas those in medium- and large-sized humic (color 30–90 mg Pt/L) and humus-rich lakes (color >90 mg Pt/L) were characterized by decreased abundance and subsequently low PUFA content. The abundance, taxon richness, and nutritional quality of the communities were also strongly related to latitude. Lakes with lower pH were characterized by lower benthic invertebrate diversity and low frequency of taxa with high somatic EPA and DHA content. The complexity of littoral habitats dominated by various macrophyte assemblages creates an environment that favors higher benthic abundance and increased presence of taxonomic groups with high PUFA content. Nutritional quality of benthic invertebrates for upper trophic levels can be modulated by a complex interplay between external stressors and abiotic factors that typically shape the structure of littoral benthic communities.

Supplementing branched-chain volatile fatty acids in dual-flow cultures varying in dietary forage and corn oil concentrations. I: Digestibility, microbial protein, and prokaryotic community structure

Branched-chain amino acids are deaminated by amylolytic bacteria to branched-chain volatile fatty acids (BCVFA), which are growth factors for cellulolytic bacteria. Our objective was to determine the dietary conditions that would increase the uptake of BCVFA by rumen bacteria. We hypothesized that increased forage would increase cellulolytic bacterial abundance and incorporation of BCVFA into their structure. Supplemental polyunsaturated fatty acids, supplied via corn oil (CO), should inhibit cellulolytic bacteria growth, but we hypothesized that additional BCVFA would alleviate that inhibition. Further, supplemental BCVFA should increase neutral detergent fiber degradation and efficiency of bacterial protein synthesis more with the high forage and low polyunsaturated fatty acid dietary combination. The study was an incomplete block design with 8 dual-flow continuous cultures used in 4 periods with 8 treatments (n = 4 per treatment) arranged as a 2 × 2 × 2 factorial. The factors were: high forage (HF) or low forage (LF; 67 or 33%), without or with supplemental CO (3% dry matter), and without or with 2.15 mmol/d (which included 5 mg/d of 13C each of BCVFA isovalerate, isobutyrate, and 2-methylbutyrate). The isonitrogenous diets consisted of 33:67 alfalfa:orchardgrass pellet, and was replaced with a concentrate pellet that mainly consisted of ground corn, soybean meal, and soybean hulls for the LF diet. The main effect of supplementing BCVFA increased neutral detergent fiber (NDF) degradability by 7.6%, and CO increased NDF degradability only in LF diets. Supplemental BCVFA increased bacterial N by 1.5 g/kg organic matter truly degraded (6.6%) and 0.05 g/g truly degraded N (6.5%). The relative sequence abundance decreased with LF for Fibrobacter succinogenes, Ruminococcus flavefaciens, and genus Butyrivibrio compared with HF. Recovery of the total 13C dose in bacterial pellets decreased from 144 µg/ mg with HF to 98.9 µg/ mg with LF. Although isotope recovery in bacteria was greater with HF, BCVFA supplementation increased NDF degradability and efficiency of microbial protein synthesis under all dietary conditions. Therefore, supplemental BCVFA has potential to improve feed efficiency in dairy cows even with dietary conditions that might otherwise inhibit cellulolytic bacteria.

Fatty acid desaturase 2 determines the lipidomic landscape and steroidogenic function of the adrenal gland.

Corticosteroids regulate vital processes, including stress responses, systemic metabolism, and blood pressure. Here, we show that corticosteroid synthesis is related to the polyunsaturated fatty acid (PUFA) content of mitochondrial phospholipids in adrenocortical cells. Inhibition of the rate-limiting enzyme of PUFA synthesis, fatty acid desaturase 2 (FADS2), leads to perturbations in the mitochondrial lipidome and diminishes steroidogenesis. Consistently, the adrenocortical mitochondria of Fads2-/- mice fed a diet with low PUFA concentration are structurally impaired and corticoid levels are decreased. On the contrary, FADS2 expression is elevated in the adrenal cortex of obese mice, and plasma corticosterone is increased, which can be counteracted by dietary supplementation with the FADS2 inhibitor SC-26192 or icosapent ethyl, an eicosapentaenoic acid ethyl ester. In humans, FADS2 expression is elevated in aldosterone-producing adenomas compared to non-active adenomas or nontumorous adrenocortical tissue and correlates with expression of steroidogenic genes. Our data demonstrate that FADS2-mediated PUFA synthesis determines adrenocortical steroidogenesis in health and disease.

FAAH Pro129Thr Variant Is Associated with Increased Cholesterol Levels in Normal-Weight Metabolically Unhealthy Subjects.

Introduction: The endocannabinoid system (ECS) plays an integral role in maintaining metabolic homeostasis, where an hyperactivation has been related with serum lipid alterations. The biological effects of ECS are limited by the activation of the endocannabinoid-degrading enzyme fatty acid amide hydrolase (FAAH) and by polyunsaturated fatty acid (PUFA) intake as precursors. The FAAH Pro129Thr variant has been associated with obesity in some populations. However, the association with metabolic phenotypes in the Mexican population has not been studied. This study aimed to analyze the association of the FAAH Pro129Thr variant with serum lipids and diet in Mexican adults with different metabolic phenotypes. Methods: This is a cross-sectional study with 306 subjects between 18 and 65 years of age. They were classified with normal weight (NW) or excess weight (EW) according to their body mass index (BMI). The EW group included individuals with overweight or obesity (BMI 25-39.9 kg/m2). The individuals were classified into two metabolic phenotypes, metabolically healthy and metabolically unhealthy (MUH), using the homeostatic model assessment of insulin resistance and the National Cholesterol Education Program-adenosine triphosphate III cutoff points for blood pressure, triglycerides, high-density lipoprotein cholesterol, and fasting glucose. Subjects with ≥2 of 5 altered parameters were classified as MUH. The FAAH Pro129Thr variant was determined by allelic discrimination with TaqMan® probes. Results: The total cholesterol and very low-density lipoprotein cholesterol levels were associated with the FAAH Pro129Thr variant in NW-MUH subjects. Moreover, a lower PUFA intake was found in EW-MUH subjects with the FAAH variant. Conclusions: FAAH Pro129Thr variant has an important role in lipid metabolism, especially in NW-MUH subjects. By contrast, a low dietary intake of endocannabinoid PUFA precursors may partly counteract the development of the altered lipid profile associated with overweight/obesity.

Biochemical and gene expression differences associated with higher fat deposition in Krškopolje pigs in comparison with lean hybrid pigs

Krškopolje pig is the only preserved local pig breed in Slovenia. Compared to lean pig genotypes, it is characterised by greater capacity for lipid deposition. The underlying mechanisms responsible for a greater fat deposition in this breed have not yet been investigated. Therefore, the main objective of the present study was to determine differences in gene expression and lipid metabolism between Krškopolje pigs and lean hybrid pigs reared in the same environmental conditions. Despite a greater fat deposition, activities of lipogenic enzymes were lower in Krškopolje than in lean hybrid pigs (i.e. malic enzyme and glucose-6-phosphate dehydrogenase displayed 2.6- and 2.0-fold lower activity measured on tissue postmortem, respectively). Krškopolje pigs had also lower polyunsaturated fatty acid (PUFA) and higher saturated (SFA) and monounsaturated fatty acids (MUFA) content than their lean-type peers. Consistent with a higher MUFA, overexpression of the stearoyl-CoA desaturase gene was determined in Krškopolje pigs compared to the lean hybrid pigs. In accordance with higher adiposity, Krškopolje pigs exhibited higher expression of genes involved in lipogenesis (ACACA, FASN), adipogenesis (PPARγ) and energy homeostasis (LEP) than lean hybrid pigs. Regarding the sex effect in Krškopolje pigs, females exhibited higher lipogenic enzyme activities than surgically castrated males, while no significant differences were observed for fat quantity and quality traits or gene expression. In conclusion, Krškopolje pigs are characterised by a higher fat deposition associated with higher SFA and MUFA content, a higher expression of genes involved in lipogenesis, and lower activity of lipogenic enzymes compared to lean-type hybrid pigs.

Dietary polyunsaturated fatty acids intake, air pollution, and the risk of lung cancer: A prospective study in UK biobank

BackgroundEpidemiological evidence on the association between specific types of polyunsaturated fatty acids (PUFAs) intake and lung cancer risk is limited. However, whether dietary-specific PUFAs intake can modify the association between air pollutants and incident lung cancer remains unknown. MethodsCox proportional hazard models and restricted cubic spline regression were used to evaluate the associations of omega-3 PUFAs, omega-6 PUFAs and the ratio of omega-6 PUFAs to omega-3 PUFAs intake with lung cancer risk. Furthermore, we evaluated the associations between air pollutants and incident lung cancer, and whether dietary-specific PUFAs intake would modify the relationship using stratification analyses. ResultsThis study found significant associations between the risk of lung cancer and omega-3 PUFAs intake (hazard ratio [HR], 0.82; 95 % confidence interval [CI], 0.73–0.93; per 1 g/d), and omega-6 PUFAs intake (HR, 0.98; 95 % CI, 0.96–0.99; per 1 g/d). We did not observe an association between the omega-6 to omega-3 PUFAs intake ratio and incident lung cancer. With regard to air pollution, omega-3 PUFAs intake attenuated the positive relationship between nitrogen oxides (NOx) pollution and lung cancer risk, and an increased incidence of lung cancer was found only in the low omega-3 PUFAs intake group (p < 0.05). Surprisingly, PUFAs intake (regardless of omega-3 PUFAs, omega-6 PUFAs, or in total) reinforced the pro-carcinogenic effects of PM2.5 on lung cancer, and a positive association between PM2.5 pollutants and incident lung cancer was observed only in the high PUFAs groups (p < 0.05). ConclusionsHigher dietary omega-3 and omega-6 PUFAs intake was associated with a decreased risk of lung cancer in the study population. As omega-3 PUFAs have different modification effects on NOX and PM2.5 air pollution related lung cancer incidence, precautions should be taken when using omega-3 PUFAs as health-promoting dietary supplements, especially in high PM2.5 burden regions.
