Single-shot off-axis full-field optical coherence tomography

Full field optical coherence tomography (FF-OCT) enables high-resolution in-depth imaging within turbid media. In this work, we present a simple approach which combines FF-OCT with off-axis interferometry for reconstruction of en-face images. With low spatial and temporal coherence illumination, this method is able to extract an FF-OCT image from only one interference acquisition. This method is described, and the proof-of-concept is demonstrated through the observation of scattering samples such as organic and ex vivo biomedical samples.

Ptychographic imaging with partially coherent plasma EUV sources

Abstract We report on high-resolution lens-less imaging experiments based on ptychographic scanning coherent diffractive imaging (CDI) method employing compact plasma sources developed for extreme ultraviolet (EUV) lithography applications. Two kinds of discharge sources were used in our experiments: a hollow-cathode-triggered pinch plasma source operated with oxygen and for the first time a laser-assisted discharge EUV source with a liquid tin target. Ptychographic reconstructions of different samples were achieved by applying constraint relaxation to the algorithm. Our ptychography algorithms can handle low spatial coherence and broadband illumination as well as compensate for the residual background due to plasma radiation in the visible spectral range. Image resolution down to 100 nm is demonstrated even for sparse objects, and it is limited presently by the sample structure contrast and the available coherent photon flux. We could extract material properties by the reconstruction of the complex exit-wave field, gaining additional information compared to electron microscopy or CDI with longer-wavelength high harmonic laser sources. Our results show that compact plasma-based EUV light sources of only partial spatial and temporal coherence can be effectively used for lens-less imaging applications. The reported methods may be applied in combination with reflectometry and scatterometry for high-resolution EUV metrology.

Compact interferometric module for full-field interferometric phase microscopy with low spatial coherence illumination.

We propose a compact and external off-axis interferometric module that can achieve interference with low spatial coherence illumination over the entire field of view. The interferometer is easy to align and stable and can be connected to the output of an existing microscope illuminated with a low spatial coherence light source, thus allowing quantitative phase imaging with a low degree of spatial noise. We demonstrate the imaging and the interference properties of the proposed interferometric module and use it for quantitative phase imaging of reflective samples.

Coherence switching of a degenerate VECSEL for multimodality imaging

We demonstrate a VECSEL (vertical external cavity surface emitting laser) based degenerate source with an adjustable degree of spatial coherence that is electrically pumped, mechanically compact and supports continuous-wave emission. The laser operation can be switched between a large number of mutually incoherent spatial modes and few-mode operation at little power loss. This technology allows multimodality imaging, where low spatial coherence illumination is used for traditional high-speed video-microscopy and high spatial coherence illumination is used to extract dynamic information of flow processes. The initial demonstration is performed on imaging embryo heart function in Xenopus, which is an important animal model for human heart disease.

Numerically focused full-field swept-source optical coherence microscopy with low spatial coherence illumination

We propose a 3D imaging technique based on the combination of full-field swept-source optical coherence microscopy (FF-SSOCM) with low spatial coherence illumination and a special numerical processing that allows for numerically focused coherent-noise-free imaging without mechanical scanning in longitudinal or transversal directions. We show, both theoretically and experimentally, that the blurring effects arising in FF-SSOCM due to defocus can be corrected by appropriate numerical processing even when low spatial coherence illumination is used. A FF-SSOCM system was built for testing the performance of this technique. Coherent-noise-free imaging of a sample with longitudinal extent exceeding the optical depth of field is demonstrated without displacement of the sample or any optical element.

Low-coherence enhanced backscattering from highly forward scattering media

We present a theoretical basis for calculation of the angular profile of the coherent backscattering intensity under low spatial coherence illumination. We take into account two contributions to the intensity, namely, the diffusion contribution and the contribution from the waves that experience the small-angle multiple scattering before and after single deflection in the backward direction. The latter contribution describes transport of light at subdiffusion length scales and is responsible for the wings of the backscattering angular profile. Our results are in good agreement with data of Monte-Carlo simulations and experiment.

Enhancement factor in low-coherence enhanced backscattering and its applications for characterizing experimental skin carcinogenesis

We experimentally study potential mechanisms by which the enhancement factor in low-coherence enhanced backscattering (LEBS) can probe subtle variations in radial intensity distribution in weakly scattering media. We use enhanced backscattering of light by implementing either (1) low spatial coherence illumination or (2) multiple spatially independent detections using a microlens array under spatially coherent illumination. We show that the enhancement factor in these configurations is a measure of the integrated intensity within the localized coherence or detection area, which can exhibit strong dependence on small perturbations in scattering properties. To further evaluate the utility of the LEBS enhancement factor, we use a well-established animal model of cutaneous two-stage chemical carcinogenesis. In this pilot study, we demonstrate that the LEBS enhancement factor can be substantially altered at a stage of preneoplasia. Our animal result supports the idea that early carcinogenesis can cause subtle alterations in the scattering properties that can be captured by the LEBS enhancement factor. Thus, the LEBS enhancement factor has the potential as an easily measurable biomarker in skin carcinogenesis.

Circular polarization memory effect in low-coherence enhanced backscattering of light

We experimentally study the propagation of circularly polarized light in the subdiffusion regime by exploiting enhanced backscattering [(EBS), also known as coherent backscattering] of light under low spatial coherence illumination. We demonstrate for the first time, to the best of our knowledge, that a circular polarization memory effect exists in EBS over a large range of scatterers' sizes in this regime. We show that low-coherence EBS signals from the helicity preserving and orthogonal helicity channels cross over as the mean free path length of light in media varies, and that the cross point indicates the transition from multiple to double scattering in EBS.

Modeling low-coherence enhanced backscattering using Monte Carlo simulation

Constructive interference between coherent waves traveling time-reversed paths in a random medium gives rise to the enhancement of light scattering observed in directions close to backscattering. This phenomenon is known as enhanced backscattering (EBS). According to diffusion theory, the angular width of an EBS cone is proportional to the ratio of the wavelength of light lambda to the transport mean-free-path length l(s)* of a random medium. In biological media a large l(s)* approximately 0.5-2 mm >> lambda results in an extremely small (approximately 0.001 degrees ) angular width of the EBS cone, making the experimental observation of such narrow peaks difficult. Recently, the feasibility of observing EBS under low spatial coherence illumination (spatial coherence length Lsc << l(s)*) was demonstrated. Low spatial coherence behaves as a spatial filter rejecting longer path lengths and thus resulting in an increase of more than 100 times in the angular width of low coherence EBS (LEBS) cones. However, a conventional diffusion approximation-based model of EBS has not been able to explain such a dramatic increase in LEBS width. We present a photon random walk model of LEBS by using Monte Carlo simulation to elucidate the mechanism accounting for the unprecedented broadening of the LEBS peaks. Typically, the exit angles of the scattered photons are not considered in modeling EBS in the diffusion regime. We show that small exit angles are highly sensitive to low-order scattering, which is crucial for accurate modeling of LEBS. Our results show that the predictions of the model are in excellent agreement with the experimental data.

Origin of low-coherence enhanced backscattering

The origin of low-coherence enhanced backscattering (EBS) of light in random media when the spatial coherence length of illumination is much smaller than the transport mean free path has been poorly understood. We report that in weakly scattering discrete random media low-coherence EBS originates from time-reversed paths of double scattering. Low spatial coherence illumination dephases the time-reversed waves outside its finite coherence area, which isolates the minimal number of scattering events in EBS from higher-order scattering. Moreover, we show the first experimental evidence that the minimal number of scattering events in EBS is double scattering, which has been hypothesized since the first observation of EBS.

Low-coherence enhanced backscattering: review of principles and applications for colon cancer screening

The phenomenon of enhanced backscattering (EBS) of light, also known as coherent backscattering (CBS) of light, has been the object of intensive investigation in nonbiological media over the last two decades. However, there have been only a few attempts to explore EBS for tissue characterization and diagnosis. We have recently made progress in the EBS measurements in tissue by taking advantage of low spatial coherence illumination, which has led us to the development of low-coherence enhanced backscattering (LEBS) spectroscopy. In this work, we review the current state of research on LEBS. After a brief discussion of the basic principle of EBS and LEBS, we present an overview of the unique features of LEBS for tissue characterization, and show that LEBS enables depth-selective spectroscopic assessment of mucosal tissue. Then, we demonstrate the potential of LEBS spectroscopy for predicting the risk of colon carcinogenesis and colonoscopy-free screening for colorectal cancer (CRC).

Coherent backscattering spectroscopy.

Coherent backscattering (CBS) of light in random media has been previously investigated by use of coherent light sources. Here we report a novel method of CBS measurement that combines low spatial coherence, broadband illumination, and spectrally resolved detection. We show that low spatial coherence illumination leads to an anomalously broad CBS peak and a dramatic speckle reduction; the latter is further facilitated by low temporal coherence detection. Thus CBS can be observed in biological tissue and other media that previously were beyond the reach of conventional CBS measurements. We also demonstrate, for the first time to our knowledge, spectroscopic analysis of CBS. CBS spectroscopy may find important applications in probing random media such as biological tissue in which depth-selective measurements are crucial.
