Antibody desolvation with sodium chloride and acetonitrile generates bioactive protein nanoparticles.

About 30% of the FDA approved drugs in 2021 were protein-based therapeutics. However, therapeutic proteins can be unstable and rapidly eliminated from the blood, compared to conventional drugs. Furthermore, on-target but off-tumor protein binding can lead to off-tumor toxicity, lowering the maximum tolerated dose. Thus, for effective treatment therapeutic proteins often require continuous or frequent administration. To improve protein stability, delivery and release, proteins can be encapsulated inside drug delivery systems. These drug delivery systems protect the protein from degradation during (targeted) transport, prevent premature release and allow for long-term, sustained release. However, thus far achieving high protein loading in drug delivery systems remains challenging. Here, the use of protein desolvation with acetonitrile as an intermediate step to concentrate monoclonal antibodies for use in drug delivery systems is reported. Specifically, trastuzumab, daratumumab and atezolizumab were desolvated with high yield (∼90%) into protein nanoparticles below 100 nm with a low polydispersity index (<0.2). Their size could be controlled by the addition of low concentrations of sodium chloride between 0.5 and 2 mM. Protein particles could be redissolved in aqueous solutions and redissolved antibodies retained their binding activity as evaluated in cell binding assays and exemplified for trastuzumab in an ELISA.

Extraction and Characterization of Type I Collagen from Parrotfish (Scarus sordidus Forsskål, 1775) Scale solubilized with the Aid of Acetic Acid and Pepsin.

Waste from marine fish processing is an important source of valuable products. Fish collagen is considered a alternative biomaterial due to its excellent properties, and it is widely used for industrial purposes. Thus, this present study aimed to characterize acid and pepsin-soluble collagens from the waste of parrotfish (Scarus sordidus Forsskål, 1775) scales. The yields (p > 0.05) of acid-soluble collagen (ASC-PFS) and pepsin-soluble collagen (PSC-PFS) were 1.17 g/100 g and 1.00 g/100 g, respectively. Both collagen samples were categorized as type I owing to the presence of two alpha chain subunits (α1 and α1) after being confirmed by a sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). Under the fourier transform infrared (FTIR) test, the triple helical structure of type I collagens from the ASC-PFS and PSC-PFS was maintained. Moreover, the study of UV visible spectra and X-ray diffraction (XRD) showed the similarity of collagens derived from different fish species, and the thermostability (T max) evaluation of all extracted collagens was in the range of 36.22-37.78°C, and their values were comparable to previous research on the fish scale collagens. The effect of various pH and sodium chloride (NaCl) treatments on solubility exhibited that the ASC-PFS and PSC-PFS were highly soluble in an acidic condition (pH < 5.0) and low concentration of sodium chloride (<30 g/L). Taken together, collagens extracted from parrotfish scale waste can be an alternative source for industries.

Effect of monovalent electrolyte solutions on the human tear ferning pattern.

To investigate the effect of the addition of a low concentration of sodium chloride (NaCl) and potassium chloride (KCl) solutions on the tear ferning (TF) patterns of tears collected from humans. A tear sample (20 μL) was collected from the right eye of 23 males and 7 females (25.4 ± 6.6 years). The tears were collected in one sitting for healthy subjects (N = 13). For dry eye participants (N = 17), the tear samples were collected in two separate settings with five minutes gap in between. A sample (1 μL) from each tear was dried on a glass slide, and the obtained ferns were observed using a microscope and graded using the five-point TF grading scale. Mixtures of tear samples (0.5 μL) and different volumes (0.5-2.5 μL) of each electrolyte (10-30 mg in 100 mL of water) solution were prepared, and their TF patterns were recorded and compared with those of the corresponding pure tears. Significant improvements (Wilcoxon test, P < 0.001) have been seen in the TF grades of the tear samples after the addition of NaCl and KCl solutions. A significant difference (Wilcoxon test, P = 0.016) was found between the TF grades when NaCl and KCl solutions were added to the tear samples. The TF grades of pure tears collected from dry-eye subjects ranged from 2.1 to 3.5, based on the five points grading scale, and decreased to be in the range of 0.4 to 1.6 after the addition of electrolyte solutions. While the TF grades of pure tears collected from normal-eye ranged from 1.2 to 1.9 and improved after the addition of electrolyte solution to be in the range of 0.4 to 1.5. The TF test was used in vitro to assess the impact of the addition of a low concentration of sodium and potassium chloride solutions on tears collected from humans. The TF grades of human tears significantly improved after the addition of either sodium or potassium chloride solution. The mechanism for the improvement in TF grades due to the addition of electrolyte solutions must be investigated.

Effects of Low Concentrations of NaCl and EG on Hydrate Formation Kinetics and Morphology in the Presence of SDS

The research on gas hydrates can be divided into two subfields: risk prevention and control of pipeline blockage by hydrates and industrial applications of solidified natural gas (SNG) in gas storage and transportation, seawater desalination, and gas recovery. The two opposing properties of hydrates have stimulated research into promoting and inhibiting methods. A considerable number of studies have reported that the same type of additive can play a promoting or inhibiting effect at different concentrations. This study evaluated the kinetic effects of low concentrations of ethylene glycol (EG) and sodium chloride (NaCl) on methane hydrate production in sodium dodecyl sulfate (SDS) solution. The results showed that both NaCl and EG had kinetic inhibitory effects on the hydrate reaction in SDS solution, and the inhibitory effects gradually increased with the increase of the concentration. Among them, the inhibitory effect of NaCl was stronger than that of EG at the same content. 5 wt % NaCl can reduce the reaction rate of hydrate in SDS solution by 81%, but 0.1 wt % NaCl helped SDS to act on hydrate growth, and the rate of hydrate growth stage was increased by 100%. When 0.1 wt % NaCl or 5 wt % EG existed in the SDS solution, the hydrate mainly grew in the bulk solution and on the inner wall surface of the container, forming a peak-like structure below the interface, and 5 wt % EG also formed a ridge-like structure above the interface. Exploring the macroscopic formation characteristics of hydrate is helpful to industrial process optimization and predicting the additive concentration in the pipeline. This work emphasizes that the selection of substances can be broadened and the rational use of resources can be achieved as much as possible when selecting promoters that are conducive to hydrate formation kinetics or inhibitors that control hydrate growth.

Physicochemical and Microstructural Analyses of Pepsin-Soluble Collagens Derived from Lizardfish (Saurida tumbil Bloch, 1795) Skin, Bone and Scales.

Reducing food waste is critical for sustainability. In the case of fish processing, more than sixty percent of by-products are generated as waste. Lizardfish (Saurida tumbil Bloch, 1795) is an economically important species for surimi production. To address waste disposal and maximize income, an effective utilization of fish by-products is essential. This study aims to isolate and characterize pepsin-soluble collagens from the skin, bone and scales of lizardfish. Significant differences (p < 0.05) in the yields of collagen were noted with the highest yield recorded in pepsin-soluble skin collagen (PSSC) (3.50 ± 0.11%), followed by pepsin-soluble bone collagen (PSBC) (3.26 ± 0.10%) and pepsin-soluble scales collagen (PSCC) (0.60 ± 0.65%). Through SDS–polyacrylamide gel electrophoresis, the presence of two alpha chains were noted and classified as type I. From Fourier transform infrared spectroscopy (FTIR) analysis, the triple-helix structure of the collagen was maintained. The X-ray diffraction and UV visible spectra characteristics of the lizardfish collagens in this study are similar to the previously reported fish collagens. In terms of thermostability, PSSC (Tmax = 43.89 °C) had higher thermostability in comparison to PSBC (Tmax = 31.75 °C) and PSCC (Tmax = 30.54 °C). All pepsin-soluble collagens were highly soluble (>70%) in acidic conditions (particularly at pH 4.0) and at low sodium chloride concentrations (0–30 g/L). Microstructural analysis depicted that all extracted collagens were multi-layered, irregular, dense, sheet-like films linked by random coiled filaments. Overall, pepsin-soluble collagens from lizardfish skin, bone and scales could serve as potential alternative sources of collagens.

Sweet basil can be grown hydroponically at low phosphorus and high sodium chloride concentration: Effect on plant and nutrient solution management

Agronomic inputs in agriculture easily exceed crop needs causing environment pollution. Phosphorus (P) is a macronutrient largely used in agriculture as a mineral fertilizer that requires an accurate management because it is a finite and non-renewable resource. Growing plants hydroponically can optimize plant nutrition achieving high crop nutrient use efficiency. However, irrigation water of poor quality (saline) interferes with crop performance. In this study, the effects of different P and NaCl levels in the root zone were evaluated on basil plants grown in a deep liquid culture system in a growth chamber. Two experiments with different P (from 0.1 to 1.0 mol m−3) and NaCl (from 0 to 60 mol m−3) concentrations in the nutrient solution were carried out. Plant growth, leaf gas exchanges and fluorescence, uptake of minerals and water, and leaf quality (content of minerals and pigments, and antioxidant capacity) were analysed. The most relevant morphological plant response to the low P (i.e., roughly 0.1 x of a standard nutrient solution) concentration consisted in a greater root/shoot ratio. However, plants growing at low P concentration did not show any reduction in yield and quality. Sodium chloride did not significantly affect plants yield and quality up to a concentration of 30 mol m−3 confirming the tolerance of basil to moderate saline stress. Significant reduction in the total biomass, water uptake and photosynthesis instead occurred at NaCl 60 mol m−3. These results confirmed the hypotheses that basil can be grown hydroponically at low P and high NaCl concentration with proper management of the nutrient solution. The absence of significant interactions between P and NaCl suggests that these two factors can be managed independently with potential benefits for crop economic and environmental sustainability.

Screening of salt tolerance traits and the salt tolerance evaluation method in &lt;em&gt;Brassica napus&lt;/em&gt; at the seed germination stage

Soil salinity is one of the major abiotic stresses that negatively affect plant growth and agricultural productivity. For many crop species, the germination stage is one of the most sensitive stages to salinity stress. This study evaluated salt tolerance in 200 Brassica napus L. germplasms using hierarchical cluster analysis based on multiple morphological parameters, including germination rate, root length, fresh weight of root, shoot length, fresh weight of shoot, and total fresh weight. Membership function was used as a comprehensive index to select and evaluate salt tolerance of these germplasms, identifying 8 highly salt-tolerant germplasms, 40 salt-tolerant germplasms, 65 moderate salt-tolerant germplasms, 52 salt-sensitive germplasms, and 35 highly saltsensitive germplasms lines. The responses of rapeseed germplasm to salt stress indicate differences in morphological parameters. Furthermore, NaCl showed a positive effect on total fresh weight and biomass production of some germplasms at a concentration of 100 mmol L–1. Since the correlation value of salt tolerance with total fresh weight was highest under 200 mmol L–1 NaCl, it can be considered the most reliable parameter to evaluate salt tolerance. Therefore, the findings of this study can be applied as an effective and reliable method for mass screening and evaluation of Brassica napus germplasm at the germination stage for breeding salt-tolerant rapeseed genotypes. Highlights - The salinity tolerance of 200 varieties of B. napus germplasms was investigated. - B. napus is more vulnerable to saline conditions during the germination and early reproductive stages than the vegetative and flowering periods. - Based on hierarchical cluster analysis, there was a wide variability of salinity tolerance among rapeseed germplasm. - Low concentration of sodium chloride had a positive effect on shoot and root growth, germination and total weight in some B. napus seedlings. - Total fresh weight can be utilized as the most efficient index for mass screening of salt tolerance in B. napus germplasms at the germination stage.

Metagenetic and Volatilomic Approaches to Elucidate the Effect of Lactiplantibacillus plantarum Starter Cultures on Sicilian Table Olives.

The present study aimed to evaluate the effect of selected Lactiplantibacillus plantarum strains on both microbiota composition and volatile organic compound profile of Sicilian table olives. Two mixed cultures, named O1 and O2, were set up for pilot-plan scale fermentations at 5% of NaCl. Uninoculated table olives at 5 and 8% (C5 and C8) of salt were used as control. The fermentation process was monitored until 80 days through a dual approach, which included both classical microbiological and 16S amplicon-based sequencing and volatilomics analyses. Compared with control samples (C5 and C8), experimental samples, inoculated with starter cultures (O1 and O2), exhibited a faster acidification with a more pronounced drop in pH. Metagenetics data revealed significant differences of microbiota composition among samples, highlighting the dominance of lactobacilli in both experimental samples; a high occurrence of Enterobacter genus only in control samples with 5% of NaCl; and the presence of Bacteroides, Faecalibacterium, Klebsiella, and Raoultella genera only in control samples with 8% of NaCl. Furthermore, microbiota composition dynamics, through the fermentation process, significantly affected the volatile organic compounds of the final products, whereas no compounds involved in off-odors metabolites were detected in all samples investigated. In conclusion, the addition of the proposed starter cultures and the use of low concentrations of sodium chloride positively affected the microbiota and volatile organic compounds, ensuring the microbiological safety and the pleasant flavors of the final product.

Conversion of xylose into furfural over MC-SnOx and NaCl catalysts in a biphasic system

Furfural is a promising platform chemical that can be catalyzed from lignocellulose biomass. In this study, a novel micro-mesoporous carbon supported tin oxide catalyst (MC-SnOx) was developed to convert xylose into furfural in a low sodium chloride (NaCl) concentration and acid-free biphasic system. The catalysts synthesized by annealing from 400 °C to 600 °C for 3.5h were characterized by BET, SEM, XPS, XRD, NH3-TPD and FTIR techniques. The factor that mostly affected the catalytic performance was the acid concentration of the catalysts, and the best catalytic performance was achieved by MC-SnOx annealed at 450 °C. Further increasing the annealing temperature can cause reduction, volatilization and aggregation of Sn species, which can finally affect acid concentration and performance of the catalyst. In addition, a synergistic catalytic effect was found between MC-SnOx and NaCl and effectivity of low concentration of NaCl indicated the potential of applying seawater or wastewater containing NaCl as a low-cost reaction solvent and NaCl source. To balance the catalytic performance, cost, equipment safety and environmental concerns, a reasonable furfural yield of 53.9% was achieved over the MC-SnOx-450 and NaCl (0.2 M) in biphasic system under mild conditions (180 °C and 20 min) with a good catalyst reusability.

Selected Metabolites Profiling of Staphylococcus aureus Following Exposure to Low Temperature and Elevated Sodium Chloride.

Staphylococcus aureus is one of the main foodborne pathogens that can cause food poisoning. Due to this reason, one of the essential aspects of food safety focuses on bacterial adaptation and proliferation under preservative conditions. This study was aimed to determine the metabolic changes that can occur following the exposure of S. aureus to either low temperature conditions or elevated concentrations of sodium chloride (NaCl). The results revealed that most of the metabolites measured were reduced in cold-stressed cells, when compared to reference controls. The major reduction was observed in nucleotides and organic acids, whereas mannitol was significantly increased in response to low temperature. However, when S. aureus was exposed to elevated NaCl, a significant increase was observed in the metabolite levels, particularly purine and pyrimidine bases along with organic acids. The majority of carbohydrates remained constant in the cells grown under ideal conditions and those exposed to elevated NaCl concentrations. Partial least square discriminate analysis (PLS-DA) of the metabolomic data indicated that both, prolonged cold stress and osmotic stress conditions, generated cells with different metabolic profiles, in comparison to the reference controls. These results provide evidence that, when bacterial cells exposed to low temperatures or high concentrations of NaCl, experience in situ homeostatic alterations to adapt to new environmental conditions. These data supported the hypothesis that changes in metabolic homeostasis were critical to the adaptive processes required for survival under alterations in the environmental conditions.

Differential Effects of Growth Medium Salinity on Biofilm Formation of Two Salmonella enterica Strains

Salmonella enterica is a prominent foodborne pathogen, including diverse serotypes that are prolific biofilm formers. Its ability to form biofilm can be affected by multiple environmental factors. In this study, the effect of salinity on biofilm formation by S. enterica was evaluated by using two recently isolated strains of Salmonella serotypes Enteritidis and Newport. Although supplementing the growth medium with a low concentration (0.5 to 2%) of sodium chloride (NaCl) slightly enhanced biofilm formation for the strain S. enterica serovar Enteritidis 110, it sharply reduced or abolished biofilm formation by the strain S. enterica serovar Newport 193. This differential effect of salinity on S. enterica strains of different serotypes was poorly correlated with inhibition of planktonic growth but strongly correlated with cell motility. Examining genes known to affect biofilm formation showed that the expression of adrA, csgD, and fliC, which encode proteins required for surface adhesion and cell motility, was significantly downregulated with salinity increase in Salmonella Newport 193 but not in Salmonella Enteritidis 110. Therefore, it is plausible that the differential effect of salinity on biofilm formation by Salmonella Enteritidis 110 and Salmonella Newport 193 resulted from the differential regulation to genes required for cell adherence and motility.

Effect of some environmental conditions on the growth of rhizobium species

Rhizobium species are involved in symbiotic relationship which can be exploited in agriculture to enhance crop and pasture growth without the addition of nitrogen fertilizers. However, a number of environmental factors are known to affect the symbiotic efficiency of Rhizobium. This current study evaluated tolerance of Rhizobium species isolated from root nodules of cowpea obtained from Akure, Nigeria to variation in temperature, pH levels, salt concentrations, heavy metals as well as antibiotics. Three Rhizobium species were isolated from root nodules of Vignaunguiculata and Phaseolus vulgaris after 34 days of planting yeast extract mannitol agar (YEMA). Their tolerance to environmental factors such as temperature, pH, sodium chloride, heavy metals as well as antibiotics was determined. All the isolates grew very well at 28oC, moderately at 37oC but their growth was hampered at 50o C. Maximal growth was observed at neutral pH. However, Sinorhizobium sp showed high tolerance to pH 7.0. Also, the isolates showed high tolerance to low concentration of sodium chloride (0.1%) and heavy metals with reduction in their optical density at higher concentrations. Rhizobium sp showed high level of tolerance at 4% compared to Mesorhizobium sp. and Sinorhizobium sp. Rhizobium spp. showed resistance to all the antibiotics investigated while Mesorhizobium sp. was sensitive to only to pefloxacin and sparfloxacin, while Sinorhizobium sp. was only sensitive to pefloxacin. The isolates in this study can be assessed for their suitability as inoculants forcowpea in soils below temperature 50oC and at neutral pH.&#x0D; J. bio-sci. 26: 15-24, 2018

Evaluation of Terpolymer-Gel Systems Crosslinked by Polyethylenimine for Conformance Improvement in High-Temperature Reservoirs

SummaryA terpolymer-gel system using low toxic polyethylenimine (PEI) as the crosslinker was developed for conformance improvement in high-temperature reservoirs. Suitable gelation time (GT), gel strength, and thermal stability could be obtained by selecting PEI molecular weight and adjusting terpolymer concentrations. With the increase of terpolymer concentration, GT decreases and the gel strength increases. However, in this research, the effect of PEI concentration on the gelation performance was much less obvious than that of the polymer concentration. Very low concentrations of sodium chloride (NaCl) can slightly shorten the GT. After critical concentrations were reached, the authors determined that the ions will delay the crosslinking reaction. Moreover, the addition of sodium carbonate (Na2CO3) can also lengthen GT. The gel systems were able to maintain thermal stability at 150°C. Uniformly distributed 3D network microstructures and the small size of the gel-grid pores made the network structure maintain thermal stability. The use of the terpolymer-gel-system gelation mechanism crosslinked by PEI can help petroleum engineers better understand and apply this terpolymer-gel system.

The Efficacy of an Electrolysed Water Formulation on Biofilms.

Electrolysed water is a basic process whereby an electric current is passed through deionised water containing a low concentration of sodium chloride in an electrolysis chamber, which results in a more complex chemistry resulting in the production of a strong bactericidal and fungicidal solution at the anode. This microbicidal solution contains hypochlorous acid that is fast-acting and environmentally safe, as upon bacterial killing, the equilibrium shifts from hypochlorous acid back to salt and water. Other antimicrobial agents produced in this process include sodium hypochlorite and chlorine. The use of electrolysed water formulations in wound care to control wound bioburden is underway. However, there is limited evidence of the efficacy of electrolysed water on the control of biofilms, which are renowned for their tolerance to a variety of antimicrobials. Therefore this study aimed to assess a new electrolysed water formulation on in vitro Staphylococcus aureus and Pseudomonas aeruginosa biofilms. Results showed that the electrolysed water formulation effectively reduced biofilm in all models following a 15min contact time. Microbial cell counts confirmed the reduction biofilm bacteria. Additional cytotoxicity using L929 fibroblasts confirmed that a 50% and 25% dilution of the electrolysed water formulation was non-cytotoxic to cells. In conclusion, this study has confirmed that the application of a new electrolysed water product effectively removed biofilm after a short exposure time. The use of this technology as a wound cleanser may help to control existing biofilms in complicated, non-healing wounds.

Effects of Environmental Stresses and in Vitro Digestion on the Release of Tocotrienols Encapsulated Within Chitosan-Alginate Microcapsules.

Considering the health benefits of tocotrienols, continuous works have been done on the encapsulation and delivery of these compounds. In this study, we encapsulated tocotrienols in chitosan-alginate microcapsules and evaluated their release profile. Generally, these tocotrienols microcapsules (TM) displayed high thermal stability. When subjected to pH adjustments (pH 1-9), we observed that the release of tocotrienols was the highest (33.78 ± 0.18%) under basic conditions. The TM were also unstable against the effect of ionic strength, with a high release (70.73 ± 0.04%) of tocotrienols even at a low sodium chloride concentration (50 mM). As for the individual isomers, δ-tocotrienol was the most sensitive to pH and ionic strength. In contrast, β-/γ-tocotrienols were the most ionic-stable isomers but more responsive toward thermal treatment. Simulated gastrointestinal model showed that the chitosan-alginate-based TM could be used to retain tocotrienols in the gastric and subsequently release them in the intestines for possible absorption.

Nutritional Value and Potential Chemical Food Safety Hazards of Selected Traditional and Conventional Pork Hams from Poland

Consumers no longer look for food characterized only by health safety and proper nutritional value. As a result, an increased interest in traditional and regional food can be observed. The aim of this paper is to analyze the results of a comparative analysis of three types of hams: traditional products registered on the List of Traditional Products of the Polish Ministry of Agriculture and Rural Development, then products whose names, retail prices, appearance, and manufacturer’s description suggest traditional methods of production, and finally conventional hams. The results show that traditional hams are characterized by the highest nutritional value among the analyzed groups of products. They have the lowest water and a high protein content. In addition, traditional hams are distinguished by a low concentration of sodium chloride and no addition of phosphates. The residues of nitrites and nitrates indicate their moderate use in manufacturing processes.

Assessment of apparent effectiveness of chemical egg disinfectants for improved artificial hatching in Oreochromis karongae (Pisces: Cichlidae)

Oreochromis karongae is one of the three key Tilapia species locally known as ‘Chambo’ which is endemic to Lake Malawi. The species is favored by consumers and due to high demand its catches have significantly declined over time. Aquaculture holds the potential to supplement catches as well as produce seed for restocking purposes. However, seed production of O.karongae in artificial hatcheries has registered little success due to high egg and fry mortalities. Therefore, a study was conducted to compare the apparent effectiveness of three chemical egg disinfectants in an attempt to improve hatching success and reduce fry mortality in O. karongae in an artificial re-circulating incubation system at the National Aquaculture Center, Domasi, Malawi. Batches of 200 stage II eggs were immersed for 5 minutes in 50ml solutions of (1) 50mg/l Potassium Permanganate, (2) 500mg/l Formalin, (3) 1000mg/l Sodium Chloride, (4) 5000mg/l Sodium Chloride, while treatment (5) was a control without any chemical treatment. Each treatment was replicated three times in 1.2 L plastic incubation jars with continuous water flow rate of 0.17 l/s operated at 10% daily water replacement. There was significantly low total mortality and high hatchability in treatments (1), (2) and (3) than (4) and (5) (p<0.05). The results provided evidence that pre-incubation egg treatment using chemicals has potential to significantly reduce mortalities and increase fry production in an artificial re-circulating incubation system. It is recommended, based on the findings that lower concentrations of sodium chloride, being a safer and less toxic chemical can be used for disinfecting O.karongae eggs. Future studies should focus on undertaking a toxicity test with various levels of the chemical disinfectants to identify optimum doses. This should be coupled with microbiological assays to validate the apparent effectiveness found in this study and identify the key microbial species that are responsible for mortalities during incubation of O.karongae eggs in a re-circulating system. Compression coating technique has been used in formulation of chronotherapeutic drug delivery systems with pulsatile carvedilol release with polyethylene oxide as controlling release agent. FMEA, risk analysis tool, was applied within Quality by Design approach with aim to detect process and formulation parameters affecting the carvedilol release profile from compression coated tablets. It gives Risk Priority Numbers (RPNs) for each failure mode. Also, using experimental designs, statistical significance of the formulation parameters influence was estimated. High RPNs in case of the lag time as critical quality attribute (CQA) was obtained for polymer molecular weight, compression of coat and low concentration of sodium chloride. For percent of released carvedilol from coated tablets (Q), second CQA, RPNs were high for low concentration of sodium chloride, sodium starch glycolate and crospovidone, polymer molecular weight and also for compression of the tablet coat. Experiments performed according to experimental plans, showed statistically significant influence of Polyox® WSR N60K and sodium chloride concentration on lag time, and concentration of polymer, sodium chloride, mannitol and type of superdisintegrant on Q. These studies demonstrate that FMEA may be a useful tool for Formulation by Design of compression coated tablets.

Impact of low-concentrated acidic electrolysed water obtained by membrane electrolysis on the decontamination of meat microbiota

Abstract The influence of acidic electrolysed water (AEW) treatment on inactivation of pure bacterial cultures inoculated onto the surface of agarised media and surface microbiota of pork meat were examined. Low-concentrated AEW (low concentration of sodium chloride and low current electrolysis) was generated by electrolysis (5 or 10 min) of 0.001% or 0.01% NaCl solution. The number of viable microorganisms was determined using a plate count method. The effect of AEW on bacterial cell morphology were investigated using scanning electron microscopy (SEM). After treatment with AEW, a significant, about 3.00 log reduction of Pseudomonas fluorescens, Yersinia enterocolitica, Escherichia coli, Staphylococcus aureus, Bacillus subtilis, Listeria monocytogenes, and Micrococcus luteus populations was observed. In the AEW treatment of pork, the highest reduction of total number of microorganisms (2.1 log reduction), yeast and moulds (2.5-2.6 log reduction), and psychrotrophs (more than 1 log reduction) was observed after spraying with 0.001% NaCl subjected to 10 min electrolysis. SEM revealed disruption and lysis of E. coli and S. aureus cells treated with AEW, suggesting a bactericidal effect. Higher available chlorine concentration (0.37-8.45 mg/L), redox potential (863.1-1049.8 mV), and lower pH (2.73-3.70) had an influence on the shape of bacteria and the number of breaks in the bacterial membrane.

Gelation Study on a Hydrophobically Associating Polymer/Polyethylenimine Gel System for Water Shut-off Treatment

A gel system has been widely used in many mature oilfields for water shut-off treatment. In the present study, the hydrophobically associating polymer (HAP) was cross-linked with the polyethylenimine (PEI) to form a HAP/PEI gel system which contains 0.35 wt % HAP and 0.60 wt % PEI. Gelation behaviors of the HAP/PEI gel formed in bottle and in core were studied, respectively. Results show that the gelation behaviors of the gel system were greatly affected by the concentration of HAP and that of PEI. The addition of the sodium chloride or calcium chloride generally resulted in a decrease of the apparent viscosity and an extension of the gelation time. However, a low concentration of sodium chloride led to a slight increase of the gel viscosity in bottle due to the intensification of the hydrophobic association. The rock skeleton of the core had a great effect on the gelation behavior.
