Development of Stabilizing Solution for Long-Term Storage of Bacteriophages at Room Temperature and Application to Control Foodborne Pathogens.

Bacteriophages (phages) have gained considerable attention as effective antimicrobial agents that infect and kill pathogenic bacteria. Based on this feature, phages have been increasingly used to achieve food safety. They are stored in a medium or buffer to ensure stability; however, they cannot be directly applied to food under these conditions due to reasons such as regulatory considerations and concerns about marketability. This study developed a stabilizing solution that allowed the maintenance of phage activity for extended periods at room temperature while being directly applicable to food. The stability of phages stored in distilled water was relatively low. However, adding a stabilizer composed of sugars and salts improved the survival rates of phages significantly, resulting in stability for up to 48 weeks at room temperature. When Escherichia coli O157:H7-contaminated vegetables were washed with tap water containing phages, the phages reduced the pathogenic E. coli count by over 90% compared with washing with tap water alone. Additionally, when pathogenic E. coli-contaminated vegetables were placed in a phage-coated container and exposed to water, the coating of the container dissolved, releasing phages and lysing the pathogenic E. coli. This led to a significant 90% reduction in pathogenic E. coli contamination compared to that after water rinsing. These results suggest an effective and economical method for maintaining phage activity and establishing the potential for commercialization through application in the food industry.

A simple long term storage solution for human tissues as continuous source of DNA/RNA for PCR analysis

Aim: In molecular biology, there is a need for tissue storage solutions for long term analysis in order to have a suitable source of nucleic acids (RNA and DNA). There are many reports to store the tissue samples in different solutions like ethanol, DMSO as well as with storage in liquid nitrogen, but a suitable storage solution for tissue at room temperature is still missing. Therefore, a solution called Genekam Tissue Storage Solution (GTSS) is developed, which can be used to store the tissues at room temperature for a long time. Materials and Methods: Human tissues were stored in this solution for 4 years. as these are tested with conventional and real time PCR for the presence of internal controls after the isolation of RNA and DNA over the period of 4 years, along with spectrometric testing. Results: Results of internal controls for human housekeeping genes like beta-globin and beta-actin were successful in nucleic acid isolations. Spectrophotometric values of all nucleic acid isolation range from 1.03 to 33.8 ng/µl (1.03 to 33.8 µg/ml). Conclusion: In this work, it may be the first time about the successful development of a simple long-term noncryogenic storage solution for human tissues at room temperature as a continuous source of DNA and RNA for molecular biological analysis.

Climate performance of liquefied biomethane with carbon dioxide utilization or storage

In the process of upgrading biogas to biomethane for gas grid injection or use as a vehicle fuel, biogenic carbon dioxide (CO₂) is separated and normally emitted to the atmosphere. Meanwhile, there are a number of ways of utilizing CO₂ to reduce the dependency on fossil carbon sources. This article assesses the climate performance of liquefied biomethane for road transport with different options for utilization or storage of CO₂. The analysis is done from a life cycle perspective, covering the required and avoided processes from biogas production to the end use of biomethane and CO₂. The results show that all of the studied options for CO₂ utilization can improve the climate performance of biomethane, in some cases contributing to negative CO₂ emissions. One of the best options, from a climate impact perspective, is to use the CO₂ internally to produce more methane, although continuous supply of hydrogen from renewable sources can be a challenge. Another option that stands out is concrete curing, where CO₂ can both replace conventional steam curing and be stored for a long time in mineral form. Storing CO₂ in geological formations can also lead to negative CO₂ emissions. However, with such long-term storage solutions, opportunities to recycle biogenic CO₂ are lost, together with the possibility of de-fossilizing processes that require carbon, such as chemical production and horticulture.

Microscale Electrochemical Corrosion of Uranium Oxide Particles.

Understanding the corrosion of spent nuclear fuel is important for the development of long-term storage solutions. However, the risk of radiation contamination presents challenges for experimental analysis. Adapted from the system for analysis at the liquid-vacuum interface (SALVI), we developed a miniaturized uranium oxide (UO2)-attached working electrode (WE) to reduce contamination risk. To protect UO2 particles in a miniatured electrochemical cell, a thin layer of Nafion was formed on the surface. Atomic force microscopy (AFM) shows a dense layer of UO2 particles and indicates their participation in electrochemical reactions. Particles remain intact on the electrode surface with slight redistribution. X-ray photoelectron spectroscopy (XPS) reveals a difference in the distribution of U(IV), U(V), and U(VI) between pristine and corroded UO2 electrodes. The presence of U(V)/U(VI) on the corroded electrode surface demonstrates that electrochemically driven UO2 oxidation can be studied using these cells. Our observations of U(V) in the micro-electrode due to the selective semi-permeability of Nafion suggest that interfacial water plays a key role, potentially simulating a water-lean scenario in fuel storage conditions. This novel approach offers analytical reproducibility, design flexibility, a small footprint, and a low irradiation dose, while separating the α-effect. This approach provides a valuable microscale electrochemical platform for spent fuel corrosion studies with minimal radiological materials and the potential for diverse configurations.

The Effect of Choline Salt Addition to Trehalose Solution for Long-Term Storage of Dried and Viable Nuclei from Fully Grown Oocytes.

Although drying techniques are exciting alternatives to cryopreservation, it remains challenging to maintain tightly controlled temperatures and humidity levels during storage of dried products. The objective of this study was to determine if the addition of choline acetate to trehalose solution could enable a wider range of storage conditions for preservation of nuclei from fully grown oocytes, by allowing temporary humidity excursions (>44% relative humidity) that may lead to crystallization of trehalose and loss of DNA integrity. Using domestic cat germinal vesicle oocytes as a model, we characterized the recovery as well as the integrity of samples after microwave-assisted dehydration. Exposure to choline acetate alone did not impair the germinal vesicle's DNA integrity and only had a negative impact on the chromatin configuration. Choline acetate addition enabled us to reach lower moisture contents after 25 min of microwave-assisted drying. Sample recovery after rehydration was also better in the presence of choline acetate. The integrity of the germinal vesicle's DNA was not affected, while the chromatin configuration was impaired by the presence of choline acetate during dehydration. Importantly, choline acetate addition helped to maintain an amorphous state (absence of detrimental crystallization) during excursion from ideal humidity conditions.

Direct measurement of hydrogen relative permeability hysteresis for underground hydrogen storage

Underground Hydrogen Storage (UHS) is a long-term storage solution which utilises hydrogen as an energy carrier to store large-scale excess renewable energy in the subsurface. Key petrophysical properties, such as wettability and interfacial tension of hydrogen and water have been discovered which provide a practical basis for simulation models. Our work directly measures relative permeability hysteresis during co-injection core floods of hydrogen and water with a capillary number of Nc=2.92×10−7. Segmented micro-CT images are used to determine saturation in the Bentheimer sandstone. Our results demonstrate that hydrogen relative permeability in sandstones is very low, with an end-point relative permeability of krg0=0.049 at Swi= 0.24. Capillary trapping results in residual gas saturation of Sgr= 0.4. Wettability analysis confirms the system is water-wet, with contact angles of 57° at Sgr and 44° at Swi. These results improve the reliability of numerical simulations and demonstrate the possible challenges involved with the commercial application of UHS.

Preservation of black grapes by isochoric freezing

Fruits are perishable. It's crucial to have an efficient preservation technique that may extend storage duration while maintaining the physical quality and nutritional values to avoid wastage. The majority of long-term storage solutions for fruits use refrigeration. In this study, we evaluate the potential of isochoric freezing as an alternative method of preservation for black grapes (Vitis vinifera L.). We compare the properties of black grapes preserved for 7 days in trehalose solution at −4 °C in isochoric conditions (average pressure 34.2 MPa) with those of fresh black grapes and with grapes preserved isobarically in four conditions (room temperature, in the fridge, in the freezer, and in a plastic bag filled with trehalose solution). The results indicate that grapes preserved by isochoric freezing at temperatures below the freezing point of water do not lose weight; on the contrary, they resulted in a very small (2%) weight gain. Freezing under isochoric conditions did not result in significant changes in terms of macroscopic appearance, colour, firmness, °Brix values, or pH. We consider that isochoric freezing has the potential to be used as a preservation method for grapes while maintaining physicochemical parameters similar to those of fresh fruits.

Evaluation of Storage Solutions for DNA Preservation in Various Conditions

DNA should be protected from factors such as temperature fluctuation, hydrolysis, and oxidative damage, to be preserved intact. Thus far, DNA has been conventionally stored in freezers to avoid the influence of these factors. However, as the required storage time and number of samples increase, so do the spatial and financial burden. DNAstable and DNAstable Plus are commercial solutions for long-term storage of DNA at room temperature. In this study, DNA extracted from human blood was stored at different temperatures (room temperature, 60 °C, −80 °C, −20 °C, and 4 °C), under hydration and illumination conditions, and with or without long-term storage solutions. After 1, 3, 6, and 12 months of storage, we analyzed the quantity and preservation of STR loci of the stored DNA. Storage of DNA for 1 year at 60 °C using DNAstable® Plus, simulating 16 years of storage at room temperature, had a preservative effect similar to that of freezing in dried state. On the other hand, DNAstable® was inefficient compared to conventional methods. Storage at ambient temperature using DNAstable® Plus thus presents a cost- and space-effective alternative for long-term storage, even for concentrations of DNA lower than 0.5 ng/μL.

Design, construction, and validation of an in-situ groundwater trace element analyzer with applications in carbon storage

It is estimated that carbon emissions should reach net-zero by 2050 to meet important climate targets. Carbon capture is likely necessary to reach these targets, requiring a long-term storage solution such as geological carbon sequestration. However, as with any subsurface activity, leakage can occur, potentially impacting groundwater quality near the storage site. Rapid detection is essential to mitigate damage to this resource. Since CO2 will acidify groundwater, the concentrations of acid soluble minerals and associated cations will increase. Thus, an in-situ, real-time element analysis system based on laser-induced breakdown spectroscopy (LIBS) is under development to monitor these elements. The system splits the traditional LIBS system into a miniature, all-optical sensor head built around a passively Q-switch laser fiber coupled to a control unit. Previous work has validated the LIBS technique for use at high pressure as well as the split system design. In this work, a fieldable prototype sensor is developed and tested in an onsite monitoring well where trace elements concentrations (approx. 0–3 ppm) were tracked over 20 days. These concentrations varied in response to local rainfall, diluting with increased rain, demonstrating the ability of a LIBS-based sensor to track trace elements under real-world conditions.

Damages and stress responses in sperm cells and other germplasms during dehydration and storage at nonfreezing temperatures for fertility preservation.

Long-term preservation of sperm, oocytes, and gonadal tissues at ambient temperatures has the potential to lower the costs and simplify biobanking in human reproductive medicine, as well as for the management of animal populations. Over the past decades, different dehydration protocols and long-term storage solutions at nonfreezing temperatures have been explored, mainly for mammalian sperm cells. Oocytes and gonadal tissues are more challenging to dehydrate so little to no progress have been made. Currently, the detrimental effects of the drying process itself are better characterized than the impact of long-term storage at nonfreezing temperatures. While structural and functional properties of germ cells can be preserved after dehydration, a long list of damages and stresses in nuclei, organelles, and cytoplasmic membranes have been reported and sometimes mitigated. Characterizing those damages and better understanding the response of germ cells and tissues to the stress of dehydration is fundamental. It will contribute to the development of optimal protocols while proving the safety of alternativestorage options for fertility preservation. The objective of this review is to (1) document the types of damages and stress responses, as well as their mitigation in cells dried with different techniques, and (2) propose new research directions.

The Use of Pectins As Part of a Cryoprotective Solution For Long-term Storage of Human Platelet Concentrates

BACKGROUND: Pectins have unique properties and great potential to become an indispensable component of cryoprotective environment for platelet freezing. OBJECTIVE: To investigate the possibility of including pectins (apple pectin AU-701, tanacetan) into the composition of a cryoprotective solution for platelets during low-temperature storage. MATERIALS AND METHODS: Samples of platelet concentrates (PC) were frozen under the protection of complex solutions and stored in an electric freezer at -80 °C for 1 and 6 months. RESULT: The study showed that of the basic cryoprotectants, the best effect in the preservation of PC was with dimethylacetamide (DMAC). The use of pectins as an additive to the base solution of DMAC statistically improves the preservation of PC after exposure to low temperatures (-80 ° C) for 30 and 180 days. CONCLUSIONS: We conclude that DMAC is more promising as a basis for the development of a new combined cryoprotectant for PC freezing. Moreover, the chemical structure of pectin determines the level of its cryoprotective action in relation to the preservation of PC.

Experimental and numerical characterization of hydrogen jet fires

Compressed hydrogen gas is considered the most convenient and robust technological solution for long-term storage. However, several safety concerns are still under investigation. This work presents an experimental and numerical characterization of the jet flame produced after the accidental release from a high-pressure tank containing pure hydrogen at pressures ranging from 90 to 450 bar and release diameters ranging from 1 to 5 mm. Results are expressed in terms of temperature history and flame length. The complete set of measurements has been reported in the supplementary materials. Both integral and discrete models were employed. Besides, the computational fluid dynamic was integrated with finite reaction rate and accurate thermodynamic properties (from the ab initio approach) and showed excellent agreement with experimental data.

Highly Sensitive and Durable Organic Photodiodes Based on Long-Term Storable NiOx Nanoparticles.

Organic optoelectronic devices that can be fabricated at low cost have attracted considerable attention because they can absorb light over a wide frequency range and have high conversion efficiency, as well as being lightweight and flexible. Moreover, their performance can be significantly affected by the choice of the charge-selective interlayer material. Nonstoichiometric nickel oxide (NiOx) is an excellent material for the hole-transporting layer (HTL) of organic optoelectronic devices because of the good alignment of its valence band position with the highest occupied molecular orbital level of many p-type polymers. Herein, we report a simple low-temperature process for the synthesis of NiOx nanoparticles (NPs) that can be well dispersed in solution for long-term storage and easily used to form thin NiOx NP layers. NiOx NP-based organic photodiode (OPD) devices demonstrated high specific detectivity (D*) values of 1012-1013 jones under various light intensities and negative biases. The D* value of the NiOx NP-based OPD device was 4 times higher than that of a conventional poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate) (PEDOT:PSS)-based device, an enhancement that originated mainly from the 16 times decreased leakage current. The NiOx NP-based OPD device demonstrated better reliability over a wide range of light intensities and operational biases in comparison to a device with a conventional sol-gel-processed NiOx film. More importantly, the NiOx NP-based OPD showed long-term device stability superior to those of the PEDOT:PSS and sol-gel-processed NiOx-based devices. We highlight that our low-temperature solution-processable NiOx NP-based HTL could become a crucial component in the fabrication of stable high-performance OPDs.

The Influence of Hyper-Alkaline Leachate on a Generic Host Rock Composition for a Nuclear Waste Repository: Experimental Assessment and Modelling of Novel Variable Porosity and Surface Area

Deep geological disposal is the preferred solution for long-term storage of radioactive waste in many countries. In a deep repository, cementitious materials are widely used in the structure and buffer/backfill of the repository for the stabilisation of the hazardous materials. The cement acts as a physical barrier and also contributes chemically to waste containment by buffering the groundwater to a high pH, limiting the solubility of many radionuclides. This paper describes an experimental and modelling study which evaluates the geochemical interaction between young cement leachate (YCL, pH = 13) and a generic hard rock (in this case Hollington sandstone, representing a ‘hard’ host rock) during permeation with the leachate, as it drives mineralogical changes in the system. One-dimensional reactive transport was modelled using a mixing cell approach within the PHREEQC geochemical code to identify the essential parameters and understand and scale up the effect of variations in these parameters on the observed geochemical processes. This study also focused on the effects of variable porosity, reactive surface area and pore volume on improving the modelling of rock alteration in the system compared to conventional models that assume constant values for these properties. The numerical results showed that the interaction between the injected hyper-alkaline leachate and the sandstone sample results in a series of mineralogical reactions. The main processes were the dissolution of quartz, kaolinite and k-feldspar which was coupled with the precipitation of calcium silicate hydrate gel and tobermorite-14A (C–S–H), prehnite (hydrated silicate), saponite-Mg (smectite clay) and mesolite (Na–Ca zeolite). The simulation showed that the overall porosity of the system increased as primary minerals dissolve and no stable precipitation of the secondary C–S–H /C–A–S–H phases was predicted. The variable porosity scenario provides a better fitting to experimental data and more detailed trends of chemistry change within the column. The time and the number of moles of precipitated secondary phases were also improved which was related to greater exposure surface area of the minerals in the sandstone sample to the YCL.Article HighlightsThe drop in calcium, aluminium and silicate concentrations is mainly due to the formation of calcium silicate hydrate and zeolite minerals as secondary phases. The simulation showed that the overall porosity of the system increased as primary minerals dissolve and no stable precipitation of the secondary C–S–H /C–A–S–H phases was predicted.The dissolution of primary minerals and the precipitation of secondary C–S–H phases had a minimal effect on the pH values, and this was controlled mainly by the initial fluid chemistry.The variable porosity scenario provides a better fitting to experimental data and more detailed trends of chemistry change within the column.

Development and Demonstration of a Decentralised Hydrogen rSOC System

The quality of life and economic prosperity of the population depends to a large extent on a secure and sustainable electricity supply. In order to ensure this for generations to come, a comprehensive integration of renewable, decentralized electrical energy sources is necessary. Such developments are already evident in the world's largest markets. Inevitably, electricity production is increasingly being distributed over larger areas and energy supply systems are tending towards decentralization. Traditional role distributions, such as centralized feed-in and load adjustment by large power plants and decentralized demand, are being replaced by decentralized feeders at all grid levels and prosumers. Security of supply can only be achieved by balancing fluctuating output and demand. Therefore, in order to achieve the EU's 2050 climate targets, in addition to flexibility on the consumer side and sector coupling strategies, increased balancing options in the form of storage systems are required. Currently available storage technologies such as pumped storage power plants, etc. can be considered as complementary solutions. In Europe, their capacities are limited by almost exhausted utilization potentials. Since the fluctuating generation profiles also contain a significant seasonal component, long-term storage solutions are essential. In order to offer appropriate long-term storage services, the additional use of gaseous energy carriers enabled by PtG (Power-to-Gas) technologies is the only alternative.Currently, however, PtG storage technologies are still in the development and demonstration phase, so that further research is required primarily at the system but also at the component level. Another major issue is the commercial viability related to full-load operating hours and installation cost of PtG technologies, if the operation is only given when surplus electricity is available. Fuel cell technologies like reversible solid oxide cell systems (rSOC) that combine both electrolysis and electricity generation in one unit offer great potentials to address these issues.Within the Austrian funded R&D project FIRST (Fully Integrated Reversible Solid oxide cell system) a compact, reversible, high-temperature solid oxide (rSOC) system with a rated power of 15 kWel in SOEC operation and a rated power of 5 kWel in SOFC operation is being developed and demonstrated.In order to realize a compact and thus economically operable system, the Balance of Plant (BoP) components are being optimized for both SOEC and SOFC operation compared to two separate units. In addition, the stand alone system targets a fast switching between gas and electricity production and enables intelligent linking with electricity, gas and heat grids.The reversible solid oxide cell (rSOC) demonstrator shall be capable to produce H2 with > 70 % efficiency, or electricity with > 50% efficiency, in a simplified system with low operating costs. Flexible operation by rapidly switching between electrolysis and fuel cell modes within minutes to enable optimal use of electricity from fluctuating energy sources such as wind and solar. Load forecasting models are applied to respond in advance to changes in demand or yield, bringing new system control strategies to bear.The project aims to test the usability of the technology beyond test bed operation under real operating conditions as a long-term application. Therefore, the results will be relevant to industrial stakeholders as well as utilities.Within this work the status of the FIRST project will be presented including results from the simulative investigation and optimization within a dynamic model.

Photosystem I integrated into mesoporous microspheres has enhanced stability and photoactivity in biohybrid solar cells.

Isolated proteins, especially membrane proteins, are susceptible to aggregation and activity loss after purification. For therapeutics and biosensors usage, protein stability and longevity are especially important. It has been demonstrated that photosystem I (PSI) can be successfully integrated into biohybrid electronic devices to take advantage of its strong light-driven reducing potential (−1.2V vs. the Standard Hydrogen Electrode). Most devices utilize PSI isolated in a nanosize detergent micelle, which is difficult to visualize, quantitate, and manipulate. Isolated PSI is also susceptible to aggregation and/or loss of activity, especially after freeze/thaw cycles. CaCO3 microspheres (CCMs) have been shown to be a robust method of protein encapsulation for industrial and pharmaceutical applications, increasing the stability and activity of the encapsulated protein. However, CCMs have not been utilized with any membrane protein(s) to date. Herein, we examine the encapsulation of detergent-solubilized PSI in CCMs yielding uniform, monodisperse, mesoporous microspheres. This study reports both the first encapsulation of a membrane protein and also the largest protein to date stabilized by CCMs. These microspheres retain their spectral properties and lumenal surface exposure and are active when integrated into hybrid biophotovoltaic devices. CCMs may be a robust yet simple solution for long-term storage of large membrane proteins, showing success for very large, multisubunit complexes like PSI.

Designing an Event Store for a\xa0Modern Three-layer Storage Hierarchy

Event stores face the difficult challenge of continuously ingesting massive temporal data streams while satisfying demanding query and recovery requirements. Many of today’s systems deal with multiple hardware-based trade-offs. For instance, long-term storage solutions balance keeping data in cheap secondary media (SSDs, HDDs) and performance-oriented main-memory caches. As an alternative, in-memory systems focus on performance, while sacrificing monetary costs, and, to some degree, recovery guarantees. The advent of persistent memory (PMem) led to a multitude of novel research proposals aiming to alleviate those trade-offs in various fields. So far, however, there is no proposal for a PMem-powered specialized event store.Based on ChronicleDB, we will present several complementary approaches for a three-layer architecture featuring main memory, PMem, and secondary storage. We enhance some of ChronicleDB’s components with PMem for better insertion and query performance as well as better recovery guarantees. At the same time, the three-layer architecture aims to keep the overall dollar cost of a system low. The limitations and opportunities of a PMem-enhanced event store serve as important groundwork for comprehensive system design exploiting a modern storage hierarchy.

Metrological Suitability of Functionalized Epitaxial Graphene.

This work presents one solution for long-term storage of epitaxial graphene (EG) in air, namely through the functionalization of millimeter-scale devices with chromium tricarbonyl - Cr(CO)3. The carrier density may be tuned reproducibly by annealing below 400 K due to the presence of Cr(CO)3. All tuning is easily reversible with exposure to air, with the idle, in-air, carrier density always being close to the Dirac point. Precision measurements in the quantum Hall regime indicate no detrimental effects from the treatment, validating the pursuit of developing air-stable EG-based QHR devices.

Radiation-induced molecular hydrogen gas generation in the presence of aluminum alloy 1100

The United States government currently manages nearly 13 metric tons of aluminum-clad spent nuclear fuel (SNF) without a long-term storage solution, so a fundamental understanding of corrosion processes occurring on aluminum alloy surfaces is of utmost importance to plan for extended (>50 years) interim dry storage of aluminum-clad SNF. While thermal and chemical corrosion processes are well characterized for aluminum, radiation effects are not. To help understand the impacts of radiation on aluminum-clad SNF, the radiation-induced molecular hydrogen gas (H2) generation from pristine and pre-corroded aluminum alloy 1100 (Al-1100) coupons has been studied. Corrosion of coupons was achieved by submerging coupons in water at 95 °C for 29 days, yielding a ~5 μm boehmite/bayerite oxide film. Pre-corroded coupons were exposed to cobalt-60 gamma radiation to absorbed doses of up to 1.0 MGy under a variety of conditions: cover-gas composition (argon, nitrogen, or air), relative humidity (0, 50, and 100%), and temperature (ambient, 100, and 200 °C). Post-irradiation measurements demonstrated that the yield of H2 was directly attributable to the presence of the Al-1100 coupons and their physisorbed water with dependence on absorbed gamma dose, relative humidity, and cover-gas composition. No H2 was quantified in the presence of air, while both nitrogen and argon environments afforded higher H2 yields with increasing relative humidities. This was attributed to the greater availability of adsorbed water for radiolytic processes. Irradiation of pre-corroded Al-1100 coupons at different temperatures under 0% relative humidity argon conditions yielded statistically equivalent H2 yields for ambient temperatures and 100 °C. However, irradiation at 200 °C promoted a 3 to 4-fold increase in the yield of H2, possibly due to the transformation of bayerite to boehmite and/or improved efficiency of H• and H2 release from oxide surfaces.
