Bioactive proteins and antioxidant peptides from Litsea cubeba fruit meal: Preparation, characterization and ameliorating function on high-fat diet-induced NAFLD through regulating lipid metabolism, oxidative stress and inflammatory response

Non-alcoholic fatty liver disease (NAFLD) plays an increasingly significant threat to human health. In this study, the processing by-products of Litsea cubeba fruit meal were defatted by ultrasound-assisted methods, then the acetone-precipitated protein of L. cubeba (LCP) was obtained and structural analysis was performed. LCP was hydrolyzed by a two-step sequential hydrolysis method using alcalase and papain. Subsequently, antioxidant peptide fraction (IV2) was isolated and identified from the resultant hydrolysate through membrane ultrafiltration, Sephadex G-15 chromatography, and liquid chromatograph mass spectrometer (LC-MS). Animal experimentation indicated the potential of IV2 to mitigate hepatic steatosis. Moreover, IV2 could effectively reduce oxidative stress-induced damage by modulating the Keap1-Nrf2 pathway to activate downstream heme oxygenase-1 (HO-1) and NAD(P) H quinone oxidoreductase 1 (NQO1). Integrating metabolomics and transcriptomics revealed enrichment in pathways associated with glycerolipid metabolism and fatty acid β-oxidation, suggesting the principal mechanisms underlying IV2's ameliorative effects on NAFLD. Transcriptome sequencing identified 3092 up-regulated and 3010 down-regulated genes following IV2 treatment. Interaction analyses based on different lipid compositions (DELs) and differentially expressed genes (DEGs) indicated that IV2 primarily alleviated hepatic steatosis by modulating peroxisome proliferator-activated receptor α (PPAR-α) related pathways, thereby augmenting fatty acid β-oxidation within liver cells. These results indicate that IV2 shows potential in improving high-fat diet (HFD)-induced NAFLD, with improved fatty acid β-oxidation and reduced triglyceride biosynthesis emerging as underlying mechanisms.

Peroxymonosulfate activation over (Cu+ decorated g-C3N4)/Bi2WO6 composites with S-scheme heterojunction for RhB degradation

Catalytic activation of peroxymonosulfate (PMS) is one of the important research areas in the development of advanced oxidation technology for wastewater treatment. At present, the development of high-performance catalysts for PMS activation has become a research hotspot. Herein, (Cu+ decorated g-C3N4)/Bi2WO6 ((Cu+/g-C3N4)/Bi2WO6) composite was prepared successively by calcination and solvothermal method, which was used in visible light-assisted PMS activation for the degradation of rhodamine B (RhB). The construction of S-scheme heterojunctions between Cu+ decorated g-C3N4 and Bi2WO6 facilitated the activation of PMS, leading to a RhB degradation ratio of 98.46 % within 50 min. The corresponding reaction kinetic was significantly improved by a factor of 2.32 and 7.94 compared with that of CCN and BWO, respectively. The enhanced performance of the (Cu+/g-C3N4)/Bi2WO6 can be attributed to the successful construction of S-scheme heterojunction, which facilitates the separation of photogenerated carriers and accelerates the PMS activation to produce abundant active species. Furthermore, the (Cu+/g-C3N4)/Bi2WO6 composites exhibit strong stability and reusability following three cycles. In addition, the intermediates produced during the RhB degradation process were investigated by liquid chromatograph mass spectrometer (LC-MS). This study provides a new insight into the preparation of S-scheme photocatalysts for the photocatalytic-PMS activation system that will achieve efficient removal of dyes from wastewater.

Occurrence and profiles of bisphenol A and its analogues in marine shrimps from Tianjin and their implications for human exposure

Bisphenols, a class of compounds with endocrine-disrupting characteristics, encompassing its effects on reproduction, nervous and immune systems. Diet represents the principal route of bisphenols intake, prompting concerns about dietary exposure. In this study, the occurrence and content of nine bisphenols in 96 marine shrimps from Tianjin were detected and analysed using ultra high performance liquid chromatography mass spectrometer and solid-phase extraction. The analytical method was validated to demonstrate reliable analytical performance. The results revealed that bisphenol A was detected in the majority of the samples, with the highest detection rate among the nine bisphenols, followed by bisphenol S, while the remaining bisphenols were not detected. A discernible age-related trend was identified in the assessment of dietary exposure, whereby exposure levels increased with decreasing age. Furthermore, regional discrepancies were observed, yet no gender-based distinctions were discerned. The estimated daily intake to total bisphenols through marine shrimp in this study was lower than the current tolerable daily intake set by the European Food Safety Authority, and it can be assumed that the risk of exposure to the nine bisphenols investigated through dietary is acceptable. However, given the recently proposed draft values by the European Food Safety Authority and the higher exposures observed in children and teenagers, further surveillance and dietary guidance is required. To our knowledge, this is the first study in Tianjin to evaluate the health risks associated with bisphenol A and its analogues via marine shrimp, which can monitor dietary safety and protect population health.

Efficiency and mechanistic insights of photocatalytic degradation and sterilization utilizing g-C3N4/WO3-x Z-scheme heterojunction under full-spectrum light

Designing highly efficient full-spectrum responsive Z-scheme heterojunction for wastewater decontamination and disinfection is urgently essential. Herein, a novel Z-scheme g-C3N4/WO3-x heterojunction was successfully synthesized by a facile wet impregnation method for the highly efficient photocatalytic degradation and sterilization. Benefiting from the accelerated charge separation, broadened light absorption to near infrared region, and appropriate band energy, optimized g-C3N4/WO3-x heterojunction exhibited photocatalytic degradation of 86.3 % and 97.4 % tetracycline in 2 h under visible light and full-spectrum light, respectively. Meanwhile, 6.5-log of Escherichia coli strains could also be inactivated by optimized g-C3N4/WO3-x heterojunction within 45 min and 15 min under visible light and full-spectrum light, respectively. Impressively, it also exhibited desirable anti-interference ability and satisfying photocatalytic degradation capacity with various key experimental parameters. The plausible degradation pathways of TC and the main active species involved in the photocatalysis were investigated based on liquid chromatography-mass spectrometer (LC-MS) and electron paramagnetic resonance (EPR) analysis. Mineralization process of TC was further confirmed by three-dimensional fluorescence spectra (3D EMMs). Besides, Toxicity Estimation Software Tool (T.E.S.T.) and Ecological Structure-Activity Relationships (ECOSAR) program were applied to predict the noxiousness of TC and its potential intermediates. The results suggested that the excitotoxicity of intermediates generally decreased relative to the pristine TC. Finally, we propose a plausible enhancement mechanism of photocatalytic degradation and sterilization. This work sheds a new light on the design of novel full-spectrum responsive Z-scheme heterojunction for environmental remediation.

TGM2-Mediated Autophagy Contributes to the Radio-Resistance of Non-Small Cell Lung Cancer Stem-like Cells.

Objectives: Cancer cells with 'stemness' are generally resistant to chemoradiotherapy. This study aims to compare the differences in radiation sensitivity of A549 and CD44+A549 stem-like cells to X-rays and carbon ion radiation (C-ions), and to find a target that can kill cancer stem-like cells (CSCs) of non-small cell lung cancer (NSCLC). Methods: The study used two cell lines (A549 and CD44+A549). The tumorigenicity of cells was tested with animal experiments. The cells were irradiated with X-rays and C-ions. Cell viability was detected using the CCK-8 and EdU assay. A liquid chromatograph-mass spectrometer (LC-MS) helped detect metabolic differences. Protein and mRNA expression were detected using a Western blot, reverse transcription-quantitative (RT-qPCR), and PCR array. The autophagic activity was monitored with a CYTO-ID® Autophagy Detection Kit 2.0. Immunofluorescence and co-immunoprecipitation helped to observe the localization and interaction relationships. Results: First, we verified the radio-resistance of CD44+A549 stem-like cells. LC-MS indicated the difference in autophagy between the two cells, followed by establishing a correlation between the radio-resistance and autophagy. Subsequently, the PCR array proved that TGM2 is significantly upregulated in CD44+A549 stem-like cells. Moreover, the TGM2 knockdown by small interfering RNA could decrease the radio-resistance of CD44+A549 cells. Bioinformatic analyses and experiments showed that TGM2 is correlated with the expression of CD44 and LC3B. Additionally, TGM2 could directly interact with LC3B. Conclusions: We established the CD44-TGM2-LC3 axis: CD44 mediates radio-resistance of CD44+A549 stem-like cells through TGM2 regulation of autophagy. Our study may provide new biomarkers and strategies to alleviate the radio-resistance of CSCs in NSCLC.

Liquid Chromatography Mass Spectrometric Method and a Fluorometric 96 Well Plate Assay for Determination of Thiamine in Salmonid Eggs.

Thiamine deficiency is a large contributor to reduced reproduction success among salmonids throughout the northern hemisphere. In Scandinavia, this reproduction disorder is known as M74; while in North America, it is known as early mortality syndrome (EMS). The disorder fluctuates in magnitude from year to year. During years with high prevalence of the disorder, salmonid hatcheries that stock various aquatic systems to maintain the population size experience difficulties filling their quotas without thiamine treatment of alevins. The disorder is monitored both by observing the survival rate and by measuring the thiamine content of prefertilized eggs in the hatcheries. Here, a simple extraction procedure is presented, which allows for quantitative determination of the various phosphorylated forms of thiamine using liquid chromatography mass spectrometry but also allows for extraction in 96 deep-well plates and measurement of the total thiamine content using fluorescence monitoring with a plate reader, following oxidation of thiamine to thiochrome. The latter procedure could also be integrated into a highly portal system where the thiochrome is determined using the DeNovix QFX analyzer. The newly developed extraction procedure and cleanup method for fluorescence measurement represent the most versatile and simple methods to date for monitoring of thiamine in salmonid eggs. The methods produced accurate and precise data with quantification limits below the limit where the deficiency causes 100% lethality.

Unbalanced Expression of Structural Genes in Carotenoid Pathway Contributes to the Flower Color Formation of the Osmanthus Cultivar 'Yanzhi Hong'.

Carotenoids are important natural pigments that are responsible for the fruit and flower colors of many plants. The composition and content of carotenoid can greatly influence the color phenotype of plants. However, the regulatory mechanism underling the divergent behaviors of carotenoid accumulation, especially in flower, remains unclear. In this study, a new cultivar Osmanthus fragrans 'Yanzhi Hong' was used to study the regulation of carotenoid pigmentation in flower. Liquid chromatograph-mass spectrometer (LC-MS) analysis showed that β-carotene, phytoene, lycopene, γ-carotene, and lutein were the top five pigments enriched in the petals of 'Yanzhi Hong'. Through transcriptome analysis, we found that the expression of the structural genes in carotenoid pathway was imbalanced: most of the structural genes responsible for lycopene biosynthesis were highly expressed throughout the flower developmental stages, while those for lycopene metabolism kept at a relatively lower level. The downregulation of LYCE, especially at the late developmental stages, suppressed the conversion from lycopene to α-carotene but promoted the accumulation of β-carotene, which had great effect on the carotenoid composition of 'Yanzhi Hong'. Ethylene response factor (ERF), WRKY, basic helix-loop-helix (bHLH), v-myb avian myeloblastosis viral oncogene homolog (MYB), N-Acetylcysteine (NAC), auxin response factor (ARF), and other transcription factors (TFs) have participated in the flower color regulation of 'Yanzhi Hong', which formed co-expression networks with the structural genes and functioned in multiple links of the carotenoid pathway. The results suggested that the cyclization of lycopene is a key link in determining flower color. The modification of the related TFs will break the expression balance between the upstream and downstream genes and greatly influence the carotenoid profile in flowers, which can be further used for creating colorful plant germplasms.

BiOBr/FeMoO4 composite achieves oxygen vacancy concentration adjustment to promote persulfate activation degradation of organic pollutants in saline water

The investigation about the mechanism of crystal plane regulation on the generation of oxygen vacancies remains a challenge. In this paper, BiOBr/FeMoO4 composites were synthesized by precise control of crystal plane growth, and it exhibited the enhanced concentration of oxygen vacancies due to lower formation energy of oxygen vacancies. The composite performs higher photo-Fenton-like ability for degrading oxytetracycline hydrochloride (OTC). Structural analyses and theoretical calculations reveal that crystal plane regulation induces significant changes in oxygen vacancy concentration. The BiOBr/FeMoO4/peroxydisulphate (PDS) /light system, which dominated by the non-radical pathway, degraded 96.8 % ± 1.0 % of OTC within 30 min. The activation mechanism of the system and the degradation pathway of OTC were elucidated. The intermediates in the degradation process of OTC were evaluated using liquid chromatograph-mass spectrometer (LC-MS), toxicity evaluation software tool (T.E.S.T) and soybean germination experiments. This work offers novel insights into the pivotal role of crystal plane directional regulation in the quantitative generation of oxygen vacancies.

Enhanced visible light responsive piezoelectric photocatalysis based on Bi2S3 coated BaTiO3 nanorods heterostructures

Although the presence of the built-in electric field will solve the problem of carrier complexation in photocatalytic systems to some extent. However, free carriers will quickly shield the stabilized electric field and lose its effect. Therefore, how to introduce the dynamic piezoelectric field into the photocatalytic system has become an imminent problem. Herein, we developed an overcoated, visible light responsive, piezoelectric-assisted photocatalytic system by depositing Bi2S3 photocatalysts with a narrow-band system onto the surface of highly piezo-responsive BaTiO3 nanorods (BTO NRs). The heterojunction structure, bound by Bi-O chemical bonding, enhances carrier transport efficiency under the influence of the piezoelectric field. In the degradation experiments, the first-order rate constant for the degradation of chlortetracycline hydrochloride (CTC) in the BTO NRs/Bi2S3 system with the optimal complex ratio was 0.0276 min−1, which was 3.1 and 7.8 times higher than that of BTO NRs and Bi2S3, respectively. Additionally, we deduced the degradation pathways of CTC through a combination of Density functional theory (DFT) calculations and Liquid Chromatograph Mass Spectrometer (LC-MS), evaluating the toxicity of the intermediates. This complex system, featuring a highly photo-responsive semiconductor as a photo-acceptor deposited on a piezoelectric semiconductor surface providing a dynamic built-in electric field, enhances carrier separation efficiency under optimal light energy utilization conditions. These findings present novel and effective strategies for addressing two primary challenges in photocatalytic systems: low spectral utilization and significant photogenerated carrier complexation.

Transcriptomics and metabolomics analyses of graphene oxide toxicity on porcine alveolar macrophages

Graphene oxide (GO) is a type of nanomaterial widely used in tissue engineering, photocatalysis, and biomedicine. GO has been found to produce adverse effects on a broad range of cells and tissues. However, the molecular mechanisms underlying GO toxicity still remain to be explored. In this study, using porcine alveolar macrophages as a study model, we explored the toxic effects of GO and performed genome-wide detection of genes and metabolites associated with GO exposure using RNA-seq and liquid chromatograph mass spectrometer techniques. GO exposure significantly inhibited cell viability and induced apoptosis and oxidative stress in porcine alveolar macrophages. Further, GO exposure promoted cellular inflammation by upregulating the expression of pro-inflammatory cytokines (IL-6, IL-8, and IL-12). Transcriptomic analysis of GO-exposed cells revealed 424 differentially expressed genes. Functional enrichment analysis showed that the differentially expressed genes were significantly enriched in the pathways of Ribosome and oxidative phosphorylation (OXPHOS). In addition, metabolic analysis identified 203 differential metabolites, and these metabolites were significantly enriched in biosynthesis of cofactors, purine metabolism, and nucleotide metabolism. Integrative analyses of transcriptome and metabolome showed that OXPHOS was the most significantly enriched pathway and the involved genes were downregulated. This study revealed the toxic effects of GO on porcine alveolar macrophages and provided global insights to the metabolomic and transcriptomic alterations related to GO exposure. The results contributed to our understanding of the molecular mechanism of GO, and may further promote the detection of biomarkers for the prediction and control of GO toxicity.

Decomposition of tetracycline with peroxymonosulfate activated by an ordered hierarchical pores Fe-N/C catalyst rich in FeNx sites: Insights into singlet oxygen mechanism

FeNx sites show significant superiority in the degradation of antibiotic contaminants, but there are difficulties in successfully constructing highly dispersed FeNx sites. In this work, Fe-N/C catalysts doped with hierarchical ordered pores structure and FeNx active sites were prepared by metal node exchange strategy. Among them, Fe-N/C-2 had more excite species for peroxymonosulfate (PMS) owing to the synergy between Fe and N coordination, which achieved 89.36 % tetracycline (TC) removal rate within 20 min, and its reaction kinetic (k = 0.245 min−1) is 7.66 times more than original PMS system. Moreover, the material exhibited excellent activation performance in a variety of complex water environments, such as the coexistence of multiple anions, strong acids and alkalis (pH = 2.90 ∼ 10.73), and natural organic matter interference. Chemical bond detection confirmed the successful formation of FeNx sites, free radical trapping experiments showed that FeNx was the main catalytic site, and 1O2 was the main active component. Furthermore, the reaction principle of the FeNx site catalyzing the production of 1O2 from PMS was revealed in detail through mechanism exploration. Next, the probable pathways of TC decomposition were elaborated in terms of the intermediates identified by Liquid Chromatograph Mass Spectrometer (LC-MS). This study emphasized the significant potential of synthesizing Fe-N/C catalysts with activated FeNx sites, elucidated mechanism of FeNx in PMS activation, and confirmed its feasibility in TC degradation.

New antidepressant mechanism of Yueju Pill: Increasing ghrelin level by inhibiting gastric mTOR/S6K signaling pathway and sensitizing hippocampal GHS-R

Background and aimYueju Pill (YJ) not only has good antidepressant effect but also can effectively treat digestive system diseases. However,it remains unclear whether the mechanism of antidepressant action of YJ is related to the peripheral digestive system. The purpose of this study was to elucidate the antidepressant mechanism of YJ on ghrelin level based on gastric mTOR/S6K signal pathway and sensitized hippocampal Ghrelin/GHS-R system in CUMS mice. Experimental procedureThe depression model was induced by chronic unpredictable mild stress (CUMS) and social isolation. The antidepressant effect of YJ was observed by behavioral experiment and hemodynamic experiments. Ghrelin levels in in hippocampus and blood were measured by Elisa kit, and the mRNA of ghrelin in mice stomach was measured by Real-time Quantitative PCR (RT-qPCR). The activation level of gastric mTOR/S6K signal pathway was detected by Western Blot (WB). Rapamycin (Rapa) and L-Leucine (L-Leu) were used to verify the effects of YJ on the synthesis and release of ghrelin. The activity of GHS-R in hippocampus was observed by immunofluorescence. Hippocampal neuronal damage was evaluated by HE staining and Nissl staining. The level of central neurotransmitter was measured by liquid chromatograph mass spectrometer (LC-MS). Results and conclusionYJ ameliorates CUMS-induced depressive-like behavior by inhibiting the gastric mTOR/S6K signaling pathway and increasing GHR expression in the mouse stomach. However, these effects of YJ could be resisted by L-Leu (a mTOR receptor agonist). Further studies have shown that YJ can sensitize the Ghrelin/GHS-R system in the hippocampus, with significant neuroprotective effects, and is also involved in regulating the levels of key neurotransmitters (5-hydroxytryptamine, Dopamine and γ-aminobutyric acid) in depressive-like states.

Novel metabolic and lipidomic biomarkers of sarcopenia.

The pathophysiology of sarcopenia is complex and multifactorial and has not been fully elucidated. The impact of resistance training and nutritional support (RTNS) on metabolomics and lipodomics in older adults with sarcopenia remains uncertain. This study aimed to explore potential biomarkers of sarcopenia and clinical indicators of RTNS in older sarcopenic adults. Older individuals diagnosed with sarcopenia through routine health checkups at a community hospital were recruited for a 12-week randomized controlled trial focusing on RTNS. Plasma metabolomic and lipidomic profiles of 45 patients with sarcopenia and 47 matched controls were analysed using 1H-nuclear magnetic resonance (1H-NMR) and liquid chromatography-mass spectrometer (LC-MS). At baseline, the patient and control groups had similar age, sex, and height distribution. The patient group had significantly lower weight, BMI, grip strength, gait speed, skeletal muscle index, lean mass of both the upper and lower limbs, and lower limb bone mass. There was a significant difference in 12 metabolites between the control and patient groups. They are isoleucine (patient/control fold change [FC]=0.86±0.04, P=0.0005), carnitine (FC=1.05±0.01, P=0.0110), 1-methylhistamine/3-methylhistamine (FC=1.24±0.14, P=0.0039), creatinine (FC=0.71±0.04, P<0.0001), carnosine (FC=0.71±0.04, P=0.0007), ureidopropionic acid (FC=0.61±0.10, P=0.0107), uric acid (FC=0.88±0.03, P=0.0083), PC (18:2/20:0) (FC=0.69±0.03, P=0.0010), PC (20:2/18:0) (FC=0.70±0.06, P=0.0014), PC (18:1/20:1) (FC=0.74±0.05, P=0.0015), PI 32:1 (FC=4.72±0.17, P=0.0006), and PI 34:3 (FC=1.88±0.13, P=0.0003). Among them, carnitine, 1-methylhistamine/3-methylhistamine, creatinine, ureidopropionic acid, uric acid, PI 32:1, and PI 34:3 were first identified. Notably, PI 32:1 had highest diagnostic accuracy (0.938) for sarcopenia. 1-Methylhistamine/3-methylhistamine, carnosine, PC (18:2/20:0), PI 32:1, and PI 34:3 levels were not different from the control group after RTNS. These metabolites are involved in amino acid metabolism, lipid metabolism, and the PI3K-AKT/mTOR signalling pathway through the ingenuity pathway analysis. These findings provide information on metabolic changes, lipid perturbations, and the role of RTNS in patients with sarcopenia. They reveal new insights into its pathological mechanisms and potential therapies.

Mechanistic investigation of azo dye removal from carbon-deficient dyeing wastewater using horizontal-vertical constructed wetlands

Azo dye degradation can be achieved by simulating a series of anaerobic and aerobic conditions within the constructed wetland (CW) system. The current investigation evaluated the effectiveness of a baffled horizontal-vertical CW system, planted with Typha angustifolia, simulating anaerobic-aerobic conditions to treat carbon-deficient synthetic dyeing wastewater containing 100 mg/L Reactive Yellow 145 (RY145) azo dye. In the absence of an available carbon source in dyeing wastewater, an optimum quantity of sodium acetate was supplemented as the substrate for microbial degradation of RY145. Influent dyeing wastewater characteristics were 5555 ADMI colour, 461 mg/L chemical oxygen demand (COD) and 39 mg/L total nitrogen (TN). During the operation period, the CW system achieved 97% colour, 87% COD, 95% ammonium nitrogen (NH4+-N) and 71% TN removals at 4 d hydraulic retention time (HRT). Favourable environmental conditions, such as low redox conditions and substrate availability in horizontal CW, contributed to a significant reduction in colour (96%). Most TN reduction (67%) happened in horizontal CW by denitrification and plant assimilation. The metagenomic study revealed that Proteobacteria, Bacteroidetes, Chloroflexi and Firmicutes were responsible for pollutant degradation within horizontal CW. The UV–visible spectra and high-resolution liquid chromatograph mass spectrometer (HR-LCMS) analysis confirmed that dye degradation intermediates generated from the breakage of azo bonds were eliminated in vertical CW with high redox conditions. The results of the phytotoxicity and fish toxicity experiments demonstrated a substantial toxicity reduction in the CW system-treated effluent.

Research Note: Metabolomics revealed the causes of the formation of chicken structural blue earlobes

The earlobes of chickens exhibit a range of colors, but there has been relatively little research on the formation of structural blue earlobes. Previous results showed that the structural color earlobes were related to the interplay between melanin and collagen in light reflection. To investigate the metabolic differences in these earlobe colors, we conducted nontargeted liquid chromatograph mass spectrometer (LC-MS) for metabolomic sequencing on structural blue (Green and Blue groups) and nonstructural color (Black group) earlobes tissue of Jiangshan black-bone chickens. The content detection in earlobe tissues of different groups shows that there were significant differences in melanin and collagen content between the Black and Green group. The metabolome identified a total of 6,102 mass spectroscopic peaks and ultimately identified 919 annotated metabolites. Variable importance in the projection (VIP) analysis identified the common differential expressed metabolites (DMs) "Tyr Thr Ala Glu" among the 3 groups. By combining those DMs with differentially expressed genes (DEGs) in our previous transcriptome data from the same sample, and associated with KEGG pathway analysis, multiple pathways related to melanogenesis and collagen metabolism were enriched across the 3 groups. By analyzing the metabolites and genes in these pathways, as well as the interaction network diagram of DEGs, we identified some key genes, Wnt Family Member 6 (WNT6), Transcription Factor 7 (TCF7), Proopiomelanocortin (POMC) and Calcium/Calmodulin Dependent Protein Kinase II Alpha (CAMK2A), and some key DMs like DG (11M3/9M5/0:0) and gentisic acid. The differential gene expression and metabolic levels affect the production of melanin and collagen, leading to differences in the content in melanin and the thickness of the collagen layer between earlobe colors, while the thickness of the collagen layer could affect light scattering, ultimately resulting in different colored earlobes in Jiangshan black-bone chickens.

Targeted screening and profiling of massive components of colistimethate sodium by two-dimensional-liquid chromatography-mass spectrometry based on self-constructed compound database

In-depth study of the components of polymyxins is the key to controlling the quality of this class of antibiotics. Similarities and variations of components present significant analytical challenges. A two-dimensional (2D) liquid chromatography-mass spectrometric (LC-MS) method was established for screening and comprehensive profiling of compositions of the antibiotic colistimethate sodium (CMS). A high concentration of phosphate buffer mobile phase was used in the first-dimensional LC system to get the components well separated. For efficient and high-accuracy screening of CMS, a targeted method based on a self-constructed high resolution mass spectrum database of CMS components was established. The database was built based on the commercial MassHunter Personal Compound Database and Library (PCDL) software and its accuracy of the compound matching result was verified with 6 known components before being applied to genuine sample screening. On this basis, the unknown peaks in the CMS chromatograms were deduced and assigned. The molecular formula, group composition and origins of a total of 99 compounds, of which the combined area percentage accounted for more than 95% of CMS components, were deduced by this 2D-LC-MS method combined with the MassHunter PCDL. This profiling method was highly efficient and could distinguish hundreds of components within 3 h, providing reliable results for quality control of this kind of complex drugs.

Study on the fourof Pfaffia glomerata based on the metabolomics technology and comparison of Dangshen () in the equivalent substitution prescription.

To explore the four Qiof Pfaffia glomerata (PG) and endow this foreign folk herb with the properties of Chinese medicine, make it Chinese medicinal and localized, and could be used as a Chinese medicine. The normal group, six cold herb groups, six hot herb groups, six cool herb groups, PG prescription group (PGPG), and Dangshen (Radix Codonopsis) prescription group (CPPG) were prepared with corresponding concentrations of water extracts, these herb extracts were administered by gavage to Sprague-Dawley rats, and the 12 h urine at night on the 29th day of the SD rats in each group were collected, Liquid Chromatograph Mass Spectrometer system was used to analyze them, the best discriminant models for the medicinal properties of cold-hot and cold-cool were set up, so as to the medicinal properties of PGPG, CPPG and PG were predicted. Based on the Progenesis QI, Human Metabolome Database, Kyoto Encyclopedia of Genes and Genomes, MetaboAnalyst 5.0 database, we enriched metabolic pathway and classification mechanism of medicinal properties of cold-cool Chinese herbs and the molecular mechanism of PG prescription. We established a best model of cold-hot herbal discrimination in the positive ion mode, then the probability that PGPG was predicted cold property was 88.9%. Furthermore, a model of cold-cool herbal discrimination was established, then the probability of PGPG containing the cool property was 77.8%. In addition, typical cold and cool herbs mainly affected nine biomarkers such as tyrosine-proline, (R)-3',7-Dihydroxy-2',4'-dimethoxyisoflavane in rats. The regulation trend of PGPG on markers was basically as same as the cool herbs and mainly involved in regulating the two pathways of cytochrome P450 and purine metabolism. The results showed PGPG had a cool medicinal property as same as CPPG, and the regulation trend of PGPG on markers was consistent with cool herbs. Therefore, the medicine properties PG and CP should be consistent, and the Traditional Chinese Medicine property of PG was predicted to be neutral.

Chemically bonded CdBiO2Br/BiOI heterojunction with strong interfacial electric field for enhanced photocatalysis

Semiconductor-based photocatalysis shows a large potential for contaminant purification, however, a single semiconductor photocatalyst suffers from low utilization efficiency of photogenerated carriers, leading to inferior photocatalytic activity. Herein, CdBiO2Br@BiOI (CBOB@BiOI) heterojunction with a close interface interaction is prepared by a facile precipitation method at the ambient atmosphere. X-ray photoelectron spectroscopy and Density Functional Theory calculations on work function consistently disclose that the formation of CdBiI bonds at the interface induces the electron migration from BiOI to CdBiO2Br, which allows CBOB@BiOI heterojunction to have a strong interfacial electric field (IEF) for efficiently separating photocharges. In-situ Kelvin-probe Force Microscopy measurement demonstrates that CBOB@BiOI heterojunction shows a larger surface potential than CdBiO2Br and BiOI in dark and a potential decrease under illumination, which can be ascribed to the photoinduced electron migration within heterojunction driven by the IEF. Thus, the CBOB@BiOI heterojunction shows a much more efficient photocatalytic activity for the breakdown of tetracycline hydrochloride (TC), which is 2.44 and 3.99 times higher than CdBiO2Br and BiOI, respectively. Electron Paramagnetic Resonance test also confirms that the CBOB@BiOI heterojunction produces much more superoxide radicals and holes as reactive species than CdBiO2Br and BiOI. Furthermore, the latent degradation pathways of TC are investigated by Liquid Chromatograph Mass Spectrometer. This work may provide new ideas for constructing intimately bonded heterojunction photocatalysts with a strong IEF for efficient photocatalytic activity.

Platelet-derived Growth Factor Receptor-α Induces Contraction Knots and Inflammatory Pain-like Behavior in a Rat Model of Myofascial Trigger Points.

Myofascial trigger points (MTrPs) are the primary etiological characteristics of chronic myofascial pain syndrome. Receptor tyrosine kinases (RTKs) are associated with signal transduction in the central mechanisms of chronic pain, but the role of RTKs in the peripheral mechanisms of MTrPs remains unclear. The current study aimed to identify RTKs expression in MTrPs and elucidate the molecular mechanisms through which platelet-derived growth factor receptor-α (PDGFR-α) induces contraction knots and inflammatory pain-like behavior in a rat model of myofascial trigger points. MTrPs tissue samples were obtained from the trapezius muscles of patients with myofascial pain syndrome through needle biopsy, and PDGFR-α activation was analyzed by microarray, enzyme-linked immunosorbent assay, and histological staining. Sprague-Dawley rats (male and female) were used to investigate PDGFR-α signaling, assessing pain-like behaviors with Randall-Selitto and nest-building tests. Muscle fiber and sarcomere morphologies were observed using histology and electron microscopy. The PDGFR-α binding protein was identified by coimmunoprecipitation, liquid chromatograph mass spectrometer, and molecular docking. PDGFR-α-related protein or gene levels, muscle contraction, and inflammatory markers were determined by Western blot and reverse-transcription quantitative polymerase chain reaction. PDGFR-α phosphorylation levels were elevated in the MTrPs tissues of individuals with trapezius muscle pain and were positively correlated with pain intensity. In rats, PDGFR-α activation caused pain-like behaviors and muscle contraction via the Janus kinase 2/signal transducer and activator of transcription-3 (JAK2/STAT3) pathway. JAK2/STAT3 inhibitors reversed the pain-like behaviors and muscle contraction induced by PDGFR-α activation. Collagen type I α 1 (COL1A1) binds to PDGFR-α and promotes its phosphorylation, which contributed to pain-like behaviors and muscle contraction. COL1A1-induced phosphorylation of PDGFR-α and the subsequent activation of the JAK2/STAT3 pathway may induce dysfunctional muscle contraction and increased nociception at MTrPs.

Boosted antibiotic elimination over 2D/2D mesoporous CeO2/BiOCl S-scheme photocatalyst

Exploring efficient photocatalysts holds significant importance for the prevention and treatment of antibiotic pollution. This research reports a 2D/2D CeO2/BiOCl S-scheme photocatalyst constructed by mesoporous CeO2 and BiOCl nanosheets (NSs). Efficient charge separation and transfer was achieved by the synergistic effect of large-area contact and S-scheme charge transfer mechanism, which significantly enhanced the photocatalytic performance of CeO2/BiOCl. Experimental results demonstrated that the CeO2/BiOCl photocatalyst displayed superior photocatalytic activity to the individual components, which could efficiently eliminate four types of antibiotics, namely tetracycline (TC), chlortetracycline (CTC), doxycycline (DOC), and oxytetracycline (OTC) under UV light irradiation. Particularly, the sample of 0.4CeO2/BiOCl exhibited the highest catalytic efficiency, with a TC degradation rate constant of 0.01524 min−1, which was 1.85 and 22.74 times higher than that of single CeO2 and BiOCl, respectively. Moreover, liquid chromatograph-mass spectrometer (LC-MS) analysis is employed to reveal the degradation pathways of TC. This work offers an insightful idea for the fabrication of novel S-scheme photocatalysts applied in environmental purification.
