Nuclear SUN1 stabilizes endothelial cell junctions via microtubules to regulate blood vessel formation.

Endothelial cells line all blood vessels, where they coordinate blood vessel formation and the blood-tissue barrier via regulation of cell-cell junctions. The nucleus also regulates endothelial cell behaviors, but it is unclear how the nucleus contributes to endothelial cell activities at the cell periphery. Here, we show that the nuclear-localized linker of the nucleoskeleton and cytoskeleton (LINC) complex protein SUN1 regulates vascular sprouting and endothelial cell-cell junction morphology and function. Loss of murine endothelial Sun1 impaired blood vessel formation and destabilized junctions, angiogenic sprouts formed but retracted in SUN1-depleted sprouts, and zebrafish vessels lacking Sun1b had aberrant junctions and defective cell-cell connections. At the cellular level, SUN1 stabilized endothelial cell-cell junctions, promoted junction function, and regulated contractility. Mechanistically, SUN1 depletion altered cell behaviors via the cytoskeleton without changing transcriptional profiles. Reduced peripheral microtubule density, fewer junction contacts, and increased catastrophes accompanied SUN1 loss, and microtubule depolymerization phenocopied effects on junctions. Depletion of GEF-H1, a microtubule-regulated Rho activator, or the LINC complex protein nesprin-1 rescued defective junctions of SUN1-depleted endothelial cells. Thus, endothelial SUN1 regulates peripheral cell-cell junctions from the nucleus via LINC complex-based microtubule interactions that affect peripheral microtubule dynamics and Rho-regulated contractility, and this long-range regulation is important for proper blood vessel sprouting and junction integrity.

Force transmission and SUN-KASH higher-order assembly in the LINC complex models.

The linker of the nucleoskeleton and cytoskeleton (LINC) complex comprises of SUN (Sad-1 and UNC-84) and KASH (Klarsicht, ANC-1, SYNE homology) domain proteins, which connect the inner and outer nuclear membrane, thereby mediating the transfer of physical forces between the cytoskeleton and the nucleoskeleton. While impaired SUN and KASH proteins are associated with structural and functional cell defects, the precise mechanism of force transduction and the assembly of these proteins remain unknown. Recent studies suggested a higher order assembly of SUN and KASH instead of a more widely accepted linear trimer model for the LINC complex. In the present study, we use molecular dynamics simulations to investigate the mechanism of force transfer across the two proposed models of LINC complex assembly, namely the 3:3 linear trimer model and the 6:6 higher-order model. Employing steered molecular dynamics simulations with forces at different rates and directions on various structures, we examine the structural stability of the two models under various biologically relevant conditions. Our results suggest that both models can withstand and transfer significant levels of force while retaining their structural integrity. However, some of the structures were more stable under specific forces compared to other structures. Slower pulling rates resulted in higher mean square fluctuations of the proteins compared to the fast pulling. Interestingly, the higher-order assembly tends to provide an additional range of motion flexibility and might be more suitable for describing the interaction between SUN and KASH under compressive and shear forces. These findings offer insights into how the SUN and KASH proteins maintain the structural integrity of the nuclear membrane.

Two-step regulation of centromere distribution by condensin II and the nuclear envelope proteins.

The arrangement of centromeres within the nucleus differs among species and cell types. However, neither the mechanisms determining centromere distribution nor its biological significance are currently well understood. In this study, we demonstrate the importance of centromere distribution for the maintenance of genome integrity through the cytogenic and molecular analysis of mutants defective in centromere distribution. We propose a two-step regulatory mechanism that shapes the non-Rabl-like centromere distribution in Arabidopsis thaliana through condensin II and the linker of the nucleoskeleton and cytoskeleton (LINC) complex. Condensin II is enriched at centromeres and, in cooperation with the LINC complex, induces the scattering of centromeres around the nuclear periphery during late anaphase/telophase. After entering interphase, the positions of the scattered centromeres are then stabilized by nuclear lamina proteins of the CROWDED NUCLEI (CRWN) family. We also found that, despite their strong impact on centromere distribution, condensin II and CRWN proteins have little effect on chromatin organization involved in the control of gene expression, indicating a robustness of chromatin organization regardless of the type of centromere distribution.

Novel bi-allelic variants in KASH5 are associated with meiotic arrest and non-obstructive azoospermia.

KASH5 is an essential component of the LINC (linker of the nucleoskeleton and cytoskeleton) complex that regulates chromosome movements and nuclear envelope (NE) remodeling in mouse spermatocytes during meiosis prophase I, but its expression and function in human cells, as well as its association with male infertility are largely unknown. In this study, a novel heterozygous copy number variation (CNV) (seq [GRCh37] del(19) (19q13.33) chr19: g.49894043-49903011del) and a heterozygous loss of function variant (NM_144688: c.979_980del: p.R327Sfs*21) in human KASH5 were identified in a non-obstructive azoospermia (NOA)-affected patient and in his infertile sister by whole-exome sequencing and CNV array. Spermatogenesis in the proband was arrested at zygotene-like stage with a deficiency in homolog pairing and synapsis. KASH5 protein expression in human spermatocytes was evaluated and reported first in this study. Single-cell RNA sequencing demonstrated that the LINC complex and associated genes in human and mouse shared a similar expression pattern, indicating a conserved mechanism in the regulation of chromosome movements and NE remodeling. Kash5 knockout mouse displayed similar phenotypes, including a meiotic arrest at a zygotene-like stage and impaired pairing and synapsis. Collectively, we have identified novel rare variants within human KASH5 in patients with NOA and meiosis arrest. Our study expands the knowledge of KASH5 and associated proteins in regulating human meiosis prophase I progress and provides new insight into the genetic etiology of NOA.

The LINC Complex Assists the Nuclear Import of Mechanosensitive Transcriptional Regulators.

Mechanical forces play pivotal roles in directing cell functions and fate. To elicit gene expression, either intrinsic or extrinsic mechanical information are transmitted into the nucleus beyond the nuclear envelope via at least two distinct pathways, possibly more. The first and well-known pathway utilizes the canonical nuclear transport of mechanoresponsive transcriptional regulators through the nuclear pore complex, which is an exclusive route for macromolecular trafficking between the cytoplasm and nucleoplasm. The second pathway depends on the linker of the nucleoskeleton and cytoskeleton (LINC) complex, which is a molecular bridge traversing the nuclear envelope between the cytoskeleton and nucleoskeleton. This protein complex is a central component in mechanotransduction at the nuclear envelope that transmits mechanical information from the cytoskeleton into the nucleus to influence the nuclear structure, nuclear stiffness, chromatin organization, and gene expression. Besides the mechanical force transducing function, recent increasing evidence shows that the LINC complex plays a role in controlling nucleocytoplasmic transport of mechanoresponsive transcriptional regulators. Here we discuss recent findings regarding the contribution of the LINC complex to the regulation of intracellular localization of the most-notable mechanosensitive transcriptional regulators, β-catenin, YAP, and TAZ.

Interphase microtubules in nuclear organization and genome maintenance.

Microtubules are major cytoskeletal components mediating fundamental cellular processes, including cell division. Recent evidence suggests that microtubules also regulate the nucleus during the cell cycle's interphase stage. Deciphering such roles of microtubules should uncover direct crosstalk between the nucleus and cytoplasm, impacting genome function and organismal health. Here, we review emerging roles for microtubules in interphase genome regulation. We explore how microtubules exert cytoplasmic forces on the nucleus or transport molecular cargo, including DNA, into or within the nucleus. We also describe how microtubules perform these functions by establishing transient or stable connections with nuclear envelope elements. Lastly, we discuss how the regulation of the nucleus by microtubules impacts genome organization and repair. Together, the literature indicates that interphase microtubules are critical regulators of nuclear structure and genome stability.

The SUN1 splicing variants SUN1_888 and SUN1_916 differentially regulate nucleolar structure.

The nucleolar structure is highly dynamic and strictly regulated in response to internal cues, such as metabolic rates, and to external cues, such as mechanical forces applied to cells. Although the multilayered nucleolar structure is largely determined by the liquid-like properties of RNA and proteins, the mechanisms regulating the morphology and number of nucleoli remain elusive. The linker of the nucleoskeleton and cytoskeleton (LINC) complex comprises inner nuclear membrane Sad1/UNC-84 (SUN) proteins and outer nuclear membrane-localized nesprins. We previously showed that the depletion of SUN1 proteins affects nucleolar morphologies. This study focuses on the function of SUN1 splicing variants in determining nucleolar morphology. An RNA interference strategy showed that the predominantly expressed variants, SUN1_888 and SUN1_916, were crucial for nucleolar morphology but functionally distinct. In addition, the depletion of either SUN1_888 or SUN1_916 altered the chromatin structure and affected the distribution of histone modifications. Based on these results, we propose a model in which the LINC complex plays a role in modulating nucleolar morphology and numbers via chromatin.

Mps2 links Csm4 and Mps3 to form a telomere-associated LINC complex in budding yeast.

The linker of the nucleoskeleton and cytoskeleton (LINC) complex is composed of two transmembrane proteins: the KASH domain protein localized to the outer nuclear membrane and the SUN domain protein to the inner nuclear membrane. In budding yeast, the sole SUN domain protein, Mps3, is thought to pair with either Csm4 or Mps2, two KASH-like proteins, to form two separate LINC complexes. Here, we show that Mps2 mediates the interaction between Csm4 and Mps3 to form a heterotrimeric telomere-associated LINC (t-LINC) complex in budding yeast meiosis. Mps2 binds to Csm4 and Mps3, and all three are localized to the telomere. Telomeric localization of Csm4 depends on both Mps2 and Mps3; in contrast, Mps2's localization depends on Mps3 but not Csm4. Mps2-mediated t-LINC complex regulates telomere movement and meiotic recombination. By ectopically expressing CSM4 in vegetative yeast cells, we reconstitute the heterotrimeric t-LINC complex and demonstrate its ability to tether telomeres. Our findings therefore reveal the heterotrimeric composition of the t-LINC complex in budding yeast and have implications for understanding variant LINC complex formation.

An Emerin LEM-Domain Mutation Impairs Cell Response to Mechanical Stress.

Emerin is a nuclear envelope protein that contributes to genome organization and cell mechanics. Through its N-terminal LAP2-emerin-MAN1 (LEM)-domain, emerin interacts with the DNA-binding protein barrier-to-autointegration (BAF). Emerin also binds to members of the linker of the nucleoskeleton and cytoskeleton (LINC) complex. Mutations in the gene encoding emerin are responsible for the majority of cases of X-linked Emery-Dreifuss muscular dystrophy (X-EDMD). Most of these mutations lead to an absence of emerin. A few missense and short deletion mutations in the disordered region of emerin are also associated with X-EDMD. More recently, missense and short deletion mutations P22L, ∆K37 and T43I were discovered in emerin LEM-domain, associated with isolated atrial cardiac defects (ACD). Here we reveal which defects, at both the molecular and cellular levels, are elicited by these LEM-domain mutations. Whereas ΔK37 mutation impaired the correct folding of the LEM-domain, P22L and T43I had no impact on the 3D structure of emerin. Surprisingly, all three mutants bound to BAF, albeit with a weaker affinity in the case of ΔK37. In human myofibroblasts derived from a patient’s fibroblasts, emerin ∆K37 was correctly localized at the inner nuclear membrane, but was present at a significantly lower level, indicating that this mutant is abnormally degraded. Moreover, SUN2 was reduced, and these cells were defective in producing actin stress fibers when grown on a stiff substrate and after cyclic stretches. Altogether, our data suggest that the main effect of mutation ΔK37 is to perturb emerin function within the LINC complex in response to mechanical stress.

Role of KASH domain lengths in the regulation of LINC complexes.

The linker of the nucleoskeleton and cytoskeleton (LINC) complex is formed by the conserved interactions between Sad-1 and UNC-84 (SUN) and Klarsicht, ANC-1, SYNE homology (KASH) domain proteins, providing a physical coupling between the nucleoskeleton and cytoskeleton that mediates the transfer of physical forces across the nuclear envelope. The LINC complex can perform distinct cellular functions by pairing various KASH domain proteins with the same SUN domain protein. For example, in Caenorhabditis elegans, SUN protein UNC-84 binds to two KASH proteins UNC-83 and ANC-1 to mediate nuclear migration and anchorage, respectively. In addition to distinct cytoplasmic domains, the luminal KASH domain also varies among KASH domain proteins of distinct functions. In this study, we combined in vivo C. elegans genetics and in silico molecular dynamics simulations to understand the relation between the length and amino acid composition of the luminal KASH domain, and the function of the SUN–KASH complex. We show that longer KASH domains can withstand and transfer higher forces and interact with the membrane through a conserved membrane proximal EEDY domain that is unique to longer KASH domains. In agreement with our models, our in vivo results show that swapping the KASH domains of ANC-1 and UNC-83, or shortening the KASH domain of ANC-1, both result in a nuclear anchorage defect in C. elegans.

Length of KASH Domains Affect Linc Complex Functions

The linker of the nucleoskeleton and cytoskeleton (LINC) complexes are composed of inner nuclear membrane Sad1/UNC-84 (SUN, and outer nuclear membrane Klarsicht/ANC-1/SYNE homology (KASH) proteins. LINC complexes span the physical barrier of the nuclear envelope and regulate the physical connection between the interior of the nucleus and the cytoplasm during various cellular functions. The large cytoplasmic domains of KASH proteins bind to various elements of the cytoskeleton, whereas their 10-30 amino acid KASH domains reside in the perinuclear space (PNS) where they bind to SUN proteins. Interestingly, different KASH domain proteins can independently bind to the same SUN protein to mediate distinct cellular functions. For example, in mammals the SUN protein SUN1 can transiently associate with KASH5 in the PNS to mediate meiotic chromosome movement. Alternatively, SUN1 can bind to Nesprin1/2 to mediate nuclear movement. Similarly, in Caenorhabditis elegans (C. elegans) SUN protein UNC-84 transiently binds to KASH protein UNC-83 in embryonic hypodermal cells to mediate nuclear migration. Later, the same SUN protein UNC-84 independently binds to a different KASH protein ANC-1 to anchor of the nucleus in place for several days. In this work, we combined in vivo C. elegans genetics and in silico molecular dynamics simulations to explore the role of KASH domain length on the function of the SUN-KASH complex and its response to mechanical forces. Chain reaction: LINC complexes and nuclear positioning.

Nuclear positioning plays an essential role in defining cell architecture and behaviour in both development and disease, and nuclear location frequently adjusts according to internal and external cues. For instance, during periods of migration in many cell types, the nucleus may be actively repositioned behind the microtubule-organising centre. Nuclear movement, for the most part, is dependent upon coupling of the cytoskeleton to the nuclear periphery. This is accomplished largely through SUN and KASH domain proteins, which together assemble to form LINC (linker of the nucleoskeleton and cytoskeleton) complexes spanning the nuclear envelope. SUN proteins of the inner nuclear membrane provide a connection to nuclear structures while acting as a tether for outer nuclear membrane KASH proteins. The latter contain binding sites for diverse cytoskeletal components. Recent publications highlight new aspects of LINC complex regulation revealing that the interplay between SUN and KASH partners can strongly influence how the nucleus functionally engages with different branches of the cytoskeleton.

Mechanical Role of Nesprin-1-Mediated Nucleus-Actin Filament Binding in Cyclic Stretch-Induced Fibroblast Elongation.

The intracellular mechanical link tethering the nucleus to the cytoskeleton has been suggested to be the linker of the nucleoskeleton and cytoskeleton (LINC) complex. Previous studies have reported that knockdown of nesprin-1, a component of the LINC complex that directly binds to actin filaments, suppresses cellular morphological response to mechanical stimuli. The relation between nesprin-1 knockdown and cellular morphological changes, however, remains unclear. In this study, we examined the mechanical role of nucleus-actin filament binding in morphological changes of fibroblasts exposed to cyclic stretching. After exposure to 10% cyclic stretching for 6h, fibroblasts transfected with nesprin-1-specific small interfering RNA showed fewer elongated shapes compared with non-transfected cells. To further examine the mechanical role of the nucleus and nucleus-bound actin filaments, we applied cyclic stretching to fibroblasts treated with Trichostatin A (TSA), which decreases nuclear stiffness and thereby reduces nucleus-binding actin filament tension. TSA-treatment was found to induce more rounded cellular shapes than those of non-treated cells under both static and cyclic stretching conditions. These results suggest that the tension of nucleus-bound actin filaments plays an important role in the formation of elongated fibroblasts under cyclic stretching and that nesprin-1 knockdown causes a decrease of tension in nucleus-associated actin filaments.

Mechanosensing Defects in Nuclear Envelope Related Disorders

Recent data indicate that mutations in nuclear envelope genes cause both defects in mechanotransduction signaling and force transmission across the nuclear envelop. A central role in this mechanosensory process has been attributed to A-type lamins, which together with the LInker of the Nucleoskeleton and Cytoskeleton (LINC)-complex enables force transmission across the nuclear envelop. Whereas a basic picture is that extracellular mechanical forces are transmitted from outside the cell to the nucleus, we hypothesize that mutations in A-type lamin (LMNA) or in nesprin can affect the mechanical transmission from the nuclear envelop to the extracellular matrix. Human myoblasts with either LMNA or nesprin-1 mutations were cultured on soft substrates (12 kPa) and compared to control (WT) myoblasts. On soft surface, LMNA and nesprin-1 mutated myoblasts exhibited enlarged and increased number of focal adhesions, increased stress fibers and enlarged cell spreading area compared with WT. Further, nesprin-1 mutant exerted significantly higher traction forces on the substrate compared with WT myoblasts. These abnormalities were greatly reduced after treatment with Y27632, or SU6656, which inhibit Rho-associated protein kinase or the Src family kinase respectively, suggesting an abnormal activation of Rho-dependent Src pathway in mutant cells. Treatment with the MLCK inhibitor ML7 also significantly reduced the spreading area in mutant cells but without modifying the number and distribution of perinuclear actin stress fibers. We concluded that the integrity of the LINC complex and the lamina is required for proper regulation of the cytoplasmic actin contractility in soft substrates, through an apparent Src-dependent ROCK pathway.

Mechanical Regulation of Nuclear Structure and Function

Mechanical loading induces both nuclear distortion and alterations in gene expression in a variety of cell types. Mechanotransduction is the process by which extracellular mechanical forces can activate a number of well-studied cytoplasmic signaling cascades. Inevitably, such signals are transduced to the nucleus and induce transcription factor-mediated changes in gene expression. However, gene expression also can be regulated through alterations in nuclear architecture, providing direct control of genome function. One putative transduction mechanism for this phenomenon involves alterations in nuclear architecture that result from the mechanical perturbation of the cell. This perturbation is associated with direct mechanical strain or osmotic stress, which is transferred to the nucleus. This review describes the current state of knowledge relating the nuclear architecture and the transfer of mechanical forces to the nucleus mediated by the cytoskeleton, the nucleoskeleton, and the LINC (linker of the nucleoskeleton and cytoskeleton) complex. Moreover, remodeling of the nucleus induces alterations in nuclear stiffness, which may be associated with cell differentiation. These phenomena are discussed in relation to the potential influence of nuclear architecture-mediated mechanoregulation of transcription and cell fate.

The nuclear periphery

In recent years, definitions of the nuclear periphery have become increasingly blurred with the realization that nuclear envelope (NE) and nuclear pore complex (NPC) components interact with and influence the behavior of both nuclear and cytoplasmic structures. The Minisymposium on the Nuclear Periphery reflected this evolving outlook by bringing together an eclectic group of researchers working on a variety of problems related to the biology of the NE/NPC. A. Szymborska (Ellenberg laboratory, European Molecular Biology Laboratory, Heidelberg) described how emerging techniques in superresolution light microscopy can be used to define the disposition of NPC components with precision in the range of nanometers. This approach permitted the first visualization of the radial substructure of the NPC by light microscopy and was able to resolve distinct members of an NPC subcomplex (the Nup107-160/Y complex). This has provided new insights into the orientation of this complex within the NPC. L. Lu (Kirchhausen laboratory, Harvard Medical School, Boston) raised the controversial issue of how NPCs become incorporated into the reforming NE at the end of mitosis. Using a combination of three-dimensional live-cell imaging and electron microscope tomography, he showed that newly segregated chromatids become coated by planar membrane sheets derived from the peripheral endoplasmic reticulum prior to NPC assembly. This suggests that mitotic and interphase NPC assembly may follow similar pathways (Lu et al., 2011 ). D. Osorio (Gomes laboratory, Universite Pierre et Marie Curie, Paris) described a small interfering RNA (siRNA)-based screen that revealed a role for Ndc1, an NPC membrane protein, in the positioning of the nucleus relative to the centrosome during fibroblast migration. His data indicate that the interaction of Ndc1 with Sun2, a constituent of LINC (LInker of the Nucleoskeleton and Cytoskeleton) complexes might regulate nuclear positioning and cell migration. D. Hu (Vallee laboratory, Columbia University, New York) focused on the cell cycle–dependent nuclear oscillation in neural progenitor cells. This process, termed interkinetic nuclear migration (INM), involves dynein-driven migration of the nucleus toward the apical surface of the neocortex in G2 (Tsai et al., 2010 ). Interestingly, in vivo siRNA-mediated depletion of factors previously implicated in dynein recruitment to the NE during G2 in nonneuronal cells, namely BicD2 (Splinter et al., 2010 ) and Nup133 (Bolhy et al., 2011 ), successively impaired INM in G2. These findings strongly support a role for dynein recruitment to the NE in apical nuclear migration during INM and provide insight into the mechanism of cell cycle control. H. Horn (Stewart laboratory, Institute of Medical Biology, Singapore) reported on the role of nesprin-4 (Nesp4), an outer nuclear membrane protein and LINC complex component. Nesp4 functions as an adaptor for kinesin-1 and is expressed in certain epithelial cells, including hair cells of the inner ear (Roux et al., 2009 ). Derivation of Nesp4-deficient mice reveals that this protein is required for the basal positioning of nuclei in outer hair cells. In the absence of Nesp4, outer hair cells are lost through apoptosis, resulting in progressive hearing loss. Y. Kim (Zheng laboratory, Carnegie Institute for Science, Baltimore) unexpectedly reported that lamin B1− and B2− double-knockout embryonic stem cells (ESCs), despite expressing no other type of lamin (A/C or B3), are indistinguishable from wild-type cells and are fully capable of differentiating toward the trophectoderm lineage. Furthermore, gene-expression analyses revealed that B-type lamins are not required for the silencing of their bound genes that are required for lineage specification in differentiating ESCs. Remarkably, lamin B double-knockout mice develop to term, albeit with defects in multiple organs (Kim et al., 2011 ). Clearly, this study is going to change our view of nuclear lamina function in both health and disease. Taken together, the studies described in this minisymposium impact a number of areas of cell and developmental biology. There is little doubt that they will provide the foundation for future work in this rapidly evolving field. We look forward to seeing the fruits of this work at future ASCB meetings.

Dysfunctional Connections Between the Nucleus and the Actin and Microtubule Networks in Laminopathic Models

Laminopathies encompass a wide array of human diseases associated to scattered mutations along LMNA, a single gene encoding A-type lamins. How such genetic alterations translate to cellular defects and generate such diverse disease phenotypes remains enigmatic. Recent work has identified nuclear envelope proteins—emerin and the linker of the nucleoskeleton and cytoskeleton (LINC) complex—which connect the nuclear lamina to the cytoskeleton. Here we quantitatively examine the composition of the nuclear envelope, as well as the architecture and functions of the cytoskeleton in cells derived from two laminopathic mouse models, including Hutchinson-Gilford progeria syndrome ( Lmna L530P/L530P) and Emery-Dreifuss muscular dystrophy ( Lmna −/−). Cells derived from the overtly aphenotypical model of X-linked Emery-Dreifuss muscular dystrophy ( Emd −/y) were also included. We find that the centrosome is detached from the nucleus, preventing centrosome polarization in cells under flow—defects that are mediated by the loss of emerin from the nuclear envelope. Moreover, while basal actin and focal adhesion structure are mildly affected, RhoA activation, cell-substratum adhesion, and cytoplasmic elasticity are greatly lowered, exclusively in laminopathic models in which the LINC complex is disrupted. These results indicate a new function for emerin in cell polarization and suggest that laminopathies are not directly associated with cells’ inability to polarize, but rather with cytoplasmic softening and weakened adhesion mediated by the disruption of the LINC complex across the nuclear envelope.
