Acute administration of methylphenidate differentially affects cortical processing of emotional facial expressions in attention-deficit hyperactivity disorder children as studied by functional near-infrared spectroscopy.

Significance: It has been reported that children with attention-deficit hyperactivity disorder (ADHD) have impairment in the recognition of angry but not of happy facial expressions, and they show atypical cortical activation patterns in response to facial expressions. However, little is known about neural mechanisms underlying the impaired recognition of facial expressions in school-aged children with ADHD and the effects of acute medication on their processing of facial expressions.Aim: We aimed to investigate the possibility that acute administration of methylphenidate (MPH) affects processing of facial expressions in ADHD children.Approach: We measured the hemodynamic changes in the bilateral temporo-occipital areas of ADHD children observing the happy and angry facial expressions before and 1.5 h after MPH or placebo administration in a randomized, double-blind, placebo-controlled, crossover design study.Results: We found that, regardless of medication, happy expressions induced increased oxyhemoglobin (oxy-Hb) responses in the right inferior occipital region but not in the superior temporal region. For angry expressions, oxy-Hb responses increased after MPH administration, but not after placebo administration, in the left inferior occipital area, whereas there was no significant activation before MPH administration.Conclusions: Our results suggest that (1) ADHD children consistently recruit the right inferior occipital regions to process happy expressions and (2) MPH administration to ADHD children enhances cortical activation in the left inferior occipital regions when they process angry expressions.

Poor Sleep Quality Associates With Decreased Functional and Structural Brain Connectivity in Normative Aging: A MRI Multimodal Approach.

Sleep is a ubiquitous phenomenon, essential to the organism homeostasis. Notwithstanding, there has been an increasing concern with its disruption, not only within the context of pathological conditions, such as neurologic and psychiatric diseases, but also in health. In fact, sleep complaints are becoming particularly common, especially in middle-aged and older adults, which may suggest an underlying susceptibility to sleep quality loss and/or its consequences. Thus, a whole-brain modeling approach to study the shifts in the system can cast broader light on sleep quality mechanisms and its associated morbidities. Following this line, we sought to determine the association between the standard self-reported measure of sleep quality, the Pittsburgh Sleep Quality Index (PSQI) and brain correlates in a normative aging cohort. To this purpose, 86 participants (age range 52–87 years) provided information regarding sociodemographic parameters, subjective sleep quality and associated psychological variables. A multimodal magnetic resonance imaging (MRI) approach was used, with whole-brain functional and structural connectomes being derived from resting-state functional connectivity (FC) and probabilistic white matter tractography (structural connectivity, SC). Brain regional volumes and white matter properties associations were also explored. Results show that poor sleep quality was associated with a decrease in FC and SC of distinct networks, overlapping in right superior temporal pole, left middle temporal and left inferior occipital regions. Age displayed important associations with volumetric changes in the cerebellum cortex and white matter, thalamus, hippocampus, right putamen, left supramarginal and left lingual regions. Overall, results suggest that not only the PSQI global score may act as a proxy of changes in FC/SC in middle-aged and older individuals, but also that the age-related regional volumetric changes may be associated to an adjustment of brain connectivity. These findings may also represent a step further in the comprehension of the role of sleep disturbance in disease, since the networks found share regions that have been shown to be affected in pathologies, such as depression and Alzheimer's disease.

Word or Word-like? Dissociating Orthographic Typicality from Lexicality in the Left Occipito-temporal Cortex

Prior lesion and functional imaging studies have highlighted the importance of the left ventral occipito-temporal (LvOT) cortex for visual word recognition. Within this area, there is a posterior-anterior hierarchy of subregions that are specialized for different stages of orthographic processing. The aim of the present fMRI study was to dissociate the effects of subword orthographic typicality (e.g., cider [high] vs. cynic [low]) from the effect of lexicality (e.g., pollen [word] vs. pillen [pseudoword]). We therefore orthogonally manipulated the orthographic typicality of written words and pseudowords (nonwords and pseudohomophones) in a visual lexical decision task. Consistent with previous studies, we identified greater activation for pseudowords than words (i.e., an effect of lexicality) in posterior LvOT cortex. In addition, we revealed higher activation for atypical than typical strings, irrespective of lexicality, in a left inferior occipital region that is posterior to LvOT cortex. When lexical decisions were made more difficult in the context of pseudohomophone foils, left anterior temporal activation also increased for atypical relative to typical strings. The latter finding agrees with the behavior of patients with progressive anterior temporal lobe degeneration, who have particular difficulty recognizing words with atypical orthography. The most novel outcome of this study is that, within a distributed network of regions supporting orthographic processing, we have identified a left inferior occipital region that is particularly sensitive to the typicality of subword orthographic patterns.

Face repetition effects in implicit and explicit memory tests as measured by fMRI.

Recent parallels between neurophysiological and neuroimaging findings suggest that repeated stimulus processing produces decreased responses in brain regions associated with that processing--a 'repetition suppression' effect. In the present study, volunteers performed two tasks on repeated presentation of famous and unfamiliar faces during functional magnetic resonance imaging (fMRI). In the implicit task, they made fame-judgements (regardless of repetition); in the explicit task, they made episodic recognition judgements (regardless of familiarity). Only in the implicit task was repetition suppression observed: for famous faces in a right lateral fusiform region, and for both famous and unfamiliar faces in a left inferior occipital region. Repetition suppression is therefore not an automatic consequence of repeated perceptual processing of stimuli.

Regional cerebral blood flow patterns related to verification of low- and high-imagery sentences

Although several SPECT studies have shown left inferior occipital activation with visual imagery, the reliability of this finding remains doubtful. The present study replicated two conditions of a previous SPECT-study exploring rCBF in subjects verifying the correctness of low- and high-imagery sentences. As in the previous study, verification of high-imagery sentences was associated with higher flow rates in the left inferior occipital region and lower flow rates in the right anterior frontal region. By contrast, a bilateral increase of thalamic flow rates with imagery was not replicated, and there was a higher flow rate in the left inferior temporal region with low-imagery sentences which had not been found in the original study.

Contributions of occipital and temporal brain regions to visual and acoustic imagery—A spect study

Regional cerebral blood flow (rCBF) was assessed by means of HMPAO-SPECT in two experimental groups. In a control condition both groups listened to abstract words, in the experimental condition they heard five names of objects. One group was advised to form visual images of the objects, the other group was advised to form acoustic images of the sounds made by these objects. Post-experimental questionnaires revealed that most of the subjects in the acoustic imagery condition had had visual images in addition to the acoustic ones. Both imagery conditions lead to approximately equal increases of rCBF in the left inferior occipital region and in the left thalamus. Flow increases in both hippocampal regions and the right inferior and superior temporal regions were larger in the acoustic than in the visual imagery condition. It is concluded that only the activation of left inferior occipital and left thalamic regions can be interpreted as being related to modality-specific visual aspects of imagery.

Cerebral correlates of imagining colours, faces and a map—I. SPECT of regional cerebral blood flow

The distribution of regional cerebral blood flow (rCBF) was assessed by single photon emission computerized tomography (SPECT) in subjects during a resting state and during imagining either colours or faces or a route on a map. Twelve out of 30 subjects reported the spontaneous occurrence of mental visual images during the resting state. In these subjects flow in both orbitofrontal regions was higher than in those subjects who had not experienced spontaneous imagery. Voluntary imagery led to an increase of regional flow indices in basal temporal regions of both hemispheres and to a rightwards shift of global hemispheric asymmetry. The local changes were distinctly more marked with faces than with any of the other two stimuli. Imagining faces was also the only condition that led to an increase of activity in the left inferior occipital region which has been suggested by previous studies as being a crucial area for visual imagery. it is concluded that the observed differences of rCBF patterns between imagery conditions are related to the amount of information conveyed by the mental image. In contrast to the results of a companion study on DC- shifts accompanying imagery there was no effect of the visual versus spatial character of the images.

Regional cerebral blood flow patterns in visual imagery

Regional cerebral blood flow patterns were investigated by means of single photon emission computerized tomography in subjects who solved cognitive tasks which either did or did not require the use of visual imagery. In a first experiment judging the correctness of visual imagery sentences like “a grapefruit is bigger than an orange” led to increases of regional activity in inferior temporal and in the left inferior-occipital region when compared to blood flow patterns elicited by judgements about low imagery sentences or by responding differentially to “yes” and “no”. Motor imagery sentences did not cause such an increase. In a second experiment a condition in which visual images were used for counting the corners of letters was compared to a condition in which subjects internally rehearsed the alphabet. The only difference concerned the inferior frontal regions which showed higher activity in rehearsing the alphabet. However, activity in inferior temporal and inferior occipital regions showed a positive correlation to the self-rated vividness of the visual images in the corner counting condition. The results of both experiments yield converging evidence that visual imagery is selectively related to activity of inferior-temporal and occipital regions. They thus support the hypothesis that the cerebral correlate of visual imagery is different from that of non-imaginal thinking.
