Semantic representations in inferior frontal and lateral temporal cortex during picture naming, reading, and repetition.

Reading, naming, and repetition are classical neuropsychological tasks widely used in the clinic and psycholinguistic research. While reading and repetition can be accomplished by following a direct or an indirect route, pictures can be named only by means of semantic mediation. By means of fMRI multivariate pattern analysis, we evaluated whether this well-established fundamental difference at the cognitive level is associated at the brain level with a difference in the degree to which semantic representations are activated during these tasks. Semantic similarity between words was estimated based on a word association model. Twenty subjects participated in an event-related fMRI study where the three tasks were presented in pseudo-random order. Linear discriminant analysis of fMRI patterns identified a set of regions that allow to discriminate between words at a high level of word-specificity across tasks. Representational similarity analysis was used to determine whether semantic similarity was represented in these regions and whether this depended on the task performed. The similarity between neural patterns of the left Brodmann area 45 (BA45) and of the superior portion of the left supramarginal gyrus correlated with the similarity in meaning between entities during picture naming. In both regions, no significant effects were seen for repetition or reading. The semantic similarity effect during picture naming was significantly larger than the similarity effect during the two other tasks. In contrast, several regions including left anterior superior temporal gyrus and left ventral BA44/frontal operculum, among others, coded for semantic similarity in a task-independent manner. These findings provide new evidence for the dynamic, task-dependent nature of semantic representations in the left BA45 and a more task-independent nature of the representational activation in the lateral temporal cortex and ventral BA44/frontal operculum.

Evidence for a Spoken Word Lexicon in the Auditory Ventral Stream.

The existence of a neural representation for whole words (i.e., a lexicon) is a common feature of many models of speech processing. Prior studies have provided evidence for a visual lexicon containing representations of whole written words in an area of the ventral visual stream known as the visual word form area. Similar experimental support for an auditory lexicon containing representations of spoken words has yet to be shown. Using functional magnetic resonance imaging rapid adaptation techniques, we provide evidence for an auditory lexicon in the auditory word form area in the human left anterior superior temporal gyrus that contains representations highly selective for individual spoken words. Furthermore, we show that familiarization with novel auditory words sharpens the selectivity of their representations in the auditory word form area. These findings reveal strong parallels in how the brain represents written and spoken words, showing convergent processing strategies across modalities in the visual and auditory ventral streams.

Temporal hierarchy of cortical responses reflects core-belt-parabelt organization of auditory cortex in musicians.

Human auditory cortex (AC) organization resembles the core-belt-parabelt organization in nonhuman primates. Previous studies assessed mostly spatial characteristics; however, temporal aspects were little considered so far. We employed co-registration of functional magnetic resonance imaging (fMRI) and magnetoencephalography (MEG) in musicians with and without absolute pitch (AP) to achieve spatial and temporal segregation of human auditory responses. First, individual fMRI activations induced by complex harmonic tones were consistently identified in four distinct regions-of-interest within AC, namely in medial Heschl's gyrus (HG), lateral HG, anterior superior temporal gyrus (STG), and planum temporale (PT). Second, we analyzed the temporal dynamics of individual MEG responses at the location of corresponding fMRI activations. In the AP group, the auditory evoked P2 onset occurred ~25ms earlier in the right as compared with the left PT and ~15ms earlier in the right as compared with the left anterior STG. This effect was consistent at the individual level and correlated with AP proficiency. Based on the combined application of MEG and fMRI measurements, we were able for the first time to demonstrate a characteristic temporal hierarchy ("chronotopy") of human auditory regions in relation to specific auditory abilities, reflecting the prediction for serial processing from nonhuman studies.

Developmental organization of neural dynamics supporting auditory perception

Purpose: A prominent view of language acquisition involves learning to ignore irrelevant auditory signals through functional reorganization, enabling more efficient processing of relevant information. Yet, few studies have characterized the neural spatiotemporal dynamics supporting rapid detection and subsequent disregard of irrelevant auditory information, in the developing brain. To address this unknown, the present study modeled the developmental acquisition of cost-efficient neural dynamics for auditory processing, using intracranial electrocorticographic responses measured in individuals receiving standard-of-care treatment for drug-resistant, focal epilepsy. We also provided evidence demonstrating the maturation of an anterior-to-posterior functional division within the superior-temporal gyrus (STG), which is known to exist in the adult STG. Methods: We studied 32 patients undergoing extraoperative electrocorticography (age range: eight months to 28 years) and analyzed 2,039 intracranial electrode sites outside the seizure onset zone, interictal spike-generating areas, and MRI lesions. Patients were given forward (normal) speech sounds, backward-played speech sounds, and signal-correlated noises during a task-free condition. We then quantified sound processing-related neural costs at given time windows using high-gamma amplitude at 70–110 Hz and animated the group-level high-gamma dynamics on a spatially normalized three-dimensional brain surface. Finally, we determined if age independently contributed to high-gamma dynamics across brain regions and time windows. Results: Group-level analysis of noise-related neural costs in the STG revealed developmental enhancement of early high-gamma augmentation and diminution of delayed augmentation. Analysis of speech-related high-gamma activity demonstrated an anterior-to-posterior functional parcellation in the STG. The left anterior STG showed sustained augmentation throughout stimulus presentation, whereas the left posterior STG showed transient augmentation after stimulus onset. We found a double dissociation between the locations and developmental changes in speech sound-related high-gamma dynamics. Early left anterior STG high-gamma augmentation (i.e., within 200 ms post-stimulus onset) showed developmental enhancement, whereas delayed left posterior STG high-gamma augmentation declined with development. Conclusions: Our observations support the model that, with age, the human STG refines neural dynamics to rapidly detect and subsequently disregard uninformative acoustic noises. Our study also supports the notion that the anterior-to-posterior functional division within the left STG is gradually strengthened for efficient speech-sound perception after birth.

Weighted RSA: An Improved Framework on the Perception of Audio-visual Affective Speech in Left Insula and Superior Temporal Gyrus

Being able to accurately perceive the emotion expressed by the facial or verbal expression from others is critical to successful social interaction. However, only few studies examined the multimodal interactions on speech emotion, and there is no consistence in studies on the speech emotion perception. It remains unclear, how the speech emotion of different valence is perceived on the multimodal stimuli by our human brain. In this paper, we conducted a functional magnetic resonance imaging (fMRI) study with an event-related design, using dynamic facial expressions and emotional speech stimuli to express different emotions, in order to explore the perception mechanism of speech emotion in audio-visual modality. The representational similarity analysis (RSA), whole-brain searchlight analysis, and conjunction analysis of emotion were used to interpret the representation of speech emotion in different aspects. Significantly, a weighted RSA approach was creatively proposed to evaluate the contribution of each candidate model to the best fitted model and provided a supplement to RSA. The results of weighted RSA indicated that the fitted models were superior to all candidate models and the weights could be used to explain the representation of ROIs. The bilateral amygdala has been shown to be associated with the processing of both positive and negative emotions except neutral emotion. It is indicated that the left posterior insula and the left anterior superior temporal gyrus (STG) play important roles in the perception of multimodal speech emotion.

Neural Correlates of Audiovisual Temporal Binding Window in Individuals With Schizotypal and Autistic Traits: Evidence From Resting-State Functional Connectivity.

Temporal proximity is an important clue for multisensory integration. Previous evidence indicates that individuals with autism and schizophrenia are more likely to integrate multisensory inputs over a longer temporal binding window (TBW). However, whether such deficits in audiovisual temporal integration extend to subclinical populations with high schizotypal and autistic traits are unclear. Using audiovisual simultaneity judgment (SJ) tasks for nonspeech and speech stimuli, our results suggested that the width of the audiovisual TBW was not significantly correlated with self-reported schizotypal and autistic traits in a group of young adults. Functional magnetic resonance imaging (fMRI) resting-state activity was also acquired to explore the neural correlates underlying inter-individual variability of TBW width. Across the entire sample, stronger resting-state functional connectivity (rsFC) between the left superior temporal cortex and the left precuneus, and weaker rsFC between the left cerebellum and the right dorsal lateral prefrontal cortex were correlated with a narrower TBW for speech stimuli. Meanwhile, stronger rsFC between the left anterior superior temporal gyrus and the right inferior temporal gyrus was correlated with a wider audiovisual TBW for non-speech stimuli. The TBW-related rsFC was not affected by levels of subclinical traits. In conclusion, this study indicates that audiovisual temporal processing may not be affected by autistic and schizotypal traits and rsFC between brain regions responding to multisensory information and timing may account for the inter-individual difference in TBW width. LAY SUMMARY: Individuals with ASD and schizophrenia are more likely to perceive asynchronous auditory and visual events as occurring simultaneously even if they are well separated in time. We investigated whether similar difficulties in audiovisual temporal processing were present in subclinical populations with high autistic and schizotypal traits. We found that the ability to detect audiovisual asynchrony was not affected by different levels of autistic and schizotypal traits. We also found that connectivity of some brain regions engaging in multisensory and timing tasks might explain an individual's tendency to bind multisensory information within a wide or narrow time window. Effects of socioeconomic status in predicting reading outcomes for children: The mediation of spoken language network

The present longitudinal study investigated the effects of early childhood socioeconomic status on language-related resting-state functional connectivity and reading outcome in adolescence. Seventy-nine children participated in this study. Socioeconomic status was measured via parent questionnaire measuring parental education and family income at 1 month. At age 14, resting-state fMRI data and reading-related behavioral data of the children were collected. Resting-state functional connectivity (RSFC) analysis was performed based on four regions of interest, including the left inferior frontal gyrus (L.IFG), left anterior superior temporal gyrus (L.aSTG), left posterior superior temporal gyrus (L.pSTG) and right anterior superior temporal gyrus (R.aSTG). Significant associations were found between parental education and the language-related RSFC, including the RSFC of L.IFG-L.aSTG and the RSFC of L.aSTG-L.pSTG, while no association was found between family income and language-related RSFC. Furthermore, the parental education-associated functional connectivity patterns (i.e., L.IFG-L.aSTG and L.aSTG-L.pSTG) were found to be positively correlated with children’s reading skills (word list reading and sentence reading fluency). Finally, path analyses indicated that the intrinsic brain connectivity between L.aSTG and L.pSTG influenced the relationship between parental education and children’s reading outcomes.

Local syntactic violations evoke fast mismatch-related neural activity detected by optical neuroimaging.

It remains to be investigated whether syntax-related mismatch activity would be evoked in event-related optical signals by syntactic violations that deviate from our language knowledge and expectations. In the current study, we have employed fast optical neuroimaging with a frequency-domain oximeter to examine whether syntactic violations of English bare infinitives in the non-finite complement clause would trigger syntax-related mismatch effects. Recorded sentences of bare or full infinitive structures (without or with the 'to' infinitival marker) with syntactically correct or incorrect versions and non-syntactic lexical items (verbs) were presented to native speakers of English (n = 8) during silent movie viewing as a passive oddball task. The analysis of source strength (i.e., minimum norm current amplitudes) revealed that the syntactic category violations of bare object infinitives led to significantly more robust optical mismatch effects than the other syntactic violation and non-structural, lexical elements. This mismatch response had a peak latency of 186ms in the left anterior superior temporal gyrus. In combination with our prior MEG report (Kubota et al. in Neurosci Lett 662:195-204, 2018), the present optical neuroimaging findings show that syntactic marking (unmarked-to-marked) violations of the bare object infinitive against the rule of the mental grammar enhance the signal strength exactly in the same manner seen with MEG scanning, including the peak latency of mismatch activity and the activated area of the brain.

The role of left vs. right superior temporal gyrus in speech perception: An fMRI-guided TMS study

Debate continues regarding the necessary role of right superior temporal gyrus (STG) regions in sublexical speech perception given the bilateral STG activation often observed in fMRI studies. To evaluate the causal roles, TMS pulses were delivered to inhibit and disrupt neuronal activity at the left and right STG regions during a nonword discrimination task based on peak activations from a blocked fMRI paradigm assessing speech vs. nonspeech perception (N = 20). Relative to a control region located in the posterior occipital lobe, TMS to the left anterior STG (laSTG) led to significantly worse accuracy, whereas TMS to the left posterior STG (lpSTG) and right anterior STG (raSTG) did not. Although the disruption from TMS was significantly greater for the laSTG than for raSTG, the difference in accuracy between the laSTG and lpSTG did not reach significance. The results argue for a causal role of the laSTG but not raSTG in speech perception. Further research is needed to establish the source of the differences between the laSTG and lpSTG.

Representation of associative and affective semantic similarity of abstract words in the lateral temporal perisylvian language regions

The examination of semantic cognition has traditionally identified word concreteness as well as valence as two of the principal dimensions in the representation of conceptual knowledge. More recently, corpus-based vector space models as well as graph-theoretical analysis of large-scale task-related behavioural responses have revolutionized our insight into how the meaning of words is structured. In this fMRI study, we apply representational similarity analysis to investigate the conceptual representation of abstract words. Brain activity patterns were related to a cued-association based graph as well as to a vector-based co-occurrence model of word meaning. Twenty-six subjects (19 females and 7 males) performed an overt repetition task during fMRI. First, we performed a searchlight classification procedure to identify regions where activity is discriminable between abstract and concrete words. These regions were left inferior frontal gyrus, the upper and lower bank of the superior temporal sulcus bilaterally, posterior middle temporal gyrus and left fusiform gyrus. Representational Similarity Analysis demonstrated that for abstract words, the similarity of activity patterns in the cortex surrounding the superior temporal sulcus bilaterally and in the left anterior superior temporal gyrus reflects the similarity in word meaning. These effects were strongest for semantic similarity derived from the cued association-based graph and for affective similarity derived from either of the two models. The latter effect was mainly driven by positive valence words. This research highlights the close neurobiological link between the information structure of abstract and affective word content and the similarity in activity pattern in the lateral and anterior temporal language system.

Aging-related differences in the cortical network subserving intelligible speech

Language communication is crucial throughout the lifespan. The current study investigated how aging affects the brain network subserving intelligible speech. Using functional magnetic resonance imaging, we compared brain responses to intelligible and unintelligible speech between older and young adults. Univariate and multivariate analyses revealed reduced brain activation and lower regional pattern distinctions in response to intelligible versus unintelligible speech in the left anterior superior temporal gyrus (aSTG) and the left inferior frontal gyrus (IFG) in the older compared with young adults. Notably, the functional connectivity between the left IFG and the left angular gyrus (AG) was increased and a significantly enhanced bidirectional effective connectivity between the left aSTG and the left AG was observed in the older adults for processing speech intelligibility. Our study revealed aging-related differences in the cortical activity for intelligible speech and suggested that increased frontal-temporal-parietal functional integration may help facilitate spoken language processing in older adults.

Neural Mechanisms for Monitoring and Halting of Spoken Word Production.

During conversation, speakers monitor their own and others' output so they can alter their production adaptively, including halting it if needed. We investigated the neural mechanisms of monitoring and halting in spoken word production by employing a modified stop signal task during fMRI. Healthy participants named target pictures and withheld their naming response when presented with infrequent auditory words as stop signals. We also investigated whether the speech comprehension system monitors inner (i.e., prearticulatory) speech via the output of phonological word form encoding as proposed by the perceptual loop theory [Levelt, W. J. M. Speaking: From intention to articulation. Cambridge, MA: MIT Press, 1989] by presenting stop signals phonologically similar to the target picture name (e.g., cabbage-CAMEL). The contrast of successful halting versus naming revealed extensive BOLD signal responses in bilateral inferior frontal gyrus, preSMA, and superior temporal gyrus. Successful versus unsuccessful halting of speech was associated with increased BOLD signal bilaterally in the posterior middle temporal, frontal, and parietal lobes and decreases bilaterally in the posterior and left anterior superior temporal gyrus and right inferior frontal gyrus. These results show, for the first time, the neural mechanisms engaged during both monitoring and interrupting speech production. However, we failed to observe any differential effects of phonological similarity in either the behavioral or neural data, indicating monitoring of inner versus external speech might involve different mechanisms.

Correlates of anomia in non-semantic variants of primary progressive aphasia converge over time

To track neural correlates of naming performance with disease progression, we estimated key areas affected in nonfluent/agrammatic (nfvPPA) and logopenic (lvPPA) primary progressive aphasia variants over time and changes in naming correlates over time. Twenty-nine non-semantic PPA participants (17 nfvPPA and 12 lvPPA) were selected based upon current diagnostic criteria and PiB-PET status and conducted a confrontation-naming task and a structural MRI. Linear mixed-effect models implemented in FreeSurfer were used for tracking cortical thickness and epicenters of atrophy over time. Using averaged cortical thickness of epicenters and naming performance as variables of interest, two sets of multivariate analyses were conducted to compare atrophy progression and naming correlates across groups. While all PPA participants demonstrated naming deterioration and progressive cortical thinning in the left temporal lobe and the left inferior frontal gyrus, the lvPPA cohort showed greater naming deterioration and thinning in the left posterior inferior parietal cortex over time than it did the nfvPPA cohort. The multivariate analyses confirmed a widespread cortical thinning in lvPPA over time, but a more rapid thinning in the right superior frontal gyrus of nfvPPA participants. Impaired naming correlated with common cortical regions in both groups. These regions included the left anterior superior temporal gyrus and the posterior middle temporal gyrus, which was primarily affected in lvPPA. Non-semantic PPA variants initially present with separate epicenters of atrophy and different spatial-temporal patterns of neurodegeneration over time, but the common involvement in key cortical regions of the left temporal lobe accounts for naming deterioration in both groups.

Neural correlates of semantic and phonological processing revealed by functional connectivity patterns in the language network

Semantics and phonology are fundamental components of language. Neuroimaging studies have identified a language network (LN) that is distributed through multiple regions and exhibits preferential responses to semantic and phonological information. However, it is unclear how these regions work collaboratively to support the processing of these components. In the present study, we first defined the LN as voxels that responded more to sentences than to strings of Chinese pseudo-characters. We subsequently used a voxel-based global brain connectivity method based on resting-state functional connectivity (FC) to characterize the neural correlates of semantic and phonological processing. We specifically correlated the within-network connectivity (WNC) of each voxel in the LN with the participants’ scores on the semantic and phonological components extracted from a battery of reading tests via principal component analysis. We found that individuals with stronger WNC in the left posterior superior temporal gyrus (lpSTG) and anterior superior temporal gyrus (laSTG) were better at semantic and phonological processing, respectively. Furthermore, the FC of the lpSTG with the laSTG and bilateral fusiform gyrus mainly contributed to semantic processing, whereas the FC of the laSTG with the left posterior middle temporal gyrus and inferior frontal gyrus largely contributed to phonological processing. Importantly, the semantic and phonological subnetworks overlapped in the laSTG, the WNC of which correlated with the participants’ performances during semantic-phonological interactions. Our study revealed the hub and subnetwork for semantic and phonological processing, respectively, and highlighted the role of the laSTG in semantic-phonological interactions.

The Functional Neuroanatomy of Lexical Tone Perception: An Activation Likelihood Estimation Meta-Analysis.

In tonal language such as Chinese, lexical tone serves as a phonemic feature in determining word meaning. Meanwhile, it is close to prosody in terms of suprasegmental pitch variations and larynx-based articulation. The important yet mixed nature of lexical tone has evoked considerable studies, but no consensus has been reached on its functional neuroanatomy. This meta-analysis aimed at uncovering the neural network of lexical tone perception in comparison with that of phoneme and prosody in a unified framework. Independent Activation Likelihood Estimation meta-analyses were conducted for different linguistic elements: lexical tone by native tonal language speakers, lexical tone by non-tonal language speakers, phoneme, word-level prosody, and sentence-level prosody. Results showed that lexical tone and prosody studies demonstrated more extensive activations in the right than the left auditory cortex, whereas the opposite pattern was found for phoneme studies. Only tonal language speakers consistently recruited the left anterior superior temporal gyrus (STG) for processing lexical tone, an area implicated in phoneme processing and word-form recognition. Moreover, an anterior-lateral to posterior-medial gradient of activation as a function of element timescale was revealed in the right STG, in which the activation for lexical tone lied between that for phoneme and that for prosody. Another topological pattern was shown on the left precentral gyrus (preCG), with the activation for lexical tone overlapped with that for prosody but ventral to that for phoneme. These findings provide evidence that the neural network for lexical tone perception is hybrid with those for phoneme and prosody. That is, resembling prosody, lexical tone perception, regardless of language experience, involved right auditory cortex, with activation localized between sites engaged by phonemic and prosodic processing, suggesting a hierarchical organization of representations in the right auditory cortex. For tonal language speakers, lexical tone additionally engaged the left STG lexical mapping network, consistent with the phonemic representation. Similarly, when processing lexical tone, only tonal language speakers engaged the left preCG site implicated in prosody perception, consistent with tonal language speakers having stronger articulatory representations for lexical tone in the laryngeal sensorimotor network. A dynamic dual-stream model for lexical tone perception was proposed and discussed.

Tau PET imaging predicts cognition in atypical variants of Alzheimer's disease.

Accumulation of paired helical filament tau contributes to neurodegeneration in Alzheimer's disease (AD). 18 F-flortaucipir is a positron emission tomography (PET) radioligand sensitive to tau in AD, but its clinical utility will depend in part on its ability to predict cognitive symptoms in diverse dementia phenotypes associated with selective, regional uptake. We examined associations between 18 F-flortaucipir and cognition in 14 mildly-impaired patients (12 with cerebrospinal fluid analytes consistent with AD pathology) who had amnestic (n = 5) and non-amnestic AD syndromes, including posterior cortical atrophy (PCA, n = 5) and logopenic-variant primary progressive aphasia (lvPPA, n = 4). Amnestic AD patients had deficits in memory; lvPPA in language; and both amnestic AD and PCA patients in visuospatial function. Associations with cognition were tested using sparse regression and compared to associations in anatomical regions-of-interest (ROIs). 18 F-flortaucipir uptake was expected to show regionally-specific correlations with each domain. In multivariate analyses, uptake was elevated in neocortical areas specifically associated with amnestic and non-amnestic syndromes. Uptake in left anterior superior temporal gyrus accounted for 67% of the variance in language performance. Uptake in right lingual gyrus predicted 85% of the variance in visuospatial performance. Memory was predicted by uptake in right fusiform gyrus and cuneus as well as a cluster comprising right anterior hippocampus and amygdala; this eigenvector explained 57% of the variance in patients' scores. These results provide converging evidence for associations between 18 F-flortaucipir uptake, tau pathology, and patients' cognitive symptoms.

Auditory cortical delta-entrainment interacts with oscillatory power in multiple fronto-parietal networks

The timing of slow auditory cortical activity aligns to the rhythmic fluctuations in speech. This entrainment is considered to be a marker of the prosodic and syllabic encoding of speech, and has been shown to correlate with intelligibility. Yet, whether and how auditory cortical entrainment is influenced by the activity in other speech–relevant areas remains unknown. Using source-localized MEG data, we quantified the dependency of auditory entrainment on the state of oscillatory activity in fronto-parietal regions. We found that delta band entrainment interacted with the oscillatory activity in three distinct networks. First, entrainment in the left anterior superior temporal gyrus (STG) was modulated by beta power in orbitofrontal areas, possibly reflecting predictive top-down modulations of auditory encoding. Second, entrainment in the left Heschl's Gyrus and anterior STG was dependent on alpha power in central areas, in line with the importance of motor structures for phonological analysis. And third, entrainment in the right posterior STG modulated theta power in parietal areas, consistent with the engagement of semantic memory. These results illustrate the topographical network interactions of auditory delta entrainment and reveal distinct cross-frequency mechanisms by which entrainment can interact with different cognitive processes underlying speech perception.

Development of the Intrinsic Language Network in Preschool Children from Ages 3 to 5 Years.

Resting state studies of spontaneous fluctuations in the functional magnetic resonance imaging (fMRI) blood oxygen level dependent signal have shown great potential in mapping the intrinsic functional connectivity of the human brain underlying cognitive functions. The aim of the present study was to explore the developmental changes in functional networks of the developing human brain exemplified with the language network in typically developing preschool children. To this end, resting-sate fMRI data were obtained from native Chinese children at ages of 3 and 5 years, 15 in each age group. Resting-state functional connectivity (RSFC) was analyzed for four regions of interest; these are the left and right anterior superior temporal gyrus (aSTG), left posterior superior temporal gyrus (pSTG), and left inferior frontal gyrus (IFG). The comparison of these RSFC maps between 3- and 5-year-olds revealed that RSFC decreases in the right aSTG and increases in the left hemisphere between aSTG seed and IFG, between pSTG seed and IFG, as well as between IFG seed and posterior superior temporal sulcus. In a subsequent analysis, functional asymmetry of the language network seeding in aSTG, pSTG and IFG was further investigated. The results showed an increase of left lateralization in both RSFC of pSTG and of IFG from ages 3 to 5 years. The IFG showed a leftward lateralized trend in 3-year-olds, while pSTG demonstrated rightward asymmetry in 5-year-olds. These findings suggest clear developmental trajectories of the language network between 3- and 5-year-olds revealed as a function of age, characterized by increasing long-range connections and dynamic hemispheric lateralization with age. Our study provides new insights into the developmental changes of a well-established functional network in young children and also offers a basis for future cross-culture and cross-age studies of the resting-state language network.

Predictive Brain Mechanisms in Sound-to-Meaning Mapping during Speech Processing.

In daily communication via spoken language, one of the core processes is understanding the words being used. Effortless and efficient information exchange via speech relies not only on the identification of individual spoken words, but also on the contextual information giving rise to expected meanings. Despite the accumulating evidence for the bottom-up perception of auditory input, it is still not fully understood how the top-down modulation is achieved in the extensive frontotemporal cortical network. Here, we provide a comprehensive description of the neural substrates underlying sound-to-meaning mapping and demonstrate how the dual-stream model functions in the modulation of expectations, allowing for a better understanding of how the human brain accomplishes sound-to-meaning mapping in a predictive manner.

Atypical white-matter microstructure in congenitally deaf adults: A region of interest and tractography study using diffusion-tensor imaging

Considerable research documents the cross-modal reorganization of auditory cortices as a consequence of congenital deafness, with remapped functions that include visual and somatosensory processing of both linguistic and nonlinguistic information. Structural changes accompany this cross-modal neuroplasticity, but precisely which specific structural changes accompany congenital and early deafness and whether there are group differences in hemispheric asymmetries remain to be established. Here, we used diffusion tensor imaging (DTI) to examine microstructural white matter changes accompanying cross-modal reorganization in 23 deaf adults who were genetically, profoundly, and congenitally deaf, having learned sign language from infancy with 26 hearing controls who participated in our previous fMRI studies of cross-modal neuroplasticity. In contrast to prior literature using a whole-brain approach, we introduce a semiautomatic method for demarcating auditory regions in which regions of interest (ROIs) are defined on the normalized white matter skeleton for all participants, projected into each participants native space, and manually constrained to anatomical boundaries. White-matter ROIs were left and right Heschl's gyrus (HG), left and right anterior superior temporal gyrus (aSTG), left and right posterior superior temporal gyrus (pSTG), as well as one tractography-defined region in the splenium of the corpus callosum connecting homologous left and right superior temporal regions (pCC). Within these regions, we measured fractional anisotropy (FA), radial diffusivity (RD), axial diffusivity (AD), and white-matter volume. Congenitally deaf adults had reduced FA and volume in white matter structures underlying bilateral HG, aSTG, pSTG, and reduced FA in pCC. In HG and pCC, this reduction in FA corresponded with increased RD, but differences in aSTG and pSTG could not be localized to alterations in RD or AD. Direct statistical tests of hemispheric asymmetries in these differences indicated the most prominent effects in pSTG, where the largest differences between groups occurred in the right hemisphere. Other regions did not show significant hemispheric asymmetries in group differences. Taken together, these results indicate that atypical white matter microstructure and reduced volume underlies regions of superior temporal primary and association auditory cortex and introduce a robust method for quantifying volumetric and white matter microstructural differences that can be applied to future studies of special populations.
