Novel Nanocomposites for Luminescent Thermometry with Two Different Modalities.

In this work, we successfully integrated fluorescent nanodiamonds (FNDs) and lanthanide ion-doped upconversion nanoparticles (UCNPs) in a nanocomposite structure for simultaneous optical temperature sensing. The effective integration of FND and UCNP shells was confirmed by employing high-resolution TEM imaging, X-ray diffraction, and dual-excitation optical spectroscopy. Furthermore, the synthesized ND@UCNP nanocomposites were tested by making simultaneous optical temperature measurements, and the detected temperatures showed excellent agreement within their sensitivity limit. The simultaneous measurement of temperature using two different modalities having different sensing physics but with the same composite nanoparticles inside is expected to greatly improve the confidence of nanoscale temperature measurements. This should resolve some of the controversy surrounding nanoscale temperature measurements in biological applications.

Engineering DNA on the Surface of Upconversion Nanoparticles for Bioanalysis and Therapeutics

Surface modification of inorganic nanomaterials with biomolecules has enabled the development of composites integrated with extensive properties. Lanthanide ion-doped upconversion nanoparticles (UCNPs) are one class of inorganic nanomaterials showing optical properties that convert photons of lower energy into higher energy. Additionally, DNA oligonucleotides have exhibited powerful capabilities for organizing various nanomaterials with versatile topological configurations. Through rational design and nanotechnology, DNA-based UCNPs offer predesigned functionality and potential. To fully harness the capabilities of UCNPs integrated with DNA, various DNA-UCNP composites have been developed for diagnosis and therapeutics. In this review, beginning with the introduction of the UCNPs and the conjugation of DNA strands on the surface of UCNPs, we present an overview of the recent progress of DNA-UCNP composites while focusing on their applications for bioanalysis and therapeutics.

Application of lanthanide-doped upconversion nanoparticles for cancer treatment: a review.

With the excellent ability to transform near-infrared light to localized visible or UV light, thereby achieving deep tissue penetration, lanthanide ion-doped upconversion nanoparticles (UCNP) have emerged as one of the most striking nanoscale materials for more effective and safer cancer treatment. Up to now, UCNPs combined with photosensitive components have been widely used in the delivery of chemotherapy drugs, photodynamic therapy and photothermal therapy. Applications in these directions are reviewed in this article. We also highlight microenvironmental tumor monitoring and precise targeted therapies. Thenwe briefly summarize some new trends and the existing challenges for UCNPs. We hope this review can provide new ideas for future cancer treatment based on UCNPs.

Porous Upconversion Nanostructures as Bimodal Biomedical Imaging Contrast Agents

Lanthanide ion doped upconversion nanoparticles (UCNPs) hold great promise as multimodal contrast agents for a range of medical imaging techniques, including optical bioimaging (OBI), magnetic resonance imaging (MRI), and computed tomography (CT). However, it is challenging to obtain UCNPs with both maximal contrast enhancement effects for both OBI and MRI simultaneously owing to the dilemma in the size of UCNPs. UCNPs in large dimensions contain more photonic Ln ions and less surface defects, which is favored for high luminescent emissions, while small UCNPs with high specific surface areas allow a higher proportion of paramagnetic Ln ions to be more accessible to water molecules, which offers enhanced contrast in MRI. In this work, we report the novel design of core−porous shell UCNPs with both high luminescent emissions and magnetic relaxivities as potential dual-modal contrast agents. The core–porous shell UCNPs were fabricated via the selective etching of the inert shell of NaYF4: 30%Gd at the active core of NaYF4: 20%Yb, 1%Er. Their morphology and composition were carefully characterized using transmission electron microscopy (TEM), energy-dispersive X-ray spectroscopy, X-ray diffraction, and high resolution TEM. Their photoluminescent and magnetic resonance properties were experimentally determined and compared for the core, core–dense shell, and core–porous shell UCNPs. Core–porous shell UCNPs were found to display bright luminescence and superior MRI contrast enhancement, thus showing great potential as bimodal OBI and MRI contrast agents.

Video-rate upconversion display from optimized lanthanide ion doped upconversion nanoparticles.

Volumetric displays that create bright image points within a transparent bulk are one of the most attractive technologies in everyday life. Lanthanide ion doped upconversion nanoparticles (UCNPs) are promising luminescent nanomaterials for background free, full-colour volumetric displays of transparent bulk materials. However, video-rate display using UCNPs has been limited by their low emission intensity. Herein, we developed a video-rate upconversion display system with much enhanced brightness. The integral emission intensity of the single UCNPs was fully employed for video-rate display. It was maximized by optimizing the emitter concentration and, more importantly, by temporally synchronizing the scanning time of the excitation light to the the raised emission time of the single UCNPs. The excitation power dependent emission response and emission time decay curves were systematically characterized for the single UCNPs with various emitter concentrations from 0.5% to 6%. 1%Tm3+ doped UCNPs presented the highest integral emission intensity. By embedding this UCNPs into a polyvinyl acetate (PVA) film, we achieved a two-dimensional (2D) upconversion display with a frame rate of 29 Hz for 35 by 50 pixels. This work demonstrates that the temporal response as well as the integral emission intensity enable video-rate upconversion display.

Coherent power amplification of third-order harmonic femtosecond pulses at thin-film up-conversion nanoparticles

Third harmonic generation (THG) is a nonlinear optical process attractive in high-resolution interfacial studies, sub-wavelength light manipulation, and bio-molecular detection due to its capability of converting low-energy quanta into a quantum of a higher energy. One of the limitations in utilizing THG is its low power conversion efficiency; thus, various THG enhancement methods have been researched by involving plasmonic coupling effects or utilizing electric band gap resonances at quantum dots or two-dimensional materials. Meanwhile, lanthanide ion-doped up-conversion nanoparticles (UCNPs) can be excited by a multi-photon process similar to THG, but its interaction or resonance with THG has not been studied to date. In this Communication, we demonstrate the first coherent amplification of third-order harmonic femtosecond pulses at multi-layered UCNP thin-film with an amplification factor of 7.8. This amplification is made by the resonance interaction of incident femtosecond laser field, generated third-order harmonics, and the electric band gaps of UCNPs. The power contribution of the third-order harmonic and the up-conversion luminescence (UCL) is strongly dependent on the sample geometry due to the reabsorption effect. For in-depth understanding of the emission characteristics, spectral-domain, time-domain, radio-frequency (RF) domain, and polarization-dependence analysis were addressed. This coherent amplification of third harmonic (TH) at UCNP thin-films enables us to attain higher power, shorter wavelength, and ultra-short femtosecond pulses generated from a simple thin-film structure near to the target samples, which will pave a way to an ultrafast short-wavelength laser platform for material characterization, sub-wavelength photonics, and biomolecular detection.

Surface Functionalisation of Upconversion Nanoparticles with Different Moieties for Biomedical Applications

Lanthanide ion-doped upconversion nanoparticles (UCNPs) that can convert low-energy infrared photons into high-energy visible and ultraviolet photons, are becoming highly sought-after for advanced biomedical and biophotonics applications. Their unique luminescent properties enable UCNPs to be applied for diagnosis, including biolabeling, biosensing, bioimaging, and multiple imaging modality, as well as therapeutic treatments including photothermal and photodynamic therapy, bio-reductive chemotherapy and drug delivery. For the employment of the inorganic nanomaterials into biological environments, it is critical to bridge the gap in between nanoparticles and biomolecules via surface modifications and subsequent functionalisation. This work reviews the various ways to surface modify and functionalise UCNPs so as to impart different functional molecular groups to the UCNPs surfaces for a broad range of applications in biomedical areas. We discussed commonly used base functionalities, including carboxyl, amino and thiol moieties that are typically imparted to UCNP surfaces so as to provide further functional capacity.

Upconversion nanocomposites for photo-based cancer theranostics.

Lanthanide ion doped upconversion nanoparticles (UCNPs) are known to be able to convert long-wavelength excitation light (usually 980 nm) into high-energy ultraviolet (UV) or visible emissions, and they have attracted significant attention because of their distinct photochemical properties including sharp emission bands, low autofluorescence, high tissue penetration depth, inertness to ambient interference and minimal photodamage to tissues. Until now, UCNPs have shown great potential in various realms including bioimaging, biosensing and biomedical applications. Especially in recent years, UCNP based nanocomposites have been found to be promising tools for multi-modal imaging and low-invasive photo-based therapy of tumors. In this review, we summarize the recent achievement and progress of UCNP based multifunctional nanoplatforms for bioimaging and cancer phototheranostics, including photodynamic therapy (PDT) and photothermal therapy (PTT). Furthermore, some emerging trends, future directions as well as challenges in this rapidly growing field are discussed.

Functionalized Upconversion Nanoparticles: Versatile Nanoplatforms for Translational Research

The design, application, and translation of targeted multimodality molecular imaging probes based on nanotechnology have attracted increasing attentions during the last decade and will continue to play vital roles in cancer diagnosis and personalized medicine. With the growing awareness of drawbacks of traditional organic dyes and quantum dots, biocompatible lanthanide ion doped upconversion nanoparticles have emerged as promising candidates for clinically translatable imaging probes, owing to their unique features that are suitable for future targeted multimodal imaging in living subjects. In this review, we summarized the recent advances in the field of functionalized upconversion nanoparticles (f-UCNP) for biological imaging and therapy in vivo, and discussed the future research directions, obstacles ahead, and the potential use of f-UCNP in translational research.

Approaches to the Multicolor Tuning of Lanthanide-Ion Doped Upconversion Nanoparticles

In recent years, lanthanide-ion doped nanocrystals have attracted considerable attention for its promising applications in multiplexed biological labeling. These materials can convert near-infrared (NIR) light into visible and offer low autofluorescence, high resistance to photobleaching, high penetration depth and large anti-Stokes shifts. With the development of these techniques, the ability to manipulate multicolor output has become more important for its biological and photovoltaic applications. This review mainly focuses on the recent development of various approaches for the multicolor tuning of lanthanide-ion doped upconversion nanoparticles.
