Effect of biosolids characteristics on retention and release behavior of azithromycin and ciprofloxacin

Azithromycin (AZ) and ciprofloxacin (CIP) are commonly prescribed antibiotics frequently detected in municipal biosolids and identified by the USEPA as contaminants of emerging concern. The land application of municipal biosolids is an agronomically beneficial practice but is also a potential pathway of CIP and AZ release into the environment. Understanding retention-release behavior is crucial for assessing the environmental fate of and risks from land-applied biosolids-borne target antibiotics. Here, we used batch equilibrations to assess retention and release of environmentally relevant concentrations of CIP and AZ in ten different biosolids. The biosolids included Class A and Class B materials with a range of physiochemical characteristics (e.g. pH, cation exchange capacity (CEC), organic matter content (OM), and iron (Fe) and aluminum (Al)) expected to influence retention and release of AZ and CIP. Retention was linear (R2 > 0.99 for AZ and >0.96 for CIP) and sorption coefficients (Kd) ranged from 52 to 370 L kg−1 for AZ and 430–2300 L kg−1 for CIP. Desorption also varied but was highly hysteretic, with hysteresis coefficients (H) ranging 0.01 to 0.15 for AZ and ≤0.01 for CIP, suggesting limited bioaccessibility. The penalized and shrinkage method least absolute shrinkage and selection operator (LASSO) was used to produce models describing AZ and CIP sorption behavior based on any given biosolids physiochemical characteristics. Multiple linear regression analysis linked AZ sorption behavior to total Fe content, likely due to a predisposition of AZ to participate in reactions with in situ Fe species. CIP sorption behavior was linked to oxalate extractable Al and total phosphorus (P) content, suggesting CIP bonding with amorphous forms of Al and a potential relationship between CIP sorption to biosolids and biosolids production processes, as manifested by correlation of CIP sorption with total P content.

Long term carbon sequestration potential of biosolids-amended copper and molybdenum mine tailings following mine site reclamation

Management and reclamation of industrial mine sites for carbon (C) sequestration is an emerging technique for offsetting anthropogenic C emissions. Land application of municipal biosolids is an effective method for amending closed tailings storage facilities and providing the nutrients to establish a vegetative cover. Biosolids applications can influence the C sequestration potential of tailings and other mine wastes at the onset of reclamation by initiating soil development processes and enhancing primary productivity, thereby leading to increased accumulation of soil organic carbon (SOC) over time. The short term ecological benefits of biosolids applications are well understood, but the long-term (>10 years) effects of biosolids on reclaimed mine soils are under-researched. The objective of this long-term study was to determine the effects of biosolids applied in 1998 at increasing rates (0, 150 and 250 dry Mg ha−1) on the C sequestration potential of a copper and molybdenum mine tailings site in the southern interior of British Columbia, Canada that is currently undergoing reclamation to a pasture-based ecosystem. We assessed changes in C pools, plant productivity and select soil physiochemical parameters at an established research site at the Bethlehem Tailings Storage Facility over a 13-year period spanning from 1998 to 2011. Tailings total C and N concentrations increased with time and were highest when biosolids were applied at 250 Mg ha−1. Carbon pools increased with increasing biosolids application and ranged from 23 to 155 Mg C ha−1 after 13 years of reclamation. The net SOC sequestration rates (i.e. the C sequestration potential) ranged from 0.72 to 6.3 Mg C ha −1 yr −1 and were highest at the 250 Mg ha−1 application rate. The C storage efficiency was higher in the 150 Mg ha−1 treatment (0.74 Mg C stored per Mg of biosolids applied), indicating that lower application rates of biosolids are more efficient at storing C than higher application rates. Aboveground plant biomass was substantially higher on biosolid-amended tailings (6 and 6.7 Mg ha−1 for B150 and B250, respectively) compared to the unamended tailings (0.39 Mg ha−1), which suggests that the increase in C pools was a direct result of organic matter inputs from enhanced plant productivity. The tailings were naturally high in Cu and Mo, and when amended with biosolids at a rate of 250 Mg ha−1, elevated levels of Zn (as compared to federal soil quality guidelines) were detected. The unamended tailings increased in alkalinity with time, whereas the pH of the biosolid-amended tailings remained stable around neutral. This study demonstrated that a single application of biosolids can facilitate plant production and the accumulation of SOC on mine tailings for more than a decade, and supports the use of biosolids for promoting long-term C sequestration on reclaimed mine sites in similar environments.

Monitoring Bacteroides spp. markers, nutrients, metals and Escherichia coli in soil and leachate after land application of three types of municipal biosolids

A lysimeter-based field study was done to monitor the transfer of culturable Escherichia coli, general (ALLBAC), human (Hf183) and swine (PIG-BAC-1) specific 16S rRNA Bacteroides spp. markers, nutrients and metals through soils and leachate over time following land application of a CP1/Class A as well as two CP2/Class B municipal biosolids (MBs). Hf183 markers were detected up to six days following application in soils receiving dewatered and liquid MBs, but not in leachate, suggesting their use in source tracking is better suited for recent pollution events. The CP2/Class B biosolids and swine manure contributed the highest microbial load with E. coli loads (between 2.5 and 3.7 log CFU (100 mL)−1) being greater than North American concentration recommendations for safe recreational water. ALLBAC persisted in soils and leachate receiving all treatments and was detected prior to amendment application demonstrating its unsuitability for identifying the presence of fecal pollution. A significant increase in NO3–N (for Lystek and dewatered MBs) and total-P (for dewatered and liquid MBs) in leachate was observed in plots receiving the CP1/Class A and CP2/Class B type MBs which exceeded North American guidelines, suggesting impact to surface water. Metal (As, Cd, Cr, Co, Cu, Pb, Mo, Ni, Se, Zn and Hg) transfer was negligible in soil and leachate samples receiving all treatments. This study is one of the first to examine the fate of E. coli and Bacteroides spp. markers in situ following the land application of MBs where surface runoff does not apply.

Occurrence and Fate of Perfluorochemicals in Soil Following the Land Application of Municipal Biosolids

The recent implementation of soil and drinking water screening guidance values for two perfluorochemicals (PFCs), perfluorooctanoate (PFOA) and perfluorooctane sulfonate (PFOS) by the U.S. Environmental Protection Agency (EPA), reflects the growing concerns regarding the presence of these persistent and bioaccumulative chemicals in the natural environment. Previous work has established the potential risk to the environment from the land application of industrially contaminated biosolids, but studies focusing on environmental risk from land application of typical municipal biosolids are lacking. Thus, the present study investigated the occurrence and fate of PFCs from land-applied municipal biosolids by evaluating the levels, mass balance, desorption, and transport of PFCs in soils receiving application of municipal biosolids at various loading rates. This study is the first to report levels of PFCs in agricultural soils amended with typical municipal biosolids. PFOS was the dominant PFC in both biosolids (80-219 ng/g) and biosolids-amended soil (2-483 ng/g). Concentrations of all PFCs in soil increased linearly with increasing biosolids loading rate. These data were used to develop a model for predicting PFC soil concentrations in soils amended with typical municipal biosolids using cumulative biosolids loading rates. Mass balance calculations comparing PFCs applied vs those recovered in the surface soil interval indicated the potential transformation of PFC precursors. Laboratory desorption experiments indicated that the leaching potential of PFCs decreases with increasing chain length and that previously derived organic-carbon normalized partition coefficients may not be accurate predictors of the desorption of long-chain PFCs from biosolids-amended soils. Trace levels of PFCs were also detected in soil cores from biosolids-amended soils to depths of 120 cm, suggesting potential movement of these compounds within the soil profile over time and confirming the higher transport potential for short-chain PFCs in soils amended with municipal biosolids.

Persistence of triclocarban and triclosan in soils after land application of biosolids and bioaccumulation in Eisenia foetida

The presence of the antimicrobial chemicals triclocarban (TCC) and triclosan (TCS) in municipal biosolids has raised concerns about the potential impacts of these chemicals on soil ecosystems following land application of municipal biosolids. The relative persistence of TCC and TCS in agricultural fields receiving yearly applications of biosolids at six different loading rates over a three-year period was investigated. Soil and biosolids samples were collected, extracted, and analyzed for TCC and TCS using liquid chromatography-tandem mass spectrometry. In addition, the potential for bioaccumulation of TCC and TCS from the biosolids-amended soils was assessed over 28 d in the earthworm Eisenia foetida. Standard 28-d bioaccumulation tests were conducted for three biosolids loading rates from two sites, representing agronomic and twice the agronomic rates of biosolids application plots as well as control plots receiving no applications of biosolids. Additional bioaccumulation kinetic data were collected for the soils receiving the high biosolids loadings to ensure attainment of quasi steady-state conditions. The results indicate that TCC is relatively more persistent in biosolids-amended soil than TCS. In addition, TCC bioaccumulated in E. foetida, reaching body burdens of 25 ± 4 and 133 ± 17 ng/g(ww) in worms exposed for 28 d to the two soils amended with biosolids at agronomic rates. The 28-d organic carbon and lipid-normalized biota soil accumulation factors (BSAFs) were calculated for TCC and ranged from 0.22 ± 0.12 to 0.71 ± 0.13. These findings suggest that TCC bioaccumulation is somewhat consistent with the traditional hydrophobic organic contaminant (HOC) partitioning paradigm. However, these data also suggest substantially reduced bioavailability of TCC in biosolids-amended soils compared with HOC partitioning theory.

Trace Organic Chemicals in Biosolieds-Amended Soils: State-of-the-Science Review

The presence of trace organic chemicals (TOrCs) in municipal biosolids in the U.S. has received considerable attention by the public and scientific community over the last several years. Of particular concern is whether the presence of TOrCs in biosolids results in significant risks to public health and the environment upon land application of the biosolids. While the U.S. Environmental Protection Agency has evaluated the risks associated with dioxins present in biosolids-amended soils, to date, no other TOrCs, particularly those of emerging concern, have been subjected to complete risk assessments with respect to the land application of municipal biosolids. This study identified the scientific data gaps that should be filled if appropriate risk assessments are to be conducted. The focus of this effort was to compile data from published peer-reviewed literature that addresses the occurrence, mobility, persistence, bioaccumulation, toxicity, and microbial impacts of biosolids-borne TOrCs in soils. In addition, an evaluation of the current modeling approaches employed to evaluate the risks of biosolids-borne TOrCs was conducted. This title belongs to WERF Research Report Series . Effects of land application of municipal biosolids on nitrogen-fixing bacteria in agricultural soil

Effects of municipal biosolids on microbial N2-fixation in agricultural soil were assessed in a 3-month laboratory study which included analysis of metals, pharmaceuticals, and personal care products. Reference agricultural soil was amended with organic manure or municipal biosolids from a southern Ontario wastewater plant, with a biosolids-only treatment included to evaluate metabolic activity in this inoculum. Microbial N2-fixation in reference and manure-amended soils were similar (p = 0.144) over 3 months and lower than in biosolids-amended soil (p = 0.001); however, differences among soil treatments decreased over time, with no significant difference at test termination. In general, one-time application of biosolids caused short-term stimulation of N2-fixing activity with a return to reference conditions within 3 months. Although no residual effects were detected, biosolids introduced elevated levels of metals, pharmaceuticals, personal care products (particularly analgesics), and viable bacteria; long-term effects caused by repeat applications (as commonly practiced) requires further investigation.

Pharmaceutical and personal care products in tile drainage following surface spreading and injection of dewatered municipal biosolids to an agricultural field

Land application of municipal biosolids can be a source of environmental contamination by pharmaceutical and personal care products (PPCPs). This study examined PPCP concentrations/temporally discrete mass loads in agricultural tile drainage systems where two applications of biosolids had previously taken place. The field plots received liquid municipal biosolids (LMB) in the fall of 2005 at an application rate of ∼ 93,500 L ha − 1 , and a second land application was conducted using dewatered municipal biosolids (DMB) applied at a rate of ∼ 8 Mt dw ha − 1 in the summer of 2006. The DMB land application treatments consisted of direct injection (DI) of the DMB beneath the soil surface at a nominal depth of ∼ 0.11 m, and surface spreading (SS) plus subsequent tillage incorporation of DMB in the topsoil (∼ 0.10 m depth). The PPCPs examined included eight pharmaceuticals (acetaminophen, fluoxetine, ibuprofen, gemfibrozil, naproxen, carbamazepine, atenolol, sulfamethoxazole), the nicotine metabolite cotinine, and two antibacterial personal care products triclosan and triclocarban. Residues of naproxen, cotinine, atenolol and triclosan originating from the fall 2005 LMB application were detected in tile water nearly nine months after application (triclocarban was not measured in 2005). There were no significant differences ( p > 0.05) in PPCP mass loads among the two DMB land application treatments (i.e., SS vs. DI); although, average PPCP mass loads late in the study season (> 100 days after application) were consistently higher for the DI treatment relative to the SS treatment. While the concentration of triclosan (∼ 14,000 ng g − 1 dw) in DMB was about twice that of triclocarban (∼ 8000 ng g − 1 dw), the average tile water concentrations for triclosan were much higher (43 ± 5 ng L − 1 ) than they were for triclocarban (0.73 ± 0.14 ng L − 1 ). Triclosan concentrations (maximum observed in 2006 ∼ 235 ng L − 1 ) in tile water resulting from land applications may warrant attention from a toxicological perspective.

Detection of the antimicrobials triclocarban and triclosan in agricultural soils following land application of municipal biosolids

The occurrence of the antimicrobials triclocarban (TCC) and triclosan (TCS) was investigated in agricultural soils following land application of biosolids using liquid chromatography–tandem mass spectrometry (LC–MS–MS) with negative ion multimode ionization. The method detection limits were 0.58 ng TCC/g soil, 3.08 ng TCC/g biosolids, 0.05 ng TCS/g soil and 0.11 ng TCS/g biosolids and the average recovery from all of the sample matrices was >95%. Antimicrobial concentrations in biosolids from three Michigan wastewater treatment plants (WWTPs) ranged from 4890 to 9280 ng/g, and from 90 to 7060 ng/g, for TCC and TCS respectively. Antimicrobial analysis of soil samples, collected over two years, from ten agricultural sites previously amended with biosolids, indicated TCC was present at higher concentrations (1.24–7.01 ng/g and 1.20–65.10 ng/g in 2007 and 2008) compared to TCS (0.16–1.02 ng/g and from the method detection limit, <0.05–0.28 ng/g in 2007 and 2008). Soil antimicrobial concentrations could not be correlated to any soil characteristic, or to the time of last biosolids application, which occurred in either 2003, 2004 or 2007. To our knowledge, our data represent the first report of TCC, and the first comparison of TCC and TCS concentrations, in biosolids-amended agricultural soils. Such information is important because approximately 50% of US biosolids are land applied, therefore, any downstream effects of either antimicrobial are likely to be widespread.

Pharmaceutical and personal care products in tile drainage following land application of municipal biosolids

Land application of municipal biosolids (sewage) is a common farming practice in many parts of the world. There is potential for transport of pharmaceuticals and personal care products (PPCPs) from agricultural fields to adjacent surface waters via tile drainage systems. In this study, liquid municipal biosolids (LMB) (total solids = 11,933 mg L − 1 ), supplemented with selected PPCPs and the fluorescent dye tracer rhodamine WT (RWT), were applied to tile drained fields using two land application approaches. Objectives included evaluating the relative benefits of land application practices with respect to reducing PPCP loadings to tile drains, evaluating PPCP persistence in tile water, and determining whether rhodamine WT can be used to estimate PPCP mass loads in tile. The PPCPs examined included an antibacterial agent used in personal care products (triclosan), a metabolite of nicotine (cotinine), and a variety of drugs including two sulfonamide antimicrobials (sulfapyridine, sulfamethoxazole), a β-blocker (atenolol), an anti-epileptic (carbamazepine), an antidepressant (fluoxetine), analgesic/anti-inflammatories (acetaminophen, naproxen, ibuprofen), and a lipid-regulator (gemfibrozil). Maximum observed PPCP concentrations in the spiked LMB were about 10 3 ng g − 1 dry weight. PPCPs were shown to move rapidly via soil macropores to tile drains within minutes of the land application. Maximum observed PPCP concentrations in tile effluent associated with the LMB application-induced tile flow event were ~ 10 1 to 10 3 ng L − 1 . PPCP mass loads, for the application-induced tile-hydrograph event, were significantly ( p < 0.1) higher for surface spreading over non-tilled soil (incorporation tillage occurring 20 h post-application), relative to aerating soil immediately prior to surface spreading using an AerWay® slurry deposition system. PPCP concentrations that were detected above the limit of quantitation (LOQ) in tile water during several precipitation-induced tile flow events that occurred post-application, included: triclosan (max. ~ 1.5 × 10 2 ng L − 1 ), carbamazepine (max. ~ 7 × 10 1 ng L − 1 ), atenolol (max ~ 4 × 10 1 ng L − 1 ), and cotinine (max ~ 2 × 10 1 ng L − 1 ). In spite of their presence in biosolids, the other PPCPs were not observed above LOQ concentrations during these events. PPCP concentrations were predicted from RWT concentrations over a 40 day study period. Tile mass loads as a percent of PPCP mass applied to soil ranged from 4.2% ± SD of 9.2% to 7.1% ± 10.9% for the AerWay® system and surface spreading plus incorporation treatments, respectively.

Bioaerosols from Land‐Applied Biosolids: Issues and Needs

Bioaerosols are a vehicle for the dissemination of human and animal pathogens. Because of land-filling costs and the ban on ocean dumping of municipal biosolids, land application of biosolids and animal manure is increasing all over the globe. There is no doubt that the creation, generation, and disposal of human and animal wastes increases the aerosolization potential of a wide variety of microbial pathogens and related pollutants. In an attempt to address public health issues associated with the land application of municipal biosolids, the U.S. National Research Council (Washington, D.C.) published a report on this issue in 2002. This paper focuses on the current information and technology gaps related to estimating the public health risks associated with bioaerosols during the land application of biosolids.

Effects of Unseeded Areas on Species Richness of Coal Mines Reclaimed with Municipal Biosolids

Abstract Land application of municipal biosolids on coal mine spoils can benefit vegetation establishment in mine reclamation. However, the application of biosolids leads to domination by early‐successional species, such as grasses, and low establishment of woody and volunteer species, thus reducing potential for forestry as a postmining land use. In this experiment, tree seedlings were planted in strips (0.6‐, 1‐, and 4‐m wide) that were not seeded with grasses, and the effects of unseeded strip width on seedling growth and species richness were assessed. Planted seedling mortality was high; therefore, the effect of unseeded strip width on seedling growth could not be determined. However, it was found that natural plant invasion and species richness were highest in the 4‐m unseeded strips. The practice of leaving 4‐m‐wide unseeded strips in mine reclamation with biosolids in the eastern United States, along with the improvement of tree seedling planting practices and planting stock, would help promote a more species‐rich plant community that could be utilized for forestry or a variety of other postmining land uses.
