Maternal co-exposure to mercury and perfluoroalkyl acid isomers and their associations with child neurodevelopment in a Canadian birth cohort

BackgroundPerfluoroalkyl acids (PFAAs) within the broader class of per- and polyfluoroalkyl substances (PFAS) are present in human serum as isomer mixtures, but epidemiological studies have yet to address isomer-specific associations with child development and behavior. ObjectivesTo examine associations between prenatal exposure to 25 PFAAs, including perfluorooctane sulfonate (PFOS) and perfluorooctanoate (PFOA) isomers, and child neurodevelopment among 490 mother–child pairs in a prospective Canadian birth cohort, the Alberta Pregnancy Outcomes and Nutrition (APrON) study. To consider the influence of a classic neurotoxicant, total mercury (THg), based on its likelihood of co-exposure with PFAAs from common dietary sources. MethodsMaternal blood samples were collected in the second trimester and child neurodevelopment was assessed at 2 years of age using the Bayley Scales of Infant and Toddler Development, 3rd Edition (Bayley-III). Linear or curvilinear multiple regression models were used to examine associations between exposures and neurodevelopment outcomes. ResultsSelect PFAAs were associated with lower Cognitive composite scores, including perfluoroheptanoate (PFHpA) (β = −0.88, 95% confidence interval (CI): −1.7, −0.06) and perfluorododecanoate (PFDoA) (β = −2.0, 95% CI: −3.9, −0.01). Non-linear relationships revealed associations of total PFOS (β = −4.4, 95% CI: −8.3, −0.43), and linear-PFOS (β = −4.0, 95% CI: −7.5, −0.57) and 1m-PFOS (β = −1.8, 95% CI: −3.3, −0.24) isomers with lower Language composite scores. Although there was no effect modification, including THg interaction terms in PFAA models revealed negative associations between perfluorononanoate (PFNA) and Motor (β = −3.3, 95% CI: −6.2, −0.33) and Social-Emotional (β = −3.0, 95% CI: −5.6, −0.40) composite scores. DiscussionThese findings reinforce previous reports of adverse effects of maternal PFAA exposure during pregnancy on child neurodevelopment. The unique hazards posed from isomers of PFOS justify isomer-specific analysis in future studies. To control for possible confounding, mercury co-exposure may be considered in studies of PFAAs.

Penetration of Perfluorooctanesulfonate Isomers and Their Alternatives from Maternal Blood to Milk and Its Associations with Chemical Properties and Milk Primary Components.

Perfluorooctanesulfonate (PFOS) and its alternatives, including chlorinated polyfluorinated ether sulfonates (Cl-PFESAs), are mainly detected per- and polyfluoroalkyl substances (PFAS) in human samples such as milk. However, the mechanism of their blood to milk transfer was not well studied. Here, 145 paired maternal serum and human milk samples were analyzed for six PFOS isomers and Cl-PFESAs to evaluate the transfer efficiency from maternal serum to human milk (TEHM/MS). Besides physicochemical properties, this study for the first time evaluated the influencing effects of the primary components in human milk (carbohydrate, lipid, and protein) on TEHM/MS of PFAS. No significant association was observed between TEHM/MS and the albumin binding affinity of the compounds (p = 0.601), but TEHM/MS was significantly negatively correlated with the logarithmic octanol-water partition coefficients (r2 = 0.853, p = 0.001), the logarithmic membrane-water partition coefficients (r2 = 0.679, p = 0.012), and the carbohydrate contents in human milk. The effect of carbohydrate was further confirmed using in vitro tests. The negative associations between TEHM/MS and hydrophobicity, membrane passive permeability, and the carbohydrate content in human milk consistently indicated that passive diffusion through the paracellular route might be the main transfer pathway for PFOS and Cl-PFESAs from blood to milk in humans.

Apparent Half-Lives of Chlorinated-Perfluorooctane Sulfonate and Perfluorooctane Sulfonate Isomers in Aviation Firefighters.

Elevated levels of perfluorooctane sulfonate (PFOS) and elevated detection frequency of chloro-substituted PFOS have been reported in Australian firefighters with historical exposure to aqueous-film forming foam (AFFF). The aim of this study is to estimate the apparent half-lives of Cl-PFOS and PFOS isomers in firefighters following the end of exposure to 3M-AFFF. Paired serum samples from 120 firefighters, collected approximately five years apart, were analyzed for 8-Cl-PFOS (8-chloroperfluoro-1-octanesulfonic acid) and PFOS isomers via targeted LC-MS/MS. Apparent half-life was estimated by assuming a first order-elimination model. Cl-PFOS was detected in 93% of all initial serum samples (<LOQ-1.09 ng/mL). The average half-life of Cl-PFOS among the firefighters was 5.0 years. Branched PFOS isomers made up 55% of the total isomer concentration at the initial sampling timepoint. Five years later, the proportion of branched PFOS isomers was greater (65%). The longest average half-life (11.5 years) was estimated for "1m-PFOS". Other isomers had average half-lives ranging from 4.0 to 7.5 years. Marked differences in half-lives between PFOS isomers suggest that the elimination rate of "total PFOS" (sum of all PFOS isomers) is non-linear. This is the first study to report the serum concentrations and apparent half-life of Cl-PFOS in humans.

Estimation of Transport Parameters of Perfluoroalkyl Acids (PFAAs) in Unsaturated Porous Media: Critical Experimental and Modeling Improvements.

Predicting the transport of perfluoroalkyl acids (PFAAs) in the vadose zone is critically important for PFAA site cleanup and risk mitigation. PFAAs exhibit several unusual and poorly understood transport behaviors, including partitioning to the air-water interface, which is currently the subject of debate. This study develops a novel use of quasi-saturated (residual air saturation) column experiments to estimate chemical partitioning parameters of both linear and branched perfluorooctane sulfonate (PFOS) in unsaturated soils. The ratio of linear-to-branched air-water interfacial partitioning constants for all six experiments was 1.62 ± 0.24, indicating significantly greater partitioning of linear PFOS isomers at the air-water interface. Standard breakthrough curve analysis and numerical inversion of HYDRUS models support the application of a Freundlich isotherm for PFOS air-water interfacial partitioning below a critical reference concentration (CRC). Data from this study and previously reported unsaturated column data on perfluorooctanoate (PFOA) were reevaluated to examine unsaturated systems for transport nonidealities. This reanalysis suggests both transport nonidealities and Freundlich isotherm behavior for PFOA below the CRC using drainage-based column methods, contrary to the assertions of the original authors. Finally, a combined Freundlich-Langmuir isotherm was proposed to describe PFAA air-water interfacial partitioning across the full range of relevant PFAA concentrations.

Per-and polyfluoroalkyl substances, epigenetic ageand DNA methylation: across-sectional study of firefighters.

Background: Per- and polyfluoroalkyl substances (PFASs) are persistent chemicals that firefighters encounter. Epigenetic modifications, including DNA methylation, could serve as PFASstoxicity biomarkers. Methods: With a sample size of197 firefighters, we quantified theserum concentrations of nine PFASs, blood leukocyte DNA methylation and epigenetic age indicators via the EPIC array. We examined theassociations between PFASswith epigenetic age, site- and region-specific DNA methylation, adjusting for confounders. Results: Perfluorohexane sulfonate, perfluorooctanoate (PFOA)and the sum of branched isomers of perfluorooctane sulfonate (Sm-PFOS) were associated with accelerated epigenetic age. Branched PFOA, linear PFOS, perfluorononanoate, perfluorodecanoateand perfluoroundecanoate were associated with differentially methylated loci and regions. Conclusion: PFASs concentrations areassociated with accelerated epigenetic age and locus-specific DNA methylation. The implications for PFASs toxicity merit further investigation.

Serum perfluoroalkyl substances and breast cancer risk in Japanese women: A case-control study

BackgroundPerfluoroalkyl substances (PFASs) may contribute to causing breast cancer; however, associations between exposure to PFASs and risk of breast cancer are controversial. ObjectivesIn the present study, we newly distinguished branched isomers of PFASs from their linear isomers and aimed to investigate the association between serum PFAS concentrations and breast cancer risk in Japanese women. MethodsWe used a case-control design to study 405 eligible matched pairs attending four hospitals in Nagano Prefecture, Japan from May 2001 to September 2005. We used in-port arylation gas-chromatography mass spectrometry with negative chemical ionization to measure serum concentrations of 20 PFAS congeners. We calculated multivariable-adjusted odds ratios (ORs) and 95% confidence intervals (CIs) of breast cancer and its hormone-receptor subtypes by quartiles or tertiles of serum PFASs. ResultsAfter multivariable adjustment for breast cancer risk factors, we found that serum concentrations of 20 PFAS congeners were significantly inversely associated with risk of breast cancer. Comparing the extreme quartiles of linear isomers of perfluorooctane sulfonate or perfluorooctanoic acid, ORs were 0.15 (95% CI: 0.07, 0.33 P for trend <0.0001) and 0.21 95% CI: 0.10, 0.44 P for trend <0.0001). Among postmenopausal women, whereas we found the linear isomer of perfluorotridecanoic acid to be inversely associated with breast cancer risk, a medium degree of exposure to the branched isomer of perfluorotridecanoic acid was associated with a marginally increased risk of breast cancer (OR [95% CI] = 1.74 [0.98, 3.09]). DiscussionIn our case-control study, we found overall no association between serum PFAS concentrations and increased risk of breast cancer. Many inverse associations between serum PFAS concentrations and breast cancer risk were found.

Sorption of Perfluorooctane sulfonate (PFOS) including its isomers on hydrargillite as a function of pH, humic substances and Na2SO4

Perfluorooctane sulfonate (PFOS) is a persistent organic pollutant (POP) and emergent contaminant that are widespread in the environment. Understanding the mechanisms controlling the distribution of PFOS and its isomers between hydrargillite and the water phase is important in order to study their redistribution and mobility in the environment. This study investigated the effects of pH, humic acid, fulvic acid and Na2SO4 on sorption of PFOS isomers to hydrargillite. A mixture of PFOS isomers was spiked into water and hydrargillite was added to the system and shaken for one day; the system was tested with different aqueous composition. Concentrations of PFOS isomers in the aqueous phase were quantified using an ultra-performance liquid chromatograph coupled to a triple quadrupole mass spectrometer. Our results showed that the distribution coefficients of PFOS isomers were found to be 0.76, 0.71, 0.93 and 0.90 at pH 6.5, for 3-/4-/5- PFOS, 6-/2-PFOS, L-PFOS and total PFOS respectively. The distribution coefficients increased at lower pH and decreased at alkaline conditions. The presence of humic substances (HS) increased the sorption slightly at the environmental pH of 6.5, although a competition effect was observed during acidic conditions. A tendency of PFOS distribution to hydrargillite in the presence of Na2SO4 was like its behavior in the presence of HS although the mechanisms behind the sorption were interpreted differently. This study revealed that L-PFOS was readily sorbed when no other chemicals were added or in 20 mg/L FA or 100 mg/L Na2SO4. We suggest that an increase in PFOS sorption in the presence of HS may be due to hydrophobic mechanisms while Na2SO4 contributed to increased sorption through ionic strength effects.

Analysis of Perfluorooctanesulfonate Isomers and Other Perfluorinated Alkyl Acids in Serum by In-Port Arylation Gas Chromatography Negative Chemical Ionization–Mass Spectrometry

Perfluorooctanesulfonate (PFOS) is a common environmental pollutant and has been detected in various environmental media. Several major isomers have been identified, and their profiles provide important signatures from the source of contamination. Gas chromatography has a significant resolving power for the isomers. However, methods for derivatizing sulfonates for a sensitive analysis are limited. In this study, in-port derivatization with diaryl iodonium salts into aryl sulfonates was used. A technical mixture of PFOS was separated on gas chromatography to afford 11 isomers. In electron-capture negative ionization, the PFOS derivatives form specific fragmented anions (sulfinate anions). The derivatization was also applied to other per- or polyfluoroalkyl acids (PFASs; 13 perfluoroalkyl carboxylic acids and 10 sulfonates). Analysis of human serum samples was achieved with as little as 50 μL of sample and indicated the presence of PFOS isomers and PFASs. The detection limits of PFOS isomers were 0.02–0.09 ng mL–¹ with sufficient recoveries (85%–98%), and those for other PFASs ranged from 0.01–1 ng mL–¹ (recoveries, 80%–100%), which are suitable for the analysis of human serum samples.

Atmospheric perfluoroalkyl acid occurrence and isomer profiles in Beijing, China

The airborne occurrence, isomer profiles, and phase distribution of perfluoroalkyl acids (PFAAs), including perfluoroalkyl carboxylates (PFCAs) and sulfonates (PFSAs), have received little scientific attention to date. Here we collected gaseous and particulate phase (PM2.5) samples in China, between June and November 2013, using alkalized annular denuders and downstream filters toavoid sampling artefacts associated with traditional air sampling. We analysed the concentrations of 18 linear PFAAs and the branched isomers of perfluorooctanoate (PFOA) and perfluorooctane sulfonate (PFOS). Concentrations of total PFAAs were dominated by PFCAs, with a range of 6.6–610 pg/m3 in the gaseous phase and 2.3–290 pg/m3 in the particulate phase. Concentrations of total PFCAs were higher in summer than winter in both phases. Branched PFOA isomers accounted for 10–22% of total PFOA in the gaseous phase and 13–24% in the particulate phase, which is close to, but slightly lower than, their abundance in the commercial PFOA mixtures manufactured using the electrochemical fluorination (ECF) process. In contract, branched PFOS isomers accounted for 26-63% of total PFOS in the gaseous phase and 39-77% in the particulate phase, which is much higher than their abundance in commercial PFOS mixtures manufactured by ECF. Most PFCAs had mean particle-associated fractions (Φ) higher than 0.5. PFHxS had a much higher mean Φ (0.65) than linear PFOS (0.31). We hypothesise that PFAAs observed in Beijing air may originate from the local water bodies through processes such as aerosol generation, although transformation of precursors also contribute.

Perfluoroalkyls/polyfluoroalkyl substances and dental caries experience in children, ages 3–11 years, National Health and Nutrition Examination Survey, 2013–2014

The objective of this research is to determine the association of seven perfluoroalkyl and polyfluoroalkyl substances versus dental caries experience in US children, ages 3-11 years. A cross-sectional study design was used in the analysis of National Health and Nutrition Examination Survey 2013-2014 serological data of perfluoroalkyl and polyfluoroalkyl substances. The seven perfluoroalkyl and polyfluoroalkyl substances were: 2-(N-methyl-perfluorooctane sulfonamide) acetic acid; perfluorodecanoic acid; perfluorononanoic acid; perfluorohexane sulfonic acid; linear isomers of perfluorooctanoate; linear perfluorooctane sulfonate; and monomethyl branched isomers of perfluorooctane sulfonate. Two summative variables were created: monomethyl branch isomers of perfluorooctane sulfonic acid with linear isomer of perfluorooctane and branch isomers of perfluorooctanoate with linear isomer perfluorooctonate. In unadjusted logistic regression, in which the comparison was between the less than 75th percentile reference group and the 75th and above percentile group, higher perfluorodecanoic acid was associated with dental caries experience [unadjusted odds ratio: 1.79 (95% CI: 1.19, 2.46; P = 0.0069); adjusted odds ratio: 1.54 (95% CI: 1.03, 2.30; P = 0.0385)]. Of the seven examined perfluoroalkyl and polyfluoroalkyl substances, only perfluorodecanoic acid had an association with dental caries experience in an unadjusted model and adjusted logistic regression model.

Longitudinal analysis reveals early-pregnancy associations between perfluoroalkyl sulfonates and thyroid hormone status in a Canadian prospective birth cohort

Serum perfluoroalkyl acids (PFAAs) have been linked to disruption of maternal thyroid hormone homeostasis, but results have varied between studies which we hypothesized was due to timing of the thyroid hormone measurements, variability in PFAA isomer patterns, or presence of other stressors. In a longitudinal study design, we investigated the time-dependency of associations between PFAA isomers and thyroid hormones during pregnancy and post-partum while considering thyroid peroxidase antibody (TPOAb) status and mercury (Hg) co-exposure. In participants of a prospective Canadian birth cohort (n = 494), free thyroxine (FT4), free triiodothyronine (FT3), thyroid stimulating hormone (TSH) and TPOAb were quantified in maternal plasma collected in each trimester and 3-months postpartum, and 25 PFAAs (15 linear and 10 branched) and Hg were quantified in samples collected during the second trimester. Perfluorohexane sulfonate (PFHxS) and total branched isomers of perfluorooctane sulfonate (PFOS) were positively associated with TSH in mixed-effect models, with strongest associations early in gestation. Throughout pregnancy and post-partum, PFHxS was inversely associated with FT4, consistent with elevated TSH, while Hg was inversely associated with FT3. In TPOAb-positive women, negative associations were found between PFUnA and FT4, and 1m-PFOS and TSH, supporting previous studies that thyroid disorder could increase susceptibility to PFAA-mediated hormone dysregulation. Hg did not confound associations but was a significant interaction term, revealing further positive associations between PFOS isomers (∑3m+4m-PFOS) and TSH. Higher perfluoroalkyl sulfonate exposures were associated with higher TSH and/or lower FT4, strongly suggestive that PFHxS and branched PFOS isomers are risk factors for subclinical maternal hypothyroidism. Isomer-specific analysis is important in future studies, as crude measures of ‘total-PFOS’ masked the associations of branched isomers. A concerning result was for PFHxS which had consistent negative associations with FT4 at all time points and a positive association with TSH in early pregnancy when fetal development is most sensitive to disruption.

Isomers of perfluoroalkyl substances and overweight status among Chinese by sex status: Isomers of C8 Health Project in China

Previous investigations on the associations of polyfluoroalkyl substances (PFASs) with overweight/obesity are mixed. Moreover, little information has been reported about the association between isomers of PFASs with body mass index (BMI), waist circumference (WC) or overweight. To address this shortcoming in the literature, we conducted a study involving 1612 Chinese adults (1204 men and 408 women), ages 22–96 years old, from Shenyang, China, to analyze serum isomers of perfluorooctanesulfonate (PFOS), perfluorooctanoate (PFOA), and other PFASs. Height, weight and WC were measured by a standardized protocol of WHO. Results indicated that increased serum concentrations of all (both branched and linear) isomers of PFASs were associated with a higher prevalence of overweight, and these associations were more pronounced in women. The adjusted odds ratios (ORs) from logistic regression analyses among women were 1.45 (95% confidence interval [CI]: 1.06, 1.99) for linear PFOS isomers, 1.33 (95% CI: 1.00, 1.77) for branched PFOS isomers, 1.39 (95% CI: 1.06, 1.81) for 3 + 4 + 5m PFOS, 1.54 (95% CI: 1.08, 2.21) for linear PFOA isomers, and 1.62 (95% CI: 1.05, 2.51) for branched PFOA isomers, respectively. Associations with increased WC were yielded a similar pattern. Linear regression models also showed positive associations between PFASs and BMI or WC. In conclusion, this study suggests that PFASs and their isomers are positively associated with overweight or increased WC, and the associations are stronger in women. Furthermore, PFOA and its isomers displayed the most robust obesogenic associations.

Renal function and isomers of perfluorooctanoate (PFOA) and perfluorooctanesulfonate (PFOS): Isomers of C8 Health Project in China

Perfluoroalkyl substances (PFASs) are widely-utilized synthetic chemicals commonly found in industrial and consumer products. Previous studies have examined associations between PFASs and renal function, yet the results are mixed. Moreover, evidence on the associations of isomers of PFASs with renal function in population from high polluted areas is scant. To help to address this data gap, we used high performance liquid chromatography-mass spectrometry to measure serum isomers of perfluorooctanoate (PFOA), perfluorooctanesulfonate (PFOS), and other PFASs from 1612 adults residing in Shenyang, China, and characterized their associations with estimated glomerular filtration rate (eGFR) and chronic kidney disease (CKD). Results showed that after adjusted for multiple confounding factors, most of the higher fluorinated PFASs, except for PFOA and PFDA, were negatively associated with eGFR and positively associated with CKD. Compared with linear PFOS (n-PFOS), branched PFOS isomers (Br-PFOS) were more strongly associated with eGFR (Br-PFOS; β = −1.22, 95%CI: 2.02, −0.42; p = 0.003 vs. n-PFOS; β = −0.16, 95%CI: 0.98, 0.65; p = 0.691) and CKD (Br-PFOS; OR = 1.27; 95% CI: 1.02, 1.58; p = 0.037 vs. n-PFOS; OR = 0.98; 95% CI: 0.80, 1.20; p = 0.834). In conclusion, branched PFOS isomers were negatively associated with renal function whereas their linear counterparts were not. Given widespread exposure to PFASs, potential nephrotoxic effects are of great public health concern, Furthermore, longitudinal research on the potential nephrotoxic effects of PFASs isomers will be necessary to more definitively assess the risk.

Protein-specific distribution patterns of perfluoroalkyl acids in egg yolk and albumen samples around a fluorochemical facility

In this study, eggs from free-range and barn chickens in farms around a fluorochemical facility were collected to assess the distribution profiles of perfluoroalkyl acids (PFAAs), including isomers of perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA), and perfluorohexane sulfonate (PFHxS), in egg yolk and albumen. The results revealed that the concentrations of PFAAs in yolks were significantly higher than those in albumen. All 17 PFAAs examined could be detected in yolks, showing decreasing concentrations with increasing distance from the fluorochemical facility. The three predominant compounds in yolks were perfluorobutanoic acid (PFBA, mean concentration 81.4 ng/g ww), PFOS (28.0 ng/g ww), and PFOA (4.83 ng/g ww), and this result is consistent with the product structure of the facility. Moreover, n-PFOA, n-PFOS, and n-PFHxS were the dominant contaminants in yolk, with mean concentrations of 4.75, 25.7, and 4.29 ng/g ww, respectively. In albumen, PFBA was still the predominant PFAA congener (mean concentration = 3.93 ng/g ww), followed by PFOA. Docking analysis indicated that the PFAAs presented higher binding abilities with the low density lipoprotein, high density lipoprotein, and vitellin proteins in yolk than that with ovalbumin albumen proteins, which might be the main factor influencing the possible difference in distributions of PFAAs in yolk and albumen.

Isomer-specific biotransformation of perfluoroalkyl sulfonamide compounds in aerobic soil

As an important reservoir of pollutants, soil may play a critical role in altering isomer ratios of perfluorooctane sulfonate (PFOS) or PFOS precursors (PrePFOS) via microbial processes, but this possibility has not yet been investigated, as well as the feasibility of using PFOS isomer ratio for source tracking in PFOS contaminated sites. In the present study, N‑ethyl perfluorooctane sulfonamide ethanol (EtFOSE) of the technical grade was incubated in soil microcosms for 105 days to examine isomer-specific transformation processes. Experimental data combined with a mathematical model suggest new biotransformation pathways leading to PFOS, including a direct pathway to produce PFOS via hydrolysis of the sulfonamide bond. A similar rate of biotransformation was observed for EtFOSE with an estimated half-life of 8.7 and 9.6 days for the branched and linear isomers, respectively, without statistical difference. Two transformation intermediates, N‑ethyl perfluorooctanoic acid (EtFOSAA) and perfluorooctane sulfonamide (FOSA), also showed preferential biotransformation of branched isomers. On the contrary, one intermediate N‑ethyl perfluorooctane sulfonamide (EtFOSA) showed the preferred transformation of the linear isomer with an estimated half-life of 80.8 and 11.2 days for the branched and linear isomers, respectively. As PFOS is likely to be generated through more than one pathway or one precursor, its final isomer ratio is collectively determined by several upstream reactions, each having specific isomer-specific transformation kinetics. Though the soil showed enrichment of branched PFOS isomers during the 4-month incubation, compared to PFOS standards, some uncertainty arises in concluding preferential generation of branched PFOS from its precursors, due to the lack of standards for branched PreFOS. The complexity of isomer-specific biotransformation only reinforced the challenge of applying the PFOS isomer ratio for source tracking in environmental microbial systems.

Probing mechanisms for bioaccumulation of perfluoroalkyl acids in carp (Cyprinus carpio): Impacts of protein binding affinities and elimination pathways

With regulations on the manufacture and usage of perfluorooctanoate (PFOA), perfluorooctane sulfonate (PFOS) and related compounds, short-chain perfluoroalkyl acids (PFAAs) are increasingly being used as alternatives. However, there are limited studies on their bioaccumulation mechanisms, especially for short-chain PFAAs. In this study, we examined the binding affinity of PFAAs with fish serum proteins and tissue distributions of perfluoroalkyl carboxylates (C7-C11 PFCAs) and perfluoroalkyl sulfonates (C4, C6, and C8 PFSAs) in carp (Cyprinus carpio), including the isomers of PFOS and perfluorohexane sulfonate (PFHxS). For both PFCAs and PFSAs, the fish serum protein binding constant (KA) and bioconcentration factor (BCF) increased with an increase in the carbon chain length. PFHxS (C6 PFSA) had a much higher KA but displayed a much lower BCF than those of C7-C11 PFCAs. It indicated that not only fish blood proteins, but also other proteins in the liver and kidney, mediated the accumulation of PFAAs in fish. The lowest concentration ratios of PFHxS in liver to blood and in kidney to blood suggested that it could not be effectively transported to liver and kidney by fatty acid binding proteins and organic anion transporters. PFOS and PFHxS displayed different elimination pathways, although their linear (n-) isomers were accumulated more in fish than the corresponding branched (br-) isomers. The n-PFOS was eliminated more via the feces but br-PFOS was eliminated more via the urine; while the opposite trend was observed for PFHxS isomers.

Characterization of Nine Isomers in Commercial Samples of Perfluoroethylcyclohexanesulfonate and of Some Minor Components Including PFOS Isomers.

Electrochemical fluorination of 4-ethylbenzenesulfonyl halides produces a mixture of compounds that has found extensive use as an erosion inhibitor in aircraft hydraulic fluids. This paper reports a study of the composition of commercial samples of this material from two industrial scale manufacturers in terms of the structures and relative concentrations of their components, the major of which is perfluoroethylcyclohexanesulfonate (PFECHS). Fractionation of one of these mixtures by column chromatography produced fractions in which all significant components were of sufficient purity to allow assignment of their structures by 19F NMR spectroscopy. Assessment of the relevant signals in the 19F NMR spectra of the commercial mixtures by integration revealed the presence of 14 constituents at levels ≥0.5% in terms of molar contributions. Ten of these involve five pairs of geometric isomers, including cis- and trans-PFECHS which accounted for between 55% and 60% of the components present. Three constituents were determined to be isomers of perfluorooctanesulfonate (PFOS), two branched and the other linear PFOS itself. The availability of samples of the various components also allowed us to identify the compounds responsible for the peaks observed when the commercial samples were analyzed by LC/MS using either C18 or perfluorophenyl stationary phases.

Perfluoroalkyl Acids (PFAAs) in Serum from 2-4-Month-Old Infants: Influence of Maternal Serum Concentration, Gestational Age, Breast-Feeding, and Contaminated Drinking Water.

Little is known about factors influencing infant perfluorinated alkyl acid (PFAA) concentrations. Associations between serum PFAA concentrations in 2-4-month-old infants ( n = 101) and determinants were investigated by multiple linear regression and general linear model analysis. In exclusively breast-fed infants, maternal serum PFAA concentrations 3 weeks after delivery explained 13% (perfluoroundecanoic acid, PFUnDA) to 73% (perfluorohexanesulfonate, PFHxS) of infant PFAA concentration variation. Median infant/maternal ratios decreased with increasing PFAA carbon chain length from 2.8 for perfluoroheptanoic acid and perfluorooctanoic acid (PFOA) to 0.53 for PFUnDA and from 1.2 to 0.69 for PFHxS and perfluorooctanesulfonate (PFOS). Infant PFOA, perfluorononanoic acid (PFNA), and PFOS levels increased 0.7-1.2% per day of gestational age. Bottle-fed infants had mean concentrations of PFAAs 2 times lower than and a mean percentage of branched (%br) PFOS isomers 1.3 times higher than those of exclusively breast-fed infants. PFOA, PFNA, and PFHxS levels increased 8-11% per week of exclusive breast-feeding. Infants living in an area receiving PFAA-contaminated drinking water had 3-fold higher mean perfluorobutanesulfonate (PFBS) and PFHxS concentrations and higher mean %br PFHxS. Prenatal PFAA exposure and postnatal PFAA exposure significantly contribute to infant PFAA serum concentrations, depending on PFAA carbon chain length. Moderately PFBS- and PFHxS-contaminated drinking water is an important indirect exposure source.

Association among total serum isomers of perfluorinated chemicals, glucose homeostasis, lipid profiles, serum protein and metabolic syndrome in adults: NHANES, 2013–2014

Perfluorinated chemicals (PFCs) have been used widely in consumer products manufacture. Recent in vitro as well as animal studies have found that there are different toxicity and pharmacokinetic profiles between isomers of perfluorooctanoic acid (PFOA) and/or perfluorooctane sulfonate (PFOS). However, the differential effects of linear or branched PFOA/PFOS isomers on human beings have never been reported. Herein, we examined 1871 adult subjects (age older than 18 years) from the National Health and Nutrition Examination Survey (NHANES) 2013–2014 to determine the association between the isomers of PFOA/PFOS and serum biochemistry profiles, including glucose, lipids, protein and components of metabolic syndrome (MS). The results showed that for PFOA, increased linear PFOA was associated with increases in total cholesterol, serum albumin and an enhancement of β cell function as well as a decrease in the serum globulin. Increased branched PFOA was significantly associated with increased fasting glucose. All isomers of PFOA were positively associated with high-density lipoprotein-cholesterol (HDL-C) and negatively associated with glycohemoglobin (HbA1C). The branched PFOS was positively associated with β cell function and inversely associated with serum globulin. Both linear and branched isomers of PFOS were positively associated with the total protein and albumin. The increased branched PFOA was associated with less HDL-C insufficiency defined by the National Cholesterol Education Program Third Adult Treatment Panel (NCEP-ATP III) MS criteria, whereas the increased concentrations of serum total and linear PFOS were associated with less hypertriglyceridemia by the NCEP-ATP III. In conclusion, serum isomers of PFOA and PFOS were associated with glucose homeostasis, serum protein as well as lipid profiles; they were also indicators of MS. This may suggest that there is a distinct difference in the toxicokinetics of the isomers of PFOA and PFOS. Further clinical and animal studies are warranted to clarify the putative causal relationships between isomers and biochemical alterations.

Isomers of perfluorooctanesulfonate (PFOS) in cord serum and birth outcomes in China: Guangzhou Birth Cohort Study

Prior investigations on the associations of polyfluoroalkyl substances (PFASs) with fetal growth are mixed. Moreover, little research has accrued pertaining to the association between isomers of PFASs with gestational age and birth weight. To address this gap and present novel information, we conducted a study including 321 pairs of mothers and their infants recruited from Guangzhou, China. High performance liquid chromatography-mass spectrometry was utilized to analyze isomers of perfluorooctanesulfonate (PFOS), perfluorooctanoate (PFOA) along with other PFAS levels in cord serum samples. Mothers' and infants' characteristics were gathered from medical records. The resulting data revealed that higher PFOS, PFOA and isomers of PFOS were associated with lower birth weight. Per ln-unit (ng/mL) increase in cord serum total branched PFOS isomers was associated with a 126.3g (95% CI: −195.9, −56.8) reduction in the weight of infants at birth, while an ln-unit (ng/mL) increase of serum linear PFOS isomers (n-PFOS) was associated with a 57.2g (95% CI: −103.1, −11.3) reduction in the weight of infants at birth upon the subsequent adjustment for potential confounding variables. Notably, the association between cord PFAS level and birth weight was more pronounced in male infants. Furthermore, a positive association among branched PFOS isomers (1m-PFOS and 3+4+5m-PFOS) and gestational age was found. No associations could be found among other PFASs in conjunction with gestational age or birth weight. In conclusion, this investigation suggests that higher PFAS concentrations are associated with lower birth weight, and branched PFOS isomers show greater impact on infant birth weight than linear PFOS.
