Zn(II)-catalysed enantioselective addition of alcohols and tert-butyl hydroperoxide to isatin-derived N-Boc ketimines.

Using binaphthyl-proline-based chiral ligands, Zn(II)-catalysed addition of alcohols and tert-butyl hydroperoxide to isatin-derived N-Boc ketimines provided the isatin-derived C3 N,O-aminals in up to 99% yield and up to 99% ee. The reactions could be carried out under mild conditions and a gram-scale reaction could be realized without the loss of the yield and enantioselectivity.

Organocatalytic Asymmetric Synthesis of Spirooxindole Embedded Oxazolidines.

The first organocatalytic asymmetric synthesis of spirooxindole embedded oxazolidines has been developed via a domino reaction involving hemiaminal formation, followed by an unprecedented aza-Michael reaction between isatin derived N-Boc ketimines and γ-hydroxy enones. A quinine derived bifunctional squaramide catalyst was found to be efficient for this reaction, and the products were obtained in good diastereoselectivity and with high enantioselectivity.

An enantioselective aza-Friedel-Crafts reaction of 5-aminoisoxazoles with isatin-derived N-Boc ketimines.

By employing a chiral phosphoric acid as a catalyst, an enantioselective aza-Friedel-Crafts reaction of 5-aminoisoxazoles with isatin-derived N-Boc ketimines was realized. The reaction provided a wide variety of novel 3-isoxazole 3-amino-oxindoles with good yields (up to 99%) and moderate to good enantioselectivities (up to 99%). The absolute configuration of one product was assigned by X-ray crystal structural analysis and a plausible reaction mechanism was proposed. In addition, a scale-up reaction was performed successfully. Finally, one product was subjected to Suzuki-Miyaura coupling with phenylboronic acid to afford the product in a moderate yield without erosion of the enantioselectivity.

Rhodium-Catalyzed Enantioselective Addition of Arylboroxines to Isatin-Derived N-Boc Ketimines Using Chiral Phosphite-Olefin Ligands: Asymmetric Synthesis of 3-Aryl-3-amino-2-oxindoles.

An efficient rhodium-catalyzed enantioselective arylation of isatin-derived N-Boc ketimines with arylboroxines has been developed using H8-BINOL-derived phosphite-olefin as a chiral ligand. This method allows access to a broad variety of valuable tetrasubstituted 3-amino-2-oxindole derivatives in good yields (up to 98%) with excellent enantioselectivities (up to 98% ee).

L-tert-Leucine derived urea-ammonium salts: Efficient bifunctional phase transfer catalysts for highly diastereo- and enantioselective aza-Henry reaction of isatin-derived N-Boc ketimines with α-aryl nitromethanes

An efficient way of aza-Henry reaction between isatin-derived N-Boc ketimines and α-aryl nitromethanes catalyzed by bifunctional phase transfer catalysts with a quaternary ammonium center derived from L-tert-Leucine has been developed. A series of 3-substituted 3-amino-oxindoles were constructed by this catalytic protocol in excellent yields (90–99%), with high enantioselectivities (83–95%) and diastereoselectivities (79:21–97:3). The asymmetric aza-Henry reaction of N-Boc amidosulfones and α-aryl nitromethanes were also investigated and gave the corresponding products in high to excellent yields (72–97%) with high enantioselectivities (up to 99%) and diastereoselectivities (up to >99:1).

Isatin N-protected ketimines with nitromethane catalyzed by chiral binol linked monomeric macrocyclic Cu(II)–salen complex

Chiral Cu-1B generated in situ was used as an efficient catalyst for the synthesis of β-nitroamines in high yield (88%) with excellent enantioselectivity (ee up to 99%) at RT in absence of co-catalyst via asymmetric aza-Henry reaction of various isatin derived N-Boc ketimines with nitromethane. This catalytic system did not work well with other nitroalkanes under the above optimized reaction conditions. To examine this catalytic behaviour, quantum chemical DFT calculations were performed with the nucleophiles (CH2NO2− and CH3CHNO2−) for the conversion of 1a to 2a using macrocyclic Cu-1B complex. The DFT calculated results have shown that the reaction with CH2NO2− is more favourable than the corresponding CH3CHNO2−. The calculated activation barriers suggest that the reaction with CH2NO2− is ∼8.0 kcal/mol energetically favoured than CH3CHNO2−. This catalytic protocol was further used to obtain chiral β-diamines (a building block for pharmaceuticals) at gram scale. In order to elucidate the reaction mechanism of asymmetric aza Henry reaction kinetic experiments were performed with different concentrations of the catalyst Cu-1B, nitromethane and 1g as the representative substrate. The reaction of isatin N-Boc ketimine was first order with respect to the concentration of the catalyst and the nitromethane but did not depend on the initial concentration of the substrate. A possible mechanism for the aza Henry reaction was proposed.

N-Sulfonyl Bisimidazoline Ligands and Their Applications in Pd(II)-Catalyzed Asymmetric Addition toward α-Tertiary Amines.

A new class of chiral N-sulfonyl bisimidazoline (Bim) ligands have been designed, prepared, and applied in Pd(II)-catalyzed asymmetric addition of arylboronic acids to isatin-derived N-Boc ketimines. The combination of Pd(OCOCF3)2 and N-tosyl Bim ligand shows high catalytic activity and excellent asymmetric induction, enabling asymmetric addition to offer α-tertiary amines with generally good to high yields and excellent enantioselectivity (up to 96% yield, 96% ee). This asymmetric Pd(II) catalysis can tolerate air conditions, providing a practical and operationally simple protocol toward the construction of an enantioenriched α-tertiary stereocenter.

Asymmetric phase-transfer catalysts bearing multiple hydrogen-bonding donors: Synthesis and application in nitro-Mannich reaction of isatin-derived N-Boc ketimines

A series of bifunctional asymmetric phase-transfer catalysts bearing multiple hydrogen-bonding donors derived from cinchona alkaloids are synthesized, and successfully applied to asymmetric nitro-Mannich of isatin-derived N-Boc ketimines. The products 3-substituted 3-amino-oxindoles were constructed in excellent yields (96–99%) and good enantioselectivities (up to 95% ee).

Recyclable chiral Cu(II) macrocyclic salen complex catalyzed enantioselective aza-Henry reaction of isatin derived N-Boc ketimines and its application for the synthesis of β-diamine

Recyclable chiral Cu(II) macrocyclic salen complex Cu(II)-7 generated in situ efficiently catalyzed asymmetric aza-Henry reaction of various isatin derived N-Boc ketimines with nitromethane as nucleophile at RT to give a high yield (88%) of β-nitro amines with excellent chiral induction (ee, up to 99%) with the added advantage of six times catalyst recyclability. This catalytic system also worked well with nitroethane and nitropentane to furnish the corresponding products in moderate yields with high enantioselectivities for major diastereomers. This protocol is also used for the synthesis of enantiomerically pure (S)-β-diamines via asymmetric aza-Henry reaction of N-Boc ketimine (1b) in two steps in good yield with high enantioselectivity.

An Organocatalytic Asymmetric Friedel-Crafts Addition/Fluorination Sequence: Construction of Oxindole-Pyrazolone Conjugates Bearing Vicinal Tetrasubstituted Stereocenters.

A highly efficient and practical one-pot sequential process, consisting of an organocatalytic enantioselective Friedel-Crafts-type addition of 4-nonsubstituted pyrazolones to isatin-derived N-Boc ketimines and a subsequent diastereoselective fluorination of the pyrazolone moiety, is developed. This reaction sequence delivers novel oxindole-pyrazolone adducts featuring vicinal tetrasubstituted stereocenters with a 0.5 mol % catalyst loading in high yield with excellent enantio- and diastereocontrol. Notably, chloro, bromo, and thioether functionalities can be readily incorporated, rendering a broad diversity of the product.

The 2′-phenyl cinchonidine thiourea-catalyzed asymmetric addition of alcohols to isatin-derived N-Boc ketimines

Optically active isatin-derived N,O-aminals were obtained through the catalytic asymmetric addition of various alcohols to isatin-derived N-Boc ketimines. Using 2′-phenyl cinchonidine thiourea as the catalyst, adducts were obtained in 96% yield and with up to 92% ee.

An organocatalytic Mannich/denitration reaction for the asymmetric synthesis of 3-ethylacetate-substitued 3-amino-2-oxindoles: formal synthesis of AG-041R.

The highly enantioselective organocatalytic addition of ethyl nitroacetate to isatin-derived N-Boc ketimines (Boc = tert-butoxycarbonyl), followed by the removal of the nitro group, is described. The scalable reaction sequence leads to the title compounds as important intermediates of pyrroloindoline alkaloids and related drugs in excellent yields and enantioselectivities. The synthesis of the hexahydrofurano[2,3-b]indole skeleton, the spirocarbamate oxindole unit, and the formal synthesis of AG-041R have been carried out to demonstrate the synthetic utility of this protocol.

The origin for highly enantioselective induction of 1-naphthol to isatin-derived N-Boc ketimines catalyzed by quinine thiourea catalyst: an experimental and computational study

Rhodium-catalyzed three-component reaction of 3-diazooxindoles with indoles and isatin-derived ketimines: a facile and versatile approach to functionalized 3,3′,3′′-trisindoles

A simple, facile and efficient Rh2(OAc)4-catalyzed three-component reaction of 3-diazooxindoles with indoles and isatin-derived N-Boc ketimines towards a variety of functionalized 3,3',3''-trisindoles in high yields with moderate to excellent diastereoselectivities has been developed. This methodology provides an ideal approach for the direct introduction of indole and oxindole into an isatin moiety at the 3-position.

Catalytic enantioselective addition of alcohols to isatin-derived N-Boc ketimines

The enantioselective addition of alcohols to isatin-derived N-Boc ketimines was investigated for the first time. Isatin-derived N,O-aminals were obtained in excellent yields with moderate-to-good enantioselectivities (up to 78% ee) in the presence of a quinine-based bifunctional catalyst.

The highly enantioselective addition of indoles and pyrroles to isatins-derived N-Boc ketimines catalyzed by chiral phosphoric acids

The first asymmetric aza-Friedel-Crafts reaction of indoles and pyrroles with isatin-derived N-Boc ketimines catalyzed by chiral phosphoric acids is reported. In general, derivatives of substituted 3-amino-2-oxindoles were obtained with excellent enantioselectivities and high yields.

New challenges in quantum chemistry: Quests for accurate calculations for large molecular systems.

As quantum chemistry plays a more and more central role in many complicated chemical problems, it has become necessary to obtain accurate results for large molecular systems. Conventional quantum chemistry methods are either too expensive to apply to large systems or too approximate for the results to be reliable, and they fail to satisfy this requirement. A variety of different approaches is being developed with the aim of achieving this goal: local correlation methods; divide-and-conquer methods; linear-scaling density functional methods based on the fast multipole and other approximations; effective potential methods; and hybrid methods. ONIOM (our N-layered integrated molecular orbital plus molecular mechanics method), developed by the authors, is a hybrid method in which a large molecular system is divided into onion-skin-like layers, and different quantum chemistry/molecular mechanics methods are used for different parts of the system; the results are combined to extrapolatively estimate the results of high-level calculation for the real system. Several applications of ONIOM will be discussed.
