Involvement of OA1, an Intracellular GPCR, and Gαi3, Its Binding Protein, in Melanosomal Biogenesis and Optic Pathway Formation

Ocular albinism type 1 (OA1) is characterized by abnormalities in retinal pigment epithelium (RPE) melanosomes and misrouting of optic axons. The OA1 gene encodes a G-protein-coupled receptor (GPCR) that coimmunoprecipitates with the G alpha i-subunit of heterotrimeric G-proteins from human melanocyte extracts. This study was undertaken to test whether one of the G alpha i proteins, G alpha i3, signals in the same pathway as OA1 to regulate melanosome biogenesis and axonal growth through the optic chiasm. Adult G alpha i3(-/-) and Oa1(-/-) mice were compared with their respective control mice (129Sv and B6/NCrl) to study the effects of the loss of G alpha i3 or Oa1 function. Light and electron microscopy were used to analyze the morphology of the retina and the size and density of RPE melanosomes, electroretinograms to study retinal function, and retrograde labeling to investigate the size of the uncrossed optic pathway. Although G alpha i3(-/-) and Oa1(-/-) photoreceptors were comparable to those of the corresponding control retinas, the density of their RPE melanosomes was significantly lower than in control RPEs. In addition, the RPE cells of G alpha i3(-/-) and Oa1(-/-) mice showed abnormal melanosomes that were far larger than the largest 129Sv and B6/NCrl melanosomes, respectively. Although G alpha i3(-/-) and Oa1(-/-) mice had normal results on electroretinography, retrograde labeling showed a significant reduction from control in the size of their ipsilateral retinofugal projections. These results indicate that G alpha i3, like Oa1, plays an important role in melanosome biogenesis. Furthermore, they suggest a common Oa1-G alpha i3 signaling pathway that ultimately affects axonal growth through the optic chiasm.

Effect of Bcl-2 overexpression on establishment of ipsilateral retinocollicular projection in mice

During perinatal development in rodents, ipsilateral retinofugal projection spreading over the superior colliculus is eventually restricted to the rostromedial region. Since this restriction is accompanied by the apoptotic death of more than half of the retinal ganglion cells (RGCs), cell death is believed to play a major role in the restriction of transient ipsilateral projection from the retina to the superior colliculus. To determine the role of RGC death in the establishment of ipsilateral retinofugal projection, we examined the projection pattern in the superior colliculus and the dorsal lateral geniculate nucleus of transgenic mice overexpressing the human bcl-2 gene, which protects against cell death in the CNS. Retrograde labeling of RGCs showed that the number of ipsilaterally projecting RGCs in adult transgenic mice was approximately twice that in adult wild-type mice, indicating that the naturally occurring death of RGCs was prevented in these mutant mice. However, anterograde labeling of ipsilateral retinofugal pathways revealed that the innervation of retinogeniculate and retinocollicular projections was as restricted in transgenic mice as in wild-type mice. From these results we suggest that restriction of ipsilateral retinofugal projection during development is due to retraction or elimination of excessive terminals rather than to naturally occurring RGC death.

The Visual System of the Florida Garfish, <i>Lepisosteus platyrhincus</i>(Ginglymodi) (Part 2 of 2)

Infusion of cobaltous-lysine into the optic nerve of juvenile Florida garfish reveals that the preoptic area, pretectum, thalamus and the mediorostral and ventrolateral poles of the optic tectum each receive bilateral, monosynaptic input. The bilateral input to the mediorostral pole of the tectum projects via the dorsal optic tract and terminates in the rostral half of the tectum. The bilateral input to the ventrolateral tectum projects via the ventral optic tract and extends the whole length of the tectum. The incidence of direct ipsilateral input to the tectum suggests binocular vision may play a functional role in the survival of this species. In order to test this hypothesis, we examined the retinal distribution of ipsilateral retinal ganglion cells that project to the mediorostral tectum and the size and location of the binocular visual field. Cell distribution was examined by retrograde labelling with rhodamine-conjugated dextran amine delivered by microinjections into the mediorostral pole of the right optic tectum. In the ipsilateral retina, ganglion cells are distributed in a narrow temporoventral area with a mean of 0.44±0.14 x 10³ cells per mm² apposing the retinal margin. In the contralateral retina, ganglion cells are also distributed within this temporoventral region with a mean peak density of 2.33 ± 0.47 x 10³ cells per mm². Three broad classes of ipsilaterally projecting retinal ganglion cells (orthotopic cells within the ganglion cell layer, displaced cells within the inner nuclear layer, and giant cells within the ganglion and inner nuclear layers) are identified, intermingled in both the ipsilateral and contralateral retinae after retrograde labelling from the mediorostral pole of the tectum. Ophthalmoscopic mapping of the monocular and binocular visual fields reveals two small frontal wedges of binocular overlap. A dorsal wedge (12° wide) extends from approximately 7° above the horizontal axis to 10° beyond the vertical axis, and a ventral wedge (20° wide) extends approximately 10° below the horizontal axis to 15° beyond the vertical axis. The dorsal binocular visual field is subtended by the temporoventral region of the retina possessing both ipsilaterally and contralaterally projecting ganglion cells which terminate in the mediorostral pole of the optic tectum. Therefore, the partial decussation of retinal ganglion cell axons at the optic chiasm brings information from corresponding regions of the binocular visual field into register in the mediorostral pole of the optic tectum. In addition, the size of the binocular visual field matches the relative proportion of the retina containing ipsilaterally projecting ganglion cells. The ontogeny and phylogeny of ipsilateral retinofugal projections in actinopterygians is discussed in relation to the putative secondary pathways mediating binocular vision.

An experimental re-evaluation of the primary visual system of the European chameleon, Chamaeleo chameleon.

The retinofugal projections of the chameleon were investigated by means of autoradiography, horseradish peroxidase and fluorescent techniques after intraocular injection of tracers. An ipsilateral contingent of visual fibers and projections is absent. The retinal fibers decussate completely in alternating fascicles at the optic chiasma and course to terminate in two hypothalamic nuclei (nucleus suprachiasmaticus and nucleus opticus periventricularis hypothalami posterior), six thalamic nuclei (nucleus ovalis, nucleus geniculatus lateralis dorsalis partes lateralis and medialis, nucleus geniculatus lateralis ventralis, lateral part of nucleus dorsolateralis anterior, and nucleus ventrobasalis), four pretectal nuclei (nucleus griseus tectalis, nucleus lentiformis mesencephali, nucleus geniculatus pretectalis and nucleus posterodorsalis), the optic tectum (stratum griseum et fibrosum superficiale) and the tegmental nucleus opticus tegmenti. Our findings are, in general, compatible with previous descriptions of the primary visual system in other species of lizards. However, they indicate three features particular to chameleons: first, the total absence of an ipsilateral retinofugal projection; second, the existence of an additional hypothalamic visual center located in the posterior mediodorsal hypothalamus; and third, the large size of nucleus opticus tegmenti. These features are discussed in terms of the taxonomic position of chameleons with respect to other lizards.

Adult bony fish possess only a small ipsilateral retinofugal projection, if any. Experimental manipulation, such as unilateral enucleation, can lead to an enhancement of this projection. We examined the patterns of, as well as the conditions for the development and maintenance of an enhanced ipsilateral retinofugal projection (EIRP) after nerve crush, after enucleation, and after various combinations of both types of surgery in juvenile and adult <i>Haplochromis burtoni </i>(Cichlidae). Retinal projections were labeled either unilaterally with horseradish perixodase, or with the lipophilic fluorescent dye DiI in aldehyde-fixed animals, or bilaterally with differently colored fluorescent dextran amines. Unilateral nerve crush always leads to the regeneration of retinofugal fibers to the contralateral tectum but spares some contralateral diencephalic nuclei. In addition, unilateral or bilateral nerve crush in many cases, and unilateral enucleation in some cases, leads to the development of an EIRP to the ipsilateral diencephalon and tectum. This EIRP persists (4 months and longer postoperatively) in only 10% of the unilaterally enucleated animals, in none of the animals subjected to unilateral nerve crush and in 79% of the animals subjected to bilateral nerve crush. All unilaterally enucleated animals in which the remaining, contralateral optic nerve was crushed develop and maintain an EIRP. These data suggest that nerve crush alone is sufficient to cause regenerating fibers to project, at least transiently, to the ipsilateral side of the brain. When the normal contralateral projection is either absent or in the process of regeneration, an EIRP can be maintained. In the latter case, alternate bands or patches of ipsi- and contralateral fibers in the tectum may result. Ipsilateral fibers follow unusual pathways by recrossing at the rostral diencephalon. Likewise, regenerating contralateral retinal fibers grow differently in this area; here, where the optic-nerve projection is reorganized into the optic tract, many regenerating fibers are deflected to the ipsilateral side of the brain. Despite atypical routes taken by some fibers, the EIRP nevertheless ends only in specific retinorecipient areas. An EIRP develops independently of the age of the animal, independently of the time lapse between enucleation and nerve lesion, and independently of persisting debris. The effects of ablation of visual cortex in neonatal rabbits on the organization of retinothalamic and retinopretectal projections

Primary visual cortex was ablated unilaterally in neonatal rabbits. Following a survival of 2–4 months, retrogade degeneration of the dorsal lateral geniculate nucleus (LGd) was assessed, and reorganization of retinofugal pathways was studied using methods of anretrograde transport of [3H]proline or of horseradish peroxidase. A complete lesion of primary visual cortex resulted in complete retrograde degeneration of the LGd with no sparing of any class of neurons. The terminations of retinofugal axons in the pretectum and thalamus were compared with those observed in normal animals. No major reorganization of ipsilateral retinofugal projections was observed in either the thalamus and pretectum ipsilateral to the ablated cortex, or in the thalamus and pretectum contralateral to the ablated cortex. However, contralateral retinofugal projections to the pretectum ipsilateral to the ablated cortex were significantly different from normal. In the thalamus, the projections to the lateral posterior nucleus were expanded in area and increased in density. In the pretectum, the projections to the rostral pretectal areas were greatly increased in area, especially in the region of the olivary pretectal nucleus and posterior pretectal nucleus. However, the density of these projections was not increased relative to normal. Consideration of these results in relation to other published data on the anatomical consequences of neonatal visual cortex lesions, both in mammals which show behavioral sparing following neonatal visual cortex lesions and in mammals which, like the rabbit, show no behavioral sparing, suggests that: (1) behavioral sparing may correlate with patterns of survival or death of neurons in the thalamus and retina; and (2) reorganization of retinofugal pathways is not necessarily associated with behavioral sparing.

Effects of neonatal intraocular colchicine on synaptogenesis and on the retention of the ipsilateral retinofugal projection within the superior colliculus

A normally transient ipsilateral retinofugal projection exists in the rat but is retained following eye removal because of the loss of competitive interaction between crossed and uncrossed fibers. To further explore this phenomenon, colchicine (10(-3)M) was injected into the right eye of newborn albino rats to partially suppress axonal transport in optic fibers, alter the developmental time course of retinofugal synaptic terminals and determine if this would in turn extend the period of survival of the ipsilateral projection. Measurements of the number of fibers in the nerve were also made to insure that colchicine was not lethal to the retinofugal projection. Projections into the superior colliculus were demonstrated by anterograde movement of HRP from the left eye. TMB histochemistry revealed dense labeling of the contralateral retino-recipient layers at 5 dpn in untreated or saline-injected controls. The ipsilateral projection was seen as a lighter band of activity across the colliculus which was most concentrated in the antero-medial quadrant. This pathway was transient and degenerated by 10 dpn, except for a few antero-medial fibers. Animals treated with colchicine demonstrated a retention of this pathway through 20 dpn. A concomitant quantitative analysis of synaptic development within the superior colliculus revealed populations of boutons with round (R) and flattened (F) vesicles, as well as multiple junctional (MJ) and serial (S) complexes, most of which were specialized R boutons. The various synaptic categories displayed specific ratios unique to the different stages of maturation. Intraocular colchicine reduced the ratio of R, MJ and S boutons to F terminals between 5-15 dpn (P less than 0.01). By 20 dpn, the proportions of MJ and S boutons remained depressed but the normal ratio of R to F boutons was restored. Areal determinations of each synaptic profile included in the counts revealed a significant reduction in the size of MJ synaptic profiles examined in colchicine-treated animals and this may have been reflected in the slight loss of tectal volume (6-9%). Removal of the left eye and assessment of degenerating boutons showed that the expanded ipsilateral projection was not sufficiently dense to produce such a restoration. It thus appears that colchicine delayed the growth of the R population, but the effect was reversible and this category of boutons continued to develop, albeit on a later time course.(ABSTRACT TRUNCATED AT 400 WORDS)

Postnatal development of ipsilateral retino-geniculate projections in normal albino rats and the effects of removal of one eye at birth.

The postnatal development of ipsilateral retinofugal projections to the lateral geniculate body in normal albino rats, and in rats unilaterally enucleated at birth has been examined. At postnatal ages ranging from 1 day to 6 months, horseradish peroxidase was injected into one eye of normal rats and into the remaining eye of neonatally enucleated animals. After approximately 20 hours, the animals were perfused, the brains sectioned and reaction product visualised using tetramethylbenzidine. Ipsilateral retinal projections to the lateral geniculate body in normal animals were extensive on postnatal day 1 and became reduced over the next few days, the adult pattern being established between days 9 and 12. In the enucleated group, the terminal fields of the ipsilateral projections to the lateral geniculate body from the remaining eye remained larger and displayed a greater density of terminal labelling than in age-matched controls. In addition, the ipsilateral terminal field in the dorsal lateral geniculate nucleus occupied a more lateral position than in control animals. These findings support previous suggestions that the abnormally large ipsilateral retino-fugal projections observed in adult rats following removal of one eye, at or close to, birth, result from a failure of the ipsilateral projection to become restricted and that terminal or preterminal sprouting of retinal axons may also make a small contribution to the formation of the exuberant ipsilateral projection.

The development and restriction of the ipsilateral retinofugal projection in the chick

Although it is generally believed that the central projections of the retina in birds are entirely crossed, using wheat germ agglutinin-conjugated horseradish peroxidase (WGA-HRP) as an anterograde tracer, we have found that in normal posthatched chicks there is a small ipsilateral retinofugal projection to the diencephalon and midbrain. Most of the ipsilateral fibers appear to be directed to the lateral anterior and dorsolateral anterior nuclei of the thalamus, to the pretectal region, and to the ectomammillary nucleus and the adjoining nucleus externus. Even in the best preparations the numbers of ipsilateral fibers are so small that it is hardly surprising that they have been overlooked in previous axonal degeneration and autoradiographic experiments. A significantly larger ipsilateral retinal projection develops during the second week of incubation. The ipsilaterally directed fibers can be first seen on the fifth day of incubation and their numbers appear to increase until about embryonic day 12. At this stage the projection involves substantially more fibers than at hatching and is also more extensive in its distribution; in fact, in its general organization (but not its size) it closely parallels the normal crossed retinofugal system, contributing fibers to essentially all the primary visual relay nuclei in the diencephalon and midbrain and to much of the optic tectum, where the densest projection is to its caudomedial aspect. During the second week of incubation there is also a small number of retinal fibers, which after crossing in the optic chiasm, recross the midline in the posterior and tectal commissures (and also in the tectal roof plate), before ending in the pretectal region of the ipsilateral side. In addition, there is a markedly aberrant projection from the retina into the contralateral optic nerve. Most of the ipsilateral retinal fibers are eliminated between the twelfth and sixteenth days of incubation, and by day 17 the ipsilateral projection is reduced to its mature form. The progressive reduction in the ipsilateral projection occurs at a time when it is known (from other studies) that there is an appreciable loss of retinal ganglion cells; but whether the reduction is due to neuronal death or to the selective elimination of ipsilateral axon collaterals remains to be determined. The existence of a significant ipsilateral retinofugal component early in development, probably accounts, in part, for the distinctive and persistent ipsilateral projection that occurs if one eye is removed during the first few days of incubation.

Transient retinofugal pathways in the developing chick.

The central retinal projections have been examined in normal chick embryos early in development to determine if a transient ipsilateral projection is present. Anterograde transport of HRP identified using tetramethyl benzidine as a chromogen and degeneration techniques were used on embryos of successive ages between 6 and 16 days of incubation to show the central distribution of the retinal axons from one eye. Besides the anticipated contralateral projection, a small projection was identified to primary visual nuclei on the side of the brain ipsilateral to the injected or lesioned eye in embryos between days 6 and 12 of incubation. A projection from the injected eye into the contralateral optic nerve was also identified in a number of embryos. By embryonic day 15 the retinal projection to the ipsilateral side of the brain and into the contralateral optic nerve had disappeared. This loss of the anomalous projections coincides with a period of substantial cell death in the retinal ganglion cell layer. It appears, therefore, that in the chick, like the rat, the ipsilateral retinofugal projection resulting from an embryonic enucleation may in part be due to retention of a normal ipsilateral projection.

Origins of crossed and uncrossed retinal projections in pigmented and albino mice.

The extent of the binocular cortical field in albino mice, as revealed by recording from single cells, was almost normal; although the input from the ipsilateral eye was weaker than normal, most cells were driven from both eyes. By backfilling retinal ganglion cells from one optic tract with horseradish peroxidase we examined the origins of the retinofugal projections. Filled cells ipsilateral to the injected tract were concentrated in a crescent-shaped area bordering the inferior temperal retina. In black mice this area constituted 20% of the total retinal area, in albinos 17%. In black mice we counted nearly 1,000 labeled cells in the ipsilateral retina, or 2.6% of all cells filled in both eyes. Albinos had about one-third fewer filled cells ipsilaterally than black mice. Four percent of all ipsilaterally filled cells in black mice and 8% in albinos were scattered outside of the crescent region. The density of ipsilaterally projecting cells was uniform throughout the crescent region in black mice, but decreased toward the central retina in albinos. In retinas contralateral to the injection up to 39,000 cells were filled-about two-thirds of the cells in the ganglion-cell layer whose cytoplasm contained conspicuous Nissl substance. Depending on classification of unfilled cells as ganglion cells or interneurons, we estimated a total of 48,000 to 65,000 ganglion cells to exist in the retina. The size distribution of ipsilaterally projecting ganglion cells was similar in albinos and normals. Ipsilaterally projecting ganglion cells were on average 1.8-3 times larger in volume than contralaterally projecting ones in both types of mice. Displaced ganglion cells were relatively more common in ipsilateral retinofugal projections: 21% of all ipsilateral ganglion cells were displaced versus less than 1% of all the contralateral ganglion cells in black mice. In albinos only 13% of the ganglion cells in the ipsilateral retina were displaced. The overall reduction in ipsilaterally projecting cells in albinos was reflected twice as much in displaced ganglion cells as in normally placed ones.
